이에 따라, 환경의 영향을 크게 받지 않아 배양이 쉬우며 생산되는 색소의 품질이 상대적으로 균일하며 연중 계속적으로 안정적인 색소의 생산을 가능케 하는, 미세조류를 이용한 제아잔틴 및 루테인의 생산 방법에 관한 연구가 계속되어 오고 있다. 그러나, 미세조류가 생산해 내는 색소에는 제아잔틴이 함유되어 있지 않거나, 제아잔틴 또는 루테인 이외에도 다른 종류의 색소가 너무 많이 함유되어 있어 다시 제아잔틴, 루테인을 분리하고 정제해야만 하는 단점이 있어, 보다 높은 효율로 제아잔틴 또는 루테인을 생산해 낼 수 있는 특징이 있는 신규한 미세조류를 개발해야 할 필요성이 높아지고 있는 실정이다.
본 발명은 종래 메리골드와 같은 식물로부터 추출하여 생산해 왔던 제아잔틴(zeaxanthin) 및 루테인(lutein) 색소를 다량 생산할 수 있는 신규한 미세조류와 이의 제아잔틴 및 루테인 생산 용도를 제공하는 것을 목적으로 한다.
또한, 본 발명은 상기와 같은 신규한 미세조류를 이용하여 제아잔틴 또는 루테인을 생산하는 방법을 제공하는 것을 목적으로 한다.
상기의 목적을 달성하기 위하여, 본 발명의 일 측면은 수탁번호 KCTC 14308BP로 기탁된 클로렐라 속 HS-LZ(Chlorella sp. HS-LZ) 미세조류를 제공한다.
본 발명의 다른 측면은 수탁번호 KCTC 14308BP로 기탁된 클로렐라 속 HS-LZ(Chlorella sp. HS-LZ) 미세조류를 배양하는 단계; 및 상기 미세조류의 배양액으로부터 색소를 얻는 단계를 포함하는, 제아잔틴 또는 루테인의 생산방법을 제공한다.
본 발명의 다른 측면은 수탁번호 KCTC 14308BP로 기탁된 클로렐라 속 HS-LZ(Chlorella sp. HS-LZ) 미세조류를 포함하는, 제아잔틴 또는 루테인 생산용 조성물을 제공한다.

본 발명의 신규한 클로렐라 속 HS-LZ 미세조류는, 일반적인 클로렐라들과는 달리 해수, 담수 및 기수에서 모두 성장이 가능하며 높은 성장속도를 나타내는바, 바이오매스 생산성이 높고 색소나 지방의 생산량이 우수한 특징이 있다. 상기 HS-LZ 미세조류는 종래 클로렐라 속 HS2 미세조류와는 달리 제아잔틴을 생산할 수 있는 특징이 있으며, HS-LZ 미세조류가 생산하는 색소는 제아잔틴 및 루테인이 대부분을 차지하고 있어 엽록소 a를 비롯한 다른 종류의 색소는 거의 함유되어 있지 않아, 제아잔틴 및 루테인을 분리, 정제하기 위한 과정이 대폭 간소화될 수 있다는 장점이 있다.
종래 제아잔틴 또는 루테인을 생산하기 위해 이용하던 방법인 메리골드 꽃잎으로부터 색소를 추출하는 방법과 비교할 때, 본 발명의 미세조류를 이용한 제아잔틴 또는 루테인의 생산방법은 별도의 노동력이 필요한 수확과정이 생략되며 상기 미세조류가 단세포 생물이므로 이에 의한 산물이 균질한 특징이 있어 안정적인 색소 생산이 가능하다는 장점이 있다.
나아가, 본 발명의 HS-LZ 미세조류는 일정 조건 하에서는 제아잔틴 및 루테인의 생산성이 현저하게 높아져 높은 효율로 안정적인 색소 생산이 가능하다는 장점이 있다.
다만, 본 발명의 효과는 상기에서 언급한 효과로 제한되지 아니하며, 언급되지 않은 또 다른 효과들은 하기의 기재로부터 당업자에게 명확히 이해될 수 있을 것이다.
도 1은 클로렐라 속 HS2 미세조류에 EMS를 처리하여 돌연변이를 유도하고 고체배지에 도말한 것으로, 미세조류 집락 별로 배지의 색상 변화를 확인한 것이다.
도 2는 EMS를 처리하여 고체배지가 녹색이 아닌 다른 색으로 변화한 클로렐라 속 HS2 미세조류 돌연변이체 집락을, 다시 액체배지에서 배양하면서 빛이 주어지지 않는 조건(dark)과 빛이 주어지는 조건(light)으로 배양한 결과이다.

이하, 본 발명을 상세히 설명한다.
1. 신규 클로렐라 속 미세조류
본 발명은 신규 클로렐라 속 미세조류를 제공한다.
본 발명의 상기 미세조류는 수탁번호 KCTC 14308BP로 기탁된 클로렐라 속 HS-LZ(Chlorella sp. HS-LZ) 미세조류 이다.
상기 클로렐라 속 HS-LZ는 클로렐라 속 HS2 미세조류(수탁번호 KCTC 13108BP)로부터 개량되어 만들어진 신규한 미세조류로, 상기 클로렐라 속 HS2 미세조류는 다양한 탄소원이 주어진 환경이나 담수, 해수 및 기수 환경에서도 높은 성장속도를 나타내고 바이오매스, 색소, 지질 생산성이 우수한 특징이 있는 것이므로, 본 발명의 클로렐라 속 HS-LZ 미세조류도 상기 클로렐라 속 HS2 미세조류의 장점과 특성을 나타낼 수 있다.
상기 클로렐라 속 HS-LZ 미세조류는 서열번호 1의 염기서열을 포함하는 폴리뉴클레오티드를 포함하는 것일 수 있으며, 구체적으로 상기 클로렐라 속 HS-LZ 미세조류의 18s rDNA가 서열번호 1의 염기서열을 포함하는 것일 수 있다. 상기 서열번호 1의 염기서열은 클로렐라 속 HS2 미세조류의 18s rDNA 염기서열과 동일한 것으로, 이에 따라 본 발명의 클로렐라 속 HS-LZ 미세조류를 클로렐라 속으로 분류되는 것으로 동정할 수 있다.
상기 클로렐라 속 HS-LZ 미세조류는 제아잔틴(zeaxanthin)을 생산할 수 있다. 종래의 클로렐라 미세조류, 예컨대 클로렐라 속 HS2 미세조류는 제아잔틴을 생산하지 못하는 것과는 달리, 본 발명의 클로렐라 속 HS-LZ 미세조류는 제아잔틴을 생산할 수 있다.
상기 클로렐라 속 HS-LZ 미세조류는 루테인(lutein)을 생산할 수 있다. 본 발명의 클로렐라 속 HS-LZ 미세조류는 종래의 클로렐라 미세조류, 예컨대 클로렐라 속 HS2 미세조류와 비교할 때 루테인의 생산성이 더 높은 것일 수 있다.
상기 제아잔틴 및 루테인은 안구 건강에 도움을 주는 활성이 있는 것으로 알려져 높은 경제적 가치를 가지는 색소로, 본 발명의 미세조류는 상기 색소들을 높은 순도로 생산할 수 있는 특징이 있는바, 제아잔틴 또는 루테인을 생산하기 위한 용도로 유용하게 이용될 수 있으며 나아가 안질환의 예방, 치료나 눈 건강의 개선을 위한 용도로 이용될 수 있다.
상기 클로렐라 속 HS-LZ 미세조류는 상기 제아잔틴 또는 루테인을 생산하여 세포 내에 축적하거나, 세포 외부로 방출할 수 있다.
상기 클로렐라 속 HS-LZ 미세조류가 생산하는 색소는, 색소 100 중량부에 대하여 제아잔틴 및 루테인 중 적어도 하나를 90 중량부 이상의 함량으로 함유하는 것일 수 있다. 구체적으로, 상기 색소는 색소 100 중량부에 대하여 제아잔틴 및/또는 루테인을 92 중량부 이상, 95 중량부 이상, 96 중량부 이상, 97 중량부 이상, 98 중량부 이상, 99 중량부 이상, 또는 99.5 중량부 이상의 함량으로 함유하는 것일 수 있으며, 제아잔틴 및 루테인 이외에 다른 색소는 함유하지 않는 것일 수 있다. 상기 클로렐라 속 HS-LZ 미세조류가 생산하는 색소는, 엽록소 a, 엽록소 b, 네오잔틴(neoxanthin) 및 비올라잔틴(violaxanthin)으로 이루어진 군으로부터 선택되는 적어도 하나를 함유하지 않는 것일 수 있다. 종래의 클로렐라 미세조류, 예컨대 클로렐라 속 HS2 미세조류는 제아잔틴을 생산하지 못하거나, 제아잔틴을 생산하더라도 제아잔틴 및 루테인 이외에 다른 색소들(상기 엽록소 a, 엽록소 b, 네오잔틴, 비올라잔틴 등)도 다량으로 생산하여 전체 색소 중 제아잔틴 및 루테인 이외의 색소가 10 중량부 초과의 함량으로 포함될 수 있다. 이와는 달리, 본 발명의 클로렐라 속 HS-LZ 미세조류가 생산하는 색소는 제아잔틴 및 루테인 중 적어도 하나를 상기 범위의 함량으로 함유하고 있는바, 제아잔틴 및/또는 루테인이 대부분을 차지하여 제아잔틴 또는 루테인을 수득하기 위해 이를 분리, 정제하는 과정이 간소화되거나 생략될 수 있다는 장점이 있다.
상기 클로렐라 속 HS-LZ 미세조류는, 상기 미세조류 배양액 건조 중량 당 제아잔틴을 1 ㎎/g 내지 5 ㎎/g 생산하는 것일 수 있으며, 구체적으로 1.2 ㎎/g 내지 4.5 ㎎/g, 1.5㎎/g 내지 4 ㎎/g, 1.9 ㎎/g 내지 3.6 ㎎/g, 1.8 ㎎/g 내지 2.2 ㎎/g, 1.9 ㎎/g 내지 2 ㎎/g, 3 ㎎/g 내지 4 ㎎/g, 3.3 ㎎/g 내지 3.8 ㎎/g 또는 3.5 ㎎/g 내지 3.7 ㎎/g의 제아잔틴을 생산하는 것일 수 있다.
상기 클로렐라 속 HS-LZ 미세조류는, 상기 미세조류 배양액 건조 중량 당 루테인을 0.2 ㎎/g 내지 3 ㎎/g 생산하는 것일 수 있으며, 구체적으로 0.5 ㎎/g 내지 2.5 ㎎/g, 0.6 ㎎/g 내지 2 ㎎/g, 0.7 ㎎/g 내지 1.5 ㎎/g, 0.5 ㎎/g 내지 1 ㎎/g, 0.6 ㎎/g 내지 0.9 ㎎/g, 0.7 ㎎/g 내지 0.9 ㎎/g, 1.2 ㎎/g 내지 1.5 ㎎/g 또는 1.3 ㎎/g 내지 1.4 ㎎/g의 루테인을 생산하는 것일 수 있다.
2. 제아잔틴 또는 루테인의 생산방법
본 발명은 제아잔틴 또는 루테인을 생산하는 방법을 제공한다.
본 발명의 상기 제아잔틴 또는 루테인의 생산방법은, 수탁번호 KCTC 14308BP로 기탁된 클로렐라 속 HS-LZ(Chlorella
sp. HS-LZ) 미세조류를 배양하는 단계; 및 상기 미세조류의 배양액으로부터 색소를 얻는 단계를 포함한다.
상기 클로렐라 속 HS-LZ 미세조류에 대한 설명은 앞서 '1. 신규 클로렐라 속 미세조류'에서 이에 대해 설명한 것과 동일하다.
본 발명의 클로렐라 속 HS-LZ 미세조류는 제아잔틴 또는 루테인을 고순도로 생산할 수 있는 특징이 있는바, 상기 클로렐라 속 HS-LZ 미세조류를 배양하는 단계를 거쳐 이로부터 색소를 추출하는 단계를 거칠 경우, 고함량의 제아잔틴 또는 루테인을 생산할 수 있다. 상기 클로렐라 속 HS-LZ 미세조류가 생산하는 색소의 성분은 제아잔틴 및 루테인이 대부분이며 엽록소 a, 엽록소 b, 네오잔틴, 비올라잔틴 등 다른 색소는 거의 함유되어 있지 않으므로, 제아잔틴 또는 루테인만을 높은 순도로 생산할 수 있는 장점이 있다.
상기 클로렐라 속 HS-LZ 미세조류를 배양하는 단계는, 빛이 주어지는 조건 또는 빛이 주어지지 않는 조건에서 배양하는 것일 수 있다. 본 발명의 클로렐라 속 HS-LZ 미세조류는 빛의 존재 유무와 무관하게 제아잔틴 또는 루테인을 생산할 수 있는바, 빛이 주어지는 조건에서 배양하는 것이 일반적인 다른 클로렐라 미세조류와는 달리, 빛이 주어지지 않는 암조건에서도 제아잔틴 또는 루테인의 생산이 가능하다는 장점이 있다.
상기 클로렐라 속 HS-LZ 미세조류를 배양하는 단계는, 통상적인 클로렐라의 배양 배지에서 배양하는 것일 수 있으며, 예컨대 당류, 아미노산, 유기산 등이 함유된 배지에서 배양하는 것일 수 있다. 구체적으로, 상기 배지는 글루코오스, 덱스트로오스, 수크로오스, 과당, 말토오스, 락토오스, 당밀(molasses), 글루탐산, 말산, 시트르산 등을 포함하는 것일 수 있다. 상기 클로렐라 속 HS-LZ 미세조류는 질소원 및 무기염을 더 포함하는 배지에서 배양될 수 있다. 상기 질소원은 질산염, 아질산염, 요소, 암모늄 클로라이드, 암모니아, 펩톤, 트립톤, 효모 추출물(yeast extract), 비프 추출물(beef extract), 맥아 추출물(malt extract), 감자 추출물(potato extract) 또는 NPK 비료일 수 있고 상기 무기염은 Na+, K+, Mg2+ 또는 Ca2+일 수 있으나, 이에 제한되지 않는다. 상기 배지는 통상적으로 사용될 수 있는 식물용 복합비료, 미생물 발효용 복합배지, BG11 배지 등일 수 있다. 상기 클로렐라 속 HS-LZ 미세조류를 배양하기 위한 배지에 포함되는 물은 해수, 기수 또는 담수일 수 있으며 예를 들어 담수일 수 있으나 이에 제한되는 것은 아니며, 상기 배지에 포함되는 물의 종류는 제아잔틴 또는 루테인을 생산하기 위한 공정, 클로렐라 속 HS-LZ 미세조류의 배양 조건 등을 고려하여 적절하게 선택될 수 있다.
또한, 상기 클로렐라 속 HS-LZ 미세조류를 배양하는 단계는, 상기 미세조류를 유기탄소원이 존재하지 않는 조건에서 3일 내지 14일 동안 배양하는 것을 포함하는 것일 수 있으며, 예를 들어 3일 내지 10일, 4일 내지 9일, 또는 5일 내지 8일 동안 배양하는 것일 수 있다. 상기 미세조류를 유기탄소원이 존재하지 않는 스트레스 조건에서 배양하는 경우, 상기 미세조류의 색소 생산량이 증가될 수 있으며 제아잔틴 및 루테인의 생산량이 증가될 수 있다. 상기 클로렐라 속 HS-LZ 미세조류를 배양하는 단계는 유기탄소원이 존재하는 배지에서 배양하여 세포 수를 증가시킨 후, 다시 유기탄소원이 존재하지 않는 조건에서 배양함으로써 제아잔틴 및 루테인의 생산량을 현저하게 증가시킬 수 있다.
상기 색소를 얻는 단계는, 상기 배양액 내 미세조류 세포를 파쇄하는 단계를 포함하는 것일 수 있다. 상기 미세조류 세포를 파쇄하는 방법은 세포 내 색소 성분, 구체적으로 제아잔틴 또는 루테인이 파괴되거나 활성을 잃지 않도록 세포를 파쇄하는 것이라면 어떤 방법이든 이용될 수 있으며, 예컨대 세포에 초음파를 처리(sonication)하여 파쇄하는 것일 수 있다.
상기 색소를 얻는 단계는, 상기 미세조류로부터 색소를 추출하는 단계를 포함하는 것일 수 있다. 상기 색소의 추출은, 당업자에게 공지된 임의의 추출 방법을 이용할 수 있으며, 추출 방법으로는 용매 추출법(저온 암조건 추출 포함), 초음파 추출법, 여과법 및 환류 추출법 등 종래 알려진 통상적인 추출 방법을 모두 사용할 수 있으며, 구체적으로 용매 추출법이나 환류 추출법을 이용함으로써 제조할 수 있다. 상기 추출 과정은 수회 반복할 수 있으며, 이후에 농축 또는 동결건조 등의 단계를 추가적으로 거칠 수 있다. 구체적으로, 수득한 추출물을 감압 농축하여 농축액을 얻고, 상기 농축액을 동결건조시킨 후 분쇄기를 이용하여 고농도의 추출 분말을 제조할 수 있다. 추출 용매로는 헥산, 벤젠, 톨루엔, 디에틸에테르, 클로로포름, 에틸아세테이트, 메틸렌 클로라이드, 1,4-디옥산, 테트라하이드로퓨란, 아세톤, 아세토니트릴, 디메틸포름아마이드, 디메틸설폭사이드, 아세트산, 메탄올, 에탄올, 프로필렌글라이콜, 프로필알코올, 부틸렌글라이콜, 부틸알코올, 글리세린 등을 포함하는 탄소수 1 내지 4의 알코올류 등이 사용될 수 있으나 이에 한정되지 않는다.
본 발명의 제아잔틴 또는 루테인의 생산방법은, 엽록소 a, 엽록소 b, 네오잔틴(neoxanthin) 및 비올라잔틴(violaxanthin)을 제거하는 단계를 포함하지 않는 것일 수 있다. 즉, 제아잔틴 및 루테인만을 별도로 분리하는 단계를 포함하지 않는 것일 수 있다. 상기 클로렐라 속 HS-LZ 미세조류가 생산하는 색소에는 제아잔틴 및 루테인 이외의 다른 색소가 함유되어 있지 않거나 매우 미량으로 함유될 수 있는바, 엽록소 a, 엽록소 b, 네오잔틴, 비올라잔틴 등의 색소를 제거하는 과정을 거치지 않고도 제아잔틴 및 루테인을 높은 순도로 생산하여 수득할 수 있다. 다만, 상기 색소로부터 제아잔틴 및 루테인을 더 높은 순도로 수득하기 위한 경우, 필요에 따라 제아잔틴 및 루테인을 분리하는 단계를 더 거칠 수 있으며, 이 경우 고성능 액체 크로마토그래피(HPLC)를 이용할 수 있으나 이에 제한되는 것은 아니다. 또한, 필요에 따라 제아잔틴 및 루테인 중 어느 하나만을 생산하고자 하는 경우, 제아잔틴 또는 루테인을 분리하는 단계를 더 거칠 수 있다.
3. 클로렐라 속 HS-LZ 미세조류의 용도
본 발명은 클로렐라 속 HS-LZ(Chlorella

. HS-LZ) 미세조류의 제아잔틴 또는 루테인의 생산 용도를 제공한다.
본 발명의 제아잔틴 또는 루테인 생산용 조성물은, 수탁번호 KCTC 14308BP로 기탁된 클로렐라 속 HS-LZ(Chlorella sp. HS-LZ) 미세조류를 포함한다.
상기 클로렐라 속 HS-LZ 미세조류에 대한 설명은 앞서 '1. 신규 클로렐라 속 미세조류'에서 이에 대해 설명한 것과 동일하다. 상기 클로렐라 속 HS-LZ 미세조류는 제아잔틴 또는 루테인을 생산할 수 있는 미세조류이며, 생산하는 색소들 중 제아잔틴 및 루테인을 제외한 다른 색소는 거의 생산하지 않으므로, 별도의 제아잔틴 또는 루테인의 분리, 정제 과정이 간소화되거나 생략될 수 있어 고순도의 제아잔틴 또는 루테인을 간편하게 생산할 수 있다. 또한, 클로렐라 속 HS-LZ 미세조류로부터 생산되는 루테인은, 메리골드에 의해 생산되는 루테인과는 달리 루테인 분자의 양쪽 말단에 지방산이 연결되어 있지 않으므로 이를 제거하는 과정이 수반되지 않는다는 장점이 있다. 이러한 클로렐라 속 HS-LZ 미세조류의 특징을 고려할 때, 상기 미세조류를 포함하는 조성물은 제아잔틴 또는 루테인을 생산하기 위해 유용하게 이용될 수 있다.

본 발명의 안질환의 예방 또는 치료용 약학 조성물, 및 안질환의 예방 또는 개선용 건강기능식품은, 수탁번호 KCTC 14308BP로 기탁된 클로렐라 속 HS-LZ(Chlorella sp. HS-LZ) 미세조류 또는 이의 추출물을 포함한다.
앞서 기재한 것처럼, 상기 클로렐라 속 HS-LZ 미세조류 또는 이의 추출물은 제아잔틴 또는 루테인을 포함하고 있으므로, 상기 조성물은 제아잔틴 또는 루테인이 나타내는 생리적 활성에 따라 다양한 용도로 이용될 수 있으며, 구체적으로 안질환을 예방, 치료 또는 개선하기 위해 이용될 수 있다.

상기 예방은 질환 또는 장애에 걸릴 위험을 감소시키는 것을 의미하며, 질환에 노출되거나 질환에 걸리기는 쉽지만 아직 질환에 걸리거나 질환의 증후를 나타내지 않는 대상에서 질병의 1종 이상의 임상적 증후가 진행되지 않도록 하여 질환의 발병을 억제하거나 지연시키는 모든 행위를 의미한다.
상기 치료는 질환 또는 장애를 경감시키는 것을 의미하며, 질환 또는 이의 1종 이상의 임상적 증후의 진행을 저지 또는 감소시켜 질환의 증세를 호전시키거나 이롭게 변경하는 모든 행위를 의미한다.
상기 개선은 질환 또는 장애의 증세가 호전되도록 하거나 이롭게 되도록 하는 모든 행위를 의미한다.
본 발명의 상기 약학 조성물은 가축, 인간 등을 포함하는 포유동물에 경구 또는 비경구로 투여, 예를 들어 정맥 및 동맥 내, 근육 내, 피하, 복강 내, 점막 또는 국소(예, 점안), 안구, 경피 등에 적용될 수 있다.
상기 약학 조성물은 경구 투여용 제형, 예를 들면 정제, 트로키제(troches), 로진지(lozenge), 수용성 또는 유성현탁액, 조제분말 또는 과립, 에멀젼, 하드 또는 소프트 캅셀, 시럽 또는 엘릭실제(elixirs) 등으로 제제화될 수 있다. 정제 및 캡슐 등의 제형으로 제제화하기 위해 락토오스, 사카로오스, 솔비톨, 만니톨, 전분, 아밀로펙틴, 셀룰로오스 또는 젤라틴과 같은 결합제; 디칼슘 포스페이트와 같은 부형제; 옥수수 전분 또는 고구마 전분과 같은 붕괴제; 스테아린산 마그네슘, 스테아린산 칼슘, 스테아릴푸마르산 나트륨 또는 폴리에틸렌글리콜 왁스와 같은 윤활유가 함유될 수 있다. 캡슐제형의 경우는 상기에서 언급한 물질 이외에도 지방유와 같은 액체 담체를 함유할 수 있다.
본 발명의 약학 조성물을 비경구 투여시에는 피하주사, 정맥주사, 근육 내 주사, 흉부 내 주사 주입 방식과 점막, 또는 국소에 적용되는데, 분산제, 좌제, 분제, 에어로졸(비강 스프레이 또는 흡입제), 점안제, 겔, 현탁액제(수성, 또는 비수성 액상 현탁액, 수중유 에멀젼 또는 유중수 에멀젼), 용액제 등 비경구 투여에 적합한 액상 투여 형태 등에 의한다. 비경구 투여용 제형으로 제제화하기 위해서는 상기 조성물을 안정제 또는 완충제와 함께 물에서 혼합하여 용액으로 제조하고 이는 앰플 또는 바이알의 단위 투여형으로 제제화될 수 있다.
본 발명의 클로렐라 속 HS-LZ 미세조류 또는 이의 추출물의 유효투입량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양화될 수 있지만, 일반적으로 성인 환자 체중 1 ㎏ 당 1 내지 20 ㎎/일이고, 구체적으로 5 내지 10 ㎎/일이며, 의사 또는 약사의 판단에 따라 일정 시간 간격으로 1일 수회, 구체적으로 하루 2 회 내지 3 회 분할 투여될 수 있다.
본 발명의 건강기능식품은 상기 클로렐라 속 HS-LZ 미세조류 또는 이의 추출물을 총 중량 중 0.01 내지 50 %(w/w), 구체적으로 1 내지 30 %(w/w) 범위로 사용할 수 있다. 예를 들면, 클로렐라 속 HS-LZ 미세조류 또는 이의 추출물을 통상적인 기호성 식품, 즉 라면, 생면 등의 면류, 두부, 시리얼, 빵류, 츄잉 껌, 사탕, 과자류 등에 첨가하여 통상적으로 알려진 방법에 의하여 각종 식품을 제조할 수 있고, 식용가능한 식품 첨가물로서 적용할 수 있고, 또한, 정제, 과립제, 환제, 경질캅셀제, 연질캅셀제 또는 액제 제형 등 일반적인 제형으로 제형화될 수 있으며, 생즙, 파우치, 음료, 또는 다류로 제조될 수 있고, 상기한 성분 이외에 다른 성분은 제형에 따라 본 기술 분야의 통상의 기술자가 적절하게 선택하여 배합할 수 있다.
이하, 본 발명을 실시예에 의하여 상세히 설명한다.
단, 하기 실시예는 본 발명을 구체적으로 예시하는 것이며, 본 발명의 내용이 하기 실시예에 의해 한정되지 아니한다.

실시예
1] 클로렐라 속 HS-
LZ

) 미세조류의 제조
본 발명의 클로렐라 속 HS-LZ(Chlorella sp. HS-LZ) 미세조류는 클로렐라 속 HS2(Chlorella sp. HS2) 미세조류를 대상으로 인위적인 돌연변이를 유발시킴으로써 제조하였으며, 이를 통해 제아잔틴(zeaxanthin) 및 루테인(lutein)의 생산성이 우수한 특징이 있는 미세조류를 제조할 수 있었다.
개량을 위한 모균주로 사용된 상기 클로렐라 속 HS2 미세조류는 담수, 해수 및 기수 환경 모두에서 성장이 가능하며 우수한 성장속도를 나타내 바이오매스, 색소 및 지질의 생산성이 다른 미세조류에 비해 우수한 특징이 있다. 상기 클로렐라 속 HS2 미세조류는 한국생명공학연구원에 수탁번호 KCTC 13108BP로 기탁된 것을 이용하였다.
상기 클로렐라 속 HS2 미세조류를 암(dark) 조건에서 배양한 후 3일 동안 연속으로 계대배양하여 최대한 세포 연령이 낮은 세포들을 수득하였다. 10 ㎖의 배양 배지에 107개의 HS2 미세조류 세포를 접종하고, 여기에 1%(w/v)의 EMS(ethyl methanesulfonate, Sigma-Aldrich, Cat. No. M0880)를 40 mM의 농도로 처리한 후 2시간 동안 배양하여 돌연변이를 유도하였다. 그 후, BD DifcoTM Yeast mold Agar 고체배지(271210, BD Difco, 미국) 각 1개당 103개의 세포를 도말하고 총 50개의 고체배지에서 색 변화가 발생한 콜로니를 선별하여 다시 스트리킹 함으로써, 후보 변이체 미세조류 집락들을 선별하였다(도 1). 기존 HS2 미세조류 배지의 경우 짙은 녹색을 나타내는데, 생산하는 색소의 조성이 변화되어 배지의 색상도 함께 옅은 녹색 내지 황색으로 변화된 배지를 선별한 것이다.
고체배지에서의 색상 변화를 통해 선별한 변이체 미세조류들을 다시 탄소원으로 포도당을 첨가한 BD DifcoTM Yeast mold 액체배지(271120, BD Difco, 미국)에서 다시 배양하면서, 광(light)조건과 암(dark)조건을 각각 모두 설정하여 배지의 색상 변화를 추가로 관찰하였다.
그 결과, 빛이 주어진 광조건과 빛을 차단한 암조건에서 모두, 액체배지의 색상이 짙은 황색으로 변화된 7개의 돌연변이체를 선별할 수 있었으며(도 2), 이를 통해 본 발명의 클로렐라 속 HS-LZ 미세조류 및 다른 변이체 M2 내지 M7 미세조류를 제조해 수득할 수 있었다.
[실시예 2] 클로렐라 속 HS-LZ 미세조류의 색소 성분 확인
상기 실시예 1을 통해 제조한 본 발명의 클로렐라 속 HS-LZ 미세조류와 다른 변이체 M2 내지 M7, 그리고 야생형의 클로렐라 속 HS2 미세조류를 대상으로, 이들이 생산하는 색소의 성분과 함량을 분석함으로써, 본 발명의 HS-LZ 미세조류의 제아잔틴, 루테인 생산성을 확인하였다.
구체적으로, 상기 HS-LZ 미세조류, M2 내지 M7 변이체 및 야생형 HS2 미세조류의 배양액 2 ㎖을 4 ℃에서 분당 10,000 rpm의 속도로 3분 동안 원심분리하여 상등액을 제거한 다음, 침전된 세포만을 동결건조하였다. 동결건조된 세포 1 g에 100 ㎖의 에탄올을 첨가하여 색소의 추출을 수행하였다. 그리고, 고성능 액체 크로마토그래피(HPLC, Agilent 1260 Infinity Binary LC system) 를 통해 각 미세조류로부터 생산된 색소의 성분 및 함량을 분석하였다(컬럼: Waters Spherisorb® S5 ODS1 4.6 × 250 ㎜, 5 μm Cartridge Column, 컬럼온도: 40 ℃, 주입량: 20 ㎕, 유속: 1.2 ㎖/min, 이동상 A: acetonitrile 84%, methanol 2%, 0.1 M Tris-HCl 14%, 이동상 B: methanol 68%, ethyl acetate 32%, 검출기: Diode Array Detector).
그 결과, 야생형의 클로렐라 속 HS2 미세조류의 경우, 색소의 성분으로 루테인을 함유하고 있는 것으로 나타났으나 제아잔틴은 확인되지 않았으며, 루테인 이외에도 네오잔틴(neoxanthin), 비올라잔틴(violaxanthin), 엽록소 a 및 엽록소 b 등 다른 색소 성분들을 다량 생산하는 것으로 측정되었다. 이와는 달리, 실시예 1을 통해 선별한 변이체 M2 내지 M7 및 본 발명의 클로렐라 속 HS-LZ 미세조류의 경우 HPLC 분석 결과 루테인 및 제아잔틴의 피크만이 나타나, 다른 종류의 색소는 거의 생산하지 않고 루테인 및 제아잔틴이 색소 함량의 대부분을 차지함을 확인할 수 있었다(도 3).

클로렐라 속 HS-LZ 미세조류의 제아잔틴 및 루테인 생산성을 확인하였다. 본 발명의 HS-LZ 미세조류, 상기 변이체 M2 내지 M7 및 야생형의 클로렐라 속 HS2 미세조류의 배양액을 건조한 다음, 상기 실시예 2를 통해 분석한 각 색소 함량을 바탕으로 세포 배양액의 건조 중량 당 함유된 색소의 양을 산출하여 비교하였다. 또한, 본 발명의 HS-LZ 미세조류를 유기탄소원인 포도당이 모두 소모된 상태에서 7일간 추가 배양함으로써 스트레스 조건을 가한 상태로 색소의 성분 및 함량을 측정하여, 마찬가지로 세포 배양액의 건조 중량 당 색소의 양을 산출하여 비교하였다(도 4).
그 결과, 본 발명의 HS-LZ 미세조류 및 M2 내지 M7 변이체들은 색소의 성분으로 제아잔틴 및 루테인만을 생산하며 다른 색소들은 거의 함유하지 않는 것을 확인할 수 있었으며, 특히 제아잔틴의 생산량이 우수함을 확인할 수 있었다. 구체적으로, 본 발명의 HS-LZ 미세조류는 배양액 건조 중량 당 제아잔틴을 1.94 mg/g, 루테인을 0.79 mg/g 생산할 수 있는 것으로 측정되었다. 이와는 달리, 야생형의 HS2미세조류의 경우, 루테인은 일부 생산하나 제아잔틴은 생산하지 못하며 엽록소 a를 가장 많이 생산하는 것을 확인할 수 있었다. 상기와 같은 실험 결과를 통해, 본 발명의 클로렐라 속 HS-LZ 미세조류는, 기존의 클로렐라 속 HS2 미세조류와는 달리, 제아잔틴을 생산할 수 있는 특성이 있으며 생산된 색소량 중 많은 양이 제아잔틴이므로 제아잔틴의 생산성이 우수함을 확인할 수 있었다. 또한, HS-LZ 미세조류는 제아잔틴과 함께 루테인은 생산할 수 있으나, 그 외 엽록소 a를 비롯한 네오잔틴, 비올라잔틴 등 다른 색소들은 생산하지 않는 것을 확인함으로써, 제아잔틴 및 루테인을 별도로 분리, 정제하는 과정이 필요치 않거나 간소화될 수 있다는 장점이 있음을 확인할 수 있었다.
나아가, 본 발명의 HS-LZ 미세조류는 유기탄소원이 소모된 상태에서 배양하는 것과 같이 스트레스 조건 하에서 배양할 경우, 색소 생산성이 현저하게 향상됨을 확인할 수 있었다. 이를 통해, 본 발명의 클로렐라 속 HS-LZ는 배양액 건조 중량 당 제아잔틴을 최대 3.56 mg/g, 루테인을 최대 1.35 mg/g까지 생산할 수 있는 특징이 있어 이들 색소에 대한 생산성이 매우 우수한 미세조류임을 확인할 수 있었다.
이상에서 본 발명은 기재된 실시예에 대해서만 상세히 설명되었지만 본 발명의 기술사상 범위 내에서 다양한 변형 및 수정이 가능함은 당업자에게 있어서 명백한 것이며, 이러한 변형 및 수정이 첨부된 청구범위에 속함은 당연한 것이다.

- 청구항 1에 있어서,상기 클로렐라 속 HS-LZ 미세조류는 제아잔틴(zeaxanthin)을 생산할 수 있는 것인, 미세조류.
- 청구항 1에 있어서,상기 클로렐라 속 HS-LZ 미세조류가 생산하는 색소는, 색소 100 중량부에 대하여 제아잔틴 및 루테인 중 적어도 하나를 90 중량부 이상의 함량으로 함유하는 것인, 미세조류.
- 청구항 1에 있어서,상기 클로렐라 속 HS-LZ 미세조류가 생산하는 색소는, 엽록소 a, 엽록소 b, 네오잔틴(neoxanthin) 및 비올라잔틴(violaxanthin)으로 이루어진 군으로부터 선택되는 적어도 하나를 함유하지 않는 것인, 미세조류.
