Abstract
Description
(前処理工程)
ステンレス製寸胴鍋に、サトウキビの搾りかすであるバガス3.2kg(含水率50質量%)及び90℃の0.5%(w/w)水酸化ナトリウム水溶液20Lを添加し、2時間混合することによって、前処理を行った。前処理後の混合液を不溶分と液分に分離して、液分を約20L得た。これを2回繰り返し、40Lの液分(前処理液)を得た。
上記の前処理液全量に対して、35%(w/w)塩酸を475mL添加し、pHを3.0に調整した。これを酸性前処理液とした。酸性前処理液に、ろ過助剤として珪藻土を395g(前処理液全量基準で1%(w/w)になるように)添加し、フィルタープレスでろ過することにより、不溶性成分を除去し、ろ液を38kg得た。
ろ過工程により得られたろ液を、芳香族系合成吸着剤(ダイヤイオンSP-850、三菱ケミカル株式会社製)383mLを充填したカラム(カラム容量:1L)に、流速7.6L/h(SV=20)の条件で通液した。その後、合成吸着剤の体積の10倍量の水で洗浄し、60%(v/v)エタノール水溶液をSV=2で766g通液して溶出し、溶出画分を得た。溶出画分を、48%(w/w)水酸化ナトリウム水溶液を用いてpH6.7に調整し、ロータリーエバポレーターにより10倍の濃度に濃縮したのち、一晩凍結乾燥して、ポリフェノール組成物として、茶褐色の粉末を23g得た。
表1に示した処方に従い各成分を混合し、クリームベースを調製した。表1中、「%」は「w/w%」を意味する。調製したクリームベース100重量部に対してバガス由来ポリフェノール組成物1重量部を添加及び混合したものを試験クリームとした。また、クリームベースそのものを対照クリームとした。
なお、使用した成分は以下のとおりである。
Finsolv Tn(Innospec Performance Chemicals Europe Limited製):安息香酸アルキル
Pemulen(登録商標) TR-1(Lubrizol製):ゲル化・増粘剤/ポリマー、高分子乳化剤
Silsense(登録商標) DW-18(Lubrizol製):シリコーン、乳化剤
Novemer(登録商標) EC-1(Lubrizol製):アクリル酸/アクリルアミド共重合体、ミネラルオイル、POEソルビタントリオレエート
Germaben(登録商標) II-E(Ashland製):静菌剤
健康な18~60歳の被験者112名(平均年齢:43.36±10.24歳)に対して、二重盲検無作為化比較試験を実施した。被験者は、サトウキビ抽出物,ポリフェノール及びフェルラ酸に対するアレルギー歴がなく、皮膚疾患がなく、レーザー,ボトックス又は顔面への化学薬品注入を2週間以内に実施しておらず、免疫抑制剤又はステロイド剤を2週間以内に服用しておらず、免疫不全疾患がなく、かつ事前にインフォームドコンセントを得たものである。また、被験者数(112名)は、非特許文献(J.Cosmet.Dermatol.,2020年,19巻,671-676頁)を参考にして、有意水準95%及び検定力90%とし、被験者20~25%が脱落すると仮定して、決定した。
データの統計解析は、SPSSバージョン17.0(SPSS Co.,Ltd.,Bangkok,Thailand)を用いて行った。対応のあるt検定(p<0.05)を用いて、試験群(試験クリームを塗布した側)と対照群(対照クリームを塗布した側)とを比較した。
図1は、滑らかさ(SEsm)の評価結果を示すグラフである。SEsmの値が小さいほど、皮膚が滑らかであることを意味する。各クリームの塗布を開始する前(Before)、滑らかさ(SEsm)に有意差はなかったが、塗布開始から4週間後(After 4weeks)の時点では、試験クリームを塗布した顔の皮膚のSEsmの値(148.85±78.46)は、対照クリームを塗布した顔の皮膚のSEsmの値(159.32±86.88)よりも有意に低かった(p=0.014)。すなわち、試験クリームを塗布した顔の皮膚は、対照クリームを塗布した顔の皮膚よりも滑らかになった。
Claims (11)
- バガス由来ポリフェノール組成物を有効成分として含有する、皮膚状態改善剤。
- 前記バガス由来ポリフェノール組成物は、
水酸化ナトリウム水溶液、水酸化カリウム水溶液、及びアンモニア水溶液からなる群より選ばれる少なくとも1種のアルカリ溶液を用いてバガスを前処理し、前処理液を得る工程と、
前記前処理液のpHを塩酸で酸性に調整してからろ過し、ろ液を回収する工程と、
前記ろ液を、芳香族系合成吸着剤が充填されたカラムに通液し、前記芳香族系合成吸着剤に吸着した成分をエタノール及び水の混合溶媒で溶出して溶出画分をポリフェノール組成物として得る工程と、
を備える方法により得られるものである、請求項1に記載の皮膚状態改善剤。 - 前記アルカリ溶液の温度は60~100℃である、請求項2に記載の皮膚状態改善剤。
- 前記アルカリ溶液は水酸化ナトリウム水溶液である、請求項2又は3に記載の皮膚状態改善剤。
- 前記水酸化ナトリウム水溶液の濃度は、0.1~1.0質量%である、請求項4に記載の皮膚状態改善剤。
- 前記芳香族系合成吸着剤はスチレン-ジビニルベンゼン系樹脂からなる、請求項2~5のいずれか一項に記載の皮膚状態改善剤。
- 皮膚の滑らかさ向上用、皮膚のシワ低減用、皮膚の色素沈着抑制用、又は皮膚の水分量増加用である、請求項1~6のいずれか一項に記載の皮膚状態改善剤。
- バガス由来ポリフェノール組成物を有効成分として含有する、皮膚の滑らかさ向上剤。
- バガス由来ポリフェノール組成物を有効成分として含有する、皮膚のシワ低減剤。
- バガス由来ポリフェノール組成物を有効成分として含有する、皮膚の色素沈着抑制剤。
- バガス由来ポリフェノール組成物を有効成分として含有する、皮膚の水分量増加剤。
