Abstract

"ENSAIO HOMOGêNEO DE HIBRIDIZAçãO DUPLEX OU TRIPLEX POR MEIO DE MúLTIPLAS MEDIçõES SOB CONDIçõES VARIADAS". A invenção apresenta métodos de ensaio homogêneos para hibridização de ácidos nucleicos, detecção e avaliação. O ensaio inclui obter sinais a partir de uma amostra de teste ambos antes como durante a aplicação de voltage à amostra de teste e correlacionar os sinais, cada um dos quais é indicativo da afinidade de ligação da sonda e do alvo de cada um. O ensaio permite determinar uma extensão de correspondência entre a sonda e o alvo, a medida que a voltage pode ser calibrada de maneira a desestabilizar de forma significativa qualquer hibridização, exceto hibridização perfeitamente complementar. Os sinais cuja magnitude está correlacionada com a afinidade de ligação pode ser condutância elétrica e/ou intensidade de fluorescência. Preferivelmente, ambos os pares de sinais são medidos e comparados de maneira a aumentar a confiabilidade do ensaio. O ensaio pode detectar hibridização específica entre sondas de fita única e alvos de fita dupla não desnaturados para formar triplexes, evitando assim a necessidade de desnaturar os alvos. Os métodos de ensaio podem também ser aplicados a complexos de hibridização duplex.
