Verfahren zur Darstellung eines Barbitursäurederivates. Gegenstand des vorliegenden Patentes ist ein Verfahren zur Darstellung von 1-Methyl- 5-äthyl-5-phenyl-barbitursäure. Das Verfah ren ist dadurch gekennzeichnet, dass man auf 2 - Imino@- 3 -cyan - 5 - äthyl - 5 - phenylbar bitur- säure ein Methylierungsmittel einwirken lässt und das Methylierungsprodukt der Ver- seifung unterwirft.
Als Methylierungsmittel verwendet man zum Beispiel Methyljodid, Dimethylsulfat oder einen aromatischen Sulfonsäuiemethylester in .Gegenwart von Alkali zweckmässig Alkalialkoholat. Die Re aktion wird normalerweise in Gegenwart eines Löse- oder Verdünnemittels ausgeführt. Zweckmässig kommen dafür Alkohole zur Verwendung. Als Verseifungsmittel kommen vorzugsweise verdünnte Mineralsäuren in Frage.
Das in der beschriebenen Weise erhält liche neue Produkt bildet Kristalle vom Schmelzpunkt <B>176'.</B> Es ist in Wasser prak- tisch unlöslich, löst sich aber in wässerigen Alkalien. Es soll therapeutische Anwendung finden.
<I>Beispiel:</I> 256 Teile 3-Cyan-2-imino-5-phenyl-5- äthylbarbitursäure löst man in 3000, Teilen n-Natronlauge und fügt hierzu unter leb haftem Rühren 20 Teile Dimethylsulfat bei einer 40' nicht übersteigenden Temperatur. Wenn alles Dimethylsulfat in Reaktion ge treten ist, säuert man die alkalische Lösung mit verdünnter Schwefelsäure eiskalt an und hebert die saure Flüssigkeit von dem erst klebrig ausfallenden, bei längerem Reiben aber kristallinisch werdenden Rückstand ab.
Die so gewonnene 1.-Methyl-2-imino-3-cyan- 5 - phenyl - 5 - äthylbarbitursäure von F. 150 bis 151 (aus Methanol umgelöst) kocht man nunmehr mit der fünf- bis sechs.faehen Menge ihres Gewichtes. 20%iger Schwefel säure 5 Stunden am Rüekflusskühler. Nach dem Erkalten saugt man die als schweres Kristallmehl abgeschiedene' 1-Mssthyl-5-phe- nyl-5-äthlylbarbitursäure ab, wäscht mit Wasser nach und löst sie aus der dreizehn fachen Menge Alkohol um.
Man erhält sie so in Form eines mikrokristallinen farblosen Pulvers vom Schmelzpunkt<B>176</B> .
Die als Ausgangsmaterial dienende 3 Cyan - 2 - imino -5-phenyl-5-äthylbarbitursäure kann man zum Beispiel wie folgt erhalten: 46 Teile Natriummetall löst man in 300 Teilen Methanol, gibt hierzu 92 Teile Di- cyan.diamid und 264 Teile Phenyläthylmalon- säurediäthylester und erhitzt das Gemenge etwa 10 Stunden am Rückflusskühler auf 70: bis<B>80'.</B> Hierauf destilliert man das Me thanol ab, löst den Rückstand in 200.0 Teilen Wasser und säuert die mit Fis gekühlte Lö sung mit verdünnter Schwefelsäure bis zur schwach kongosauren Reaktion an.
Man saugt den abgeschiedenen kristallinischen Niederschlag scharf ab, wäscht ihn mit Was ser nach und trocknet ihn im Dampfbad. Die so erhaltene 3:-Cyan-2-imino-5-phenyl-5-äthyl- barbitursäure bildet nach dem Umlösen aus verdünntem Alkohol farblose Nadeln vom F. 223 .
Man kann die Methyliierung auch un mittelbar in der bei der Herstellung,des Aus gangsmaterials anfallenden Lösung vorneh men. So lässt man zum Beispiel ohne den Methylalkohol abzudestillieren, in die Lö sung unter Rühren 140 Teile Dimethylsul- fat einfliessen. und trägt dafür Sorge, dass die Temperatur während des Zulaufes<B>50'</B> nicht übersteigt.
Wenn die Reaktionstempe- ratur zurückgegangen ist, destilliert man dann zum Beispiel den Methylalkohol auf dem Wasserbad ab, versetzt den Rückstand mit der sechsfachen Menge seines Gewichtes 20%iger wässeriger Schwefelsäure und kocht 6 Stunden unter Rühren am Rückflusskühler. Die weitere Aufarbeitung erfolgt wie oben.

<I> Example: </I> 256 parts of 3-cyano-2-imino-5-phenyl-5-ethylbarbituric acid are dissolved in 3000 parts of sodium hydroxide solution and 20 parts of dimethyl sulfate are not added to this while stirring vigorously at a rate of 40 ' excessive temperature.
