炎症性皮膚疾患の予防、治療又は改善剤の提供。 油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂を有効成分とする炎症性皮膚疾患の予防、治療又は改善剤。
Description
本発明は、アトピー性皮膚炎等の炎症性皮膚疾患の予防、治療又は改善剤に関する。
アトピー性皮膚炎(AD)は炎症を伴う皮膚疾患の一つで、遺伝的要因や環境要因により引き起こされる疾患である。近年の研究により、アトピー性皮膚炎の病態形成には、免疫学的異常と皮膚バリア機能の欠損の両方が関わっていることが明らかとなっている。皮膚のバリア機能は、主に角質層やその表面を覆う皮脂膜により担われ、それらが正常に機能することで体内からの水分損失を防ぎ、外界からの様々な刺激や物質の侵入を防いでいる。ところが、アトピー性皮膚炎の患者に多くみられるアトピックドライスキンにおいては皮膚バリア機能が破綻し、環境アレルゲンや刺激の侵入と湿疹の発症、悪化、痒みの誘発が悪循環的に作用して、病態の慢性化、難治化を引き起こしている。皮膚バリア機能の異常は、尋常性乾癬や他の類似の皮膚状態の患者でも顕著である。そのため、アトピー性皮膚炎等の治療にあたっては、炎症抑制と共に皮膚バリア機能の正常化により症状を寛解状態へ導くことが重要となる。
α-リノレン酸(C18:3、ALA)は、亜麻仁油やシソ実油に多く含まれるω3系高度不飽和脂肪酸である。α-リノレン酸は、抗動脈硬化作用、高血圧作用、抗アレルギー作用等があることが報告されている。また、亜麻仁油の摂取で、乾燥・敏感肌の経表皮水分蒸散量(TEWL)の値が改善することが報告されている(非特許文献1)。
一方、ジアシルグリセロールを高濃度に含む油脂は、食後の血中トリグリセリド(中性脂肪)の増加を抑制し、体内への蓄積性が少ない等の生理作用を有することが報告されている。例えば、特許文献1では、構成脂肪酸の15~90重量%が炭素数20未満のω3系不飽和脂肪酸であり、シス型ω3系不飽和脂肪酸/(シス型ω6系不飽和脂肪酸+飽和脂肪酸+トランス型不飽和脂肪酸)の重量比が1~6であるジグリセリドを60~100重量%含有する油脂組成物は、内臓脂肪燃焼性及び体脂肪燃焼性等に優れていることが報告されている。
しかしながら、構成脂肪酸としてα-リノレン酸を多く含むジアシルグリセロール含有油脂がアトピー性皮膚炎等の炎症性皮膚疾患の予防又は治療に有用であることは知られていない。
一方、ジアシルグリセロールを高濃度に含む油脂は、食後の血中トリグリセリド(中性脂肪)の増加を抑制し、体内への蓄積性が少ない等の生理作用を有することが報告されている。例えば、特許文献1では、構成脂肪酸の15~90重量%が炭素数20未満のω3系不飽和脂肪酸であり、シス型ω3系不飽和脂肪酸/(シス型ω6系不飽和脂肪酸+飽和脂肪酸+トランス型不飽和脂肪酸)の重量比が1~6であるジグリセリドを60~100重量%含有する油脂組成物は、内臓脂肪燃焼性及び体脂肪燃焼性等に優れていることが報告されている。
しかしながら、構成脂肪酸としてα-リノレン酸を多く含むジアシルグリセロール含有油脂がアトピー性皮膚炎等の炎症性皮膚疾患の予防又は治療に有用であることは知られていない。

すなわち、本発明は、以下の1)~12)に係るものである。
1)油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂を有効成分とする炎症性皮膚疾患の予防、治療又は改善剤。
2)油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂を有効成分とする皮膚バリア機能改善剤。
3)油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂を有効成分とする炎症性皮膚疾患の予防又は改善用食品。
4)油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂を有効成分とする皮膚バリア機能改善用食品。
5)炎症性皮膚疾患の予防、治療又は改善剤を製造するための、油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂の使用。
6)炎症性皮膚疾患の予防、治療又は改善に使用するための、油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂。
7)炎症性皮膚疾患の予防、治療又は改善するための、油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂の非治療的使用。
8)皮膚バリア機能改善剤を製造するための、油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂の使用。

本発明は、炎症性皮膚疾患の予防、治療又は改善剤を提供することに関する。
本発明者は、アトピー性皮膚炎モデルマウスを用いて、α-リノレン酸を多く含むジアシルグリセロール含有油脂の有効性を評価した結果、α-リノレン酸を多く含むジアシルグリセロール含有油脂を添加した餌で飼育したマウスでは皮膚炎スコアと耳介の肥厚が抑制され、また、経表皮水分蒸散量(TEWL)の上昇が抑制され、また、炎症性皮膚疾患に係る各種サイトカインの上昇が抑制され、当該油脂はアトピー性皮膚炎等の炎症性皮膚疾患の予防又は治療に有用であることを見出した。
本発明によれば、皮膚炎の抑制と皮膚バリア機能の改善により炎症性皮膚疾患の予防、治療又は改善に有効な医薬品、食品等が提供される。
本発明において「油脂」とは、脂肪酸とグリセリンとのエステル化合物を指し、モノアシルグリセロール、ジアシルグリセロール及びトリアシルグリセロールのいずれか1種以上を含むものである。油脂の種類に特に制限はなく、食用油脂として使用できるものであれば何れでもよい。
本発明に用いる油脂は、油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂である。以下、本明細書において「油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂」を単に「本発明の油脂」と記載することがある。
本発明の油脂を構成する脂肪酸中のα-リノレン酸の含有量は40質量%以上であるが、生理効果の点から、好ましくは45質量%以上、より好ましくは50質量%以上、更に好ましくは52質量%以上であり、また、酸化安定性の点から、好ましくは80質量%以下、より好ましくは70質量%以下、更に好ましくは60質量%以下である。
油脂を構成する脂肪酸中のα-リノレン酸の含有量は、40質量%以上80質量%以下、好ましくは45質量%以上80質量%以下、より好ましくは50質量%以上80質量%以下、より好ましくは50質量%以上70質量%以下、更に好ましくは52質量%以上70質量%以下、更に好ましくは52質量%以上60質量%以下である。なお、本明細書における脂肪酸量は遊離脂肪酸換算量である。
油脂を構成する脂肪酸中のα-リノレン酸の含有量は、40質量%以上80質量%以下、好ましくは45質量%以上80質量%以下、より好ましくは50質量%以上80質量%以下、より好ましくは50質量%以上70質量%以下、更に好ましくは52質量%以上70質量%以下、更に好ましくは52質量%以上60質量%以下である。なお、本明細書における脂肪酸量は遊離脂肪酸換算量である。
本発明において、油脂を構成するα-リノレン酸以外の構成脂肪酸としては、特に限定されず、飽和脂肪酸又は不飽和脂肪酸のいずれであってもよい。
油脂の風味・工業的生産性の点からは、油脂を構成する脂肪酸中の不飽和脂肪酸の含有量は、好ましくは60質量%以上100質量%以下、より好ましくは70質量%以上100質量%以下、更に好ましくは75質量%以上99質量%以下、更に好ましくは80質量%以上98質量%以下である。不飽和脂肪酸の炭素数は、生理効果の点から、好ましくは14~24、より好ましくは16~22である。
油脂の風味・工業的生産性の点からは、油脂を構成する脂肪酸中の不飽和脂肪酸の含有量は、好ましくは60質量%以上100質量%以下、より好ましくは70質量%以上100質量%以下、更に好ましくは75質量%以上99質量%以下、更に好ましくは80質量%以上98質量%以下である。不飽和脂肪酸の炭素数は、生理効果の点から、好ましくは14~24、より好ましくは16~22である。
油脂を構成する脂肪酸中のリノール酸(C18:2)の含有量は、工業的生産性の点から、好ましくは5質量%以上、より好ましくは10質量%以上であり、また、生理効果の点から、好ましくは40質量%以下、より好ましくは30質量%以下、更に好ましくは20質量%以下である。
油脂を構成する脂肪酸中のリノール酸(C18:2)の含有量は、好ましくは5質量%以上40質量%以下であり、より好ましくは10質量%以上30質量%以下、更に好ましくは10質量%以上20質量%以下である。
油脂を構成する脂肪酸中のリノール酸(C18:2)の含有量は、好ましくは5質量%以上40質量%以下であり、より好ましくは10質量%以上30質量%以下、更に好ましくは10質量%以上20質量%以下である。
また、油脂を構成する脂肪酸中のオレイン酸(C18:1)の含有量は、工業的生産性の点から、好ましくは10質量%以上であり、また、生理効果の点から、好ましくは50質量%以下、より好ましくは40質量%以下、更に好ましくは30質量%以下である。
油脂を構成する脂肪酸中のオレイン酸(C18:1)の含有量は、好ましくは10質量%以上50質量%以下であり、より好ましくは10質量%以上40質量%以下、更に好ましくは10質量%以上30質量%以下である。
油脂を構成する脂肪酸中のオレイン酸(C18:1)の含有量は、好ましくは10質量%以上50質量%以下であり、より好ましくは10質量%以上40質量%以下、更に好ましくは10質量%以上30質量%以下である。
油脂を構成する脂肪酸中の飽和脂肪酸の合計含有量は、外観、生理効果、油脂の工業的生産性の点から、好ましくは6.0質量%以下、より好ましくは5.5質量%以下、更に好ましくは5.0質量%以下である。また、工業的な生産性の点から、好ましくは0.5質量%以上である。
油脂を構成する脂肪酸中の飽和脂肪酸の合計含有量は、好ましくは0.5質量%以上6.0質量%以下、更に好ましくは0.5質量%以上5.5質量%以下、更に好ましくは0.5質量%以上5.0質量%以下である。
飽和脂肪酸の種類は特に限定されないが、好ましくは炭素数14~24の飽和脂肪酸、より好ましくは炭素数16~22の飽和脂肪酸、更に好ましくは炭素数16、18、20の飽和脂肪酸である。
油脂を構成する脂肪酸中の飽和脂肪酸の合計含有量は、好ましくは0.5質量%以上6.0質量%以下、更に好ましくは0.5質量%以上5.5質量%以下、更に好ましくは0.5質量%以上5.0質量%以下である。
飽和脂肪酸の種類は特に限定されないが、好ましくは炭素数14~24の飽和脂肪酸、より好ましくは炭素数16~22の飽和脂肪酸、更に好ましくは炭素数16、18、20の飽和脂肪酸である。
本発明の油脂中のジアシルグリセロールの含有量は25質量%以上であるが、効果を有効に発現する点、生理効果の点から、好ましくは30質量%以上、より好ましくは50質量%以上、更に好ましくは55質量%以上、更に好ましくは60質量%以上、更に好ましくは65質量%以上、更に好ましくは70質量%以上、更に好ましくは80質量%以上、更に好ましくは83質量%以上、更に好ましくは84質量%以上であり、また、好ましくは96質量%以下、より好ましくは95質量%以下、更に好ましくは94質量%以下である。
油脂中のジアシルグリセロールの含有量は、好ましくは25質量%以上96質量%以下、より好ましくは30質量%以上96質量%以下、更に好ましくは50質量%以上96質量%以下、更に好ましくは50質量%以上95質量%以下、更に好ましくは55質量%以上95質量%以下、更に好ましくは60質量%以上95質量%以下、更に好ましくは65質量%以上95質量%以下、更に好ましくは65質量%以上94質量%以下、更に好ましくは70質量%以上94質量%以下、更に好ましくは80質量%以上94質量%以下、更に好ましくは83質量%以上94質量%以下、更に好ましくは84質量%以上94質量%以下である。
油脂中のジアシルグリセロールの含有量は、好ましくは25質量%以上96質量%以下、より好ましくは30質量%以上96質量%以下、更に好ましくは50質量%以上96質量%以下、更に好ましくは50質量%以上95質量%以下、更に好ましくは55質量%以上95質量%以下、更に好ましくは60質量%以上95質量%以下、更に好ましくは65質量%以上95質量%以下、更に好ましくは65質量%以上94質量%以下、更に好ましくは70質量%以上94質量%以下、更に好ましくは80質量%以上94質量%以下、更に好ましくは83質量%以上94質量%以下、更に好ましくは84質量%以上94質量%以下である。
本発明において、ジアシルグリセロールを構成する脂肪酸は飽和脂肪酸又は不飽和脂肪酸のいずれであってもよいが、ジアシルグリセロールを構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であることが効果を有効に発現する点から好ましい。
ジアシルグリセロールを構成する脂肪酸中のα-リノレン酸の含有量は、同様の点から、好ましくは45質量%以上、より好ましくは50質量%以上、更に好ましくは52質量%以上であり、また、酸化安定性の点から、好ましくは80質量%以下、より好ましくは70質量%以下、更に好ましくは60質量%以下である。
ジアシルグリセロールを構成する脂肪酸中のα-リノレン酸の含有量は、40質量%以上80質量%以下、好ましくは45質量%以上80質量%以下、より好ましくは50質量%以上80質量%以下、より好ましくは50質量%以上70質量%以下、更に好ましくは52質量%以上70質量%以下、更に好ましくは52質量%以上60質量%以下である。
ジアシルグリセロールを構成する脂肪酸中のα-リノレン酸の含有量は、同様の点から、好ましくは45質量%以上、より好ましくは50質量%以上、更に好ましくは52質量%以上であり、また、酸化安定性の点から、好ましくは80質量%以下、より好ましくは70質量%以下、更に好ましくは60質量%以下である。
ジアシルグリセロールを構成する脂肪酸中のα-リノレン酸の含有量は、40質量%以上80質量%以下、好ましくは45質量%以上80質量%以下、より好ましくは50質量%以上80質量%以下、より好ましくは50質量%以上70質量%以下、更に好ましくは52質量%以上70質量%以下、更に好ましくは52質量%以上60質量%以下である。
本発明の油脂は、トリアシルグリセロールを含有していてもよく、その含有量は、油脂の工業的生産性の点から、好ましくは1質量%以上、より好ましくは2質量%以上、更に好ましくは5質量%以上であり、また、好ましくは75質量%以下、より好ましくは72質量%以下、より好ましくは50質量%以下、より好ましくは25質量%以下である。
油脂中のトリアシルグリセロールの含有量は、好ましくは1質量%以上75質量%以下、より好ましくは2質量%以上75質量%以下、更に好ましくは2質量%以上72質量%以下、更に好ましくは5質量%以上72質量%以下、更に好ましくは5質量%以上50質量%以下、更に好ましくは5質量%以上25質量%以下である。
また、油脂中のモノアシルグリセロールの含有量は、風味、油脂の工業的生産性の点から、好ましくは3質量%以下であり、より好ましくは2質量%以下、更に好ましくは1.51質量%以下であり、また、好ましくは0質量%超である。油脂中のモノアシルグリセロールの含有量は、0質量%でもよい。
トリアシルグリセロール、ジアシルグリセロール及びモノアシルグリセロールの脂肪酸組成は同じであることが、油脂の工業的生産性の点で好ましい。
油脂中のトリアシルグリセロールの含有量は、好ましくは1質量%以上75質量%以下、より好ましくは2質量%以上75質量%以下、更に好ましくは2質量%以上72質量%以下、更に好ましくは5質量%以上72質量%以下、更に好ましくは5質量%以上50質量%以下、更に好ましくは5質量%以上25質量%以下である。
また、油脂中のモノアシルグリセロールの含有量は、風味、油脂の工業的生産性の点から、好ましくは3質量%以下であり、より好ましくは2質量%以下、更に好ましくは1.51質量%以下であり、また、好ましくは0質量%超である。油脂中のモノアシルグリセロールの含有量は、0質量%でもよい。
トリアシルグリセロール、ジアシルグリセロール及びモノアシルグリセロールの脂肪酸組成は同じであることが、油脂の工業的生産性の点で好ましい。
本発明の油脂は、不純物として遊離脂肪酸又はその塩を含有していてもよい。油脂中の遊離脂肪酸又はその塩の含有量は、風味の点から、好ましくは3質量%以下、より好ましくは2質量%以下、更に好ましくは1質量%以下であり、また、好ましくは0質量%超である。油脂中の遊離脂肪酸又はその塩の含有量は、0質量%でもよい。
本発明の油脂は、常法に従って、油脂由来の脂肪酸とグリセリンとのエステル化反応、油脂とグリセリンとのエステル交換反応(グリセロリシス)等により得ることができる。必要に応じて通常の食用油脂を混合してもよい。
エステル化反応とグリセロリシス反応は、アルカリ金属又はその合金、アルカリ金属又はアルカリ土類金属の酸化物や水酸化物、アルカリ金属又はアルカリ土類金属のアルコキシド等の化学触媒を用いる化学法と、リパーゼ等の酵素を用いる酵素法とに大別される。
なかでも、脂肪酸組成を制御する点から、後述する油脂を加水分解して得られる脂肪酸(分別脂肪酸)とグリセリンとのエステル化反応が好ましい。
エステル化反応とグリセロリシス反応は、アルカリ金属又はその合金、アルカリ金属又はアルカリ土類金属の酸化物や水酸化物、アルカリ金属又はアルカリ土類金属のアルコキシド等の化学触媒を用いる化学法と、リパーゼ等の酵素を用いる酵素法とに大別される。
なかでも、脂肪酸組成を制御する点から、後述する油脂を加水分解して得られる脂肪酸(分別脂肪酸)とグリセリンとのエステル化反応が好ましい。
本発明において、油脂の起源は、食用油脂として使用できるものであれば特に制限はなく、植物性油脂、動物性油脂のいずれでもよい。例えば、大豆油、菜種油、サフラワー油、米油、コーン油、ヒマワリ油、綿実油、オリーブ油、ゴマ油、落花生油、ハトムギ油、小麦胚芽油、シソ油、アマニ油、エゴマ油、チアシード油、サチャインチ油、クルミ油、キウイ種子油、サルビア種子油、ブドウ種子油、マカデミアナッツ油、ヘーゼルナッツ油、カボチャ種子油、椿油、茶実油、ボラージ油、パーム油、パームオレイン、パームステアリン、やし油、パーム核油、カカオ脂、サル脂、シア脂、藻油等の植物性油脂;魚油、アザラシ油、ラード、牛脂、バター脂等の動物性油脂;あるいはこれらのエステル交換油、水素添加油、分別油等の油脂類を挙げることができる。
これらの油は、それぞれ単独で用いてもよく、あるいは適宜混合して用いてもよい。なかでも、使用性の点から、植物性油脂を用いるのが好ましく、更に低温耐性に優れた液状油脂を用いるのが好ましく、更にシソ油、アマニ油及びエゴマ油からなる群より選ばれる1種又は2種以上の油脂を用いるのが、α-リノレン酸を豊富に含むため好ましい。なお、液状油脂とは、基準油脂分析試験法2.3.8-27による冷却試験を実施した場合、20℃で液状である油脂をいう。また、食用油脂は、精製工程を経た精製油脂であるのが好ましい。

油脂由来の脂肪酸は、油脂を加水分解して得ることができる。油脂を加水分解する方法としては、高温高圧分解法と酵素分解法が挙げられる。高温高圧分解法とは、油脂に水を加えて、高温、高圧の条件で反応することにより、脂肪酸とグリセリンを得る方法である。また、酵素分解法とは、油脂に水を加えて、油脂加水分解酵素を触媒として用い、低温の条件で反応することにより、脂肪酸とグリセリンを得る方法である。
加水分解反応は、常法に従って行うことができる。

油脂の加水分解後は、加水分解反応物を分別して固体を除去することが好ましい。分別方法としては、溶剤分別法、自然分別法(ドライ分別法)、湿潤剤分別法が挙げられる。
析出した固体の除去手段としては、静置分離、濾過、遠心分離、脂肪酸に湿潤剤水溶液を混合し分離する方法等が挙げられる。

油脂由来の脂肪酸とグリセリンとのエステル化反応は、酵素法により温和な条件で行うのが風味等の点で優れており好ましい。
酵素の使用量は、酵素の活性を考慮して適宜決定することができるが、反応速度を向上する点から、固定化酵素を使用する場合は、エステル化反応原料の合計質量に対して、好ましくは1~30質量%、より好ましくは2~20質量%である。
エステル化反応の反応温度は、反応速度を向上する点、酵素の失活を抑制する点から、好ましくは0~100℃、より好ましくは20~80℃、更に好ましくは30~60℃である。また、反応時間は、工業的な生産性の点から、好ましくは15時間以内、より好ましくは1~12時間、更に好ましくは2~10時間である。
脂肪酸とグリセリンとの接触手段としては、浸漬、攪拌、固定化リパーゼを充填したカラムにポンプ等で通液する方法等が挙げられる。
酵素の使用量は、酵素の活性を考慮して適宜決定することができるが、反応速度を向上する点から、固定化酵素を使用する場合は、エステル化反応原料の合計質量に対して、好ましくは1~30質量%、より好ましくは2~20質量%である。
エステル化反応の反応温度は、反応速度を向上する点、酵素の失活を抑制する点から、好ましくは0~100℃、より好ましくは20~80℃、更に好ましくは30~60℃である。また、反応時間は、工業的な生産性の点から、好ましくは15時間以内、より好ましくは1~12時間、更に好ましくは2~10時間である。

エステル化反応の後は、通常油脂に対して用いられる精製工程を行ってもよい。具体的には、蒸留処理、酸処理、水洗、脱色、脱臭等の工程を挙げることができる。
後記実施例に示すとおり、本発明の油脂をアトピー性皮膚炎モデルマウスに投与すると、アトピー性皮膚炎の進行の程度を表す皮膚炎スコアと耳介の肥厚が抑制され、また、経表皮水分蒸散量(TEWL)の上昇が抑制された。加えて、皮膚炎誘導部位の皮膚におけるmRNA発現解析を行ったところ、本発明の油脂を摂取させた群では、主要なTh2サイトカインであるIL-4、自然免疫担当細胞やケラチノサイトから分泌されるサイトカインであるIL-1β、IL-6、TSLP、IL-33の発現上昇が抑制された。
本発明の油脂によって発現上昇が抑制された上記サイトカインは、アトピー性皮膚炎のみならず脂漏性皮膚炎、接触性皮膚炎、乾癬、皮脂欠乏性湿疹、結節性紅斑等の種々の炎症性皮膚疾患における炎症応答やバリア機能の低下に寄与していることが報告されている(参考文献; Adalsteinsson JA, Kaushik S, et al. Exp Dermatol. 2020 May;29(5):481-489、Lee HY, et al. Mediators Inflamm. 2013;2013:916497、Belge K, et al. F1000Prime Rep. 2014 Jan 2;6:4、Hanel KH, et al. Int J Mol Sci. 2013 Mar 26;14(4):6720-45、Polycarpou A, et al. Front Immunol. 2017 Mar 13;8:233)。従って、本発明の油脂は、炎症性皮膚疾患の予防、治療又は改善剤、皮膚バリア機能改善剤(以下、「炎症性皮膚疾患の予防、治療又は改善剤等」とも称す)となり得、炎症性皮膚疾患の予防、治療又は改善のため、皮膚バリア機能の改善のために使用することができ、また炎症性皮膚疾患の予防、治療又は改善剤等を製造するために使用することができる。
ここで、「使用」は、ヒト若しくは非ヒト動物への使用であり得、治療的使用であっても非治療的使用であってもよい。「非治療的」とは、医療行為を含まない、すなわち人間を手術、治療又は診断する方法を含まない、より具体的には医師、又は医療従事者もしくは医師の指示を受けた者が人間に対して手術、治療又は診断を実施する方法を含まない概念である。
本発明の油脂によって発現上昇が抑制された上記サイトカインは、アトピー性皮膚炎のみならず脂漏性皮膚炎、接触性皮膚炎、乾癬、皮脂欠乏性湿疹、結節性紅斑等の種々の炎症性皮膚疾患における炎症応答やバリア機能の低下に寄与していることが報告されている(参考文献; Adalsteinsson JA, Kaushik S, et al. Exp Dermatol. 2020 May;29(5):481-489、Lee HY, et al. Mediators Inflamm. 2013;2013:916497、Belge K, et al. F1000Prime Rep. 2014 Jan 2;6:4、Hanel KH, et al. Int J Mol Sci. 2013 Mar 26;14(4):6720-45、Polycarpou A, et al. Front Immunol. 2017 Mar 13;8:233)。従って、本発明の油脂は、炎症性皮膚疾患の予防、治療又は改善剤、皮膚バリア機能改善剤(以下、「炎症性皮膚疾患の予防、治療又は改善剤等」とも称す)となり得、炎症性皮膚疾患の予防、治療又は改善のため、皮膚バリア機能の改善のために使用することができ、また炎症性皮膚疾患の予防、治療又は改善剤等を製造するために使用することができる。

本明細書において、対象となる「炎症性皮膚疾患」の例としては、アトピー性皮膚炎、脂漏性皮膚炎、接触性皮膚炎、乾癬、皮脂欠乏性湿疹、結節性紅斑、新生児座瘡・座瘡様発疹、汗疹、毛包炎、乳児湿疹、乾燥肌等が挙げられる。なかでも、アトピー性皮膚炎に好適である。「アトピー性皮膚炎」は、増悪と寛解を繰り返す、痒みを伴う湿疹を主病変とする慢性に経過する疾患を指す。
「皮膚バリア機能」とは、皮膚の内側からの水分の蒸散を防ぐ機能及び外界から皮膚への物質の侵入を防ぐ機能を意味する。本発明においては、皮膚の内側からの水分の蒸散を防ぐ機能(水分透過バリア機能)の改善に適する。皮膚バリア機能の改善により、皮膚バリア機能の低下に起因する乾皮症(皮脂欠乏性)、皮脂欠乏性湿疹等の治療又は改善が可能となる。
「予防」とは、個体における疾患若しくは症状の発症の防止又は遅延、あるいは個体の疾患若しくは症状の発症の危険性を低下させることをいう。
「改善」とは、疾患、症状又は状態の好転、疾患、症状又は状態の悪化の防止又は遅延、あるいは疾患又は症状の進行の逆転、防止又は遅延をいう。
また、「治療」には、疾患の完全治癒に加えて、症状を改善させることが包含される。
「改善」とは、疾患、症状又は状態の好転、疾患、症状又は状態の悪化の防止又は遅延、あるいは疾患又は症状の進行の逆転、防止又は遅延をいう。
また、「治療」には、疾患の完全治癒に加えて、症状を改善させることが包含される。
本発明の炎症性皮膚疾患の予防、治療又は改善剤等は、それ自体、炎症性皮膚疾患の予防、治療又は改善のため、皮膚バリア機能の改善のための医薬品、医薬部外品、化粧品、食品又は飼料となり、また当該医薬品、医薬部外品、化粧品、食品又は飼料に配合して使用される素材又は製剤となり得る。
当該医薬品(医薬部外品を含む、以下同じ)は、本発明の油脂を、炎症性皮膚疾患の予防、治療又は改善のため、皮膚バリア機能の改善のための有効成分として含有する。更に、該医薬品は、該有効成分の機能が失われない限りにおいて、必要に応じて薬学的に許容される担体、又は他の有効成分、薬理成分等を含有していてもよい。
医薬品は任意の投与形態で投与され得る。投与形態としては、例えば、錠剤(チュアブル錠等を含む)、カプセル剤、顆粒剤、散剤、トローチ剤等の経口固形製剤、内服液剤、シロップ剤等の経口液状製剤、注射剤、坐剤、吸入薬、経皮吸収剤、外用剤等の非経口投与製剤が挙げられる。好ましい投与形態は経口投与である。形態は使用目的に応じて大きさを任意に調節することができる。
このような種々の剤形の製剤は、必要に応じて、薬学的に許容される担体、例えば、賦形剤、結合剤、増量剤、崩壊剤、界面活性剤、滑沢剤、分散剤、緩衝剤、浸透圧調整剤、pH調整剤、乳化剤、防腐剤、安定剤、保存剤、増粘剤、流動性改善剤、嬌味剤、発泡剤、香料、被膜剤、希釈剤等や、他の薬効成分等と適宜組み合わせて、定法に従って調製することができる。
医薬品は任意の投与形態で投与され得る。投与形態としては、例えば、錠剤(チュアブル錠等を含む)、カプセル剤、顆粒剤、散剤、トローチ剤等の経口固形製剤、内服液剤、シロップ剤等の経口液状製剤、注射剤、坐剤、吸入薬、経皮吸収剤、外用剤等の非経口投与製剤が挙げられる。好ましい投与形態は経口投与である。形態は使用目的に応じて大きさを任意に調節することができる。
このような種々の剤形の製剤は、必要に応じて、薬学的に許容される担体、例えば、賦形剤、結合剤、増量剤、崩壊剤、界面活性剤、滑沢剤、分散剤、緩衝剤、浸透圧調整剤、pH調整剤、乳化剤、防腐剤、安定剤、保存剤、増粘剤、流動性改善剤、嬌味剤、発泡剤、香料、被膜剤、希釈剤等や、他の薬効成分等と適宜組み合わせて、定法に従って調製することができる。
当該化粧品は、本発明の油脂を、炎症性皮膚疾患の予防又は改善のため、皮膚バリア機能の改善のための有効成分として含有する。さらに、該化粧品は、該有効成分の機能が失われない限りにおいて、必要に応じて化粧料に許容される担体、又は他の有効成分、薬効成分、化粧成分等を含有していてもよい。
本発明の油脂を含む化粧品の好ましい例としては、顔、ボディ用の化粧料(例えば、ローション、ゲル、クリーム、パック等)、メークアップ用化粧料、顔又はボディ用の洗浄料等が挙げられる。
斯かる化粧品の各製剤は、常法に従って製造することができる。化粧料に許容される担体としては、例えば、各種油剤、界面活性剤、ゲル化剤、緩衝剤、防腐剤、酸化防止剤、溶剤、分散剤、キレート剤、増粘剤、紫外線吸収剤、乳化安定剤、pH調整剤、色素、香料等が挙げられる。
他の有効成分、薬効成分、化粧成分としては、例えば、植物抽出物、殺菌剤、保湿剤、抗炎症剤、抗菌剤、角質溶解剤、清涼剤、抗脂漏剤、洗浄剤、メークアップ成分等が挙げられる。
本発明の油脂を含む化粧品の好ましい例としては、顔、ボディ用の化粧料(例えば、ローション、ゲル、クリーム、パック等)、メークアップ用化粧料、顔又はボディ用の洗浄料等が挙げられる。
斯かる化粧品の各製剤は、常法に従って製造することができる。化粧料に許容される担体としては、例えば、各種油剤、界面活性剤、ゲル化剤、緩衝剤、防腐剤、酸化防止剤、溶剤、分散剤、キレート剤、増粘剤、紫外線吸収剤、乳化安定剤、pH調整剤、色素、香料等が挙げられる。
他の有効成分、薬効成分、化粧成分としては、例えば、植物抽出物、殺菌剤、保湿剤、抗炎症剤、抗菌剤、角質溶解剤、清涼剤、抗脂漏剤、洗浄剤、メークアップ成分等が挙げられる。
当該食品は、本発明の油脂を、炎症性皮膚疾患の予防又は改善のため、皮膚バリア機能の改善のための有効成分として含有する。
食品には、炎症性皮膚疾患にみられる症状の予防又は改善、或いは皮膚バリア機能の改善を訴求とし、必要に応じてその旨の表示が許可又は届出された食品(特定保健用食品、機能性表示食品、特別用途食品等)が含まれる。表示の例としては、「肌の水分保持に役立ち、乾燥を緩和する機能がある」、「肌の不快感を改善する機能がある」等がある。機能表示が許可又は届出された食品は、一般の食品と区別することができる。
食品の形態は、固形、半固形又は液状(例えば飲料)であり得る。例としては、各種食品組成物(パン類、ケーキ類、麺類、菓子類、冷凍食品、アイスクリーム類、あめ類、ふりかけ類、スープ類、乳製品、シェイク、飲料、調味料等)、更には、上述した経口投与製剤と同様の形態(顆粒剤、粉剤、錠剤、カプセル剤、マイクロカプセル剤、トローチ剤等の固形製剤)の栄養補給用組成物が挙げられる。
種々の形態の食品は、本発明に油脂を、任意の食品材料、若しくは他の有効成分、又は食品に許容される添加物(例えば、溶剤、軟化剤、油、乳化剤、防腐剤、酸味料、甘味料、苦味料、pH調整剤、安定剤、着色剤、紫外線吸収剤、酸化防止剤、保湿剤、増粘剤、固着剤、分散剤、流動性改善剤、湿潤剤、香科、調味料、風味調整剤)等と適宜組み合わせて、定法に従って調製することができる。
食品には、炎症性皮膚疾患にみられる症状の予防又は改善、或いは皮膚バリア機能の改善を訴求とし、必要に応じてその旨の表示が許可又は届出された食品(特定保健用食品、機能性表示食品、特別用途食品等)が含まれる。表示の例としては、「肌の水分保持に役立ち、乾燥を緩和する機能がある」、「肌の不快感を改善する機能がある」等がある。機能表示が許可又は届出された食品は、一般の食品と区別することができる。

種々の形態の食品は、本発明に油脂を、任意の食品材料、若しくは他の有効成分、又は食品に許容される添加物(例えば、溶剤、軟化剤、油、乳化剤、防腐剤、酸味料、甘味料、苦味料、pH調整剤、安定剤、着色剤、紫外線吸収剤、酸化防止剤、保湿剤、増粘剤、固着剤、分散剤、流動性改善剤、湿潤剤、香科、調味料、風味調整剤)等と適宜組み合わせて、定法に従って調製することができる。
当該飼料は、本発明の油脂を、炎症性皮膚疾患の予防又は改善のため、皮膚バリア機能の改善のための有効成分として含有する。
飼料の形態としては、好ましくはペレット状、フレーク状、マッシュ状又はリキッド状であり、例えば、牛、豚、鶏、羊、馬等に用いる家畜用飼料、ウサギ、ラット、マウス等に用いる小動物用飼料、犬、猫、小鳥等に用いるペットフード等が挙げられる。
飼料は、本発明の油脂を、他の飼料材料、例えば、肉類、蛋白質、穀物類、ぬか類、粕類、糖類、野菜、ビタミン類、ミネラル類、ゲル化剤、保型剤、pH調整剤、調味料、防腐剤、栄養補強剤等と適宜組み合わせて、定法に従って調製することができる。
飼料の形態としては、好ましくはペレット状、フレーク状、マッシュ状又はリキッド状であり、例えば、牛、豚、鶏、羊、馬等に用いる家畜用飼料、ウサギ、ラット、マウス等に用いる小動物用飼料、犬、猫、小鳥等に用いるペットフード等が挙げられる。
飼料は、本発明の油脂を、他の飼料材料、例えば、肉類、蛋白質、穀物類、ぬか類、粕類、糖類、野菜、ビタミン類、ミネラル類、ゲル化剤、保型剤、pH調整剤、調味料、防腐剤、栄養補強剤等と適宜組み合わせて、定法に従って調製することができる。
製剤中の本発明の油脂の含有量は、製剤の形態に応じて異なるため一概には言えないが、例えば製剤の総量を基準として、好ましくは0.5質量%以上であり、また、好ましくは99.8質量%以下である。
本発明の油脂の投与量又は使用量は、本発明の効果を達成できる量であり得る。当該投与量又は使用量は、対象の種、体重、性別、年齢、状態、又はその他の要因に従って変動し得るが、経口投与(摂取)の場合には、成人(60kg)1人当たり1回、本発明の油脂として、例えば0.3~15gである。
上記製剤は、任意の投与計画に従って投与又は使用され得るが、1日に1回~複数回に分けて、1日間以上、好ましくは7日間以上、より好ましくは14日間以上、よりさらに好ましくは30日間以上、反復・継続して投与又は使用し得る。

本発明の炎症性皮膚疾患の予防、治療又は改善剤等は、油脂組成物の形態として投与又は使用されることが好ましい。
炎症性皮膚疾患の予防、治療又は改善剤等が油脂組成物の形態である場合、油脂組成物中の本発明の油脂の含有量は、使用性の点から、好ましくは90質量%以上、より好ましくは95質量%以上であり、また、好ましくは100質量%以下、より好ましくは99.9質量%以下である。
炎症性皮膚疾患の予防、治療又は改善剤等が油脂組成物の形態である場合、油脂組成物中の本発明の油脂の含有量は、使用性の点から、好ましくは90質量%以上、より好ましくは95質量%以上であり、また、好ましくは100質量%以下、より好ましくは99.9質量%以下である。
油脂組成物は、風味、酸化安定性、着色抑制等の点から、抗酸化剤を含有するのが好ましい。油脂組成物中の抗酸化剤の含有量は、風味、酸化安定性、着色抑制等の点から、好ましくは0.005質量%以上1.0質量%以下、より好ましくは0.04質量%以上0.75質量%以下、更に好ましくは0.08質量%以上0.5質量%以下である。
抗酸化剤としては、食品に使用するものであれば特に制限はないが、天然抗酸化剤、レシチン、トコフェロール、アスコルビルパルミテート、アスコルビルステアレート、ジブチルヒドロキシトルエン(BHT)及びブチルヒドロキシアニソール(BHA)等から選ばれる少なくとも1種が好ましい。
抗酸化剤としては、食品に使用するものであれば特に制限はないが、天然抗酸化剤、レシチン、トコフェロール、アスコルビルパルミテート、アスコルビルステアレート、ジブチルヒドロキシトルエン(BHT)及びブチルヒドロキシアニソール(BHA)等から選ばれる少なくとも1種が好ましい。
本発明の炎症性皮膚疾患の予防、治療又は改善剤等を投与又は使用する対象としては、炎症性皮膚疾患、好ましくはアトピー性皮膚炎の予防や治療、改善を所望するヒト、皮膚バリア機能の改善を所望するヒト等が挙げられる。非ヒト動物としては、類人猿、その他霊長類等の非ヒト哺乳動物等が挙げられる。
上述した実施形態に関し、本発明は以下の態様をさらに開示する。
<1>油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂を有効成分とする炎症性皮膚疾患の予防、治療又は改善剤。
<2>油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂を有効成分とする皮膚バリア機能改善剤。
<3>油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂を有効成分とする炎症性皮膚疾患の予防又は改善用食品。
<4>油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂を有効成分とする皮膚バリア機能改善用食品。
<5>油脂を構成する脂肪酸中のα-リノレン酸の含有量が、好ましくは45質量%以上、より好ましくは50質量%以上、更に好ましくは52質量%以上であり、また、好ましくは80質量%以下、より好ましくは70質量%以下、更に好ましくは60質量%以下であり、また、40質量%以上80質量%以下、好ましくは45質量%以上80質量%以下、より好ましくは50質量%以上80質量%以下、より好ましくは50質量%以上70質量%以下、更に好ましくは52質量%以上70質量%以下、更に好ましくは52質量%以上60質量%以下である<1>~<4>のいずれかに記載の剤又は食品。
<6>油脂を構成する脂肪酸中のリノール酸(C18:2)の含有量が、好ましくは5質量%以上、より好ましくは10質量%以上であり、また、好ましくは40質量%以下、より好ましくは30質量%以下、更に好ましくは20質量%以下であり、また、好ましくは5質量%以上40質量%以下であり、より好ましくは10質量%以上30質量%以下、更に好ましくは10質量%以上20質量%以下である<1>~<5>のいずれかに記載の剤又は食品。

実施例1
〔油脂及び特殊飼料の調製〕
アマニ油(Archer Daniels Midland社製)を酵素により加水分解して得られた脂肪酸を、冷却及び遠心分離により分別して分別脂肪酸を得た。得られた分別脂肪酸300質量部とグリセリン47質量部を混合し、市販の固定化1,3位選択リパーゼ(ノボザイム社)を触媒としてエステル化反応を行った。固定化酵素を濾別した後、反応終了品を分子蒸留し、酸処理及び水洗を行い、処理油を得た。処理油を脱臭後、抗酸化剤を添加して、アマニ油由来ジアシルグリセロール(DAG)含有油脂(lin-DAG)を調製した。
〔油脂及び特殊飼料の調製〕
アマニ油(Archer Daniels Midland社製)を酵素により加水分解して得られた脂肪酸を、冷却及び遠心分離により分別して分別脂肪酸を得た。得られた分別脂肪酸300質量部とグリセリン47質量部を混合し、市販の固定化1,3位選択リパーゼ(ノボザイム社)を触媒としてエステル化反応を行った。固定化酵素を濾別した後、反応終了品を分子蒸留し、酸処理及び水洗を行い、処理油を得た。処理油を脱臭後、抗酸化剤を添加して、アマニ油由来ジアシルグリセロール(DAG)含有油脂(lin-DAG)を調製した。
上記方法にて調製したアマニ油由来DAG油脂(lin-DAG)、アマニ油(lin-TAG、サミット製油製)、及びコントロールとして大豆油(soy-TAG、日清オイリオグループ(株)製)を用意した。なお、lin-TAGおよびsoy-TAGにも、lin-DAGと同じ抗酸化剤を同濃度添加した。
マウス飼料として用いられるAIN-93G(オリエンタル酵母工業株式会社)から元々含まれる大豆油(7質量%)及びコーンスターチ(3質量%)を除去したものに対して、上記いずれかの油脂を10質量%添加することにより、3種類の特殊飼料を調製した。
lin-DAG、lin-TAG及びsoy-TAGのグリセリド組成及び脂肪酸組成を表1に示す。なお、表1の脂肪酸組成におけるC18:3の不飽和脂肪酸は、α-リノレン酸である。
マウス飼料として用いられるAIN-93G(オリエンタル酵母工業株式会社)から元々含まれる大豆油(7質量%)及びコーンスターチ(3質量%)を除去したものに対して、上記いずれかの油脂を10質量%添加することにより、3種類の特殊飼料を調製した。
lin-DAG、lin-TAG及びsoy-TAGのグリセリド組成及び脂肪酸組成を表1に示す。なお、表1の脂肪酸組成におけるC18:3の不飽和脂肪酸は、α-リノレン酸である。
グリセリド組成及び脂肪酸組成の分析方法は以下のとおりである。
(i)油脂のグリセリド組成
ガラス製サンプル瓶に、油脂サンプル約10mgとトリメチルシリル化剤(「シリル化剤TH」、関東化学製)0.5mLを加え、密栓し、70℃で15分間加熱した。これに水1.0mLとヘキサン1.5mLを加え、振とうした。静置後、上層をガスクロマトグラフィー(GLC)に供して分析した。

〔試験概要〕
(1)NC/Nga雄性マウス32匹を、非感作群(soy-TAG(-)群)と経皮感作群(soy-TAG(+)群、lin-TAG(+)群及びlin-DAG(+)群)の4群に群分けし(各群、N=8)、経皮感作群のマウスに対して前記3種類の特殊飼料のいずれかをそれぞれ自由摂取させた。なお、非感作群のマウスにはsoy-TAG含有の特殊飼料を自由摂食させた。30日後、経皮感作群のマウスに対し、コナヒョウヒダニ由来アレルゲンを含有するビオスタAD軟膏(株式会社ビオスタ)100mgをマウス頚背部および耳介部に均一に塗布した。1回目のビオスタAD塗布から4日後、マウス頚背部および耳介部に4質量% ドデシル硫酸ナトリウム(SDS)水溶液 150uLを均一に塗布し、荒れ肌を誘導した後、再度ビオスタAD軟膏100mgを同一部位に均一に塗布した。非感作群のマウスに対しては、ビオスタAD軟膏の塗布は行わず、SDS水溶液の塗布のみ行った。この操作を1週間に2回の頻度で、さらに4回行いアトピー性皮膚炎を誘導した。初回のビオスタAD軟膏塗布から22日後、マウスをイソフルラン吸入による麻酔下にて、心採血により安楽死させ、皮膚炎誘導部位である頚背部の皮膚を採取した。初回のビオスタAD軟膏塗布前及び塗布後7、14、21日後に目視による皮膚炎スコアの評価、耳介厚の測定、Tewameter(登録商標)TM300(Courage + Khazaka electronic GmbH)による経表皮水分蒸散量(TEWL)の測定、皮膚炎誘導部位の皮膚における各種サイトカインのmRNA発現解析を行った。

皮膚炎スコア(ADスコア)の評価は、下記表3の各指標に従った。各指標、i)発赤・出血、(ii)痂皮形成・乾燥、(iii)浮腫、(iv)擦傷・組織欠損の4項目(各0-3点)を評価し、得られたスコアの合計値(0-12点)を最終的な皮膚炎スコアとした。
皮膚炎誘導部位の皮膚におけるmRNA発現解析は以下に従って行った。
採取した頚背部の皮膚から、RNeasy Mini Kit(QIAGEN)を用いて推奨プロトコルに従いTotal RNAを抽出した。得られたTotal RNAから、SuperScriptTM VILOTM cDNA Synthesis Kit (Thermo Fisher Scientific)を用いて推奨プロトコルに従いcDNA合成した。その後、得られたcDNAを用いて、real-time PCRを行うことで、皮膚炎誘導症部位の皮膚に含まれる各種サイトカインのmRNAの発現解析を行った。この時、real-time PCRではTaqmanTM Fast Universal PCR Master Mix(2×)(Applied Biosystems)と各遺伝子固有のTaqmanTM probeを用いてサンプルを調製することで、mRNA発現量の検出を行った。各種サイトカイン遺伝子の発現解析で用いたTaqmanTM probeを下記表4に示す。また、RPLP0を内在性コントロールとして用いた。

(2)結果
図1、図2及び図3に示すように、非感作群のsoy-TAG(-)群に比較し、経皮感作群のsoy-TAG及びlin-TAGを含有する特殊飼料を摂取させたsoy-TAG(+)群及びlin-TAG(+)群においては皮膚炎スコア、耳介の肥厚及び経表皮水分蒸散量(TEWL)の上昇が見られたが、lin-DAGを含有する特殊飼料を摂取させたlin-DAG(+)群においては、皮膚炎スコア、耳介の肥厚及び経TEWLの全てが有意に抑制された。
また、図4に示すように、lin-DAG(+)群の皮膚炎誘導部位の皮膚において、IL-4、IL-1β、IL-6、TSLP、IL-33の発現上昇の有意な抑制が確認された。
これらサイトカインは、脂漏性皮膚炎、接触性皮膚炎、乾癬、皮脂欠乏性湿疹、結節性紅斑、新生児座瘡・座瘡様発疹、汗疹、毛包炎、乳児湿疹、乾燥肌等の種々の炎症性皮膚疾患における炎症応答やバリア機能の低下に寄与していることが報告されている。従って、本発明の油脂は、アトピー性皮膚炎を始めとする、脂漏性皮膚炎、接触性皮膚炎、乾癬、皮脂欠乏性湿疹、結節性紅斑、新生児座瘡・座瘡様発疹、汗疹、毛包炎、乳児湿疹、乾燥肌等の種々の炎症性皮膚疾患の予防、治療又は改善剤等に有用であるといえる。
図1、図2及び図3に示すように、非感作群のsoy-TAG(-)群に比較し、経皮感作群のsoy-TAG及びlin-TAGを含有する特殊飼料を摂取させたsoy-TAG(+)群及びlin-TAG(+)群においては皮膚炎スコア、耳介の肥厚及び経表皮水分蒸散量(TEWL)の上昇が見られたが、lin-DAGを含有する特殊飼料を摂取させたlin-DAG(+)群においては、皮膚炎スコア、耳介の肥厚及び経TEWLの全てが有意に抑制された。
また、図4に示すように、lin-DAG(+)群の皮膚炎誘導部位の皮膚において、IL-4、IL-1β、IL-6、TSLP、IL-33の発現上昇の有意な抑制が確認された。
これらサイトカインは、脂漏性皮膚炎、接触性皮膚炎、乾癬、皮脂欠乏性湿疹、結節性紅斑、新生児座瘡・座瘡様発疹、汗疹、毛包炎、乳児湿疹、乾燥肌等の種々の炎症性皮膚疾患における炎症応答やバリア機能の低下に寄与していることが報告されている。従って、本発明の油脂は、アトピー性皮膚炎を始めとする、脂漏性皮膚炎、接触性皮膚炎、乾癬、皮脂欠乏性湿疹、結節性紅斑、新生児座瘡・座瘡様発疹、汗疹、毛包炎、乳児湿疹、乾燥肌等の種々の炎症性皮膚疾患の予防、治療又は改善剤等に有用であるといえる。
Claims (36)
- 油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂を有効成分とする炎症性皮膚疾患の予防、治療又は改善剤。
- 炎症性皮膚疾患がアトピー性皮膚炎、脂漏性皮膚炎、接触性皮膚炎、乾癬、皮脂欠乏性湿疹、結節性紅斑、新生児座瘡・座瘡様発疹、汗疹、毛包炎、乳児湿疹又は乾燥肌である請求項1記載の炎症性皮膚疾患の予防、治療又は改善剤。
- ジアシルグリセロールを構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上である請求項1又は2記載の炎症性皮膚疾患の予防、治療又は改善剤。
- 油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂を有効成分とする皮膚バリア機能改善剤。
- 皮膚バリア機能が水分透過バリア機能である請求項4記載の皮膚バリア機能改善剤。
- ジアシルグリセロールを構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上である請求項4又は5記載の皮膚バリア機能改善剤。
- 油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂を有効成分とする炎症性皮膚疾患の予防又は改善用食品。
- 炎症性皮膚疾患がアトピー性皮膚炎、脂漏性皮膚炎、接触性皮膚炎、乾癬、皮脂欠乏性湿疹、結節性紅斑、新生児座瘡・座瘡様発疹、汗疹、毛包炎、乳児湿疹又は乾燥肌である請求項7記載の炎症性皮膚疾患の予防又は改善用食品。
- ジアシルグリセロールを構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上である請求項7又は8記載の炎症性皮膚疾患の予防又は改善用食品。
- 油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂を有効成分とする皮膚バリア機能改善用食品。
- 皮膚バリア機能が水分透過バリア機能である請求項10記載の皮膚バリア機能改善用食品。
- ジアシルグリセロールを構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上である請求項10又は11記載の皮膚バリア機能改善用食品。
- 炎症性皮膚疾患の予防、治療又は改善剤を製造するための、油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂の使用。
- 炎症性皮膚疾患がアトピー性皮膚炎、脂漏性皮膚炎、接触性皮膚炎、乾癬、皮脂欠乏性湿疹、結節性紅斑、新生児座瘡・座瘡様発疹、汗疹、毛包炎、乳児湿疹又は乾燥肌である請求項13記載の使用。
- ジアシルグリセロールを構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上である請求項13又は14記載の使用。
- 皮膚バリア機能改善剤を製造するための、油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂の使用。
- 皮膚バリア機能が水分透過バリア機能である請求項16記載の使用。
- ジアシルグリセロールを構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上である請求項16又は17記載の使用。
- 炎症性皮膚疾患の予防、治療又は改善に使用するための、油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂。
- 炎症性皮膚疾患がアトピー性皮膚炎、脂漏性皮膚炎、接触性皮膚炎、乾癬、皮脂欠乏性湿疹、結節性紅斑、新生児座瘡・座瘡様発疹、汗疹、毛包炎、乳児湿疹又は乾燥肌である請求項19記載の油脂。
- ジアシルグリセロールを構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上である請求項19又は20記載の油脂。
- 皮膚バリア機能改善に使用するための、油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂。
- 皮膚バリア機能が水分透過バリア機能である請求項22記載の油脂。
- ジアシルグリセロールを構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上である請求項22又は23記載の油脂。
- 炎症性皮膚疾患の予防、治療又は改善するための、油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂の非治療的使用。
- 炎症性皮膚疾患がアトピー性皮膚炎、脂漏性皮膚炎、接触性皮膚炎、乾癬、皮脂欠乏性湿疹、結節性紅斑、新生児座瘡・座瘡様発疹、汗疹、毛包炎、乳児湿疹又は乾燥肌である請求項25記載の非治療的使用。
- ジアシルグリセロールを構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上である請求項25又は26記載の非治療的使用。
- 皮膚バリア機能を改善するための、油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂の非治療的使用。
- 皮膚バリア機能が水分透過バリア機能である請求項28記載の非治療的使用。
- ジアシルグリセロールを構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上である請求項28又は29記載の非治療的使用。
- 油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂を対象に投与又は適用する、炎症性皮膚疾患の予防、治療又は改善方法。
- 炎症性皮膚疾患がアトピー性皮膚炎、脂漏性皮膚炎、接触性皮膚炎、乾癬、皮脂欠乏性湿疹、結節性紅斑、新生児座瘡・座瘡様発疹、汗疹、毛包炎、乳児湿疹又は乾燥肌である請求項31記載の方法。
- ジアシルグリセロールを構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上である請求項31又は32記載の方法。
- 油脂を構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上であり、ジアシルグリセロールの含有量が25質量%以上である油脂を対象に投与又は適用する、皮膚バリア機能の改善方法。
- 皮膚バリア機能が水分透過バリア機能である請求項34記載の方法。
- ジアシルグリセロールを構成する脂肪酸中のα-リノレン酸の含有量が40質量%以上である請求項34又は35記載の方法。
