<B>Verfahren zur Herstellung eines</B> Biguanid#Derivates. Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung eines Bi- guanid-Derivates, nämlich des Nl-p-Chlor- phenyl-NS-isopropyl-biguanids, welches als Chemotherapeutikum oder als Zwischenpro dukt für die Herstellung von Chemotherapeu- tika wertvoll ist.
Das erfindungsgemässe Verfahren ist da durch gekennzeichnet, dass man Isopropyl- guanyl-thioharnstoff mit p-Chlor-anilin in Ge genwart eines Eiitschwefelungsmittels zur Um setzung bringt.
Der als Ausgangsmaterial für das erfin dungsgemässe Verfahren vei-%vendete Isopro- pylguanyl-thioharnstoff kann durch Umset zung von Isopropyldicyandiamid mit Schwe felwasserstoff hergestellt werden.
Als Entschwefelungsmittel können bei spielsweise die Oxyde und die Salze der Schwermetalle, insbesondere v an Blei, Kupfer, Silber und Quecksilber, verwendet werden.
Bei der Ausführung des erfindungsgemässen Verfahrens werden die Reaktionsteilnehmer vorzugsweise zusammen erhitzt, und zwar zweckmässig in Gegenwart eines Lösungs- bzw. Verdünnungsmittels. Der Isopropylguany 1- t.hioharnstoff kann in freier Form oder in Form seiner Salze, beispielsweise des Hydro chlorids, verwendet werden.
Im letzteren Fall wird der Thioharnstoff zweckmässig in situ durch Zugabe einer äquivalenten Menge einer basischen Substanz, wie z. B. Natriummethylat oder Natriumäthylat, in Freiheit gesetzt. Ausserdem kann man auch das p-Chlor-anilin, sofern dies erwünscht ist, in Form eines Sal zes verwenden.
Das Nl-p-Chlor - phenyl-N ' - isopropyl - bi- guanid, welches nach dem erfindungsgemässen Verfahren erhalten wird, ist als Chemothera- peutikum oder als Zwischenprodukt für Chemotherapeutika wertvoll. Es ist insbeson dere als Antimalariamittel nützlich. Es ist eine starke Base, die mit organischen und an organischen Säuren beständige Salze bildet, die in vielen Fällen in Wasser leicht löslich sind.
Diese Salze lassen sich dadurch erhalten, dass man das Biguanid in einer wässerigen Lösung der Säure löst und hierauf das Wasser abdampft. Zweckmässiger werden sie in trok- kener Form hergestellt, indem man die Kom ponenten in einem organischen Lösungsmittel, beispielsweise in Aceton oder in einem Al kohol, in welchem die Salze schwer löslich sind, vermischt.
Auf diese Weise kann man leicht beispielsweise die Salze mit Essigsäure, Milchsäure, Methansulfonsäure, Methylendi- sa.licy1säure, Methylen-bis-2,3-oxynaphthoe- säure und Salzsäure herstellen.
Im folgenden Beispiel bedeuten die Teile Gewichtsteile.
<I>Beispiel:</I> Ein, Gemisch. von 2,1 Teilen p-Chlor-anilin- hydrochlorid, 2,0 Teilen Isopropylgüanyl- thioharnstoff, 2,7 Teilen Mereuriozy d und 25 Teilen Wasser wird während 2 Stunden unter RLüekfluss gerührt. Es wird hierauf auf 50 bis 80 C abgekühlt Lmd mit 6 Teilen 35pro- zentiger Salzsäure versetzt.
Man filtriert dann das Gemisch und behandelt das Filtrat so lange mit. wässerigem Ammoniak, bis es gegen Kongorot nur noch schwach sauer reagiert. Die feste Substanz wird abfiltriert Lmd aus Wasser umkristallisiert. Man erhält so Nl-p- Chlor - phenyl - N5 - isopropyl-biguanid-hydro- chlorid vom Smp. 243 bis 214 C, welches nach einer üblichen Methode in die freie Base vom Smp. 129 C übergeführt wird.
<B> Process for the preparation of a </B> biguanide derivative. The process according to the invention is characterized in that isopropylguanyl-thiourea is reacted with p-chloro-aniline in the presence of a sulfurizing agent.
The isopropylguanyl thiourea used as starting material for the process according to the invention can be prepared by reacting isopropyl dicyandiamide with hydrogen sulphide.
The oxides and salts of heavy metals, especially lead, copper, silver and mercury, can be used as desulphurising agents.
When carrying out the process according to the invention, the reactants are preferably heated together, expediently in the presence of a solvent or diluent. Isopropylguany 1-t.hiourea can be used in free form or in the form of its salts, for example hydrochloride.
In the latter case, the thiourea is expediently in situ by adding an equivalent amount of a basic substance, such as. B. sodium methylate or sodium ethylate set free. In addition, you can also use the p-chloro-aniline, if so desired, in the form of a salt.
The Nl-p-chloro-phenyl-N'-isopropyl-biguanide, which is obtained by the process according to the invention, is valuable as a chemotherapeutic or as an intermediate for chemotherapeutic agents. It is particularly useful as an antimalarial agent. It is a strong base that forms salts with organic and organic acids, which in many cases are easily soluble in water.
These salts can be obtained by dissolving the biguanide in an aqueous solution of the acid and then evaporating the water. They are more expediently prepared in dry form by mixing the components in an organic solvent, for example in acetone or in an alcohol in which the salts are sparingly soluble.
In this way, for example, the salts with acetic acid, lactic acid, methanesulfonic acid, methylenediic acid, methylene-bis-2,3-oxynaphthoic acid and hydrochloric acid can easily be prepared.
