Description
Description METHOD FOR IMPROVING COLD RESISTANCE OF CHRYSANTHEMUM MORIFOLIUM RMAT. BY MELATONIN Technical Field The present invention relates to a method for improving cold resistance of Chrysanthemum morifolium Rmat. by melatonin, and belongs to the fields of cell biology and biology. Background Chrysanthemum morifolium Rmat., originating from China, is one of the top ten traditional flowers in China and one of the four major cut flowers in the world. Chrysanthemum morifolium Rmat. is loved by people for bright colors and diverse flower types. At present, more than 3,000 varieties of Chrysanthemum morifolium Rmat. are grown in China, and the cultivated area reaches 8,900 hectares. Chrysanthemum morifolium Rmat. is mainly used for landscaping, ground covering, interior decoration, tea making, medicine preparation, etc. Chrysanthemum morifolium Rmat. is a cold-resistant short-day herbaceous plant. Chrysanthemum morifolium Rmat. blooms in autumn when all the other flowers wither and the weather gets colder, so different scales of Chrysanthemum morifolium Rmat. exhibitions are held in autumn across the country. However, as the overground part of Chrysanthemum morifolium Rmat. is vulnerable to freezing damage, the ornamental period of Chrysanthemum morifolium Rmat. is shortened, and the Chrysanthemum morifolium Rmat. exhibitions cannot be held for a long time, which restricts the increase of economic benefits thereof. Therefore, it is very important to improve the cold resistance of Chrysanthemum morifolium Rmat. In the research of cold resistance of Chrysanthemum morifolium Rmat., researchers have done a lot of research work. Zheng Lu et al. have studied the relationship between nutritional characteristics and cold resistance of Chrysanthemum morifolium Rmat.; Ander-son et al. have studied the relationship between the number of foot buds of Chrysanthemum morifolium Rmat. in autumn and cold resistance of different varieties; and Xu Ying and Chen Fadi have studied the relationship between low-temperature treatment and cold resistant adaptability of 8 different varieties of Chrysanthemum morifolium Rmat. However, no study related to effect of melatonin on the cold resistance of Chrysanthemum morifolium Rmat. has been reported.
Melatonin (MT) is also known as pineal hormone, and is named as"melatonin" because of the ability of lightening the skin color of certain fishes, reptiles and amphibians. Melatonin is an indoleamine essential for life. The molecular formula of melatonin is C 13N 2 H 160 2 , and the chemical name thereof is N-acetyl-5 methoxytryptamine. Melatonin is found in most living organisms, including primitive single-celled organisms, microorganisms and algae as well as vertebrates and plants. In recent years, abundant achievements have been made in the study of plant melatonin at home and abroad, and melatonin is considered as a new plant growth regulator and biological stimulant, which can regulate the growth processes such as seed germination, root growth, flowering, leaf senescence and fruit ripening, and can also improve the stress resistance of higher plants by alleviating environmental stresses such as heavy metal, salt and alkali ions, ultraviolet radiation, low temperature and drought and biotic stresses such as pathogenic bacteria and pests. Studies have shown that at low temperature, melatonin can improve the ability to resist cold damage by regulating the synthesis of polyamines in plant cells, thus protecting the cells. When Aloe vera (Haw.) Berg cultured at 25°C is transferred to an environment of 4°C, the content of melatonin is increased. Exogenous melatonin can alleviate the apoptosis of carrot suspension cell line induced by low temperature stress. This suggests that at low temperature, melatonin can improve the ability to resist cold damage by regulating the synthesis of polyamines in plant cells, thus protecting the cells. However, no study related to effect of melatonin on the cold resistance of Chrysanthemum morifolium Rmat. has been reported. Therefore, it is of important practical significance to spray a melatonin solution on leaves of Chrysanthemum morifolium Rmat. to improve cold resistance and obtain plants with high cold resistance. Summary The purpose of the present invention is to obtain a method for improving cold resistance of Chrysanthemum morifolium Rmat. by melatonin, so as to solve the above problem. The present invention can solve the problem by adopting the following technical solution: A method for improving cold resistance of Chrysanthemum morifolium Rmat. by melatonin, comprising the following steps:
In a plastic greenhouse where a Chrysanthemum morifolium Rmat. variety "Huangju" is grown, when the minimum temperature in the plastic greenhouse begins to reach 7°C-8°C, spraying a melatonin solution with a concentration of 0.1 mmol/L on leaves of Chrysanthemum morifolium Rmat. at about 9:00 a.m. according to a standard that no solution drips from the leaves, and spraying the solution once every other 3 days for a total of 3 times. The result shows that plants of Chrysanthemum morifolium Rmat. sprayed with the melatonin solution can grow normally, whereas plants of Chrysanthemum morifolium Rmat. that are not sprayed with the melatonin solution suffer from freezing damage, and outer petals of ligulate flowers become discolored. This indicates that melatonin can improve the cold resistance of Chrysanthemum morifolium Rmat. Beneficial effects: by using the above method, the cold resistance of the whole plants of Chrysanthemum morifolium Rmat. is obviously improved. The effect of freezing damage resistance of the Chrysanthemum morifolium Rmat. in the plastic greenhouse in early winter is higher than 95%. The present invention provides an effective method for improving cold resistance of Chrysanthemum morifolium Rmat. by melatonin, whereby plants of Chrysanthemum morifolium Rmat. that are able to resist frost damage and have prolonged ornamental period in early winter are obtained. Description of Drawings Fig. 1 A plant of Chrysanthemum morifolium Rmat. suffering from freezing damage (control) Fig. 2 A plant of Chrysanthemum morifolium Rmat. not suffering from freezing damage (treatment) Fig. 3 Flowers of Chrysanthemum morifolium Rmat. not suffering from freezing damage (treatment) and suffering from freezing damage (control) Detailed Description To easily understand the means, the creative feature, the purpose and the effect realized by the present invention, the present invention is further elaborated below. By using the method of spraying melatonin, plants of Chrysanthemum morifolium Rmat. with higher cold resistance and reduced early frost damage are obtained. After 3 years of continual spraying of melatonin and follow-up observation of early frost damage on plants of Chrysanthemum morifolium Rmat., it is found that the phenomena of leave and flower withering caused by frost damage are obviously reduced. Thereby, a method for improving cold resistance of Chrysanthemum morifolium Rmat. is obtained. The present invention provides a method for improving cold resistance of Chrysanthemum morifolium Rmat. by melatonin.
