Verfahren zur Darstellune eines Barbitursäurederivates. Durch das Patent Nr. 163696 und Patent Nr. 169179 werden Verfahren zur Darstellung der 1,5-Dimethyl-5(d-1,2-cyclohexenyl)-bar-bi- tursäure geschützt.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Darstellung derselben Bar- bitursäureverbindung. Das Verfahren ist da durch gekennzeichnet, dass man auf 2-Imino- 3-cyan-5-methyl-5 (d-1,2-cyclohexenyl)-barbi- tursäure ein Methylierungsmittel einwirken lässt und das Methylierungsprodukt mit einem Verseifungsmittel behandelt. Als Methylie- rungsmittel verwendet man z.
B. Methyljodid, Dimethylsulfat oder einen aromatischen Sul- fonsäuremethylester in Gegenwart von Alkali, zweckmässig Alkalialkoholat. Die Reaktion wird normalerweise in Gegenwart eines Löse- oder Verdünnemittels ausgeführt. Zweckmässig kommen dafür Alkohole zur Verwendung. Als Verseifungsmittel kommen vorzugsweise ver dünnte Mineralsäuren in Frage.
Das in der beschriebenen Weise erhält liche Produkt bildet Kristalle vom Schmelz- punkt 146 und ist mit dem im Hauptpatent Nr. 163696 beschriebenen Produkt identisch. Es ist in Wasser praktisch unlöslich, löst sich aber in wässerigen Alkalien. Es soll thera peutische Anwendung finden.
Beispiel 46 Teile Natrium löst man in 300 Teilen Methanol, gibt hierzu 92 Teile Dicyandiamid und 254 'feile (d-1,2-Cyclohexerryl)=methyl- malonsäurediäthylester (Kp2.108-110 ) und erhitzt etwa 10 Stunden auf<B>70-80'</B> am Rück- ilusskühler. Zu der erkalteten;
die 2-Imino 3-cyarr-5-methyl-5-(d-1,2-cyclohexenyl)-barbi- tursäure enthaltenden Lösung fügt man hier auf unter Rühren 140 Teile Dimethylsulfat; wobei man dafür Sorge trägt, dass die Tem peratur nicht über<B>55'</B> steigt. Nach mehr stündigem Stehen destilliert man den grössten Teil des Methanols im Vakuum ab und ver kocht den Rückstand mit 1000 Teilen 25 /oiger Schwefelsäure 8 Stunden lang am Rück fiusskühler. Die abgeschiedene 1,5-Dimethyl- 5-(d-1,2-cyclohexenyl)-barbitursäure wird ab gesaugt, gewaschen und nach dem Trocknen aus Essigester umgelöst. F. 146 .
Process for the preparation of a barbituric acid derivative. Processes for the preparation of 1,5-dimethyl-5 (d-1,2-cyclohexenyl) -baric acid are protected by patent no. 163696 and patent no. 169179.
The subject of the present patent is a process for the preparation of the same barbituric acid compound. The process is characterized in that a methylating agent is allowed to act on 2-imino-3-cyano-5-methyl-5 (d-1,2-cyclohexenyl) barbaric acid and the methylation product is treated with a saponifying agent. The methylation agent used is z.
B. methyl iodide, dimethyl sulfate or an aromatic sulfonic acid methyl ester in the presence of alkali, expediently alkali metal alcoholate. The reaction is usually carried out in the presence of a solvent or diluent. Alcohols are expediently used for this. The saponification agents are preferably dilute mineral acids.
The product obtained in the manner described forms crystals with a melting point of 146 and is identical to the product described in main patent no. It is practically insoluble in water but dissolves in aqueous alkalis. It should be used therapeutically.
EXAMPLE 46 parts of sodium are dissolved in 300 parts of methanol, 92 parts of dicyandiamide and 254 parts (d-1,2-cyclohexerryl) = methyl-malonic acid diethyl ester (boiling point 2.108-110) are added and the mixture is heated to 70 for about 10 hours -80 '</B> on the reflux cooler. To the cold;
the solution containing 2-imino 3-cyarr-5-methyl-5- (d-1,2-cyclohexenyl) barbituric acid is added to 140 parts of dimethyl sulfate with stirring; Care must be taken that the temperature does not rise above <B> 55 '</B>. After standing for several hours, most of the methanol is distilled off in vacuo and the residue is boiled with 1000 parts of 25% sulfuric acid for 8 hours on a reflux condenser. The deposited 1,5-dimethyl-5- (d-1,2-cyclohexenyl) barbituric acid is sucked off, washed and, after drying, redissolved from ethyl acetate. F. 146.
