Abstract
본 발명은 온도감응성 히알루론산 비드 겔에 관한 것으로, 상세하게 상기 온도감응성 히알루론산 구형 비드 겔은 고순도이면서 균일하고, 선택적인 크기의 투명한 구형 형태를 지닐 뿐만 아니라, 121℃ 멸균조건에서도 그 형태가 분해되지 않는 특성을 가지며, 생체적합성, 점탄성 및 열과 효소에 대한 안정성 등이 매우 우수하다.
Description
본 발명은 온도감응성 히알루론산 비드 겔에 관한 것으로, 히알루론산, 가교제 및 알칼리 수용액을 혼합하여 구형 겔을 제조한 후 삼원공중합은 혼합하여 온도감응성 히알루론산 비드 겔 제조할 수 있다.
가교된 히알루론산으로 구성된 비드를 제조하는 일반적으로 널리 알려진 방법들로는 화학적 방법과 물리적 방법으로 혼합하여 사용하였으며, 비드를 분리하는 방법으로는 미세 노즐을 통해 침전을 하거나, 외부를 지질화 및 캡슐화를 진행하는 방법, 가용화 된 생분해성 고분자를 계면활성제를 이용하여 분산 후 용매 추출 증발에 의해 수득하는 방법 등이 알려져 있다.
물리적 방법으로 입자의 균일도를 높이기 위해서는 impeller의 모양 및 교반 속도에 영향을 받는데 대부분 고속의 교반 속도를 유지해야한다. 미세 노즐을 통한 방법은 석류열매와 유사한 외관을 만들어내 완벽한 구 형태를 유지하지 못하고, 외부를 지질화 및 캡슐화를 통해 구형 비드를 수득했을 경우는 물리적 외력에 파손되어 내부의 액상이 흘러나오고, 용매 추출 증발법을 사용시 인체에 유해한 계면활성제와 유기용매가 필수적으로 사용되어 최종산물에 대한 완벽한 제거가 필요하다.
그 외 가교 된 히알루론산으로 구성된 비드를 제조하는 일반적인 방법은 양이온을 나타내는 물질을 과량 사용하여 음이온 히알루론산과의 이온결합을 통해서 불용성 침전물 형태로 제조하거나, 과량의 가교제를 사용하여 불용성 형태를 만든 후 건조하여 물리적으로 입자화 시키거나, 또는, 히알루론산의 카르복실기에 소수성 물질을 결합시켜 유기용매에 녹인 후 상 분리되는 두 개의 다른 용매에서 유화제등을 사용하여 마이크로비드를 형성시키고 용매 추출법 등으로 마이크로 비드 들을 회수하는 방법을 나타나고 있다. BDDE에 의해 가교 된 히알루론산 비드의 제조 관련 문헌에서는 알칼리 수용액에 히알루론산을 10℃에서 overnight 용해시켜 분자량 저하가 일어난 후, 과량의 BDDE(히알루론산 100mg 당 BDDE 20mg, 약 가교 반응 site 대비 약 40 mol% 정도)를 첨가 한 후, olive oil 교반 반응 후, filer를 통해 회수 하고, 70% 아세톤 수용액, 증류수, PBS(phosphate buffer saline, 인산염완충식염수)로 정제하는 제조 방법으로 20 ~ 30%에 가까운 높은 화학적 변형률과 낮은 팽윤도 18.99 ~ 23.02로 너무 단단하고 불균일한 모양을 지니고 있고, 응용분야에서 사용에 한계를 보이고 있다.
또한, 가교제로 알렌드로네이트(ALD), 아데노신 디포스페이트(ADP) 및 아데노신 트리포스페이트(ATP)로 이루어진 군의 리간드와 가교반응을 시켜서 가교 된 히알루론산을 만들고, 이를 용해시킨 후, 다가 양이온에 떨어 뜨러 나노/마이크로비드를 제조하는 방법으로, 안전성 및 안정성에 대한 문제로 인해 응용분야 사용에 한계를 보이고 있다.
또한, 폴리에틸렌옥사이드-폴리프로필렌옥사이드-폴리에틸렌옥사이드 삼원 공중합체 15~30중량%, 수불용상 히알루론산 2.5~4.5중량%(가교제 0.02~0.1 중량%), 소듐알지네이트 0.1~1중량%로 구성이 된 조직유착 방지용 하이드로겔 조성물 특허가 있는데, 가교 된 히알루론산이 구형이 아닌 무정형 겔이고, 폴리에틸렌옥사이드-폴리프로필렌옥사이드-폴리에틸렌옥사이드 삼원 공중합체은 일반적으로 온도감응성 특성을 보유하기 위한 일반적인 범위인 15~30중량%로 보이고 있다.
하지만, 히알루론산은 친수성이 상당히 높은 물질로서 일반적으로 알려진 상기 방법으로 투명하고, 팽윤도가 높으면서도 균일한 구형 비드를 제조하는 데에는 그 동안 한계를 가지고 있었다. 오일 상 내에서 수용액상의 히알루론산은 친수성이 상당히 높은 관계로 용매 증발법으로 수분을 제거하기가 어렵고, 용매 추출법으로 유기용매를 첨가할 경우, 입자가 서로 엉기는 현상을 보여서 균일한 크기의 비드를 제조할 수가 없었다. 또한, 현재까지 알려진 히알루론산의 비드 제조방법은 양이온을 나타내는 물질을 과량 사용하여 음이온 히알루론산과 이온결합을 통해서 불용성 침전물 형태로 제조하거나, 과량의 가교제를 사용하여 불용성 형태를 만든 후 건조하여 물리적으로 입자화 시키거나, 또는 히알루론산의 카르복실기에 소수성 물질을 결합시켜 유기용매에 녹인 후 용매 추출법으로 제조하는 것들이었는데, 이는 입자의 크기가 상당히 작고, 불균일한 모양을 가지면서, 수용액상에 팽윤 정도가 매우 낮은 단단한 불투명한 입자형태로서 응용분야에서 한계를 보여왔다.
위와 같이 히알루론산의 독특한 물질 특성으로 인해, 종래 기술들은 균일한 구형 및 투명성을 유지하면서도, 수용액상에서 팽윤 정도가 매우 높은 비드를 제조하는 공정에는 적용할 수가 없었다.
또한, 폴리에틸렌옥사이드(PEO)-폴리프로필렌옥사이드(PPO)-폴리에틸렌옥사이드(PEO) 삼블록 공중합체(triblock copolymer)를 사용하여 온도감응성 특성을 부여하기 위해서는 13중량% ~ 30중량%의 고함량을 적용해서 제조할 수밖에 없는 한계가 있다.
본 발명의 목적은 균일한 구형 모양 및 크기 조절, 가교를 통한 히알루론산 구형 비드 겔의 분해기간 조절, 우수한 물리적인 안정성을 가짐으로써 체내, 체외의 다양한 환경에서도 생체적합성이 우수하여 의료기기, 제약, 화장품, 식품 산업 등에 유용하게 사용될 수 있는 투명하고, 팽윤도가 높으면서도 균일한 구형의 온도감응성 히알루론산 구형 비드 겔을 제공하는 것이다.
상기 목적을 달성하기 위하여, 본 발명은 히알루론산 및 삼원 공중합체를 포함하고, 상기 온도감응성 히알루론산 비드 겔은 주파수 스윕(frequency sweep)을 통해 1Hz에서 저장 탄성률(G', Pa) 값이 37℃에서 25℃보다 2배 이상인 것을 특징으로 하는, 온도감응성 히알루론산 비드 겔을 제공한다.
본 발명의 온도감응성 히알루론산 구형 비드 겔은 고순도이면서 균일하고, 선택적인 크기의 투명한 구형 형태를 지닐 뿐만 아니라, 아니라 121℃ 멸균조건에서도 그 형태가 분해되지 않는 특성을 가지며, 생체적합성, 점탄성 및 열과 효소에 대한 안정성 등이 매우 우수하다.

본 발명은 온도감응성 히알루론산 구형 비드 겔 제조 시 오일상에서 유화제를 사용하지 않고 히알루론산 용액과 오일의 종류 및 상대 비율로 비드 크기를 조절하고, 에폭시 반응기를 둘 이상 가지고 있는 소량의 가교제(polyfunctional epoxide)와 화학적으로 결합시킨 히알루론산 구형의 비드 겔을 제공한다.
본 발명은 히알루론산 및 삼원 공중합체를 포함하는 온도감응성 히알루론산 비드 겔을 제공한다.
상기 히알루론산은 히알루론산 또는 그의 염을 의미하는데, 히알루론산 염에는 히알루론산 나트륨, 히알루론산 칼륨, 히알루론산 칼슘, 히알루론산 마그네슘, 히알루론산 아연, 히알루론산 코발트 등과 같은 무기염과, 히알루론산 테트라부틸암모늄 등과 같은 유기염이 모두 포함한다.
상기 히알루론산 또는 그의 염의 분자량은 특별히 제한되지 않으며 100,000 내지 5,000,000 달톤(dalton)인 것이 바람직하다.
상기 삼원 공중합체는 1~500 kDa의 범위의 폴리에틸렌옥사이드-폴리프로필렌옥사이드-폴리에틸렌옥사이드일 수 있다.
상기 온도감응성 히알루론산 비드 겔은 삼원 공중합체를 8 내지 12중량%로 포함할 수 있다.
상기 온도감응성 히알루론산 비드 겔은 주파수 스윕(frequency sweep)을 통해 1Hz 및 37℃에서 저장 탄성률(G', Pa) 값이 500 내지 3000 Pa 또는 700 내지 2000 Pa을 나타낼 수 있다.
또한, 상기 온도감응성 히알루론산 비드 겔은 주파수 스윕(frequency sweep)을 통해 1Hz 및 25℃에서 저장 탄성률(G', Pa) 값이 50 내지 500 Pa 또는 100 내지 400 Pa을 나타낼 수 있다.
상기 온도감응성 히알루론산 비드 겔은 주파수 스윕(frequency sweep)을 통해 1Hz에서 저장 탄성률(storage modulus; G', Pa) 값이 37℃에서 25℃보다 2배 이상, 3배 이상, 4배 이상 또는 5 내지 10배일 수 있다.
상기 온도감응성 히알루론산 비드 겔의 평균 직경은 50 ~ 2,000 ㎛가 바람직하다. 히알루론산 구형 비드 겔의 크기에 따라서 의학적으로 적용되는 범위를 다양화할 수 있다.
상기 온도감응성 히알루론산 비드 겔은 10~25 N의 압력을 가하는 경우에도 형태를 유지할 수 있다.
본 발명의 온도감응성 히알루론산 구형 비드 겔은 투명한 구형 형태로 선택된 크기와 분해 기간를 갖고 우수한 점탄성을 나타내며, 생체적합성이 우수하며, 열과 효소에 대하여 매우 안정하다.
구체적으로, 상기 온도감응성 히알루론산 비드 겔은 120~125℃의 온도에서 멸균이 가능하고, 체내에 도입된 상태에서 어떠한 부작용도 없을 뿐만 아니라, 주름개선 필러, 성형보조제, 관절주사제, 유착방지제로 사용할 수 있는 물성을 제공할 수 있으며, 약물전달체 또는 세포전달체로써 특정한 약물 또는 세포를 특정 생체부위에 전달하는 매개체(vehicle)로도 사용될 수 있고, 조직공학용 세포지지체 등으로도 사용될 수 있다.
본 발명의 온도감응성 히알루론산 구형 비드 겔은 종래의 제조 방법 즉, 구형 비드의 외부를 지질화 및 캡슐화를 진행하는 방법, 유화제 및 계면활성제 등을 사용하는 방법, 과량의 가교제를 사용하여 불용성 형태를 만든 후 건조하여 물리적으로 입자화 방법, 소수성 물질을 결합시켜 유기용매에 녹인 후 상분리 하는 제조방법은 적용하지 않고 제조하는 것이 특징이다.
또한, 본 발명의 온도감응성 히알루론산 비드 겔은 히알루론산, 가교제 및 알칼리 수용액을 혼합하여 반응시키는 제1 단계; 상기 반응 혼합물을 오일에 분산시킨 후 가교시켜 구형 겔을 제조하는 제2 단계; 상기 제조된 구형 겔을 회수하여 미반응물 및 불순물을 제거하여 정제시키는 제3 단계; 및 상기 정제된 구형 겔을 삼원 공중합체를 혼합하여 조제하는 제4 단계;를 거쳐 제조된 것일 수 있다.
상기 제1 단계는 히알루론산, 가교제 및 알칼리 수용액을 혼합하여 반응시키는 단계로, 히알루론산을 알킬리 수용액에 넣고, 가교제를 첨가하고 균질하게 혼합하여 반응물을 반응시킬 수 있다.
상기 히알루론산은 산의 형태와 그의 염을 모두 포함하는 개념이므로, 본 발명의 제조방법에서 "히알루론산"은 히알루론산, 히알루론산 염, 및 히알루론산과 히알루론산 염의 혼합물을 모두 포함한다.
상기 알칼리 수용액은 특별히 제한되는 것은 아니지만, 구체적으로 수산화 나트륨, 수산화칼륨, 암모니아수와 같은 시약이 사용된다. 상기 알칼리 수용액의 농도는 특별히 제한되지 않으며 농도는 0.1 내지 10N인 것이 히알루론산의 용해 및 가교 반응을 위해 바람직하다.

상기 가교제는 히알루론산 또는 그의 염의 알코올기와 반응하여 에테르 결합을 형성하여 히알루론산 또는 그의 염 간에 가교가 이루어진다. 에폭시 작용기와 알코올과의 반응은 고온과 알칼리 상태에서 반응이 진행되는 것으로 알려져 있고, 반응 진행에 따라서 히알루론산 또는 그의 염에 붙어있게 되므로 부산물은 발생하지 않는다. 단, 반응 효율이 낮을 경우, 히알루론산 또는 그의 염과 결합한 가교제의 여러 에폭시 작용기중 미반응의 에폭시 작용기가 히알루론산 또는 그의 염에 남아있게 되며, 이러한 잔량의 미반응 에폭시 작용기는 향후 생체 내 물질과 반응할 위험이 있어 이를 최소화하거나 에폭시 작용기를 가수분해하여 비활성작용기로 변환시키는 추가의 반응과 공정이 필요하다. 또한, 과량의 가교제를 첨가하는 경우는 미반응의 가교제가 히알루론산 또는 그의 염의 가교 물질에 남아있을 위험성이 매우 크다. 따라서 이러한 위험을 최소화하기 위하여, 본 발명에 기술된 바와 같이 적당한 양의 가교제를 사용하여 고효율의 반응을 유도하는 것이 히알루론산 구형 비드 겔의 생체적합성의 관점에서 뿐만 아니라 제조공정효율 측면에서도 매우 바람직하다.
상기 가교제가 균일하게 혼합된 히알루론산 또는 그의 염 알칼리 수용액을 다양한 종류의 오일과 혼합한 후 적절히 교반하고 일정 온도에서 가교 반응을 시켜 주게 된다. 오일 상에서 분산되어 있는 히알루론산 또는 그의 염 알칼리수용액은 용매가 증발하면서 서서히 고농도로 바뀌게 되고 이에 따라 실질적인 반응농도가 증가하면서 반응의 효율성을 극대화 시킬 수 있어 소량의 가교제를 사용하여 우수한 물성을 지닌 히알루론산 구형 비드 겔을 제조할 수 있다. 따라서, 상기 기술된 히알루론산 가교 반응 공정은 낮은 농도로 가교제가 혼합되고 용매가 증발함에 따라 가교 반응은 상대적으로 고농도에서 이루어지기 때문에 낮은 농도의가교제가 사용되었을 때의 장점과 고농도의 가교제가 사용되었을 때의 장점을 동시에 취할 수 있다. 이러한 히알루론산 구형 비드 겔 제조방법은 견고하고 특별한 물성을 지닌 히알루론산 구형 비드 겔을 제조할 수 있을 뿐만 아니라 가교 반응의 효율성을 동시에 가질 수 있다.
상기 제1 단계에서 가교제는 히알루론산의 반복단위를 기준으로 1 내지 30당량%의 비율로 혼합된 것일 수 있다. 상기 가교제의 농도가 상기 범위이면 팽윤도가 높으면서도 형태 유지가 잘되고 안전한 비드겔이므로 바람직하다. 반면, 1 당량% 미만이면 쉽게 분해가 되고 형태가 유지되지 못하는 단점으로 인해서 바람직하지 못하고, 30당량% 초과이면 높은 가교제 사용으로 인한 부작용 빈도수가 증가하고 또한, 팽윤도가 낮은 단단한 물성으로 인해 사용 편의성이 떨어져서 바람직하지 못하다.
또한, 상기 제1 단계에서 히알루론산의 농도는 0.1 내지 10 N의 알칼리 수용액에 대해서 1 내지 30중량%의 비율로 혼합될 수 있다. 히알루론산 또는 그 염의 농도가 상기 범위이면 균질한 혼합 및 반응이 가능하고, 팽윤도가 높으면서 형태 유지가 가능한 비드겔이 가능하므로 바람직하다. 반면, 1 중량% 미만이면 가교가 제대로 되지 않아서 형태 유지가 어려워 서 바람직하지 못하고, 30중량% 초과이면 너무 높은 농도로 인해 균질한 혼합이 어려워서 불균일한 반응이 일어나고 너무 높은 가교로 인해 단단한 특성이 나타나므로 바람직하지 못하다.
상기 반응 혼합물의 가교 반응 시 온도는 특별히 제한되고 있지 않지만 5℃내지 50℃인 것이 바람직하고 15℃ ~ 40℃이 더욱 바람직하다. 또한, 반응 용액에서 가교 결합의 생성을 위한 지속시간은 특별히 제한되지 않으나 반응 시간이 길어질수록 히알루론산이 알칼리 조건에 의해 가수 분해되어 제조된 히알루론산 구형 비드 겔의 물리적 점탄성이 낮아지는 문제점이 있으므로, 제1 단계의 반응 시간은 30분 내지 24 시간일 수 있다.
상기 제2 단계는 상기 반응물을 오일에 분산시킨 후 가교시켜 구형 겔을 제조하는 단계이다. 구체적으로, 상기 제2 단계의 반응 시간은 1시간 내지 96시간일 수 있다.
상기 제1 단계의 가교 반응 후, 오일에 분산되어 있는 상태에서 다시 가교 반응을 통해 투명하고, 팽윤도가 매우 높은 균일한 구형 형태의 온도감응성 히알루론산 구형 비드 겔을 형성하고, 별도의 유화제, 계면활성제 및 가용화제 등을 사용하지 않고, 오일과 히알루론산 요액의 비율을 조절하여 제조할 수 있다.
상기 오일은 미네랄 오일 (mineral oil), 미디움 체인 트리글라이세라이드 (medium chain triglycerides), 맨헤이든유 (menhaden oil), 아마인유 (linseed oil), 코코넛 오일 (coconut oil), 대구 오일 (cod oil, fish oil), 대두유 (soya bean oil), 고래기름 (whale oil), 목화 씨 오일 (cotton seed oil), 참깨 씨 오일 (sesame seed oil), 땅콩 기름 (peanut oil), 올리브 오일 (olive oil), 라드 기름 (lard oil), 야자유 (palms oil), 옥수수 기름 (corn oil), 채유 (rapeseed oil), 골유 (bone oil), 챠이나 우드 오일 (china wood oil) 및 피마자유 (castor oil)로 이루어진 군에서 선택된 하나 이상이다.
상기 제2 단계는 상기 반응 혼합물과 오일은 1:1 내지 1:9의 중량비로 혼합될 수 있다. 히알루론산 또는 그의 염 알칼리 수용액과 오일의 혼합 중량비가 상기 범위이면 생산성이 우수하면서 균일한 비드겔이 가능하므로 바람직하다. 반면, 히알루론산 또는 그의 염 알칼리 수용액과 오일의 혼합중량비가 1:1 미만이면 히알루론산 또는 그의 염 알카리 수용액이 오일보다 많아서 뭉치는 현상으로 인해 균일하게 오일상에 분산이 되기 어려워서 바람직하지 못하고, 1:9를 초과하면 히알루론산 또는 그의 염 알카리 수용액이 오일상에 균일하게 분산이 잘 되기는 하나 부피 대비 생산성에서 떨어지는 단점으로 인해서 바람직하지 못하다.
상기와 같이 상기 반응 혼합물을 다양한 종류의 오일과 혼합 시 그 혼합시간과 수용액 대비 오일 비율을 조절하여 히알루론산 구형 비드 겔의 물성을 조절할 수도 있다.
유화제가 첨가되지 않는 상태에서 히알루론산 구형 비드 겔의 크기는 교반 속도, 오일의 종류 및 수용액 대비 오일의 비율을 통해서 조절이 되고 있는데, 오일 상의 점도가 높을수록 히알루론산 구형 비드 겔의 크기가 작아지는 결과를 보이고 있다. 이는 상대적으로 오일 상 보다 높은 점도를 보이고 있는 히알루론산 및 그의 염 알칼리 수용액이 두 개의 상 간의 점도 차이가 줄어 들면서 보다 원활하게 분산이 되고 있기 때문으로 보인다.
또한, 비드의 크기는 교반 속도가 증가할수록, 히알루론산 및 그의 염 알칼리 수용액의 점도가 낮을수록 작아지고 있다.
상기 제3 단계는 상기 제2 단계에서 제조된 구형 겔을 회수하여 미반응물 및 불순물을 제거하는 정제단계이다.
상기 제3 단계는 (a) 상기 제2 단계에 제조된 구형 겔을 증류수, 산성용액, 산성염이 첨가된 수용액 및 인산염완충식염수로 이루어진 군에서 선택된 하나 이상을 사용하여 1차 세척하는 단계; (b) 상기 1차 세척된 구형 겔을 유기용매 수용액으로 2차 세척하는 단계; 및 (c) 상기 2차 세척된 구형 겔을 증류수, 산성용액, 산성염이 첨가된 수용액 및 인산염완충식염수 중에서 선택된 하나 이상을 사용하여 3차 세척하는 단계를 거쳐 구형 겔을 정제시킬 수 있다.
상기 (a) 및 (c) 단계는 독립적으로 증류수나, 산성용액 또는 산성염이 첨가된 수용액 및 인산염완충식염수를 이용하여 수회 세척을 수행하여 미반응의 가교제와 수산화 이온을 제거한다.
상기 염이 첨가된 수용액을 사용함으로써 미반응물의 세척 효율을 증대시킬 수 있는데, 이 경우 염화나트륨 수용액, 인산 수용액, 인산염이 포함된 염화나트륨 수용액이 가장 적합하다.
상기 유기용매는 디메틸설폭사이드(DMSO), 디메틸포름아마이드(DMF), 아세트나이트릴, 메탄올, 테트라하이드로퓨란(THF), 아세톤, 아세톤 수용액 및 탄소수 2 내지 6의 알코올 중에서 선택된 하나 이상을 포함할 수 있다. 상기 (b) 단계에서, 상기 유기용매 수용액은 오일을 제거하는 역할을 한다.
상기 유기용매 수용액은 유기용매 대비 증류수의 중량비가 9:1 내지 1:9일 수 있다. 유기용매 대비 증류수의 중량비가 상기 범위이면 적절하게 비드겔이 팽윤하면서 오일 및 불순물 제거가 용이하므로 바람직하다. 반면, 유기용매 대비 증류수의 중량비가 9:1 이상이면 높은 유기용매 중량비로 인해 비드겔이 충분히 팽윤하지 못하고 그로 인해서 오일 및 불순물 제거가 용이하지 못하게 되어서 바람직하지 못하고, 1:9 미만이면 낮은 유기용매 중량비로 인해 오일 및 불순물 제거가 용이하지 못하게 되어서 바람직하지 못하다.
상기와 같이 상기 반응물을 유기용매와 오일 상에 섞이는 특성에 따라 제조된 히알루론산 구형 비드 겔의 물성이 달라질 수 있고, 유기용매의 극성지표가 증류수와 가까울수록 제조된 히알루론산 구형 비드 겔의 투명도는 증가하고 복합 점도는 낮아짐을 알 수 있어서 다양한 물성의 히알루론산 구형 비드 겔을 제조할 수 있다.
상기 제4 단계는 상기 정제된 구형 겔과 삼원 공중합체를 혼합하여 온도감응성 히알루론산 비드 겔을 조제하는 단계이다.
구체적으로, 상기 정제된 구형 겔을 완충용액에 팽윤시킨 후 삼원 공중합체와 혼합하여 팽윤된 히알루론산 비드 겔에 삼원 공중합체를 침투시키고, 120~125℃의 온도로 멸균시켜 온도감응성 히알루론산 구형 비드 겔을 제조할 수 있다.
또는, 상기 정제된 구형 겔을 유기용매 또는 유기용매 수용액을 이용하여 히알루론산 구형 비드 겔을 침전 상태로 회수하고 질소, 공기, 열, 감압을 통하여 건조한 후 의약용 적용 목적에 따라 온도감응성 히알루론산 구형 비드 겔을 크기 형태별로 분류하고, 완충용액으로 팽윤시키고, 온도감응성 합성고분자를 혼합 조제하고, 120~125℃의 온도로 멸균시켜 온도감응성 히알루론산 구형 비드 겔을 제조할 수 있다.
상기 히알루론산 구형 비드 겔 침전에 사용하는 유기용매에는 디메틸설폭사이드(DMSO), 디메틸포름아마이드(DMF), 아세트나이트릴, 메탄올, 테트라하이드로퓨란(THF), 아세톤, 아세톤 수용액, 에탄올 등의 탄소수 2 내지 6의 알코올 또는 알코올 수용액을 사용할 수 있다. 또한, 상기 완충용액은 인산염완충식염수(Phosphate buffer saline, PBS)일 수 있다.
상기 제4 단계는 삼원 공중합체를 80~120 mg/ml 또는 90~110 mg/ml의 농도로 혼합할 수 있다.
상기 삼원 공중합체는 1~500 kDa의 범위의 폴리에틸렌옥사이드-폴리프로필렌옥사이드-폴리에틸렌옥사이드일 수 있다.
이하에서는 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예
[실시예 1] 온도감응성 히알루론산 구형 비드 겔 제조
50 mg/ml의 농도로 히알루론산 나트륨 (분자량: 1,000,000)을 0.25 N 166mL NaOH 용액에 넣고, 동시에 부탄다이올다이글라이시딜에테르 (BDDE)를 0.4183g첨가하였다 (히알루론산 반복단위에 대한 BDDE의 당량비: 10 mol%). 30분 동안 자공전원심믹서를 통해 혼합을 시켜 준 후, 40℃에서, 1시간 동안 1단계 반응을 시켜준다. 상기 반응물을 332g의 MCT 오일상에 떨어뜨린다. 700rpm 속도로 교반시켜 주면서 40℃에서 24 시간 동안 2단계 반응을 시켜 주었다.
24 시간 후, 500 ml의 아이소프로판올을 서서히 첨가시켜 혼합액에서 분산되어 있는 히알루론산 비드들을 회수시켜 주었다. 아이소프로판올로 3회 세척하였다. 회수된 히알루론산 비드들을 각 5L PBS로 5회 세척하였다. 세척이 된 히알루론산 비드에 아이소프로판올 1L을 첨가하여 침전을 시켜준다. 3회 반복하여 아이소프로판올 교체를 실시하였다. 침전물을 30% 아이소프로판올 1.5L을 첨가하여 1시간 동안 팽윤을 시켜준다. 팽윤 후, 3회 반복하여 아이소프로판올 교체를 실시하였다. 침전물을 다시 각 5L PBS로 5회 세척하였다.
세척된 히알루론산 구형 비드 겔의 일부는 PBS에 팽윤 시킨 후, 비드 겔에 Pluronic F127을 100mg/mL이 되도록 첨가한 후, 121℃ 멸균하여 온도감응성 히알루론산 구형 비드 겔을 제조하고, 다른 히알루론산 구형 비드 겔 일부는 유기용매 침전공정, 질소가스 건조공정, 분류공정, PBS 재 팽윤 공정, 121℃ 멸균공정을 거쳐 용도별로 적합한 크기 및 형태의 히알루론산 구형 비드 겔을 제조하였다.
[비교예 1] 히알루론산 구형 비드 겔 제조

24 시간 후, 500 ml의 아이소프로판올을 서서히 첨가시켜 혼합액에서 분산되어 있는 히알루론산 비드들을 회수시켜 주었다. 아이소프로판올로 3회 세척하였다. 회수된 히알루론산 비드들을 각 5L PBS로 5회 세척하였다. 세척이 된 히알루론산 비드에 아이소프로판올 1L을 첨가하여 침전을 시켜준다. 3회 반복하여 아이소프로판올 교체를 실시하였다. 침전물을 30% 아이소프로판올 1.5L을 첨가하여 1시간 동안 팽윤을 시켜준다. 팽윤 후, 3회 반복하여 아이소프로판올 교체를 실시하였다. 침전물을 다시 각 5L PBS로 5회 세척하였다.
세척된 히알루론산 구형 비드 겔의 일부는 PBS에 팽윤 시킨 후, 비드 겔에 Pluronic F127을 150mg/mL이 되도록 첨가한 후, 121℃ 멸균하여 온도감응성 히알루론산 구형 비드 겔을 제조하고, 다른 히알루론산 구형 비드 겔 일부는 유기용매 침전공정, 질소가스 건조공정, 분류공정, PBS 재 팽윤 공정, 121℃ 멸균공정을 거쳐 용도별로 적합한 크기 및 형태의 히알루론산 구형 비드 겔을 제조하였다.
[실험예] 실시예 1에서 제조된 온도감응성 히알루론산 구형 비드 겔과 비교예 1, 2, 3, 4, 5에서 제조된 히알루론산 구형 비드 겔의 물성 비교
실시예 1 및 비교예 1, 2, 3, 4, 5에서 제조된 온도감응성 하알루론산 비드 겔이 구형임을 확인하였다(도 2).
실시예 1 및 비교예 1, 2, 3, 4, 5에서 제조된 온도감응성 히알루론산 구형 비드 겔의 유변학적 특성을 확인하기 위하여 레오미터 (rheometer)를 사용하여 25℃에서 40℃까지 temperature sweep(1Hz, frequency)을 통해 온도에 따른 storage modulus(G', Pa)의 값 변화를 확인하였다(도면 1, 표 1). 실시예 1에서 온도에 따른 물성변화가 크게 일어나고 있음을 확인하였고, 25℃와 37℃에서 frequency sweep을 통해 0.02 ~ 1 Hz의 진동수에서 (complex modulus, Storage modulus, Loss modulus)을 측정하였고(도면 3), 그 결과 37℃에서 높은 물성을 보이고 있음을 확인할 수 있었다.
도 3을 참조하면, 1에서 0.02Hz 전 영역에서 저장탄성율(Storage modulus)이 손실탄성율(Loss modulus)보다 높은 값을 유지하고 있어, 탄성율이 높아서 형태 유지력이 좋은 우수한 점탄성을 보이고 있고, 온도가 증가하면서 저장탄성율(Storage modulus, Pa)이 105.4에서 1,023로 증가되고 있어 겔 타입으로 조직 수복용 재료로서 우수한 성능을 보이고 있음을 확인할 수 있었다.

실시예 1 및 비교예 1, 2, 3, 4, 5에서 제조된 온도감응성 히알루론산 구형 비드 겔의 Extrusion force를 측정한 결과(도면 5), 실시예 1의 경우 27G needle 장착 시 7.14 ± 2.39N, 비교예 1의 경우 27G needle 장착 시 10.31 ± 2.55N, 비교예 2의 경우 27G needle 장착 시 8.79 ± 2.55N, 비교예 3의 경우 27G needle 장착 시 10.09 ± 2.49N, 비교예 4의 경우 27G needle 장착 시 12.91 ± 1.09N, 비교예 5의 경우 27G needle 장착 시 12.79 ± 2.15N 값을 보이고 있어, 현재 시장에서 판매가 되고 있는 주요 제품들의 Extrusion force인 10 ~ 20 N값과 비교하여 낮은 결과를 보이고 있어 사용성이 우수한 결과를 보이고 있다.실시예 1에서 제조된 온도감응성 히알루론산 구형 비드 겔을 27G Needle 통과 후에도 모양이 변하지 않고 구형의 형태를 잘 유지하고 있음을 확인하였다(도면 5).
이는 구형 비드의 형태가 needle을 장착하여 시술 후에도, 도 5에서 최대 압출력인 25N에서도 모양이 깨지지 않고, 구형의 형태 유지가 가능할 정도로 비드 겔의 탄성이 우수하여 시술 부위에서 외부 충격이 가해도 구형의 모양 유지가 가능함을 알 수 있다.
이상으로 본 발명의 특정한 부분을 상세히 기술한 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적인 기술은 단지 바람직한 구현 예일 뿐이며, 이에 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항과 그의 등가물에 의하여 정의된다고 할 것이다.
본 발명의 범위는 후술하는 특허청구범위에 의하여 나타내어지며, 특허청구범위의 의미 및 범위 그리고 그 균등 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.
Claims (12)
- 히알루론산 및 삼원 공중합체를 포함하고,상기 온도감응성 히알루론산 비드 겔은 주파수 스윕(frequency sweep)을 통해 1Hz에서 저장 탄성률(G', Pa) 값이 37℃에서 25℃보다 2배 이상인 것을 특징으로 하는, 온도감응성 히알루론산 비드 겔.
- 제 1 항에 있어서,상기 온도감응성 히알루론산 비드 겔은 구형의 형태이고,상기 온도감응성 히알루론산 비드 겔의 평균 직경은 50 내지 2000 ㎛인 것을 특징으로 하는 온도감응성 히알루론산 비드 겔.
- 제 1 항에 있어서,상기 온도감응성 히알루론산 비드 겔은 삼원 공중합체를 8 내지 12중량%로 포함하는 온도감응성 히알루론산 비드 겔.
- 제 1 항에 있어서,상기 삼원 공중합체는 1~500 kDa의 범위의 폴리에틸렌옥사이드-폴리프로필렌옥사이드-폴리에틸렌옥사이드인 것을 특징으로 하는, 온도감응성 히알루론산 비드 겔.
- 제 1 항에 있어서,상기 온도감응성 히알루론산 비드 겔은 히알루론산, 가교제 및 알칼리 수용액을 혼합한 반응 혼합물을 오일에 분산시킨 후 가교시켜 제조한 구형 겔; 및 삼원 공중합체를 혼합하여 제조된 것을 특징으로 하는, 온도감응성 히알루론산 비드 겔.
- 제 5 항에 있어서,상기 구형 겔은 상기 반응 혼합물을 오일에 분산시킨 후 가교시켜 구형 겔을 제조한 후 상기 구형 겔을 회수하여 미반응물 및 불순물을 제거하여 정제시켜 제조된 것을 특징으로 하는, 온도감응성 히알루론산 비드 겔.
- 제 6 항에 있어서,상기 구형 겔은 증류수, 산성용액, 산성염이 첨가된 수용액 및 인산염완충식염수로 이루어진 군에서 선택된 하나 이상을 사용하여 1차 세척하고, 상기 1차 세척된 구형 겔을 유기용매 수용액으로 2차 세척한 후, 상기 2차 세척된 구형 겔을 증류수, 산성용액, 산성염이 첨가된 수용액 및 인산염완충식염수 중에서 선택된 하나 이상을 사용하여 3차 세척하여 정제시키는 것을 특징으로 하는 온도감응성 히알루론산 비드 겔.
- 제 5 항에 있어서,상기 가교제는 히알루론산의 반복단위를 기준으로 1 내지 30당량%의 비율로 혼합된 것을 특징으로 하는, 온도감응성 히알루론산 비드 겔.
- 제 5 항에 있어서,상기 히알루론산의 농도는 0.1 내지 10 N의 알칼리 수용액에 대해서 1 내지 30중량%의 비율로 혼합된 것을 특징으로 하는, 온도감응성 히알루론산 비드 겔.
- 제 5 항에 있어서,상기 반응 혼합물과 오일은 1:1 내지 1:9의 중량비로 혼합된 것을 특징으로 하는, 온도감응성 히알루론산 비드 겔.
- 제 10 항에 있어서,상기 유기용매 수용액은 유기용매 대비 증류수의 중량비가 9:1 내지 1:9인 것을 특징으로 하는, 온도감응성 히알루론산 비드 겔.
- 제 10 항에 있어서,상기 유기용매는 디메틸설폭사이드(DMSO), 디메틸포름아마이드(DMF), 아세트나이트릴, 메탄올, 테트라하이드로퓨란(THF), 아세톤, 아세톤 수용액 및 탄소수 2 내지 6의 알코올 중에서 선택된 하나 이상인 것을 특징으로 하는, 온도감응성 히알루론산 비드 겔.
