Abstract
본 발명은 염증성 질환의 치료 또는 예방을 위한 서방형 주사용 조성물 및 이의 제조 방법에 관한 것으로, 칸나비디올을 포함하는 마이크로 입자를 이용하여, 1개월 이상 칸나비디올을 지속 방출하여 염증성 질환의 치료 또는 예방의 효과를 나타낼 수 있다. 또한, 독성이 없고 장기간 사용으로 인한 부작용이 없어 장시간 지속 사용이 가능한 칸나비디올을 포함하는 서방형 주사용 조성물로, 1회 주사로 1개월 이상 염증성 질환의 치료 또는 예방 효과를 나타낼 수 있어 복용 편의성을 크게 개선할 수 있다.
Description
본 발명은 염증성 질환의 치료 또는 예방을 위한 서방형 주사용 조성물 및 이의 제조 방법에 관한 것이다.
골관절염(osteoarthritis)은 무릎 연골 손상 및 관절 윤활액의 이상으로 인하여 발생하는 퇴행성 질환으로서, 윤활 관절(synovial joint)에서 연골과 주위골에 퇴행성 변화가 나타나서 생기는 관절염을 말한다. 연골은 관절에서 뼈의 끝 부분을 덮고 있는 미끄러운 조직으로 건강한 연골은 뼈가 서로 움직임으로서 발생하는 충격의 흡수를 돕는다. 골관절염에서는 연골의 상층이 분해되어 마모되는데 이는 연골 아래 뼈들이 직접적으로 접촉하는 것을 허용하게 된다. 이러한 직접적인 접촉은 통증과 부기 및 관절의 운동 손실을 초래하며 그 관절은 시간 이 지남에 따라 정상적인 형상을 잃을 수 있고 골증식증(osteophytosis)에 이를 수 있다. 또한 뼈나 연골의 부스러기가 떨어져나가 관절 공간 내에 떠다닐 수 있으며, 이는 더 큰 통증과 손상을 초래하게 된다.
골관절염에 걸린 사람은 흔히 관절 통증과 운동의 감소를 경험한다. 다른 형태들의 관절염과 달리, 골관절염은 내부기관이 아닌 관절에만 영향을 미치게 된다. 골관절염은 관절염의 가장 흔한 유형이며 두 번째로 흔한 형태의 관절염으로는 자가면역계 이상이 원인인 류머티스 관절염이 있다.
현재 골관절염의 치료를 위해서는, 약물요법, 운동요법 또는 수술요법 등이 사용될 수 있다고 알려져 있다. 이 중 약물 요법으로 아세트아미노펜 (acetaminophen), 비스테로이드성 항염제 (nonsteroidal anti-inflammatory drugs, NSAIDs), 코르티코스테로이드 (corticosteroids) 등의 약물이 사용되고 있다.
하지만 대부분의 약물들은 염증을 치료하여 통증을 완화시키는 것으로 근본적인 연골 퇴행 억제를 하지 못하고 오히려 연골 손상을 가속화시키거나 심혈관, 위장관, 신장, 간 등에 부작용이 문제로 지적되고 있다.
이에 골관절염의 치료 또는 개선을 위한 치료제의 개발이 필요하며, 1회 주사를 통해 장시간 지속적인 약물 방출을 통해 골관절염의 치료 또는 예방 효과를 높일 수 있는 제품에 대한 개발이 필요하다.

본 발명의 목적은 염증성 질환의 치료 또는 예방을 위한 서방형 주사용 조성물 및 이의 제조 방법을 제공하는 것이다.
본 발명의 다른 목적은 칸나비디올을 포함하는 마이크로 입자를 이용하여, 1개월 이상 칸나비디올을 지속 방출하여 염증성 질환의 치료 또는 예방의 효과를 나타낼 수 있는 서방형 주사용 조성물을 제공하는 것이다.
본 발명의 다른 목적은 독성이 없고 장기간 사용으로 인한 부작용이 없어 장시간 지속 사용이 가능한 칸나비디올을 포함하는 서방형 주사용 조성물로, 1회 주사로 1개월 이상 염증성 질환의 치료 또는 예방 효과를 나타낼 수 있어 복용 편의성을 크게 개선한 서방형 주사용 조성물의 제조 방법을 제공하는 것이다.
상기 목적을 달성하기 위하여, 본 발명은 염증성 질환의 치료 또는 예방을 위한 서방형 주사용 조성물로 칸나비디올(CBD, Cannabidiol)을 1개월 이상 지속적으로 방출하는 마이크로 입자를 포함하며, 상기 마이크로 입자는 칸나비디올 및 생분해성 고분자를 포함할 수 있다.
상기 마이크로 입자는 칸나비디올 및 생분해성 고분자를 1:3 내지 1:10의 중량 비율로 포함할 수 있다.
상기 생분해성 고분자는 폴리락트산, 폴리락타이드, 폴리락틱-코-글리콜산, 폴리락타이드-코-글리콜라이드(PLGA), 폴리포스파진, 폴리이미노카보네이트, 폴리포스포에스테르, 폴리안하이드라이드, 폴리오르쏘에스테르, 폴리카프로락톤, 폴리하이드록시발레이트, 폴리하이드록시부티레이트, 폴리아미노산 및 이들의 조합으로 이루어진 군으로부터 선택될 수 있다.
상기 폴리락타이드-코-글리콜라이드는 락타이드:글리콜라이드의 단량체 비율이 60:40 내지 90:10일 수 있다.
상기 염증성 질환은 골관절염일 수 있다.
상기 마이크로 입자는 하기 방출 시험에 의해 18시간 경과 후, 전체 CBD의 중량 대비하여, 35% 내지 80% 방출되는 것일 수 있다:
[방출 시험 조건]
용출 시험액은 10 mM PBS 완충액 (pH7.4) 1L에 Tween 20을 0.05 %(w/v)가 되도록 혼합하여 혼합 용액을 제조하고, 상기 혼합 용액에 아지드화나트륨을 0.02 %(w/v), 아스크로브산나트륨 0.0625 %(w/v)가 되도록 혼합하여 제조하였다.
상기 용출 시험액 100mL를 주사약병에 넣고, 상기 마이크로 입자를 넣어 주었으며, 이때 마이크로 입자는 CBD의 총 중량이 15mg이 되도록 하였다. 상기 주사약병을 완전히 밀봉시킨 후, 50 ℃를 유지하며, 120 rpm 속도로 진탕하여 시험하였으며, 용출 개시 18 시간이 경과되면, 주사약병을 꺼내어 3 분간 방치한 후 주사기를 이용하여 시험액 1 mL를 정확하게 취하여, 3000 rpm으로 3 분간 원심분리 후 상층액을 검액으로 하여 CBD의 방출을 확인하였다.
본 발명의 다른 일 실시예에 따른 염증성 질환의 치료 또는 개선을 위한 서방형 주사용 조성물의 제조 방법은 칸나비디올(CBD) 및 생분해성 고분자를 유기 용매에 용해하여 유상 용액을 제조하는 단계; 계면활성제를 물에 용해하여 수상 용액을 제조하는 단계; 및 상기 유상 용액 및 수상 용액을 혼합하여 에멀젼을 형성하는 단계를 포함할 수 있다.
상기 제조된 에멀젼은 수상 용액 내로 수득하여 잔류 용매를 제거하는 단계; 상기 잔류 용매가 제거된 에멀젼을 세척 및 동결 건조하여 마이크로 입자를 제조하는 단계; 및 상기 동결 건조한 마이크로 입자를 주사 용수와 혼합하는 단계를 추가로 포함할 수 있다.
상기 유상 용액 및 수상 용액은 각 마이크로 채널에 주입하여 흐르게 하며, 상기 유상 용액의 흐름 및 수상 용액의 흐름이 서로 교차하는 지점에서 칸나비디올 및 서방화제를 포함하는 에멀젼을 형성할 수 있다.
상기 유상 용액은 부틸하이드록시톨루엔(BHT)를 추가로 포함할 수 있다.
본 발명은 칸나비디올을 포함하는 마이크로 입자를 이용하여, 1개월 이상 칸나비디올을 지속 방출하여 염증성 질환의 치료 또는 예방의 효과를 나타낼 수 있다.
또한, 전신노출 시 안전역이 넓은 것으로 알려진 칸나비디올을 포함하는 서방형 주사용 조성물로, 장기간 사용으로 인한 부작용이 적고 1회 주사로 1개월 이상 염증성 질환의 치료 또는 예방 효과를 나타낼 수 있어 복용 편의성을 크게 개선할 수 있다.

본 발명은 칸나비디올(CBD, Cannabidiol)을 1개월 이상 지속적으로 방출하는 마이크로 입자를 포함하며, 상기 마이크로 입자는 칸나비디올 및 생분해성 고분자를 포함하는, 염증성 질환의 치료 또는 예방을 위한 서방형 주사용 조성물에 관한 것이다.
이하, 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있도록 본 발명의 실시예에 대하여 상세히 설명한다. 그러나 본 발명은 여러 가지 상이한 형태로 구현될 수 있으며 여기에서 설명하는 실시예에 한정되지 않는다.
본 발명의 칸나비디올(CBD)은 대마에서 분리된 생리활성 물질이며, 대마에서 직접 추출하거나, 추출한 CBD을 이용할 수 있으며, 화학적 합성에 의해 제조될 수 있으며, 이의 염도 모두 포함하는 것으로, 상기 예시에 제한되지 않는다.
대마는 삼으로도 불리며, 종래 옷감으로 활용되었다. 보다 구체적으로 삼의 섬유는 직물, 모기장, 밧줄, 어망, 제지원료 등에 이용되고, 삼의 열매는 기름을 짜서 식용으로 하거나 등불기름, 비누, 바니시, 페인트 등에 이용되고 깻묵은 사료와 비료로 이용된다.
대마의 대표적인 성분 중 하나인 정신 활성 화학 물질인 델타-9-테트라하이드로칸나비놀 (delta-9-tetrahydrocannabinol, THC)은 Cannabis sativa 또는 Cannabis indica 식물 유래 성분으로 일반적으로 마리화나(대마초)라 불리며 불법 약물로 지정하여 관리하는 국가들이 있다. 불법약물로 지정된 국가의 경우 특별한 허가를 받지 아니한 경우, 보관, 취급 등이 불법행위로 규정하고 있다. 또다른 대표 성분은 칸나비디올(cannabidiol, CBD)로 THC에 반해 향정신성 작용이 없는 것으로 알려져 있다. 따라서 의약품 및 화장품 등 사용에 대해 최근 다양한 연구가 진행되고 있다.
세계보건기구(WHO)는 대마 추출물을 원료로 한 칸나비디올(CBD, Cannabidiol) 오일이 뇌전증(간질), 알츠하이머(치매) 등에 효능이 있다고 밝힌 보고서를 발표하였다.
세계도핑방지기구는 ‘2018년 금지목록 국제표준’을 펴내며 올해부터 CBD를 금지 약물에서 제외하였다. CBD는 운동선수들이 통증 치료를 목적으로 널리 쓰는 약물로, 해시시, 마리화나 등 대마초에서 얻을 수 있는 다른 물질은 사용을 금지했지만 의료 목적으로 쓰이는 CBD 오일은 허용하였다.
상기와 같이 다양한 국가에서 의료 목적의 CBD에 대한 사용이 허용되고 있는 실정이다.
다만, 상술한 바와 같이 CBD를 의료 목적으로 사용할 때, CBD 오일 상태로 이용하고 있어 특정 질병의 치료 또는 예방을 위해선, 장시간 지속적인 복용을 해야 하는 점에서, 복용 편의성을 높인 제형은 없는 실정이다.
이에 본 발명에서는, CBD를 이용하여 염증성 질환의 치료 또는 예방을 위한 용도로 이용될 수 있는 주사용 조성물이며, 특히, 1회 주사를 통해 1개월 이상 지속적으로 CBD를 방출할 수 있는 마이크로 입자를 포함하는 것을 특징으로 한다.
상기 마이크로 입자는 CBD 및 생분해성 고분자를 포함하는 것을 특징으로 한다.
이에, 본 발명의 마이크로 입자를 이용하여 주사제로 활용하는 경우, 1회 투여에 의해 지속적인 CBD의 투여 효과를 나타낼 수 있다. 구체적으로, 1회 투여에 의해 1개월 이상 동안 혈중 CBD의 농도를 유효 농도 이상으로 유지할 수 있으며, 1회 투여에 의해 수 개월 동안 혈중 CBD의 농도를 유효 농도 이상으로 유지할 수 있으며, 1회 투여에 의해 3개월 동안 혈중 CBD의 농도를 유효 농도 이상으로 유지할 수 있으며, 1회 투여에 의해 6개월 동안 혈중 CBD의 농도를 유효 농도 이상으로 유지할 수 있으며, 1회 투여에 의해 12개월 동안 혈중 CBD의 농도를 유효 농도 이상으로 유지할 수 있다.
즉, 1개월 이상, 3개월 이상, 6개월 이상, 12개월 이상 지속적인 CBD의 방출 효과를 나타낼 수 있다.
상기와 같은 지속적인 CBD의 방출 효과를 나타내는 것은, 서방화제, 즉 생분해성 고분자의 분해 속도에 영향을 받을 수 있다.
상기 생분해성 고분자는 폴리락트산, 폴리락타이드, 폴리락틱-코-글리콜산, 폴리락타이드-코-글리콜라이드(PLGA), 폴리포스파진, 폴리이미노카보네이트, 폴리포스포에스테르, 폴리안하이드라이드, 폴리오르쏘에스테르, 폴리카프로락톤, 폴리하이드록시발레이트, 폴리하이드록시부티레이트, 폴리아미노산 및 이들의 조합으로 이루어진 군으로부터 선택될 수 있다.
상기 일 예시로, 폴리락티드-코-글리코리드는 락티드에 대한 글리콜리드의 몰비가 약 40:60 내지 약 90:10이며, 약 40:60 내지 약 85:15이며, 약 40:60 내지 약 80:20이며, 약 40:60 내지 약 75:25이며, 약 45:55 내지 약 90:10이며, 약 45:55 내지 약 80:20일 수 있으나, 상기 예시에 제한되는 것은 아니지만, 바람직하게는 폴리락티드-코-글리코리드에서 락티드에 대한 글리콜리드의 몰비는 약 75:25 또는 약 50:50일 수 있다. 상기 락티드에 대한 글리콜라이드의 몰비의 범위를 갖는 폴리락티드-코-글리코리드를 서방화제로 이용하는 경우, 1개월 이상 체내에서 지속적으로 CBD를 방출할 수 있다.
상기 생분해성 고분자는 1종 이상의 폴리락티드 및 1종 이상의 폴리락티드-코-글리코리드를 포함할 수 있다. 본 발명에서 생분해성 고분자는 예를 들어, 폴리락티드 2종, 폴리락티드 1종과 폴리락티드-코-글리코리드 1종, 폴리락티드-코-글리코리드 2종, 폴리락티드 3종, 폴리락티드 2종과 폴리락티드-코-글리코리드 1종, 폴리락티드 1종과 폴리락티드-코-글리코리드 2종 등의 조합을 포함할 수 있고, 특히, 폴리락티드 1종과 폴리락티드-코-글리코리드 1종, 또는 폴리락티드-코-글리코리드 2종을 포함할 수 있으나, 이에 제한되지 않는다.
상기 생분해성 고분자는 2종 이상의 폴리락티드-코-글리코리드를 포함할 수 있다. 상기 생분해성 고분자는 1종의 폴리락티드-코-글리코리드 및 1종의 폴리락타이드를 포함할 수 있으나, 상기 예시에 국한되지 않고 1개월 이상 체내에서 지속적으로 CBD를 방출할 수 있는 것은 제한 없이 모두 사용이 가능하다.
상기 마이크로 입자는 CBD 및 생분해성 고분자를 1:3 내지 1:10의 중량비율로 포함하며, 1:3.5 내지 1:9.5의 중량비율로 포함하며, 1:4 내지 1:9의 중량 비율로 포함될 수 있다. 상기 범위 내로 포함하는 마이크로 입자는 이를 주사용 조성물로 이용할 때, 주사 가능한 용량 범위 내에서 CBD를 지속적으로 방출하는 서방형 주사제로 이용할 수 있다. 즉, 생분해성 고분자 대비 CBD를 상기 범위 값 미만으로 포함하는 경우, CBD를 주사제로 투여하기 위한 총 용량을 맞추기 위해서 주사용 조성물의 양이 증가하게 되고, 이는 사람에게 1회 투여 가능한 주사제의 양을 초과하는 문제가 발생하게 된다. 또한, 생분해성 고분자 대비 CBD를 상기 범위 값을 초과하여 포함하는 경우, CBD가 균일하게 분포하는 마이크로 입자로의 제조가 용이하지 않다.
상기 염증성 질환은 골관절염일 수 있다. 다른 염증성 질환에 대해서도 이용이 가능하며, CBD에 의한 염증성 질환의 치료 또는 개선 효과가 입증된 질병은 제한 없이 모두 적용될 수 있으나, 바람직하게는 골관절염에 대한 치료 또는 개선을 위한 용도로 이용될 수 있다.
상기 마이크로 입자는 하기 방출 시험에 의해 18시간 경과 후, 전체 CBD의 중량 대비하여, 35% 내지 80% 방출되는 것일 수 있다:

상기 CBD 방출 시험은, 용출 시험액을 이용하여 CBD의 지속 방출에 대한 것을 확인한 것으로, 상술한 바와 같이 CBD를 포함하는 마이크로 입자가 1개월 이상 지속적으로 CBD를 방출하기 위해선, 상기 방출 시험에 의해, 18시간 경과 시점에, 전체 CBD의 중량 대비하여 35% 내지 80% 방출되어야 하며, 40% 내지 78% 방츨되어야 하며, 50% 내지 75% 방출되어야 한다. 상기 범위 내에서 CBD가 방출될 때, 마이크로 입자가 1개월 이상 지속적으로 CBD의 방출 효과를 나타낼 수 있다. 즉, 상기 범위 값 미만으로 방출되는 경우, 초기 CBD의 방출 효과가 너무 미비하여 CBD에 의한 염증성 질환의 치료 또는 개선 효과를 나타낼 수 없으며, 상기 범위 값을 초과하여 방출되는 경우는, CBD의 방출양이 지나치게 많아, 1개월 이상 지속적인 CBD 방출 효과를 나타낼 수 없다.
또한, 본 발명의 마이크로 입자는 부틸하이드록시톨루엔(BHT)를 추가로 포함할 수 있다.

상기 화학식으로 표시되는 화합물인 CBD는 공기 중에서 산소와 접촉에 의해 산화가 발생하지 않는 안정적인 화합물이나, 본 발명에서와 같이, 생분해성 고분자를 포함하는 포함하는 마이크로 입자로 제형화하는 경우, CBD의 산화가 발생하는 문제가 있다.
상기와 같이 CBD가 산화되는 경우, 앞서 설명한 바와 같이 노란색으로 색상이 변화될 수 있으며, 산화에 의해 화합물의 구조가 상이해짐에 따라, 효능의 저하가 발생할 수 있다.
상기 부틸하이드록시톨루엔(BHT)는 마이크로 입자 내에 포함되어 CBD의 산화를 방지할 수 있다. 즉, 마이크로 입자 내에서 CBD를 대신하여 부틸하이드록시톨루엔(BHT)가 산화됨으로 인해, 칸나비디올의 산화를 방지할 수 있다.
본 발명의 마이크로 입자는, 후술하는 바와 같이 구형의 마이크로 입자로, 제조 공정에 의해 용매가 완전히 제거되면, 칸나비디올, 서방화제 및 부틸하이드록시톨루엔(BHT)가 균일하게 분포할 수 있다.
이때, 상기 마이크로 입자가 산소와 접촉하는 환경 하에서, CBD의 산화 반응이 일어나게 되는 것이나, 상기 CBD에 비해, 부틸하이드록시톨루엔(BHT)의 산화 반응이 우선하여 일어나게 되어, CBD의 산화 반응을 방지할 수 있다.
상기와 같은 작용에 의해 CBD의 산화 반응을 방지할 수 있는 것이기는 하나, 상기 부틸하이드록시톨루엔(BHT) 이외의 항산화제에 의해 이러한 산화 방지 효과가 나타나는 것은 아니며, 부틸하이드록시톨루엔(BHT)를 포함하는 경우에서, CBD에 앞서 부틸하이드록시톨루엔(BHT)가 산화되어 CBD의 산화 방지 효과가 나타날 수 있다.
상기 부틸하이드록시톨루엔(BHT)는 칸나비디올 및 생분해성 고분자의 전체 중량에 대해, 0.11 중량% 내지 9.9 중량%로 포함되며, 0.2 중량% 내지 9 중량%로 포함되며, 0.3 중량% 내지 8 중량%로 포함되며, 0.5 중량% 내지 5 중량%로 포함될 수 있으며, 상기 범위 내에서 구형의 마이크로 입자로 제조가 가능하고, CBD의 적정 함량 범위를 유지하여 서방형 제형으로 이용이 가능하고, 부틸하이드록시톨루엔(BHT)에 의한 CBD의 산화 방지 효과를 통해 안정성을 높일 수 있다.
본 발명의 다른 일 실시예에 따른 칸나비디올을 포함하는 서방형 주사 조성물은, 상기 칸나비디올을 포함하는 마이크로 입자 및 현탁 용제를 포함할 수 있다.
상기 주사용 조성물은 현탁 용제를 포함할 수 있고, 상기 현탁 용제는 등장화제, 현탁화제 및 용제를 포함한다.
보다 구체적으로, 상기 등장화제는 D-만니톨(D-Mannitol), 말티톨(Maltitol), 솔비톨(Sorbitol), 락티톨(Lactitol), 자일리톨(Xylitol), 염화나트륨(Sodium chloride) 및 이의 혼합으로 이루어진 군으로부터 선택될 수 있으며, 바람직하게는 D-만니톨이지만, 상기 예시에 국한되지 않는다.
상기 현탁화제는 카르복시메틸셀룰로오스나트륨(Soduim Carboxymethylcellulose), 폴리소르베이트80(Polysorbate 80), 녹말(starch), 녹말 유도체, 다가알콜류, 키토산(chitosan), 키토산 유도체, 셀룰로스(cellulose), 셀룰로스 유도체, 콜라겐(collagen), 젤라틴 (gelatin), 히알루론산(hyaluronic acid, HA), 알긴산(alginic acid), 알진(algin), 펙틴(pectin), 카라기난(carrageenan), 콘드로이틴(chondroitin), 콘드로이틴 설페이트(chondroitin sulfate), 덱스트란(dextran), 덱스트란 설페이트(dextran sulfate), 폴리라이신(polylysine), 티틴(titin), 피브린(fibrin), 아가로스 (agarose), 플루란(fluran), 잔탄검(xanthan gum) 및 이의 혼합으로 이루어진 군으로부터 선택되며, 바람직하게는 카르복시메틸셀룰로오스나트륨 및 폴리소르베이트 80이지만, 상기 예시에 국한되지 않는다.
상기 용제는 주사용수(Injection water)를 이용할 수 있으며, 주사용수로 사용가능한 용제는 제한 없이 모두 사용 가능하다.
본 발명의 다른 일 실시예에 따른 칸나비디올(CBD)을 포함하는 마이크로 입자의 제조 방법은 CBD 및 생분해성 고분자를 유기 용매에 용해하여 유상 용액을 제조하는 단계; 계면활성제를 물에 용해하여 수상 용액을 제조하는 단계; 및 상기 유상 용액 및 수상 용액을 혼합하여 에멀젼을 형성하는 단계를 포함할 수 있다.
상기 제조 방법은 CBD, 서방화제 및 항산화제를 유기 용매에 용해하여 제조한 유상 용액과 계면활성제를 포함하는 수상 용액을 이용하여, 에멀젼을 형성할 수 있다.
후술하는 바와 같이 본 발명은, 마이크로 채널을 이용한 미세 유체법에 의해 상기 에멀젼을 제조하는 것이기는 하나, 상기 제조 방법에 제한되지 않고, 용매 증발법, 멤브레인법 등의 마이크로 입자 제조 방법을 모두 적용할 수 있다.
상기 유기 용매는 CBD 및 생분해성 고분자를 완전히 용해시킬 수 있는 것을 이용할 수 있다. 구체적으로, 클로로포름, 클로로에탄, 디클로로에탄, 디클로로메탄, 트리클로로에탄, 염화메틸렌, 메탄올 및 이들의 혼합물로 이루어진 군으로부터 선택된 어느 하나 이상의 것이며, 바람직하게는 염화메틸렌, 메탄올 및 이들의 혼합에서 이루어진 군에서 선택될 수 있다. 상기 유기 용매는 CBD 및 생분해성 고분자를 완전히 용해시킬 수 있는 것은 상기 예시에 국한되지 않고, 당업자가 쉽게 선택할 수 있는 유기 용매라면 모두 사용 가능하다.
상기 유기 용매 내 CBD 및 생분해성 고분자는 1:3 내지 1:10의 중량비율로 포함하며, 1:3.5 내지 1:9.5의 중량비율로 포함하며, 1:4 내지 1:9의 중량 비율로 포함될 수 있다.
상기 유상 용액 내의 생분해성 고분자는 10 내지 20 중량% 포함하며, 바람직하게는 12 내지 18 중량%이며, 보다 바람직하게는 15 중량%이나, 상기 예시에 국한되지 않는다. 상기 범위 값 미만으로 포함하는 경우는 에멀젼의 제조가 불가하며, 상기 범위 값을 초과하여 포함하는 경우에는 유상 용액의 점도가 너무 높아 에멀젼으로 제조가 용이하지 않은 문제가 있다.
상기 생분해성 고분자는 폴리락트산, 폴리락타이드, 폴리락틱-코-글리콜산, 폴리락타이드-코-글리콜라이드(PLGA), 폴리포스파진, 폴리이미노카보네이트, 폴리포스포에스테르, 폴리안하이드라이드, 폴리오르쏘에스테르, 폴리카프로락톤, 폴리하이드록시발레이트, 폴리하이드록시부티레이트, 폴리아미노산 및 이들의 조합으로 이루어진 군으로부터 선택되며, 바람직하게는 폴리락타이드-코-글리콜라이드(PLGA) 및/또는 폴리락타이드(PLA)이지만, 상기 예시에 국한되지 않는다.
상기 유상 용액에는 부틸하이드록시톨루엔(BHT)를 추가로 포함할 수 있다. 상술한 바와 같이, 상기 부틸하이드록시톨루엔(BHT)은 유상 용액에 포함되어, 마이크로 입자 내 포함될 수 있고, 이로 인해 CBD의 산화를 방지하는 역할을 수행할 수 있다. 상기 부틸하이드록시톨루엔(BHT)는 칸나비디올 및 생분해성 고분자의 전체 중량에 대해, 0.11 중량% 내지 9.9 중량%로 포함되며, 0.2 중량% 내지 9 중량%로 포함되며, 0.3 중량% 내지 8 중량%로 포함되며, 0.5 중량% 내지 5 중량%로 포함될 수 있다.
상기 수상 용액에 포함되는 계면활성제는 0.1 내지 1.0 중량%이며, 0.2 내지 0.5 중량%이며, 0.25 중량%로 포함될 수 있다. 나머지는 모두 물이다.
상기 계면활성제는 메틸셀룰로오스, 폴리비닐피롤리돈, 레시틴, 젤라틴, 폴리비닐알코올, 소르비탄 모노올레이트(예를 들어, 스판 80(SpanTM 80) 등), 폴리옥시에틸렌 소르비탄 지방산 에스테르(예를 들어, 트윈 80(TweenTM 80) 등), 폴리옥시에틸렌 피마자유 유도체, 라우릴 황산 나트륨, 스테아르산 나트륨, 에스테르 아민, 리니어 디아민, 패티 아민 및 이들의 혼합물로 이루어진 군으로부터 선택된 어느 하나 이상의 것이며, 바람직하게는 폴리비닐알코올일 수 있으나, 상기 예시에 국한되지 않고 완전한 구 형상의 에멀젼으로 제조할 수 있는 계면활성제는 모두 사용 가능하다.
앞서 설명한 바와 같이 유상 용액 및 수상 용액을 각각 제조한 후, 이를 이용하여 에멀젼을 제조하는 방법은 제한되지 않는다.
다만, 본 발명에서는 미세 유체법에 의해 마이크로 입자를 제조하는 방법에 대해 설명하고자 한다.
상기 미세 유체법을 이용하기 위한 마이크로 칩은, 웨이퍼 또는 유리 기판 상에 형성될 수 있다. 상기 마이크로 칩에는 마이크로 채널이 형성되며, 상기 마이크로 채널은 보다 구체적으로, 유상 용액이 흐르는 채널, 수상 용액이 흐르는 채널 및 이송 채널을 포함하며, 상기 유상 용액이 흐르는 채널 및 수상 용액이 흐르는 채널은 하나의 지점에서 서로 만나도록 형성되며, 상기 이송 채널의 일단은 2개의 채널이 접합하는 부분에 연결될 수 있다.
상기 유상 용액이 흐르는 채널 및 수상 용액이 흐르는 채널에는 각각 유상 용액 및 수상 용액을 주입하기 위한 주입부가 결합되어 있으며, 상기 이동 채널의 일단에는 에멀젼을 포함하는 용액을 회수하기 위한 회수부가 결합될 수 있다.
상기 마이크로 채널은 유리 기판, 실리콘 웨이퍼 또는 고분자 필름으로 이루어진 군으로부터 선택된 소재에 형성될 수 있으나, 상기 소재의 예시는 상기 예시에 국한되지 않고, 마이크로 채널의 형성이 가능한 소재는 모두 사용 가능하다.
상기 고분자 필름은 폴리이미드(Polyimide), 폴리에틸렌(Polyethylene), 플루오르화에틸렌프로필렌(Fluorinated ethylene propylene), 폴리프로필렌(Polypropylene), 폴리에틸렌 테레프탈레이트(Polyethylene terephthalate), 폴리에틸렌 나프탈레이트(Polyethylene naphthalate), 폴리술폰(Polysulfone) 및 이들의 혼합으로 이루어진 군으로부터 선택될 수 있으나, 상기 예시에 국한되지 않는다.
일 예시로, 실리콘 웨이퍼에 e-beam evaporator를 이용하여 알루미늄을 증착하며, 포토리소그래피(photolithography) 기법을 이용하여 포토레지스트(photoresist)를 알루미늄 위에 패터닝한다. 이후, 포토레지스트를 마스크로 이용하여 알루미늄 식각(etching)하고, 포토레지스트를 제거한 후 알루미늄을 마스크로 하여 실리콘을 DRIE(deep ion reactive etching)로 에칭하고, 알루미늄 제거 후 웨이퍼 위에 유리를 양극 접합하여 밀봉하여, 상기의 마이크로 채널을 제조한다.
상기의 마이크로 채널은 평균 직경이 60 내지 150㎛이며, 바람직하게는 80 내지 100㎛이지만, 예시에 국한되지 않는다. 상기 범위 값 미만의 평균 직경을 갖는 마이크로 채널을 이용하는 경우, 직경이 너무 작은 에멀전으로 제조되어, 유효한 약물의 방출 및 생체내 흡수에 영향을 미칠 수 있다.
또한, 마이크로 채널의 평균 직경이 상기 범위 값을 초과하는 경우, 제조된 마이크로 입자의 평균 크기가 120㎛ 초과하게 되고, 주사제로 투여 시 이물감 및 통증이 증가될 수 있으며 마이크로 채널의 직경이 커질 수록 제조된 입자의 입도분포가 커져 균일한 입도의 마이크로 입자를 제조하기 어렵다.
또한, 상기 마이크로 채널의 평균 직경은 입자의 평균 직경과 밀접하게 관련되지만, 유상 용액 및 수상 용액의 유량비(μl/min)와도 밀접한 관련이 있다.
또한, 상기 마이크로 채널의 단면 폭(w) 및 단면의 높이(d)는 제조되는 마이크로 입자의 평균 직경(d')과 밀접한 관련이 있다. 상기 마이크로 채널 단면의 폭(w)은 마이크로 입자의 평균 직경(d')에 대해 0.7 내지 1.3의 비율 범위이며, 마이크로 채널 단면의 높이(d)는 마이크로 입자의 평균 직경(d')에 대해 0.7 내지 1.3의 비율 범위이다.
즉, 제조하고자 하는 마이크로 입자의 평균 직경(d')이 결정되면, 이에 따라, 마이크로 채널 단면의 폭(w) 및 높이(d)의 길이는 d'의 0.7 내지 1.3의 비율 범위로 설정해야만, 원하는 크기의 마이크로 입자로의 제조가 가능하다.
상기 마이크로 칩을 이용하여 마이크로 입자를 제조하기 위해, 상기 유상 용액이 흐르는 채널에는 유상 용액을 주입하고, 수상 용액이 흐르는 채널에는 수상 용액을 주입하여 상기 두 개의 채널이 접합하는 부분에서 에멀젼을 형성할 수 있다.
상기 유상 용액 및 수상 용액을 각각 마이크로 채널로 주입하는 경우, 유상 용액 및 수상 용액의 유량비는 1:10 내지 1:50이며, 1:15 내지 1:40이며, 1:15 내지 1:30이며, 1:15 내지 1:25일 수 있다. 상기와 같이 유상 용액 및 수상 용액의 유량 비를 조절하여 균일한 직경을 갖는 마이크로 입자로의 제조할 수 있다.
상기 접합하는 부분에서 형성된 에멀젼은 앞서 제조한 수상 용액, 즉 계면활성제 및 물의 혼합 용액을 이용하여 수득할 수 있다. 상기 제조된 수상 용액은 마이크로 채널로 주입하여 에멀젼의 형성에도 이용하고, 수조에 채워 수집된 에멀젼들이 뭉치는 현상을 방지하는데 이용될 수 있다.
상기 수조 내 수집된 에멀젼은 잔류 유기 용매를 제거할 수 있다. 상기 잔류 유기 용매를 제거하는 단계는, 일정한 온도 조건 및 교반 속도로 교반하여, 에멀젼의 내부에 존재하는 잔류 유기 용매를 증발시켜 제거할 수 있다. 이때, 교반 조건은 15 내지 20℃에서 50 내지 70분 동안 1차 교반하는 단계; 20 내지 40℃에서 50 내지 70분 동안 2차 교반하는 단계; 및 40 내지 60℃에서 1 내지 3시간 동안 3차 교반하는 것이다.
상기 교반 속도는, 1차 내지 3차 교반 단계 모두 동일하게 진행하였으며, 교반 속도는 300 내지 500 rpm이며, 400rpm일 수 있다.
상기와 같이, 온도 조건을 교반 공정의 진행에 따라, 점진적으로 상승시켜 교반하는 것을 특징으로 하며, 온도를 단계적으로 상승시킴에 따라, 에멀젼의 내부에 존재하는 유기 용매의 증발 속도를 조절할 수 있다.
유상 용액 및 수상 용액이 마이크로 채널을 흐를 때의 온도 또한 5 내지 20℃이며, 바람직하게는 10℃이다. 즉, 마이크로 채널을 흐르고, 교차점을 형성하여 에멀젼을 생성한 이후, 수집된 에멀젼을 1차 교반할 때 까지는 일정하게 5 내지 20℃로 저온을 유지한다. 에멀젼의 제조 과정에서 저온을 유지해야만, 구형의 입자를 제조 및 유지할 수 있다. 즉, 저온 조건이 아닌 경우에는 일정한 구 형상의 입자를 제조하기 어려운 문제가 발생한다.
이후, 2차 교반 공정은 온도를 점진적으로 상승시키고, 교반 시간을 늘려, 에멀젼의 내부에 존재하는 유기 용매가 서서히 표면으로 이동하여, 표면에서 유기 용매가 증발됨에 따라, 에멀젼의 성상에 미치는 영향을 최소화할 수 있다. 즉, 급격하게 유기 용매가 증발되는 경우, 유기 용매의 증발에 의해 최종적으로 제조된 마이크로 입자의 표면이 매끈하지 못하고, 기공이 형성되는 문제가 발생할 수 있다. 이러한 문제를 방지하고자, 상기와 같이 온도 조건을 점진적으로 상승시키고, 교반 공정을 진행하는 시간 또한 증가시켜, 유기 용매의 증발 속도를 조절할 수 있고, 이러한 유기 용매의 증발 속도 조절로 인해, 제조된 마이크로 입자의 표면 성상을 제어할 수 있다.
상기 3차 교반 공정은 에멀젼 내 유기용매가 외부 수상으로 추출된 이후 외부 수상을 유기용매 끓는점 근처의 온도로 높여 교반함으로써 포화된 유기용매를 수상으로부터 제거하여 에멀젼 내 잔류 유기용매 제거가 용이하게 한다.
마지막으로 잔류 유기 용매를 제거한 마이크로 입자를 세척 및 건조하는 단계로, 교반하여 표면의 유기 용매를 모두 제거한 마이크로 입자를 제균 여과된 정제수로 수 차례 세척하여 마이크로 입자에 잔존하는 계면활성제를 제거하고, 이후 동결 건조한다.
최종적으로 생성된 마이크로 입자는 구형의 생분해성 고분자로 이루어진 마이크로 입자에 CBD가 고르게 분포되어 있는 형태이며, CBD 및 생분해성 고분자를 1:3 내지 1:10의 중량비율로 포함할 수 있다.
또한, 앞서 설명한 바와 같이, 최종적으로 생성된 마이크로 입자는 CBD 이외에 부틸하이드록시톨루엔(BHT)가 고르게 분포하고 있어, 공기 중의 산소와 접촉 시, 부틸하이드록시톨루엔(BHT)가 우선하여 산화되어 CBD의 산화를 방지할 수 있다.
상기 마이크로 입자 내에 포함된 CBD 및 생분해성 고분자의 중량 비율은 에멀젼의 제조 시, 유상 용액에서의 중량 비율과 동일한데, 이는 마이크로 채널을 통과하여 에멀젼을 형성하고, 에멀젼 내 유기 용매를 모두 제거함에 따라, 유상 용액 내에서의 중량 비율과 동일한 비율로 CBD 및 생분해성 고분자를 함유한 마이크로 입자를 제조할 수 있다.
상기 마이크로 입자는 평균 직경이 30 내지 70㎛이며, 30 내지 65㎛이며, 30 내지 60㎛일 수 있다. 또한 평균 직경에 대한 표준 편차가 1 내지 30㎛이며, 1 내지 20㎛이며, 1 내지 10㎛이며, 1 내지 7㎛일 수 있다. 상기 직경 범위 내에서 균일한 입자로 제조가 가능함을 확인할 수 있으며, 상기 균일한 입자들을 이용하여 서방형 주사제로 사용 시, 이물감을 줄여서, 투약 편의성을 높일 수 있으며, 체내 주입 시 초기 과방출을 방지하고 지속적인 CBD 방출 효과를 나타낼 수 있다.
제조예
마이크로 입자의 제조
1) 유상 용액 제조
칸나비디올, 생분해성 고분자로 락타이드·글리콜라이드 공중합체, 락타이드 공중합체 단독 또는 혼합 및 부틸하이드록시톨루엔(BHT)를 염화메틸렌에 (Methylene Chloride) 용해하여 유상 용액을 제조하였다. 이때, 상기 유상 용액 내의 생분해성 고분자는 15중량%의 비율로 포함하며 칸나비디올 및 생분해성 고분자의 중량 비는 1:3 내지 1:10이다.
2) 수상 용액 제조
폴리비닐알콜을 물에 용해하여 수상 용액을 제조하였으며 수상 용액 내 폴리비닐알콜은 0.25중량%로 포함하였다.
3) 마이크로 입자 제조
상기 유상 용액 및 수상 용액을 실리콘 웨이퍼 상에 형성된 마이크로 채널에 주입하며 마이크로 입자를 제조하였다. 이때의 유상 대 수상의 유량비는 1:20이며, 온도 조건은 10.0℃로 유지하였다. 생산된 마이크로 입자는 수상 용액이 담긴 수조 내 수집되며, 10.0℃에서 1시간, 30.0℃에서 1시간, 45.0℃에서 2시간 400rpm 속도로 교반하였다.
4) 세척 및 수득
교반 완료한 마이크로 입자는 제균 여과된 정제수로 세척하고 동결건조한 후 파우더 형태로 수득하였다.
하기 표 1에 기재된 성분 및 함량으로 후술하는 방법으로 미립구를 제조하였다.

실험예 1
CBD 방출 실험
1) 표준액 (Standard solution) 제조
칸나비디올 표준품을 약 15mg 취하여 100mL 볼륨 플라스크에 넣는다. 희석액을 (아세토니트릴) 약 50mL 투입하고 10분간 초음파 처리하였다. 충분히 식힌 후 시험액으로 표선을 맞추고 0.45μm 필터로 (수용성 PTFE) 필터링한 후 표준액으로 사용하였다(농도: 0.15mg/mL).
2) 용출 시험액의 제조
10 mM PBS 완충액 (pH7.4) 1 L에 Tween 20을 0.05 %(w/v)가 되도록 한 뒤 상기 혼합액에 아지드화나트륨을 0.02 %(w/v) 및 아스크로브산나트륨0.0625 %(w/v)이 되도록 혼합하여 용출 시험액을 제조하였다.
3) 검액 (Test solution) 제조
각 검체는 주성분으로써 15mg이 되도록 정밀하게 달아 주사약병에 넣고 용출시험액 100 mL를 정확하게 넣어 밀봉시킨 후 50 ℃ 부근의 일정한 온도로 120 rpm 속도로 진탕하여 시험하였다. 주사 약병은 가로로 눕혀서 넣었다. 용출 개시 2 시간, 18 시간 및 72 시간마다 검체가 든 주사약병을 꺼내어 3 분간 검액 속 검체가 가라앉도록 방치한 후 주사기를 이용하여 시험액 1 mL를 정확하게 취하여, 3000 rpm으로 3 분간 원심분리 후 상층액을 검액으로 하였다.
4) HPLC조건
- 검출기 (Detector): UV (220nm)
- 컬럼 (Column): ODS column (4.6*150mm, 5μm)

상기 실험 조건에 의해 CBD의 용출 실험을 진행한 결과는 도 1 및 표 2와 같다:

상기 실험 결과에 의하면, 비교예 1은 2시간 경과 시점에 44.5%의 CBD가 방출되고, 18시간이면 82.6%, 72시간에는 97.9%로 방출이 거의 완료되는 것을 확인할 수 있다. 이는 초기 CBD의 방출량이 커서 장시간 지속적인 CBD의 방출 효과를 나타내지 못하는 문제가 있다.
또한, 비교예 2의 경우는 72시간 경과 시점에 47.9%로 방출되어 장시간 지속 방출의 효과를 나타낼 수 있기는 하나, 2시간 경과 시점에 3%의 CBD만 방출되어 초기 CBD의 방출량이 너무 적어, CBD에 의해 염증성 질환의 치료 및 개선 효과가 미비한 문제가 있다.
반면, 실시예 1 내지 5의 경우는, 2시간 경과 시점에 적절한 정도의 CBD 방출 효과를 나타낼 수 있으며, 72시간 경과 시점에도 방출이 완전히 완료되지 않아 장시간 지속적인 약물 방출 효과를 나타냄을 확인할 수 있다.
실험예 2
Rat 약동학 시험
Rat 약동학의 실험을 진행하기 위한 실험 조건은 하기와 같다:

대조군으로 CBD Isolate Solution을 이용하였다. 실험 동물은 생후 6주령의 수컷 SD rat을 코아텍에서 구입하여 일주일간 적응시킨 후, 시험을 진행하며 실험식이는 실험동물용 일반 사료(오리엔트)를 공급하고 물과 사료는 자유롭게 섭취할 수 있도록 사육하였다.
일반증상
실험기간 중, 모든 동물에 대하여 매일 2회 (오전, 오후) 일반증상의 변화를 관찰하였으며, 체중은 도입 시, 군 분리 시 측정하였다.
혈액 샘플링
순화가 완료된 rat에 시험물질 SC 투여 전 0 time 및 시험물질 투여 후 4hr, 8hr, 24hr, 3d, 5d, 1w, 2w, 4w, 6w, 8w, 12w 총 14번의 각각의 time point에 jugular vein으로부터 혈액을 500 ~ 1000μL 채취한 후 즉시 1000g에서 10분간 원심분리하여 serum을 따로 분리하고 -70℃에 동결 보관하였다.
상기 실험에 의해, 실시예 1 내지 5의 마이크로 입자에 대한 pK 측정한 결과는 하기 표 4와 도 2와 같다:

(단위 μg/mL)상기 실험 결과에 의하면, 본 발명의 실시예 1 내지 5는 최소 1달 이상 혈중 CBD 농도가 유지되는 것을 확인할 수 있다. 즉 실시예 1에서는 4주 시점에 0.036 μg/mL이고, 실시예 4에서도 0.782 μg/mL로 나타남을 확인하였다.
또한, 실시예 2 내지 5의 경우는, 7주에도 혈중 CBD 농도를 확인할 수 있는 점에서 지속적인 CBD 방출 효과가 나타남을 확인할 수 있다.
실험예 3
골관절염(OA)에 대한 치료 실험
순화가 완료된 rat를 군분리 후 좌측 하지 무릎관절 주변을 깨끗이 제모한 후 골관절염 유발물질인 MIA (Monosodium iodoacetate, Sigma, USA)를 0.9% 멸균 saline으로 60 mg/mL의 농도로 조제하여 BD Ultra-Fine™ II Insulin Syringe를 이용하여 50㎕씩 관절강내에 투여하였다. 우측 무릎 관절에는 동량의 0.9% 멸균 saline만을 투여하였다. MIA 투여 2주 후, 관절강 내 하기 표 5와 같은 시험물질 투여한 후, 치료 효능에 대해 평가를 진행하였다:

투여 방법
CBD solution 및 실시예 2 내지 4 군에는 MIA투여후 14일 후에 좌측 무릎 관절강내에 투여하고, 우측 무릎관절에는 동량의 0.9% 멸균 saline만을 투여하였다. Normal control 및 Positive control 군에도 동량의 0.9% saline을 투여하였다.
Dose level selection
- CBD solution: 기존 문헌상 MIA-OA model에서의 효능 농도 300 ug/joint 선정하였다.
- 실시예 2 내지 3: 미립구 볼륨 및 Rat 관절강내 투여 가능량(30 ul) 고려 시, maximum feasible dose level (1-2 mg/joint)
체중 측정 결과
시험기간 중 체중 측정을 측정한 결과는 도 3과 같다. Positive control에서 체중 증가량이 가장 낮고, CBD solution 및 실시예 2 내지 4의 투여군에서 Positive control 대비 증체량 증가하는 경향이 나타났다. 실시예 3 및 실시예 4 투여군에서 통계적으로 유의미하게 Positive control 대비 증체량 증가하였다. OA 유도 시 통증 및 보행 장애로 인한 사료섭취량 감소가 일반적으로 관찰되었으며, 이로 인해 체중이 감소하는 경향을 나타냈으나, CBD를 투여 시 통증감소로 사료섭취 활동이 개선됨에 따라 Positive control 대비 증체량 증가함을 확인하였으며, 특히 실시예 3 및 실시예 4에서 이러한 경향이 크게 나타났다.
하지 체중 부하 측정
하지 체중 부하 측정은 MIA 투여전 (0일), MIA 투여 후 6일, 13일 (시험약물 투여 전)에 측정하고, 시험약물 투여 후 2시간, 4시간, 3일, 6일, 10일, 13일, 20일, 27일차에 Incapacitance tester (Ugo Basile, Italy)를 사용하여 오른쪽, 왼쪽 발을 각각 측정하였다. Rat의 배가 기기의 센서에 닿지 않은 상태에서 양쪽의 발무게(g)를 각각 측정하였다. 측정 결과는 좌, 우측 발에서 측정된 무게의 ratio (%, Left/Right)로 분석하였다.
각 측정일에 하지 양측의 무게(g) 측정하였다. 관절염이 유발될 경우, 유발측의 (좌측) 체중 부하 감소하였다. 체중부하값은 하기와 같은 식에 의해 계산하였다:
체중부하값 = 정상 하지 무게 (우측) / 관절염 유발 하지 무게 (좌측)
상대적통증비율(%) = (각 군의 체중부하 평균값/정상군의 체중부하 평균값) X 100
시험 결과는 도 4와 같다:
Positive control은 정상군인 Normal control 대비 현저한 weight bearing 변화로 관절염이 유발하였음을 확인하였다. 투여 2시간째 CBD solution 및 실시예 2 내지 4 투여군 모두 Positive control(PC) 대조군과 대비하여 현저한 체중부하가 개선되었다.
CBD solution 투여군은, 투여 2시간째 이후 PC 대조군과 대비하여 차이가 나타나지 않았다. 실시예 4 투여군은 투여 14일째 이후 PC 대조군 대비 체중부하가 개선되는 경향이 나타났으며, 14차 및 28일차 대조군 대비 통계적으로 유의미한 개선됨이 나타났다. 실시예 3 투여군은 투여 14일 및 21일째 PC 대조군 대비 통증 감소하는 경향이 나타났으며, 14일차에서 유의성을 확인하였다.
발회피 역치 시험(Paw withdrawal threshold)
회피반응 검사는 MIA 투여전 (0일), MIA 투여 후 7일, 14일 (시험약물 투여 전)에 측정하고, 시험약물 투여 후 2시간, 4시간, 4일, 7일, 11일, 14일, 21일, 28일차에 electronic von-Frey aesthesiometer (IITC Life science, USA)를 이용하여 기계적 자극을 가한 후 발의 회피반응을 평가하였다. 발의 회피 반응을 측정하기 위해 rat을 철망위에 투명한 아크릴 통에 넣고 20분 정도 새로운 환경에 적응하도록 하였다. 측정방법은 electronic von-Frey filament를 발바닥에 직각으로 적용하여 급격한 회피 반응이 일어났을 threshold (g force) 값을 측정하였다. 측정 결과는 좌, 우측 발에서 측정된 g force의 ratio (Left/Right)로 분석하였다.
Paw withdrawal threshold ratio = Left threshold (유발측) / Right threshold (정상측), 관절염이 완화될 경우, 좌-우 비율이 동등해지는 방향으로 변화 (ratio 1)
시험 결과는 도 5 내지 12와 같다.
Positive control(PC) 대조군은 Normal control인 정상군 대비 현저한 역치가 증가하였다. 투여 2시간째 CBD solution 및 실시예 2 내지 4 투여군은 모두 PC 대조군 대비 현저한 반응 개선이 나타났다. 구체적으로 CBD solution 투여군은 투여 2시간째 PC 대조군 대비 감소하는 경향이 나타났으며, 이후 대조군 대비 차이가 없었다. 실시예 2 투여군은 투여 4일차 이후에도 지속적인 개선 경향이 나타났으며, 4일/21일/28일차 통계적 유의미하게 개선되었다. 실시예 4 투여군은 투여 4일차 이후에도 지속적인 개선 경향이 나타났으며, 4일/11일차 통계적 유의미하게 개선되었다. 마지막으로 실시예 3 투여군은 투여 4일차 이후에도 지속적인 개선 경향이 나타났으며, 11/14/21일차 통계적 유의미하게 개선되었다.
하지 체중 부하 측정 시험 및 발 회피 역치 시험의 결과에 따르면, CBD solution 투여군은 PC 대조군과 비교하여 투여 초기는 효과가 나타남을 확인할 수 있으나, 그 이후에는 PC 대조군과 차이가 없음을 확인하였다. 반면, 실시예 2 내지 4는, PC 대조군과 비교하여 장시간 지속적인 치료 효과가 나타남을 확인하여, 골관절염 모델에서 치료 및 개선 효과가 나타남은 물론, 장시간 지속적인 치료 효과가 나타남을 실험을 통해 확인하였다.
실험예 4
안정성 평가
실시예 1과 동일한 조성으로 마이크로 입자를 제조하면서, BHT를 포함하지 않은 것만 제외하고 실시예 1과 동일하게 마이크로 입자를 제조하였다.
5mL 유리 바이알에 상기 마이크로 입자를 약 100mg을 칭량하여 넣고 캡핑 또는 언캡핑 (close/open) 안정성 챔버에 입고하였다. 안정성 평가를 위해, 장기조건에서 (온도: 25±2℃, 상대습도60±5%) 진행하였으며 일일 성상변화를 관찰하여 황변여부를 확인하였다. 한편, 초기 검체 및 입고 6일차 검체는 봉입률 시험하여 함량 변화 여부를 확인하였다.
실험 결과는 도 13과 같다.
마이크로 입자를 제조한 직후에는 색상의 변화가 나타나지 않음을 확인하였으며, 6일이 경과된 후, 황변 여부를 확인한 결과에서, 유리 바이알의 캡핑 여부와 상관없이 BHT를 포함하지 않은 경우 노란색으로 색상이 변화됨을 확인하였다.
반면, 실시예 1과 같이 BHT를 1 중량%를 포함하는 경우는 색상이 변화되지 않아, 우수한 안정성을 나타냄을 확인하였다.
이상에서 본 발명의 바람직한 실시예에 대하여 상세하게 설명하였지만 본 발명의 권리범위는 이에 한정되는 것은 아니고 다음의 청구범위에서 정의하고 있는 본 발명의 기본 개념을 이용한 당업자의 여러 변형 및 개량 형태 또한 본 발명의 권리범위에 속하는 것이다.
본 발명은 염증성 질환의 치료 또는 예방을 위한 서방형 주사용 조성물 및 이의 제조 방법에 관한 것이다.
Claims (10)
- 칸나비디올(CBD, Cannabidiol)을 1개월 이상 지속적으로 방출하는 마이크로 입자를 포함하며,상기 마이크로 입자는 칸나비디올 및 생분해성 고분자를 포함하는,염증성 질환의 치료 또는 예방을 위한 서방형 주사용 조성물.
- 제1항에 있어서,상기 마이크로 입자는 칸나비디올 및 생분해성 고분자를 1:3 내지 1:10의 중량 비율로 포함하는염증성 질환의 치료 또는 예방을 위한 서방형 주사용 조성물.
- 제1항에 있어서,상기 생분해성 고분자는 폴리락트산, 폴리락타이드, 폴리락틱-코-글리콜산, 폴리락타이드-코-글리콜라이드(PLGA), 폴리포스파진, 폴리이미노카보네이트, 폴리포스포에스테르, 폴리안하이드라이드, 폴리오르쏘에스테르, 폴리카프로락톤, 폴리하이드록시발레이트, 폴리하이드록시부티레이트, 폴리아미노산 및 이들의 조합으로 이루어진 군으로부터 선택되는염증성 질환의 치료 또는 예방을 위한 서방형 주사용 조성물.
- 제3항에 있어서,상기 폴리락타이드-코-글리콜라이드는 락타이드:글리콜라이드의 단량체 비율이 40:60 내지 90:10인염증성 질환의 치료 또는 예방을 위한 서방형 주사용 조성물.
- 제1항에 있어서,상기 염증성 질환은 골관절염인염증성 질환의 치료 또는 예방을 위한 서방형 주사용 조성물.
- 제1항에 있어서,상기 마이크로 입자는 하기 방출 시험에 의해 18시간 경과 후, 전체 CBD의 중량 대비하여, 35% 내지 80% 방출되는 것인염증성 질환의 치료 또는 예방을 위한 서방형 주사용 조성물:[방출 시험 조건]용출 시험액은 10 mM PBS 완충액 (pH7.4) 1L에 Tween 20을 0.05 %(w/v)가 되도록 혼합하여 혼합 용액을 제조하고, 상기 혼합 용액에 아지드화나트륨을 0.02 %(w/v), 아스크로브산나트륨 0.0625 %(w/v)가 되도록 혼합하여 제조하였다.상기 용출 시험액 100mL를 주사약병에 넣고, 상기 마이크로 입자를 넣어 주었으며, 이때 마이크로 입자는 CBD의 총 중량이 15mg이 되도록 하였다. 상기 주사약병을 완전히 밀봉시킨 후, 50 ℃를 유지하며, 120 rpm 속도로 진탕하여 시험하였으며, 용출 개시 18 시간이 경과되면, 주사약병을 꺼내어 3 분간 방치한 후 주사기를 이용하여 시험액 1 mL를 정확하게 취하여, 3000 rpm으로 3 분간 원심분리 후 상층액을 검액으로 하여 CBD의 방출을 확인하였다.
- 칸나비디올(CBD) 및 생분해성 고분자를 유기 용매에 용해하여 유상 용액을 제조하는 단계;계면활성제를 물에 용해하여 수상 용액을 제조하는 단계; 및상기 유상 용액 및 수상 용액을 혼합하여 에멀젼을 형성하는 단계를 포함하는염증성 질환의 치료 또는 예방을 위한 서방형 주사용 조성물의 제조 방법.
- 제7항에 있어서,상기 제조된 에멀젼은 수상 용액 내로 수득하여 잔류 용매를 제거하는 단계;상기 잔류 용매가 제거된 에멀젼을 세척 및 동결 건조하여 마이크로 입자를 제조하는 단계; 및상기 동결 건조한 마이크로 입자를 주사 용수와 혼합하는 단계를 추가로 포함하는염증성 질환의 치료 또는 예방을 위한 서방형 주사용 조성물의 제조 방법.
- 제7항에 있어서,상기 유상 용액 및 수상 용액은 각 마이크로 채널에 주입하여 흐르게 하며, 상기 유상 용액의 흐름 및 수상 용액의 흐름이 서로 교차하는 지점에서 칸나비디올 및 서방화제를 포함하는 에멀젼을 형성하는염증성 질환의 치료 또는 예방을 위한 서방형 주사용 조성물의 제조 방법.
- 제7항에 있어서,상기 유상 용액은 부틸하이드록시톨루엔(BHT)를 추가로 포함하는염증성 질환의 치료 또는 예방을 위한 서방형 주사용 조성물의 제조 방법.
