Verfahren zur Herstellung eines neuen basisehen Äthers. Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung eines neuen basisehen Äthers, nämlich des 1-(m-Oxy- phenoxy)-2-methylamino-propans. Es wurde .gefunden, dass dieser Äther das vegetative Nervensystem beeinflusst. Er soll daher als Pharmazeutikum und als Zwischenprodukt zur Herstellung weiterer Derivate Verwen dung finden.
Das den Gegenstand des Patentes bildende Verfahren ist dadurch gekennzeichnet, dass man eine Verbindung der Formel

unter reduzierenden Bedingungen mit einer Verbindung der Formel CIH.=Y umsetzt, in welchen beiden Formeln eines der N ein Sauerstoffatom und das andere ein Wasserstoffatom neben einem zur reduktiven Alkylierung befähigten und dabei die -NH- Gruppe ergebenden stickstoffhaltigen Rest bedeutet.
Man kann erfindungsgemäss beispielsweise 1.-(m-Oxy-phenoxy)-2-oxo-propan mit Methyl- amin und einem Reduktionsmittel zur Reak tion bringen. Man kann dabei so vorgehen, dass das Methylamin in Gegenwart des andern Ausgangsstoffes aus Nitromethan gebildet wird. Als Reduktionsmittel kann man die für solche Fälle üblichen, wie z. B. Zink und Mineralsäuren, aktiviertes Aluminium usf., verwenden. Als weitaus ergiebigstes Reduk tionsmittel ist aber katalytisch erregter Was serstoff anzusprechen.
Als Katalysatoren sind die Metalle der Raney-Gruppe, die bekannten Platinkatalysatoren usf., verwendbar. Wird in Gegenwart von Raney-Katalysatoren redu ziert, so arbeitet man vorzugsweise unter Druck; beim Arbeiten in Gegenwart. von Pla tinkatalysatoren kann der Wasserstoff ohne Druck oder mit nur leichtem LTberdruck ange- ivendet werden.
Die Reduktion wird vorteil haft in einem Lösungsmittel, beispielsweise in niederen Alkoholen, durehgeführt. Man kann erfindungsgemäss auch 1-(m-Oxy- phenoxy)-2-amino-propan mit Formaldehyd und einem Reduktionsmittel umsetzen. Dabei kann man das 1-(m-Oxy-phenoxy)-2-amino- propan in Gegenwart des andern Ausgangs stoffes aus 1-(m-Oxy-phenoxy) -2-nitro-propan herstellen.
Eine weitere Ausführungsform des Verfahrens besteht, darin, dass man 1-(m-Oxy- phenoxy)-2-oxo-propan mit N-Methyl-benzyl- amin zusammen katalytiseh reduziert, wobei man zugleich die reduktive Alkylierung und Debenzylierung vornimmt. Das so erhaltene 1-(m-Oxy-phenoxy)-2- methylamino-propan siedet unter Zersetzung. Das Chlorhydrat der Verbindung schmilzt bei 157-158 C.
Der neue basische Äther soll als Zwischen produkt und als Pharmazeutikum Verwen dung finden.
<I>Beispiel 1:</I> 21 g 1-(m-Oxy-phenoxy)-2-oxo-propan (F: 77-78 ) und 10 g Methylamin .werden in 100 ern7- Äthanol gelöst. und zwei Stunden im Autoklav bei 25-30 C (80 atü) kataly tisch h-driert. Als Katalysator wird Raney- Nickel verwendet. Der Katalysator wird abge- nutscht, das Filtrat. eingedampft, der Rück stand in 150 em3 warmem Essigester gelöst.
und mit ätherischer Salzsäure versetzt, bis keine Fällung mehr entsteht. Nach zweimali gem Umkristallisieren erhält man das reine, bei 157-158 C schmelzende Chlorhydrat des 1-(m-Oxy-phenoxy)-2-methylamino-propans in befriedigender Ausbeute als weisse Kristalle, die leicht. löslich in Äthanol und Wasser und schwer löslich in inerten Lösungsmitteln sind.

Beispiel, <I>1:</I> 1 Mol 1-(m-Oxy-phenoxy)-2-nitro-propan und 1 Mol Formaldehyd werden unter den gleichen Bedingungen, wie im Beispiel 3 beschrieben, zusammen reduziert. Die Auf arbeitung erfolgt nach Beispiel 1. Man erhält 1-(m-Oxy-phenoxy)-2-methylamino-propan in guter Ausbeute. <I>Beispiel 5:</I> 1 Mol 1-(m-Oxy-phenoxy)-2-oxo-propan und 1 Mol N-Methyl-benzylamin werden in Gegenwart von Raney-Nickel im Autoklaven bei 30 C (80 atü) hydriert.
Die Aufarbei tung, die gleich wie in Beispiel 1 erfolgen kann, liefert 1-(m-Oxy-phenoxy)-2-methyl- amino-propan in befriedigender Ausbeute.
