<B>Verfahren</B> zur <B>Herstellung eines neuen basischen Äthers.</B> Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung eines neuen basischen Äthers, nämlich des 1-(2'-Oxy-phen- oxy)-2-isopropylamino-propans. Es wurde ge funden, dass dieser Äther das vegetative Ner vensystem beeinflusst. Er soll daher als Phar mazeutikum und als Zwischenprodukt zur Herstellung weiterer Derivate Verwendung finden.
Das den Gegenstand des Patentes bildende Verfahren ist dadurch gekennzeichnet, dass man eine Verbindung der Formel

unter reduzierenden Bedingungen mit einer Verbindung der Formel umsetzt, in welchen beiden Formeln eines der X ein Sauerstoffatom und das andere ein Wasserstoffatom neben einem zur reduktiven Alkylierung befähigten und dabei die NH- Gruppe ergebenden stickstoffhaltigen Rest be deutet.
Man kann erfindungsgemäss beispielsweise 1-(2'-Oxy-phenoxy)-2-oxo-propan mit Isopro- pylamin und einem Reduktionsmittel zur Re aktion bringen. Man kann dabei so vorgehen, dass das Isopropylamin in Gegenwart des an dern Ausgangsstoffes aus- 2-Nitro-propan ge bildet wird. Als Reduktionsmittel kann man die für solche Fälle üblichen, wie z. B. Zink und Mineralsäuren, aktiviertes Aluminium usf., verwenden.
Als weitaus ergiebigstes Re- duktionsmittel ist aber katalytiseh erregter Wasserstoff anzusprechen. Als Katalysatoren sind die Metalle der Raney-Gruppe, die be kannten Platin-Katalysatoren usf., verwend bar. Wird in Gegenwart von Raney-Katalysa- toren reduziert, so arbeitet man vorzugsweise unter Druck; beim Arbeiten in Gegenwart von Platin-Katalysatoren kann der Wasserstoff ohne Druck oder mit nur leichtem Überdruck angewendet werden.
Die Reduktion wird vor teilhaft in einem Lösungsmittel, beispielsweise in niederen Alkoholen, durchgeführt.
Man kann erfindungsgemäss auch 1-(2'- Oxy-phenoxy)-2-amino-propan mit Aceton und einem Reduktionsmittel umsetzen. Dabei kann man das 1-(2'-Oxy-phenoxy)-2-amino- propan in Gegenwart des andern Ausgangs stoffes aus 1-(2'-Oxy-phenoxy)-2-nitro-propan herstellen.
Eine weitere Ausführungsform des Verfahrens besteht darin, dass man 1-(2'-Oxy- phenoxy)-2-oxo-propan mit N-Isopropyl-ben- zylamin zusammen katalytisch reduziert, wo bei man zugleich die reduktive Alkylierung und Debenzylierung vornimmt. Das so erhaltene 1-(2'-Oxy-phenoxy)-2-iso- propylamino-propan siedet unter 12 mm bei 126-l28 C und bildet ein farbloses Öl. Das Chlorhydrat der Verbindung schmilzt bei 128 bis 129 C.
Der neue basische Äther soll als Zwischen produkt und als Pharmazeutikum Verwen dung finden.
Beispiel <I>1:</I> 20,0 g 1-(2'-Oxy-phenoxy)-2-oxo-propan und 21,0 g Isopropylamin werden in 100 em3 Äthanol gelöst und 10 Stunden im Autoklaven bei 20 C (120 atü) katalytisch hydriert.
Als Katalysator wird Raney-Nickel ver wendet.
Der Katalysator wird abgenutscht, das Fil trat eingedampft, der Rückstand in 2n-Salz- säure aufgenommen und ausgeäthert. Aus der salzsauren Lösung wird mit konz. Natronlauge die Base in Freiheit gesetzt und in Äther auf genommen.
Nach dem Trocknen und Verdampfen des Äthers wird der Rückstand bei vermindertem Druck destilliert, wobei man das unter 12 mm bei l26-128 C siedende 1-(2'-Oxy-phenoxy)- 2-isoprokylamino-propan in 55o/oiger Ausbeute gewinnt.
Das Chlorhydrat der Base schmilzt bei 128 bis 129 C.
Beispiel 2: _ 1 Mol 1-(2'-Oxy-phenoxy)-2-oxo-propan und 2,3 Mol 2-Nitro-propan werden in glei cher Weise wie unter Beispiel 1 angegeben zusammen reduziert. Man erhält so 1-(2'-Oxy- phenoxy)-2-isopropylamino-propan in befrie digender Ausbeute.
<I>Beispiel 3:</I> 1 Mol 1-(2'-Oxy-phenoxy)-2-amino-propan und 3 Mol Aceton werden in Gegenwart von Raney-Nickel bei 20 C (150 atü) hydriert.
Die Aufarbeitung erfolgt gleich wie in Beispiel 1.
Man erhält so ebenfalls 1-(2'-Oxy-phen- oxy)-2-isopropylamino-propan in guter Aus beute. <I>Beispiel 4:</I> 1 Mol 1-(2'-Oxy-phenoxy)-2-nitro-propan und 3 Mol Aceton werden unter den gleichen Bedingungen wie im Beispiel 3 beschrieben zusammen reduziert. Die Aufarbeitung er folgt nach Beispiel 1. Man erhält 1-(2'-Oxy- phenoxy)-2-isopropylamino-propan in guter Ausbeute.
<I>Beispiel 5:</I> 1 Mol 1-(2'-Oxy-phenoxy)-2-oxo-propan und 1,5 Mol N-Isopropyl-benzylamin werden in Gegenwart von Raney-Nickel im Autokla ven bei 20 C (150 atü) hydriert. Die Auf arbeitung, die gleich wie in Beispiel 1 angege ben erfolgen kann, liefert 1-(2'-Oxy-phenoxy)- 2-isopropylamino-propan in befriedigender Ausbeute.

According to the invention, for example, 1- (2'-oxy-phenoxy) -2-oxo-propane can be reacted with isopropylamine and a reducing agent. One can proceed in such a way that the isopropylamine is formed from 2-nitro-propane in the presence of the other starting material. As a reducing agent, the usual for such cases, such. By far the most productive reducing agent is catalytically excited hydrogen. As catalysts, the metals of the Raney group, the known platinum catalysts, etc., are usable. This also gives 1- (2'-oxy-phenoxy) -2-isopropylamino-propane in good yield. Example 4: 1 mol of 1- (2'-oxy-phenoxy) -2-nitro-propane and 3 mol of acetone are reduced together under the same conditions as described in Example 3. Working up he follows according to Example 1. 1- (2'-Oxyphenoxy) -2-isopropylaminopropane is obtained in good yield.
<I> Example 5: </I> 1 mol of 1- (2'-oxy-phenoxy) -2-oxo-propane and 1.5 mol of N-isopropyl-benzylamine are in the presence of Raney nickel in an autoclave at 20 C (150 atm) hydrogenated. Working up, which can be carried out in the same way as in Example 1, gives 1- (2'-oxy-phenoxy) -2-isopropylamino-propane in a satisfactory yield.
