Verfahren zur Darstellung eines Barbitursäurederivates. Es sind Verfahren zur Darstellung von N'-alkylierten Barbitursäuren bekannt. Sie bestehen zum Beispiel darin, dass man durch Kondensation disubstituierterMalonsäuren oder Cyanessigsäuren oder ihrer Derivate, wie zum Beispiel ihre Ester, Amide, Amidsäureester, Chloride und Nitrile mit geeigneten Harn stoffabkömmlingen, wie zum Beispiel alky- lierten Guanidinen, Thioharnstoffen,
Isoharn- stoffäthern und ähnlichen zunächst die ent sprechenden Zwischenprodukte herstellt und diese in üblicher Weise in die C,C-disub- stituierten N-Alkyl-barbitursäuren umwandelt.
Bei der weiteren Bearbeitung wurde nun gefunden, dass man in besonders vorteilhafter Weise N-monoalkylierte C,C-disubstituierte Barbitursäuren gewinnen kann, wenn man eine bei der Kondensation des Dicyandiamids mit 0,C - disubstituierten Malonsäuren oder Cyanessigsäuren oder deren Derivaten, wie Ester, Amide, Amidsäureester,
Chloride oder Nitrile erhältlichen Cyaniminoverbindungen in üblicher Weise mit alkylierenden Mitteln behandelt und die Alkylierungsprodukte durch Verseifen in die N-monoalkylierten C,C-di- substituierten Barbitursäuren überführt.
Vor den bereits bekannten Darstellungs arten für N-monoalkylierte Barbitursäuren hat die genannte Ausführungsform den Vor zug, dass sie zu reinen Substanzen führt und dass die Ausbeute nicht durch das Entstehen von N,N-disubstituierten Produkten beein trächtigt wird, da in den Cyaniminoverbin- dungen, wie sie durch Kondensation mit Dicyandiamid erhalten werden, nur eine Al- kylierung in 1-Stellung erfolgt.
Zweckmässig geht man bei der Alkylierung von den Salzen der Cyaniminobarbitursäuren aus; jedoch ist es nicht erforderlich, diese abzutrennen, vielmehr gelingt es, die Konden sation, Alkylierung und Verseifung in einem Arbeitsgang durchzuführen, was eine wesent liche Vereinfachung und Verbilligung des Verfahrens bedeutet.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Darstellung von 1,5-Di- methyl-5 -(d-1,2 - cyclohexenyl)-barbitursäure. Das Verfahren ist dadurch gekennzeichnet, dass man auf 2,4-Diimino-3-cyan-5-methyl-5- (d-1,2-cyclohexenyl)-barbitursäure ein Methy- lierungsmittel einwirken lässt und das llZethy- lierungsprodukt mit einem Verseifungsmittel behandelt.
Als Methylierungsmittel verwendet man zum Beispiel Methyljodid, Dimethylsulfat, oder einen aromatischen Sulfonsäuremethyl- ester in Gegenwart von Alkali, zweckmässig Alkalialkoholat. Die Reaktion wird normaler weise in Gegenwart eines Löse- oder Ver- dünnemittels ausgeführt. Zweckmässig kom men dafür Alkohole zur Verwendung. Als Verseifungsmittel kommen vorzugsweise ver dünnte Mineralsäuren in Frage.
Das in der beschriebenen Weise erhält liche Produkt bildet Kristalle vom Schmelz punkt 146 und ist mit dem im Hauptpatent Nr. 163696 beschriebenen Produkt identisch. Es ist in Wasser praktisch unlöslich, löst sich aber in wässerigen Alkalien. Es soll therapeutische Anwendung finden.
<I>Beispiel:</I> 46 Teile Natriummetall werden in 1000 Teilen trockenem Methylalkohol gelöst. Zur Lösung werden noch warm 245 Teile 2,4- Diimino- 3 -cyan-5-methyl- 5- (d-1,5-cyclohexe- nyl)-barbitursäure (Zersetzungspunkt 265o) zugegeben. Die klare Lösung wurde auf<B>15"</B> abgekühlt und mit 250 Teilen Dimethylsulfat versetzt, wobei man dafür Sorge trägt, dass die Temperatur nicht über<B>550</B> steigt.
Nach mehrstündigem Stehen wird der grösste Teil des Methanols abdestilliert und der Rückstand in 100 Teilen 25 o/oiger Schwefelsäure acht Stunden lang am Rückflusskühler gekocht. Die dann abgeschiedene 1,5-Dimethyl-5-(d- 1,2 - cyclohexenyl) - barbitursäure wird abge saugt, gewaschen und nach dem Trocknen aus Essigester umgelöst. Schmelzpunkt 146 0. Das methylierte Zwischenprodukt zeigt aus Methylalkohol umgelöst den Schmelzpunkt <B>2550.</B>

Cyanimino compounds obtainable from chlorides or nitriles are treated in the customary manner with alkylating agents and the alkylation products are converted into the N-monoalkylated C, C-di-substituted barbituric acids by saponification.
Before the already known types of representation for N-monoalkylated barbituric acids, the aforementioned embodiment has the advantage that it leads to pure substances and that the yield is not impaired by the formation of N, N-disubstituted products, since in the cyanimino compounds as obtained by condensation with dicyandiamide, only one alkylation takes place in the 1-position.
In the alkylation, it is expedient to start from the salts of the cyaniminobarbituric acids; however, it is not necessary to separate them off; rather, it is possible to carry out the condensation, alkylation and saponification in one operation, which means a substantial simplification and a lower cost of the process.
The subject of the present patent is a process for the preparation of 1,5-dimethyl-5 - (d-1,2-cyclohexenyl) barbituric acid. The process is characterized in that a methylating agent is allowed to act on 2,4-diimino-3-cyano-5-methyl-5- (d-1,2-cyclohexenyl) barbituric acid and the methylation product is allowed to act with a saponifying agent treated.
The methylating agent used is, for example, methyl iodide, dimethyl sulfate or an aromatic sulfonic acid methyl ester in the presence of an alkali, expediently an alkali metal alcoholate. The reaction is usually carried out in the presence of a solvent or thinning agent. Alcohols are expediently used for this purpose. The saponification agents are preferably dilute mineral acids.
The product obtained in the manner described forms crystals with a melting point of 146 and is identical to the product described in main patent no. 163696. It is practically insoluble in water but dissolves in aqueous alkalis. It should find therapeutic application.
<I> Example: </I> 46 parts of sodium metal are dissolved in 1000 parts of dry methyl alcohol. 245 parts of 2,4-diimino-3-cyan-5-methyl-5- (d-1,5-cyclohexenyl) barbituric acid (decomposition point 2650) are added to the solution while still warm. The clear solution was cooled to 15 "and 250 parts of dimethyl sulfate were added, care being taken that the temperature did not rise above 550.
After standing for several hours, most of the methanol is distilled off and the residue is boiled in 100 parts of 25% strength sulfuric acid for eight hours on a reflux condenser.
