Abstract

세포 투과성 펩타이드는 PTD(protein-transduction domains), MTS(membrane-translocating sequences)라고도 불리는 데, 수송대상체에 결합된 형태 또는 혼합된 형태로 세포막을 통과해 단백질, DNA, RNA 등의 운반 대상을 세포 내뿐만 아니라 세포질, 세포내 소기관, 핵 안에까지 운반할 수 있다.
종래 세포 투과성 펩타이드들은 HIV-1과 같은 바이러스 단백질로부터 유래한 서열이거나, 초파리와 같은 다른 종이 발현하는 단백질로부터 유래한 것들이거나, 종래 세포 투과성 펩타이드를 구성하는 아미노산 서열 분석을 통해 특징적인 아미노산 서열을 선정, 인공적으로 합성해 낸 아미노산 서열이라는 점에서, 인체에 적용해서 사용할 때에 면역 반응 등의 부작용을 일으킬 소지가 있었다. 또한, 전달하고자 하는 단백질의 구조 및 기능에 영향을 미칠 수 있기 때문에, 세포 내로 전달하고자 하는 생물학적 활성 물질과 연결할 때에 효율이 저하되는 문제가 있었다.
예를 들어, 당뇨병 치료제로 알려진 리라글루타이드는 인간 GLP-1과 97% 서열 상동성을 갖는 GLP-1 수용체 작용제이다. 리라글루타이드는 1일 1회, 피하 주사 요법으로서 제2형 당뇨병을 갖고 있는 환자의 혈당 조절 개선, 그리고 비만 환자의 체중 조절용 약물로서 승인되어 판매되고 있다.
이와 같은 주사제의 투여는 환자의 큰 불편을 유발하므로, 약물의 복약 순응도가 감소한다. 따라서, 당뇨 환자를 위한 치료제로서 리라글루타이드를 비침습적인 경로(경구, 비강 등)로 투여하기 위한 새로운 시도가 필요하다.
이에, 본 발명자는 리라글루타이드와 같은 GLP-1 수용체 작용제를 비침습적인 경로로 투여할 수 있는 제제를 개발하기 위해 예의 연구를 거듭한 결과, 일련의 서열을 나타내는 TCTP 단백질 (translationally controlled tumor protein) 유래의 PTC (protein transduction domain) 변이체들이 GLP-1 수용체 작용제를 효과적으로 세포 내로 침투시킬 수 있다는 것을 확인하고 본 발명을 완성하게 되었다.

고체상 펩타이드 합성(SPPS) 방법은 작은 다공성의 비드(beads)에 링커(linkers)라 불리는 기능성 유닛(functional units)을 부착하여 펩타이드 사슬을 이어 나갈 수 있도록 유도함으로써 합성을 개시할 수 있다. 액체상 방법과 달리 펩타이드는 비드와 공유 결합하여 TFA(trifluoroacetic acid)와 같은 특정 반응물에 의해 절단되기 전까지 여과(filtration) 과정에 의해 떨어져 나가는 것을 방지한다. 고체상에 부착된 펩타이드의 N-말단 아민과 N-보호 아미노산 유닛(N-protected amino acid unit)이 결합하는 보호(protection) 과정, 탈보호(deprotection) 과정, 다시 드러난 아민 그룹(amine group)과 새로운 아미노산이 결합하는 커플링(coupling) 과정의 사이클(cycle, deprotection-wash-coupling-wash)이 반복되면서 합성이 이루어지게 된다. 상기 SPPS 방법은 마이크로파(microwave) 기술을 함께 이용하여 수행할 수 있으며, 마이크로파 기술은 펩타이드 합성 과정에서 열을 가해줌으로써 각 사이클의 커플링과 탈보호에 요구되는 시간을 단축시킬 수 있다. 상기 열 에너지는 확장되는 펩타이드 사슬이 접히거나(folding) 집합체를 형성하는 것(aggregation)을 방지하고 화학적 결합을 촉진시킬 수 있다.
또한 액체상 펩타이드 합성법에 의해 본 발명의 펩타이드를 제작할 수 있으며, 이의 구체적 방법은 하기의 문헌들을 참조로 한다: US 등록특허 제 5,516,891. 또한 본 발명의 펩타이드는 상기 고체상 합성법과 액체상 합성법을 혼합하는 방법 등의 다양한 방법으로 합성 가능하며, 본 명세서에 기술된 수단에 그 제조 방법이 제한되지 않는다.
단백질 합성은 수동 기법을 이용해서 또는 자동화에 의해 수행될 수 있다. 자동화된 합성은, 예를 들어 Applied Biosystems 431A 펩티드 합성기(Perkin Elmer)를 이용해서 달성될 수 있다. 대안적으로, 다양한 단편이 별도로 화학적으로 합성되고 화학적 방법을 이용하여 조합되어 목적 분자를 제조할 수 있다.
본 발명에서 상기 펩타이드는 당업자에게 알려진 통상의 펩타이드 합성 방법 혹은 제조 방법을 통하여 D-form이나 L-form, 서열 중 일부만 D-form이나 L-form으로 구성된 펩타이드, 또는 이들의 라세미체 형태로 모두 제작하여 사용될 수 있다. 또한 펩타이드의 안정성을 높이기 위해 그 외의 당업계에 공지된 통상적인 변형이 가능하다.

'보존적 치환'이란, 어느 아미노산이 유사한 특성을 가지는 다른 아미노산으로 치환되어 당업자라면 그 펩타이드의 2차 구조 및 감수성질(hydropathic nature, 소수성 또는 친수성 성질)이 실질적으로 비변화되었다고 예측할 수 있는 치환이다. 일반적으로 하기 아미노산 군이 보존성 변화를 나타낸다: (1) ala, pro, gly, glu, asp, gln, asn, ser, thr; (2) cys, ser, tyr, thr; (3) val, ile, leu, met, ala, phe; (4) lys, arg, his; 및 (5) phe, tyr, trp, his.
변형(modification)은 본 발명의 펩타이드의 구조 내에 수행되는 것일 수 있고, 원하는(바람직한) 특징을 가지고 있는 펩타이드 변이체 또는 파생물(derivatives)을 암호화하는 기능적 분자를 수득할 수 있다. 본 발명의 상기 펩타이드와 등가(equivalent)의 또는 향상된(improved) 변이체를 제작하기 위해 펩타이드의 아미노산 서열을 변경하고자 하는 경우, 당업자는 당업계에 알려진 단백질 코돈 정보에 기초하여 하나 이상의 코돈을 변화시킬 수 있다.
또한 상기 변이체는 비보존적 변경을 포함할 수 있다. 바람직한 실시예에서, 변이체 펩타이드는 5개 아미노산 또는 그것보다 적은 아미노산의 치환, 결실 또는 부가에 의해 천연 서열과는 다를 수 있다. 변이체는 또한, 예를 들어 상기 펩타이드의 2차 구조 및 감수(hydropathic) 특성에 대해서 가지는 영향이 최소인 아미노산의 결실 또는 부가에 의해 변경될 수 있다.
본 발명에서 상기 펩타이드는 단백질의 N-말단에 시그널(또는 리더) 서열을 포함할 수 있고, 이 서열은 번역과 동시에 또는 번역 후에 그 단백질의 이송을 지시한다. 상기 펩타이드는, 또한 펩타이드의 합성, 정제 또는 동정을 용이하게 하기 위해(예를 들면 폴리His), 또는 펩타이드의 고체 지지체에 대한 결합을 증강하기 위해, 링커 서열 또는 다른 서열과 결합(컨쥬게이트)될 수 있다.
본 발명의 일양태에서 본 발명의 상기 펩타이드를 변경하기 위하여 화학선택적 결찰(chemoselective ligation)기술이 이용될 수 있으며, 예를 들어 부위 특이적인 방식 및 제어된 방식으로 중합체(polymer)를 부착하는 것에 의할 수 있다. 이러한 기술은 대표적으로 화학적 수단 또는 재조합 수단 중 하나에 의해 단백질 골격 내로 화학 선택적인 앵커(chemoselective anchor)가 결합하는 것과, 상보적인 링커를 운반하는 중합체로 후속적 변경하는 것에 의존한다. 결과적으로, 조립 공정 및 얻어진 단백질-중합체 결합체(protein-polymer conjugate)의 공유결합 구조는 제어되며, 그것에 따라 유효성 및 약물 동태학적 특성 등과 같은 약물 특성의 합리적 최적화가 가능하게 된다. 예컨대 PEG의 선택적인 부착을 허용함으로써 이들의 약동학적 특성을 개선한다.
본 발명의 상기 펩타이드에는 약학적으로 허용가능한 염의 형태가 포함이 될 수 있다. 상기 약학적으로 허용가능한 염은 예를 들어, 염산염, 황산염, 인산염, 초산염, 구연산염, 주석산염, 숙신산염, 젖산염, 말레산염, 푸마르산염, 옥살산염, 메탄술폰산염, 또는 파라톨루엔술폰산염 등을 들 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 일양태에서 상기 펩타이드는 서열번호 1 내지 10으로 이루어진 군에서 선택될 수 있으며, 바람직하게는 서열번호 3 내지 5로 이루어진 군에서 선택될 수 있으며, 가장 바람직하게는 서열번호 3의 펩타이드일 수 있다.
본 발명의 일 실시예에 따르면, 서열번호 1 내지 10의 아미노산 서열로 이루어진 펩타이드는 종래의 TAT 단백질의 투과율과 비교하여 탁월한 세포막 투과도를 나타내며, 처리농도가 높을수록, 처리시간이 길수록 세포 내 투과율은 급격히 증가하는 양상을 나타내는 것으로 확인되었다.
본 발명에서 상기 GLP-1 수용체 작용제는 GLP-1(7-37)의 변이체인 펩티드 또는 화합물을 의미한다. GLP-1(7-37)은 서열 HAEGTFTSDV SSYLEGQAAKEFIAWLVKGRG (서열번호 12)를 가진다. 상기 "변이체"는 하나 이상의 아미노산 치환, 삭제, 부가 및/또는 삽입을 포함하는 펩티드 또는 화합물을 의미한다.
한 구체예에서 상기 GLP-1 수용체 작용제는 GLP-1(7-37)의 전체 길이에 걸쳐 GLP-1(7-37)에 대한 적어도 60%, 65%, 70%, 80% 또는 90%의 서열 동일성을 나타내는 펩티드 또는 화합물일 수 있다.
그 종류가 특별히 제한되지 않으나, 상기 GLP-1 수용체 작용제는 바람직하게는 리라글루타이드(liraglutide), 세마글루타이드(semaglutide), 엑세나타이드(exenatide) 또는 이의 조합일 수 있으며, 가장 바람직하게는 리라글루타이드 또는 이의 염일 수 있다.
본 발명에서 상기 리라글루타이드(liraglutide)은 빅토자 또는 삭센다라는 상표명으로 판매되고 있는 제2형 당뇨병을 가진 성인의 혈당 조절용 또는 성인 체중 조절용 약물로서, 식이요법 및 운동의 보조제로 이용되는 GLP-1 수용체 작용제이다. 리라글루타이드는 34번 위치의 라이신이 아르기닌으로 치환되고 글루타모일 스페이서를 통해 26번 위치의 라이신에 팔미토일 그룹이 결합된, 자연적으로 발생한 인간 GLP-1(7-37)의 지속작용 유사체이다.
본 발명이 제공하는 상기 복합체는 리라글루타이드 등 GLP-1 수용체 작용제를 세포막을 통해 이동시키는 세포막투과 전달체(transmembrane carrier)로서의 기능을 나타낸다.
본 발명에서 상기 세포 투과성 펩타이드는 매우 작은 펩타이드이므로 혹시 발생할 수 있는 GLP-1 수용체 작용제 대한 생물학적 간섭을 최소화할 수 있다. 본 발명이 제공하는 상기 펩타이드와 GLP-1 수용체 작용제 복합체는 정맥내(intravein), 복막내(intraperitoneal), 근육내(intramuscular), 피하내(subcutaneous), 피내(intradermal), 비내(nasal), 점막내(mucosal), 흡입(inhalation) 및 경구(oral) 등의 경로로 주입함으로써 생체 내 전달될 수 있다.
본 발명에 있어서 복합체는 하나 이상의 펩타이드에 하나 이상의 GLP-1 수용체 작용제가 포함된 것을 의미한다. 본 발명에 있어서, 상기 "복합체"에서 상기 펩타이드와 GLP-1 수용체 작용제는 1 : 1, 1 : 다 (多), 다 (多) : 1, 다 (多) : 다 (多)의 비율로 포함된 것일 수 있다. 예를 들어, 상기 펩타이드와 상기 GLP-1 수용체 작용제는 1 : 1 내지 1 : 100, 또는 100 : 1 내지 1 : 1의 몰비로 포함된 것일 수 있다. 그러나 상기 몰비는 예시적인 것이며 이에 제한되지 않는다. 본 발명에서 상기 복합체는 상기 펩타이드와 GLP-1 수용체 작용제가 단순히 혼합 (mixing)된 것, 펩타이드와 GLP-1 수용체 작용제가 혼합되어 형성된 것, 또는 이들이 화학결합에 의해 연결되거나 컨쥬게이션 되어 생성된 것을 모두 포함한다. 또한 복합체는 물리적 결합, 화학적 결합, 공유 결합, 비공유 결합으로 연결되거나, 매개체를 이용하여 통합된 또는 융합된 형태로 연결될 수 있다. 예를 들어, GLP-1 수용체 작용제가 단백질인 경우, 해당 단백질 서열에 상기 세포 투과성 펩타이드의 코딩 서열을 추가하여 여러 링커와 연결한 후 융합 단백질 (fusion protein)의 형태로 세포를 투과하거나 조직, 혈액으로 운반될 수 있으나 복합체를 형성하는 방법에는 제한이 없다. 또한, 상기 GLP-1 수용체 작용제는 상기 펩타이드의 N-말단 또는 C-말단에 결합될 수 있다.
본 발명의 일 양태로서, 상기 '복합체'는 상기 펩타이드와 GLP-1 수용체 작용제가 단순히 혼합 (mixture)된 상태일 수 있다.
본 발명의 다른 구현예로서, 상기 "복합체"는 상기 펩타이드와 GLP-1 수용체 작용제가 혼합 (mixture)에 의해 복합체를 이룬 것일 수 있다. 예를 들어, 상기 펩타이드와 GLP-1 수용체 작용제는 완충액에 용해되어 일정 몰비로 혼합될 수 있으며, 혼합 후 안정화를 위해 일정한 온도, 예를 들어 상온에서, 또는 4 냉장 온도에서 일정 시간, 예를 들어 1 초 내지 1 시간 동안 방치될 수 있다. 상기 온도 및 시간은 예시적인 것이며 이에 제한되지 않는다.
본 발명의 또 다른 구현예로서, 상기 "복합체"는 상기 펩타이드와 GLP-1 수용체 작용제가 화학결합 (chemical bond)에 의해 서로 연결된 것일 수 있다. 예를 들어, 상기 화학결합은 공유 결합 또는 비공유 결합이 될 수 있다. 바람직하게는 상기 펩타이드와 GLP-1 수용체 작용제는 비공유결합에 의해 결합된 것일 수 있으며, 이는 본 발명의 세포 투과성 펩타이드 및 상기 GLP-1 수용체 작용제가 동일 용매에서 혼합되어 형성되는 것일 수 있다. 비공유결합의 바람직한 예는 수소결합, 이온결합, 반데르발스 상호작용 또는 소수성 상호작용의 각각 또는 이들의 조합일 수 있다.

그러나 화학 결합의 종류 및 사용되는 링커 또는 작용기의 종류는 상술한 것에 제한되지 않는다.

본 발명의 복합체에는 펩타이드와 GLP-1 수용체 작용제 이외에 특정 세포나 특정 세포 내 소기관에 타겟팅할 수 있는 부위, pH 변화, 특정 효소의 수준, 온도 등 특정 환경에서 펩타이드의 부위가 활성화되도록 하는 부위, 특정 환경 조건에서 펩타이드와 GLP-1 수용체 작용제가 분리되도록 하는 부위, 펩타이드와 GLP-1 수용체 작용제가 복합체를 생성할 때에 둘 사이에 링커 (linker) 부위나 chemical moiety 등 다양한 특정 부위가 부가될 수 있다. 예를 들어 국소 질병 환경 (경색, 암, 염증 등), 국소 부위의 microwave, ultrasound, radiofrequency 등이 적용되기 전까지 PTD의 기능이 감춰져 있다가 특정 환경 변화에서 PTD가 기능하여 GLP-1 수용체 작용제의 운반을 촉진하는 등 세포 혹은 환경 선택적인 다기능을 가진 GLP-1 수용체 작용제의 운반이 가능할 수 있다. 생체 내에서의 안정성을 증가시키고, 약동학적 특성을 개선하여 효능을 극대화시키거나 부작용을 줄이기 위한 다양한 부위가 추가될 수 있다. 또한, PTD의 GLP-1 수용체 작용제의 세포질로의 운반 효율을 높이기 위하여 세포질로 GLP-1 수용체 작용제의 유리를 촉진하는 인자가 추가될 수 있다. 펩타이드와 GLP-1 수용체 작용제의 복합체 내에서의 위치는 제한이 없으며, 특수한 기능을 나타낼 수 있는 추가 부위나 특수한 환경에서 복합체의 작용을 조절하거나 절단할 수 있는 부위가 포함될 수 있으며, 이 부위는 상기 예시에 한정되지 아니한다.
다양한 나노운반체 (nanocarrier)의 형태로 복합체가 형성될 수 있으며, 나노운반체로서 리포좀 (liposome)이나 마이셀 (micelle) 등 막지질 기반의 나노운반체들에 특정 세포나 분자를 표적할 수 있는 리간드나 분자 이미징을 위한 기능성기의 수식을 통해 질병의 진단과 치료에 적용할 수 있다. 본 발명에서 상기 펩타이드를 리간드로 이용해 리포좀이나 마이셀을 수식하여 리포좀이나 마이셀에 싸여져 있는 약물 혹은 나노파티클의 세포 내로의 전달을 증가시킬 수 있다.
본 발명에 있어서, 상기 복합체는 GLP-1 수용체 작용제를 생체 조직 또는 혈중으로 전달시키거나 세포 투과를 촉진시키기 위해 사용될 수 있다. 상기 복합체는 생체 조직을 구성하는 세포 또는 세포 간 연접을 통하여 전달될 수 있으나 전달 방식에는 제한이 없다.
상기 생체 조직은 하나 이상의 상피조직, 근육조직, 신경조직, 결합조직을 의미하며 각 장기는 하나 이상의 조직으로 이루어질 수 있으므로, 점막, 피부, 뇌, 폐, 간, 신장, 비장, 폐장, 심장, 위장, 대장, 소화관, 방광, 요관, 요도, 난소, 정소, 생식기, 근육, 혈액, 혈관, 림프관, 림프절, 흉선, 췌장, 부신, 갑상선, 부갑상선, 후두, 편도, 기관지, 폐포의 다양한 생체 장기가 포함될 수 있으나, 상기 예시에 제한되지 않는다.
상기 생체 조직을 구성하는 세포는 상피세포, 근육세포, 신경세포, 선세포, 신경교세포, 생식세포, 줄기세포, 중간엽세포, 중간엽줄기세포, 골아세포, 골세포, 조골세포, 파골세포, 혈구세포, 조혈세포, 폐세포, 간세포, 섬유아세포, 면역세포, 내피세포, 지방세포, 연골세포 등 다양한 세포가 포함될 수 있으나, 상기 예시에 제한되지 않는다.

본 발명의 일 양태로서, 상기 복합체는 경구제제, 주사제, 점막투여제제, 구강 점막 투여 제제, 비강 투여 제제, 안과 제제, 질 투여 제제, 흡입제, 외용제제, 및 경피 흡수제, 설하 투여 제제, 이식 제제, 및 좌제로 이루어진 군으로부터 선택된 어느 하나의 형태로 이용될 수 있으나, 이에 국한되지 않는다.

본 발명의 일 양태로서, 상기 복합체는 상기 GLP-1 수용체 작용제가 특정 기관 또는 특정 세포내 소기관으로 전달되도록 하는 신호 서열 (signal peptide)을 더 포함할 수 있다. 신호 서열이란 일반적으로 5 - 30 아미노산 길이의 짧은 펩타이드로서, 특정 세포내 소기관 (ER, 골지, 엔도솜) 내에 머물거나, 세포로부터 분비되거나, 세포막 내에 삽입된다. 본 발명의 복합체는 임의의 공지의 신호 서열을 포함할 수 있으며, 상기 신호 서열에 의해 GLP-1 수용체 작용제가 세포 내 특정 부위로 이동하도록 할 수 있으나, 상기 예시에 제한되지 않는다.
본 발명의 일 양태로서, 상기 복합체는 상기 GLP-1 수용체 작용제가 특정 기관 또는 특정 세포를 타겟팅하여 그 특정 기관 또는 특정 세포 내로 특이적으로 전달되도록 하는 물질 혹은 부위를 더 포함할 수 있다. 예를 들어, 상기 복합체는 특정 세포에 존재하는 수용체에 특이적으로 결합하는 항체를 더 포함할 수 있다. 예컨대 본 발명의 복합체에서 상기 펩타이드와 GLP-1 수용체 작용제의 복합체에 유방암세포의 HER-2 수용체에 특이적으로 결합하는 항체 (허셉틴)가 추가적으로 결합되어 있는 경우, 펩타이드-GLP-1 수용체 작용제의 복합체는 상기 항체를 통해 유방암 세포에 특이적으로 결합하고 그 세포 내로 전달될 수 있다. 이와 같이 본 발명에서 펩타이드와 GLP-1 수용체 작용제의 복합체가 특정 기관 또는 특정 세포 내로 전달될 수 있도록 하는 물질의 종류에는 제한이 없으며, 당업계에 알려진 임의의 타겟팅 분자가 본 발명에 적용될 수 있다.
본 발명의 일 양태로서, 세포 밖이나 내부에서만 세포 투과능이 활성화되는 세포 투과성 펩타이드 (Activable cell penetrating peptide)의 일환으로 pH-sensitive system, enzyme-sensitive system, light-sensitive system, oxygen-sensitive system 등을 활용하여 특정 pH, 효소활성, 빛, 산소 환경에 따라 PTD의 세포투과 효능이 발현되어 특이적인 전달을 가능하게 하는 부위 혹은 물질을 더 포함할 수 있으나, 상기 예시에 제한되지 않는다.
본 발명의 일 양태로서, 상기 복합체 또는 이를 포함하는 조성물은 상기 펩타이드 및 GLP-1 수용체 작용제가 세포 내의 엔도솜에서 더 쉽게 방출 (endosomal release, endosomal escape)되도록 하는 물질을 더 포함할 수 있다. 예를 들어, membrane-disruptive peptides, membrane-disruptive polymers, lysosomotropic agent 등이 가능하며, p53 단백질의 사합체화 도메인 (p53tet)을 상기 펩타이드에 연결시키거나, HA2 융합 펩타이드와 같은 PMAPs (pH-Dependent Membrane Active Peptides)를 상기 펩타이드에 연결시키는 등의 전략을 취할 수 있으나, 상기 예시에 제한되지 않는다.
본 발명의 일 양태로서, 상기 복합체 또는 이를 포함하는 조성물은 상기 펩타이드 혹은 GLP-1 수용체 작용제의 반감기를 증가시킬 수 있는 물질 혹은 부위를 더 포함할 수 있다.
본 발명의 일 양태로서, 상기 복합체는 상기 펩타이드와 GLP-1 수용체 작용제가 목적하는 세포로 전달되기 전까지 체내에서 분해되지 않도록 하는 제형화될 수 있다. 예를 들어, 상기 GLP-1 수용체 작용제가 위장 또는 소장 상피에서 흡수되도록 하는 것이 목적인 경우, 상기 펩타이드와 GLP-1 수용체 작용제가 소화관의 위장 또는 소장 상피에 도착하기까지 분해되거나 다른 임의의 세포로 흡수되지 않도록 제형화될 수 있다.

본 발명에 따른 약학적 조성물은 상기 복합체를 단독으로 함유하거나 약학적으로 허용되는 담체와 함께 적합한 형태로 제형화 될 수 있으며, 부형제 또는 희석제를 추가로 함유할 수 있다. 상기 담체로는 모든 종류의 용매, 분산매질, 수중유 또는 유중수 에멀젼, 수성 조성물, 리포좀, 마이크로비드 및 마이크로좀이 포함된다.
약학적으로 허용되는 담체로는 예컨대, 비경구 투여용 담체를 추가로 포함할 수 있다. 비경구 투여용 담체는 물, 적합한 오일, 식염수, 수성 글루코오스 및 글리콜 등을 포함할 수 있으며, 안정화제 및 보존제를 추가로 포함할 수 있다. 적합한 안정화제로는 아황산수소나트륨, 아황산나트륨 또는 아스코르브산과 같은 항산화제가 있다. 적합한 보존제로는 벤즈알코늄 클로라이드, 메틸-또는 프로필-파라벤 및 클로로부탄올이 있다. 본 발명의 약학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현택제 등을 추가로 포함할 수 있다. 그 밖의 약학적으로 허용되는 담체 및 제제는 당업계에 공지되어 있는 것을 참고로 할 수 있다.
본 발명의 조성물은 인간을 비롯한 포유동물에 어떠한 방법으로도 투여할 수 있다. 예를 들면, 비경구적으로 투여할 수 있다. 비경구적인 투여방법으로는 이에 한정되지는 않으나, 정맥내, 근육내, 동맥내, 골수내, 경막내, 심장내, 경피, 피하, 복강내, 비강내, 장관, 국소, 설하 또는 직장내 투여일 수 있다.
본 발명의 약학적 조성물은 상술한 바와 같은 투여 경로에 따라 비경구 투여용 제제로 제형화 할 수 있다.
비경구 투여용 제제의 경우에는 주사제, 크림제, 로션제, 외용연고제, 오일제, 보습제, 겔제, 에어로졸 및 비강 흡입제의 형태로 당업계에 공지된 방법으로 제형화할 수 있다. 이들 제형은 모든 제약 화학에 일반적으로 공지된 처방서에 기재되어 있다.
본 발명의 조성물의 총 유효량은 단일 투여량(single dose)으로 환자에게 투여될 수 있으며, 다중 투여량(multiple dose)으로 장기간 투여되는 분할 치료 방법(fractionated treatment protocol)에 의해 투여될 수 있다. 본 발명의 약학적 조성물은 질환의 정도에 따라 유효성분의 함량을 달리할 수 있다. 바람직하게 본 발명의 약학적 조성물의 바람직한 전체 용량은 1일당 환자 체중 1㎏ 당 약 0.01㎍ 내지 10,000mg, 가장 바람직하게는 0.1㎍ 내지 500mg일 수 있다. 그러나 상기 약학적 조성물의 용량은 제제화 방법, 투여 경로 및 치료 횟수뿐만 아니라 환자의 연령, 체중, 건강 상태, 성별, 질환의 중증도, 식이 및 배설율등 다양한 요인들을 고려하여 환자에 대한 유효 투여량이 결정되는 것이므로, 이러한 점을 고려할 때 당 분야의 통상적인 지식을 가진 자라면 본 발명의 조성물의 적절한 유효 투여량을 결정할 수 있을 것이다. 본 발명에 따른 약학적 조성물은 본 발명의 효과를 보이는 한 그 제형, 투여 경로 및 투여 방법에 특별히 제한되지 아니한다.
본 발명에서 상기 조성물은 사용목적 내지 양상에 따라 함량은 크게 제한되지 않으며, 예를 들면 조성물 총 중량을 기준으로 0.01~99 중량%, 바람직하게는 0.5~50 중량%, 더 바람직하게는 1~30 중량%일 수 있다. 또한, 본 발명에 따른 약학적 조성물은 유효성분 외에 약학적으로 허용가능한 담체, 부형제 또는 희석제와 같은 첨가제를 더 포함할 수 있다. 본 발명의 약학적 조성물은 본 발명의 방법으로 제조된 세포 투과성 펩타이드 및 GLP-1 수용체 작용제를 0.1 내지 99.9 중량%로 포함하고, 담체를 99.9% 내지 0.1 중량%로 포함할 수 있다. 또한, 상기 조성물의 세포 투과성 펩타이드 및 GLP-1 수용체 작용제는 세포 투과성 펩타이드를 0.01 내지 99.99 중량%로 포함하고, GLP-1 수용체 작용제를 99.99% 내지 0.01 중량%로 포함할 수 있다.

세포 투과성 펩타이드 및 GLP-1 수용체 작용제를 포함하는 본 발명의 복합체는 GLP-1 수용체 작용제의 세포막 투과능을 개선하여 비침습적인 방법에 따라 약물을 투여하는 것을 가능하게 하거나, 약물을 세포, 조직, 혈액 등 생체 내로 효과적으로 전달할 수 있다.
도 1a 내지 1f는 본 발명의 펩타이드들을 다양한 농도로 BEAS-2B 세포 또는 HeLa 세포에 처리하고, 15 분 후(1a, 1b), 30분 후 (1c, 1d) 및 2시간 후(1e, 1f) 세포 내 투과정도를 분석한 결과이다.
도 2a 및 2b는 본 발명의 펩타이드를 다양한 농도로 BEAS-2B 세포 또는 HeLa 세포에 처리하고, BEAS-2B 세포에서 30 분 후 형광 현미경으로 분석하였고(2a), HeLa 세포에서 2 시간 후 형광 현미경으로 분석한 결과(2b)이다.
도 3은 본 발명의 CPP 후보군과 리라글루타이드의 결합 비율을 확인하기 위해 표면 플라드몬 공명(surface plasmon resonance, SPR) 분석한 결과이다.
도 4는 인간 유래 대장암 세포인 Caco-2 세포주에서 세포 투과성 펩타이드와 리라글루타이드의 복합체를 처리한 후, 세포 투과능을 평가한 결과이다.
도 5는 세포 투과성 펩타이드와 리라글루타이드의 복합체를 HeLa 세포에 처리하고, 1 시간 30 분 후 세포 내 투과 정도를 형광 현미경으로 관찰하고 이를 정량화하여 나타낸 그래프이다.

이하, 본 발명을 하기 실시예에 의해 상세히 설명한다. 단, 하기 실시예는 본 발명을 예시하기 위한 것일 뿐, 본 발명이 이들에 의해 제한되는 것은 아니다.
실시예 1: 세포 투과성 펩타이드의 제작
세포 투과성을 나타내는 펩타이드를 제작하기 위해 하기 표 1에 나타낸 서열번호 1 내지 10의 각 펩타이드를 순도 95% 이상이 되도록 제작하였다. 이 펩타이드 합성은 아이큐어펩젠㈜에 의뢰하여 진행하였다.

실시예 2: 세포 투과성 펩타이드의 세포 내 투과 양상 확인
본 발명의 펩타이드의 세포 내 투과 양상을 형광 현미경으로 확인하였다.
이를 위하여 상기 실시예 1에서 제작한 서열번호 1 내지 10의 각 펩타이드의 아미노 말단에 로다민(Rhodamine) 형광 염료를 표지(labelling) 하였다. BEAS-2B 세포 또는 HeLa 세포에 다양한 농도의 BCP1 내지 BCP10의 펩타이드를 처리한 후 15분, 30분 및 2시간 경과 후 세포 내 펩타이드의 농도를 평가하였다. 대조군으로는 TAT 펩타이드(GRKKRRQRRR)(서열번호 11)를 사용하였다.
도 1에 나타낸 바와 같이, 본 발명의 BCP1 내지 BCP10의 펩타이드는 모두 대조군으로 사용된 TAT보다 월등히 향상된 세포 투과도를 나타낸 것으로 확인되었다
그 다음으로, 10μM 과 50μM의 농도로 펩타이드 BCP3, BCP4, BCP5를 BEAS-2B 세포 또는 HeLa 세포에 처리하였고, 대조군 TAT도 같은 농도로 처리하였다. 단, 세포 접종시 세포 배양 슬라이드에 접종하였는데, 이는 형광 현미경 관찰시 플라스틱은 형광간섭현상을 유발하기 때문이다. 세척 후 mounting medium with DAPI를 사용하여 유리 커버 글라스를 슬라이드 글라스에 밀착시켜 형광 현미경 400배로 시료를 관찰하였다.
도 2에 나타낸 바와 같이, BCP3, BCP4 및 BCP5는 모두 대조군으로 사용된 TAT보다 월등히 향상된 세포 투과도를 나타내는 것을 확인할 수 있었다.
실시예 3: 세포 투과성 펩타이드 및 리라글루타이드의 결합 비율 평가
세포 투과성 펩타이드(CPP) BCP3, BCP4 및 BCP5와 리라글루타이드의 상호결합 및 흡착 또는 해리 정도를 통하여 결합 비율을 확인하기 위해 표면 플라즈몬 공명(surface plasmon resonance, SPR) 분석을 진행하였다.
분석은 Dextran chip에 각 CPP를 고정하고 리라글루타이드를 통과시킬 때 분자상호결합에 의한 흡착 정도를 측정하였다.
그 결과, 하기 표 2 및 도 3에 나타낸 바와 같이, BCP3, BCP4 및 BCP5 모두 리라글루타이드와 높은 결합력을 나타내었으며, 이 중에서 특히 BCP3과 리라글루타이드와의 결합력이 가장 높은 것으로 확인되었다.
실시예 4: 세포 투과성 펩타이드 및 리라글루타이드 복합체의 세포막 투과능 평가
인간 유래 대장암세포인 Caco-2 세포주를 준비하고, 12-웰 플레이트에 인서트를 장착하여 각 웰 당 1 X 105개씩 분주한 다음, 세포배양액 (DMEM)으로 하여, 37℃의 가습조건이 유지되는 5% CO2 인큐베이터에서 21일간 배양하였다.
2일 마다 새로운 세포 배양액으로 교체해주고, 21~28일 후 세포를 분주한 웰에 투과능 실험을 진행하였다. HBSS에 리라글루타이드와 세포 투과성 펩타이드 BCP3, BCP4 또는 BCP5를 몰 비율에 맞게 각각 혼합하여 각 웰에 리라글루타이드의 농도가 100μM이 되게 제조하였다.
각 세포 투과성 펩타이드와 리라글루타이드의 혼합물은 인서트 위쪽 donor에 500uL 처리하였으며, 약물 처리 후 5시간 동안 1시간마다 인서트 아래쪽 acceptor에 200uL씩 얻어 HPLC로 분석했다.
그 결과, 도 4에 나타낸 바와 같이 리라글루타이드 단독군(LRG)보다 세포 투과성 펩타이드 및 리라글루타이드 혼합물의 투과도가 향상된 것으로 나타났고, 이 중에서 BCP3과 리라글루타이드를 1:1의 몰비로 혼합한 복합체의 투과능이 가장 높은것으로 나타났다.
실시예 5: 세포 투과성 펩타이드 및 리라글루타이드 복합체의 세포 내 투과 양상
자궁경부암 세포주인 HeLa 세포주를 준비하고, 세포막을 투과하여 세포 내 축척된 리라글루타이드를 검출하기 위해, 리라글루타이드의 N-말단에 형광물질인 FITC(fluorescein isothicyanate)로 라벨시켰다.
그 다음 상기 실시예 4에서 투과능이 제일 높았던 서열번호 BCP3과 리라글루타이드 복합체를 1:0.5 및 1:1의 몰비로 각각 혼합하여 혼합물을 제조하였고, 각각 HeLa 세포주에 1 시간 30 분 동안 처리하였다.
배양이 종결된 후, HeLa 세포의 세포막에 남아있는 혼합물을 제거하기 위해 PBS로 3 회 세척하였다. 세포 내 투과 양상을 형광현미경으로 확인한 후, 마이크로 플레이트 형광 판독기(SYNERGY H1 microplate reader, BIO-TEK)를 사용하여 방출(emission) 518nm, 여기(excitation) 493nm에서 측정하여 세포 내로 들어간 리라글루타이드의 지표인 FITC의 형광도를 측정하였다.
그 결과, 도 5에 나타낸 바와 같이 리라글루타이드 단독군(LRG)보다 BCP3와 리라글루타이드 복합체 처리군에서 높은 수준의 형광이 관찰되어 BCP3와 리라글루타이드 혼합체가 자궁경부암세포의 세포막을 잘 투과함을 알 수 있었다.
실시예 6: 독성평가
상기 제조한 각 복합체가 세포독성을 나타내지 않아 치료제로 이용할 수 있는지 여부를 확인하였다.
실시예 4와 같이 각 세포 투과성 펩타이드와 리라글루타이드를 몰 비율에 맞게 각각 혼합하여 혼합물을 HeLa 세포주에 24시간 동안 처리한 후, CCK-8 (cell counting kit-8)를 사용하여 세포 독성을 확인하였다.
그 결과, 도 6에 나타낸 바와 같이 각 복합체가 세포독성을 나타내지 않는 것으로 확인되었다.
세포 투과성 펩타이드 및 GLP-1 수용체 작용제를 포함하는 본 발명의 복합체는 GLP-1 수용체 작용제의 세포막 투과능을 개선하여 비침습적인 방법에 따라 약물을 투여하는 것을 가능하게 하거나, 약물을 세포, 조직, 혈액 등 생체 내로 효과적으로 전달할 수 있어 산업상 이용가능성이 매우 높다.
Claims (17)
- 하기의 아미노산 서열로 이루어진 세포 투과성 펩타이드 및 GLP-1 수용체 작용제(Glucagon like peptide-1 receptor agonist)를 포함하는 복합체:MIIFR-R1-R2-R3-R4-R5-R6상기 식에서,R1은 A, V, I, L, S, F, K 및 R로 이루어진 군에서 선택된 어느 하나의 아미노산이고,R2는 S, L, F, T 및 Y로 이루어진 군에서 선택된 어느 하나의 아미노산이며,R3는 E, L, A 및 R로 이루어진 군에서 선택된 어느 하나의 아미노산이고,R4는 Q, H, T, L, D 및 R로 이루어진 군에서 선택된 어느 하나의 아미노산이며,R5는 L, S, V, A, K 및 H로 이루어진 군에서 선택된 어느 하나의 아미노산이고,R6는 DK, EK, QK, NK, KK 및 FK로 이루어진 군에서 선택된 어느 하나의 디펩타이드이다.
- 제1항에 있어서, 상기 세포 투과성 펩타이드는 서열번호 1 내지 10으로 이루어진 군에서 선택되는 것을 특징으로 하는 복합체.
- 제1항에 있어서, 상기 GLP-1 수용체 작용제는 리라글루타이드(liraglutide), 세마글루타이드(semaglutide), 엑세나타이드(exenatide) 또는 이의 조합인 것을 특징으로 하는 복합체.
- 제1항에 있어서, 상기 GLP-1 수용체 작용제는 상기 세포 투과성 펩타이드에 공유결합 또는 비공유결합에 의해 결합된 것을 특징으로 하는 복합체.
- 제4항에 있어서, 상기 비공유결합은 수소결합, 이온결합, 반데르발스 상호작용 및 소수성 상호작용 중 어느 하나 이상인 것을 특징으로 하는 복합체.
- 제1항에 있어서, 상기 복합체는 상기 세포 투과성 펩타이드 및 상기 GLP-1 수용체 작용제가 혼합되어 형성되는 것을 특징으로 하는 복합체.
- 제1항에 있어서, 상기 복합체는 GLP-1 수용체 작용제를 생체 조직 또는 혈중으로 전달시키거나, 또는 세포 투과를 촉진시키기 위한 것을 특징으로 하는 복합체.
- 하기의 아미노산 서열로 이루어진 세포 투과성 펩타이드 및 GLP-1 수용체 작용제(Glucagon like peptide-1 receptor agonist)를 혼합하는 단계를 포함하는, 제1항에 따른 복합체 제조방법:MIIFR-R1-R2-R3-R4-R5-R6상기 식에서,R1은 A, V, I, L, S, F, K 및 R로 이루어진 군에서 선택된 어느 하나의 아미노산이고,R2는 S, L, F, T 및 Y로 이루어진 군에서 선택된 어느 하나의 아미노산이며,R3는 E, L, A 및 R로 이루어진 군에서 선택된 어느 하나의 아미노산이고,R4는 Q, H, T, L, D 및 R로 이루어진 군에서 선택된 어느 하나의 아미노산이며,R5는 L, S, V, A, K 및 H로 이루어진 군에서 선택된 어느 하나의 아미노산이고,R6는 DK, EK, QK, NK, KK 및 FK로 이루어진 군에서 선택된 어느 하나의 디펩타이드이다.
- 제1항 내지 제7항 중 어느 한 항에 따른 복합체를 유효성분으로 포함하는 당뇨병 또는 비만 예방 또는 치료용 약학적 조성물.
- 제1항 내지 제7항 중 어느 한 항에 따른 복합체를 유효성분으로 포함하는 경구 투여용 조성물.
- 제1항 내지 제7항 중 어느 한 항에 따른 복합체를 유효성분으로 포함하는 비강 투여용 조성물.
- 제1항 내지 제7항 중 어느 한 항에 따른 복합체를 유효성분으로 포함하는 경피 투여용 조성물.
- 하기의 아미노산 서열로 이루어진 세포 투과성 펩타이드를 유효성분으로 포함하는 GLP-1 수용체 작용제의 경구 투여용 제제 제조용 조성물:MIIFR-R1-R2-R3-R4-R5-R6상기 식에서,R1은 A, V, I, L, S, F, K 및 R로 이루어진 군에서 선택된 어느 하나의 아미노산이고,R2는 S, L, F, T 및 Y로 이루어진 군에서 선택된 어느 하나의 아미노산이며,R3는 E, L, A 및 R로 이루어진 군에서 선택된 어느 하나의 아미노산이고,R4는 Q, H, T, L, D 및 R로 이루어진 군에서 선택된 어느 하나의 아미노산이며,R5는 L, S, V, A, K 및 H로 이루어진 군에서 선택된 어느 하나의 아미노산이고,R6는 DK, EK, QK, NK, KK 및 FK로 이루어진 군에서 선택된 어느 하나의 디펩타이드이다.

- 당뇨병 또는 비만 치료용 제제를 제조하기 위한 제1항 내지 제7항 중 어느 한 항에 따른 복합체의 용도.
- 제1항 내지 제7항 중 어느 한 항에 따른 복합체를 유효성분으로 포함하는 조성물의 유효량을 이를 필요로 하는 개체에 투여하는 것을 포함하는 당뇨병 또는 비만을 치료하는 방법.
