La présente invention a pour objet un nouveau procédé de préparation d'amides.
En général, on obtient des amides organiques en traitant directement les esters organiques correspondants par l'ammoniac ou une amine. Mais dans le cas de certains esters organiques, tels que les esters qui sont sujets à altération ou décomposition en milieu alcalin ou en présence d'ammoniac ou d'amines, ce procédé particulier de synthèse des amides ne peut pas être appliqué facilement.
On a découvert maintenant un procédé perfectionné de conversion d'esters en amides qui n'est pas sujet aux limitations des procédés antérieurement décrits.Autre- ment dit, ce procédé perfectionné peut facilement s'appli- quer aux esters organiques qui se décomposeraient dans les conditions usuelles de transformation.
Suivant l'invention, on propose un procédé de préparation d'amides répondant à la formule:

Les conditions de la réaction ne sont pas spécia- lement déterminantes. On mélange l'ester organique à du formiate d'ammonium ou à un sel formiate dtune amine primaire ou secondaire, et on chauffe le mélange à une tèmpérature d'environ 10-1690 C suivant la nature et les propriétés physiques de l'ester. Le temps nécessaire peut varier d'environ 10 minutes à 15 heures. On isole le produit final du mélange réactionnel de façon usuelle et on peut le purifier en le recristallisant par l'alcool de façon usuelle.
Pour constituer l'un des réactifs, on peut utili- ser non seulement le formiate d'ammonium mais les sels formiates d'amines primaires ou secondaires comme le formia- te de méthylammonium ou le formiate de diméthylammonium.
Dans les exemples qui suivent, on décrira parti- culièrement l'invention à propos de la préparation d'une nouvelle amide tétracyclique , la dédiméthylamino-6-démé- thyl-12o(-désoxy-anhydrochlorotétracycline, qui possède une activité antimicrobienne utile sur divers microorganis- mes d'essais représentatifs. Mais l'invention n'est aucune- ment limitée à la préparation des amides tétracylcines, citées seulement à titre d'eexemple, car on peut facile- ment appliquer l'invention lorsqu'il s'agit de convertir en amide correspondante d'autres esters représentés par la formule générale ci-dessus .
On décrira l'invention plus en détail à propos des exemples-précis qui suivent.

EXEMPLE 1.- Préparation de la dédiméthylamino-6-déméthyl-12-désoxy- anhydrochlorotétracycline.-

The present invention relates to a new process for the preparation of amides.
In general, organic amides are obtained by directly treating the corresponding organic esters with ammonia or an amine. But in the case of certain organic esters, such as esters which are subject to alteration or decomposition in an alkaline medium or in the presence of ammonia or amines, this particular method of synthesizing amides cannot be easily applied.
We have now discovered an improved method of converting esters to amides which is not subject to the limitations of the previously described methods. In other words, this improved method can easily be applied to organic esters which would decompose in organic esters. usual processing conditions.
