Process for preparing an aromatic aeylamidodiol, nitrated in the nucleus. In the main patent, a process for preparing P-1-p-nitrophenyl-2-dichloroacetamidopropane-1,3-diol has been described. The present patent relates to a process for the preparation of another aromatic acylamidodiol, nitrated in the nucleus.
This process is characterized in that one submits to 1-me partial hydrolysis, in the presence of water, the pseudo structural form of a triacylated nitroaminodiol of formula
EMI0001.0011
wherein R 1 and R 3 are acyl radicals, under conditions such that! U-1-p-nitrophenyl-2-bromoacetamidopropane-1,3-diol is formed. The latter is a new substance, crystallized, having a melting point (130-131 C (racemic form).
Either the racemic form or one of the optically active forms, preferably the levogy form of the starting triacylated nitroaminodiol, can be subjected to said hydrolysis. Preferably, the hydrolysis is carried out by means of an alkali metal hydroxide dissolved in an aqueous liquid containing an organic solvent miscible with water.
When carrying out the process, the temperature is usually kept below 50 ° C, and it is preferable to use as a water-miscible organic solvent an aliphatic ketone such as acetone or a water-miscible alcohol. like ethyl alcohol. The best results are obtained with an aqueous liquid consisting of equal parts of water and of an organic solvent miscible with water.
Usually, Yf-1-p-nitrophenyl-2-bromoacetamido-1,3-diacetoxypropane is employed as the starting material.
The product obtained by the process according to the present patent, especially the racemic form and the levorotatory form, has therapeutic value as an antibiotic and is suitable for the preparation of other compounds having antibiotic activity.
Example 1: Is dissolved, at 0 C, 1 g of dl-P-1-p-nitro-phenyl-2-bromoacetamido-1,3-diacetoxypropane in a mixture composed of 50 ems of acetone and 50 em3 of a 0.2 n NaOH solution. After allowing the reaction mixture to stand for one hour, this mixture is neutralized with sulfuric acid and evaporated to dryness under reduced pressure.
Then, the residue is extracted with several hot portions of ethylene dichloride, these combined extracts are concentrated and cooled to obtain dl-P-1-p-nitro-phenyl2-bromoacetamidopropane-1,3-diol, from the point fusion 130-131 C.
This product, which can be purified by recrystallization from a mixture of ethyl acetate and petroleum ether, or else from ethylene dichloride, corresponds to the formula

Example 2:
Is dissolved at 0 C, 1 g of (1) - 1-p-nitro-phenyl-2-bromoacetamido-1,3-diacetoxypropane in a mixture composed of 50 cms of acetone and an equal volume of a 0.2 n solution of NaOH. After allowing the reaction mixture to stand for one hour, it is neutralized with hydrochloric acid and evaporated to dryness under reduced pressure.
The residue is extracted with several hot portions of ethylene dichloride, the extracts then concentrated and cooled to obtain (1) - 1-p-nitrophenyl 2-bromoacetamidopropane-1,3-diol. If desired, this compound can be further purified by recrystallization from ethylene chloride or from a mixture of ethyl acetate and petroleum ether; melting point: 166-168 C; (a) D in alcohol = -3 6.
