Abstract
본 발명은 그래핀 필름 상에 바이러스 수용체부가 카벤 화합물을 포함하는 링커부를 통해 고정화된 그래핀 채널 부재와 이를 포함하는 그래핀 트랜지스터 및 바이오 센서에 관한 것이다.
Description
본 발명은 바이러스 수용체를 포함하는 그래핀 채널 부재 및 이를 포함하는 센서에 관한 것이다.
최근 코비드-19(COVID-19)와 같은 전염병이 대유행함에 따라 바이러스 진단 방법으로, 검사 결과의 정확도가 높은 PCR(Polymerase Chain Reaction) 방법과 진단 속도가 빠른 LFA(Lateral Flow Assay) 를 이용하고 있다. 그러나 PCR 방법을 이용한 진단은 결과의 신뢰도가 높으나 동시간에 측정할 수 있는 샘플이 50개 미만이며 진단에 필요한 측정 시간이 약 1시간 정도 소요되어서 증가하는 감염 의심자 수 대비 검사자 수를 따라가지 못하는 단점이 있다.
또한 LFA를 이용한 진단은 진단 속도는 빠르나 검사 결과의 정확도가 PCR 방법보다 낮다는 단점을 가지고 있어서, 신속하면서도 정확도가 높은 코비드-19의 진단 방법의 개발에 많은 관심이 주목되고 있다.
특히 위와 같이 바이러스 진단의 문제를 극복하기 위한 노력이 많이 있었으나, 기존의 항체를 이용한 진단 방법의 대부분은 검사 정확도가 70% 정도로 낮으며, 뉴클레오티드를 이용한 진단 방법의 경우에는 한 번에 검사 가능한 수가 적어서, 팬데믹(pandemic) 상황에서 진단 방법으로 활용하기에는 적절하지 못한 문제가 있다.
나아가 바이러스 진단을 위해서 인간의 타액(혈액 또는 분비액 등)으로부터 얻어진 검체를 보관 및 이동하기 위한 바이러스 검체 수송 배지(UTM, Universal Transport Medium)를 이용하는데, 이러한 UTM을 이용하는 경우에는 센서의 노이즈 때문에 정확한 검출이 불가능한 문제가 있다.
감염률이 매우 높은 코비드-19와 같은 바이러스는 감염된 후에 개인적인 회복력에 따라 죽음으로 이르게 하기에 이에 대한 예방책이 반드시 필요하다. 따라서 팬데믹과 같은 고위험 상황에서 검사 정확도가 높고 동시에 다수의 검사가 가능한 진단 방법에 대한 연구가 지속되고 있다.
본 발명은 위와 같은 기존의 바이러스 진단 방법의 한계를 극복하기 위해 안출된 것으로서, SARS-CoV 바이러스, MERS-CoV 바이러스, SARS-CoV-2 바이러스(COVID-19 바이러스), H1N1 인플루엔자 바이러스 등 또는 이들의 변이체(변이 바이러스) 등의 여러 종류의 바이러스를 UTM(Universal Transport Medium) 하에서도 신속하고 정확하게 진단하기 위한 범용 바이러스 수용체를 그래핀 필름 상에 고정화시킨 그래핀 채널 부재 및 이를 포함하는 그래핀 트랜지스터와 바이오센서를 제공하고자 한다.
본 발명의 일 측면은, 그래핀 필름; 및 바이러스 수용체를 포함하는 바이러스 수용체부;를 포함하고, 상기 바이러스 수용체부는 하기 화학식 1 또는 하기 화학식 2로 표시되는 카벤 화합물을 포함하는 링커부를 통해 상기 그래핀 필름 상에 고정화된, 그래핀 채널 부재를 제공한다.

또한 본 발명의 다른 측면은, 기판; 전술한 그래핀 채널 부재; 및 한 쌍의 전극;을 포함하는 그래핀 트랜지스터를 제공한다.
또한 본 발명의 또 다른 측면은, 상기 그래핀 트랜지스터를 포함하는 바이오센서를 제공한다.
본 발명의 그래핀 채널 부재는 SARS-CoV 바이러스, MERS-CoV 바이러스, SARS-CoV-2 바이러스(COVID-19 바이러스), H1N1 인플루엔자 바이러스 등 또는 이들의 변이체(변이 바이러스) 등의 여러 종류의 바이러스를 UTM(Universal Transport Medium) 시약에서도 높은 선택성, 높은 민감도, 높은 안정성, 및/또는 높은 정확도를 가지고 검출해 낼 수 있는 효과가 있다. 또한 종래 진단 방법에 비하여 매우 빠른 시간(약 1분 이내)에 바이러스 검출이 가능하여, 현장에서 실시간으로 바이러스 진단이 가능한 장점이 있다.
또한 SARS-CoV 바이러스, MERS-CoV 바이러스, SARS-CoV-2 바이러스(COVID-19 바이러스), H1N1 인플루엔자 바이러스 등 또는 이들의 변이체(변이 바이러스) 등의 바이러스를 하나의 센서로 동시에 독립적으로 검출해 낼 수 있어서, 다양한 바이러스에 대한 신속하고 정확한 진단이 가능한 효과가 있다.

이하, 본 발명을 상세히 설명한다.
1.
그래핀 채널 부재
본 발명의 일 측면은 그래핀 채널 부재를 제공한다.
본 발명의 상기 그래핀 채널 부재는 그래핀 필름 및 바이러스 수용체부를 포함한다.
상기 그래핀 필름은 상기 바이러스 수용체를 지지하기 위한 것으로서, 그래핀층이 단층 또는 다층(multi-layer)으로 적층된 형태로 구성될 수 있다. 상기 그래핀층이 다층으로 형성되는 경우 그래핀 필름 자체의 표면 저항의 감소되어 이를 이용한 그래핀 채널 부재가 적용된 트랜지스터 등과 같은 소자의 민감도가 저하될 수 있다. 따라서 상기 그래핀 필름은 단층의 그래핀층으로 이루어진 것이 보다 바람직하다. 특히, 상기 그래핀 필름의 두께는 0.1 내지 1 ㎚일 수 있고, 이는 단층의 그래핀 필름의 두께를 의미할 수 있다. 상기 그래핀 필름의 두께가 상기 범위를 만족하는 경우에는 높은 전도도 및 높은 전하 이동도의 성질을 가지게 되어, 이를 이용한 그래핀 채널 부재를 이용하여 민감도가 높은 트랜지스터 등의 소자를 구현할 수 있게 된다.
한편, 상기 그래핀 필름은 패턴화될 수 있고, 구체적으로는 미세 패턴화될 수 있다. 예를 들어, 상기 그래핀 필름은 원 또는 삼각형, 사각형, 오각형, 육각형(허니콤(honeycomb)) 등의 다각형의 형태로 다양하게 패턴화될 수 있다. 상기와 같이 그래핀 필름을 패턴화함으로써 상기 그래핀 필름의 형상을 다양화활 수 있기 때문에, 이를 이용한 그래핀 채널 부재가 다양한 형태를 갖는 소자에 디자인적인 제약 없이 적용될 수 있다.
상기 바이러스 수용체부는 바이러스에 대한 수용체를 포함할 수 있다. 상기 바이러스에 선택적으로 반응하는 수용체일 수 있고, 예컨대 바이러스가 숙주세포를 감염시킬 때 특이적으로 결합하는 숙주세포에 존재하는 수용체일 수 있으며, 따라서 상기 바이러스 수용체는 숙주세포의 세포 표면, 세포막 등에 위치하는 화합물, 단백질일 수 있다. 특히, 상기 바이러스는 SARS-CoV 바이러스, MERS-CoV 바이러스, SARS-CoV-2 바이러스(COVID-19 바이러스), H1N1 인플루엔자 바이러스 등 또는 이들의 변이체(변이 바이러스)일 수 있고, 상기 바이러스 수용체는 ACE2(Angiotensin-converting enzyme 2), DPP4(Dipeptidyl peptidase-4), NRP1(Neuropilin-1), 시알산 등 또는 이들 중에서 선택되는 적어도 하나일 수 있으나 이에 한정되는 것은 아니하고 상기 바이러스들의 특정 구조 또는 단백질과 특이적으로 결합할 수 있는 물질이라면 모두 포함할 수 있다.
상기 ACE2는 안지오텐신 전환효소 2(Angiotensin-converting enzyme 2)라는 효소로서, 안지오텐신 전환효소와 상동하는 제1형 막관통 단백질이며, SARS-CoV 바이러스, SARS-CoV-2 바이러스(COVID-19 바이러스) 등이나 이들의 변이체(변이 바이러스)가 숙주세포로 침입할 때 결합하는 수용체이다. 또한, 상기 DDP4는 디펩티딜 펩티다아제 4(Dipeptidyl peptidase-4, CD26)로 알려진 단백질로서, 제2형 막관통 단백질이며, MERS-CoV 바이러스나 이의 변이체(변이 바이러스)가 숙주세포로 침입할 때 결합하는 수용체이다. 또한, 상기 NRP1은 뉴로필린-1 단백질로서, SARS-CoV-2 바이러스(COVID-19 바이러스)나 이의 변이체(변이 바이러스)가 숙주세포로 침입할 때 결합하는 수용체이다. 또한, 상기 시알산은 숙주세포의 표면에 위치하여 H1N1 인플루엔자 바이러스가 숙주세포로 침입할 때 결합하는 수용체이고, 상기 시알산은 시일산 그 자체일 수도 있고, 시알릴락토오스(sialyllactose) 등과 같은 시알산의 유도체일 수도 있으나, 이에 한정되는 것은 아니다.
한편, 상기 바이러스 수용체는 형질전환된 대장균으로부터 제조되는 것일 수 있다. 예를 들어, 단백질에 해당되는 바이러스 수용체인 ACE2, DPP4, NRP1 등은 이들 단백질을 암호화하는 뉴클레오티드를 대장균 등과 같은 숙주세포 내로 형질도입하여 제조되는 것일 수 있다. 구체적으로, 상기와 같은 형질도입된 숙주세포를 배양하여 상기 바이러스 수용체들을 발현시킨 후에, 상기 숙주세포를 용해시키고, 상기 용해물로부터 상기 바이러스 수용체를 분리 및/또는 정제하여 얻을 수 있다.
또한, 바이러스 수용체부는 상기 바이러스 수용체가 지질 및 지지체 단백질에 의하여 나노 디스크의 형태로 구성된 것일 수 있다. 상기 나노 디스크는 상기 바이러스 수용체가 지질을 관통한 형태로 상기 지질이 상기 바이러스 수용체를 감싸는 형태일 수 있고, 지지체 단백질이 상기 바이러스 수용체를 수용한 지질의 가장자리를 감싸는 형태일 수 있다. 구체적으로, 상기 바이러스 수용체는 지지체 단백질(membrane scaffold protein, MSP) 및 지질(Lipid)을 혼합하여 만들어진 지지체 안에 안정적으로 고정될 수 있고, 상기 나노 디스크의 단면의 평균 직경은 5㎚ 내지 20㎚ 또는 7㎚ 내지 15㎚일 수 있으나, 상기 지질과 지지체 단백질의 소재를 적절히 선택하여 상기 나노 디스크의 크기와 특성을 조절할 수 있다.
상기 지질은 POPC(1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine), POPG(1-palmitoyl-2-oleoyl-sn-glycero-3-phosphoglycerol) 등이거나 이들 중에 선택되는 적어도 하나 또는 이들의 혼합물일 수 있으며, 막 환경과 유사하게 제조하기 위해 POPC 및 POPG를 적절한 비율로 조절하여 혼합할 수 있다.
상기 지지체 단백질은 수용체 및 지질을 감싸기 위한 단백질이면 제한되지 않고 이용될 수 있으며, 환경 안정성이 높고 수용체 및 지질을 감싸는 능력이 우수한 단백질이라면 특별한 제한 없이 이용될 수 있다.
상기와 같이, 상기 바이러스 수용체부가 나노 디스크의 형태로 제조되는 경우, 생체 내의 생체 환경과 유사한 원래의 수용체 구조를 모방할 수 있어서 다양한 주변 환경에서 안정성이 우수할 뿐만 아니라 민감도도 높아서 검출 대상 바이러스에 대한 선택성이 향상된다. 따라서 이를 적용한 그래핀 트랜지스터 또는 바이오센서의 성능 유지 기간이 길어질 수 있다. 예를 들어, 통상 단백질 형태만의 수용체 사용시 성능 유지 기간이 7 내지 10일 정도인 반면, 본 발명과 같은 나노 디스크 기반의 수용체의 경우에는 약 30일까지 동일 성능의 유지가 가능한 장점이 있다.
한편, 상기 바이러스 수용체부에는 ACE2, DPP4, NRP1, 시알산 등 중 어느 하나의 수용체만이 포함된 것일 수도 있고, 이들 중 둘 이상의 조합 또는 이들 모두가 포함된 것일 수도 있다.
상기 바이러스 수용체부는 상기 그래핀 필름 상에 고정된다. 상기 고정은 물리적 결합 또는 화학적 결합을 통해 이루어질 수 있고, 특히 상기 바이러스 수용체부는 링커부를 매개로 하여 화학적으로 고정될 수 있다. 즉 상기 링커부는 상기 바이러스 수용체부를 상기 그래핀 필름에 고정화시키기 위한 매개체의 역할을 할 수 있다.
이 때, 상기 링커부는 하기 화학식 1 또는 하기 화학식 2로 표시되는 것일 수 있다.

상기 A는 질소(N) 원자를 포함하는 탄소수 1 내지 20의 알킬기 또는 질소(N) 원자를 포함하는 탄소수 2 내지 30의 헤테로아릴기이다. 구체적으로 상기 A는 아자이드(azide), 프탈이미드(phthalimide), 또는 아민(amine)일 수 있다.
본 발명에 있어서, "인접한" 기는 해당 치환기가 치환된 원자와 직접 연결된 원자에 치환된 치환기, 해당 치환기와 입체구조적으로 가장 가깝게 위치한 치환기, 또는 해당 치환기가 치환된 원자에 치환된 다른 치환기를 의미할 수 있다. 예컨대, 벤젠고리에서 오쏘(ortho)위치로 치환된 2개의 치환기 및 지방족 고리에서 동일 탄소에 치환된 2개의 치환기는 서로 "인접한"기로 해석될 수 있다.

또한, 상기 알킬기, 시클로알킬기, 아릴기, 헤테로아릴기 또는 탄화수소 고리는 다시 알킬기, 시클로알킬기, 아릴기 또는 헤테로아릴기로 치환 또는 비치환될 수 있다.
상기 화학식 1 또는 화학식 2로 표시되는 카벤 화합물이 링커부로 이용되는 경우, 상기 카벤 화합물의 비공유전자쌍이 상기 그래핀 필름과 화학적 결합(공유결합)될 수 있고, 이를 위해 예컨대 상기 그래핀 필름에 상기 카벤 화합물을 처리 한 후에 100℃ 이상의 온도를 가하여 반응시킬 수 있다.
상기 수용체부의 바이러스 수용체는 상기 링커부와 아미드 결합을 통해서 화학적으로 결합시킬 수 있다. 예를 들어, EDC-NHS 반응, DMTMM 반응, 또는 글루타르알데히드(Glutaraldehyde) 반응 등 커플링제를 이용한 방법, 또는 클릭(click) 반응 등의 화학 반응을 통해 고정할 수 있다.
상기와 같이 링커부를 통해 상기 바이러스 수용체부가 상기 그래핀 필름에 고정화되면, 상기 바이러스 수용체부가 외부 환경의 변화로부터 보다 안정해지는, 외부 환경 변화에 대한 안정성이 향상되는 장점이 있다. 특히 UTM 시약에 대한 안정성이 향상된다.
한편, 상기 그래핀 필름이 패턴화된 경우, 상기 패턴화된 그래핀 필름의 표면에 상기 바이러스 수용체가 결합되어 있을 수 있다. 이 때 전술한 것과 같이 상기 바이러스 수용체부가 상기 화학식 1 또는 화학식 2로 표시되는 카벤 화합물을 링커부로 하여 상기 패턴화된 그래핀 필름에 화학적 결합으로 고정화되어 있을 수 있다.
상기와 같이 그래핀 필름 상에 상기 화학식 1 또는 화학식 2로 표시되는 카벤 화합물을 링커부로 하여 상기 바이러스 수용체부가 고정화된 본 발명의 그래핀 채널 부재는, 그래핀 층을 이용하기 때문에 게이트에 전압이 가해지지 않은 오프 상태에서도 높은 전류가 흘러 작동 전류의 온/오프 비율이 매우 낮으므로 보다 민감도가 높은 소자를 구현하는데 적용될 수 있을 뿐만 아니라, 외부 환경 변화에 대해서도 매우 높은 안정성을 나타내므로 이용편의성이나 활용성이 높은 소자를 구현할 수 있는 장점이 있다. 아울러, 상기 그래핀 필름에 바이러스 수용체부에 바이러스 수용체로 ACE2, DPP4, NRP1, 시알산을 모두 고정화시킨 경우에는 SARS-CoV 바이러스, MERS-CoV 바이러스, SARS-CoV-2 바이러스(COVID-19 바이러스), H1N1 인플루엔자 바이러스 등 또는 이들의 변이체(변이 바이러스)의 실시간 다중 검출이 가능하다는 장점도 있다.

그래핀 트랜지스터
또한 본 발명의 다른 측면은, 상기 그래핀 채널 부재를 포함하는 그래핀 트랜지스터를 제공한다.
본 발명의 그래핀 트랜지스터는, 기판; 전술한 그래핀 채널 부재; 및 한 쌍의 전극;을 포함한다.
상기 기판은 본 발명의 그래핀 트랜지스터의 구성들이 지지되는 지지대로서의 역할을 하는 것으로서, Si 기판, 유리 기판, GaN 기판, 실리카(SiO2) 기판 등의 절연성 무기물 기판, Ni, Cu, W 등의 금속 기판 또는 플라스틱 기판(예를 들어, 폴리에틸렌나프탈레이트(polyethylene naphthalate, PEN) 등) 등을 사용할 수 있으며, 절연성 기판을 사용하는 경우, 그래핀 채널 부재와의 친화력이 우수한 점에서, 실리카(SiO2) 기판, 또는 실리콘 웨이퍼인 것이 바람직하다. 또한, 상기 기판은 그래핀의 증착이 가능한 다양한 물질 중에서 선택될 수 있으며, 예를 들어 실리콘-게르마늄, 실리콘 카바이드(SiC) 등의 물질로 구성될 수 있고, 에피택셜(epitaxial) 층, 실리콘-온-절연체(silicon-on-insulator) 층, 반도체-온-절연체(semiconductor-on-insulator) 층 등을 포함할 수 있다.
상기 그래핀 채널 부재는 상기 기판 상에 형성될 수 있다.
구체적으로는 상기 기판 상에 탄화수소 가스를 탄소 공급원으로 하여 화학 기상 증착법으로 그래핀을 성장시켜 그래핀 필름을 형성하는 것일 수 있다.
상기 그래핀 필름은, 예를 들면 화학 기상 증착법을 이용하여 형성할 수 있으며, 이를 이용하면 뛰어난 결정질을 갖는 단층 내지 수층의 그래핀을 대면적으로 얻을 수 있다. 상기 화학 기상 증착법은 기판 표면에 높은 운동 에너지를 갖는 기체 또는 증기 형태의 탄소 전구체를 흡착, 분해 또는 반응시켜 탄소 원자로 분리시키고, 해당 탄소 원자들이 서로 원자간 결합을 이루게 함으로써 그래핀을 성장시키는 방법이다.
상기 화학 기상 증착법은 PECVD(Plasma Enhanced Chemical Vapor Deposition), APCVD(Atmospheric Pressure Chemical Vapor Deposition) 및 LPCVD(Low Pressure Chemical Vapor Deposition)로 이루어진 군으로부터 선택되는 적어도 어느 하나일 수 있으며, 넓은 면적에 결점을 최소화하여 증착이 가능한 점에서 상기 화학 기상 증착법은 LPCVD인 것이 바람직하다.
상기 화학 기상 증착의 구체적인 방법으로서, 예를 들면 니켈, 구리, 알루미늄, 철 등의 금속 촉매를 스퍼터링 장치 및 전자빔 증발 장치를 이용하여 산화 실리콘층을 가지는 웨이퍼 상에 증착시켜 금속 촉매층을 형성하고, 이를 CH4, C2H2 등의 탄소를 포함하는 가스와 함께 반응기에 넣고 가열하여, 금속 촉매층에 탄소가 흡수되도록 하고, 이를 냉각하여 상기 금속 촉매층으로부터 탄소를 분리시켜 결정화시킨 후, 최종적으로 상기 금속 촉매층을 제거함으로써 그래핀 필름을 형성할 수 있다.
다만, 상기 그래핀 필름을 형성하는 방법은 화학 기상 증착법에 한정되는 것은 아니며, 여러 가지 방법을 이용하여 그래핀 필름을 형성할 수 있다.
예를 들어, 여러 층으로 구성된 흑연 결정에서 기계적인 힘으로 한 층을 벗겨내어 그래핀을 만드는 물리적 박리법, 산화-환원 특성을 활용한 화학적 박리법 또는 SiC와 같이 탄소가 결정에 흡착되거나 포함되어 있는 재료를 1,500℃의 고온 상태에서 열처리하는 에피텍셜 합성법을 이용하여 그래핀 필름을 형성시킬 수 있다.
상기 한 쌍의 전극은 그래핀 채널 부재에 전압을 인가하기 위해 상기 그래핀 필름 상에서 서로 이격되어 형성되는 소스 전극과 드레인 전극일 수 있다.
이러한 소스 전극과 드레인 전극은 상기 그래핀 필름을 통하여 전기적으로 연결될 수 있고, 도전성을 가지는 물질을 포함할 수 있으며, 예를 들어 금속, 금속 합금, 전도성 금속 산화물 또는 전도성 금속 질화물 등으로 형성될 수 있다.
상기 소스 전극과 드레인 전극은 각각 독립적으로 Cu, Co, Bi, Be, Ag, Al, Au, Hf, Cr, In, Mn, Mo, Mg, Ni, Nb, Pb, Pd, Pt, Re, Rh, Sb, Ta, Te, Ti, W, V, Zr, Zn 및 이들의 조합으로 이루어진 군으로부터 선택되는 적어도 하나를 포함할 수 있으나 이에 한정되는 것은 아니며, 그래핀과의 접촉성 및 식각의 용이성 측면에서, Au, 또는 Cr/Au 합금인 것이 바람직하다.
상기 한 쌍의 전극은 당업계에 공지된 방법으로 형성할 수 있으나, 예를 들어, 포토리소그라피(photolithography), 열증착 공정(Thermal Deposition), 전자빔증착 공정(E-beam Deposition), PECVD(Plasma Enhanced Chemical Vapor Deposition), LPCVD(Low Pressure Chemical Vapor Deposition), PVD(Physical Vapor Deposition), 스퍼터링(sputtering), 원자층 증착 공정(ALD, Atomic Layer Deposition) 등의 증착 방법에 의하여 형성할 수 있다.
상기 그래핀 채널 부재는 상기 그래핀 필름 상에 바이러스 수용체를 포함하는 바이러스 수용체부가 상기 카벤 화합물을 포함하는 링커부를 매개로 결합(고정화)되어 있는 것일 수 있다. 상기 그래핀 채널 부재, 바이러스 수용체, 바이러스 수용체부, 카벤 화합물, 링커부, 그래핀 필름에 관해서는 전술한 내용이 동일하게 적용될 수 있다.
이 때 상기 바이러스 수용체인 ACE2, DPP4, NRP1, 시알산 등이 각각 별개의 그래핀 채널 부재에 고정된 것일 수도 있고, 상기 바이러스 수용체들이 모두 하나의 그래핀 채널 부재에 고정된 것일 수도 있다.
상기 바이러스 수용체부가 상기 링커부를 통해 상기 그래핀 필름 상에 결합(고정화)되는 경우, 상기 그래핀 필름 상에 결합된 링커부는 단일층의 형태로 링커층을 형성할 수 있고, 상기 링커부에 고정된 바이러스 수용체부 역시 단일층의 형태로 바이러스 수용체층을 형성할 수 있다.
상기 링커층이 단일층으로 형성되는 경우, 그래핀 본연의 우수한 전하 이동도, 투명도 및/또는 유연성을 가질 뿐만 아니라, 외부의 비특이적 전하들의 접근에 의한 노이즈 신호를 차단할 수 있는 효과가 있다.
또한, 상기 링커층은 그 두께가 0.1 내지 2㎚일 수 있다. 상기 링커층의 두께가 0.1 ㎚보다 얇은 경우 저항이 증가하는 문제점이 있고, 2㎚보다 두꺼운 경우 링커들 간의 상호 작용이 많아져서 수용체의 고정화 효율이 감소되는 문제가 있다.
또한 상기 그래핀 필름 상에 결합된 링커부가 상기 화학식 1 또는 화학식 2로 표시되는 카벤 화합물을 포함하는 경우에는 UTM 시약 하에서도 노이즈 없이 신속하고 정확하게 바이러스를 진단할 수 있다. 특히 상기 그래핀 트랜지스터는 UTM 시약 하에서도 안정하여서, 전기적 이력 현상에 의해 발생된 전압의 차이가 0.5V 이하일 수 있고, 구체적으로는 0.4V 이하일 수 있다. 그에 따라 본 발명의 그래핀 트랜지스터는 UTM 시약에서도 노이즈가 최소화되어, UTM 시약에서 곧바로 바이러스 진단이 가능한 효과가 있다.
3.
바이러스 검출용 바이오센서
본 발명의 또 다른 측면은, 전술한 그래핀 트랜지스터를 포함하는 바이오센서를 제공한다.
본 발명에 따른 바이오센서는, 전기장 효과에 의해 소스 전극 및 드레인 전극 사이의 그래핀 필름에 흐르는 전류가 변하는 반도체 특성을 이용한 것이다.
구체적으로, 그래핀 필름의 표면에 형성된 바이러스 수용체부가 각각의 바이러스 수용체들에 특이적으로 결합하는 바이러스와 반응하면, 주변의 전기장에 변화가 일어나며, 이로 인해 소스 전극과 드레인 전극 사이의 그래핀 필름에 흐르는 전류 값이 함께 변하고, 이러한 전류의 변화를 측정하는 방식으로 표적물을 검출할 수 있다.
이러한 센서는, 위와 같은 그래핀 트랜지스터를 이용함으로써 민감도, 특이성, 신속성 및/또는 휴대성이 우수하며, 특히, 그래핀 필름을 채널층으로 사용함으로써 그래핀의 높은 전하 캐리어 이동도와 전도도 특성으로 인하여 우수한 민감도와 실시간 감지 성능을 가지고, 이에 따라 SARS-CoV 바이러스, MERS-CoV 바이러스, SARS-CoV-2 바이러스(COVID-19 바이러스), H1N1 인플루엔자 바이러스 등 또는 이들의 변이체(변이 바이러스)의 검출 한계를 개선시켜서 신속하고 높은 민감도 및/또는 높은 정확성을 가지는 검출이 가능한 효과가 있다.
한편, 본 발명의 바이오센서는 ACE2, DPP4, NRP1 및 시알산을 모두 포함하는 그래핀 채널이 구비된 그래핀 트랜지스터를 이용할 수도 있고, 상기 ACE2, DPP4, NRP1 및 시알산 각각을 별개로 포함하는 4종의 그래핀 트랜지스터를 집적하고 각각의 그래핀 트랜지스터에서의 전기적 신호를 모두 수신하여 분석할 수 있도록 하나로 모듈화하여 이용할 수도 있다. 이처럼 바이러스 수용체로 ACE2, DPP4, NRP1 및 시알산을 모두 이용하여 바이오센서를 구성하는 경우, ACE2가 고정화된 부분에서만 저항 변화가 나타나는 경우에는 SARS CoV 바이러스 진단을 확인할 수 있고, ACE2 수용체가 고정화된 부분 및 뉴로필린 수용체가 고정화된 부분에서 저항 변화가 나타나는 경우에는 SARS CoV-2 바이러스 진단을 확인할 수 있으며, DDP4 수용체가 고정화된 부분에서만 저항 변화가 나타나는 경우에는 MERS CoV 바이러스 진단을 확인할 수 있으며, 시알산 함유 수용체가 고정화된 부분에서만 저항 변화가 나타나는 경우에는 H1N1 인플루엔자 바이러스 진단을 확인할 수 있다. 따라서 SARS-CoV 바이러스, MERS-CoV 바이러스, SARS-CoV-2 바이러스(COVID-19 바이러스), H1N1 인플루엔자 바이러스 등 또는 이들의 변이체(변이 바이러스)의 실시간 다중 검출이 가능하게 된다.
또한, 위와 같이 링커부를 그래핀 트랜지스터 내의 그래핀 필름에 형성시켜 이에 결합된 바이러스 수용체부를 그래핀 트랜지스터의 채널 영역에 존재시키는 경우에는 그래핀 채널 표면의 안정성이 향상될 뿐만 아니라, UTM 시약에서 직접 바이러스 검사를 수행하는 경우에도, UTM 시약에 의한 노이즈가 최소화 되어서, 신속하고 정확한 진단이 가능한 효과가 있다.
나아가 전술한 그래핀 트랜지스터는 소형화가 가능하여, 장소의 제약 없이 현장에서 실시간으로 진단이 가능하여 매우 간편하고 정확하게 바이러스 진단 및 검사가 가능하다.
이하에서, 바람직한 실시예를 들어 본 발명을 더욱 상세하게 설명한다.
그러나 이들 실시예는 본 발명을 보다 구체적으로 설명하기 위한 것으로서, 본 발명의 범위가 이에 의하여 한정되는 것은 아니다.
[실시예 1]
바이러스 수용체부의 제조
[1-1]
ACE2, DPP4 및 NRP1 나노 디스크의 제작
인간 ACE2(Angiotensin-converting enzyme 2)를 암호화하는 유전자(GenBank Accession No. NM_001371415.1), 인간 DPP4(Dipeptidyl peptidase-4)를 암호화하는 유전자(GenBank Accession No. NM_001935.4), 그리고 인간 NRP1(Neuropilin-1)을 암호화하는 유전자(GenBank Accession No. AY249243.1)을 각각 pET-21a(+)벡터(Invitrogen, 미국) 에 삽입하고, E. Coli BL21(DE3) (Thermo Fisher, 미국)에 형질전환한 다음, 상기 형질전환체를 37℃에서 50μg/mL의 앰피실린이 포함된 1L의 LB(Luria-Bertani) 배지로 OD600 값이 0.5에 도달할 때까지 배양하였다.
그런 다음, IPTG(isopropyl thiogalactoside)를 최종 농도가 1nM이 되도록 첨가하고 3시간 동안 배양하여, 상기 ACE2, DPP4 및 NRP1 각각의 과발현을 유도하였다. 상기와 같이 ACE2, DPP4 및 NRP1이 각각 과발현된 형질전환체를 4℃에서 7000g로 20분 동안 원심분리하고, 용해 완충액(lysis buffer)(20mM Tris-HCl, 0.5M NaCl, 20mM 이미다졸, pH8.0)으로 재현탁한 다음, 초음파를 처리하여(sonication)(5s on/off, 5 min) 상기 형질전환체의 세포를 파쇄하였다.
상기와 같은 세포 파쇄물을 12000rpm으로 20분간 초원심분리하여 얻은 침전물에 N-라우로일사르코신(N-lauroylsarcosine)을 처리하여 밤새 반응시켜 가용화시킨 다음, HisTrapTM 컬럼(Sigma-Aldrich, 미국)이 장착된 AKTA pure(Cytiva)를 이용하여 FPLC(Fast protein liquid chromatography)를 수행함으로써, ACE2, DPP4, NRP1을 분리 및 정제하였다. 상기와 같이 분리 및 정제된 3종의 단백질들을 대상으로 웨스턴 블럿을 수행하여, 도 2에 도시된 바와 같이 ACE2, DPP4 및 NRP1 각각이 모두 고순도로 정제되었음을 확인하였다.
상기와 같이 정제된 3종의 수용체 각각을, POPC(1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine)와 POPG(1-palmitoyl-2-oleoyl-sn-glycero-3-phosphoglycerol)를 1:1의 몰비로 클로로포름에 용해시키고 질소 가스와 진공을 이용하여 건조시킨 다음, 상기 건조물을 HEPES 완충용액으로 재현탁시켰다. 상기와 같이 얻어진 재현탁물에 MSP(Membrane scaffold protein)를 혼합하고, 크기-배제 크로마토그래피를 수행하여 나노디스크화 되지 못한 단백질들을 제거함으로써, 지지체 내에 상기 3종의 단백질들 각각이 안정적으로 고정된 나노 디스크를 제조하였다.

시알산의 준비
말단에 아민기가 도입된 아미노엑실-6'-시알릴락토오스를 ㈜진켐에 의뢰하여 합성하였다.
[실시예 2]
링커 화합물의 제조
하기 반응식 1에 따라, 4-아미노-3-니트로페놀을 Br-PEGn-Br과 반응시켜, 링커부인 카벤 화합물을 제조하였다.

특히, n이 11인 Br-PEG11-Br을 이용하여, 하기와 같은 화학식 4의 화학 구조를 갖는 카벤 화합물 1을 각각 제조하였다.

[실시예 3]
바이러스 검출용 바이오센서의 제조
[3-1]
그래핀 트랜지스터의 제조
구리 호일을 챔버 내에서 1000℃까지 가열하고, 8sccm으로 H2를 주입하여 90mTorr로 30분(20분의 프리-어닐링과 10분의 안정화) 동안 유지한 후, 20sccm으로 CH4를 추가로 주입하여 총 압력이 560mTorr인 상태로 40분 동안 유지한 다음, 이를 35℃/min로 200℃까지 냉각시키고 로(furnace)를 상온까지 냉각시켜, 상기 구리 호일 상에 단일층의 그래핀 필름(그래핀층)을 형성하였다. 상기와 같이 형성된 그래핀 필름 상에, 6000rpm의 속도로 폴리메틸메타아크릴레이트 용액(950 PMMA A4, 4% in anisole)(MicroChem Corp.)을 스핀 코팅하고, 식각제(etchant)를 이용하여 상기 폴리메틸메타아크릴레이트가 코팅된 그래핀 필름을 상기 구리 호일로부터 분리하였다. 상기와 같이 분리된 그래핀 필름을 10분 동안 탈이온 증류수에 침지하여, 상기 그래핀 필름에 남아 있는 잔여 식각제 성분들을 제거하였다. 상기와 같이 세척된 그래핀 필름을 기판인 실리콘 웨이퍼로 전사한 다음, 상기 그래핀 필름 상에 아세톤을 투하하여 상기 그래핀 필름을 코팅하고 있던 PMMA를 녹여 제거함으로써, 실리콘 웨이퍼 상에 그래핀 필름을 형성하였다. 이 때 투명성은 97.8%로 유지되었다. 상기와 같이 실리콘 웨이퍼 상에 형성된 그래핀 필름에, 포지티브 포토레지스트(AZ5214, Clariant Corp)를 스핀 코팅한 다음, UV 노광, 베이킹 및 현상 과정을 거쳐, 상기 그래핀 필름을 패턴화하였다.
상기와 같이 패턴화되어 정렬된 그래핀 필름의 양 말단에 RIE(Reactive Ion Etching)(oxygen plasma treatment) 방법을 통해 전극 패턴(폭/길이=W/L=1, L=100 ㎛ 채널 길이)을 형성한 다음, 포토레지스트 스핀 코팅, UV 노광, 베이킹 및 현상 과정을 거쳐 상기 그래핀 필름의 일부에 전극(W/L=5, L=100 ㎛ 채널 길이)이 형성된 그래핀 트랜지스터를 제조하였다.

링커부의 형성
상기 실시예 2에서 제조된 링커 화합물인 카벤 화합물 1 50mg이 용해된 10mL의 THF(tetrahydrofuran), 상기 실시예 [3-1]에서 제조한 그래핀 트랜지스터를 침지시키고, 2mM의 TBAF(Tetrabutylammonium fluoride)을 혼합한 후 상온에서 30분 동안 반응시켰다. 이어서 THF로 세척하고 용매를 제거하여, 상기 그래핀 트랜지스터의 그래핀 필름 상에 링커부를 형성하였다.

바이러스 수용체부의 연결
EDC(1-Ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride)와 NHS(N-hydroxysuccinimide)를 pH4.5의 MES(2-(N-morpholino)ethanesulfonic acid) 완충용액에 각각 1mg씩 용해시킨 뒤, 상기 혼합 용액(EDC-NHS 용액)을 상기 실시예 [3-2]에서 링커부가 고정된 그래핀 트랜지스터의 그래핀 필름 상에 처리하고 상온에서 30분 동안 반응시켰다. 그런 다음, 상기 실시예 [1-1]에서 제작한 ACE2 나노 디스크, DPP4 나노 디스크 및 NRP1 나노 디스크와 상기 실시예 [1-2]에서 준비한 시알산을 각각 처리하고 4시간 동안 반응시켜, ACE2 나노 디스크가 고정된 그래핀 트랜지스터, DPP4 나노 디스크가 고정된 그래핀 트랜지스터, NRP1 나노 디스크가 고정된 그래핀 트랜지스터 및 시알산이 고정된 그래핀 트랜지스터를 각각 제조하였다.

바이오센서의 제조
상기 실시예 [3-3]에서 제조된 4종의 그래핀 트랜지스터를 집적하고 각각의 그래핀 트랜지스터에서의 전기적 신호를 모두 수신하여 분석할 수 있도록 하나로 모듈화하여, 도 5에 도시된 바와 같은 바이러스 검출용 바이오센서를 제작하였다.
[실시예 4]
그래핀 트랜지스터 및 바이오센서의 성능 확인
[4-1]
수용체 고정화 여부 확인
각각의 수용체부는 고유의 저항값을 가지고 있고, 카벤 화합물에 결합되어 그래핀 트랜지스터에 고정화되었을 때 그래핀 트랜지스터의 저항값이 증가하게 되므로, 상기와 같이 그래핀 트랜지스터들의 전기적 저항을 측정함으로써 각각의 수용체가 상기 그래핀 트랜지스터에 고정화되었는지 여부를 확인할 수 있다.
이를 위해, 상기 실시예 [3-1]에서 제작된 그래핀 트랜지스터와 상기 실시예 [3-3]에서 수용체부를 고정화한 4종의 그래핀 트랜지스터(ACE2 나노 디스크가 고정된 그래핀 트랜지스터, DPP4 나노 디스크가 고정된 그래핀 트랜지스터, NRP1 나노 디스크가 고정된 그래핀 트랜지스터, 시알산이 고정된 그래핀 트랜지스터)에 대하여, 전압을 인가하고 그에 따른 전류의 변화를 측정하여, 수용체부의 고정화에 따른 그래핀 트랜지스터의 전기적 저항 변화를 확인하였다.
그 결과, 도 6에 도시된 바와 같이, 수용체부가 고정화된 후 그래핀 트랜지스터의 저항이 증가되었고, 각각의 수용체에 따라 4종의 그래핀 트랜지스터가 서로 다른 저항값을 나타내는 것으로 확인되었다. 상기와 같은 결과로부터 수용체부가 그래핀 필름에 안정적으로 고정되었음을 알 수 있다.

수용체부의 종류에 따른 검출 가능 표적 물질의 확인
상기 실시예 [3-3]에서 수용체부를 고정화한 4종의 그래핀 트랜지스터(ACE2 나노 디스크가 고정된 그래핀 트랜지스터, DPP4 나노 디스크가 고정된 그래핀 트랜지스터, NRP1 나노 디스크가 고정된 그래핀 트랜지스터, 시알산이 고정된 그래핀 트랜지스터)에 각각에 대하여, SARS-CoV 바이러스, MERS-CoV 바이러스, SARS-CoV-2 바이러스(COVID-19 바이러스), H1N1 인플루엔자 바이러스를 각각 처리한 후의 상기 그래핀 트랜지스터들에서의 전류 변화를 측정하였다.
그 결과, 도 7에 따르면, ACE2 나노 디스크가 고정된 그래핀 트랜지스터는 SARS-CoV 바이러스와 SARS-CoV-2 바이러스(COVID-19 바이러스)가 처리된 경우에, DPP4 나노 디스크가 고정된 그래핀 트랜지스터는 MERS-CoV 바이러스가 처리된 경우에, NRP1 나노 디스크가 고정된 그래핀 트랜지스터는 SARS-CoV-2 바이러스(COVID-19 바이러스)가 처리된 경우에, 그리고 시알산이 고정된 그래핀 트랜지스터는 H1N1 인플루엔자 바이러스가 처리된 경우에 각각 전류의 변화가 확인되어, ACE2 나노 디스크가 고정된 그래핀 트랜지스터는 SARS-CoV 바이러스와 SARS-CoV-2 바이러스(COVID-19 바이러스)를, DPP4 나노 디스크가 고정된 그래핀 트랜지스터는 MERS-CoV 바이러스를, NRP1 나노 디스크가 고정된 그래핀 트랜지스터는 SARS-CoV-2 바이러스(COVID-19 바이러스)를, 그리고 시알산이 고정된 그래핀 트랜지스터는 H1N1 인플루엔자 바이러스를 각각 선택적으로 검출할 수 있음을 확인할 수 있다.

수용체부의 종류별 표적 물질의 농도에 따른 검출 한계 확인
상기 실시예 [3-3]에서 수용체부를 고정화한 4종의 그래핀 트랜지스터(ACE2 나노 디스크가 고정된 그래핀 트랜지스터, DPP4 나노 디스크가 고정된 그래핀 트랜지스터, NRP1 나노 디스크가 고정된 그래핀 트랜지스터, 시알산이 고정된 그래핀 트랜지스터)에 각각에 대하여, 상기 실시예 [4-2]에서 확인된 검출 가능 표적 물질들을 10ng/mL 내지 100㎍/mL 범위에서 농도별로 처리하면서 상기 그래핀 트랜지스터들에서의 전류 변화를 측정하였다.
그 결과, 도 8에 도시된 바와 같이, ACE2 나노 디스크가 고정된 그래핀 트랜지스터는 SARS-CoV 바이러스나 SARS-CoV-2 바이러스(COVID-19 바이러스)가 100ng/mL의 농도로 존재하는 경우에까지, DPP4 나노 디스크가 고정된 그래핀 트랜지스터는 MERS-CoV 바이러스가 10ng/mL의 농도로 존재하는 경우에까지, NRP1 나노 디스크가 고정된 그래핀 트랜지스터는 SARS-CoV-2 바이러스(COVID-19 바이러스)가 100ng/mL의 농도로 존재하는 경우에까지, 그리고 시알산이 고정된 그래핀 트랜지스터는 H1N1 인플루엔자 바이러스가 100ng/mL의 농도로 존재하는 경우에까지, 상기 표적 물질들을 검출해 낼 수 있는 것으로 확인되었다.

수용체부의 안정성 평가
상기 실시예 [3-3]에서 수용체부를 고정화한 4종의 그래핀 트랜지스터(ACE2 나노 디스크가 고정된 그래핀 트랜지스터, DPP4 나노 디스크가 고정된 그래핀 트랜지스터, NRP1 나노 디스크가 고정된 그래핀 트랜지스터, 시알산이 고정된 그래핀 트랜지스터)에 각각에 대하여, 상기 실시예 [4-2]에서 확인된 검출 가능 표적 물질들을 100ng/mL의 농도로 2일 간격으로 10일 동안처리하면서 상기 그래핀 트랜지스터들에서의 전류 변화를 측정하였고, 1일차 측정시의 민감도(So) 대비 해당 측정일의 민감도(S)의 비율을 추적하였다. 대조군으로는, 종래 COVID-19 바이러스의 항원에 대한 항체(SARS-CoV-2(2019-nCoV) Spike Antibody, Rabbit Mab / Sino Biological)를 고정시킨 그래핀 트랜지스터를 이용하였고, 각 일차 별로 3회의 반복 실험을 통해 민감도 값을 결정하였다.
그 결과, 도 9에 도시된 바와 같이, 종래 COVID-19 바이러스의 항원에 대한 항체를 고정시킨 그래핀 트랜지스터의 경우에는 약 10일이 경과한 시점부터 민감도가 저하되기 시작한데 반해, 본 발명의 실시예 [3-3]에서 제조한 4종의 그래핀 트랜지스터들은 15일 경과한 후에도 우수한 민감도를 유지하는 것으로 확인되었다. 상기와 같은 결과로부터 본 발명의 실시예 [3-3]에서 제조한 4종의 그래핀 트랜지스터에서 수용체부가 매우 안정적으로 고정화되어 성능을 장기간 유지시킬 수 있음을 알 수 있다.
[4-5]
UTM(Universal Transport Medium)에 대한 안정성 확인
상기 실시예 [3-3]에서 수용체부를 고정화한 4종의 그래핀 트랜지스터들 중 대표적으로 ACE2 나노 디스크가 고정된 그래핀 트랜지스터에 대하여, UTM(Universal Transport Medium) 시약(UTNFS-3B-N1P)(노블바이오)를 처리하면서 상기 그래핀 트랜지스터에서의 전류 변화를 측정하였다.
상기 UTM 시약에는 버퍼, BSA(bovine serum albumin), 항생제, 젤라틴 등의 다양한 물질이 함유되어 있는데, 그럼에도 불구하고 도 10에 도시된 바와 같이, 본 발명의 상기 실시예 [3-3]에서 제작한 수용체부가 고정된 그래핀 트랜지스터들의 경우에는, UTM 시약에 포함되어 있는 다양한 물질들에 전혀 반응하지 않는 것으로 확인되었다.
상기와 같은 결과로부터, 검출 가능 표적 물질들이 UTM 시약에 포함되어 있더라도, 본 발명의 상기 실시예 [3-3]에서 제작한 수용체부가 고정된 그래핀 트랜지스터들을 이용하면 노이즈없이 표적 시료의 검출이 가능함을 알 수 있다.
[4-6]
바이오센서를 이용한 COVID-19 바이러스의 검출 가부 확인
상기 실시예 [3-4]에서 제작한 바이오센서에, 100ng/mL 농도의 SARS-CoV-2 바이러스(COVID-19 바이러스)를 각각 처리한 후의 상기 바이오센서에 존재하는 4종의 그래핀 트랜지스터들에서의 전류 변화를 측정하였다.
그 결과, 도 11에 도시된 바와 같이, 상기 실시예 [4-2]에서 확인된 것과 마찬가지로, 상기 4종의 그래핀 트랜지스터들 중 ACE2 나노 디스크가 고정된 그래핀 트랜지스터와 NRP1 나노 디스크가 고정된 그래핀 트랜지스터에서만 전류의 변화가 확인되었고, 상기와 같은 결과로부터 본 발명의 바이오센서를 이용하여 SARS-CoV-2 바이러스(COVID-19)의 진단이 가능함을 알 수 있다.
[실시예 5]
링커부가 그래핀 트랜지스터 및 바이오센서에 미치는 영향 확인
[5-1]
링커부의 종류에 따른 UTM에 대한 안정성 평가
상기 실시예 2에서 제조된 링커 화합물인 카벤 화합물 1 대신, 종래 바이오센서에서 링커부로 널리 이용되던 1,5-디아미노나프탈렌(1,5-diaminonaphthalene, DAN)을 이용하는 것을 제외하고는, 상기 실시예 3에서와 동일한 방법으로 ACE2 나노 디스크를 고정시킨 그래핀 트랜지스터를 제작하였다. 상기와 같이 제작된 ACE2 나노 디스크가 DAN을 통해 고정된 그래핀 트랜지스터에 대하여, 상기 실시예 [4-5]에서와 같이 UTM 시약(UTNFS-3B-N1P)(노블바이오)를 처리하면서 상기 그래핀 트랜지스터에서의 전류 변화를 측정하였다.
그 결과, 도 12에 도시된 바와 같이, DAN을 링커부로 이용하는 경우에는 그래핀 트랜지스터에서 전류의 변화가 나타나는 것으로 확인되었다.
상기와 같은 결과로부터, 종래 바이오 센서에서 링커부로 널리 이용되던 DAN을 적용해서는 검출 가능 표적 물질들이 UTM 시약에 포함되어 있는 경우에는 표적 물질들의 검출이 불가능하고, 카벤 화합물을 링커부로 이용하는 것이 매우 중요함을 알 수 있다.
[5-2] 카벤 화합물의 PEG 단위의 반복 수에 따른 UTM에 대한 안정성 평가
상기 실시예 2에서 제조된 링커 화합물인 카벤 화합물 1 대신, 하기 화학식 5의 화학 구조를 갖는 카벤 화합물 2 또는 화학식 6의 화학 구조를 갖는 카벤 화합물 3을 이용하는 것을 제외하고는, 상기 실시예 3에서와 동일한 방법으로 ACE2 나노 디스크를 고정시킨 그래핀 트랜지스터를 제작하였다. 이때, 하기 화학식 5의 카벤 화합물 2와 하기 화학식 6의 카벤 화합물 3은, 상기 실시예 2의 반응식 1에 따라 4-아미노-3-니트로페놀을 각각 Br-PEG3-Br 및 Br-PEG7-Br과 반응시켜 제조하였다.

상기와 같이 제작된 ACE2 나노 디스크가 카벤 화합물 2 또는 카벤 화합물 3을 통해 고정된 그래핀 트랜지스터, 그리고 상기 실시예 [3-3]에서 제작한 ACE2 나노 디스크가 카벤 화합물 1을 통해 고정된 그래핀 트랜지스터 각각에 대하여, 상기 실시예 [4-5]에서와 같이 UTM 시약(UTNFS-3B-N1P)(노블바이오)를 처리하면서 상기 그래핀 트랜지스터에서 UTM에 의하여 그래핀 채널 상의 캐리어 전하의 밀도 변화(전기적 이력 현상의 변화)를 측정하였다.
그 결과, 도 13에 따르면, 카벤 화합물 2를 이용하여 ACE2 나노 디스크를 고정한 그래핀 트랜지스터의 경우 전기적 이력 현상에 의해 약 1.3V의 전압 차이가, 카벤 화합물 3을 이용하여 ACE2 나노 디스크를 고정한 그래핀 트랜지스터의 경우 전기적 이력 현상에 의해 약 0.7V의 전압 차이가, 그리고 본 발명의 실시예 [3-1]에서 제작한 카벤 화합물 1을 이용하여 ACE2 나노 디스크를 고정한 그래핀 트랜지스터의 경우 전기적 이력 현상에 의해 약 0.4V의 전압 차이가 발생한 것으로 확인되었다.
상기와 같은 결과로부터, 카벤 화합물의 PEG 단위의 반복 수가 늘어날수록 수용체부가 고정화된 그래핀 트랜지스터에서 전기적 이력 현상에 의한 전압의 변화가 점점 줄어듦을 알 수 있는바, 결국 카벤 화합물의 PEG 단위의 반복 수가 커질수록 UTM 시약에 의한 노이즈를 최소화할 수 있음을 알 수 있다.

임상 시료를 이용한 COVID-19 바이러스 및 이의 변이체의 검출 가부 확인

임상 시료를 이용한 COVID-19 바이러스의 검출 가부 확인
경북대학교병원 진단검사의학과에서 입수한 SARS-CoV-2 바이러스(COVID-19 바이러스) 감염 환자 42명의 임상 샘플을 UTM 시료에 용해시킨 후, 상기 실시예 [3-3]에서 제작한 ACE2 나노 디스크가 고정된 그래핀 트랜지스터에 각각 처리하여 상기 그래핀 트랜지스터에서의 전류 변화를 측정하였다.
그 결과, 도 14a 내지 도 14g에 도시된 바와 같이, 42개의 임상 샘플 모두에서 전류의 변화가 확인되었고, 상기와 같은 결과로부터 본 발명의 바이오센서를 이용하여 임상적으로도 SARS-CoV-2 바이러스(COVID-19)의 진단이 가능함을 알 수 있다.

COVID-19 바이러스 변이체의 검출 가부 확인
경북대학교병원 진단검사의학과에서 입수한 SARS-CoV-2 바이러스 변이체(오미크론 변이체)를 101 pfu/mL 내지 103 pfu/mL의 농도로, 상기 실시예 [3-3]에서 제작한 ACE2 나노 디스크가 고정된 그래핀 트랜지스터에 각각 처리하여 상기 그래핀 트랜지스터에서의 전류 변화를 측정하였다.
그 결과, 도 15에 도시된 바와 같이, 상기 SARS-CoV-2 바이러스 변이체에 대해서도 농도의존적으로 전류의 변화가 발생되는 것으로 확인되었고, 상기와 같은 결과로부터 본 발명의 바이오센서를 이용하여 SARS-CoV-2 바이러스 변이체(오미크론 변이체)의 진단도 가능함을 알 수 있다.
상기에서는 본 발명의 바람직한 실시예를 예시적으로 설명하였으나, 본 발명의 범위는 상기와 같은 특정 실시예에만 한정되지 아니하며, 해당 분야에서 통상의 지식을 가진 자라면 본 발명의 청구범위에 기재된 범주 내에서 적절하게 변경이 가능할 것이다.
Claims (14)
- 그래핀 필름; 및 바이러스 수용체를 포함하는 바이러스 수용체부;를 포함하고,상기 바이러스 수용체부는 하기 화학식 1 또는 하기 화학식 2로 표시되는 카벤 화합물을 포함하는 링커부를 통해 상기 그래핀 필름 상에 고정화된, 그래핀 채널 부재.[화학식 1][화학식 2]상기 화학식 1 및 2에 있어서,R1, R2, R5 및 R6은 각각 독립적으로 수소; 알킬기, 시클로알킬기, 아릴기 및 헤테로아릴기 중에서 선택되는 적어도 하나로 치환 또는 비치환된 탄소수 1 내지 20의 알킬기; 알킬기, 시클로알킬기, 아릴기 및 헤테로아릴기 중에서 선택되는 적어도 하나로 치환 또는 비치환된 탄소수 3 내지 20의 시클로알킬기; 알킬기, 시클로알킬기, 아릴기 및 헤테로아릴기 중에서 선택되는 적어도 하나로 치환 또는 비치환된 탄소수 6 내지 30의 아릴기; 또는 알킬기, 시클로알킬기, 아릴기 및 헤테로아릴기 중에서 선택되는 적어도 하나로 치환 또는 비치환된 탄소수 2 내지 30의 헤테로아릴기이거나,R3, R4, R7, R8, R9 및 R10은 각각 독립적으로 수소, 탄소수 1 내지 20의 알킬기, 탄소수 3 내지 20의 시클로알킬기, 탄소수 6 내지 30의 아릴기, 탄소수 2 내지 30의 헤테로아릴기 또는 하기 화학식 3으로 표시되는 구조이거나, R7 내지 R10 중 서로 인접한 두 개 이상의 치환기가 결합하여 탄화수소 고리를 형성하고,R3 및 R4 중 적어도 하나는 하기 화학식 3으로 표시되는 구조이고,R7 내지 R10 중 적어도 하나는 하기 화학식 3으로 표시되는 구조이거나, R7 내지 R10 중 서로 인접한 두 개 이상의 치환기가 결합하여 탄화수소 고리를 형성하는 경우 상기 탄화수소 고리를 형성하는 탄소에 결합된 수소 중 적어도 하나는 하기 화학식 3으로 표시되는 구조로 치환되고,[화학식 3]상기 화학식 3에 있어서,n은 괄호 내 단위의 반복 수로 6 내지 30의 정수이고,A는 질소(N) 원자를 포함하는 작용기이다.
- 청구항 1에 있어서,상기 바이러스 수용체부는, 상기 바이러스 수용체, 지질 및 지지체 단백질을 포함하는 나노 디스크 형태인 것인, 그래핀 채널 부재.
- 청구항 1에 있어서,상기 바이러스 수용체는 ACE2(Angiotensin-converting enzyme 2), DPP4(Dipeptidyl peptidase-4), NRP1(Neuropilin-1) 및 시알산으로 구성되는 군에서 선택되는 적어도 하나를 포함하는 것인, 그래핀 채널 부재.
- 청구항 3에 있어서,상기 ACE2는 SARS-CoV 바이러스, SARS-CoV-2 바이러스(COVID-19 바이러스) 또는 이들의 변이체에 선택적으로 반응하고,상기 DPP4는 MERS-CoV 바이러스 또는 이의 변이체에 선택적으로 반응하고,상기 NRP1 은 SARS-CoV-2 바이러스 또는 이의 변이체에 선택적으로 반응하고,상기 시알산은 H1N1 인플루엔자 바이러스 또는 이의 변이체에 선택적으로 반응하는 것인, 그래핀 채널 부재.
- 청구항 1에 있어서,상기 화학식 3의 n은 10 내지 30의 정수인, 그래핀 채널 부재.
- 청구항 1에 있어서,상기 A의 질소 원자를 포함하는 작용기는 아자이드(azide), 프탈이미드(phthalimide) 및 아민(amine) 중에서 선택되는 적어도 하나인, 그래핀 채널 부재.
- 청구항 1에 있어서,상기 R1 및 R2 중 적어도 하나 및 상기 R5 및 R6 중 적어도 하나는 각각 독립적으로 이소프로필기 또는 벤질기인, 그래핀 채널 부재.
- 청구항 1에 있어서,상기 카벤 화합물은 상기 그래핀 필름에 공유 결합으로 결합된, 그래핀 채널 부재.
- 청구항 1에 있어서,상기 그래핀 필름은 단층 또는 이층(bi-layer)인, 그래핀 채널 부재.
- 청구항 1에 있어서,상기 그래핀 필름은 패턴화된, 그래핀 채널 부재.
- 청구항 1에 있어서,상기 그래핀 필름은 두께가 0.1 내지 1 ㎚인, 그래핀 채널 부재.
- 기판;청구항 1 내지 청구항 11 중 어느 한 항의 그래핀 채널 부재; 및한 쌍의 전극;을 포함하는 그래핀 트랜지스터.
- 청구항 12에 있어서,상기 그래핀 트랜지스터는 UTM(Universal Transport Medium) 하에서 전기적 이력 현상에 의해 발생한 전압 차이가 0.5 V 이하인, 그래핀 트랜지스터.
- 청구항 12의 그래핀 트랜지스터를 포함하는 바이오센서.
