Abstract
DISPOSITIVO PARA CONTROLAR OPTICAMENTE UMA REAÇÃO QUÍMICA EM UMA CÂMARA DE REAÇÃO QUE COMPREENDE UM FLUIDO REAGENTE, E, MÉTODO PARA CONTROLAR OPTICAMENTE UMA REAÇÃO QUÍMICA EM UMA CÂMARA DE REAÇÃO QUE COMPREENDE UM FLUIDO REAGENTE. A presente invenção se refere a um dispositivo (100) e a um método para controlar opticamente uma reação química em uma câmara de reação (149) que compreende um fluido reagente (114). Em uma modalidade preferencial, a reação química compreende um sequenciamento de ácidos nucleicos em uma grade metálica. Com base em um forte confinamento óptico de luz de excitação (110) e de luz de clivagem (112), a reação de sequenciamento pode ser lida. O sequenciamento passo-a-passo é obtido com o uso de nucleotídeos com porções de bloqueio opticamente cliváveis. Após a leitura, o nucleotídeo incorporado é separado em blocos pela luz de clivagem através do mesmo substrato. Isso assegura que apenas nucleotídeos ligados serão desbloqueados. Para evitar o superaquecimento causado pela luz de clivagem, o fluido reagente é circulado pela superfície do substrato (101).
