Abstract
Un método de producción una proteína de interés (POI) cultivando una línea celular eucariota recombinante que comprende un constructo de expresión que comprende un promotor regulable de Pichia pastoris y una molécula de ácido nucleico que codifica una POI bajo el control de la transcripción de dicho promotor, que comprende las etapas de a) cultivar la línea celular con una fuente de carbón basal que reprima el promotor, b) cultivar la línea celular con una cantidad limitada de una fuente de carbono suplementaria que reprima el promotor para inducir la producción de POI a una tasa de transcripción de al menos el 15% en comparación con el promotor pGAP nativo, y c) producir y recuperar la POI; y también un promotor regulable aislado y un respectivo sistema de expresión.
