更には、本発明の発酵乳は、長期間保管後も粘度の変化が小さく、保存安定性に優れている。このような効果が得られる理由は、10℃における硬度が0.1N以下で、10℃における粘度が5000mPa・s以下で、かつ、個数基準の50%粒子径が1.0μm以下であることにより、発酵乳中で形成されたタンパク質粒子が保存中も結合することなく安定した粒構造を保持しているためであると推測される。

タンパク質粒子の真円度が1に近いほどその、そのタンパク質粒子は真円に近いことを意味する。タンパク質粒子の面積および周長は、走査電子顕微鏡(SEM)などを用いて撮影した画像について、画像解析を行うことで求めることができる。画像解析には公知の画像解析ソフトを用いて行うことができる。画像解析ソフトとしては、ImageJ Fijiなどが挙げられる。また、本明細書において、タンパク質粒子の平均真円度とは、画像中に観測される任意に選択した30個のタンパク質粒子の真円度の算術平均値のことである。
加熱殺菌処理の具体例としては、120~150℃で2~3秒間加熱処理する超高温瞬間殺菌(UHT殺菌)法、72~75℃で連続的に15秒以上加熱処理する高温短時間殺菌(HTST殺菌)法、保持式で75℃以上で15分以上加熱処理する高温保持殺菌(HTLT殺菌)法、72℃以上で連続的に15秒以上加熱処理する高温短時間殺菌(HTST殺菌)法および135~150℃で1~4秒間加熱処理する超高温滅菌殺菌(LL殺菌)法などが挙げられる。これらの方法を2種以上組み合わせて行ってもよい。加熱殺菌処理は、HTST殺菌法またはUHT殺菌法であることが好ましい。
別の好ましい一態様として、発酵工程中において、発酵槽内の液面から約15cmの深さで発酵槽の中心近傍の部位、および、発酵槽内の液面から約15cmの深さで発酵槽の内壁近傍の部位のそれぞれの粘度が、これらの部位の粘度の平均値×80~120%の範囲となるように、混合物またはその発酵物を流動させながら発酵を行う態様が挙げられる。この態様においては、発酵槽内の混合物またはその発酵物の液深は60cmを超えることが好ましい。
別の好ましい一態様として、発酵工程中において、発酵槽内の液面から1/4の深さで発酵槽の中心近傍の部位、発酵槽内の液面から1/4の深さで発酵槽の内壁近傍の部位、発酵槽内の液面から2/4の深さで発酵槽の中心近傍の部位、発酵槽内の液面から2/4の深さで発酵槽の内壁近傍の部位、発酵槽内の液面から約3/4の深さで発酵槽の中心近傍の部位、発酵槽内の液面から約3/4の深さで発酵槽の内壁近傍の部位のそれぞれの粘度が、これらの部位の粘度の平均値×80~120%の範囲となるように、混合物またはその発酵物を流動させながら発酵を行う態様が挙げられる。

μ:粘度(Pa・s)
一態様として、撹拌翼やミキサーの回転数や周速などの駆動力を変化させる方法が挙げられる。
また、別態様として、ミキサーと撹拌翼を備えた多機能型の撹拌機などのような複数の撹拌手段を備えた撹拌機を用い、駆動させる撹拌手段の種類またはその組み合わせを変化させる方法が挙げられる。ミキサーと撹拌翼を備えた多機能型の撹拌機を用いた場合の操作の一例として、粘度が低い場合は、撹拌翼のみでの撹拌を行い、粘度が増加した場合、撹拌翼を停止あるいは、撹拌翼での撹拌を行いつつ、ミキサーを駆動させて撹拌を行うなどの方法が挙げられる。この場合においても、撹拌翼およびミキサーの回転数や周速などの駆動力を変化させてもよい。
測定試料の体積基準の90%粒子径については、粒度分布測定装置(ベックマン・コールター製のLS 13 320)を用いて、レーザー回折散乱法にて測定試料の体積基準の粒度分布を測定して求めた。
測定試料の個数基準の10%粒子径、個数基準の50%粒子径、個数基準の90%粒子径および個数基準のモード径については、粒度分布測定装置(ベックマン・コールター製のLS 13 320)を用いて、レーザー回折散乱法にて測定試料の個数基準の粒度分布を測定して求めた。
測定試料の粘度は、以下の方法で測定した。
滅菌検査用カップ(栄研化学(株)製、BE2200、容器サイズ:上径58mm、下径56mm、高さ72mm、容量100ml)に、測定試料をカップ内の液面の高さが約60mmとなる量を入れ、スリーワンモーター(型番:BL1200、新東科学(株)製、撹拌羽の半径:20mm)を用いて、周速(0.8m/s、回転数400rpmの条件で、カップ内の測定試料の、下部(液面から約55mmの深さの位置)、中部(液面から約1/2の深さの位置)および上部(液面から約20mmの深さの位置)について、それぞれ30秒ずつ撹拌したのち、攪拌羽を逆回転させて、カップ内の測定試料の、下部、中部および上部について、それぞれ30秒ずつ撹拌して、前処理を行った。前処理後の測定試料について、回転式B型粘度計(TVB25形粘度計、東機産業(株)製)を用いて、測定温度10℃で、M2ロータ、M3ロータまたはM4ロータを測定試料中に侵入及び回転(30rpm、30秒間)させて測定した。ロータについては、測定試料の粘度が500mPa・s以下の場合は、M2ロータを用い、測定試料の粘度が500~4000mPa・sの場合は、M3ロータを用い、測定試料の粘度が4000mPa・s以上の場合は、M4ロータを用いた。
測定試料の硬度は、強度試験機(島津製作所製のEZ-SX)を用いて、圧縮試験を行って測定した。具体的には以下の方法で測定した。
滅菌検査用カップ(栄研化学(株)製、BE2200、容器サイズ:上径58mm、下径56mm、高さ72mm、容量100ml)に、測定試料をカップ内の液面の高さが約60mmとなる量を入れ、上圧盤(直径10mm、部品番号346-51687-04)を測定試料の表面に配置し、ダウン試験速度1.0mm/秒、ストローク10mm、アップ試験速度1.0mm/秒、ストローク15mmで圧縮試験を行い硬度(N)を測定した。
測定試料のボストウィック粘度は、ボストウィック粘度計を用いて測定した。具体的には、ボストウィック粘度計の水準器で水平に調整し、ゲートを閉じ、ゲート手前のリザーバー部を10℃に調整した測定試料で満たした。トリガーを押し下げゲートを開いてから10秒後の測定試料が流れた長さを測定し、ボストウィック粘度を求めた。
脱脂粉乳((株)明治製)と乳タンパク粉原料(MPC4867、フォンテラ製)と原料水とを混合して、脂肪濃度0.4質量%、タンパク質濃度18.1質量%、固形分濃度33.3質量%の原料ミックスを製造した。この原料ミックスを85℃で120秒間殺菌処理して製造例1の原料ミックスを製造した。製造例1の原料ミックスの体積基準の90%粒子径は10.45μmであった。
脱脂粉乳((株)明治製)と乳タンパク粉原料(MPC4867、フォンテラ製)と、牛乳((株)明治製)と、クリーム((株)明治製)と、原料水とを混合して、脂肪濃度9.8質量%、タンパク質濃度15.1質量%、固形分濃度43.7質量%の原料ミックスを製造した。この原料ミックスを60℃まで加温したのち、均質機(GEA製、ホモゲナイザー NS3015H)を用いて均質化処理を行った。均質化処理は、1次圧10MPa、2次圧5MPaの条件で行った。均質化処理後の原料ミックスを85℃で120秒間殺菌処理して製造例2の原料ミックスを製造した。製造例2の原料ミックスの体積基準の90%粒子径は3.63μmであった。
脱脂粉乳((株)明治製)と乳タンパク粉原料(MPC4867、フォンテラ製)と、牛乳((株)明治製)と、クリーム((株)明治製)と、原料水とを混合して、脂肪濃度10.0質量%、タンパク質濃度13.0質量%、固形分濃度40.5質量%の原料ミックスを製造した。この原料ミックスを60℃まで加温したのち、均質機(GEA製、ホモゲナイザー NS3015H)を用いて均質化処理を行った。均質化処理は、1次圧10MPa、2次圧5MPaの条件で行った。均質化処理後の原料ミックスを85℃で120秒間殺菌処理して製造例2の原料ミックスを製造した。製造例3の原料ミックスの体積基準の90%粒子径は1.40μmであった。
(実施例1)
容量3.5Lの発酵槽に、製造例1の原料ミックスを2500gを入れ、発酵乳スタータとして、ブルガリア菌2038株とサーモフィルス菌1131株をそれぞれ原料ミックスの2.0質量%添加し、原料ミックスと発酵乳スタータとの混合物を、以下の表に示す撹拌回転数にて撹拌しながら発酵を行った(発酵工程)。なお、発酵工程中では、撹拌によって発酵槽内の混合物およびその発酵物には上下循環流が生じており、発酵槽内の混合物及びその発酵物は滞留やよどみはなく、ほぼ均一に撹拌されていた。また、発酵工程中では、撹拌によって発酵槽内の混合物及びその発酵物の粘度はほぼ均一となっており、発酵槽内の液面から約1/4の深さで発酵槽の中心近傍の部位、発酵槽内の液面から約1/4の深さで発酵槽の内壁近傍の部位のそれぞれの粘度が、これらの部位の粘度の平均値×80~120%の範囲であった。
次いで、発酵終了後の発酵物について、以下の表に示す撹拌回転数にて撹拌しながら、発酵物の温度が以下の表に示す品温に達するまで冷却を行い(冷却工程)、発酵乳(タンパク質濃度18.1質量%)を得た。なお、発酵工程および冷却工程において、撹拌には、翼直径0.15m、回転半径0.075mの攪拌翼を用いて行った。また、発酵工程中における原料ミックスの体積基準の90%粒子径は125μm以下であった。
得られた発酵乳は、液状発酵乳であった。また、この発酵乳の体積基準の90%粒子径は32.40μmで、個数基準の10%粒子径は0.055μmで、個数基準の50%粒子径は0.084μmで、個数基準の90%粒子径は0.146μmで、個数基準のモード径は0.073μmで、10℃における硬度は0.0092N未満で、10℃における粘度は310mPa・sで、10℃におけるボストウィック粘度は21.5cm/10秒であった。また、この発酵乳の食感は、滑らかであった。
また、得られた発酵乳について、滅菌検査用カップ(栄研化学(株)製、BE2200、容器サイズ:上径58mm、下径56mm、高さ72mm、容量100ml)に入れ、10℃の温度下で、静置下状態で保管した。保管開始から7日後の発酵乳の硬度は0.0092Nであり、保管後も硬度の低いものであった。
容量3.5Lの発酵槽に、製造例2の原料ミックスを2500gを入れ、発酵乳スタータとして、ブルガリア菌2038株とサーモフィルス菌1131株とをそれぞれ原料ミックスの2.0質量%添加し、原料ミックスと発酵乳スタータとの混合物を、以下の表に示す撹拌回転数にて撹拌しながら発酵を行った(発酵工程)。なお、発酵工程中では、撹拌によって発酵槽内の混合物およびその発酵物には上下循環流が生じており、発酵槽内の混合物及びその発酵物は滞留やよどみはなく、ほぼ均一に撹拌されていた。また、発酵工程中では、撹拌によって発酵槽内の混合物及びその発酵物の粘度はほぼ均一となっており、発酵槽内の液面から約1/4の深さで発酵槽の中心近傍の部位、発酵槽内の液面から約1/4の深さで発酵槽の内壁近傍の部位のそれぞれの粘度が、これらの部位の粘度の平均値×80~120%の範囲であった。
次いで、発酵終了後の発酵物について、以下の表に示す撹拌回転数にて撹拌しながら、発酵物の温度が以下の表に示す品温に達するまで冷却を行い(冷却工程)、発酵乳(タンパク質濃度15.1質量%)を得た。なお、発酵工程および冷却工程において、撹拌には、翼直径0.15m、回転半径0.075mの攪拌翼を用いて行った。また、発酵工程中における原料ミックスの体積基準の90%粒子径は125μm以下であった。
得られた発酵乳は、液状発酵乳であった。また、この発酵乳の体積基準の90%粒子径は28.99μmで、個数基準の10%粒子径は0.057μmで、個数基準の50%粒子径は0.088μmで、個数基準の90%粒子径は0.163μmで、個数基準のモード径は0.073μmで、10℃における硬度は0.0083N未満で、10℃における粘度は382mPa・sであった。また、この発酵乳の食感は、滑らかであった。
また、得られた発酵乳について、滅菌検査用カップ(栄研化学(株)製、BE2200、容器サイズ:上径58mm、下径56mm、高さ72mm、容量100ml)に入れ、10℃の温度下で、静置下状態で保管した。保管開始から14日後の発酵乳の硬度は0.0083Nであり、保管後も硬度の低いものであった。
容量89.1Lの発酵槽に、製造例3の原料ミックスを60000gを入れ、発酵乳スタータとして、ブルガリア菌2038株とサーモフィルス菌1131株とをそれぞれ原料ミックスの2.0質量%添加し、原料ミックスと発酵乳スタータとの混合物を、以下の表に示す撹拌回転数にて撹拌しながら発酵を行った(発酵工程)。なお、発酵工程中では、撹拌によって発酵槽内の混合物およびその発酵物には上下循環流が生じており、発酵槽内の混合物及びその発酵物は滞留やよどみはなく、ほぼ均一に撹拌されていた。また、発酵工程中では、撹拌によって発酵槽内の混合物及びその発酵物の粘度はほぼ均一となっており、発酵槽内の液面から約1/4の深さで発酵槽の中心近傍の部位、発酵槽内の液面から約1/4の深さで発酵槽の内壁近傍の部位のそれぞれの粘度が、これらの部位の粘度の平均値×80~120%の範囲であった。
次いで、発酵終了後の発酵物について、以下の表に示す撹拌回転数にて撹拌しながら、発酵物の温度が以下の表に示す品温に達するまで冷却を行い(冷却工程)、発酵乳(タンパク質濃度13.0質量%)を得た。なお、発酵工程および冷却工程において、撹拌には、翼直径0.42m、回転半径0.21mの攪拌翼を用いて行った。また、発酵工程中における原料ミックスの体積基準の90%粒子径は125μm以下であった。
得られた発酵乳は、液状発酵乳であった。また、この発酵乳の体積基準の90%粒子径は26.78μmで、個数基準の10%粒子径は0.058μmで、個数基準の50%粒子径は0.091μmで、個数基準の90%粒子径は0.178μmで、個数基準のモード径は0.081μmで、10℃における硬度は0.0117N未満で、10℃における粘度は295mPa・sであった。すなわち、冷却後にカード破壊処理などを施さなくても、粘度が低く、目視での凝集沈殿物は殆ど観測されない液状発酵乳を製造することができた。また、この発酵乳の食感は、滑らかであった。
また、得られた発酵乳について、滅菌検査用カップ(栄研化学(株)製、BE2200、容器サイズ:上径58mm、下径56mm、高さ72mm、容量100ml)に入れ、10℃の温度下で、静置下状態で保管した。保管開始から21日後の発酵乳の硬度は0.0117Nであり、保管後も硬度の低いものであった。
容量3.5Lの発酵槽に、製造例3の原料ミックスを2500gを入れ、発酵乳スタータとして、ブルガリア菌2038株とサーモフィルス菌1131株とをそれぞれ原料ミックスの2.0質量%添加し、原料ミックスと発酵乳スタータとの混合物を静置した状態で、発酵物のpHが4.9以下に達するまで発酵を行った。発酵は約43℃の温度で行った。次いで、発酵終了後の発酵物について、カード破壊を行い、次いで、均質機(三和エンジニアリング製、ホモゲナイザー L100-H2-CH)を用いて1次圧10MPa、2次圧5MPaの条件で均質化処理を行い、発酵乳を得た。得られた発酵乳の10℃における粘度は17920mPa・sで、10℃における硬度は0.05Nを超えていた、また、この発酵乳については、粘度が高く粒度分布を測定することができなかった。また、発酵時に肉眼で確認できる粗大な凝集物が生じていた。
また、得られた発酵乳について、滅菌検査用カップ(栄研化学(株)製、BE2200、容器サイズ:上径58mm、下径56mm、高さ72mm、容量100ml)に入れ、10℃の温度下で、静置下状態で保管した。保管開始から21日後の発酵乳の硬度は0.2647Nであった。
実施例3および比較例1の発酵乳について、滅菌検査用カップ(栄研化学(株)製、BE2200、容器サイズ:上径58mm、下径56mm、高さ72mm、容量100ml)に入れ、10℃の温度下で、静置下状態で保管した。保管開始から、5日後、7日後、14日後の粘度を測定した。保管後の発酵乳の粘度は、上述した粘度の測定方法に示した方法に従い測定した。下記表に、粘度変化率の値も併せて併記する。なお、粘度変化率は、初期粘度(保管0日目の発酵乳の粘度)に対する、保管後の発酵乳の粘度の比である。
Claims (8)
- タンパク質の含有量が10.0質量%以上であり、
10℃における硬度が0.1N以下であり、
10℃における粘度が5000mPa・s以下であり、
個数基準の50%粒子径が1.0μm以下である、発酵乳。 - 個数基準の50%粒子径に対する個数基準の90%粒子径の比が1.0~5.0である、請求項1に記載の発酵乳。
- 個数基準の50%粒子径と個数基準のモード径との差の絶対値が1.0μm以下である、請求項1または2に記載の発酵乳。
- 個数基準の90%粒子径と個数基準のモード径との差の絶対値が5.0μm以下である、請求項1または2に記載の発酵乳。
- 10℃における硬度が0.05N以下である、請求項1または2に記載の発酵乳。
- 10℃における粘度が2000mPa・s以下である、請求項1または2に記載の発酵乳。
- 10℃におけるボストウィック粘度が10cm/10秒以上である、請求項1または2に記載の発酵乳。
- 液状発酵乳である、請求項1または2に記載の発酵乳。
