Abstract

são fornecidos nesta invenção anticorpos anti-ctla-4 humanos ativáveis compreendendo uma cadeia pesada que compreende um domínio vh e uma cadeia leve que compreende uma porção de mascaramento (mm), uma porção clivável (cm) e um domínio vl. tais anticorpos anti-ctla-4 humanos ativáveis possuem atividade de ligação ao ctla-4 no microambiente tumoral, em que a porção de mascaramento é removido através da clivagem proteolítica da porção clivável por proteases específicas do tumor, mas apresentam ligação grandemente reduzida ao ctla-4 fora do tumor. deste modo, os anticorpos anti-ctla-4 humanos ativáveis da presente invenção retêm a atividade antitumoral enquanto reduz os efeitos colaterais associados com a atividade anti-ctla-4 fora do tumor.
