A presente invenção refere-se a métodos para modificar células T inativando ambos os genes codificando o receptor de células T e um gene de ponto de controle imunológico para desatrelar o potencial da resposta imunológica. Este método envolve o uso de endonucleases de corte raro específicas, em particular TALEnucleases (endonuclease efetora de TAL) e polinucleotídeos codificando tais polipeptídios, para atingir com precisão uma seleção de genes essenciais nas células T, que estão disponíveis a partir de doadores e a partir de cultura de células primárias. A invenção abre caminho para estratégias de imunoterapia adotiva tradicionais e acessíveis para o tratamento de câncer e infecções virais.
