CDI (Clostridium difficile Infection)는 항생제 장기 투여로 장내 정상 세균총이 파괴되면서 항생제 내성의 Clostridium difficile 균종이 과다 증식하여 염증을 일으키는 질병으로 심할 경우 사망에 이를 수도 있어 미국과 유럽 등 전세계적으로 문제가 되고 있다.
현재는 항생제를 유일한 치료제로 사용하고 있으나, 이에 대한 내성이 발생할 경우 치료가 어렵게 되는 문제가 있다.
또한, 항생제 치료가 어려울 경우 정상인의 분변을 이식하는 FMT(Fecal Microbiota Transplantation) 치료를 시행하고 있으나 재발하는 사례가 발생하고 있어 완벽한 치료법이 될 수 없다.
이에, 클로스트리디움 디피실(Clostridium difficile)를 효과적으로 제어할 수 있는 치료제의 개발이 시급한 실정이다.
한편, 망고스틴(Garcinia mangostana)은 말레이시아 원산의 무환자나무목 고추나무과에 속하는 쌍떡잎식물의 열매로, 망고스틴 열매는 폴리페놀의 일종인 잔톤(xanthone)이라는 대표적인 항산화제 성분을 함유하고 있어 뛰어난 항산화력을 지닌 것으로 알려져 있으며, 태국을 비롯한 동남아에서는 오래전부터 민간약으로서 염증이나 창상과 같은 상처의 치료 등에 사용된 것으로 알려져 있다.
이러한 기술적 배경하에서, 본 출원의 발명자들은 망고스틴 과피 추출물 또는 이의 주요성분인 알파-망고스틴 (α-mangostin), 베타-망고스틴 (β-mangostin), 감마-망고스틴 (γ-mangostin), 가르시논 C (garcinone C) 또는 가르시논 D (garcinone D)이 클로스트리디움 디피실(Clostridium difficile)를 효과적으로 제어할 수 있는 천연물임을 확인하고, 본 발명을 완성하였다.
상기와 같은 문제를 해결하기 위하여, 본 발명의 목적은 항균 조성물, 특히 클로스트리디움 디피실(Clostridium difficile)에 대한 항균 활성을 나타내는 조성물을 제공하는 것이다.
본 발명의 다른 목적은 상기 항균 조성물을 포함하는 의약품을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 항균 조성물을 포함하는 건강기능식품을 제공하는 것이다.
상기 목적을 달성하기 위하여, 본 발명은 망고스틴 과피 추출물; 또는 알파-망고스틴 (α-mangostin), 베타-망고스틴 (β-mangostin), 감마-망고스틴 (γ-mangostin), 가르시논 C (garcinone C) 및 가르시논 D (garcinone D)로 구성된 군에서 선택되는 1종 이상을 포함하는, 항균 조성물을 제공한다.
본 발명은 또한, 상기 항균 조성물을 포함하는 의약품을 제공한다.
본 발명은 더욱이, 상기 항균 조성물을 포함하는 건강기능식품을 제공한다.
본 발명에 따른 조성물은 망고스틴 부산물인 과피 추출물 또는 이의 주요 성분을 포함하며 우수한 항균 활성 특히, 클로스트리디움 디피실(Clostridium difficile)에 대한 우수한 항균 활성을 나타내어, 치료제로 사용할 수 있다.
도 1은 한천확산법을 통해 망고스틴 과피 추출물 및 그 주요 성분인 알파-망고스틴 (α-mangostin), 베타-망고스틴 (β-mangostin), 감마-망고스틴 (γ-mangostin), 가르시논 C (garcinone C) 또는 가르시논 D (garcinone D)의 클로스트리디움 디피실(Clostridium difficile)에 대한 항균 활성을 확인한 결과를 나타낸 것이다.
도 2는 생균수 측정을 통해 망고스틴 과피 추출물의 항균 활성을 확인한 결과를 나타낸 것이다.
도 3은 망고스틴 과피 추출물의 최소저해농도를 측정한 결과를 나타낸 것이다.
다른 식으로 정의되지 않는 한, 본 명세서에서 사용된 모든 기술적 및 과학적 용어들은 본 발명이 속하는 기술분야에서 숙련된 전문가에 의해서 통상적으로 이해되는 것과 동일한 의미를 갖는다. 일반적으로, 본 명세서에서 사용된 명명법은 본 기술분야에서 잘 알려져 있고 통상적으로 사용되는 것이다.
일 관점에서, 본 발명은 망고스틴 과피 추출물; 또는 알파-망고스틴 (α-mangostin), 베타-망고스틴 (β-mangostin), 감마-망고스틴 (γ-mangostin), 가르시논 C (garcinone C) 및 가르시논 D (garcinone D)로 구성된 군에서 선택되는 1종 이상을 포함하는, 항균 조성물에 관한 것이다.
본 명세서에서 "항균"은 박테리아의 성장 억제, 증식 억제 및 사멸 작용을 의미하며, "항박테리아"와 혼용되어 사용될 수 있다.
상기 항균 조성물은 특히, 클로스트리디움 디피실(Clostridium difficile)에 대한 항균 활성을 나타낼 수 있다. 본 발명에 따른 조성물은 클로스트리디움 디피실(Clostridium difficile)를 효과적으로 제어할 수 있다.
본 발명에 따른 조성물은 망고스틴 과피 추출물을 유효성분으로 포함하는 항균 조성물이다.
본 발명에 따른 망고스틴 과피 추출물은 말레이시아, 태국, 베트남, 인도네시아등에 분포된 열대 과일인 망고스틴(Garcinia mangostana), 특히 망고스틴 부산물인 망고스틴 과피의 추출물이다.
상기 추출은 통상의 추출 용매를 사용하여 추출할 수 있고, 제한없이 적절히 선택하여 실시할 수 있다. 상기 추출은 예를 들어, 망고스틴 과피를 물, 탄소수 1 내지 6의 알코올 또는 물과 탄소수 1 내지 6의 알코올의 혼합용매를 사용하여 수행될 수 있다. 상기 탄소수 1 내지 6의 알코올은 예를 들어 에탄올일 수 있다.
본 발명에 따른 조성물은 알파-망고스틴 (α-mangostin), 베타-망고스틴 (β-mangostin), 감마-망고스틴 (γ-mangostin), 가르시논 C (garcinone C) 및 가르시논 D (garcinone D)로 구성된 군에서 선택되는 1종 이상을 포함하는 항균 조성물이다.
상기 망고스틴 과피 추출물은 클로스트리디움 디피실(Clostridium difficile)에 대하여 우수한 항균 활성을 나타낸다. 본 발명에 따른 구체예에서, 미생물 성장 특히, 클로스트리디움 디피실(Clostridium difficile)을 억제할 수 있는 최소 농도를 확인하였다. 예를 들어, 상기 망고스틴 과피 추출물은 2.5ppm의 최소저해농도(MIC)를 가질 수 있다.
알파-망고스틴 (α-mangostin), 베타-망고스틴 (β-mangostin), 감마-망고스틴 (γ-mangostin), 가르시논 C (garcinone C) 또는 가르시논 D (garcinone D)는 클로스트리디움 디피실(Clostridium difficile)에 대하여 우수한 항균 활성을 나타낸다. 본 발명에 따른 구체예에서, 각 성분은 다음의 최소저해농도(MIC)를 나타낸다:
알파-망고스틴 (α-mangostin): 1.25ppm;
베타-망고스틴 (β-mangostin): 1,000ppm 초과;
감마-망고스틴 (γ-mangostin): 1.25ppm;
가르시논 C (garcinone C): 5ppm; 또는
가르시논 D (garcinone D): 5ppm.
본 발명에 따른 항균 조성물은 약학 조성물로 사용될 수 있다. 이를 기반으로, 본 발명은 상기 항균 조성물을 포함하는 의약품에 관한 것이다.
상기 조성물은 '약학적으로 허용가능한 담체'를 포함할 수 있다. 각각 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제제화될 수 있다.
상기 약제학적으로 허용가능한 담체는 당업계에서 통상적으로 사용되는 것들, 예컨대 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유 등을 포함하나 이에 국한되지 않는다. 또한, 본 발명의 약학 조성물은 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제, 기타 약제학적으로 허용가능한 첨가제를 포함한다.
상기 조성물이 경구용 고형 제제로 제제화된 경우 정제, 환제, 산제, 과립제, 캡슐제 등을 포함하며, 이러한 고형제제는 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘카보네이트(calcium carbonate), 수크로스(sucrose) 또는 락토즈, 젤라틴 등을 포함할 수 있으며, 마그네슘 스테아레이트, 탈크 같은 윤활제 등을 포함하나 이에 국한되지 않는다.
상기 조성물이 경구용 액상 제제화된 경우 현탁제, 내용액제, 유제, 시럽제 등을 포함하며, 물, 리퀴드 파라핀 등의 희석제, 습윤제, 감미제, 방향제, 보존제 등을 포함하나 이에 국한되지 않는다.
상기 조성물이 조성물이 비경구용 제제화된 경우 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조 제제, 좌제를 포함하며, 비수성 용제, 현탁제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르류 등을 포함하나 이에 국한되지 않는다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로젤라틴 등이 사용될 수 있으나 이에 국한되지 않는다.
상기 조성물은 목적하는 방법에 따라 경구 투여하거나 비경구투여(예를 들어, 정맥 내, 피하, 복강내 또는 국소에 적용)할 수 있으며, 투여량은 환자의 상태 및 체중, 질병의 정도, 약물형태, 투여경로 및 시간에 따라 다르지만, 당업자에 의해 적절하게 선택될 수 있다.
상기 조성물은 약학적으로 유효한 양으로 투여한다. "약학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효 용량 수준은 개체 종류 및 중증도, 연령, 성별, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료 기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있다. 단일 또는 다중 투여될 수 있다. 상기 요소를 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 당업자에 의해 용이하게 결정될 수 있다.
상기 조성물은 식품 조성물로 사용될 수 있다. 이를 기반으로, 본 발명은 상기 항균 조성물을 포함하는 건강기능식품에 관한 것이다.
"건강기능식품"은 건강기능식품에 관한 법률 제6727호에 따른 인체에 유용한 기능성을 가진 원료나 성분을 사용하여 제조 및 가공한 식품을 의미하며, "기능성"이라 함은 인체의 구조 및 기능에 대하여 영양소를 조절하거나 생리학적 작용 등과 같은 보건 용도에 유용한 효과를 얻을 목적으로 섭취하는 것을 의미한다.
상기 조성물은 통상의 식품 첨가물을 포함할 수 있으며, 상기 "식품 첨가물"로서의 적합 여부는 다른 규정이 없는 한, 식품의약품안전처에 승인된 식품 첨가물 공전의 총칙 및 일반시험법 등에 따라 해당 품목에 관한 규격 및 기준에 의하여 판정한다.
상기 "식품 첨가물 공전"에 수재된 품목으로는 예를 들어, 케톤류, 글리신, 구연산칼륨, 니코틴산, 계피산 등의 화학적 합성물, 감색소, 감초추출물, 결정셀룰로오스, 고량색소, 구아검 등의 천연첨가물, L-글루타민산나트륨 제제, 면류첨가알칼리제, 보존료제제, 타르색소제제 등의 혼합제제류를 들 수 있다.
상기 식품 조성물은 정제, 캅셀, 분말, 과립, 액상, 환 등의 형태로 제조 및 가공할 수 있다.
상기 정제 형태의 건강기능식품은 유효성분 이외에 부형제, 결합제, 붕해제, 및 다른 첨가제와의 혼합물을 통상의 방법으로 과립화한 다음, 활택제 등을 넣어 압축성형하거나, 상기 혼합물을 직접 압축성형 할 수 있다. 또한, 상기 정제 형태의 건강기능식품은 필요에 따라 교미제 등을 함유할 수 있으며, 필요에 따라 적당한 제피제로 제피할 수도 있다.
상기 캅셀 형태의 건강기능식품 중 경질캅셀제는 통상의 경질캅셀에 유효성분 및 부형제 등의 첨가제와의 혼합물 또는 그의 입상물 또는 제피한 입상물을 충진하여 제조할 수 있으며, 연질캅셀제는 유효성분 및 부형제 등의 첨가제와의 혼합물을 젤라틴 등 캅셀기제에 충진하여 제조할 수 있다. 상기 연질캅셀제는 필요에 따라 글리세린 또는 소르비톨 등의 가소제, 착색제, 보존제 등을 함유할 수 있다.
환 형태의 건강기능식품은 유효성분, 부형제, 결합제, 붕해제 등의 혼합물을 적당한 방법으로 성형하여 조제할 수 있으며, 필요에 따라 백당이나 다른 적당한 제피제로 제피를, 또는 전분, 탈크 또는 적당한 물질로 환의를 입힐 수도 있다.
과립형태의 건강기능식품은 유효성분, 부형제, 결합제, 붕해제 등의 혼합물을 적당한 방법으로 입상으로 제조할 수 있으며, 필요에 따라 착향제, 교미제 등을 함유할 수 있다. 과립형태의 건강기능식품은 12호(1680 μm), 14호(1410 μm) 및 45호(350 μm) 체를 써서 다음 입도시험을 할 때에 12호체를 전량 통과하고 14호체에 남는 것이 전체량의 50% 이하이고 또 45호체를 통과하는 것은 전체량의 150% 이하일 수 있다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로서, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지 않는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
제조예. 망고스틴 과피 추출물 조제
망고스틴 과피 분말(100g)을 70% 에탄올 수용액(350mL)에 녹여 45℃에서 3시간 동안 가열 교반하였다. 이후 상온에서 식힌 후 멸균된 헝겊으로 거른 후 진공 여과하였다. 감압 농축기로 여액의 용매를 제거한 후 동결건조하여 망고스틴 추출물(3.8g)을 얻었다.
실시예 1. 망고스틴 과피 추출물 및 그 주요 성분의 클로스트리디움 디피실
(Clostridium difficile)
에 대한 항균활성
망고스틴 과피 추출물 및 주요 성분(α-mangostin, β-mangostin, γ-mangostin, garcinone C, garcinone D; α-mangostin은 Sigma-Aldrich Co.에서 구매하고 그 외 화합물은 ChemFaces Co.에서 구매함)을 DMSO(dimethyl sulfoxide, 99.5%)에 최종농도 50,000ppm이 되도록 녹여 시료로 사용하였다.
고체배지에서의 항균 활성은 한천확산법(agar diffusion method)을 이용하여 수행하였다. 즉, Clostridium difficile 배양 배지인 RCM (reinforced clostridial medium) soft agar (0.7% agar 함유) 10mL에 Clostridium difficile KCTC 5009 균주 배양액을 최종농도 4% 접종한 후 RCM agar plate 위에 도말하였다. 30분 정도 굳힌 후 그 위에 시료(망고스틴 과피 추출물 및 주요 성분) 각각 2uL씩 점적하여 37℃에서 48시간 혐기적으로 배양하였다. 배양 후 점적 위치 주변에 생육저해환(inhibition zone)이 생성되었는지 확인하였다. 또한 망고스틴 과피 추출물 및 주요 성분이 장내 및 인체 유익균종에 대해서도 항균활성을 나타내는지 확인하기 위하여 몇몇 유산균종(Lactococcus lactis, Leuconostoc mesenteroides, Pediococcus pentosaceus, Lactobacillus plantarum, Lactobacillus casei, Lactobacillus paracasei, Weissella cibaria)을 대상으로 동일한 방법으로 항균활성 시험을 진행하였다.
액체배지에서의 항균활성을 측정하기 위하여 Clostridium difficile KCTC 5009 균주 배양액이 최종농도 1%로 접종된 RCM broth에 망고스틴 과피 추출물을 농도별로(최종농도 0.3125ppm - 5ppm) 첨가한 후 37℃에서 혐기적으로 배양하면서 경시적으로(0, 6, 12시간) 생균수를 측정하였다. 생균수 측정은 0.1% 펩톤수를 희석액으로 사용하여 십진희석법으로 수행하였다.
망고스틴 과피 추출물 및 주요 성분(α-mangostin, β-mangostin, γ-mangostin, garcinone C, garcinone D)의 Clostridium difficile KCTC 5009 균주에 대한 항균활성을 한천확산법으로 시험한 결과, β-mangostin을 제외한 모든 시료에서 Clostridium difficile KCTC 5009 균주에 대한 강한 항균 활성을 나타내었다. β-Mangostin의 경우 다른 시료에 비해 상대적으로 약한 항균활성을 나타내었다 (도 1).
한천확산법으로 동시에 진행된 장내 및 인체 유익균종에 대한 항균활성 시험 결과, 사용한 모든 균주에 대해서 항균활성이 관찰되지 않았다. 이는 망고스틴 과피 추출물 및 주요 성분이 장내 병원성 세균인 Clostridium difficile 균주에 대해서는 강한 항균활성을 보인 반면, 유익균에 대한 생육 저해 활성은 상대적으로 약하거나 나타나지 않아 항균 선택성을 가지며, 이는 치료제로서 아주 중요한 장점을 제공할 것으로 기대된다 (표 1).

또한, 액체배지에서의 Clostridium difficile KCTC 5009 균주 생균수를 이용한 항균활성 시험 결과, 망고스틴 과피 추출물의 경우 1.25ppm 이하에서는 대조군(무첨가군)과 비슷한 균의 생장이 관찰되고 2.5ppm 이상에서는 균 생장이 제어되는 것으로 확인되었다 (도 2).
실시예 2. 망고스틴 과피 추출물 및 그 주요 성분의
Clostridium difficile
에 대한 최소저해농도(MIC, minimal inhibitory concentration) 측정
96-well plate에 RCM broth를 첨가한 후 Clostridium difficile KCTC 5009 균주 배양액을 희석하여 최종농도 1.0 x 105 CFU/mL이 되도록 접종하였다. 여기에 망고스틴 과피 추출물 및 주요 성분들을 두 배씩 희석하여 첨가한 후 24시간 배양함. 배양액에 resazurin을 첨가하여 일정 시간(8-12시간) 반응 후 색의 변화(보라색[도 3의 A]은 균 생장 저해; 핑크[도 3의 B] 내지 무색은 균 생장)로 균의 생장 유무를 판단하였다.
망고스틴 과피 추출물 및 주요 성분의 Clostridium difficile KCTC 5009 균주에 대한 최소저해농도를 측정한 결과, 망고스틴 과피 추출물은 2.5ppm, α-mangostin은 1.25ppm, β-mangostin은 1,000ppm 초과, γ-mangostin은 1.25ppm, garcinone C는 5ppm, garcinone D는 5ppm으로 나타났다 (도 3, 표 2).

이상으로 본 발명의 내용의 특정한 부분을 상세히 기술하였는바, 당업계의 통상의 지식을 가진 자에게 있어서, 이러한 구체적 기술은 단지 바람직한 실시양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.
본 발명에 따르면, 망고스틴 부산물인 과피 추출물 또는 이의 주요 성분 또한 우수한 항균 활성 특히, 클로스트리디움 디피실(Clostridium difficile)에 대한 우수한 항균 활성을 나타내어, 치료제로 사용할 수 있다.

- 망고스틴 과피 추출물; 또는알파-망고스틴 (α-mangostin), 베타-망고스틴 (β-mangostin), 감마-망고스틴 (γ-mangostin), 가르시논 C (garcinone C) 및 가르시논 D (garcinone D)로 구성된 군에서 선택되는 1종 이상을 포함하는, 항균 조성물.
- 제1항에 있어서, 클로스트리디움 디피실(Clostridium difficile)에 대한 항균 활성을 나타내는 조성물.
- 제1항에 있어서, 상기 망고스틴 과피 추출물은 물, 탄소수 1 내지 6의 알코올 또는 물과 탄소수 1 내지 6의 알코올의 혼합용매 추출물인 조성물.
- 제1항에 있어서, 상기 망고스틴 과피 추출물은 클로스트리디움 디피실(Clostridium difficile)에 대하여 2.5ppm의 최소저해농도(MIC)를 가지는 조성물.
- 제1항에 있어서, 다음의 최소저해농도(MIC)를 가지는 조성물:알파-망고스틴 (α-mangostin): 1.25ppm;베타-망고스틴 (β-mangostin): 1,000ppm 초과;감마-망고스틴 (γ-mangostin): 1.25ppm;가르시논 C (garcinone C): 5ppm; 또는가르시논 D (garcinone D): 5ppm.
