Description
洗眼用組成物に関する。
従来、洗眼用カップ(アイカップ)を用いた洗眼方法や点眼による洗眼方法が知られている。洗眼用カップを用いた洗眼方法の一例として、洗眼用カップに洗眼液を適量(例えば5mL程)注ぎ、次いで、該カップを片側の眼周辺に押し当てて該カップ内の洗眼液と眼とを接触させ、数回まばたきを行うことにより洗眼する方法が挙げられる。点眼による洗眼方法の一例として、洗眼液を眼に直接点眼して洗眼する方法が挙げられる(例えば、特許文献1)。
アラントインは、角膜修復作用等を有する成分の一種として知られている。しかし、アラントインは加水分解されやすいため安定性が乏しく、特にpH5付近以上で分解が進みやすくなり、pH6では殆ど分解されるともいわれている。一方、前述のような洗眼液はpH5以上であることが多いため、洗眼液においてアラントインの分解を抑制することは重要である。
そこで、本開示は、アラントインの分解が抑制された洗眼用組成物を提供することを目的とする。
本発明者は、前記課題に鑑み鋭意検討を行ったところ、アラントインを含有する洗眼用組成物においてヒプロメロースを配合することにより、アラントインの分解を抑制できることを見出した。本発明は、かかる知見に基づいて更に検討を重ねることにより完成されたものである。すなわち、本開示は、次に掲げる発明を包含する。
項1.アラントイン及びヒプロメロースを含有する洗眼用組成物。
項2.更に、清涼化剤を含有する、項1に記載の洗眼用組成物。
項3.pHが5~7である、項1または2に記載の洗眼用組成物。

アラントインを含有する洗眼用組成物においてヒプロメロースを配合することにより、アラントインの分解を抑制できる。このことから、該洗眼用組成物においてアラントインの安定性を向上することができる。
以下、本開示に包含される実施形態について更に詳細に説明する。
本開示に包含される洗眼用組成物は、アラントイン及ヒプロメロースを含有する。
アラントインは、化学式C4H6N4O3で表される公知の化合物である。該洗眼用組成物中、アラントインの含有量は、本開示の効果が得られる限り制限されないが、好ましくは0.003~0.05w/v%、より好ましくは0.005~0.04w/v%、更に好ましくは0.008~0.03w/v%、特に好ましくは0.015~0.03w/v%が例示される。
ヒプロメロースは、従来公知であり、ヒドロキシプロピルメチルセルロースとも称される。該洗眼用組成物中、ヒプロメロースの含有量は、本開示の効果が得られる限り制限されないが、該組成物におけるアラントインの分解を一層抑制する観点から、好ましくは0.01~1w/v%、より好ましくは0.05~0.8w/v%、更に好ましくは0.08~0.5w/v%、特に好ましくは0.15~0.5w/v%が例示される。また、ヒプロメロースの含有量は、本開示の効果が得られる限り制限されないが、該洗眼用組成物中、アラントイン1質量部あたり、好ましくは0.01~20質量部、より好ましくは0.1~20質量部、更に好ましくは1~20質量部、特に好ましくは3.5~20質量部が例示される。
該洗眼用組成物は、更に清涼化剤を含有してもよい。清涼化剤としてメントール、カンフル、ボルネオール等が例示される。清涼化剤においてd体、l体、dl体が存在する場合、その別は問わず、いずれであってもよい。清涼化剤は1種単独で使用してもよく、2種以上を組み合わせて使用してもよい。清涼化剤を含有する場合、その含有量は制限されないが、該組成物におけるアラントインの分解を一層抑制する点から、該組成物中、好ましくは0.0001~0.05w/v%、より好ましくは0.0005~0.01w/v%、更に好ましくは0.001~0.008w/v%が例示される。また、清涼化剤の含有量は、本開示の効果が得られる限り制限されないが、該洗眼用組成物中、アラントイン1質量部あたり、好ましくは0.005~5質量部、より好ましくは0.01~1質量部、更に好ましくは0.05~1質量部が例示される。
本開示の洗眼用組成物は、点眼や洗眼における使用に許容可能な任意の他の成分を更に含有してもよい。該他の成分として、本開示を制限するものではないが、緩衝剤、安定剤、等張化剤、溶剤、pH調整剤、薬効成分、防腐剤等が例示される。該他の成分は、本開示の効果を妨げないことを限度として、目的等に応じて適宜選択すればよく、1種単独で使用してもよく、2種以上を組み合わせて使用してもよく、その配合量も適宜決定すればよい。
該他の成分の一例として緩衝剤を挙げると、緩衝剤としてホウ酸、ホウ砂等が例示される。緩衝剤は、1種単独で使用してもよく、2種以上を組み合わせて使用してもよい。例えば、該組成物がホウ酸を含有する場合、ホウ酸の含有量は、本開示の効果を妨げない限り制限されず、該組成物中、好ましくは0.1~2.5w/v%が例示され、より好ましくは0.5~2w/v%が例示される。例えば、該組成物がホウ砂を含有する場合、ホウ砂の含有量は、本開示の効果を妨げない限り制限されず、該組成物中、好ましくは0.005~0.5w/v%が例示され、より好ましくは0.005~0.05w/v%が例示される。該組成物は、ホウ酸やホウ砂以外の緩衝剤を含有してもよく、含有しなくてもよい。
該他の成分の一例として安定剤を挙げると、安定剤としてポリソルベート(ポリソルベート80、ポリソルベート60等)、ポリオキシエチレン(POE)硬化ヒマシ油(POE(80)硬化ヒマシ油、POE(60)硬化ヒマシ油等、括弧内の数字は酸化エチレンの平均付加モル数を意味する)、エデト酸ナトリウム(水和物を含む)等が例示される。これらは1種単独で使用してもよく、2種以上を組み合わせて使用してもよい。該組成物が安定剤を含有する場合、その含有量は、本開示の効果を妨げない限り制限されず、該組成物中、好ましくは0.01~0.5w/v%が例示され、より好ましくは0.05~0.5w/v%が例示される。該組成物が安定剤を含有する場合、例えばポリソルベート、ポリオキシエチレン硬化ヒマシ油及びエデト酸ナトリウムから選択されるいずれか1種のみ、または2種のみを含有してもよく、また、例えば、ポリソルベートを含有する場合、ポリオキシエチレン硬化ヒマシ油及び/またはエデト酸ナトリウムを配合しないで調製することもできる。
該他の成分の一例として薬効成分を挙げると、薬効成分として、アミノ酸類、抗炎症剤(消炎剤)、角膜表層保護剤、抗ヒスタミン剤、ビタミン類等が挙げられる。これらは1種単独で使用してもよく、2種以上を組み合わせて使用してもよい。
本開示を制限するものではないが、例えばアミノ酸類として、グルタミン酸及びその塩(グルタミン酸ナトリウム等)等のアミノ酸及びその塩;タウリンなどのアミノ酸類似物等が例示される。これらは1種単独で使用してもよく、2種以上を組み合わせて使用してもよい。本開示の洗眼用組成物は、アミノ酸及び/またはその塩を配合しないで調製することもできる。ここでいうアミノ酸とは、アミノ基とカルボキシ基の両方の官能基を持つ蛋白質の構成ユニットとなりえるアミノ酸を意味し、例えばL-アスパラギン酸等が含まれる。また、本開示の洗眼用組成物は、アミノ酸類似物を配合しないで調製することもできる。
本開示を制限するものではないが、例えば抗炎症剤として、硫酸亜鉛、乳酸亜鉛、イプシロン-アミノカプロン酸、グリチルリチン酸及びその塩(グリチルリチン酸二カリウム等)等が例示される。これらは1種単独で使用してもよく、2種以上を組み合わせて使用してもよい。本開示を制限するものではないが、該組成物は好ましくは硫酸亜鉛及び/または乳酸亜鉛を含有しない。また、該組成物は、イプシロン-アミノカプロン酸やグリチルリチン酸及びその塩を配合しないで調製することもできる。
本開示を制限するものではないが、例えば角膜表層保護剤として、コンドロイチン硫酸及びその塩、ヒアルロン酸及びその塩等が例示され、塩として好ましくはナトリウム塩等のアルカリ金属塩等が例示される。これらは1種単独で使用してもよく、2種以上を組み合わせて使用してもよい。本開示を制限するものではないが、コンドロイチン硫酸やその塩の分子量として、好ましくは重量平均分子量が0.5万~4.5万程度、より好ましくは1.5万~3万程度が例示される。また、本開示を制限するものではないが、該組成物は、コンドロイチン硫酸及びその塩ならびに/またはヒアルロン酸及びその塩を配合せず調製することもできる。
本開示を制限するものではないが、例えば抗ヒスタミン剤として、ジフェンヒドラミン及びその塩(ジフェンヒドラミン塩酸塩等)、クロルフェニラミン及びその塩(マレイン酸クロルフェニラミン等)等が例示される。これらは1種単独で使用してもよく、2種以上を組み合わせて使用してもよい。本開示を制限するものではないが、該組成物は、ジフェンヒドラミン及びその塩ならびに/またはクロルフェニラミン及びその塩を配合せず調製することもできる。
本開示の組成物が薬効成分を含有する場合、本開示の効果を妨げない限り、薬効成分の含有量は制限されず、該組成物中、薬効成分の含有量は好ましくは0.01~5w/v%、より好ましくは0.025~4w/v%、更により好ましくは0.1~3w/v%が例示される。なお、前述のアラントインは薬効成分としても公知であるが、該成分は、本開示では薬効成分の説明には包含されず、従って、薬効成分の該含有量にアラントインの含有量は含まれない。
該他の成分の一例として防腐剤を挙げると、防腐剤として第4級アンモニウム塩系防腐剤、パラオキシ安息香酸エステル類、クロロブタノール、ソルビン酸類等が例示される。第4級アンモニウム系防腐剤として、ベンザルコニウム塩化物、ベンゼトニウム塩化物等が例示される。パラオキシ安息香酸エステル類として、パラオキシ安息香酸メチル、パラオキシ安息香酸プロピル、パラオキシ安息香酸ブチル等のパラオキシ安息香酸エステル、パラオキシ安息香酸エステルの塩(ナトリウム塩等のアルカリ金属塩等)等が例示される。ソルビン酸類として、ソルビン酸、ソルビン酸の塩(ナトリウム塩、カリウム塩等のアルカリ金属塩等)等が例示される。これらは1種単独で使用してもよく、2種以上を組み合わせて使用してもよい。本開示を制限するものではないが、該組成物は好ましくは防腐剤を配合しない。本開示を制限するものではないが、本開示の洗眼用組成物は、後述の実施例から理解できる通り、防腐剤を配合せず調製することもできる。
本開示の洗眼用組成物は、アラントイン、ヒプロメロース、必要に応じて清涼化剤、他の成分を混合することにより製造することができる。
本開示の洗眼用組成物のpHは、点眼剤や洗眼剤として使用可能な範囲であれば制限されないが、室温(25℃)でpH5~8、該組成物の防腐性を向上させる観点から、好ましくはpH5.5~7、より好ましくはpH5.5~6.5、更に好ましくはpH5.5~6、特に好ましくはpH5.5~5.8が例示される。pHは、卓上型pHメーターF-52(株式会社堀場製作所製)で測定される。
本開示の洗眼用組成物の形態は、室温で液状であり、従来公知の方法と同様に眼を洗浄できる限りその使用方法は制限されず、通常、洗眼カップ(アイカップ)を用いた洗眼に使用されるか、点眼による洗眼に使用される。このため、該組成物は容器に収容され、該容器から洗眼カップに注いで使用されるか、該容器からそのまま点眼される。該組成物の粘度や浸透圧(浸透圧比)はこのようにして使用できる限り制限されない。
本開示の洗眼用組成物は、コンタクトレンズを装着した状態で使用してもよく、装着していない状態で使用してもよい。コンタクトレンズは、ハードコンタクトレンズ、ソフトコンタクトレンズの別を問わない。
本開示の洗眼用組成物は、前述のようにして使用できる限り制限されないが、洗眼カップを用いて使用される場合、通常、後述の実施例に述べるような従来公知の一般的な容器(例えば、容器本体、キャップを含む)に収容される。該容器は、容器本体とキャップとが独立しているタイプ、一体型になっているタイプを問わず、また、中栓(中蓋)があってもよく、中栓には洗眼用組成物を洗眼カップに注ぐための穴が開いていてもよい。該容器の容量も制限されないが、使い勝手が良い等の観点から、容量は好ましくは400~550mL、より好ましくは450~550mLが例示される。
洗眼カップを用いる洗眼は、通常の手順に従えばよく、例えば、洗眼カップに本開示の洗眼用組成物を前記容器から適量、例えば1眼あたり1回に4~6mLを注ぎ、次いで、該カップを片側の眼周辺に押し当てて該カップ内の洗眼用組成物と眼とを接触させ、数回まばたきを行うことにより使用される。該組成物の1日あたりの使用回数も制限されず、例えば、1眼あたり1日1~6回、2~6回、3~6回等のいずれであってもよい。このようにして眼を洗浄することができる。
また、本開示の洗眼用組成物が点眼による洗眼に使用される場合、使い勝手が良い等の観点から、該組成物は、通常、点眼容器に収容して使用される。点眼容器としては、該組成物を収容して点眼できる限り制限されず、例えば、従来公知の点眼剤や点眼型洗眼剤等に使用されている点眼容器(例えば、容器本体、中栓(ノズル)、キャップを含む)が例示される。点眼容器は、容器本体、中栓(ノズル)及びキャップのそれぞれが独立しているタイプ、容器本体、中栓(ノズル)及びキャップの少なくとも2つが一体型となっているタイプ、中栓(ノズル)に穴が設けられているタイプ、中栓(ノズル)に穴が無く、使用時にキャップを巻き締めることによって中栓(ノズル)に穴を開けて使用するタイプ等のいずれであってもよい。
点眼容器の容量は制限されないが、使い勝手が良い等の観点から、容量は8~20mLが例示され、好ましくは10~15mL、より好ましくは12~14mLが例示される。容量は、マルチドーズ型、ユニットドーズ型の別を考慮して適宜決定すればよい。
点眼による洗眼は、本開示の洗眼用組成物を、1眼あたり1回に3~8滴点眼することにより使用される。この限りにおいて制限されないが、該組成物は、1眼あたり1回に、好ましくは4~8滴、より効率良く洗眼する観点から、より好ましくは4~6滴が点眼される。1滴あたりの滴下量も制限されないが、1滴あたり、好ましくは35~40μL、より好ましくは35~37μLが例示される。また、該組成物の1日あたりの使用回数も特に制限されず、例えば、1眼あたり1日1~12回、2~8回、3~6回等のいずれであってもよい。このようにして眼を洗浄することができる。
このように、アラントインを含有する洗眼用組成物にヒプロメロースを配合することにより、アラントインの分解を抑制できる。このことから、該洗眼用組成物においてアラントインの安定性を向上することができる。また、該組成物が前述の薬効成分等を更に含有している場合は、該薬効成分等に基づく有用作用も提供することができる。
以下、実施例を示して本発明をより詳細に説明するが、本発明はこれらに限定されない。
試験例
1.洗眼用組成物の調製及び安定性評価
次の表1に示す配合割合に従い各成分を混合し、各組成物を調製した(実施例1~6、比較例1及び2)。製造直後の各組成物のpHを測定した。pHは、室温(25℃)で、卓上型pHメーターF-52(株式会社堀場製作所製)を用いて測定した。得られた組成物500mLを洗眼用カップ(アイカップ)(アイボンAL d、小林製薬株式会社製)付きの洗眼剤用容器(容量500mL、アイボンAL d、小林製薬株式会社製)に充填し蓋をしたのち、50℃、暗所で1カ月間静置保存した。また、別途、得られた各組成物10mLを点眼型洗眼剤用容器(容量13mL、ポリエチレンテレフタレート樹脂製、マルチドーズ型、伸晃化学株式会社製)に入れ、蓋をしたのち、50℃、暗所で1カ月間静置保存した。調製直後の各組成物と、50℃、暗所で1カ月間保存後の各組成物について、アラントインの含有量をHPLC(高速液体クロマトグラフィー;High Performance Liquid Chromatography)により定量した。
1.洗眼用組成物の調製及び安定性評価

調製直後の各組成物におけるアラントインの含有量を100%として、洗眼剤用容器にて保存後の各組成物におけるアラントインの割合を算出し、小数点第二位を四捨五入した値をアラントインの残存率(%)とした。
2.結果
結果を表1に示す。表1は、洗眼用カップ付き洗眼剤用容器に保存した場合の結果である。

アラントインを含有し、ヒプロメロースを含有しない比較例1の組成物では、アラントインの残存率が86.4%であり、アラントインの残存率は十分ではなかった。これに対して、アラントインにヒプロメロースを併用した実施例1及び2では、アラントインの残存率がそれぞれ90.1%、91.1%へと向上し、アラントインの安定性が向上した。一方、アラントインにヒドロキシエチルセルロースを併用した比較例2では、アラントインの残存率が85.2%であり、比較例1と比較してアラントインの安定性がむしろ低下した。
このことから、ヒプロメロースを用いることにより洗眼用組成物中のアラントインの低減を抑制でき、安定性を向上できることが確認された。また、ヒプロメロースとヒドロキシエチルセルロースとはセルロース系の増粘剤に属する点で共通するが、ヒプロメロースとアラントインとを併用した場合はアラントインの安定性を向上できるのに対して、ヒドロキシエチルセルロースとアラントインとを併用してもアラントインの安定性は向上できないことが分かった。
また、アラントイン、ヒプロメロースと共に、メントールもしくはボルネオール、またはメントールとボルネオールの両方を併用した実施例3~5では、アラントインの残存率が更に向上し、アラントインの安定性を一層向上できた。このことから、アラントイン、ヒプロメロースと共にメントール、ボルネオールといった清涼化剤を更に併用することにより、洗眼用組成物中のアラントインの安定性を更に向上できることが確認された。また、実施例6は、実施例5のpHを5.5へ変更したものであり、実施例6においてアラントインの安定性の更なる向上が認められた。また、表には示さないが、実施例3においてメントールの含有量を0.006w/v%へ増加させた場合、アラントインの安定性が更に向上した。また、実施例4においてボルネオールの含有量を0.006w/v%へ増加させた場合においても、アラントインの安定性が更に向上した。なお、点眼型洗眼剤用容器に収容した各組成物においても同様に、アラントインの安定性を向上できることが認められた。
処方例
表2に本開示の洗眼用カップ付き洗眼剤用容器に収容した洗眼用組成物の処方例を示す。表2に示す処方例も試験例と同様にしてアラントインの残存率を評価したところ、処方例1~5においてヒプロメロースを含有しない場合と比較して、処方例1~5のいずれにおいてもアラントインの安定性が向上していることが確認された。また、処方例1~5を点眼型洗眼剤用容器に収容した各組成物においても同様に、アラントインの安定性が向上していることが確認された。
表2に本開示の洗眼用カップ付き洗眼剤用容器に収容した洗眼用組成物の処方例を示す。表2に示す処方例も試験例と同様にしてアラントインの残存率を評価したところ、処方例1~5においてヒプロメロースを含有しない場合と比較して、処方例1~5のいずれにおいてもアラントインの安定性が向上していることが確認された。また、処方例1~5を点眼型洗眼剤用容器に収容した各組成物においても同様に、アラントインの安定性が向上していることが確認された。

- アラントイン及びヒプロメロースを含有する洗眼用組成物。
- 更に、清涼化剤を含有する、請求項1に記載の洗眼用組成物。
- pHが5~7である、請求項1または2に記載の洗眼用組成物。
