Abstract

本開示は、SIRT3遺伝子発現増強用組成物に関する。
皮膚老化の原因として、加齢による生理的な老化や紫外線照射による光老化が知られている。また、加齢や紫外線照射によって、活性酸素が発生し、DNAやタンパク質、細胞が損傷し、細胞死が引き起こされ、シミ等の色素沈着や、シワの形成、たるみの発生等の皮膚老化が生じることが知られている(非特許文献1、2)。光老化を抑制する効果を示す成分として、ウロリチンAが知られている(特許文献1)。さらに、ウロリチンAは、メラニンの産生抑制効果を示し、美白効果を示すことが知られている(特許文献2)。
また、ミトコンドリア機能不全及び酸化ストレスは、アルツハイマー病、パーキンソン病、ハンチントン病、筋萎縮性側索硬化症(ALS)等の神経変性疾患と因果関係があることが知られており、ウロリチンA等のウロリチン類は、ミトコンドリアの機能を向上させる効果を有することから、神経変性疾患の治療又は予防に用いられ、神経変性疾患に伴う認知障害の治療又は予防に用いられ得ることが知られている(特許文献3)。
また、騒音性難聴(NIHL)の病因の一つとして、ミトコンドリアにおける活性酸素の蓄積により引き起こされる有毛細胞傷害が知られている。さらに、マウスにおいて、サーチュイン遺伝子の一つであるSIRT3遺伝子発現を阻害すると、活性酸素分解酵素SOD2の脱アセチル化が抑制されることでSOD2の酵素活性が低下し、ミトコンドリアの酸化ストレスを引き起こし、ひいては有毛細胞において酸化的損傷を引き起こすことが示されている。したがって、SIRT3タンパク質は、活性酸素の発生を抑制する効果を有し、NIHLによる有毛細胞傷害の改善し得るため、騒音性難聴の治療や改善に有用であることが知られている(非特許文献3)。
また、加齢や認知症に伴う認知機能の低下、神経機能の低下、神経細胞の消失の多くには、酸化ストレスが関与していることが知られており、葉酸、ビタミンB12、ビタミンE、S-アデノシルメチオニン、アセチル-L-カルニチン、N-アセチルシステイン等の抗酸化物質を含む製剤の摂取により、ヒトの脳における酸化ストレスが軽減し、軽度認知障害やアルツハイマー病の初期段階において認知能力の低下を遅らせたり、改善したりすることが知られている(非特許文献4)。
このように、SIRT3遺伝子発現の増強により酸化ストレスが抑制、軽減等されることが知られており、酸化ストレスの抑制、軽減等により、皮膚老化、美白、難聴の予防や改善、認知機能の改善等の種々の効果が得られることが知られている。しかし、ウロリチンM3がSIRT3遺伝子の発現を増強することは知られていない。

本開示の一態様は、下記の構造式で示されるウロリチンM3を有効成分とする、SIRT3遺伝子発現増強用組成物である。
ウロリチンM3を得る方法は特段限定されず、市販されているものを用いてもよく、化学合成により合成して取得してもよく、微生物に生成させて取得してもよい。
市販されているウロリチンM3としては、Toronto Research Chemicals社製のもの等が挙げられる。化学合成により合成する場合は、例えば、Asian Journal of Chemistry, Vol.29, No.8, 1803-1805 (2017)に記載の方法を用いて、メチル2-ブロモバニリン酸(2-ブロモ-5-ヒドロキシ-4-メトキシ安息香酸ともいう)とレゾルシノールにより合成することができる。

本態様に係るSIRT3遺伝子発現増強用組成物(本明細書では、「本態様に係る組成物」と記載することがある。)は、ウロリチンM3を単独で含むものであってもよいし、SIRT3遺伝子の発現を増強できる限り、他の成分を含んでもよい。本態様に係る組成物が当該他の成分を含む場合には、本態様に係る組成物は、ウロリチンM3と他の成分との混合物であってもよく、それらの成分が均一でも不均一でもよい。
本態様におけるSIRT3遺伝子は、サーチュイン3遺伝子である。本態様におけるSIRT3遺伝子は、SIRT3タンパク質を生産する。
本態様に係る組成物は、摂取又は投与するものであってもよく、外用であってもよい。

SIRT3遺伝子の発現が増強することにより、SIRT3タンパク質(本明細書では、「SIRT3」と記載することがある。)が増加する。本態様に係る組成物は、SIRT3の増加のために用いることもでき、また、本開示は、ウロリチンM3を有効成分とする、SIRT3の増加のための組成物を提供することもできる。
また、SIRT3が増加することにより、活性酸素分解酵素SOD2が脱アセチル化して活性化し、活性酸素の分解が促進することが知られている。
すなわち、本態様に係る組成物は、活性酸素分解酵素SOD2の脱アセチル化のために用いることもでき、また、本開示は、ウロリチンM3を有効成分とする、活性酸素分解酵素SOD2の脱アセチル化のための組成物を提供することもできる。

抗酸化とは、生体内又は細胞内における活性酸素(ROS)の発生を抑制したり、生体内又は細胞内において過剰に発生した活性酸素(ROS)を消去したりすることにより、酸化ストレスの増加若しくは発生を抑制、軽減又は緩和することをいう。抗酸化により、生体内又は細胞内のタンパク質、脂質、核酸等の活性酸素による傷害を有効に防止することができる。
また、本態様に係る組成物は、皮膚老化の予防若しくは改善、又は美白のために用いることもでき、また、本開示は、ウロリチンM3を有効成分とする、皮膚老化の予防若しくは改善、又は美白のための組成物を提供することもできる。
紫外線照射による皮膚老化では、光による皮膚の酸化ストレスの増加によって、シワの形成が促進され、皮膚の弾力が失われ、たるみを生じたりハリがなくなったりすることが知られている。また、加齢による生理的な老化も、酸化ストレスによって促進されることが知られている。そのため、本態様に係る組成物が抗酸化作用を有することで、これらの皮膚老化の諸症状を改善又は予防することができる。
さらに、紫外線照射によって、活性酸素が発生し、DNAやタンパク質、細胞が損傷し、細胞死が引き起こされ、シミ、ソバカス等の色素沈着が生じることが知られている。そのため、本態様に係る組成物が抗酸化作用を有することで、美白効果を得ることができる。

また、酸化ストレスの増加によって、難聴が引き起こされることや、SIRT3は有毛細胞傷害を改善することが知られている。すなわち、本態様に係る組成物は、SIRT3遺伝子の発現増強によって予防又は改善され得る疾患等の予防又は改善に用いることもでき、また、本開示は、ウロリチンM3を有効成分とする、SIRT3遺伝子の発現増強によって予防又は改善され得る疾患等の予防又は改善のための組成物を提供することもできる。「改善」は、「治療」を含む。当該疾患等としては、難聴及び難聴に関連する疾患等が例示できる。すなわち、本開示は、ウロリチンM3を有効成分とする、難聴の予防若しくは改善のための組成物を提供することもできる。
本態様において、「難聴」とは、聴覚が低下した病態であり、騒音性難聴、脳梗塞による難聴、感染による難聴、老人性難聴、薬の副作用による難聴、突発性難聴等を含む。難聴は、騒音性難聴であることが好ましい。

本態様に係る組成物の好ましい態様としては、飲食品(サプリメントを含む。)、医薬品、飼料、化粧品が例示できる。すなわち、本開示により提供することができる好ましい態様として、ウロリチンM3を有効成分とする飲食品、ウロリチンM3を有効成分とする医薬品、ウロリチンM3を有効成分とする飼料、及びウロリチンM3を有効成分とする化粧品が例示できる。
ウロリチンM3を飲食品の素材として用いる場合、一般の飲食品の他、特定保健用食品、栄養補助食品、機能性食品、病者用食品、食品添加物等(これらには飲料も含まれる。)として使用できる。飲食品の形態としては、ウロリチンM3を含有する植物自体や動物自体ではない形態でもよく、例えば、適当な助剤を添加した後、慣用の手段を用いて、食用に適した形態、例えば、顆粒状、粒状、錠剤、カプセル、ペーストなどに成形して食用に供してもよく、また種々の食品、例えば、ハム、ソーセージなどの食肉加工食品、かまぼこ、ちくわなどの水産加工食品、パン、菓子、バター、粉乳、発酵乳製品に添加して使用したり、水、果汁、牛乳、清涼飲料などの飲料に添加して使用してもよい。
前記飲食品は、水、タンパク質、糖質、脂質、ビタミン類、ミネラル類、有機酸、有機塩基、果汁、フレーバー類等を主成分とすることができる。タンパク質としては、例えば、全脂粉乳、脱脂粉乳、部分脱脂粉乳、カゼイン、大豆タンパク質、鶏卵タンパク質、肉タンパク質等の動植物性タンパク質、及びこれらの加水分解物、バターなどが挙げられる。糖質としては、糖類、加工澱粉(デキストリンのほか、可溶性澱粉、ブリティッシュスターチ、酸化澱粉、澱粉エステル、澱粉エーテル等)、食物繊維などが挙げられる。脂質としては、例えば、ラード、サフラワー油、コーン油、ナタネ油、ヤシ油、これらの分別油、水素添加油、エステル交換油等の植物性油脂などが挙げられる。ビタミン類としては、例えば、ビタミンA、カロチン類、ビタミンB群、ビタミンC、ビタミンD群、ビタミンE、ビタミンK群、ビタミンP、ビタミンQ、ナイアシン、ニコチン酸、パントテン酸、ビオチン、イノシトール、コリン、葉酸などが挙げられ、ミネラル類としては、例えば、カルシウム、カリウム、マグネシウム、ナトリウム、銅、鉄、マンガン、亜鉛、セレン、乳清ミネラルなどが挙げられる。有機酸としては、例えば、リンゴ酸、クエン酸、乳酸、酒石酸などが挙げられる。これらの成分は、2種以上を組み合わせて使用してもよく、合成品及び/又はこれらを多く含む飲食品を用いてもよい。
前記飲食品は、常法に従って製造することができる。また、ウロリチンM3への配合量、配合方法、配合時期は適宜選択することができる。さらに、必要に応じて、瓶、袋、缶、箱、パック等の適宜の容器に封入することができる。
前記飲食品全量に対するウロリチンM3の含有量は、例えば、0.0001重量%以上、0.001重量%以上、または0.01重量%以上であってもよく、20重量%以下、10重量%以下、または1重量%以下であってもよく、それらの矛盾しない組み合わせであってもよい。

ウロリチンM3を医薬品の素材として用いる場合、医薬品としての適用方法は、経口投与又は非経口投与のいずれも採用することができる。投与に際しては、有効成分を経口投与、直腸内投与、注射などの投与方法に適した固体又は液体の医薬用無毒性担体と混合して、慣用の医薬製剤の形態で投与することができる。このような製剤としては、例えば、錠剤、顆粒剤、散剤、カプセル剤などの固形剤、溶液剤、懸濁剤、乳剤などの液剤、凍結乾燥製剤などが挙げられ、これらの製剤は製剤上の常套手段により調製することができる。前記の医薬用無毒性担体としては、例えば、グルコース、乳糖、ショ糖、澱粉、マンニトール、デキストリン、脂肪酸グリセリド、ポリエチレングルコール、ヒドロキシエチルデンプン、エチレングリコール、ポリオキシエチレンソルビタン脂肪酸エステル、アミノ酸、ゼラチン、アルブミン、水、生理食塩水などが挙げられる。また、必要に応じて、安定化剤、湿潤剤、乳化剤、結合剤、等張化剤などの慣用の添加剤を適宜添加することもできる。
前記医薬品全量に対するウロリチンM3の含有量は、例えば、0.0001重量%以上、0.001重量%以上、または0.01重量%以上であってもよく、20重量%以下、10重量%以下、または1重量%以下であってもよく、それらの矛盾しない組み合わせであってもよい。

対象がヒト以外の哺乳動物である場合、ウロリチンM3を飼料の素材として用いることができる。このとき、飼料原料とウロリチンM3とを、哺乳動物の種類、発育ステージ、地域などの飼育環境に応じて適宜配合してもよい。
飼料原料としては、例えば、穀物類または加工穀物類(とうもろこし、マイロ、大麦等)、糟糠類(ふすま、米糠、コーングルテンフィード等)、植物性油粕類(大豆油粕、ごま油粕、綿実油粕等)、動物性原料(脱脂粉乳、魚粉、肉骨粉等)、ミネラル類(炭酸カルシウム、リン酸カルシウム、食塩、無水ケイ酸等)、ビタミン類、アミノ酸類、ビール酵母などの酵母類、無機物質の微粉末(結晶性セルロース、タルク、シリカ等)などが挙げられる。
飼料原料としては、例えば、穀物類または加工穀物類(とうもろこし、マイロ、大麦等)、糟糠類(ふすま、米糠、コーングルテンフィード等)、植物性油粕類(大豆油粕、ごま油粕、綿実油粕等)、動物性原料(脱脂粉乳、魚粉、肉骨粉等)、ミネラル類(炭酸カルシウム、リン酸カルシウム、食塩、無水ケイ酸等)、ビタミン類、アミノ酸類、ビール酵母などの酵母類、無機物質の微粉末(結晶性セルロース、タルク、シリカ等)などが挙げられる。
前記飼料は、前記の飼料原料に、配合飼料に通常使用される賦形剤、増量剤、結合剤、増粘剤、乳化剤、着色料、香料、食品添加物、調味料等の飼料用添加剤のほか、所望によりその他の成分(抗生物質や殺菌剤、駆虫剤、防腐剤等)を配合してもよい。
前記飼料の形態は特に限定されるものではなく、例えば、粉末状、顆粒状、ペースト状、ペレット状、カプセル剤(ハードカプセル、ソフトカプセル)、錠剤等が挙げられ、愛玩動物用のペットフードや、実験動物用の飼料として用いられてもよい。

また、前記化粧品は、通常の化粧品に用いられる成分や製剤上必要な成分を適宜加えて製造することができる。このような成分としては、例えば、水溶性高分子、植物成分、保湿剤、ロウ類、炭化水素類、精油類、油脂成分(エモリエント成分)、無機塩類、色素類、香料類、微粉体、殺菌剤、防腐剤等が挙げられる。
前記化粧品は、常法に従って製造することができる。また、前記態様に係る乳化組成物の化粧品への配合量、配合方法、配合時期は適宜選択することができる。すなわち、前記態様に係る製造方法によって乳化組成物を取得した後、該乳化組成物と化粧品の原料とを配合して化粧品とする工程を経て、前記化粧品を製造してよい。前記化粧品は、必要に応じて、瓶、袋、缶、箱、パック等の適宜の容器に封入することができる。

本態様に係る化粧品全量に対するウロリチンM3の含有量は、具体的には、例えば、0.0001重量%以上20重量%以下、0.0001重量%以上10重量%以下、0.0001重量%以上1重量%以下、0.001重量%以上20重量%以下、0.001重量%以上10重量%以下、0.001重量%以上1重量%以下、0.01重量%以上20重量%以下、0.01重量%以上10重量%以下、または0.01重量%以上1重量%以下であってもよい。
本態様に係る化粧品全量に対するウロリチンM3の含有量は、具体的には、例えば、0.0001重量%以上20重量%以下、0.0001重量%以上10重量%以下、0.0001重量%以上1重量%以下、0.001重量%以上20重量%以下、0.001重量%以上10重量%以下、0.001重量%以上1重量%以下、0.01重量%以上20重量%以下、0.01重量%以上10重量%以下、または0.01重量%以上1重量%以下であってもよい。
前記化粧品の有効適用量は、対象の齢、体重、皮膚老化の程度、美白の必要性の程度等、適用経路、適用スケジュール、製剤形態などにより適宜設定されるが、それを対象に外用として適用することにより、該対象においてSIRT3遺伝子の発現が増強する限り特に制限されず、ウロリチンM3の総量として、適用部位の1cm2あたり、かつ、1回当たり、例えば、1mg以上、2mg以上、または5mg以上であってもよく、500mg以下、100mg以下、または10mg以下であってもよく、それらの矛盾しない組み合わせであってもよい。
ウロリチンM3の総量として、具体的には、例えば、適用部位一箇所の1cm2あたり、かつ、1回当たり、1mg以上500mg以下、1mg以上100mg以下、1mg以上10mg以下、2mg以上500mg以下、2mg以上100mg以下、2mg以上10mg以下、5mg以上500mg以下、5mg以上100mg以下、または5mg以上10mg以下であってもよい。
また、前記化粧品は、1日あたり1回の適用でもよいし、1日に複数回の適用でもよい。また、数日又は数週間に1回の適用であってもよい。
ウロリチンM3の総量として、具体的には、例えば、適用部位一箇所の1cm2あたり、かつ、1回当たり、1mg以上500mg以下、1mg以上100mg以下、1mg以上10mg以下、2mg以上500mg以下、2mg以上100mg以下、2mg以上10mg以下、5mg以上500mg以下、5mg以上100mg以下、または5mg以上10mg以下であってもよい。
また、前記化粧品は、1日あたり1回の適用でもよいし、1日に複数回の適用でもよい。また、数日又は数週間に1回の適用であってもよい。
以下に実施例を記載するが、いずれの実施例も、限定的な意味として解釈される実施例ではない。
〔実施例1〕HaCaT細胞のSIRT3の増加
(SIRT3遺伝子のプロモーターを導入した細胞の調製)
ヒト表皮角化細胞HaCaT細胞由来のゲノムDNAを鋳型として、配列番号1に示されるヒトSIRT3プロモーター領域の塩基配列をPCRで増幅した。なお、使用したプライマーの配列は表1に記した。プライマー配列は、左から右へ5’から3’の向きである。配列番号2及び配列番号3はそれぞれ、報告されているhSIRT3プロモーターの塩基配列情報をもとにして、末端に制限酵素AseI及びNheIの認識配列を付加したものである。使用したDNAポリメラーゼは、KOD FX(東洋紡株式会社)であり、PCRの反応条件は、94℃2分間、98℃10秒間、及び68℃1分間を1サイクルとする40サイクルの反応条件である。
(SIRT3遺伝子のプロモーターを導入した細胞の調製)
ヒト表皮角化細胞HaCaT細胞由来のゲノムDNAを鋳型として、配列番号1に示されるヒトSIRT3プロモーター領域の塩基配列をPCRで増幅した。なお、使用したプライマーの配列は表1に記した。プライマー配列は、左から右へ5’から3’の向きである。配列番号2及び配列番号3はそれぞれ、報告されているhSIRT3プロモーターの塩基配列情報をもとにして、末端に制限酵素AseI及びNheIの認識配列を付加したものである。使用したDNAポリメラーゼは、KOD FX(東洋紡株式会社)であり、PCRの反応条件は、94℃2分間、98℃10秒間、及び68℃1分間を1サイクルとする40サイクルの反応条件である。
PCRによって得られたSIRT3プロモーター断片をpGEM-T Easy Vector(プロメガ社)へTAクローニングした。その後、シーケンシングにより、pGEM-T Easy VectorにSIRT3プロモーター断片の塩基配列が組み込まれていることを確認した。その後、pGEM-T Easy vectorに組み込まれたSIRT3プロモーター断片を制限酵素AseI及びNheI消化によって切り出し、断片を得た。得られた断片を、EGFP-C3(タカラバイオ株式会社製)ベクターのCMVプロモーターを制限酵素AseI及びNheI消化で除いた部位に挿入することで、ベクターpSIRT3p-EGFPを得た。
トランスフェクションを行うために、5mLディッシュに、HaCaT細胞(ヒト表皮角化細胞;理化学研究所バイオリソースセンターより入手、細胞番号RCB1872)を6.0×105cells/dishの細胞濃度になるように10%FBS添加DMEM培地に播種し、37℃、5%CO2存在下で培養した。翌日、1.5mLチューブ中で、pSIRT3p-EGFP(15μg)を容量が300μLになるように10%FBS含有DMEM培地で希釈した後、そこに70μLのHilymax(株式会社 同仁化学研究所)を加えて室温で20分間インキュベートした。その後、全量をHaCaT細胞に添加し、48時間培養することで、トランスフェクションを行った。
pSIRT3p-EGFPが薬剤G418耐性を有することに基づき、薬剤選択を行った。すなわち、トランスフェクションしたHaCaT細胞に、薬剤G418(和光純薬工業株式会社)を最終濃度が800μg/mLとなるように添加し、37℃、5%CO2存在下で培養した。3日毎に、新しい10%FBS含有DMEM培地に置換し、薬剤G418を最終濃度が800μg/mLとなるように添加した。これにより、HaCaT細胞にpSIRT3p-EGFPが導入されたHaCaT(SIRT3p-EGFP)細胞が得られた。
(HaCaT細胞のSIRT3の増加)
HaCaT(SIRT3p-EGFP)細胞を用いて、各被験物質について、SIRT3遺伝子のプロモーターの増強効果を調べた。

Asian Journal of Chemistry, Vol.29, No.8, 1803-1805 (2017)に記載の方法により、ウロリチンM3を合成した。
また、96ウェルプレートに、HaCaT(SIRT3p-EGFP)細胞を0.6×105cells/wellの細胞濃度になるように10%FBS添加DMEM培地に播種し、37℃、5%CO2存在下で培養した。翌日、各ウェルに、それぞれDMSOに溶解した、ケンペロール(富士フイルム和光純薬株式会社)、得られたウロリチンM3、又はエラグ酸(富士フイルム和光純薬株式会社)を、最終濃度10μMとなるようにそれぞれ添加した(n=3)。また、ネガティブコントロールとして同量のDMSOを添加した。なお、ケンペロールは、ポジティブコントロールとして用いた。各被験物質を添加してから2日間、37℃、5%CO2存在下で培養した後、培養液に、細胞固定液(4%パラホルムアルデヒド、富士フイルム和光純薬株式会社)100μLを各ウェルに添加し、室温で15分間静置した。その後、各ウェルをアスピレートし、PBSで2回洗浄後、核染色溶液(Hoechst 33342、富士フイルム和光純薬株式会社)100μLを各ウェルに添加し、20分間、室温で静置した。その後、再び各ウェルをアスピレートし、PBSで2回洗浄し、さらにPBS 100μLを添加した。その後、IN Cell Analyzer 2200(GE Healthcare, Cytiva, Tokyo, Japan)を用いて各ウェルの蛍光強度を測定した。各被験物質及びポジティブコントロールのSIRT3量は、蛍光強度の値をネガティブコントロール(DMSO)におけるEGFPの蛍光強度の値で除することにより算出し、n=3におけるSIRT3量の平均値を算出した。
また、96ウェルプレートに、HaCaT(SIRT3p-EGFP)細胞を0.6×105cells/wellの細胞濃度になるように10%FBS添加DMEM培地に播種し、37℃、5%CO2存在下で培養した。翌日、各ウェルに、それぞれDMSOに溶解した、ケンペロール(富士フイルム和光純薬株式会社)、得られたウロリチンM3、又はエラグ酸(富士フイルム和光純薬株式会社)を、最終濃度10μMとなるようにそれぞれ添加した(n=3)。また、ネガティブコントロールとして同量のDMSOを添加した。なお、ケンペロールは、ポジティブコントロールとして用いた。各被験物質を添加してから2日間、37℃、5%CO2存在下で培養した後、培養液に、細胞固定液(4%パラホルムアルデヒド、富士フイルム和光純薬株式会社)100μLを各ウェルに添加し、室温で15分間静置した。その後、各ウェルをアスピレートし、PBSで2回洗浄後、核染色溶液(Hoechst 33342、富士フイルム和光純薬株式会社)100μLを各ウェルに添加し、20分間、室温で静置した。その後、再び各ウェルをアスピレートし、PBSで2回洗浄し、さらにPBS 100μLを添加した。その後、IN Cell Analyzer 2200(GE Healthcare, Cytiva, Tokyo, Japan)を用いて各ウェルの蛍光強度を測定した。各被験物質及びポジティブコントロールのSIRT3量は、蛍光強度の値をネガティブコントロール(DMSO)におけるEGFPの蛍光強度の値で除することにより算出し、n=3におけるSIRT3量の平均値を算出した。
試験の結果を図1に示す。図1の通り、ウロリチンM3は、SIRT3プロモーターを増強し、SIRT3を増加させることが示された。

- 医薬品である、請求項1又は2に記載の組成物。
- 化粧品である、請求項1又は2に記載の組成物。
