Abstract
식물 추출물을 유효성분으로 하는 호흡기 질환 예방, 개선 또는 치료용 조성물에 관한 것으로, 일 양상에 따른 조성물은 초과산화물분균등화효소(SOD)의 활성을 증가시키고, 호흡기 관련 세포의 세포 치사에 대한 보호 효과를 나타내며, 염증을 완화시켜, 호흡기 질환 예방, 개발 또는 치료하는 효과가 있다.
Description
식물 추출물을 유효성분으로 하는 미세먼지에 의한 호흡기 질환 예방, 개선 또는 치료용 조성물에 관한 것이다.
미세먼지(particulate matter: PM)는 눈에 보이지 않는 작은 물질로 대기 중에 오랫동안 떠다니거나 흩날려 내려오는 입자상 물질이며, 입자의 직경이 10 ㎛ 이하인 먼지를 미세먼지(PM10), 2.5 ㎛ 이하인 먼지는 초미세먼지라고 한다.
미세먼지의 발생원은 흙, 식물의 꽃가루 등의 자연 발생원과 연료의 연소나 자동차 배출가스 등의 인위적인 발생원으로 나눌 수 있다. 미세먼지의 크기와 성분은 매우 복잡하고 다양하며, 입자의 크기, 표면적, 화학적 조성이 건강 영향을 결정하는 것으로 알려져 있다.
2013년 세계보건기구(World Health Organization: WHO) 산하 국제 암 연구소는 지름 10 ㎛ 이하의 먼지인 미세먼지가 사람에게 충분한 발암 근거가 명확하다고 보고 하며 미세먼지를 흡연과 같이 1급 발암물질로 규정하였다.
이러한 미세먼지는 눈에 보이지 않을 만큼 매우 작기 때문에 대기 중에 머물러 있다 호흡기를 거쳐 폐 등에 침투하거나 혈관을 따라 체내로 이동하여 들어감으로써 건강에 나쁜 영향을 미칠 수도 있다. 구체적으로, 미세먼지의 노출은 심장 및 폐 관련 질환 등을 발생시키거나 악화시킬 수 있으며, 결과적으로 사망 증가에 영향을 줄 수 있다. 단기적으로는 천식 발작, 급성 기관지염, 부정맥과 같은 증상을 악화시키고 장기적으로는 심혈관질환, 호흡기질환, 폐암 발생의 위험이 증가한다.
식물유래 소재는 안전성 측면에서 우수하여 오랫동안 이용되었으며, 특히 국 내의 경우 민간에서 이용되거나 혹은 한방에서 이용되는 식물 및 생약성분을 주로 한 기능성 소재 개발이 활발히 이루어지고 있다.
그러나, 상기 식물 추출물들의 미세먼지에 의해 자극된 호흡기 질환 예방 또는 개선, 치료에 대한 용도는 알려지지 않았으며, 그에 대한 기전 연구도 이루어지지 않은 실정이다. 따라서 본 발명자들은 상기 추출물들이 가지는 미세먼지에 의해 자극된 호흡기 질환 예방 또는 개선, 치료에 대한 직접적인 효능 연구를 수행하였다.
이에, 본 발명자들은 미세먼지에 의해 자극된 호흡기를 질환을 예방, 개선 또는 치료하기 위한 조성물을 개발하고자 노력한 결과, 식물 추출물이 초과산화물분균등화효소(Superoxide dismutase: SOD)의 활성을 증가시키고, 세포 치사에 대해 보호 효과를 나타내고, 미세먼지에 의해 유발된 염증을 완화하며, 결과적으로 미세먼지에 의한 호흡기 질환 예방, 개선 또는 치료할 수 있음을 확인하여, 본 발명을 완성하였다.
일 양상은 금불초(Inula japonica) 추출물, 딱지꽃(Potentilla chinensis) 추출물, 이질풀(Geranium nepalense) 추출물, 한삼덩굴(Humulus japonicas) 추출물로 이루어진 군으로부터 선택되는 하나 이상의 추출물을 유효성분으로 포함하는 호흡기 질환 예방 또는 개선용 건강기능식품 조성물을 제공하는 것이다.
다른 양상은 금불초 추출물, 딱지꽃 추출물, 이질풀 추출물 및 한삼덩굴 추출물로 이루어진 군으로부터 선택되는 하나 이상의 추출물을 유효성분으로 포함하는 호흡기 질환 개선 또는 치료용 약학적 조성물을 제공하는 것이다.
또 다른 양상은 금불초 추출물, 딱지꽃 추출물, 이질풀 추출물 및 한삼덩굴 추출물로 이루어진 군으로부터 선택되는 하나 이상의 추출물을 유효성분으로 포함하는 호흡기 질환 개선 또는 예방용 의약외품 조성물을 제공하는 것이다.

상기 금불초는 국화과의 여러해살이풀로, 식약처에서 고시한 식품원재료에 잎이 식용가능부위로 명시되어 있는 식물일 수 있다.
상기 딱지꽃은 장미과의 여러해살이풀로, 식약처에서 고시한 식품원재료에 순, 잎이 식용가능부위로 명시되어 있는 식물일 수 있다.
상기 이질풀은 쥐손이풀과의 여러해살이풀로, 식약처에서 고시한 식품원재료에 잎이 식용가능부위로 명시되어 있는 식물일 수 있다.
상기 한삼덩굴은 삼과에 속하는 덩굴성 일년생 초본식물로, 식약처에서 고시한 식품원재료에 순, 잎이 식용가능부위로 명시되어 있는 식물일 수 있다.
일 구체예에 있어서, 상기 추출물은 MUC5AC, TRPV1(Transient receptor potential vanilloid 1), CXCL-1(C-X-C Motif Chemokine Ligand 1), IL-1α, MIP2(Macrophage Inflammatory Protein 2), IL-17, Interleukin-1β(IL-1β), IL-8 및 Tumor necrosis factor-α(TNF-α) 및 사이클로옥시제나아제-2(cyclooxygenase-2: COX-2)와 같은 호흡기/기침 관련 유전자 또는 염증성 사이토카인의 발현 또는 활성을 억제하고, 염증 매개 인자인 일산화질소(nitric oxide: NO)의 생성을 억제시킴으로써, 염증을 예방, 개선 또는 치료하는 것일 수 있다. 상기 염증은 미세먼지로 인한 염증일 수 있다.
일 구체예에 있어서, 상기 추출물은 SOD의 활성을 증가시킴으로써, 염증을 예방, 개선 또는 치료하는 것일 수 있다. 상기 SOD는 초과 산화이온을 산소와 과산화수소로 바꿔 주는 불균등화 반응을 촉매함으로서, 생체에 과잉의 활성산소를 제거하여 기관지 내 항산화능을 개선하는 역할을 한다. 상기 염증은 미세먼지로 인한 염증일 수 있다.
본 명세서에서 용어 "예방(prevention)"은 질환, 장애, 또는 그의 부수적 증상의 발병 또는 재발을 부분적으로 또는 완전히 지연시키거나 방지하거나, 질환 또는 장애의 획득 또는 재획득을 막거나, 질환 또는 장애의 획득의 위험을 감소시키는 것을 총칭한다. 상기 예방은 본 발명에 따른 조성물의 투여로 염증 또는 염증 관련 질환, 장애, 또는 증상의 발생을 억제 또는 지연시키는 모든 행위를 말한다.
본 명세서에서 용어 "개선"은 치료되는 상태와 관련된 파라미터, 예를 들면 증상의 정도를 적어도 감소시키는 모든 행위를 말한다.
본 명세서에서 용어 "치료"는 질환, 장애, 또는 그의 부수적 증상이 호전되거나 이롭게 변경되는 모든 행위를 말한다.
상기 호흡기 질환은 미세먼지에 의한 호흡기 질환일 수 있다.
본 명세서에서 용어 "미세먼지"는 아황산가스, 질소 산화물, 오존, 일산화탄소, 다환 방향족 탄화수소(Polycyclic aromatic hydrocarbon: PAH) 및 다양한 유해 중금속을 포함하는 대기오염 물질로서, 눈에 보이지 않는 작은 물질로 입경이 10 ㎛ 이하의 물질인 미세먼지와 2.5 ㎛ 이하의 물질인 초미세먼지를 모두 포함하며 석탄/석유 등 화석연료 연소에 의한 매연, 자동차 배기가스, 건설 현장 등에서 발생하는 비산먼지 및 소각장 연기 등이 발생원으로 작용한다. 1차적 발생은 고체상태의 미세먼지로 나타나고 2차적 발생은 가스상태의 물질이 공기 중의 다른 물질과 화학반응 일으켜 미세먼지로 나타나는 것을 의미한다.
상기 호흡기 질환은 호흡기 염증성 폐질환, 만성 폐쇄성 폐질환(chronic obstructive pulmonary disease; COPD), 알레르기 비염, 부비강염, 상기도 감염증, 하기도 감염증, 만성기관지염, 기관지 확장증, 폐렴, 폐결핵 후유중, 급성 호흡 궁박증후군, 낭포성 섬유증, 중이염, 폐섬유중, 천식, 폐기종, 인후염, 후두염 및 편도염으로 이루어진 군으로부터 선택되는 어느 하나 이상일 수 있으며, 미세먼지에 의해 상기 호흡기 질환이 발생하는 것일 수 있다.

일 구체예에 있어서, 상기 추출물은 금불초, 딱지꽃, 이질풀 또는 한삼덩굴의 전초, 뿌리, 줄기, 가지, 잎, 종자 및 열매로 이루어진 군으로부터 선택되는 하나 이상의 것에서 추출한 것일 수 있다. 추출에 사용된 상기 금불초, 딱지꽃, 이질풀 또는 한삼덩굴의 전체, 그 일부분, 또는 이들로부터 유래된 재료는 분쇄 또는 세절되거나 적당하게 건조된 것일 수 있다.
상기 추출물을 얻기 위한 추출방법은 본 발명이 속하는 기술분야의 통상적으로 사용되는 공지된 추출방법에 따라 제조될 수 있다. 구체적으로 냉침법, 가열 추출법, 초음파 추출법, 여과법, 가압추출법, 환류추출법, 초임계 추출법, 전기적 추출법 등 통상적으로 사용되는 추출법을 사용할 수 있으며, 통상의 추출기기, 초음파분쇄 추출기 또는 분획기를 이용할 수 있다.
또한, 필요한 경우 상기 추출 후, 당업계에 공지된 여과 및/또는 농축 및/또는 동결건조 방법을 추가적으로 사용할 수 있다. 본 발명에서 사용된 추출물의 건조물 또는 농축물은 상기와 같이 공지된 방법으로 건조 또는 농축된 것을 의미한다.
일 구체예에 있어서, 상기 추출물은 물, 알코올 또는 이들의 혼합물을 용매로 하여 추출된 것일 수 있다. 상기 알코올은 C1 내지 C6 알코올, 예를 들면 C1 내지 C4 알코올일 수 있다. C1 내지 C6 알코올은 메탄올, 에탄올, 프로판올, 이소프로판올, 1,3-프로판디올, 부탄올, 펜탄올 또는 헥산올일 수 있다.
일 구체예에 있어서, 상기 추출은 각 추출물에 대하여 상기 추출 용매를 3 내지 10(부피 또는 중량)배, 예를 들면, 3 내지 7(부피/중량)배, 3 내지 5(부피/중량)배, 5 내지 10(부피/중량)배, 또는 4 내지 10(부피/중량)배 첨가하는 것을 포함할 수 있다.
일 구체예에 있어서, 상기 용매는 물과 알코올의 혼합물 즉 알코올 수용액일 수 있으며, 예들 들면, 에탄올 수용액일 수 있다. 상기 알코올 수용액에서 알코올의 농도는 1 내지 99.5 (v/v)%, 10 내지 99.5 (v/v)%, 1 내지 70(v/v)%, 1 내지 40 (v/v)%, 5 내지 25 (v/v)%, 7 내지 20 (v/v)%, 5 내지 25 (v/v)%, 또는 10 내지 20 (v/v)%일 수 있다.
본 명세서에서 용어 "건강기능식품"은 건강보조의 목적으로 특정성분을 원료로 하거나 식품 원료에 들어있는 특정성분을 추출, 농축, 정제, 혼합 등의 방법으로 제조, 가공한 식품을 말하며, 상기 성분에 의해 생체방어, 생체리듬의 조절, 질병의 방지와 회복 등 생체조절기능을 생체에 대하여 충분히 발휘할 수 있도록 설계되고 가공된 식품을 말한다. 상기 건강기능식품 조성물은 미세먼지에 의한 피부 손상의 예방 및 개선 등과 관련된 기능을 수행할 수 있다.
상기 식품의 종류에는 특별한 제한은 없다. 상기 추출물을 첨가할 수 있는 식품의 예로는 산제, 과립제, 정제, 캡슐제, 환제, 겔, 젤리, 현탁액, 에멀젼, 시럽제, 티백제, 침출차, 껌, 캔디류 및 건강 음료로 이루어진 군으로부터 선택되는 제형 등이 있으며 통상적인 의미에서의 건강식품을 모두 포함한다.
상기 건강음료 조성물은 통상의 음료와 같이 여러 가지 감미제, 향미제 또는 천연 탄수화물 등을 추가 성분으로서 포함할 수 있다. 상기 감미제는 천연감미제 또는 합성 감미제일 수 있다. 천연감미제는 타우마틴(taumatin) 또는 스테비아(stevia) 추출물일 수 있고, 합성감미제는 사카린(saccharin) 또는 아스파르탐(aspartame)일 수 있다.
상기 천연 탄수화물은 모노사카라이드(monosaccharide), 디사카라이드(disaccharide), 폴리사카라이드(polysaccharide), 자일리톨(xylitol), 소르비톨(sorbitol) 또는 에리트리톨(erythritol)일 수 있다. 상기 모노사카라이드는 포도당 또는 과당일 수 있고, 디사카라이드는 말토오스(maltose) 또는 수크로오스(sucrose)일 수 있다. 폴리사카라이드는 덱스트린(dextrin) 또는 사이클로덱스트린(cyclodextrin)일 수 있다. 상기 천연 탄수화물의 비율은 본 발명의 조성물 100 ml 당 일반적으로 약 0.01 내지 10 g, 예를 들면, 약 0.01 내지 0.1 g일 수 있다.
상기 건강기능식품에는 식품학적으로 허용 가능한 식품 보조 첨가제를 포함할 수 있으며, 건강기능식품의 제조에 통상적으로 사용되는 적절한 담체를 포함할 수 있다.
상기 외에 본 발명의 조성물은 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산 음료에 사용되는 탄산화제 등을 포함할 수 있다. 그 밖에 본 발명의 조성물은 천연 과일주스, 과일주스 음료 및 야채 음료의 제조를 위한 과육을 포함할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 이러한 첨가제의 비율은 크게 중요하진 않지만 본 발명의 조성물 100 중량부 당 0.01 내지 0.1 중량부의 범위에서 선택되는 것이 일반적이다.
다른 양상은 금불초 추출물, 딱지꽃 추출물, 이질풀 추출물 및 한삼덩굴 추출물로 이루어진 군으로부터 선택되는 하나 이상의 추출물을 유효성분으로 포함하는 호흡기 질환 개선 또는 치료용 약학적 조성물을 제공한다.

상기 약학적 조성물은 통상의 충진제, 증량제, 결합제, 붕해제, 항응집제, 윤활제, 습윤제, pH 조절제, 영양제, 비타민, 전해질, 알긴산 및 그의 염, 펙트산 및 그의 염, 보호성 콜로라이드, 글리세린, 향료, 유화제 또는 방부제 등을 포함할 수 있다.
상기 약학적 조성물은 약학적으로 허용되는 담체를 포함할 수 있으며, 이러한 담체의 예로는 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로즈, 폴리비닐피롤리돈, 물, 메틸하이드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유, 덱스트린, 칼슘카보네이트, 프로필렌글리콜, 리퀴드 파라핀 및 생리식염수로 이루어진 군으로부터 선택된 하나 이상일 수 있다.
상기 약학적 조성물의 제형은 사용방법에 따라 달라질 수 있으며, 포유동물에 투여된 후 활성 성분의 신속, 지속 또는 지연된 방출을 제공할 수 있도록 본 발명이 속하는 기술분야에 잘 알려진 방법을 사용하여 제형화될 수 있다.
경구 투여를 위한 제제에는 정제, 연질 또는 경질 캡슐제, 환제, 산제, 현탁화제, 시럽제, 주사제 및 과립제 등이 포함되고, 이러한 제제는 하나 이상의 부형제 예를 들면, 전분, 칼슘카보네이트, 수크로스 또는 락토오스, 젤라틴 등을 섞어 제조될 수 있다. 또한, 단순한 부형제 이외에 마그네슘 스테아레이트, 탈크 같은 윤활제들도 사용될 수 있다. 비경구투여를 위한 제제는 크림, 로션제, 연고제, 경고제, 액제, 에어로솔제, 유동엑스제, 엘릭서, 침제, 향낭, 패취제 또는 주사제 등일 수 있다.
상기 방법은 임의의 동물에 적용 가능하며, 동물은 인간 및 영장류뿐 아니라, 소, 돼지, 양, 말, 개 및 고양이 등의 가축을 포함할 수 있다.
상기 치료, 예방, 또는 개선을 위한 조성물의 투여량은 투여방법, 복용자의 연령, 성별, 환자의 중증도, 상태, 체내에서 활성 성분의 흡수도, 불활성률 및 병용되는 약물을 고려하여 결정할 수 있으며, 1일 유효성분을 기준으로 하였을 때 0.1 mg/kg(체중) 내지 500 mg/kg(체중), 0.1 mg/kg(체중) 내지 400 mg/kg(체중) 또는 1 mg/kg(체중) 내지 300 mg/kg(체중)으로 투여할 수 있으며, 1회 또는 수회로 나누어 투여할 수 있으나, 이에 한정되는 것은 아니다.
또 다른 양상은 금불초 추출물, 딱지꽃 추출물, 이질풀 추출물 및 한삼덩굴 추출물로 이루어진 군으로부터 선택되는 하나 이상의 추출물을 유효성분으로 포함하는 호흡기 질환 개선 또는 예방용 의약외품 조성물을 제공한다.
상기 추출물 및 호흡기 질환은 상기한 바와 같다.
용어 "의약외품"은 사람이나 동물의 질병을 치료, 경감, 처치 또는 예방할 목적으로 사용되는 섬유, 고무제품 또는 이와 유사한 것, 인체에 대한 작용이 약하거나 인체에 직접 작용하지 아니며, 기구 또는 기계가 아닌 것과 이와 유사한 것, 감염 예방을 위하여 살균, 살충 및 이와 유사한 용도로 사용되는 제제 중 하나에 해당하는 물품으로서, 사람이나 동물의 상태 또는 질병을 진단, 치료, 경감, 처치 또는 예방할 목적으로 사용하는 물품 중 기구, 기계 또는 장치가 아닌 것 및 사람이나 동물의 구조와 기능에 약리학적 영향을 줄 목적으로 사용하는 물품 중 기구, 기계 또는 장치가 아닌 것을 제외한 물품을 의미하며, 피부 외용제 및 개인위생용품도 포함할 수 있다.
상기 추출물을 의약외품 조성물에 첨가할 경우, 상기 추출물을 그대로 첨가하거나 다른 의약외품 성분과 함께 사용할 수 있고, 통상적인 방법에 따라 적절하게 사용할 수 있다. 유효 성분의 혼합양은 사용 목적(예방, 건강 또는 치료적 처치)에 따라 적합하게 결정될 수 있다.
상기 의약외품 조성물은 특별히 이에 제한되지 않으나, 개인위생용품, 피부외용제, 가습기 충진제, 마스크, 연고제 또는 필터충진제 등일 수 있다. 상기 개인위생용품은 비누, 물티슈, 휴지, 샴푸, 치약, 모발 손질 제품, 에어프레쉬너 겔 또는 세정 겔일 수 있다.
일 양상에 따른 조성물은 초과산화물분균등화효소(SOD)의 활성을 증가시키고, 호흡기 관련 세포의 세포 치사에 대한 보호 효과를 나타내며, 염증을 완화시켜 호흡기 질환 예방, 개선 또는 치료하는 효과가 있다.

이하 실시예를 통하여 보다 상세하게 설명한다. 그러나, 이들 실시예는 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예 1. 금불초, 딱지꽃, 이질풀 및 한삼덩굴 추출물의 제조
금불초, 딱지꽃, 이질풀 및 한삼덩굴 추출물을 제조하기 위하여, 국내에서 자생하는 식물인 금불초, 딱지꽃, 이질풀 및 한삼덩굴을 구입하여 증류수로 세척한 다음, 햇빛이 없는 서늘한 곳에서 중량의 변화가 없을 때까지 완전히 건조시켰다.
그리고 완전히 건조된 상기 건조물을 균질화기를 사용하여 분쇄한 다음, 상기 분쇄물 30 g을 에탄올 및 증류수를 3:7의 중량비로 혼합한 추출용매 900 g에 첨가하였다. 이때, 분쇄물 및 추출용매의 혼합비는 중량비로 30 배수로 설정하였다. 다음으로, 상기 분쇄물과 혼합한 추출 용매를 진탕 항온 수조에 넣어서 60℃로 5시간 동안 교반추출하였다. 그리고 추출에 의해 얻어진 물질을 상온에서 와트만(No. 2) 추출 용매 여과지를 사용하여 불용성 물질을 제거하였다.
다음으로, 상기 추출물을 냉각 콘덴서가 달린 증류장치에서 감압농축 하여 추출 용매를 완전히 제거하여 금불초, 딱지꽃, 이질풀 및 한삼덩굴 추출물을 각각 수득하였다.
실험예 1. 금불초, 딱지꽃, 이질풀 및 한삼덩굴 추출물의 초과산화물 불균등화효소(Superoxide dismutase: SOD)
활성 확인
상기 실시예 1.의 금불초 추출물, 딱지꽃 추출물, 이질풀 추출물 및 한삼덩굴 추출물의 초과산화물불균등화효소 활성을 확인하기 위하여, SOD Determination kit(Sigma, MO, USA)를 이용하였다.
구체적으로, 상기 실시예1.의 각 추출물을 각각 희석한 후 20 ㎕씩 96 웰 플레이트에 분주하고, 200 ㎕의 WST(Water-soluble tetrazolium salt) 용액과 20 ㎕의 효소반응 액을 첨가하였다. 다음으로, 웰 플레이트를 37 ℃에서 20 분동안 처리한 후, microplate reader(BioTek, VT, USA)를 이용하여 450 nm에서 흡광도를 측정하였다. 그리고 측정한 흡광도를 이용하여, SOD 효소 활성률을 하기 식 1에 따라 계산하였다:

그리고, 각 추출물의 농도별 SOD 효소 활성률을 표 1에 나타내었다.

표 1에 나타낸 바와 같이, 금불초, 딱지꽃, 이질풀 및 한섬덩굴의 추출물 모두 SOD 효과적인 효소 활성을 보임을 확인하였다. 특히, 그 중에서도 딱지꽃 추출물 및 이질풀 추출물이 적은 농도로도 높은 SOD 효소 활성률을 보임을 확인하였다.
이상의 결과는 SOD가 초과 산화이온을 산소와 과산화수소로 바꿔 주는 불균등화 반응을 촉매함으로서, 생체에 과잉의 활성산소를 제거하여 기관지 내 항산화능을 개선할 수 있으므로, 일 구체예에 따른 추출물의 경우, 높은 SOD 효소 활성도로 인해 미세먼지에 의한 호흡기 질환 예방, 개선 또는 치료 효과가 있음을 의미한다.
실험예 2. 금불초, 딱지꽃, 이질풀 및 한삼덩굴 추출물의 미세먼지로 유래된 일산화질소(Nitric Oxide: NO) 생성 억제 확인
상기 실시예 1.의 금불초 추출물, 딱지꽃 추출물, 이질풀 추출물 및 한삼덩굴 추출물의 NO 감소 효과를 확인하기 위하여, 미세먼지로 염증 반응을 일으킨 후 NO의 변화를 확인하였다.
구체적으로, 대식세포인 RAW264.7 세포를 American Type Culture Collection(ATCC, MD, USA)로부터 수득하고, 상기 RAW264.7 세포를 10% FBS 및 1% antibiotic-antimycotic이 포함된 DMEM(Dulbecco's Modified Eagle's Medium) 배지에서 5% CO2 및 37℃ 조건으로 배양하였다. 그리고 배양한 세포를 6 웰 플레이트에 8.0 × 105 cells/1.5 ㎖/well의 농도로 24 시간 동안 부착시킨 후, DMEM에 미세먼지(PM10) 200 ㎍/ml를 처리하여 염증 반응을 일으켰다. 그리고 실시예 1.의 각 추출물을 0, 25, 50 또는 100 ppm의 농도로 처리하여 24 시간 동안 배양하였다. 다음으로, 배양 상등액을 100 ㎕씩 96 웰 플레이트에 분주한 후, Griess(0.1% N-(1-naphtyl) ethylenediamine, 1% sulfanilamide) 시약 100 ㎕를 혼합하고 10 분 동안 반응시킨 뒤, 발색 azo-유도체 분자의 흡광도를 540 nm에서 마이크로플레이트 리더(microplate reader, BioTek, Vermont, USA)로 측정하였다. 측정한 흡광도를 이용하여, 아질산 나트륨의 희석 범위를 각 샘플의 아질산염의 양으로 표준 곡선 변환하여 NO 생성량을 계산하였다. 그 결과를 도 1에 나타내었다.

도 1에 나타낸 바와 같이, 금불초, 딱지꽃, 이질풀 및 한섬덩굴의 추출물은 모두 농도에 비례하여 유의적으로 NO 생성을 억제함을 확인하였다. 특히, 각 추출물을 100 ppm의 농도로 처리한 경우 NO 생성량이 미세먼지를 미처리한 대조군의 수준으로 감소함을 확인하였다.
이상의 결과는 일 구체예에 따른 추출물의 경우, NO 생성을 효과적으로 저해하여, 미세먼지에 의한 호흡기 질환 예방, 개선 또는 치료 효과가 있음을 의미한다.
실험예 3. 금불초, 딱지꽃, 이질풀 및 한삼덩굴 추출물의 미세먼지로 유래된 IL-1β 발현 억제 확인
상기 실시예 1.의 금불초 추출물, 딱지꽃 추출물, 이질풀 추출물 및 한삼덩굴 추출물의 미세먼지로 유래된 IL-1β의 발현 억제 효능을 확인하기 위하여, IL-1β의 발현량을 ELISA kit(Invitrogen, CA, USA)를 이용하여 측정하였다.
구체적으로, 대식세포인 RAW264.7 세포를 American Type Culture Collection (ATCC, MD, USA)로부터 수득하고, 상기 RAW264.7 세포를 10% FBS 및 1% antibiotic-antimycotic이 포함된 DMEM 배지에서 5% CO2 및 37℃ 조건으로 배양하였다. 그리고 RAW264.7 세포를 6 웰 플레이트에 8.0 Х 105 cells/1.5 ㎖/well의 농도로 24 시간 동안 부착시킨 후 DMEM에 미세먼지(PM10) 200 ㎍/ml를 처리하여 염증 반응을 일으켰다. 그리고 실시예 1.의 각 추출물을 0, 25, 50 또는 100 ppm의 농도로 처리하여 24 시간 동안 배양하였다. 24시간 후 배양 배지를 원심 분리하여 얻어진 상층액의 IL-1β를 ELISA kit를 이용하여 정량하고, 그 결과를 도 2에 나타내었다.

도 2에 나타낸 바와 같이, 금불초 추출물, 딱지꽃 추출물, 이질풀 추출물 및 한삼덩굴 추출물은 모두 농도에 비례하여 유의적으로 IL-1β의 발현을 저해함을 확인하였다. 특히, 각 추출물을 100 ppm의 농도로 처리한 경우, 미처리 대조군 대비 IL-1β의 발현을 60 내지 80 %의 수준으로 감소시키는 것을 확인하였다.
이상의 결과는 일 구체예에 따른 추출물의 경우, IL-1β의 발현을 효과적으로 저해하여, 미세먼지에 의한 호흡기 질환 예방, 개선 또는 치료 효과가 있음을 의미한다.
실험예 4. 금불초, 딱지꽃, 이질풀 및 한삼덩굴 추출물의 미세먼지로 유래된 TNF-α발현 억제 확인
상기 실시예 1.의 금불초 추출물, 딱지꽃 추출물, 이질풀 추출물 및 한삼덩굴 추출물의 미세먼지로 유래된 TNF-α(tumor necrosis factor-α)의 발현 억제 효능을 확인하기 위하여, TNF-α의 발현량을 ELISA kit(Invitrogen, CA, USA)를 이용하여 측정하였다.
구체적으로, 대식세포인 RAW264.7 세포를 American Type Culture Collection (ATCC, MD, USA)로부터 수득하고, 상기 RAW264.7 세포를 10% FBS 및 1% antibiotic-antimycotic이 포함된 DMEM 배지에서 5% CO2 및 37℃ 조건으로 배양하였다. 그리고 RAW264.7 세포를 6 웰 플레이트에 8.0 Х 105 cells/1.5 ㎖/well의 농도로 24 시간 동안 부착시킨 후 DMEM에 미세먼지(PM10) 200 ㎍/ml를 처리하여 염증 반응을 일으켰다. 그리고 실시예 1.의 각 추출물을 0, 25, 50 또는 100 ppm의 농도로 처리하여 24 시간 동안 배양하였다. 24시간 후 배양 배지를 원심 분리하여 얻어진 상층액의 TNF-α를 ELISA kit를 이용하여 정량하고, 그 결과를 도 3에 나타내었다.

실험예 5. 금불초, 딱지꽃, 이질풀 및 한삼덩굴 추출물의 미세먼지로 유래된 COX-2 mRNA 발현 억제 확인
상기 실시예 1.의 금불초 추출물, 딱지꽃 추출물, 이질풀 추출물 및 한삼덩굴 추출물의 미세먼지로 유래된 COX-2 mRNA 발현 억제 효능을 확인하기 위하여, COX-2(cyclooxygenase-2) mRNA의 발현량을 실시간-PCR(real-time polymerase chain reaction)을 이용하여 확인하였다.
구체적으로, 대식세포인 RAW264.7 세포를 American Type Culture Collection(ATCC, MD, USA)로부터 수득하고, 10% FBS 및 1% antibiotic-antimycotic이 포함된 DMEM 배지에서 5% CO2 및 37℃ 조건으로 배양하였다. RAW264.7 세포를 6 웰 플레이트에 8.0 Х 105 cells/2.0 ㎖/well의 농도로 24 시간 동안 부착시킨 후 DMEM에 미세먼지(PM10) 200 ㎍/ml를 처리하여 염증 반응을 일으켰다. 그리고 실시예 1.의 각 추출물을 0, 25, 50 또는 100 ppm의 농도로 처리하여 24 시간 동안 배양하였다. 24시간 후 세포 내에서 RNA를 추출하여 cDNA로 합성하고, 타겟 주형(primer)을 사용하여 정량적 실시간-PCR을 실시하여 최종적으로 COX-2 유전자 발현 정도를 평가하고, 그 결과를 도 4에 나타내었다.

실험예 6. 폐암 세포에서의 미세먼지 처리에 의한 세포 독성 평가
폐암세포에서 미세먼지의 세포독성을 MTT(3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay를 통해 확인하였다.
구체적으로, 폐암세포인 A549 를 American Type Culture Collection(ATCC, MD, USA)로부터 수득하고, 10% FBS 및 1% antibiotic-antimycotic이 포함된 DMEM 배지에서 5% CO2 및 37℃ 조건으로 배양하였다. 그리고 A549 세포를 96 웰 플레이트에 1.2 Х 104 cells/200㎕/well의 농도로 24 시간 동안 안정화시켰다. 이후 미세먼지(PM10)를 다양한 농도(각, 3.12, 6.25, 12.5, 25, 50 또는 100 ㎍/ml)로 처리하여 24 시간 동안 배양하였다. 24시간 배양 후 MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay를 통해 세포 생존 정도를 측정하였다. 더 자세하게는, 최종적으로 0.5 mg/ml 농도가 될 수 있도록 MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide)용액을 각각의 세포에 처리하고 4시간 동안 배양한 후, 용액을 완전히 제거하고 DMSO(Dimethyl sulfoxide)로 용해시킨 플레이트를 590 nm의 흡광도에서 측정하였다.
도 5는 미세먼지 처리에 따른 폐암 세포에서 세포 생존율을 나타낸 그래프이다.
도 5에 나타낸 바와 같이, 미세먼지의 농도에 비례하여 유의하게 세포 생존율이 감소됨을 확인하였다. 특히, 미세먼지를 100 ㎍/ml의 농도로 처리한 경우, 미처리 대조군 대비 약 40 %가 감소한 약 60 %의 세포 생존율을 보임을 확인하였다.
실험예 7. 미세먼지에 의해 손상된 폐암 세포에서 금불초, 딱지꽃 및 한삼덩굴 추출물 처리에 따른 세포 보호 효과 확인
상기 실시예 1.의 금불초 추출물, 딱지꽃 추출물 및 한삼덩굴 추출물의 호흡기 관련 세포에 대하여 미세먼지로 인한 세포 치사로부터의 보호 효과를 확인하였다.

도 6에 나타낸 바와 같이, 미세먼지의 처리에도 불고하고 각 추출물 모두 추출물의 농도에 비례하여 유의하게 호흡기 관련 세포 생존율이 증가함을 확인하였다. 구체적으로, 미세먼지 100 ㎍/ml를 처리한 결과 미세먼지 미처리 대조군 대비 약 20 %가 감소한 약 80 %의 세포 생존율을 보였으나, 상기 미세먼지가 처리된 폐암 세포에 각 추출물을 100 ppm의 농도로 처리한 경우 미세먼지 미처리 대조군 수준으로 세포 생존율이 회복됨을 확인하였다. 특히, 딱지꽃 추출물의 경우 25 ppm의 농도로도 미처리 대조군 수준으로 세포 생존율이 회복됨을 확인하였다.
이상의 결과는 일 구체예에 따른 추출물의 경우, 미세먼지에 의한 세포 독성을 효과적으로 억제하여 호흡기 관련 세포의 생존율을 현저하게 개선시킴으로써, 호흡기 질환 예방, 개선 또는 치료 효과가 있음을 의미한다.
실험예 8. 폐암 세포에서 금불초, 딱지꽃, 이질풀 및 한삼덩굴 추출물의 미세먼지로 유래된 IL-8 mRNA 발현 억제 효능 확인
호흡기 관련 세포에서 상기 실시예 1.의 금불초 추출물, 딱지꽃 추출물, 이질풀 추출물 및 한삼덩굴 추출물의 미세먼지로 인한 IL-8 mRNA발현 억제 효능을 확인하기 위하여, IL-8 mRNA의 발현량을 실시간-PCR을 이용하여 확인하였다.

도 7에 나타낸 바와 같이, 폐암 세포에서 미세먼지의 처리에 따라 증가한 염증성 사이토카인 IL-8 mRNA의 발현이 금불초 추출물, 딱지꽃 추출물, 이질풀 추출물 및 한삼덩굴 추출물의 처리에 따라 유의적으로 감소함을 확인하였다. 구체적으로, 각 추출물을 100 ppm의 농도로 처리한 경우, 추출물 미처리 대조군 대비 약 50 내지 80 % 이상의 IL-8 mRNA 발현 억제 효과를 보임을 확인하였다.
이상의 결과는 일 구체예에 따른 추출물의 경우, 호흡기 관련 세포에서 미세먼지에 의해 유도된 IL-8 발현을 효과적으로 저해함으로써, 미세먼지에 의한 호흡기 질환 예방, 개선 또는 치료 효과가 있음을 의미한다.
실험예 9. 금불초, 딱지꽃 추출물의 생체 내 (in vivo) 효과 분석
9.1. 호흡기 손상 동물 모델 제작 및 실험 디자인
금불초 추출물, 딱지꽃 추출물의 생체 내 미세먼지 호흡기에 대한 효능을 분석하기 위해 먼저 호흡기 손상 동물 모델을 제작하였다.
7주령된 수컷 Balb/c 마우스를 다음과 같이 7개 군으로 설정하여 각 군당 8마리씩 실험하였고, 외관 상태, 행동, 모발 상태, 호흡 상태 등 마우스의 전반적인 상태를 매일 확인하였다: 대조군으로, 미세먼지혼합물로 호흡기 손상을 유도하지 않은 일반 마우스 군(rBalb/c Normal)과 미세먼지혼합물로 호흡기 손상을 유도하고 아무것도 투여하지 않은 마우스 군(rPM10_CTL)을 각각 두고, 호흡기 손상 유도된 마우스에 Dexamethasone 3mg/kg를 적용한 마우스 군(rPM10_Dexa 3mg/kg)을 양성대조군으로는 하였다. 실험군으로는 호흡기 손상 마우스에 딱지꽃 추출물을 각각 200 mg/kg, 100mg/kg 경구투여한 군(rPM10_딱지꽃 200mg/kg, rPM10_딱지꽃 100mg/kg), 금불초 추출물을 각각 200 mg/kg, 100mg/kg 경구투여한 군(rPM10_금불초 200mg/kg, rPM10_금불초 100mg/kg)을 두었다.
도 8에 나타낸 바와 같이, 호흡기 손상 미세먼지(PM10)와 Disel exhaust particles (DEP)를 일정 농도로 혼합하여 4mg/ml 미세먼지혼합물 (PM10D)을 Aluminium hydroxide에 희석시켜 Intra-Nazal-Tracheal (INT) injection 방법을 이용하여 3일 간격으로 3회 (약물투여 3일 후, 6일 후, 9일 후 intra-nasaltracheal injection) 기도를 통해 폐로 직접 주입하여 호흡기 손상 동물 모델을 제작하고, 최종 미세먼지 혼합물 주입 3일 후 희생시켰다.
9.2. 금불초 추출물, 딱지꽃 추출물의 PM10D 유도 호흡기 손상 모델에서 IL-1α, CXCL-1, MIP2, IL-17, TNF-α 발현 억제 효능 확인
PM10D 유도 호흡기 손상 동물 모델의 기관지 페포 세척액(BAL fluid; BALF)에서 상기 실시예 1.의 금불초 추출물, 딱지꽃 추출물의 IL-1α, CXCL-1(C-X-C Motif Chemokine Ligand 1), MIP2(Macrophage Inflammatory Protein 2), IL-17, TNF-α 발현 억제 효능을 확인하기 위하여 ELISA kit(R&D Systems, Minneapolis, MN, USA)를 이용하여 측정하였다.
기관지 세척을 통해 마우스의 기관지로부터 기관지 폐포 세척액을 분리하였으며, 이를 이용해 IL-1α, CXCL-1, MIP2, IL-17, TNF-α를 ELISA kit를 이용하여 분석하여 정량하고, 그 결과를 도 9에 나타내었다.

도 9에 나타낸 바와 같이, PM10D 유도 호흡기 손상 동물 모델에서 호흡기 손상군에 비해 염증성 사이토카인 IL-1α, CXCL-1, MIP2, IL-17, TNF-α의 발현이 금불초 추출물, 딱지꽃 추출물의 처리에 따라 유의적으로 감소함을 확인하였다.
이상의 결과는 금불초 추출물, 딱지꽃 추출물의 경우, PM10D 유도 호흡기 손상 모델에서 IL-1α, CXCL-1, MIP2, IL-17, TNF-α발현을 효과적으로 저해함으로써, 미세먼지에 의한 호흡기 질환 예방, 개선 또는 치료 효과가 있음을 의미한다.
9.3. 금불초 추출물, 딱지꽃 추출물의 PM10D 유도 호흡기 손상 모델에서 MUC5AC, TRPV1, CXCL-1, TNF-α, COX-2 mRNA 발현 억제 효능 확인
PM10D 유도 호흡기 손상 동물 모델에서 상기 실시예 1.의 금불초 추출물, 딱지꽃 추출물의 호흡기/기침 관련 유전자인 MUC5AC mRNA, TRPV1(Transient receptor potential vanilloid 1) mRNA, CXCL-1 mRNA, TNF-α mRNA, COX-2 mRNA, NOS-II(Nitric oxide synthase II) mRNA 발현 억제 효능을 확인하기 위하여, MUC5AC mRNA, TRPV1 mRNA, CXCL-1 mRNA, TNF-a mRNA, COX-2 mRNA, NOS-II mRNA 의 발현량을 실시간-PCR을 이용하여 확인하였다.
구체적으로, 폐 조직을 TRIzol reagent 용액(Thermo Fisher Scientific Inc., Waltham, MA, USA)을 이용하여 RNA 분리 후 추출하였다. 추출한 RNA를 RTase, Dntp, BUFFER, Oligo-Dt를 넣은 후 42 ℃ water bath에서 2 시간 동안 반응시켜 cDNA를 합성하였다. 합성된 cDNA에 2 Х SYBR green PCR Master Mix (Applied Biosystems, Grand Island, NY, USA), 200 nM primers, rox dye를 넣은 후 각 cycle은 95℃ 15초, 55℃ 15초, 72℃ 15초의 조건으로 Applied Biosystems 7500 Real-Time PCR system (Applied Biosystems, Grand Island, NY, USA)를 이용하여 PCR을 수행하였다. 각 유전자의 발현은 housekeeping 유전자 glyceraldehyde-3-phosphatedehydrogenase (GAPDH)를 internal control로 사용하여 상대적 정량 값을 비교 분석하였다.

도 10에 나타낸 바와 같이, PM10D 유도 호흡기 손상 동물 모델에서 호흡기 손상군에 비해 호흡기/기침 관련 유전자 MUC5AC mRNA, TRPV1 mRNA, CXCL-1 mRNA, TNF-a mRNA, COX-2 mRNA, NOS-II mRNA 의 발현이 금불초 추출물, 딱지꽃 추출물의 처리에 따라 유의적으로 감소함을 확인하였다.
이상의 결과는 금불초 추출물, 딱지꽃 추출물의 경우, PM10D 유도 호흡기 손상 모델에서 MUC5AC, TRPV1, CXCL-1, TNF-a, COX-2, NOS-II mRNA발현을 효과적으로 저해함으로써, 미세먼지에 의한 호흡기 질환 예방, 개선 또는 치료 효과가 있음을 의미한다.
9.4. 금불초 추출물, 딱지꽃 추출물의 PM10D 유도 호흡기 손상 모델에서 폐 조직 손상에 대한 효능 확인
PM10D 유도 호흡기 손상 동물 모델에서 금불초 추출물, 딱지꽃 추출물의 폐조직 손상에 대한 효능을 확인하기 위하여 광학현미경(Light microscope, Nikon, Japan)을 이용하여 관찰하였다.
구체적으로, 폐손상의 정도를 측정하기 위해 폐조직을 10% Neutral buffered formalin에 24시간 동안 고정시킨 뒤, graded alcohol로 탈수시키고 파라핀에 포매하여 block을 제작한 뒤, Microtome을 이용하여 4μm두께로 조직 절편을 얻어 hematoxylin & eosin (H&E)염색과 Masson's trichrome(M-T)염색을 실시한 다음 광학현미경 배율에서 관찰하였다.
도 12는 PM10D 유도 호흡기 손상 동물 모델에서 금불초 추출물, 딱지꽃 추출물의 처리에 따른 폐조직 손상에 대한 효과를 나타낸 사진이다.
도 12에 나타낸 바와 같이, PM10D 유도 호흡기 손상 동물 모델에서 호흡기 손상군에 비해 미세먼지 복합물의 침착, 기도의 두께, 염증면역세포의 침윤과 과립구세포, 활성화된 대식세포, 콜라겐 침착 및 침윤정도가 금불초 추출물, 딱지꽃 추출물의 처리에 따라 유의적으로 감소함을 확인하였다.
이상의 결과는 금불초 추출물, 딱지꽃 추출물의 경우, PM10D 유도 호흡기 손상 모델에서 미세먼지 복합물의 침착, 기도의 두께, 염증면역세포의 침윤과 과립구세포, 활성화된 대식세포, 콜라겐 침착 침윤정도를 효과적으로 저해함으로써, 미세먼지에 의한 호흡기 질환 예방, 개선 또는 치료 효과가 있음을 의미한다.
제조예 1. 정제의 제조
상기 실시예 1.의 추출물을 하기 표 2의 성분비로 혼합하고, 통상의 정제 제조방법에 따라서 타정하여 정제를 제조하였다. 제조된 정제는 모든 제형예 시험 조건에서 안정함을 확인하였다.

상기 제조예 1 내지 4의 조성비는 일반적으로 적합한 성분을 혼합하여 제형예로 조성하였지만, 필요에 따라서 그 배합비 및 원료를 임의로 변경 실시하여도 무방하다.
본 발명의 추출물은 모든 제형예 시험 조건에서 안정하므로 제형의 안정성에는 문제가 없었다.
Claims (14)
- 금불초(Inula japonica) 추출물, 딱지꽃(Potentilla chinensis) 추출물, 이질풀(Geranium nepalense) 추출물 및 한삼덩굴(Humulus japonicas) 추출물로 이루어진 군으로부터 선택되는 어느 하나 이상의 추출물을 유효성분으로 포함하는 호흡기 질환 개선 또는 예방용 건강기능식품 조성물.
- 청구항 1에 있어서,상기 추출물은 MUC5AC, TRPV1, CXCL-1, IL-1α, MIP2, IL-17, IL-1β, IL-8, TNF-α및 COX-2로 이루어진 군으로부터 선택되는 어느 하나 이상의 발현 또는 활성을 억제하는 것인, 건강기능식품 조성물.
- 청구항 1에 있어서,상기 추출물은 일산화질소(nitric oxide: NO)의 생성을 억제하는 것인, 건강기능식품 조성물.
- 청구항 1에 있어서,상기 추출물은 초과산화물 불균등화효소(superoxide dismutase: SOD)의 활성을 증가시키는 것인, 건강기능식품 조성물.
- 청구항 1에 있어서,상기 호흡기 질환은 호흡기 염증성 폐질환, 만성 폐쇄성 폐질환(chronic obstructive pulmonary disease; COPD), 알레르기 비염, 부비강염, 상기도 감염증, 하기도 감염증, 만성기관지염, 기관지 확장증, 폐렴, 폐결핵 후유중, 급성 호흡 궁박증후군, 낭포성 섬유증, 중이염, 폐섬유중, 천식, 폐기종, 인후염, 후두염 및 편도염으로 이루어진 군으로부터 선택되는 어느 하나 이상인, 건강기능식품 조성물.
- 청구항 1에 있어서,상기 호흡기 질환은 미세먼지에 의한 호흡기 질환인, 건강기능식품 조성물.
- 청구항 1에 있어서,상기 추출물을 조성물 총 중량 대비 0.0001 중량% 내지 99.0 중량%로 포함하는, 건강기능식품 조성물.
- 청구항 1에 있어서,상기 추출물은 금불초, 딱지꽃, 이질풀 또는 한삼덩굴의 전초, 뿌리, 줄기, 가지, 잎, 종자 및 열매로 이루어진 군으로부터 선택되는 하나 이상의 것에서 추출한 것인, 건강기능식품 조성물.
- 청구항 1에 있어서,상기 추출물은 물, 알코올 또는 이들의 혼합 용매로 추출된 것인, 건강기능식품 조성물.
- 청구항 1에 있어서,상기 건강기능식품은 산제, 과립제, 정제, 캡슐제, 환제, 겔, 젤리, 현탁액, 에멀젼, 시럽제, 티백제, 침출차, 껌, 캔디류 및 건강 음료로 이루어진 군으로부터 선택되는 제형을 갖는, 건강기능식품 조성물.
- 금불초(Inula japonica) 추출물, 딱지꽃(Potentilla chinensis) 추출물, 이질풀(Geranium nepalense) 추출물 및 한삼덩굴(Humulus japonicas) 추출물로 이루어진 군으로부터 선택되는 하나 이상의 추출물을 유효성분으로 포함하는 호흡기 질환 개선 또는 치료용 약학적 조성물.
- 금불초(Inula japonica) 추출물, 딱지꽃(Potentilla chinensis) 추출물, 이질풀(Geranium nepalense) 추출물 및 한삼덩굴(Humulus japonicas) 추출물로 이루어진 군으로부터 선택되는 하나 이상의 추출물을 유효성분으로 포함하는 호흡기 질환 개선 또는 예방용 의약외품 조성물.
- 유효량의 금불초(Inula japonica) 추출물, 딱지꽃(Potentilla chinensis) 추출물, 이질풀(Geranium nepalense) 추출물 및 한삼덩굴(Humulus japonicas) 추출물로 이루어진 군으로부터 선택되는 하나 이상의 추출물을 그를 필요로 하는 개체에 투여하는 단계를 포함하는 호흡기 질환을 예방, 개선 또는 치료하는 방법.
- 금불초(Inula japonica) 추출물, 딱지꽃(Potentilla chinensis) 추출물, 이질풀(Geranium nepalense) 추출물 및 한삼덩굴(Humulus japonicas) 추출물로 이루어진 군으로부터 선택되는 하나 이상의 추출물을 호흡기 질환 예방, 개선 또는 치료용 조성물의 제조에 사용하기 위한 용도.
