Abstract

La presente se refiere a un proceso para preparar 1,1,3-trimetilindan-4-amina de la fórmula (1), o una de sus sales, enriquecida en uno de sus dos enantiómeros, en particular el enantiómero (R), que comprende la separación quiral de una 2,2,4-trimetil-1,2,3,4-tetrahidroquinolina opcionalmente sustituida de la fórmula (3). La presente también se refiere a un proceso para preparar amidas ópticamente activas de la fórmula (2) partiendo de dichos compuestos de la fórmula (1). Reivindicación 1: Un proceso para preparar el compuesto 1,1,3-trimetilindan-4-amina de la fórmula (1), o una de sus sales, enriquecido en el enantiómero (R) caracterizado porque comprende las siguientes etapas: a) proporcionar una mezcla racémica del compuesto 2,2,4-trimetil-1,2,3,4-tetrahidroquinolina opcionalmente sustituido de la fórmula (3a) que comprende el enantiómero (R) de la fórmula (4a) y el enantiómero (S) de la fórmula (5a), b) tratar dicha mezcla racémica del compuesto de la fórmula (3a) según una secuencia operativa A o una secuencia operativa B, en donde: la secuencia operativa A comprende: b1) convertir mediante una reacción de protección la mezcla racémica del compuesto de la fórmula (3a) en una mezcla racémica del compuesto de la fórmula (3b) que tiene un grupo protector PG para proteger el átomo de nitrógeno, en donde dicha mezcla racémica del compuesto de la fórmula (3b) comprende el enantiómero (R) de la fórmula (4b) y el enantiómero (S) de la fórmula (5b), b2) separar dicho enantiómero (R) de la fórmula (4b) de dicho enantiómero (S) de la fórmula (5b); la secuencia operativa B comprende: b3) separar dicho enantiómero (R) de la fórmula (4a) de dicho enantiómero (S) de la fórmula (5a); b4) convertir por medio de una reacción de protección dicho enantiómero (R) de la fórmula (4a) en un enantiómero (R) de la fórmula (4b); c) someter dicho enantiómero (R) de la fórmula (4b) obtenido en la etapa b2 o en la etapa b4 a una reacción de transposición ácida para formar el compuesto de la fórmula (6); d) sustituir el grupo PG de dicho compuesto de la fórmula (6) con un átomo de hidrógeno para obtener el enantiómero (R) del compuesto de la fórmula (1); donde en las fórmulas mencionadas: * indica un átomo de carbono asimétrico; X es un átomo de halógeno; n es un número entero seleccionado entre: 0, 1, 2 y 3; PG es un grupo -COR¹ o un grupo -COOR², R¹ representa un grupo alquilo C₁₋₆ lineal o ramificado; R² representa un grupo alquilo C₁₋₆ lineal o ramificado o un grupo bencilo.
