Abstract
본 발명은 피부상재 미생물 조절용 조성물에 관한 것으로, 보다 구체적으로는 니신, 라이소자임 및 유산균 발효물을 포함하는 피부상재 미생물 조절용 조성물 및 이를 유효성분으로 포함하는 화장료 조성물을 제공하는 것을 목적으로 한다.
Description
본 발명은 피부상재 미생물 조절용 조성물에 관한 것으로, 보다 구체적으로는 니신, 라이소자임 및 유산균 발효물을 포함하는 피부상재 미생물 조절용 조성물에 관한 것이다.
일반적으로 피부에 상재하는 미생물은 단위면적(cm2)당 1,000여 가지 종류가 존재하는 것으로 알려져 있다. 피부에 상재하는 이러한 미생물들의 다양한 반응과 균형이 피부 건강과 높은 연관성을 지닌다는 연구가 이어지고 있다.
피부에 상재하는 미생물 중에는 피부 화농성균, 여드름균 등과 같이 피부에 유해한 미생물과 피부 락토바실러스, 피부 바실러스 등과 같이 피부에 유익한 미생물이 공존하고 있다.
피부 유해균의 활성이 높은 경우, 여드름 등 여러 가지 피부 트러블이 발생할 수 있고, 건강한 피부일수록 피부 유익균의 활성이 높은 것은 여러 실험을 통해 입증된 바 있다.
현재 시판되고 있는 기능성 화장품 또는 피부 질환 치료용 제제의 경우 피부 유해균을 억제하기 위한 항균 성능만을 포함하고 있어 피부 유익균까지 억제하게 되어 다른 피부 트러블을 야기하는 경우도 있다.
따라서 피부 유익균의 활성을 촉진하면서 피부 유해균을 억제할 수 있는 피부상재 미생물을 제어할 수 있는 기술에 대한 요구가 절실한 상태이다.
상기와 같은 문제를 해결하기 위하여, 본 발명의 일측면은 피부상재 미생물 조절용 조성물을 제공하는 것이다.
본 발명의 일 실시예에 따른 피부상재 미생물 조절용 조성물은, 니신(Nisin), 라이소자임(Lysozyme) 및 유산균 발효물을 포함할 수 있다.
상기 유산균 발효물은 유산균을 발효 및 배양하기 위하여 적절한 배지를 선택할 수 있다. 기술 분야에서 일반적으로 사용하고 있는 배지라면 제한 없이 사용이 가능하다.
본 발명의 일 실시예에 따른 피부상재 미생물 조절용 조성물에 있어서, 상기 유산균은 락토바실러스 파라플란타룸(
Lactobacillus paraplantarum) shebah-401 균주(기탁번호 KCTC 14088BP)일 수 있다.
하나의 바람직한 예에서, 상기 니신 및 상기 라이소자임은 중량비로 0.1 : 1 내지 1 : 1 범위로 포함될 수 있다.
상기 니신은 상기 유산균 발효물 1 ml에 0.1 mg 내지 500 mg 범위로 포함될 수 있고, 상기 라이소자임은 상기 유산균 발효물 1 ml에 1 mg 내지 500 mg 범위로 포함될 수 있다.
본 발명의 일 실시예에 따른 피부상재 미생물 조절용 조성물에 있어서, 상기 유산균 발효물은 원심분리 및 여과를 통해 유산균체를 제거한 것일 수 있다.
본 발명의 일 실시예에 따른 화장료 조성물은, 상기 피부상재 미생물 조절용 조성물을 유효성분으로 포함할 수 있다.
본 발명에 있어서, "유효성분으로 포함하는"의 의미는, 화장료 조성물로서 피부 개선 효과를 나타낼 수 있는, 예컨데 여드름 개선, 항염, 피부 트러블 완화 등의 기능성을 나타낼 수 있는 정도의 유효량을 함유하는 것을 의미한다.
본 발명의 일 실시예에 따른 화장료 조성물에 있어서, 상기 화장료 조성물은 여드름 개선용일 수 있고, 피부 트러블 완화 및 진정용일 수 있다.
상기 화장료 조성물은 화장품 제제에 있어서 수용가능한 담체를 포함할 수 있다. 여기서 "화장품 제제에 있어서 수용가능한 담체"란 화장품 제제에 포함될 수 있는 이미 공지되어 사용되고 있는 화합물 또는 조성물이거나 앞으로 개발될 화합물 또는 조성물로서 피부와의 접촉 시 인체가 적응 가능한 범위를 넘어서는 독성, 불안정성 또는 자극성이 없는 것을 의미한다.
상기 담체는 본 발명의 화장료 조성물에 그것의 전체 중량에 대하여 약 1 중량% 내지 약 99.99 중량%, 바람직하게는 조성물 전체 중량의 약 90 중량% 내지 약 99.99 중량%로 포함될 수 있다. 그러나 상기 비율은 본 발명의 화장료 조성물이 제조되는 제형에 따라 또 그것의 구체적인 적용 부위(얼굴, 목 등)나 그것의 바람직한 적용량 등에 따라 달라지는 것이기 때문에, 상기 비율은 어떠한 측면으로든 본 발명의 범위를 제한하는 것으로 이해되어서는 아니된다.
상기 담체로는 알코올, 오일, 계면활성제, 지방산, 실리콘 오일, 습윤제, 보습제, 점성 변형제, 유제, 안정제, 자외선차단제, 자외선흡수제, 발색제, 향료 등이 예시될 수 있다. 상기 알코올, 오일, 계면활성제, 지방산, 실리콘 오일, 습윤제, 보습제, 점성 변형제, 유제, 안정제, 자외선차단제, 자외선흡수제, 발색제, 향료로 사용될 수 있는 화합물/조성물 등은 이미 당업계에 공지되어 있기 때문에 당업자라면 적절한 해당 물질/조성물을 선택하여 사용할 수 있다.
본 발명에 따른 화장료 조성물은, 다양한 형태로 제조될 수 있는데, 예컨대, 화장수, 에센스, 젤, 에멀전, 로션, 크림(수중유적형, 유중수적형, 다중상), 용액, 현탁액(무수 및 수계), 무수 생성물(오일 및 글리콜계), 마스크, 팩, 분말, 또는 젤라틴 등의 피막이 있는 캡슐(소프트 캡슐 또는 하드 캡슐) 제형 등의 형태로 제조될 수 있다.
본 발명에 있어서의 피부는 얼굴뿐만 아니라, 두피, 전신도 포함되는 개념으로, 이러한 두피에 적용될 수 있는 제형으로, 샴푸, 린스, 트리트먼트, 발모제 등이 있고, 전신에 적용될 수 있는 바디클렌저 등의 제형으로 제조될 수 있다.
본 발명의 실시예들에 따른 피부상재 미생물 조절용 조성물은 피부상재 유해균을 억제하고 피부상재 유익균을 활성화시킬 수 있다.
도 1은 본 발명의 실험예 1의 결과를 나타낸 그래프이다.
도 2는 본 발명의 실험예 2의 결과를 나타낸 그래프이다.
도 3은 본 발명의 실험예 3의 결과를 나타낸 그래프이다.
본문에 개시되어 있는 본 발명의 실시예들에 대해서, 특정한 구조적 내지 기능적 설명들은 단지 본 발명의 실시예를 설명하기 위한 목적으로 예시된 것으로, 본 발명의 실시예들은 다양한 형태로 실시될 수 있으며 본문에 설명된 실시예들에 한정되는 것으로 해석되어서는 아니 된다.
본 발명은 다양한 변경을 가할 수 있고 여러 가지 형태를 가질 수 있는바, 특정 실시예들을 도면에 예시하고 본문에 상세하게 설명하고자 한다. 그러나 이는 본 발명을 특정한 개시 형태에 대해 한정하려는 것이 아니며, 본 발명의 사상 및 기술 범위에 포함되는 모든 변경, 균등물 내지 대체물을 포함하는 것으로 이해되어야 한다.

어떤 구성요소가 다른 구성요소에 "연결되어" 있다거나 "접속되어" 있다고 언급된 때에는, 그 다른 구성요소에 직접적으로 연결되어 있거나 또는 접속되어 있을 수도 있지만, 중간에 다른 구성요소가 존재할 수도 있다고 이해되어야 할 것이다. 반면에, 어떤 구성요소가 다른 구성요소에 "직접 연결되어" 있다거나 "직접 접속되어" 있다고 언급된 때에는, 중간에 다른 구성요소가 존재하지 않는 것으로 이해되어야 할 것이다. 구성요소들 간의 관계를 설명하는 다른 표현들, 즉 "~사이에"와 "바로 ~사이에" 또는 "~에 이웃하는"과 "~에 직접 이웃하는" 등도 마찬가지로 해석되어야 한다.
본 출원에서 사용한 용어는 단지 특정한 실시예를 설명하기 위해 사용된 것으로, 본 발명을 한정하려는 의도가 아니다. 단수의 표현은 문맥상 명백하게 다르게 뜻하지 않는 한, 복수의 표현을 포함한다. 본 출원에서, "포함하다" 또는 "가지다" 등의 용어는 설시된 특징, 숫자, 단계, 동작, 구성요소, 부분품 또는 이들을 조합한 것이 존재함을 지정하려는 것이지, 하나 또는 그 이상의 다른 특징들이나 숫자, 단계, 동작, 구성요소, 부분품 또는 이들을 조합한 것들의 존재 또는 부가 가능성을 미리 배제하지 않는 것으로 이해되어야 한다.
다르게 정의되지 않는 한, 기술적이거나 과학적인 용어를 포함해서 여기서 사용되는 모든 용어들은 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자에 의해 일반적으로 이해되는 것과 동일한 의미이다. 일반적으로 사용되는 사전에 정의되어 있는 것과 같은 용어들은 관련 기술의 문맥상 가지는 의미와 일치하는 의미인 것으로 해석되어야 하며, 본 출원에서 명백하게 정의하지 않는 한, 이상적이거나 과도하게 형식적인 의미로 해석되지 않는다.
이하, 첨부한 도면들을 참조하여, 본 발명의 바람직한 실시예를 보다 상세하게 설명하고자 한다. 도면상의 동일한 구성요소에 대해서는 동일한 참조부호를 사용하고 동일한 구성요소에 대해서 중복된 설명은 생략한다.
본 발명에서는 피부상재 미생물 중에서 피부 화농성균, 피부 여드름균 등의 유해균을 억제하면서 피부 락토바실러스, 피부 바실러스 등의 유익균은 활성화시키는 피부상재 미생물 조절용 조성물을 개발하고자 하였다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
실시예 1
락토바실러스 파라플란타룸(
Lactobacillus paraplantarum) shebah-401 균주(기탁번호 KCTC 14088BP)를 MCT 배지에서 발효(O.D600=1.0)한 후, 원심분리와 여과 과정을 거쳐 유산균체를 제거한 발효물에 시중에 판매되는 니신을 발효물 1 ml당 0.1 mg, 시중에 판매되는 라이소자임을 발효물 1 ml당 1 mg이 되도록 녹여서 조성물을 제작하였다.
실험예 1
피부 유해균인
Staphylococcus aureus(KCCM 12256)를 Trypticase Soy Agar (DifcoCo., USA)에 접종하여 37 ℃에서 24시간 동안 배양한 후, 균주를 Trypticase Soy Agar 배지 plate (90 ㅧ 15 mm)에 100 ㎕ 도말하였다. 이후, 실시예 1의 조성물 100 ㎕, 150 ㎕, 200 ㎕, 250 ㎕, 300 ㎕, 350 ㎕, 400 ㎕를 멸균 disc-paper(지름 6.0 mm)에 점적하였고, 양성대조군(Positive control)으로 항세균제인 ampicillin 0.05 mg을 멸균 disc-paper에 점적하였고, 음성대조군(Negative control)으로 MRS 배지 400 ㎕를 멸균 disc-paper에 점적하였으며, 점적된 멸균 disc-paper들을 균주가 도말된 plate에 올려놓았다. 이후, 37 ℃에서 24시간 동안 배양을 실시하였고, 생육저지환의 크기를 측정하여 항균활성을 평가하였다.
실험예 2
피부 유해균인
Propionibacterium acnes(KCTC 3314)를 Brain Heart Infusion Agar (DifcoCo., USA)에 접종하여 37℃에서 24시간 동안 배양한 후, 균주를 Brain Heart Infusion Agar 배지 plate (90 ㅧ 15 mm)에 100 ㎕ 도말하였다. 이후, 실시예 1의 조성물 100 ㎕, 150 ㎕, 200 ㎕, 250 ㎕, 300 ㎕, 350 ㎕, 400 ㎕를 멸균 disc-paper(지름 6.0 mm)에 점적하였고, 양성대조군(Positive control)으로 항세균제인 ampicillin 0.05 mg을 멸균 disc-paper에 점적하였고, 음성대조군(Negative control)으로 MRS 배지 400 ㎕를 멸균 disc-paper에 점적하였으며, 점적된 멸균 disc-paper들을 균주가 도말된 plate에 올려놓았다. 이후, 37 ℃에서 24시간 동안 배양을 실시하였고, 생육저지환의 크기를 측정하여 항균활성을 평가하였다.
도 1은 실험예 1의 결과를 나타낸 그래프이고, 도 2는 실험예 2의 결과를 나타낸 그래프이다. 도 1 및 도 2를 참조하면, 실험예 1 및 실험예 2 모두 실시예 1의 조성물의 함량에 비례하여 생육저지환의 크기가 커지는 것을 확인할 수 있다. 따라서 실시예 1의 조성물이 피부 유해균인
Staphylococcus aureus(KCCM 12256) 및
Propionibacterium acnes(KCTC 3314)에 대하여 항균 성능을 가지는 것으로 판단된다.
실시예 2
실시예 1의 조성물을 화장품 전체 중량 대비 1.1 중량% 포함하고, 크림 제형에 적합한 담체를 사용하여 크림 형태의 화장품을 제조하였다.
실험예 3
실시예 2의 화장품을 사용하기 전 얼굴 피부에서 피부 미생물을 swab kit을 사용하여 회수하였다. 또한, 실시예 2의 화장품을 매일 얼굴 전체에 도포하는 방식으로 30일 동안 사용한 후, 얼굴 피부에서 피부 미생물을 swab kit을 사용하여 회수하였다. 이렇게 회수된 피부상재 미생물로부터 Nucleospin
® Tissue XS를 이용하여 genome을 추출하였다. 또한, 회수된 피부상재 미생물들 사이에 존재하는
Lactobacillus rhamnosus(유익균),
Bacillus Licheniformis(유익균),
Staphylococcus aureus(유해균),
Propionibacterium acnes(유해균) 미생물 총량은 타겟 미생물의 16S rRNA에 특이적으로 존재하는 염기서열을 선택적으로 증폭할 수 있는 프라이머를 이용하여 PCR 기법을 통해 확인하였고, 전체 미생물의 총량은 미생물 genome내에 존재하는 16S rRNA 전체를 증폭할 수 있는 Universial 프라이머를 이용하여 PCR 기법을 통해 확인하였다. PCR을 완료한 후 각 PCR product들은 전기영동을 통해 분별하였고, 각 밴드의 정량은 densitometry를 이용하여 수행되어졌다. 실시예 2의 화장품 사용 전과 사용 후 총 균체량 내 해당 균주량 비율은 "해당 균주에 대한 density 값/총 균주에 대한 density 값*100"으로 계산하였다.
도 3은 실험예 3의 결과를 나타낸 그래프로, 구체적으로 피부 유익균의 상대 변화량 및 피부 유해균의 상대 변화량을 나타낸 그래프이다. 도 3을 참조하면, 피부 유익균인
Lactobacillus rhamnosus 및
Bacillus Licheniformis는 실시예 2의 화장품을 사용하기 전보다 유의적으로 균주의 상대량이 증가하였다. 이에 반해, 피부 유해균인
Staphylococcus aureus 및
Propionibacterium acnes는 실시예 2의 화장품을 사용하기 전보다 유의적으로 균주의 상대량이 감소되었다.
이러한 결과는 실시예 1의 조성물이 피부 유익균의 성장을 효과적으로 증가시키고, 반대로 피부 유해균의 성장을 효과적으로 억제할 수 있다는 것을 의미한다.
Claims (9)
- 니신, 라이소자임 및 유산균 발효물을 포함하는 것을 특징으로 하는 피부상재 미생물 조절용 조성물.
- 제1항에 있어서,상기 유산균은 락토바실러스 파라플란타룸( Lactobacillus paraplantarum) shebah-401 균주(기탁번호 KCTC 14088BP)인 것을 특징으로 하는 피부상재 미생물 조절용 조성물.
- 제1항에 있어서,상기 니신 및 상기 라이소자임은 중량비로 0.1 : 1 내지 1 : 1 범위로 포함되는 것을 특징으로 하는 피부상재 미생물 조절용 조성물.
- 제1항에 있어서,상기 니신은 상기 유산균 발효물 1 ml에 0.1 mg 내지 500 mg 범위로 포함되는 것을 특징으로 하는 피부상재 미생물 조절용 조성물.
- 제1항에 있어서,상기 라이소자임은 상기 유산균 발효물 1 ml에 1 mg 내지 500 mg 범위로 포함되는 것을 특징으로 하는 피부상재 미생물 조절용 조성물.
- 제1항에 있어서,상기 유산균 발효물은 원심분리를 통해 유산균체를 제거한 것을 특징으로 하는 피부상재 미생물 조절용 조성물.
- 제1항 내지 제6항 중 어느 한 항의 상기 피부상재 미생물 조절용 조성물을 유효성분으로 포함하는 화장료 조성물.
- 제7항에 있어서,상기 화장료 조성물은 여드름 개선용인 것을 특징으로 하는 화장료 조성물.
- 제7항에 있어서,상기 화장료 조성물은 피부 트러블 완화 및 진정용인 것을 특징으로 하는 화장료 조성물.
