Procédé de préparation d'un mélange des isomères de la N-(diméthylamino- méthyl-éthyl)-phénothiazine. Les dérivés de la phénothiazine, substitués sur l'atome d'azote cyclique par un reste di- aleoylaminoaleoyle, sont déjà connus et, en particulier, le brevet français N 917595 et ses additions (auxquels correspondent les brevets suisses N s 257879,<B>261889</B> et 261890)
ont dé crit un procédé de préparation de produits de ce type par action d'un halogénure de di- alcoylaminoalcoyle sur la phénothiazine (ou sur une phénothiazine substituée sur un des atomes de carbone par un ou des restes alcoyles on alcoxy) en présence d'un accepteur d'acide halogéné, en particulier d'amidure de sodium.
Il a maintenant été trouvé que de tels dérivés de la phénothiazine peuvent être obte nus en faisant réagir un halogénure de di- alcoylaminoalcoyle avec le complexe formé par réaction clé la pliénothiazine (ou d'une phéno- thiazine substituée) avec un composé halogéno- oi1gano-magnésien (réactif de Grignard).
On peut ainsi préparer entre autres les dérivés clé la phénothiazine contenant en posi tion 10 un reste -(CRIR.)"-N(Ra) \\ dans lequel Rl et R2 sont identiques ou diffé rents et représentent un atome d'hydrogène ou un groupe méthyle, R..- représente un groupe méthyle ou éthyle et n. est égal à.
2 oii 3 et tel que la chaîne -(CRIR2),ï , dans laquelle les différents groupes (CR,R2) peu vent être identiques ou différents, contienne au plus 3 atomes au total; le noyau phéno- thiazinique peut en outre être substitué par un reste méthoxy en position 3, c'est-à-dire en position méta par rapport. à l'atome de soufre.
Le présent brevet a pour objet un procédé de préparation d'un mélange des isomères de I a N- (diméthyl - anaino - méthyl - éthyl) - phéno- thiazine. Ce procédé est caractérisé en ce que l'on fait réagir un halogéno 2-diméthylamino- 1-propane avec le complexe formé par réaction de la phénothiazine avec un composé halogéno- organo-magnésium (réactif de Grignard).
Le mélange ainsi obtenu possède des pro priétés antihistaminiques, hypnotiques et anti- algiques; il trouve donc des applications en thérapeutique.
Le réactif de Grignard utilisé peut être du type alcoylique, arylique ou arylalcoylique; sa nature spécifique ne présente en effet. aucune importance, à condition qu'il forme avec la phénothiazine servant de matière de départ un complexe du type PMgX, dans lequel P représente le noyau de la phénothiazine et X un atome d'halogène. Les conditions pour la formation de ce complexe sont celles générale ment utilisées pour la réaction de Grignard. Ainsi, les réactifs peuvent être chauffés ensem ble dans un solvant inerte anhydre, tel par exemple qu'un hydrocarbure comme le ben zène.
Lors de la réaction de l'halogéno-2-dimé- thylamino-1-propane avec ledit complexe, il se produit une isomérisation avec formation si multanée des deux isomères de la N-(diniéthyl- ainino-méthy 1-étliv 1) -phénothiazine.
Voici, à titre d'exemple, comment. le pré sent procédé peut être exécuté en pratique On ajoute, en 1-'espace d'une heure et en agitant sons atmosphère d'hydrogène, une solu tion de 6,2 g de phénothiazine dans<B>100</B> cm' de benzène sec chaud à un réactif de Grignard préparé à partir de 1 g de ,magnésium, de 6.2 g d'iodure de méthyle et de 20 em3 d'éther sec.
Après 30 minutes d'ébullition, on ajoute en une heure à. la solution maintenue bouil lante une solution de 6,2 g de chloi-o-2-dinié- thyl-amino-1-propane dans 10 eni de benzène sec et chauffe encore à l'ébullition pendant 1 heure et demie. On refroidit ensuite le mé lange réactionnel, puis le traite par une solu tion aqueuse de chlorure d'ammonium.
On ajoute du chloroforme pour dissoudre une cou che huileuse qui se forme à l'interface eau- benzène. L'extrait benzéno-chloroformique est traité avec -de l'acide chlorhydrique ?N, puis l'extrait acide est alcalinisé à 5-10 par de la soude caustique à 50%. L'huile mise en liberté est extraite au chloroforme. Après avoir chassé le solvant,
le liquide résiduel jaune et visqueux est distillé sons pression réduite et passe à 1.45-15:5 (température prise dans le bain de chauffage) sous 0,08 mm 11;g.

The present patent relates to a process for preparing a mixture of the isomers of I a N- (dimethyl - anaino - methyl - ethyl) - phenothiazine. This process is characterized in that a halo 2-dimethylamino-1-propane is reacted with the complex formed by reaction of the phenothiazine with a halo-organo-magnesium compound (Grignard's reagent).
The mixture thus obtained possesses antihistamine, hypnotic and anti-algesic properties; it therefore finds applications in therapy.
The Grignard reagent used can be of the alkyl, aryl or arylalkyl type; its specific nature does indeed present. of no importance, provided that it forms with the phenothiazine as the starting material a complex of the type PMgX, in which P is the ring of the phenothiazine and X is a halogen atom. The conditions for the formation of this complex are those generally used for the Grignard reaction. Thus, the reagents can be heated together in an anhydrous inert solvent, such as for example a hydrocarbon such as ben zene.
During the reaction of halo-2-dimethylamino-1-propane with said complex, isomerization occurs with so multaneous formation of the two isomers of N- (diniethyl-ainino-methyl 1-ethyl 1) - phenothiazine.
Here's how, as an example. the present process can be carried out in practice. A solution of 6.2 g of phenothiazine in <B> 100 </B> cm is added over 1 hour and with stirring in its atmosphere of hydrogen. 'hot dry benzene to a Grignard reagent prepared from 1 g of magnesium, 6.2 g of methyl iodide and 20 em3 of dry ether.
After 30 minutes of boiling, one adds over one hour to. the solution kept boiling a solution of 6.2 g of chloi-o-2-dinethyl-amino-1-propane in 10 ml of dry benzene and further boil for 1.5 hours. The reaction mixture is then cooled and then treated with an aqueous solution of ammonium chloride.
Chloroform is added to dissolve an oily layer which forms at the water-benzene interface. The benzeno-chloroform extract is treated with -N hydrochloric acid, then the acid extract is made 5-10 alkaline with 50% caustic soda. The released oil is extracted with chloroform.
