Abstract
본 발명은 맥주효모 추출물 및 1,3-프로판디올을 유효성분으로 포함하는 피부의 마이크로바이옴 균형 유지용 화장료 조성물에 관한 것으로, 상기 화장료 조성물은 맥주효모 추출물 및 1,3-프로판디올을 유효성분으로 포함함으로써 피부 유익균의 생장을 선택적으로 촉진시켜 피부 미생물의 균형을 유지하고 이를 통해 손상된 피부를 개선하는 등 피부 건강에 도움을 줄 수 있다.
Description
본 발명은 맥주효모 추출물 및 1,3-프로판디올을 포함하는 피부의 마이크로바이옴 균형 유지용 화장료 조성물에 관한 것이다.
마이크로바이옴(Microbiome)이란 마이크로바이오타(microbiota)와 게놈(genome)의 합성어로, 인간, 동·식물, 토양, 바다, 호수, 암벽, 대기 등 모든 환경에서 서식하거나 공존하는 미생물과 그 유전정보 전체를 포함하는 미생물 군집이라고 할 수 있으며, 최근에는 다양한 환경에서의 미생물 군집을 말하기도 한다. 또한, 식물 마이크로바이옴, 동물 마이크로바이옴, 환경 마이크로바이옴, 인체 마이크로바이옴, 장내 마이크로바이옴, 피부 마이크로바이옴 등 다양한 마이크로바이옴에 대한 연구들이 이루어지고 있으며, 이 중에서도 인체 마이크로바이옴에 대한 연구가 가장 많이 진행되고 있다.
인간의 몸은 복잡하고 생명 활동이 활발한 생태계이며, 인체의 위장 및 피부 등에는 다양한 미생물들이 존재한다. 위장 및 피부 등에는 500종 내지 1,000종 이상의 서로 다른 미생물이 공생하면서 미생물 균총을 구성한다.
피부 표면에 존재하는 미생물은 인간의 성장, 영양, 면역, 병원성에 대한 저항 등과 같은 생리 작용에 영향을 미치며, 외부 미생물, 특히, 병원성 미생물의 침입을 차단하는 역할을 한다.
그러나, 피부에 존재하는 미생물은 피부 유익균뿐만 아니라 유해균도 존재하므로 피부 건강을 위해서는 유익균의 생장은 촉진하면서 유해균의 생장은 억제시켜 피부의 마이크로바이옴 균형을 유지할 필요가 있다.

본 발명은 피부 유익균을 증가시키면서 피부 유해균을 감소시켜 피부의 마이크로바이옴 균형을 유지할 수 있는 화장료 조성물을 제공하는 것을 목적으로 한다.
본 발명의 일 양상은 맥주효모 추출물 및 1,3-프로판디올을 유효성분으로 포함하는 피부의 마이크로바이옴 균형 유지용 화장료 조성물을 제공한다.
본 발명에서 사용된 “맥주효모”는 맥주 제조 공정 중 발효가 완료된 맥주로부터 분리되는 효모를 일컫는다. 상기 맥주효모의 종류로는 사카로미세스 세레비지아에(Saccharomyces cerevisiae), 사카로미세스 파스토리아누스(Saccharomyces pastorianus), 사카로미세스 유바야누스(Saccharomyces eubayanus) 등이 있으나, 이에 제한되는 것은 아니다.
본 발명의 일 실시예에 따르면, 상기 맥주효모는 사카로미세스 세레비지아에인 것일 수 있다.
상기 사카로미세스 세레비지아에는 고대시대부터 맥주뿐만 아니라 포도주, 빵 등을 만드는데 사용되는 야생 효모로, 포도 껍질에서 분리된 것으로 여겨진다.
본 발명에서 사용된 “맥주효모 추출물”은 맥주효모의 추출 처리에 의하여 얻어지는 추출액, 추출액의 희석액이나 농축액, 추출액을 건조하여 얻어지는 건조물, 추출액의 조정제물이나 정제물, 또는 이들의 혼합물 등으로, 추출액 자체 및 추출액을 이용하여 형성 가능한 모든 제형의 추출물을 포함한다.
본 발명의 일 구체예에 따르면, 상기 맥주효모 추출물은 사카로미세스 세레비지아에로부터 당업계에 공지된 추출 방법을 제한 없이 사용하여 얻은 것일 수 있다.
보다 구체적으로, 상기 맥주효모 추출물은 사카로미세스 세레비지아에로부터 파쇄 추출, 냉침 추출, 초음파 추출, 환류 냉각 추출, 열수 추출, 가압 추출 및 용매 추출로 이루어진 군에서 선택된 하나 이상으로 추출된 것일 수 있다. 맥주효모의 효과적인 활성성분 추출을 위해 둘 이상의 추출 방법을 조합하여 사용할 수 있다.
상기 파쇄 추출은 물리적 또는 화학적인 방법을 통해 세포막을 파괴하는 추출 방법이다. 상기 파쇄 추출의 물리적인 방법으로는 블렌더(blender), 믹서(mixer), 호모지나이제(homogenizer) 등을 이용한 기계적 파쇄법이 있으며, 화학적인 방법으로는 효소, 계면활성제 등을 이용한 파쇄법이 있으나, 이에 제한되는 것은 아니다.
상기 용매 추출은 물, 탄소수 1 내지 4개의 알코올, 프로필렌글라이콜, 부틸렌글라이콜, 글리세린, 아세톤, 에틸아세테이트, 부틸아세테이트, 클로로포름, 다이에틸에테르, 다이클로로메탄, 헥산 및 이들의 혼합물로 이루어진 군에서 선택된 하나 이상의 용매로 추출된 것일 수 있다.
본 발명의 일 구체예에 따르면, 상기 맥주효모 추출물은 가열 농축, 동결 농축, 진공저온 농축, 증발 농축, 감압 농축, 상압 농축, 원심 농축 등의 농축 과정 및/또는 열풍 건조, 동결 건조, 분무 건조 등의 건조 과정이 추가적으로 수행된 것일 수 있다.
또한, 상기 맥주효모 추출물은 상술한 추출 방법에 의한 추출물뿐만 아니라 통상적인 정제 과정을 거친 추출물도 포함한다. 예를 들어, 일정한 분자량 컷-오프(cut-off) 값을 갖는 한외 여과막을 이용한 분리, 다양한 크로마토그래피 (크기, 전하, 소수성 또는 친화성에 따른 분리를 위해 제작된 것)에 의한 분리 등, 추가적으로 실시한 다양한 정제 방법을 통해 얻어진 분획도 본 발명의 추출물에 포함될 수 있다.
본 발명에서 사용된 “프로판디올(propanediol)”은 탄소끼리 단일 결합으로 사슬을 형성하는 포화 탄화수소에 두 개의 수산기(-0H)가 연결된 화합물을 의미하며, 수산기의 위치에 따라 1,2- 및 1,3- 두 개의 이성질체를 가진다.
본 발명에서 사용된 “1,3-프로판디올(1,3-propanediol)”은 화장품, 의약품의 보습 성분으로 사용되고 있으며, 식품 첨가물로써도 제한적으로 사용되고 있고, 친수성의 수산기와 소수성의 탄소 사슬을 포함하여 양친매성을 가지며 미생물의 세포막을 쉽 게 침투할 수 있어 우수한 항균력을 나타낸다.
본 발명에서 사용된 “마이크로바이옴”은 인체에 사는 세균, 진균, 바이러스 등 각종 미생물의 군집을 의미하며, “피부 마이크로바이옴”은 전형적으로 인간 피부에 서식 또는 존재하는 미생물을 지칭한다.
본 발명에 사용된 “피부의 마이크로바이옴 균형 유지”는 피부 유익균을 증식시키는 반면 피부 유해균의 증식 및/또는 활성을 억제함으로써 피부 미생물 군집의 균형을 적절히 유지시키는 것을 의미한다.
본 발명의 일 구체예에 따르면, 상기 피부의 마이크로바이옴 균형 유지는 피부 유익균을 증가시키고 피부 유해균을 감소시키는 것일 수 있다.
여기서 “피부 유익균 증가”란, 피부에 상재하거나 외부에서 유입될 수 있는 유익균의 증진을 촉진시키고 유익균의 생육을 활성화시키는 것을 의미하며, “피부 유해균 감소”란 피부에 상재하거나 외부에서 유입될 수 있는 유해균의 증진을 억제하거나 유해균의 생육을 불활성화시킴으로써 피부 미생물의 균형을 유지시키는 것을 의미한다.
본 발명의 일 구체예에 따르면, 상기 피부 유익균 및 유해균은 당업계에 공지된 피부에 서식하는 세균, 진균 등일 있으나, 이에 제한되는 것은 아니다.
보다 구체적으로, 상기 유익균은 스타필로코커스 에피더미디스(Staphylococcus epidermidis), 스트렙토코커스 뉴모니아에(Streptococcus pneumoniae), 락토바실러스 애시도필러스(Latobacillus acidophilus), 락토바실러스 람노서스(Lacotbacillus rhamnosus) 및 비피도박테리움 비피덤(Bifidobacterium bifidum)으로 이루어진 군에서 선택된 하나 이상인 것이고, 상기 유해균은 스타필로코커스 아우레우스(Staphylococcus aureus) 및 큐티박테리움 아크네스(Cutibacterium acnes)로 이루어진 군에서 선택된 하나 이상인 것일 수 있다.
상기 유익균의 일례로, 스타필로코커스 에피더미디스는 그람 양성 세균으로 일반적으로 사람이나 가축의 피부, 점막 등에 존재하며, 글리세린을 만들어 피부의 보습 효과를 지속시키며, 유기산을 합성하여 피부를 산성으로 유지시키고, 항생물질을 생산하여 스타필로코커스 아우레우스와 같은 피부 유해균의 생장을 억제시킬 수 있다.
상기 유해균의 일례로, 스타필로코커스 아우레우스는 그람 양성의 통성 혐기성 세균으로 건강한 사람이나 가축의 피부, 비강의 표면에 존재하여 식중독뿐만 아니라 피부의 화농, 중이염, 방광염 등 화농성 질환을 일으키는 원인균으로 알려져 있다. 큐티박테리움 아크네스는 방선균의 일종으로 혐기성 또는 내산소성이며, 주로 사람의 피부, 결막, 외이, 인두북, 여성 비뇨기 등에 상재하며, 모낭의 피지선에서 분비되는 지방산을 분해하면서 염증 반응을 유도하여 여드름의 원인이 되거나 기회감염균으로 작용한다.
본 발명에 따른 화장료 조성물은 맥주효모 추출물과 1,3-프로판디올의 혼합에 의한 상승 효과(synergy effect)로 인해 피부의 유익균을 선택적으로 증가시키고 유해균을 감소시켜 피부의 마이크로바이옴 균형을 유지할 수 있다.
본 발명의 일 구체예에 따르면, 상기 화장료 조성물은 맥주효모 추출물 및 1,3-프로판디올을 1 : 0.1 내지 40의 부피비로 포함하는 것일 수 있다.
상기 맥주효모 추출물 및 1,3-프로판디올의 부피비가 1 : 0.1 미만인 경우에는 1,3-프로판디올에 의한 피부 유익균의 생장 촉진 상승 효과를 기대하기 어려워 효율적으로 피부의 마이크로바이옴 균형을 유지할 수 없고, 1 : 40을 초과할 경우에는 피부 유해균뿐만 아니라 피부 유익균의 생장을 억제하여 피부의 마이크로바이옴 균형이 깨질 수 있으며 1,3-프로판디올에 의한 피부 자극이 유발될 수 있다.
본 발명에 따른 화장료 조성물은 피부의 마이크로바이옴 균형 유지를 위해 맥주효모 추출물 및 1,3-프로판디올을 1 : 0.1 내지 40의 부피비, 바람직하게는 1 : 0.5 내지 20의 부피비, 더욱 바람직하게는 1 내지 1 내지 10의 부피비로 포함할 수 있다.
또한, 본 발명의 일 구체예에 따르면, 상기 화장료 조성물은 맥주효모 추출물 및 1,3-프로판디올을 0.01 내지 40%(v/v)로 포함하는 것일 수 있다.
상기 화장료 조성물 내 맥주효모 추출물 및 1,3-프로판디올의 유효 농도가 0.01%(v/v)인 경우에는 유익균의 생장을 촉진할 수 없어, 피부의 마이크로바이옴 균형 유지 효과를 나타낼 수 없고, 궁극적으로 피부 상태 개선 효과를 나타내지 못하며, 40%(v/v)를 초과할 경우에는 유해균 및 유익균의 생장 또는 활성을 모두 억제하여 피부의 마이크로바이옴 균형이 깨지는 문제점이 있다.
본 발명에 따른 화장료 조성물은 피부의 마이크로바이옴 균형 유지를 위해 맥주효모 추출물 및 1,3-프로판디올을 0.01 내지 40%(v/v), 바람직하게는 0.05 내지 20%(v/v), 더욱 바람직하게는 0.1 내지 10%(v/v)를 포함할 수 있다.
본 발명에서 화장료 조성물은 화장품 분야에서 통상적으로 사용되는 기제, 보조제 및 첨가제를 사용하여 액체 또는 고체 형태로 제조될 수 있다. 액체 또는 고체 형태의 화장품으로는, 예를 들면, 화장수, 크림제, 로션제, 입욕제 등의 형태를 포함할 수 있고, 이에 한정되지는 않는다. 화장품 분야에서 통상적으로 사용되는 기제, 보조제 및 첨가제는 특별히 제한되지 않으며, 예를 들면, 물, 알코올, 프로필렌 글리콜, 스테아린산, 글리세롤, 세틸 알코올, 유동 파라핀 등이 있다.
본 발명의 화장료 조성물은 화장료 조성물에 통상적으로 이용되는 성분들을 포함할 수 있으며, 예컨대 항산화제, 안정화제, 용해화제, 비타민, 안료 및 향료와 같은 통상적인 보조제, 그리고 담체를 포함할 수 있다.
본 발명에서 화장료 조성물은 당업계에서 통상적으로 제조되는 어떠한 제형으로도 제조될 수 있으며, 예를 들어 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 계면활성제-함유 클렌징, 오일, 마이크로캡슐, 미세과립구, 이온형 소낭 분산제, 비이온형 소낭 분산제, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션, 스틱, 마스크팩 및 스프레이 등으로 제형화될 수 있으며, 이에 한정되는 것은 아니다. 보다 상세하게는, 유연 화장수, 영양 화장수, 영양 크림, 마사지 크림, 에센스, 아이크림, 클렌징 크림, 클렌징 폼, 클렌징 워터, 팩, 스프레이 또는 파우더의 제형으로 제조될 수 있다.
본 발명에서 화장료 조성물의 제형이 페이스트, 크림 또는 겔인 경우에는 담체 성분으로 동물성유, 식물성유, 왁스, 파라핀, 전분, 트라칸트, 셀룰로오스 유도체, 폴리에틸렌글리콜, 실리콘, 벤토나이트, 실리카, 탈크 또는 산화아연 등이 이용될 수 있다.
본 발명에서 화장료 조성물의 제형이 파우더 또는 스프레이인 경우에는 담체 성분으로서 락토스, 탈크, 실리카, 알루미늄 하이드록시드, 칼슘 실리케이트 또는 폴리아미드 파우더가 이용될 수 있고, 특히, 스프레이인 경우에는 추가적으로 클로로플루오로히드로카본, 프로판/부탄 또는 디메틸에테르와 같은 추진체를 포함할 수 있다.
본 발명에서 화장료 조성물의 제형이 용액 또는 유탁액인 경우에는 담체 성분으로서 용매, 용해화제 또는 유탁화제가 이용될 수 있고, 예를 들어 물, 에탄올, 이소프로판올, 에틸카보네이트, 에틸아세테이트, 벤질알코올, 벤질벤조에이트, 글리세롤 지방족 에스테르, 폴리에틸렌글리콜, 또는 소르비탄의 지방산 에스테르 등이 이용될 수 있다.
본 발명에서 화장료 조성물의 제형이 현탁액인 경우에는 담체 성분으로서 물, 에탄올과 같은 액상의 희석제, 에톡실화 이소스테아릴 알코올, 폴리옥시에틸렌 소르비톨 에스테르 및 폴리옥시에틸렌 소르비탄 에스테르와 같은 현탁제, 미소결정성 셀룰로오스, 알루미늄 메타히드록시드, 벤토나이트, 아가 또는 트라칸트 등이 이용될 수 있다.
본 발명에서 화장료 조성물의 제형이 계면-활성제 함유 클렌징인 경우에는 담체 성분으로서 지방족 알코올 설페이트, 지방족 알코올 에테르 설페이트, 설포숙신산 모노에스테르, 이미다졸리늄 유도체, 메틸타우레이트, 사르코 시네이트, 지방산 아미드에테르설페이트, 알킬아미도베타인, 지방족 알코올, 지방산 글리세리드, 지방산 디에탄 올아미드, 식물성유, 라놀린 유도체 또는 에톡실화 글리세롤 지방산 에스테르 등이 이용될 수 있다.
본 발명에서 화장료 조성물은 단독 또는 중복 도포하여 사용하거나, 본 발명 이외의 다른 화장료 조성물과 중복 도포하여 사용할 수 있다. 또한 본 발명의 화장료 조성물은 통상적인 사용 방법에 따라 사용될 수 있으며, 사용자의 피부 상태 또는 취향에 따라 그 사용횟수를 달리할 수 있다.
본 발명에서 화장료 조성물이 비누, 계면활성제 함유 클렌징 또는 계면활성제 비함유 클렌징 제형일 경우, 피부에 도포한 후 닦아내거나 떼거나 물로 씻어낼 수도 있다. 일예로, 비누는 액상비누, 가루비누, 고형비누 및 오일비누이며, 계면활성제 함유 클린징 제형은 클렌징 폼, 클렌징 워터, 클렌징 수건 및 클렌징 팩이며, 계면활성제 비함유 클렌징 제형은 클렌징 크림, 클렌징 로션, 클렌징 워터 및 클렌징 겔이며, 이에 한정되는 것은 아니다.
본 발명에 따른 화장료 조성물은 맥주효모 추출물 및 1,3-프로판디올을 유효성분으로 포함함으로써 피부 유익균의 생장을 선택적으로 촉진시켜 피부 미생물의 균형을 유지하고 이를 통해 손상된 피부를 개선하는 등 피부 건강에 도움을 줄 수 있다.

이하, 본 발명을 보다 상세하게 설명한다. 그러나, 이러한 설명은 본 발명의 이해를 돕기 위하여 예시적으로 제시된 것일 뿐, 본 발명의 범위가 이러한 예시적인 설명에 의하여 제한되는 것은 아니다.
준비예 1. 맥주효모 생균
맥주효모 생균(Live yeast)으로는 사카로미세스 세레비지아에(S. cerevisiae) (ATCC 2341)를 사용하였다. S. cerevisiae는 YPD(yeast extract peptone dextrose) 액체 배지를 이용하여 30℃에서 24시간 진탕배양하였으며, 배양액은 원심분리기를 통해 상등액을 제거하고 가라앉은 S. cerevisiae만을 취하였다.
준비예 2. 맥주효모 추출물
맥주효모 추출물(Yeast lysate)은 준비예 1의 맥주효모 생균 S. cerevisiae를 이용하여 준비하였다. 간략히 설명하면, 라이소자임(lysozyme) (Sigma-aldrich, 10837059001)을 이용해 S. cerevisiae를 파쇄한 후 준비예 1에서 생균 배양 시 생성된 알코올을 증발기(evaporator)를 이용하여 제거하였고, 135℃에서 6초간 살균한 뒤 분무 건조법(spray drying)으로 건조하여 맥주효모 추출물을 준비하였다.
준비예 3. 맥주효모 추출물 및 프로판디올 혼합물
1,3-프로판디올은 Sigma-Aldrich를 통해 입수하였으며, 맥주효모 추출물 및 프로판디올 혼합물은 준비예 2의 맥주효모 추출물, 1,3-프로판디올 및 증류수를 5:30:65의 부피비로 혼합하여 준비하였다.
준비예 4. 피부 유익균 및 유해균
피부 유익균으로는 스타필로코커스 에피더미디스(S. epidermidis)을 사용하였고, 피부 유해균으로는 스타필로코커스 아우레우스(S. aureus) 및 큐티박테리움 아크네스(C. acnes)를 사용하였다.
S. epidermidis (ATCC 12228) 및 S. aureus (ATCC 6538)는 TSB(tryptic soy broth) 배지에서 32℃, 18시간 배양한 후 0.85% NaCl로 희석하여 흡광도 (600 ㎚)가 0.5 ~ 0.6이 되도록 조절하여 준비하였다.
C. acnes (ATCC 6919)는 RCM(reinforced clostridial medium) 액체 배지를 이용하여 37℃, 혐기성 조건으로 4 ~ 6일간 배양한 후 0.85% NaCl로 희석해 흡광도 (600 nm)가 0.5 ~ 0.6이 되도록 조절하여 준비하였다.
실시예 1. 맥주효모 생균 및 맥주효모 추출물이 피부 유익균 생장에 미치는 영향
맥주효모 생균 및 맥주효모 추출물이 피부 유익균 생장에 미치는 영향을 평가하였다.
준비예 1의 맥주효모 생균 또는 준비예 2의 맥주효모 추출물 1%(v/v)을 포함하는 10% TSB 배지를 준비한 후 S. epidermidis를 > 106 CFU/g의 농도가 되도록 1%(v/v) 접종하였다. 이후 32℃, 4시간 배양한 후 0.85% NaCl을 이용하여 10-1 ~ 10-8로 희석하여 TSA 고체 배지에 도말하였고, 다시 32℃, 24시간 배양한 후 S. epidermidis의 콜로니(colony)를 확인하였다.
그 결과, 도 1에 나타낸 바와 같이, 맥주효모 생균은 S. epidermidis 생장에 영향을 미치지 않는 반면, 맥주효모 추출물은 S. epidermidis 생장을 촉진하였다.
실시예 2. 맥주효모 추출물이 피부 세포에 미치는 영향
맥주효모 추출물이 피부 세포에 미치는 영향을 평가하기 위해, 세포 독성 시험을 실시하였다.
케라틴 세포주인 HaCaT 세포는 DMEM 배지 (10% FBS 및 1% penicillin/ streptomycin 함유)에서 37℃에서 배양하여 사용하였다.
96웰 플레이트에 HaCaT 세포를 1 x 104 cells/well이 되도록 준비한 후 준비예 2의 맥주효모 추출물을 농도별 (31.25, 62.5, 125, 250, 500, 1000, 2000 μg/ml)로 처리하였다. 이후 37℃, 48시간 배양한 후 Quanti-MAX™ WST-8 세포 생존력 분석 키트(cell viability assay kit, Biomax)를 사용하여 450 nm에서 흡광도를 측정하여 세포 생존력을 확인하였다.
대조군으로는 맥주효모 추출물 대신 PBS를 사용하였다.
그 결과, 도 2에 나타낸 바와 같이, 맥주효모 추출물은 HaCaT 세포에 대해 독성을 나타내지 않았다.
실시예 3. 맥주효모 추출물이 피부 유익균 및 유해균의 바이오필름 형성능에 미치는 영향
맥주효모 추출물이 피부 유익균 및 유해균의 바이오필름 형성능에 미치는 영향을 평가하였다.
준비예 2의 맥주효모 추출물 1, 3 또는 5%(v/v)을 포함하는 10% TSB 배지를 준비한 후 12웰 플레이트에 1.2 ml/well씩 분주하였다. 그리고 S. epidermidis 또는 S. aureus를 > 106 CFU/g의 농도가 되도록 1%(v/v) 접종하였다. 이후 32℃, 80 rpm으로 5일간 배양하여 12웰 플레이트 벽면에 바이오필름 형성을 유도하였다. 배양액을 제거한 후 1% 크리스탈 바이올렛 용액(Crystal violet solution)을 벽면의 바이오필름이 잠기도록 1.5 ml/well 정도 분주하였고, 바이오필름이 염색될 수 있도록 30분간 실온에서 방치하였다. 그 후 크리스탈 바이올렛 용액을 제거한 후 멸균수로 3번 세척하였다. 마지막으로 96% EtOH 1.5 ml/well로 바이오필름을 녹여낸 후 흡광도 (590 nm) 측정을 통해 바이오필름 형성능을 측정하였다.
대조군으로는 맥주효모 추출물 대신 10% TSB 배지를 사용하였다.
그 결과, 도 3에 나타낸 바와 같이, 맥주효모 추출물은 S. aureus의 바이오필름 형성을 억제하는 반면, S. epidermidis의 바이오필름 형성을 선택적으로 촉진하였다.
실시예 4. 맥주효모 추출물이 피부 유익균 및 유해균 생장에 미치는 영향
맥주효모 추출물이 피부 유익균 및 유해균 생장에 미치는 영향을 평가하였다.
준비예 1의 맥주효모 생균 또는 준비예 2의 맥주효모 추출물 1%(v/v)을 포함하는 10% TSB 배지를 준비한 후 S. epidermidis, S. aureus 또는 C. acnes를 > 106 CFU/g의 농도가 되도록 1%(v/v) 접종하였다. 이후 32℃, 4시간 배양한 후 0.85% NaCl을 이용하여 10-1 ~ 10-8로 희석하여 TSA 고체 배지에 도말하였고, 다시 32℃, 24시간 배양한 후 각 균의 콜로니를 확인하였다.
그 결과, 도 4에 나타낸 바와 같이, 맥주효모 추출물은 S. aureus 및 C. acnes의 생장에 영향을 미치지 않으면서 실시예 1에서와 같이 S. epidermidis 생장만을 선택적으로 촉진하였다.
실시예 5. 맥주효모 추출물 및 프로판디올 혼합물이 피부 유익균 및 유해균 생장에 미치는 영향
맥주효모 추출물 및 프로판디올 혼합물이 피부 유익균 및 유해균 생장에 미치는 영향을 평가하였다.
준비예 3의 맥주효모 추출물 및 프로판디올 혼합물 1 또는 5%(v/v)를 10% TSB 배지에 첨가한 후 S. epidermidis 또는 S. aureus를 > 106 CFU/g의 농도로 1%(v/v) 접종하였다. 이후 32℃, 24시간 배양하고 2시간 간격으로 흡광도 (600 nm)를 측정하여 각 균의 생장률을 확인하였다.
대조군으로는 준비예 2의 맥주효모 추출물 0.05% 또는 0.25%(v/v)을 사용하였다.
그 결과, 도 5에 나타낸 바와 같이, 맥주효모 추출물 및 프로판디올 혼합물은 S. aureus 생장에 큰 영향을 미치지 않는 반면, S. epidermidis 생장을 더 촉진하였다.
실시예 6. 맥주효모 추출물 및 프로판디올 혼합물의 농도별 피부 유익균 및 유해균 생장에 미치는 영향
맥주효모 추출물 및 프로판디올 혼합물의 농도에 따른 피부 유익균 및 유해균 생장에 미치는 영향을 평가하였다.
준비예 3의 맥주효모 추출물 및 프로판디올 혼합물 0.1, 0.5, 1, 3 또는 5%(v/v)에 대해 실시예 5와 동일한 방법으로 각 균의 생장률을 확인하였다.
대조군으로는 1,3-프로판디올 : 증류수 = 30 : 70의 부피비로 포함하는 조성물 (맥주효모 추출물 제외) 5%(v/v)를 사용하였다.
그 결과, 도 6에 나타낸 바와 같이, 맥주효모 추출물 및 프로판디올 혼합물은 실시예 5에서와 같이 S. aureus 생장에 큰 영향을 미치지 않는 반면, 농도가 0.1% 이상일 때부터 S. epidermidis 생장을 촉진하며 0.1 ~ 3%에서는 큰 차이가 없으나 5%에서는 S. epidermidis 생장을 약 40% 증가시켰다.
실시예 7. 맥주효모 추출물 및 프로판디올 혼합물의 농도별 UV 스트레스에 의해 사멸된 피부 유익균 및 유해균의 생장 회복에 미치는 영향
맥주효모 추출물 및 프로판디올 혼합물의 농도를 조절하여 UV 스트레스에 의해 사멸된 피부 유익균 및 유해균의 생장 회복에 미치는 영향을 평가하였다.
준비예 3의 맥주효모 추출물 및 프로판디올 혼합물 1 또는 5%(v/v)을 포함하는 10% TSB 배지에 S. epidermidis 또는 S. aureus를 > 106 CFU/g의 농도로 1%(v/v)가 되도록 접종한 후 UV를 30분간 조사하여 균이 80% 이상 사멸한 것을 확인하였다. 이어서 32℃에서 최대 24시간 배양하며 2시간 간격으로 흡광도 (600 nm)를 측정하여 각 균의 생장률을 확인하였다.
그 결과, 도 7에 나타낸 바와 같이, 맥주효모 추출물 및 프로판디올 혼합물은 14시간 배양 후 최종 회복된 S. aureus의 양에는 영향을 끼치지 않으면서 회복 속도를 증가시킨 반면, 24시간 배양 후 최종 회복된 S. epidermidis의 양을 증가시켰다.
이제까지 본 발명에 대하여 그 바람직한 실시예들을 중심으로 살펴보았다. 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자는 본 발명이 본 발명의 본질적인 특성에서 벗어나지 않는 범위에서 변형된 형태로 구현될 수 있음을 이해할 수 있을 것이다. 그러므로 개시된 실시예들은 한정적인 관점이 아니라 설명적인 관점에서 고려되어야 한다. 본 발명의 범위는 전술한 설명이 아니라 특허청구범위에 나타나 있으며, 그와 동등한 범위 내에 있는 모든 차이점은 본 발명에 포함된 것으로 해석되어야 할 것이다.
Claims (7)
- 맥주효모 추출물 및 1,3-프로판디올을 유효성분으로 포함하는 피부의 마이크로바이옴 균형 유지용 화장료 조성물.
- 청구항 1에 있어서,상기 맥주효모 추출물은 사카로미세스 세레비지아에(Saccharomyces cerevisiae)로부터 파쇄 추출, 냉침 추출, 초음파 추출, 환류 냉각 추출, 열수 추출, 가압 추출 및 용매 추출로 이루어진 군에서 선택된 하나 이상으로 추출된 것인 화장료 조성물.
- 청구항 1에 있어서,상기 피부의 마이크로바이옴 균형 유지는 피부 유익균을 증가시키고 피부 유해균을 감소시키는 것인 화장료 조성물.
- 청구항 3에 있어서,상기 유익균은 스타필로코커스 에피더미디스(Staphylococcus epidermidis), 스트렙토코커스 뉴모니아에(Streptococcus pneumoniae), 락토바실러스 애시도필러스(Latobacillus acidophilus), 락토바실러스 람노서스(Lacotbacillus rhamnosus) 및 비피도박테리움 비피덤(Bifidobacterium bifidum)으로 이루어진 군에서 선택된 하나 이상인 것이고,상기 유해균은 스타필로코커스 아우레우스(Staphylococcus aureus) 및 큐티박테리움 아크네스(Cutibacterium acnes)로 이루어진 군에서 선택된 하나 이상인 것인 화장료 조성물.
- 청구항 1에 있어서,상기 화장료 조성물은 맥주효모 추출물 및 1,3-프로판디올을 1 : 0.1 내지 40의 부피비로 포함하는 것인 화장료 조성물.
- 청구항 1에 있어서,상기 화장료 조성물은 맥주효모 추출물 및 1,3-프로판디올을 0.01 내지 40%(v/v)로 포함하는 것인 화장료 조성물.
- 청구항 1에 있어서,상기 화장료 조성물은 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 계면활성제-함유 클렌징, 오일, 마이크로캡슐, 미세과립구, 이온형 소낭 분산제, 비이온형 소낭 분산제, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션, 스틱, 마스크팩 및 스프레이로 이루어진 군에서 선택된 하나 이상인 것인 화장료 조성물.
