Abstract
Reivindicación 1: Un método para analizar variantes de carga del factor de crecimiento endotelial vascular VEGF-Trap, que comprende: cargar una muestra de proteína en un capilar de separación que comprende una mezcla de al menos un anfolito portador, metilcelulosa y un aditivo estabilizante; aplicar un primer voltaje durante un primer período de tiempo predeterminado de manera que el anfolito portador forme un gradiente de pH dentro del capilar; aplicar un segundo voltaje durante un segundo período de tiempo predeterminado para enfocar la migración de las variantes de carga de la proteína dentro del capilar de manera que la carga general de las variantes sea neutra; y detectar y cuantificar las variantes de carga de la proteína. Reivindicación 2: El método de la reivindicación 1, en donde: la detección y cuantificación de variantes de carga comprende: medir la absorbancia para una pluralidad de isoformas de variantes de carga; segregar la pluralidad de isoformas de variantes de carga en regiones aisladas que comprenden al menos una primera región ácida (R1), una segunda región neutra (R2) y una tercera región alcalina (R3); y determinar un porcentaje de isoformas de variantes de carga que caen dentro de cada una de las regiones R1, R2 y R3. Reivindicación 20: Un tubo capilar de iCIEF configurado para su uso en un análisis de variantes de carga de VEGF-Trap, que comprende: un tubo capilar configurado para recibir una proteína, y configurado con una mezcla de anfolito portador, metilcelulosa y un aditivo estabilizante, en donde el tubo capilar incluye un revestimiento de fluorocarbono. Reivindicación 22: Un kit de iCIEF configurado para su uso en un análisis de variantes de carga de VEGF-Trap, que comprende uno o más tubos capilares de acuerdo con cualquiera de las reivindicaciones 19 - 20.
