Description
본 발명은 발효숙성노니 및 깔라만시를 유효성분으로 포함하는 조성물에 관한 것으로, 더욱 상세하게는, 발효숙성노니 및 깔라만시를 유효성분으로 포함하여 스트레스를 완화시키고, 스트레스로 인해 나타나는 증상을 예방 및 개선할 수 있는 조성물에 관한 것이다.
만병의 근원이라 불리는 스트레스는 인간이나 동물의 모든 삶의 영역에 존재한다. 스트레스는 강력한 자극으로서, 일상에서 경험하는 사소한 일부터 큰 충격을 주는 주요한 사건까지 다양한 요인에 의해 유발되고, 이러한 스트레스를 통한 자극은 피로와 권태감, 우울감 등을 유발시키고, 내·외적으로 부정적인 반응을 일으킨다.
일반적으로 신체에 미치는 자극이 일정한 정도 이상이 될 경우 신체에 손상을 주게 되는데, 이때 생체는 그 자극의 종류에 관계없이 일정한 위협에 대한 반응으로 비특이적인 전신증후군(general adaptative syndrome)을 나타내며, 이러한 현상을 스트레스라 한다(Selye, H. :The stress of life, Toronto, Longnans Green and Co., pp. 1-50, 1958). 이러한 스트레스는 다양한 나타나는데, 긴장성 두통, 편두통, 고혈압, 소화불량, 피로, 통증, 갱년기 장애, 탈모, 피부의 거칠어짐이 그 예가 된다. 또한 스트레스가 만성적으로 지속되면 각종 신경증, 위궤양 등이 나타나며, 신체의 생리활성, 면역활성 등이 저하하고, 정신적 활동을 위축시키고 정서가 불안정해져 비정상적 행동을 유발하기도 한다.
특히, 현대사회에서는 스트레스가 과도하게 발생하고 있기 때문에 스트레스는 매우 중요한 사회적 문제로 대두되고 있다.
스트레스를 유발하는 원인과 그 신체의 변화는 다양하지만 공통적으로는 교감신경계인 뇌하수체-부신계(HPA)의 기능을 항진시켜 호르몬 분비를 유발하여 부신의 기능항진과 비장의 기능저하를 일으키고, 그 결과 스트레스에 의한 모든 증상이 나타나는 것으로 알려지고 있다. 스트레스가 부가되면 비장은 면역기능의 감퇴로 인하여 무게가 현저히 줄어들며, 부신피질자극호르몬에 의하여 부신의 기능이 항진되어 무게가 증가되며, 지질감소현상이 일어나고, 코르티졸(cortisol)의 분비가 촉진된다. 코르티졸의 양은 연령 및 스트레스에 따라 증가하는 경향이 있으며 이는 또한 비만에 기여한다. 또한, 스트레스가 부가되면 부신은 호르몬을 분비하여 혈액 내에서 콜레스테롤, 혈당, 락테이트 디하이드로게나제(lactate dehydrogenase: LDH) 등을 증가시킨다.
한편 스트레스와 연관이 깊은 위 궤양의 경우에는 활성산소종이 이러한 소화기관의 궤양 생성에 관여한다는 것이 알려져 있다. 활성산소 또는 활성산소종(Reactive Oxygen Species; ROS)을 제거하기 위한 생체 방어시스템은 수퍼옥사이드 디스뮤타아제 (superoxide dismutase; SOD), 카탈라아제(catalase), 글루타치온 퍼옥시다아제(glutathione peroxidase) 등의 효소계에 의한 효소적 방어체계와 ROS나 자유라디칼의 연쇄반응을 중단 또는 종결시키는 비효소적 방어체계로 크게 구별된다. 산소를 이용하는 생물체는 수퍼옥사이드(superoxide)를 제거하는 효소인 SOD를 가지고 있어 생체는 수퍼옥사이드에 의한 손상으로부터 보호되고 있다. 카탈라아제는 조직내에서 SOD 등의 효소적 반응에 의해 생성된 H2O2를 제거하여 생체를 방어하는 기능을 나타낸다. 또한 대사과정 중 발생하는 활성산소종의 유리기를 제거할 뿐만 아니라 이들 활성산소에 의해 비가역적으로 불활성화 될 수도 있으며, 지방산화에 의해 생성된 유리기도 제거하는 것으로 보고된 바 있다. 동물조직 중 비단백 티올(thiol)의 대부분을 차지하는 글루타치온(GSH, glutathione)은 유리기제거제 역할과 H2O2 및 과산화지질을 대사시키는 GSH-Px의 기질로써 세포내 항산화제 중요한 역할을 담당하고 있다(Cho SY.et al., J. Korean Soc. Food Sci. Nutr., 32(3), pp458-463, 2003).
근래에는 스트레스 해소를 위해 다양한 방법들이 강구되고 있는데, 가장 효과적인 방법으로 운동이나 심리 치료 등이 이용되고 있다. 또한, 스트레스의 강도에 따라 약물 치료가 병행되어 스트레스로 인해 발생하는 피해를 최소화시키기 위한 방법들도 존재한다. 특히, 스트레스 완화를 위한 약물로는 신경안정제인 벤조디아제핀, 정신 안정제인 트랭퀼라이저와 같은 향정신약물 등이 이용된다.
그러나 스트레스 완화를 위해 위와 같은 안정제를 이용할 경우 중독의 위험성이 있고, 합성약품의 특성상 부작 용을 배제하기 어렵다는 문제점이 있다. 약물에 지나친 의존은 오히려 또 다른 스트레스를 불러 일으킬 수 있다는 점에서 부정적인 측면이 존재한다.
최근 한약재료를 이용하여 피로회복 혹은 스트레스 완화를 목적으로 하는 식품의 개발이 다양하게 시도되었다.
천연 한의약재를 이용한 피로회복용 조성물에 관한 연구로는 대한민국 공개특허공보 특2000-0066777호에서 천궁, 감초, 갈근, 지구자, 적양, 진피, 대조, 미나리, 및 은행잎을 추출한 추출액을 이용한 기능성 음료가 공개되었으며, 공개특허공보 제10-2005-0108856호에서는 황정, 맥문동, 구기자, 오미자를 함유하는 피로회복용 한방음료가 공개되어 있다.
스트레스 해소용 식품에 대한 연구로는 대한민국 공개특허공보 제10-2004-0006705호에서 심층수, 유기 게르마늄및 향부자, 진피, 맥아, 산조인 및 대추를 필수성분으로 하는 스트레스 해소용 건강보조식품이 공개되었으며, 공개특허공보 제10-2004-0018888호에서 홍삼, 천마, 생지황즙 등을 주원료로 하는 스트레스 해소 식품이 공개되 었다. 또한 공개특허공보 제10-2005-0080455호에서 종래의 사군자탕에 매실, 둥글레, 갈근, 계피 추출물을 추가한 항스트레스 효과를 가지는 가미사군자탕이 공개되었다.
자연계에 존재하는 식물 중에 질병의 방지와 노화 억제 등 생체조절기능을 가지는 기능성 성분들이 다량 함유되어 있다는 것이 밝혀져 천연식품소재에 관한 연구가 활발히 이루어지고 있다.
따라서, 최근에는 식물 추출물과 같이 안전성이 보장된 천연물을 이용하여 기능성 보조제를 개발하고자 하는 연구가 활발히 진행되고 있다. 이에 본 발명자들은 발효숙성노니 및 깔라만시에 관한 연구를 지속한 결과, 이들 복합물의 섭취로 스트레스 완화에 효과적인 것을 확인하고, 본 발명에 이르게 되었다.

본 발명의 하나의 목적은 발효숙성노니 또는 그 추출물 및 깔라만시 추출물을 유효성분으로 포함하는 스트레스 완화용 조성물을 제공하는데 있다.
본 발명의 다른 하나의 목적은 발효숙성노니 또는 그 추출물 및 깔라만시 추출물을 유효성분으로 포함하는 조성물의 제조방법을 제공하는데 있다.
본 발명의 또 다른 하나의 목적은 발효숙성노니 또는 그 추출물 및 깔라만시 추출물을 유효성분으로 스트레스 완화용 식품 조성물을 제공하는데 있다.
본 발명의 또 다른 하나의 목적은 발효숙성노니 또는 그 추출물 및 깔라만시 추출물을 유효성분으로 스트레스 완화용 약학적 조성물을 제공하는데 있다.

본 명세서에 개시된 발명의 기술적 사상에 따라 이루고자 하는 기술적 과제는 이상에서 언급한 문제점을 해결하기 위한 과제로 제한되지 않으며, 언급되지 않은 또 다른 과제는 아래의 기재로부터 통상의 기술자에게 명확하게 이해될 수 있을 것이다.
이를 구체적으로 설명하면 다음과 같다. 한편, 본 출원에서 개시된 각각의 설명 및 실시형태는 각각의 다른 설명 및 실시 형태에도 적용될 수 있다. 즉, 본 출원에서 개시된 다양한 요소들의 모든 조합이 본 출원의 범주에 속한다. 또한, 하기 기술된 구체적인 서술에 의하여 본 출원의 범주가 제한된다고 볼 수 없다.
상기와 같은 과제를 해결하기 위하여, 본 발명은 발효숙성노니 또는 그 추출물 및 깔라만시 추출물을 유효성분으로 포함하는 스트레스 완화용 또는 스트레스 질환의 예방 또는 치료용 조성물을 제공한다.
이하 본 발명을 상세히 설명한다.
본 발명에 있어서, "발효숙성노니"는 노니에 유산균을 접종하여 발효 및 숙성시킨 노니를 의미한다. 여기서, 노니는 그 부위에 제한없으나, 일 예시로 노니 열매를 사용할 수 있다.
"발효노니"는 노니 열매에 하기 7종의 유산균 중 어느 하나 이상을 접종시킨 발효물인 것일 수 있다:
(1) 락토바실러스 플란타럼 (Lactobacillus plantarum);
(2) 락토바실러스 파라카제이 (Lactobacillus paracasei);
(3) 락토바실러스 람노수스 (Lactobacillus rhamnosus);
(4) 락토바실러스 카제이 (Lactobacillus casei);
(5) 락토바실러스 퍼멘텀 (Lactobacillus fermentum);
(6) 락토바실러스 루테리 (Lactobacillus reuteri); 및
(7) 락토코커스 락티스 서브스피셔스 락티스 (Lactococcus lactis subsp. lactis).
상기 "발효노니"는 생물전환능이 우수한 유산균을 이용하여 발효시킴으로서 노니에 함유되어 있는 배당체 화합물인 스코폴린(Scopolin), 아스페룰사이드(Asperuloside)을 비배당체 화합물인 스코폴레틴(scopoletin), 디아세틸아스페루로시딕산 (deacetylasperulosidic acid, DAA) 및 아스페루로시딕산(asperulosidic acid)으로 생물전환될 수 있다.
상기 "발효노니" 추출물에 함유된 생물전환된 스코폴레틴의 함량은 1~200μg/mL 이고, 디아세틸아스페루로시딕산의 함량은 0.2~0.6㎎/mL 이며, 아스페루로시딕산의 함량은 0.096~1.41㎎/㎖ 이다. 발효숙성노니에도 발효노니에 함유된 스코폴레틴, 디아세틸아스페루로시딕산, 아스페룰로시딕산의 함량이 분석 범위 오차내에 동일하게 함유되어 있다.
본 발명의 "발효숙성노니"는 발효숙성노니 그 자체 또는 이로부터 얻어지는 추출물일 수 있다.
본 발명에 있어서, "발효숙성노니"는 발효숙성노니 그 자체 또는 이로부터 얻어지는 추출물일 수 있다. 일 예로, 본 발명의 발효숙성노니는 노니 열매에 유산균을 접종하여 발효 및 숙성시키고, 이러한 발효숙성된 발효숙성노니를 착즙하는 얻은 액은 추출물일 수 있다.
깔라만시(Calamansi)는 열대성 과일로, 동남아 지역에서 재배되는 열매이다. 상기 깔라만시 추출물은 상기 깔라만시로부터 과즙을 추출한 것을 의미하는바, 과즙의 추출방법은 종래 충분히 공지된 사항이므로, 그 상세한 설명은 생략한다. 또한, 별도로 추출물을 제조하는 것이 아닌, 시판 중인 깔라만시 원액을 구입하여 사용하는 것도 가능하다. 상기 깔라만시는 레몬과 비교했을 때 비타민C의 함량이 30배 이상이고, 클립토키사틴(cryptoxanthin) 헤스페리딘(hesperidin), 노빌레틴(nobiletin), 시네후린(cinehulin) 등이 다량 함유되어 있다.
본 명세서에서 "추출물"은 당업계에서 조추출물(Crude extract)로 통용되는 의미이지만, 광의적으로는 추출물을 추가적으로 분획(Fractionation)한 분획물의 의미도 포함한다. 즉, 발효숙성노니 또는 그 추출물 및 깔라만시 추출물은 상술한 추출용매를 이용하여 얻은 것뿐만 아니라, 여기에 정제과정을 추가적으로 적용하여 얻은 것도 포함한다. 예를 들어, 상기 추출물을 일정한 분자량 컷-오프(cut off) 값을 갖는 한외 여과막을 통과시켜 얻은 분획, 다양한 크로마토그래피(크기, 전하, 소수성 또는 친화성에 따른 분리를 위해 제작된 것)에 의한 분리 등 추가적으로 실시된 다양한 정제 방법을 통해 얻어진 분획도 본 발명의 발효숙성노니 및 깔라만시 추출물에 포함되는 것이다.
본 명세서에서 "유효성분으로 포함하는"이란 발효숙성노니 또는 그 추출물및 깔라만시 추출물의 효능이나 활성을 달성하는데 충분한 양을 포함하는 것을 의미한다. 본 발명은 천연식물재료인 발효숙성노니 및 깔라만시로부터 추출한 조성물로서, 과량 투여하여도 인체에 부작용이 없으므로 발효숙성노니 및 깔라만시 추출물이 본 발명의 조성물에 포함된 양적 상한은 통상의 기술자가 적절한 범위 내에서 선택하여 실시할 수 있다.
본 발명에 따른 발효숙성노니 및 깔라만시 추출물은 유기용매를 이용하여 추출하는데, 추출용매로서 사용될 수 있는 것은 아래와 같다.
우선 극성 용매로서 적합한 것은, (i) 물, (ii) 탄소수 1 내지 6의 알코올(바람직하게는, 메탄올, 에탄올, 프로판올, 부탄올, 노말-프로판올, 이소-프로판올, 노말-부탄올, 1-펜탄올, 2-부톡시에탄올 또는 에틸렌글리콜), (iii) 아세트산, (iv) DMFO(Dimethyl-formamide) 및 (v) DMSO(Dimethyl sulfoxide) 등이 있다.
그리고 비극성 용매로서 적합한 것은, 아세톤, 아세토나이트릴, 에틸 아세테이트, 메틸 아세테이트, 플루오로알 칸, 펜탄, 헥산, 2,2,4-트리메틸펜탄, 데칸, 시클로헥산, 시클로펜탄, 디이소부틸렌, 1-펜텐, 1-클로로부탄, 1-클로로펜탄, o-자일렌, 디이소프로필 에테르, 2-클로로프로판, 톨루엔, 1-클로로프로판, 클로로벤젠, 벤젠, 디에틸 에테르, 디에틸 설파이드, 클로로포름, 디클로로메탄, 1,2-디클로로에탄, 어닐린, 디에틸아민, 에테르, 사염화탄소, 메틸렌클로라이드, 석유에테르 및 THF 등이 있다.
본 발명은 추출용매로서, 대표적인 예시로, (a) 물, (b) 탄소수 1 내지 4의 무수 또는 저급 알코올(메탄올, 에탄올, 프로판올, 부탄올 등), (c) 상기 저급 알코올과 물과의 혼합용매, (d) 아세톤, (e) 에틸 아세테이트, (f) 클로로포름, (g) 부틸아세테이트, (h) 1,3-부틸렌글리콜, (i) 헥산 및 (j) 디에틸에테르 등을 사용하는 것이 바람직한데, 용이한 추출을 위해 물, 에탄올 또는 물과 에탄올의 혼합물을 이용하여 추출할 수 있다.
또한, 본 발명에서 이용되는 발효숙성노니 또는 그 추출물 및 깔라만시 추출물은 감압 증류 및 동결 건조 또는 분무 건조 등과 같은 추가적인 과정에 의해 분말 상태로 제조될 수 있다.
본 발명에 있어서, 상기 발효숙성노니 또는 그 추출물 및 깔라만시 추출물은 70:30 내지 99:1 중량비, 또는, 80:20 내지 95:5, 또는 70:30 내지 90:10 인 것을 특징으로 할 수 있다.
본 발명의 일 양태에 따르면, 본 발명은 발효숙성노니 또는 그 추출물 및 깔라만시 추출물을 유효성분으로 포함하는 스트레스 완화용 식품 조성물에 관한 것이다.
상기 식품 조성물은 일 예로, 건강기능 식품 조성물을 포함할 수 있으며, 상기 건강기능 식품 조성물은, 각종 식품류, 음료, 껌, 차, 비타민 복합제, 건강보조 식품류 등이 있고, 분말, 과립, 정제, 캡슐 또는 음료인 형태로 사용될 수 있다. 각 제형의 식품 조성물은 유효 성분 이외에 해당 분야에서 통상적으로 사용되는 성분들을 제형 또는 사용 목적에 따라 당업자가 어려움 없이 적의 선정하여 배합할 수 있으며, 다른 원료와 동시에 적용할 경우 상승 효과가 일어날 수 있다.
상기 조성물은 본 발명이 목적으로 하는 주 효과를 손상시키지 않는 범위 내에서 주 효과에 상승 효과를 줄 수 있는 다른 성분 등을 함유할 수 있다. 예를 들어, 물성 개선을 위하여 향료, 색소, 살균제, 산화 방지제, 방부제, 보습제, 점증제, 무기염류, 유화제 및 합성 고분자 물질 등의 첨가제를 더 포함할 수 있다.
본 발명의 건강기능식품 조성물을 식품첨가물로 사용하는 경우, 상기 발효숙성노니, 깔라만시를 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다. 유효성분의 혼합 양은 그의 사용 목적(예방, 건강 또는 치료적 처치)에 따라 적합하게 결정될 수 있다. 상기 식품의 종류에는 특별한 제한은 없다. 상기 발효숙성노니, 깔라만시를 첨가할 수 있는 식품의 예로는 육류, 소시지, 빵, 초콜릿, 캔디류, 스낵류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 수프, 음료수, 차, 드링크제, 알코올 음료 및 비타민 복합체 등이 있으며, 통상적인 의미에서의 건강기능식품을 모두 포함한다.
본 발명의 조성물을 건강 음료로 사용할 경우, 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상술한 천연 탄수화물은 포도당, 과당과 같은 모노사카라이드, 말토스, 슈크로스와 같은 디사카라이드, 텍스트린, 사이클로텐스트린과 같은 폴리사카라이드, 자일리톨, 소르비톨, 에리트리톨 등의 당알콜이다. 감미제로서는 타우마린, 스테비아 추출물과 같은 천연 감미제나, 사카린 또는 아스파르탐과 같은 합성 감미제 등을 사용할 수 있다.
본 발명은, 유산균을 이용하여 노니를 발효 및 숙성한 발효숙성노니를 수득하는 단계; 및 상기 발효숙성노니에 깔라만시를 추가하여 혼합하는 단계를 포함하는, 발효숙성노니 및 깔라만시를 포함하는 조성물의 제조방법을 제공한다.
본 발명의 일 양태에 따르면, 본 발명은,
(a) 유산균을 이용하여 노니를 발효 및 숙성한 발효숙성노니를 수득하는 단계; 및 (b) 상기 발효숙성노니에 깔라만시를 추가하여 숙성하는 단계를 포함하는, 발효숙성노니 또는 그 추출물 및 깔라만시 추출물을 포함하는 조성물의 제조방법을 제공한다.
본 발명의 일 양태에 따르면, 본 발명은,
(a) 노니 열매에 유산균을 접종하여 발효 및 숙성시켜 생물전환된 발효 노니를 제조하는 단계; (b) 상기 발효숙성노니를 착즙시키는 단계; 및 (c) 상기 발효숙성노니에 깔라만시를 추가하여 혼합하는 단계를 포함하는, 스트레스 완화용 조성물의 제조방법을 제공한다.
상기 발효 및 숙성은 35 내지 40℃에서 48 내지 168 시간 동안 이루어지는 것일 수 있다.
상기 숙성은 발효 과정에서 생성되는 발효액을 일정 시간 간격으로 발효물의 상부에 끼얹는 방식으로 실시하는 것일 수 있다.
상기 생물전환된 발효 노니는 노니 열매에 상기한 7종의 유산균 중 어느 한 가지 이상을 접종시킨 발효물일 수 있다.
본 발명의 일 양태에 따르면, 본 발명은 발효숙성노니 또는 그 추출물 및 깔라만시 추출물을 유효성분으로 포함하는 스트레스 완화용 또는 스트레스 질환의 예방 또는 치료용 약학적 조성물에 관한 것이다.
본 발명에 있어서, 스트레스성 호르몬인 코티졸의 분비가 억제됨을 확인함으로써, 스트레스에 의한 행동 변화 또한 억제되므로, 우울증, 불안장애, 피로증후군, 수면장애, 공황장애, 기억력 감퇴, 무기력증, 불면증, 파킨슨 질환, 및 알츠하이머 질환 등의 스트레스성 질환의 예방 또는 치료하는 효과를 나타냄을 알 수 있다.
따라서, 본 발명은 발효숙성노니 또는 그 추출물 및 깔라만시 추출물을 유효성분으로 포함하는 스트레스 질환의 예방 또는 치료용 약학적 조성물에 관한 것이다.
본 발명에 따른 약학적 조성물은 유효성분을 단독으로 함유하거나 약학적으로 허용되는 담체와 함께 적합한 형태로 제형화 될 수 있으며, 부형제 또는 희석제를 추가로 함유할 수 있다. 상기에서 '약학적으로 허용되는'이란 생리학적으로 허용되고 인간에게 투여될 때, 통상적으로 위장 장애, 현기증 등과 같은 알레르기 반응 또는 이와 유사한 반응을 일으키지 않는 비독성의 조성물을 말한다.
약학적으로 허용되는 담체로는 예컨대, 경구 투여용 담체 또는 비경구 투여용 담체를 추가로 포함할 수 있다. 경구 투여용 담체는 락토스, 전분, 셀룰로스 유도체, 마그네슘 스테아레이트, 스테아르산 등을 포함할 수 있다. 아울러, 펩티드 제제에 대한 경구투여용으로 사용되는 다양한 약물전달물질을 포함할 수 있다. 또한, 비경구 투여용 담체는 물, 적합한 오일, 식염수, 수성 글루코오스 및 글리콜 등을 포함할 수 있으며, 안정화제 및 보존제를 추가로 포함할 수 있다. 적합한 안정화제로는 아황산수소나트륨, 아황산나트륨 또는 아스코르브산과 같은 항산화제가 있다. 적합한 보존제로는 벤즈알코늄 클로라이드, 메틸-또는 프로필-파라벤 및 클로로부탄올이 있다. 본 발명의 약학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현택제 등을 추가로 포함할 수 있다. 그 밖의 약학적으로 허용되는 담체 및 제제는 다음의 문헌에 기재되어 있는 것을 참고로 할 수 있다(Remington's Pharmaceutical Sciences, 19th ed., Mack Publishing Company, Easton, PA, 1995).
본 발명의 조성물은 인간을 비롯한 포유동물에 어떠한 방법으로도 투여할 수 있다. 예를 들면, 경구 또는 비경구적으로 투여할 수 있다. 비경구적인 투여방법으로는 이에 한정되지는 않으나, 정맥내, 근육내, 동맥내, 골수내, 경막내, 심장내, 경피, 피하, 복강내, 비강내, 장관, 국소, 설하 또는 직장내 투여일 수 있다.
본 발명의 약학적 조성물은 상술한 바와 같은 투여 경로에 따라 경구 투여용 또는 비경구 투여용 제제로 제형화 할 수 있다.
경구 투여용 제제의 경우에 본 발명의 조성물은 분말, 과립, 정제, 환제, 당의정제, 캡슐제, 액제, 겔제, 시럽제, 슬러리제, 현탁액 등으로 당업계에 공지된 방법을 이용하여 제형화될 수 있다. 예를 들어, 경구용 제제는 활성성분을 고체 부형제와 배합한 다음 이를 분쇄하고 적합한 보조제를 첨가한 후 과립 혼합물로 가공함으로써 정제 또는 당의정제를 수득할 수 있다. 적합한 부형제의 예로는 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨 및 말티톨 등을 포함하는 당류와 옥수수 전분, 밀 전분, 쌀 전분 및 감자 전분 등을 포함하는 전분류, 셀룰로즈, 메틸 셀룰로즈, 나트륨 카르복시메틸셀룰로오즈 및 하이드록시프로필메틸-셀룰로즈 등을 포함하는 셀룰로즈류, 젤라틴, 폴리비닐피롤리돈 등과 같은 충전제가 포함될 수 있다. 또한, 경우에 따라 가교결합 폴리비닐피롤리돈, 한천, 알긴산 또는 나트륨 알기네이트 등을 붕해제로 첨가할 수 있다. 나아가, 본 발명의 약학적 조성물은 항응집제, 윤활제, 습윤제, 향료, 유화제 및 방부제 등을 추가로 포함할 수 있다.
비경구 투여용 제제의 경우에는 주사제, 크림제, 로션제, 외용연고제, 오일제, 보습제, 겔제, 에어로졸 및 비강 흡입제의 형태로 당업계에 공지된 방법으로 제형화할 수 있다. 이들 제형은 모든 제약 화학에 일반적으로 공지된 처방서인 문헌(Remington's Pharmaceutical Science, 19th ed., Mack Publishing Company, Easton, PA,1995)에 기재되어 있다.
본 발명의 조성물의 총 유효량은 단일 투여량(single dose)으로 환자에게 투여될 수 있으며, 다중 투여량(multiple dose)으로 장기간 투여되는 분할 치료 방법(fractionated treatment protocol)에 의해 투여될 수 있다. 본 발명의 약학적 조성물은 질환의 정도에 따라 유효성분의 함량을 달리할 수 있다. 바람직하게 본 발명의 약학적 조성물의 바람직한 전체 용량은 1일당 환자 체중 1㎏ 당 약 0.01㎍ 내지 10,000mg, 바람직하게는 0.1㎍ 내지 500mg, 가장 바람직하게는 100mg 내지 500mg일 수 있다. 그러나 상기 약학적 조성물의 용량은 제제화 방법, 투여 경로 및 치료 횟수뿐만 아니라 환자의 연령, 체중, 건강 상태, 성별, 질환의 중증도, 식이 및 배설율등 다양한 요인들을 고려하여 환자에 대한 유효 투여량이 결정되는 것이므로, 이러한 점을 고려할 때 당 분야의 통상적인 지식을 가진 자라면 본 발명의 조성물의 적절한 유효 투여량을 결정할 수 있을 것이다. 본 발명에 따른 약학적 조성물은 본 발명의 효과를 보이는 한 그 제형, 투여 경로 및 투여 방법에 특별히 제한되지 아니한다.
본 발명의 일 양태에 따르면, 본 발명은 발효숙성노니 또는 그 추출물 및 깔라만시 추출물을 포함하는 스트레스 완화용 또는 스트레스 질환의 예방 또는 치료용 의약외품 조성물에 관한 것이다.
본 발명에서 사용되는 용어 "의약외품"은 사람이나 동물의 질병을 진단, 치료, 경감, 처치 또는 예방할 목적으로 사용하는 물품 중 기구, 기계 또는 장치가 아닌 것 및 사람이나 동물의 구조와 기능에 약리학적 영향을 줄 목적으로 사용하는 물품 중 기구, 기계 또는 장치가 아닌 것을 제외한 물품을 의미하며, 일 구체예로 내복용 제제를 포함할 수 있으나 이에 제한되는 것이 아니며, 의약외품의 제제화 방법, 용량, 이용방법, 구성성분 등은 기술분야에 공지된 통상의 기술로부터 적절히 선택될 수 있다.
본 발명의 의약외품 조성물에는 상기 성분 외에 필요에 따라 약학적으로 허용 가능한 담체, 부형제 또는 희석제를 더욱 포함할 수 있다. 상기 약학적으로 허용 가능한 담체, 부형제 또는 희석제는 본 발명의 효과를 해하지 않는 한 제한되지 않으며, 예를 들어 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제, 윤활제, 감미제, 방향제, 보존제 등을 포함할 수 있다.
본 발명의 일 양태에 따르면, 본 발명은 발효숙성노니 또는 그 추출물 및 깔라만시 추출물을 포함하는 스트레스 완화용 또는 스트레스 질환의 예방 또는 치료용 사료 조성물에 관한 것이다.
본 발명에 있어서, 상기 사료 조성물은, 통상의 사료 형태로 제제화될 수 있고, 알려진 사료성분을 함꼐 포함할 수 있다. 또한, 사용되는 사료에 첨가제 형태로 가미되는, 사료첨가제로서도 사용될 수 있다. 본 발명의 사료첨가제는 사료관리법상의 보조사료에 해당하며, 탄산수소나트륨(중조), 벤토나이트 (bentonite), 산화마그네슘, 복합광물질 등의 광물질제제, 아연, 구리, 코발트, 셀레늄 등의 미량 광물질인 미네랄제제, 케로틴, 비타민 E, 비타민 A, D, E, 니코틴산, 비타민 B 복합체 등의 비타민제, 메티오닌, 리이산 등의 보호아미노산제, 지방산 칼슘염 등의 보호지방산제, 생균제(유산균제), 효모배양물, 곰팡이 발효물 등의 생균, 효모제 등이 추가로 포함될 수 있다.
본 발명의 사료 또는 사료첨가제는 포유류, 가금 및 어류를 포함하는 다수의 동물식이에 적용할 수 있다.
본 발명에 따른 스트레스 완화용 조성물에 포함된 발효숙성노니 또는 그 추출물 및 깔라만시추출물은 천연 약재 및 식품 조성물을 이용하여 스트레스를 완화시키고, 스트레스로 인해 발생되는 증상(질환)들을 예방하거나 부작용없이 효율적으로 개선시키는 효과가 있다.
또한, 상기 발효숙성노니는 노니 열매를 발효시키는 공정에서 소화 흡수를 향상시킬 수 있는 생물전환된 성분(스코폴레틴, 디아세틸아스페르로시딕산 및 아스페루로시딕산)이 분비되어, 상대적으로 소화력이 떨어지는 성인, 노인들로 하여금 소화력을 촉진시키고 배변활동을 촉진시키는 한편, 천연 원료이므로 독성과 부작용이 없어 안심하고 섭취할 수 있는 효과가 있다.

도 1은 강제수영 시험에서 본 발명의 대조군(control), 단일군 및 복합 조성물을 섭취에 의한 시험동물의 부동시간을 나타내는 그래프이다.

이하, 본 발명을 하기 실시예를 통하여 보다 상세하게 설명한다. 그러나 이들 실시예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예만으로 한정되는 것은 아니다.
실시예 1 : 스트레스 완화용 조성물 제조
본 발명에 따른 조성물은 다음과 같은 과정을 통하여 제조되었다.
1.1 : 발효숙성노니 제조
노니 열매 690kg에 복합 유산균(7종복합유산균 AON1805, (주)락토메이슨)을 접종하여, 37 ℃에서 45일 이상 발효시켰다. 발효 시작일로부터 7일마다 발효액을 10 L씩 받아 노니 열매 위쪽으로 옮겨 뿌려주면서, 발효 및 숙성을 실시하였다. 이때, 사용한 복합 유산균(7종복합유산균 AON1805, (주)락토메이슨)의 조성은 하기 표 1과 같다.
숙성이 완료된 후, 상기 발효숙성노니를 착즙하여 고체를 제거하고, 발효숙성노니추출물을 얻었다.

1.2 : 깔라만시 추출물 제조
본 실험예에서 사용한 깔라만시는 시중에 파는 깔라만시를 구매하여 사용하였다. 냉동 또는 건조 깔라만시는, 시중에 판매하는 냉동 또는 건조 깔라만시를 구입하여 사용하였다.
상기 냉동 깔라만시는 분쇄, 착즙하여 추출물을 제조하였다. 건조 깔라만시는 100g에 중량부의 10배 물을 가한다음 50℃에서 8시간 동안 2회 추출한 다음, 추출액을 여과지로 여과하였다. 여액을 37∼40℃의 수욕 상에서 rotary vacuum evaporator(EYELA A-1000S, Tokyo Rikakikai Co., Tokyo, Japan)를 사용하여 물을 제거하여 추출물을 제조하였으며, 이를 시료로 사용하였다.
상기 깔라만시의 시료별 추출 용매별 추출수율은 아래와 같다.

1.3 : 발효숙성노니 추출물과 깔라만시 추출물을 이용한 복합물 제조
상기 1.1 내지 1.2에서 제조한 발효숙성노니 추출물, 깔라만시 추출물의 배합비를 달리하여 혼합한 후, 15일간 숙성하여 복합물을 제조하였다.

실험예 1 : 본 발명의 조성물이 스트레스완화에 미치는 영향 확인
상기 실시예 1.1 내지 1.2에서 제조한 단일 추출물과 실시예 1.3에서 제조한 복합물이 스트레스완화 능력에 미치는 영향을 확인 하였다.
(1) 생쥐 시험대상
4주량의 ICR 수컷 생쥐{(주)오리엔트바이오, 대한민국}를 1주일간 적응기간을 거친 뒤 12시간 단위로 조명의 점등 및 소등이 반복되고, 실내온도 18 내지 23℃, 습도 60%를 유지하는 공간에서 1주간 적응시킨 후, 실험동물을 각각 8마리씩 실험군을 다음과 같이 분류하였다. 사료는 고형 사료를 공급하였고, 스트레스를 유도하는 과정 외에는 사료 및 급수에 제한을 두지 않았다.
실험군 1(정상 대조군, 이하 Normal 표시), 실험군 2(음성 대조군, 이하 Control 표시)은 생리식염수를 경구투여 하였다. 한편, 실험군 3(발효숙성노니 100mg/kg 투여군, 이하 FN 표시), 실험군 4(건조 깔라만시 추출물 100mg/kg 투여군, 이하 DCE 표시), 실험군 5(냉동 깔라만시 추출물 100mg/kg 투여군, 이하 FCE 표시), 실험군 6(복합물 1 100mg/kg 투여군, 이하 C 1 표시), 실험군 7(복합물 2 100mg/kg 투여군, 이하 C 2 표시), 실험군 8(복합물 3 100mg/kg 투여군, 이하 C 3 표시), 실험군 9(복합물 4 100mg/kg 투여군, 이하 C 4 표시), 실험군 10(복합물 5 100mg/kg 투여군, 이하 C 5 표시), 실험군 11(복합물 6 100mg/kg 투여군, 이하 C 6 표시) 으로 설정하였으며, 시험물질은 1회/1일 총 4주간 투여하였다.
(2) 스트레스 유도
위 시험대상의 음성대조군은 생리식염수를 실험군 1, 실험군 2, 실험군 3, 실험군 4, 실험군 5 및 실험군 6은 단일추출물 또는 복합물 투여 7일 후부터 4주간 매일 스트레스를 유도한다. 스트레스 유발 방법은 Willner 등(Wilner et al., Reduction fo sucrose preference by chronic unpredictable mild stress, 1987)의 방법을 수정하여 사용하는데, 구체적으로 단식, 단식 이후의 식이 제한(적은 양의 사료 공급), 단수, 단수 이후의 빈 물병 제공, 기울어진 사육장, 한 사육장 안에 다수의 실험동물 사육, 반짝이는 불빛, 차가운 방, 지속적인 불빛 등과 같은 예상치 못한 다양한 정신적 스트레스 상황을 조성하여 스트레스를 유발하였다.
(3) 강제수영 시험(Forced Swimming test)
시험동물 대상으로 강제수영 시험을 실시하였다. 원통형 투명 수조(지름 14 cm, 높이 20 cm)에 수심이 15 cm가 되도록 23~25℃의 물을 채운 후, 시험동물을 한 마리씩 수조 안에 넣는다. 모든 시험동물을 6분간 원통형 수조에서 강제로 수영하도록 하였다. 시작 후 2분은 적응시간으로 간주하고, 종료 전 4분간 실험동물이 원통형 수조 안에서 움직이지 않는 총 시간(이하 '부동시간'이라 한다)을 측정하여 기록하였다.
구체적으로 부동시간이라 함은 수면 위로 머리를 드는 정도의 작은 움직임을 제외하고는 어떠한 움직임도 없는 상태(부동성)를 의미한다. 부동성은 우울증의 절망 지표 중 하나로서, 스트레스 실험 동물에서 흔히 볼 수 있다.
도 1에서 보는 바와 같이 음성대조군(Control)은 정상대조군(Normal)에 비해 부동시간이 긴 것으로 조사되었다. 이에 반해 실험군 3은 정상대조군에 비해서는 다소 부동시간이 길지만, 음성대조군에 비해서는 부동시간이 80% 수준으로 짧은 것을 알 수 있다. 반면, 실험군 4 및 5는 음성대조군과 비교시 유의적인 차이를 보였으며, 발효숙성노니와 깔라만시 복합물이 투여된 실험군(6, 7, 8, 9, 10, 및 11)은 음성대조군과 대비 유의성 있게 절반 수준으로 부동시간이 감소되는 것으로 조사되었다. 따라서 발효숙성노니 또는 깔라만시 단일 추출물보다는 복합물이 부동시간 감소에 더욱 효과가 있는 것을 알 수 있다.
(4) 꼬리 매달기 시험(Tail Suspension Test)
시험동물 대상으로 꼬리 매달기 시험을 2회 반복 실시하였다. 시험동물을 바닥으로부터 15 cm 높이에서 꼬리의 1 cm 끝을 잡고 6분간 실험동물을 거꾸로 매달아 시험동물의 움직임을 관찰한다. 초기 2분은 적응기간으로 간주 하고, 종료 전 4분 동안 실험동물이 움직이지 않은 시간을 측정하여 기록하였다.
도 2에 도시된 바와 같이 음성대조군(Control)은 꼬리를 매달아 거꾸로 된 상태에서의 부동시간이 정상대조군(Normal)에 비해 긴 것으로 조사되었다. 이는 정신적 스트레스로 인해 부동시간이 증가된 것으로, 좌절감이 증가된 것을 의미한다.
실험군 3(FN)은 음성대조군(Control)에 비해 부동시간이 82% 수준으로 짧았고 비교적 몸을 움직이려고 하였으나, 실험군 4는 음성대조군에 비해 부동시간이 92% 수준으로 유의성 있는 차이를 보였다.
또한 발효숙성노니 추출물 (FN)과 깔라만시 추출물(DCE & FCE) 추출물을 일정비율로 혼합하여 제조한 복합물(1, 2, 3, 4, 5 및 6) 실험군은 모두 음성대조군 대비 유의성 있게 낮은 수준으로 조사되었다. 이를 통해 발효숙성노니 및 깔라만시 추출물이 일정비율 혼합된 복합물은 스트레스로 인한 무기력증 및 좌절감 완화에 더욱 효과가 있는 것을 알 수 있다.
실험예 2 : 부동화 스트레스유발 생쥐에서 조성물 섭취에 의한 혈중 IgA 수준 변화 시험
좁은 틀에 매일 2시간씩 21일간 움직이지 못하게 하여 스트레스를 유발시킨, 부동화 스트레스유발 8주령 C57BL/6 마우스에, 발효숙성노니 추출물 (FN), 깔라만시 추출물(CE), 발효숙성노니 추출물 (FN)과 깔라만시 추출물(DCE & FCE) 추출물을 일정비율로 혼합하여 제조한 복합물(1, 2, 3, 4, 5 및 6)을 매일 100mg/kg 씩 경구 투여하였다.
도 3에 나타난 바와 같이, 혈청중 IgA 수준을 측정한 결과로, 발효숙성노니 추출물 투여군 (FN), 복합물 1, 복합물 4 투여군이 대조군 (control)에 비하여 통계학적으로 유의성 있게 IgA 수준이 증가를 나타났다.
특히, 복합물 4 투여군에서 가장 높은 IgA 수준의 증가를 나타내었다.
실험예 3: 부동화 스트레스유발 생쥐에서 복합물 섭취에 의한 체중변화 시험
발효숙성노니 추출물 (FN), 깔라만시 추출물(CE), 발효숙성노니 추출물 (FN)과 깔라만시 추출물(DCE & FCE) 추출물을 일정비율로 혼합하여 제조한 복합물(1, 2, 3, 4, 5 및 6)을 부동화 스트레스를 매일 2시간씩 21일간 유발 8주령 C57BL/6에 매일 100 mg/kg씩 경구투여하였다.
도 4에 나타난 바와 같이, 체중변화를 측정한 결과 부동화 대조군 (control)은 정상군(Normal)에 비하여 체중 감소가 나타났고, 발효숙성노니 추출물 (FN), 복합물 1 및 4 투여군이 부동화 대조군 (control)에 비하여 체중감소가 적게 나타났다.
특히, 복합물 1 및 4에서 가장 낮은 체중감소를 나타내었다.
실험예 4: 부동화 스트레스유발 생쥐에서 복합물 섭취에 의한 혈중 코티졸(Cortisol)과 세로토닌 (Serotonin)생산량 변화 측정 시험
발효숙성노니 추출물 (FN), 깔라만시 추출물(CE), 발효숙성노니 추출물 (FN)과 깔라만시 추출물(DCE & FCE) 추출물을 일정비율로 혼합하여 제조한 복합물(1, 2, 3, 4, 5 및 6)을 부동화 스트레스를 매일 2시간씩 21일간 유발 8주령 C57BL/6에 매일 100mg/kg씩 경구투여 하였다.
혈청중 행복의 감정을 느끼게 해주는 호르몬인 세로토닌 (Serotonin) 수준을 측정한 결과, 발효숙성노니 추출물 (FN) 100 mg/kg (p<0.05) 그리고 발효숙성노니 추출물 (FN), 복합물 1 및 복합물 4 투여군이 부동화 대조군 (Control)에 비하여 통계학적으로 유의성 있게 세로토닌(Serotonin) 수준이 증가를 나타났다 (도 5).
혈청중 스트레스 호르몬인 코르티솔(Cortisol) 수준을 측정한 결과 발효숙성노니 추출물 (FN) 그리고 복합물 1 및 4 투여군에서 부동화 대조군 (control)에 비하여 통계학적으로 유의성 있게 코르티솔(Cortisol) 수준이 감소를 나타났다 (도 6).
특히, 복합물 1 및 4에서 가장 높은 세로토닌 수준의 증가와 가장 높은 코르티솔 수준의 감소를 나타내었다.
이상의 설명으로부터, 본 발명이 속하는 기술분야의 당업자는 본 발명이 그 기술적 사상이나 필수적 특징을 변경하지 않고서 다른 구체적인 형태로 실시될 수 있다는 것을 이해할 수 있을 것이다. 이와 관련하여, 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적인 것이 아닌 것으로 이해해야만 한다. 본 발명의 범위는 상기 상세한 설명보다는 후술하는 특허 청구범위의 의미 및 범위 그리고 그 등가 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.
Claims (12)
- 발효숙성노니 또는 그 추출물; 및 깔라만시 추출물을 유효성분으로 포함하는, 스트레스 완화용 또는 스트레스 질환의 예방 또는 치료용 식품 조성물.
- 제1항에 있어서,상기 발효숙성노니 또는 그 추출물; 및 깔라만시 추출물은 70:30 내지 99:1 중량비인, 스트레스 완화용 또는 스트레스 질환의 예방 또는 치료용 식품 조성물.
- 제1항에 있어서,상기 발효숙성노니 또는 그 추출물은 노니를 유산균을 이용하여 발효 및 숙성하여 얻어지는 발효숙성노니인, 스트레스 완화용 또는 스트레스 질환의 예방 또는 치료용 식품 조성물.
- 제1항 내지 제3항 중 어느 한 항에 있어서,상기 식품 조성물은 건강기능식품인, 스트레스 완화용 또는 스트레스 질환의 예방 또는 치료용 식품 조성물.
- 제1항에 있어서,상기 식품 조성물은 분말, 과립, 환, 정제, 캡슐, 캔디, 시럽 및 음료 중에서 선택된 어느 하나의 제형인, 스트레스 완화용 또는 스트레스 질환의 예방 또는 치료용 식품 조성물.
- 발효숙성노니 또는 그 추출물; 및 깔라만시 추출물을 유효성분으로 포함하는, 스트레스 완화용 또는 스트레스 질환의 예방 또는 치료용 약학적 조성물.
- 발효숙성노니 또는 그 추출물; 및 깔라만시 추출물을 유효성분으로 포함하는 스트레스 완화용 또는 스트레스 질환의 예방 또는 치료용 의약외품 조성물.
- 발효숙성노니 또는 그 추출물; 및 깔라만시 추출물을 유효성분으로 포함하는 스트레스 완화용 사료 조성물.
- (a) 유산균을 이용하여 노니를 발효 및 숙성한 발효숙성노니를 수득하는 단계; 및 (b) 상기 발효숙성노니에 깔라만시를 추가하여 숙성하는 단계를 포함하는, 발효숙성노니 또는 그 추출물 및 깔라만시 추출물을 포함하는 조성물의 제조방법.
- 제9항에 있어서,상기 (a)단계 다음에, 얻어진 발효숙성노니를 착즙하여 발효숙성노니 추출물을 얻는 단계를 더 포함하는, 조성물의 제조방법.
- 제9항의 제조방법에 따라 얻어진 조성물.
- 제11항에 있어서,상기 조성물은 스트레스 완화능을 가진 조성물.
