Abstract
본 발명은 아토피 피부염 예방 또는 치료를 위한 항염증 펩타이드에 관한 것으로, 상세하게는 서열번호 1 내지 서열번호 17로 이루어진 군에서 선택되는 아미노산 서열로 이루어지는 펩타이드 또는 이의 단편, 또는 이의 접합체, 및 상기 펩타이드 또는 이의 단편 등을 유효성분으로 포함하는 아토피 피부염 예방 또는 치료용 약학 조성물 및 화장료 조성물을 제공한다. 상기 펩타이드 또는 이의 단편, 또는 이의 접합체는 우수한 항염 활성을 가지는 바, 기존의 아토피 치료제의 부작용으로부터 안전하고 효과적으로 아토피 피부염을 예방, 개선 또는 치료할 수 있다.
Description
본 발명은 아토피 피부염 예방 또는 치료를 위한 항염증 펩타이드에 관한 것이다.
아토피 피부염(Atopic Dermatitis)은 소아에서 흔히 나타나고 성인이 되어서도 증상이 지속될 수 있는 만성 난치성 피부 염증 질환이다. 일반적으로 소양증(가려움증)과 피부 건조증, 특징적인 습진 등을 동반하며, 특히 심한 가려움증으로 자주 긁게 되면 피부 장벽이 손상되어 염증이 더욱 악화되면서 가려움증이 증가하는 악순환이 반복된다. 피부 건조증 또한, 가려움증을 유발하고 악화시킨다.
아토피 피부염은 환경적인 요인, 유전적 소인, 면역학적 이상 및 피부 보호막의 이상 등이 주요 원인으로 짐작되고 있을 뿐, 이의 발병 원인은 아직 확실하게 알려져 있지 않기에, 아토피 피부염의 그 근원을 찾아 치료하는 것이 쉽지 않다. 따라서 아토피 피부염은 완치를 목표로 하기 보다는 유발 인자를 피하고 적절한 치료를 통해 아토피 피부염의 증상을 조절해 나가는 것이 일반적이다.
현재 아토피 피부염 치료제로 스테로이드제와 칼시뉴린 억제제가 가장 많이 사용되고 있다. 그밖에 가려움증을 억제하는 항히스타민제와 이차 세균감염 방지를 위하여 항생제 등이 사용된다. 스테로이드제(부신피질호르몬제)는 크게 소염 작용과 면역 억제 작용을 하며 그 효과가 우수하지만, 장기간 바르면 피부 약화, 전신 호르몬 증상, 중독성 등의 부작용이 나타날 수 있다. 칼시뉴린 억제제는 피부가 얇고 약한 부위에 사용 가능하나, 영유아 발암 문제로 사용이 제한적인 단점이 있다. 또한, 항히스타민제는 비만세포에서 히스타민이 유리되지 못하도록 하여 가려운 증상을 경감시키나 임시방편으로 사용할 수 있으나, 장기간 사용 시 불면, 불안, 식욕감퇴 등의 부작용이 있을 수 있다.
최근에는 2001년 12월에 미국 FDA의 승인을 받은 비스테로이드성 아토피 피부염 면역 치료제 엘리델 크림 1%(성분: 피메크로리무스)이 미국 및 전세계에서 아토피 피부염 치료제로 처방 되고 있고, PDE-4 억제제 등이 출시되어 있다. 하지만 PDE-4 억제제 치료 중단 4주 후 재발률이 45%에 이르는 문제점이 있다.
이에, 이러한 부작용이 없고 안전하며 효과적인 아토피 피부염 치료제가 필요한 실정이다.
본 발명의 목적은 항염증 효과를 가진 신규 펩타이드를 제공하는 데에 있다.
본 발명의 다른 목적은 상기 펩타이드를 유효성분으로 함유하는 아토피 피부염 예방 또는 치료용 조성물을 제공하는 데에 있다.
상기의 목적을 달성하기 위하여, 본 발명은 서열번호 1 내지 서열번호 17로 이루어진 군에서 선택되는 아미노산 서열로 이루어지는 펩타이드 또는 이의 단편을 제공한다.
본 발명은 상기 펩타이드 또는 이의 단편이 생체 적합성 고분자 또는 지방산과 접합된 접합체를 제공한다.
본 발명은 상기 펩타이드 또는 이의 단편, 및 접합체로 이루어진 군에서 선택되는 하나 또는 둘 이상을 유효성분으로 포함하는 아토피 피부염 예방 또는 치료용 약학 조성물을 제공한다.
또한, 본 발명은 상기의 펩타이드 또는 이의 단편, 및 접합체로 이루어진 군에서 선택되는 하나 또는 둘 이상을 유효성분으로 포함하는 아토피 피부염 예방 또는 개선용 화장료 조성물을 제공한다.
본 발명에 따른 신규한 펩타이드 또는 이의 단편, 또는 이의 접합체는 우수한 항염증 활성을 가져, 아토피 피부염 예방, 치료 또는 개선을 위한 약학 조성물 또는 화장료 조성물로 활용할 수 있다.
상기 조성물을 활용함으로써, 기존의 아토피 치료제의 부작용으로부터 안전하고 보다 효과적으로 아토피 피부염을 예방, 치료 또는 개선할 수 있다.

이하, 본 발명을 상세하게 설명하기로 한다.
본 발명자는 신규한 펩타이드를 제조하여 이의 항염 활성을 확인하고, 아토피 피부염이 유도된 마우스 모델에서 치료 효과를 확인함으로써, 본 발명을 완성하였다.
본 발명은 서열번호 1 내지 서열번호 17로 이루어진 군에서 선택되는 아미노산 서열로 이루어지는 펩타이드 또는 이의 단편을 제공한다.
상기 펩타이드 또는 이의 단편은 면역 조절 기능 또는 항염증 활성을 가지는 것을 특징으로 한다.
또한, 상기 펩타이드 또는 이의 단편은 피부 조직으로 침투되고 세포막을 통과하는 것을 특징으로 한다.
또한, 상기 펩타이드 또는 이의 단편은 CD4 T cell의 빈도를 억제하여 사이토카인을 억제함으로써, 아토피피부염 활성을 갖는 것을 특징으로 한다.
상기 사이토카인은 IL-4, IL-13, IL-17, IL-21 및 IL-31로 이루어진 군에서 선택되는 하나 이상일 수 있으나, 이에 한정되는 것은 아니다.
상기 아미노산에 사용된 문자는 생화학 분야에서의 표준 약어 규정에 따라 다음의 아미노산을 의미한다.

본 명세서에서, "펩타이드(peptide) 또는 이의 단편"이란 아미드 결합 (또는 펩타이드 결합)으로 연결된 2개 이상의 아미노산으로 이루어진 폴리머를 의미한다.
본 발명에 따른 펩타이드 또는 이의 단편은 면역 조절 기능 또는 항염증 활성을 가질 수 있다.
본 발명의 일 실험예에 따르면, 아토피 피부염 마우스에서 증가된 IL-4R alpha, CCR3, CTLA-4 positive cell 빈도가 펩타이드 A900 처치 후에 모두 감소하였으며, IL-13, IL-4, TNF alpha, IL-17 등의 사이토카인 또한 감소하는 것을 확인할 수 있었다.
본 발명에 따른 서열번호 1 내지 서열번호 17로 이루어진 군에서 선택되는 아미노산 서열로 이루어지는 펩타이드 또는 이의 단편은 천연으로부터 유래될 수 있고, 당 분야에 공지된 화학적 합성 방법 또는 유전 공학적 방법을 이용하여 화학적으로 합성하거나 유전자 재조합 기술을 이용하여 획득될 수 있으나, 이에 제한되는 것은 아니다.
상기 공지된 화학적 합성(Creighton, Proteins; Structures and Molecular Principles, W. H. Freeman and Co., NY, 1983)의 대표적인 방법으로는 액체 또는 고체상 합성, 단편 응축, F-MOC 또는 T-BOC 화학법이 포함되나(Chemical Approaches to the Synthesis of Peptides and Proteins, Williams et al., Eds., CRC Press, Boca Raton Florida, 1997; A Practical Approach, Athert on & Sheppard, Eds., IRL Press, Oxford, England, 1989), 이에 제한되는 것은 아니다.
상기 유전공학적 방법은 통상적인 방법에 따라 상기 펩타이드 또는 이의 단편을 암호화하는 DNA 서열을 제작할 수 있다. 상기 DNA 서열은 적절한 프라이머를 사용하여 PCR 증폭함으로써 제작할 수 있다. 다른 방법으로 당업계에 공지된 표준 방법에 의해, 예컨대, 자동 DNA 합성기(Biosearch 또는 Applied Biosystems 사에서 판매하는 것)을 사용하여 DNA 서열을 합성할 수 있다.
합성된 DNA 서열은 이에 작동 가능하게 연결되어 DNA 서열의 발현을 조절하는 하나 이상의 발현 조절 서열(expression control sequence)을 포함하는 벡터에 삽입시키고, 이로부터 형성된 재조합 발현 벡터로 숙주 세포를 형질 전환시켜 형질 전환체를 생성한다. 생성된 형질 전환체를 상기 DNA 서열이 발현되기에 적절한 배지 및 조건 하에서 배양하여, 배양물로부터 상기 DNA 서열에 의해 코딩된, 실질적으로 순수한 펩타이드를 회수할 수 있다. 상기 회수는 당업계에 공지된 방법(예컨대, 크로마토그래피)을 이용하여 수행할 수 있다.
본 발명에 따른 펩타이드는 천연형 아미노산 서열을 갖는 펩타이드 뿐만 아니라 이의 아미노산 서열 변이체를 포함할 수 있다.
상기 "아미노산 서열 변이체"란, 본 발명에 따른 아미노산 서열에서 하나 이상의 아미노산 잔기가 결실, 삽입, 비보전적 또는 보전적 치환, 아미노산 유사체의 치환 또는 이들의 조합에 의하여 상이한 서열을 가지는 펩타이드를 의미하나, 상기의 변이에 의해 분자의 활성은 전체적으로 변경되지 않는다.
또한, 본 발명에 따른 펩타이드는 인산화(phosphorylation), 황화(sulfation), 아크릴화(acrylation), 당화(glycosylation), 메틸화(methylation), 파네실화(farnesylation) 등으로 수식(modification)될 수 있다.
또한, 본 발명은 서열번호 1 내지 서열번호 17로 이루어진 군에서 선택되는 아미노산 서열로 이루어지는 펩타이드 또는 이의 단편이 생체 적합성 고분자 또는 지방산과 접합(conjugation)된 접합체를 제공한다.
상기 생체 적합성 고분자는 풀루란(pullulan), 콘드로이틴황산(chondroitin sulfate), 히알루론산(hyauronic acid, HA), 글라이콜 키토산(glycol chitosan), 전분(starch), 키토산(chitosan), 덱스트란(dextran), 펙틴(pectin), 커드란(curdlan), 폴리-L-라이신(poly-L-lysine), 폴리아스파르트산(poly-aspartic acid, PAA), 폴리락틱산(polylactic acid, PLA), 폴리글리콜라이드(polyglycolide, PGA), 폴리카프로락톤(poly(ε-caprolactone), PCL), 폴리(카프로락톤-락타이드) 랜덤 공중합체(PCLA), 폴리(카프로락톤-글리콜라이드) 랜덤 공중합체(PCGA), 폴리(락타이드-글리티콜라이드) 랜덤 공중합체 (PLGA), 폴리에틸린 글라이콜(polyethylene glycol, PEG), 플루로닉 F-68(pluronic F-68) 및 플루로닉 F-127(pluronic F-127)으로 이루어진 군에서 선택되는 하나 또는 둘 이상일 수 있으나, 이에 제한되는 것은 아니다.
상기 지방산은 헥사노익산(hexanoic acid), 카프릴산(caprylic acid, C8), 카프릭산(capric acid, C10), 라우르산(lauric acid, C12), 미리스트산(myristic acid, C14), 팔미트산(palmitic acid, C16) 스테아르산(stearic acid, C18) 및 콜레스테롤(cholesterol)로 이루어진 군에서 선택되는 하나 또는 둘 이상일 수 있으나, 이에 제한되는 것은 아니다.
상기 펩타이드 또는 이의 단편을 상기 생체 적합성 고분자 또는 지방산과 접합시킴으로써 상기 펩타이드의 안정성을 증가시키는 등의 특정 목적을 수행할 수 있다. 예를 들어, 상기 펩타이드 또는 이의 단편을 생체 적합성 고분자인 폴리에틸렌 글라이콜(polyethylene glycol, PEG)과 접합(PEGylation)시켜, 펩타이드 또는 단백질에 수용성을 부여하거나, 반감기를 개선시킬 수 있다.
본 발명은 서열번호 1 내지 서열번호 17로 이루어진 군에서 선택되는 아미노산 서열로 이루어지는 펩타이드 또는 이의 단편, 및 상기 펩타이드 또는 이의 단편이 생체 적합성 고분자 또는 지방산과 접합된 접합체로 이루어진 군에서 선택되는 하나 또는 둘 이상을 유효성분으로 포함하는 아토피 피부염 예방 또는 치료용 약학 조성물을 제공한다.
상기 조성물은 경구제, 외용제, 좌제 및 주사제로 이루어진 군에서 선택되는 것을 특징으로 하나, 이에 한정되는 것은 아니다.
상기 펩타이드 또는 이의 단편, 또는 접합체는 우수한 항염증 활성을 가지는 바, 아토피 피부염 예방 또는 치료용 약학 조성물로 사용될 수 있다.
상기 펩타이드 또는 이의 단편, 또는 접합체는 전체 조성물 100 중량부에 대하여 0.001 내지 50 중량부로 포함될 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 따른 약학 조성물은 약학적 분야의 통상적인 방법에 따라 제조될 수 있다.
상기 약학 조성물은 상기 제형에 따라 약학적으로 허용가능한 적절한 담체와 배합될 수 있고, 필요에 따라, 부형제, 희석제, 분산제, 유화제, 완충제, 안정제, 결합제, 붕해제, 용제 등을 더 포함하여 제조할 수 있다.
본 명세서에서, "약학적으로 허용 가능한"이란, 상기 약학 조성물에 노출되는 세포나 인간에게 독성이 없는 것을 의미하고, 상기 적절한 담체 등은 본 발명에 따른 펩타이드 또는 이의 단편, 또는 접합체의 활성 및 특성을 저해하지 않는 것으로, 투여 형태 및 제형에 따라 달리 선택될 수 있다.
본 발명에 따른 약학 조성물은 어떠한 제형으로도 적용될 수 있고, 보다 상세하게는 통상의 방법에 따라 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 비경구형 제형로 제형화하여 사용될 수 있다.
상기 경구형 제형 중 고형 제형은 정제, 환제, 산제, 과립제, 캡슐제 등의 형태로, 적어도 하나 이상의 부형제, 예를 들면, 전분, 칼슘카보네이트, 수크로스, 락토오스, 솔비톨, 만니톨, 셀룰로오스, 젤라틴 등을 섞어 조제할 수 있고, 단순한 부형제 이외에 마그네슘 스테아레이트, 탈크 같은 윤활제들도 포함될 수 있다. 또한, 캡술제형의 경우 상기 언급한 물질 외에도 지방유와 같은 액체 담체를 더 포함할 수 있다.
상기 경구형 제형 중 액상 제형은 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다.
상기 비경구 제형은 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함될 수 있다. 비수성용제, 현탁제로는 프로필렌글리콜, 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다. 이에 제한되지 않고, 당해 기술 분야에 알려진 적합한 제제를 모두 사용 가능하다.
또한, 본 발명에 따른 약학 조성물은 치료 효능의 증진을 위해 칼슘이나 비타민 D3 등을 더 첨가할 수 있다.
본 발명에 따른 약학 조성물은 약학적으로 유효한 양으로 투여될 수 있다.
본 명세서에서, "약학적으로 유효한 양"이란, 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분하며 부작용을 일으키지 않을 정도의 양을 의미한다.
상기 약학 조성물의 유효 용량 수준은 사용 목적, 환자의 연령, 성별, 체중 및 건강 상태, 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 방법, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 배합 또는 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 달리 결정될 수 있다. 예를 들어, 일정하지는 않지만 일반적으로 0.001 내지 100mg/kg으로, 바람직하게는 0.01 내지 10mg/kg을 일일 1회 내지 수회 투여될 수 있다. 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다.
본 발명에 따른 약학 조성물은 아토피 피부염이 발생할 수 있는 임의의 동물에 투여할 수 있고, 상기 동물은 예를 들어, 인간 및 영장류뿐만 아니라 소, 돼지, 말, 개 등의 가축 등을 포함할 수 있다.
본 발명에 따른 약학 조성물은 제제 형태에 따른 적당한 투여 경로로 투여될 수 있고, 목적 조직에 도달할 수 있는 한 경구 또는 비경구의 다양한 경로를 통하여 투여될 수 있다. 투여 방법은 특히 한정할 필요 없이, 예를 들면, 경구, 직장 또는 정맥, 근육, 피하, 기관지내 흡입, 자궁내 경막 또는 뇌혈관내(intracere-broventricular) 주사 등의 통상적인 방법으로 투여될 수 있다.
본 발명에 따른 약학 조성물은 아토피 피부염의 예방 또는 치료를 위하여 단독으로 사용될 수 있고, 수술 또는 다른 약물치료 등과 병용하여 사용될 수 있다.
본 발명은 서열번호 1 내지 서열번호 17로 이루어진 군에서 선택되는 아미노산 서열로 이루어지는 펩타이드 또는 이의 단편, 및 상기 펩타이드 또는 이의 단편이 생체 적합성 고분자 또는 지방산과 접합된 접합체로 이루어진 군에서 선택되는 하나 또는 둘 이상을 유효성분으로 포함하는 아토피 피부염 예방 또는 개선용 화장료 조성물을 제공한다.
상기 펩타이드 또는 이의 단편, 또는 접합체는 우수한 항염증 활성을 가지는 바, 아토피 피부염 예방 또는 개선용 화장료 조성물로 사용될 수 있다.
상기 펩타이드 또는 이의 단편, 또는 접합체는 전체 조성물 100 중량부에 대하여 0.001 내지 50 중량부로 포함될 수 있으나, 이에 제한되는 것은 아니다.
상기 화장료 조성물은 유연화장수, 영양화장수, 영양크림, 젤, 에센스, 비누, 연고, 밤(balm) 또는 팩의 제형을 갖는 것을 특징으로 한다.
본 발명에 따른 화장료 조성물은 식품의약품안전처에 등록된 화장품 원료 리스트 및 ICID(국제화장품 원료집)에 등재되어 있는 성분 중에서 1종 이상을 더 포함할 수 있다. 예를 들어, 상기 화장료 조성물은 유기 용매, 용해제, 농축제, 겔화제, 연화제, 항산화제, 현탁화제, 안정화제, 방향제, 계면활성제, 유화제, 충전제, 금속이온 봉쇄제, 방부제, 비타민, 차단제, 습윤화제, 염료 및 안료, 친수성 또는 친유성 활성제, 또는 화장품에 통상적으로 사용되는 임의의 다른 성분과 같은 화장품 분야에서 통상적으로 사용되는 보조제를 하나 이상 더 포함할 수 있다.
상기 비타민은 수용성 또는 유용성 비타민을 포함할 수 있고, 상기 수용성 비타민은 수용성 비타민으로서 화장품에 배합 가능한 것으로, 바람직하게는 비타민 B1, 비타민 B2, 비타민 B6, 피리독신, 염산피리독신, 비타민 B12, 판토텐산, 니코틴산, 니코틴산아미드, 엽산, 비타민C, 비타민 H 등을 들 수 있다. 또한, 그들의 염 (티아민염산염, 아스코르빈산나트륨염 등)이나 유도체(아스코르빈산-2-인산나트륨염, 아스코르빈산-2-인산마그네슘염 등)도 포함될 수 있고, 상기 수용성 비타민은 미생물 변환법, 미생물의 배양물로부터의 정제법, 효소법 또는 화학 합성법 등의 통상의 방법에 의해 수득될 수 있다.
본 발명에 따른 화장료 조성물은 본 발명이 속하는 기술 분야에서 통상적으로 제조되는 어떠한 제형으로도 제조될 수 있다. 예를 들어, 화장수, 유액, 로션, 크림, 페이스트, 젤, 용액, 현탁액, 오일, 왁스, 팩, 파우더, 파운데이션, 스프레이, 계면활성제-함유 클렌징 등으로 제형화 될 수 있으나, 이에 제한되는 것은 아니다.
더욱 상세하게는, 유연 화장수, 영양 화장수, 수렴 화장수, 영양 크림, 마사지 크림, 밀크 로션, 파우더, 에센스, 아이 크림, 선로션, 선크림, 메이크업 프라이머, 메이크업 베이스, 비비크림, 분말 파운데이션, 유탁액 파운데이션, 클렌징크림, 클렌징폼, 클렌징 워터, 비누, 연고, 팩, 스틱상 제품, 밤(Balm) 타입 제품, 스프레이 또는 파우더의 제형으로 제조될 수 있다.
본 발명에 따른 화장료 조성물이 크림 또는 젤 제형인 경우에는 담체 성분으로 동물성유, 식물성유, 왁스, 파라핀, 전분, 셀룰로오스 유도체, 폴리에틸렌 글리콜, 실리콘, 벤토나이트, 실리카, 탈크 또는 산화아연 등을 더 포함할 수 있다.
상기 화장료 조성물이 용액 또는 유탁액 제형인 경우에는 용매, 용매화제 또는 유탁화제로, 물, 에탄올, 이소프로판올, 에틸카보네이트, 에틸아세테이트, 벤질알코올, 프로필렌글리콜, 글리세롤 지방족 에스테르, 폴리에틸렌글리콜 또는 소르비탄의 지방산 에스테르 등을 더 포함할 수 있다.
상기 화장료 조성물이 현탁액 제형인 경우에는 담체 성분으로 물, 에탄올 또는 프로필렌 글리콜과 같은 액상 희석제, 에톡실화 이소스테아릴 알코올, 폴리옥시에틸렌 소르비톨 에스테르 및 폴리옥시에틸렌 소르비탄 에스테르와 같은 현탁제, 미소결정성 셀룰로오스, 알루미늄 메타히드록시드, 벤토나이트, 아가 또는 트라칸트 등을 더 포함할 수 있다.
상기 화장료 조성물이 파우더 또는 스프레이 제형인 경우에는 담체 성분으로 락토스, 탈크, 실리카, 알루미늄 히드록시드, 칼슘 실리케이트 또는 폴리아미드 파우더를 더 포함할 수 있고, 특히 스프레이 제형인 경우에는 추가적으로 클로로플루오로히드로카본, 프로판/부탄 또는 디메틸에테르와 같은 추진체를 더 포함할 수 있다.
상기 화장료 조성물이 계면활성제-함유 클렌징 제형인 경우에는 담체 성분으로 지방족 알코올 설페이트, 지방족 알코올 에테르 설페이트, 설포숙신산 모노에스테르, 이세티오네이트, 이미다졸리늄 유도체, 메틸타우레이트, 사르코시네이트, 지방산 아미드 에테르 설페이트, 알킬아미도베타인, 지방족 알코올, 지방산 글리세리드, 지방산 디에탄올아미드, 식물성유, 리놀린 유도체 또는 에톡실화 글리세롤 지방산 에스테르 등을 더 포함할 수 있다.
본 발명에 따른 화장료 조성물은 단독 또는 중복으로 도포하여 사용하거나, 본 발명 이외의 다른 화장료 조성물과 중복 도포하여 사용할 수 있다. 또한, 본 발명에 따른 화장료 조성물은 통상적인 사용방법에 따라 사용될 수 있으며, 사용자의 피부 상태 또는 취향에 따라 그 사용횟수를 달리할 수 있다.

실험예
1> 아토피 피부염 유도 마우스 모델에서
펩타이드의
치료 효과 확인
실험 마우스는 12시간의 명/암 주기로, 23℃의 항온 및 55% 습도에서 병원체가 없는 상태로 사육하였다. 수일간의 적응 기간 후, 하기의 방법으로 마우스에 DNCB-유도 아토피성 피부염을 유도하였다.
먼저, 제모 크림을 이용하여 등 피부 영역의 모발을 조심스럽게 면도한 후, 피부를 민감하게 하고 아토피성 피부염을 유도하기 위해, 아세톤에 용해된 1% DNCB (Dinitrochlorobenzene) 100μL을 노출된 피부에 3일 동안 1일 1회 국소 처리하였다. 4일에서 6일 동안에는 2% DNCB 120μL을 처리하여 7일 째에는 아토피성 피부염이 발생하는 것을 확인할 수 있었다.
* 2% DNCB : 20mL 아세톤에 100mg의 DNCB 파우더를 용해시켜 제조함.
* 1% DNCB : 20mL 아세톤에 50mg의 DNCB 파우더를 용해시켜 제조함.
아토피 피부염 마우스에 항염 보습크림 투여군(Control Cream) 및 아토피 피부염 치료제인 듀피센트(Dupixent) 주사제로 처리한 양성 대조군을 펩타이드 처리군(A900 peptide)과 비교하였다.
도 1에 나타난 바와 같이, DNCB(dinitrochlorobenzene)로 유도한 아토피 피부염 마우스에 상기 펩타이드를 외용제로 발라주었을 때 아토피 피부염 증상이 호전되었고, 대조군보다 효과가 더 빨리 나타나는 것을 확인했다.
<실험예 2> 펩타이드의 항염증 활성 확인
아토피 피부염 염증을 호전시키는 효과가 있는 펩타이드가 면역 조절 기능도 있는지 확인하고자, 5일간 마우스 피부에 외용제로 투여 후 비장세포(splenocytes) 및 말초혈액세포(peripheral blood leukocytes, PBL)를 각각 분리하여 FACS용 항체로 surface staining한 후, flow cytometry로 IL-4R alpha(interleukin 4 receptor alpha), CCR3(C-C chemokine receptor type 3) 및 CTLA-4(cytotoxic T-lymphocyte-associated protein 4)를 발현하는 세포의 빈도를 분석하였다. 듀피센트(Dupixent)를 복강주사로 투여하여 대조군으로 실험하였다.
비장세포(splenocytes)를 이용한 펩타이드의 항염증 활성을 분석한 결과, 도 2에 나타난 바와 같이, 아토피 피부염 마우스에서 IL-4R alpha, CCR3 및 CTLA-4 positive cell 빈도가 정상 대조군에 비하여 증가하였고, 펩타이드 A900 처치 후에 세 molecule 모두 억제하는 경향을 보였으며, 억제 정도는 대조 투여군보다 더 효과적인 것으로 나타났다.
펩타이드의 항염증 활성을 아토피 피부염 마우스 말초혈액세포(peripheral blood leukocytes, PBL)에서 분석한 결과, 도 3에 나타난 바와 같이, A900 펩타이드 투여군에서 CD11b+Ly6G+ 및 CD83+ positive cell의 빈도는 억제되었고, Foxp3+ positive cell의 빈도는 증가하는 것으로 나타났다. CD11b+Ly6G+ 및 CD83+ positive cell의 억제 정도는 대조 투여군보다 더 효과적인 것으로 나타났다. 이는 A900 펩타이드 투여에 의한 아토피피부염 증상 완화는 호중구의 억제, CD83 억제 및 T 조절세포 증가로 인하여 조절됨을 입증한다.
더불어, 아토피피부염 마우스에 A900 펩타이드를 처리한 후, 비장세포(splenocytes)에서 세포를 분리하여 Real time PCR로 IL-4 발현 정도를 분석한 결과, 도 4에 나타난 바와 같이, A900 펩타이드 투여군에서 IL-4의 발현이 억제되는 것으로 나타났다. 이는 A900 펩타이드가 IL-4 발현을 억제함으로써, 아토피피부염 증상을 완화할 수 있음을 입증한다.
<실험예 3> 펩타이드의 피부 조직 침투성 분석
A900 펩타이드의 피부 조직 침투를 확인하기 위해, A900 펩타이드 N-말단에 형광물질 FAM(Carboxyfluorescein)을 붙인 펩타이드 A900 peptide(A900-FAM)를 ICR 4주령 마우스의 피부에 도포하였다. 오른쪽 다리 피부에는 needle scratch를 내고, 왼쪽 다리 피부에는 scratch를 내지 않고, 1μg의 펩타이드를 면봉으로 바르고 2시간 후에 피부 조직을 채취하여 동결 절단하여 confocal 현미경으로 관찰한 결과, 도 5에 나타난 바와 같이, scratch 유무와 상관없이 A900 펩타이드 처리군 모두에서 표피 및 진피 모두에서 형광을 확인하였다. 이는 A900 펩타이드의 피부 조직을 통한 침투가 가능하다는 것을 입증한다.

마우스 복강에 형광 표지된 펩타이드 3μg을 투여하고 시간의 경과에 따른 형광 발현 면역세포 빈도를 FACS로 분석한 결과, 도 6에 나타난 바와 같이, 30분 경과 시점에 50%의 복강 대식세포가 형광을 발현하였고, 1시간 경과 시점에 23%의 복강 대식세포가 형광을 발현하였으며, 3시간 경과 후부터는 형광 발현세포가 거의 감지되지 않는 것으로 나타났다. 이는 복강으로 투여된 펩타이드가 복강 내 세포의 막을 통과하여 세포 내부로 들어가서, 세포 내에서 1시간 이상 유지되고 있음을 입증한다.
<실험예 5> 세포에 펩타이드 처리 후 세포 내 발현 및 빈도 분석
5~6 주령의 수컷 ICR 마우스에서 림프절 세포(lymph node cell)를 분리하여 세포 현탁액을 배양하였다. 항-CD3(1μg/ml) 항체를 실온에서 배양 접시에 1시간 동안 처리하여 코팅한 다음 세포를 접종하였다(5 x 105 세포/well). 1시간 후, 항-CD28 항체(1㎍/ml), IL-23(20ng/ml), IL-6(20ng/ml) 및 TGF-β(5ng/ml)를 첨가하였다. 그 후, 3일 동안 배양한 세포에 A900-FAM 펩타이드(1μg/mL)를 1시간 및 24시간 동안 처리 후, 공초점 현미경 및 FACS을 통해 펩타이드의 세포 내 발현 및 발현 세포 빈도를 분석한 결과, 도 7에 나타난 바와 같이, 펩타이드 표지 형광색인 녹색으로 표지된 세포를 확인하였고, 1시간 처리 그룹에서 20%, 24시간 처리 그룹에서 40%의 세포 빈도에서 펩타이드가 발현하여, 시간의 경과에 따라 펩타이드의 발현 및 빈도가 증가하는 것으로 나타났다.
도 7에서 사용한 같은 마우스로부터 복강세포(peritoneal macrophage)를 분리하여 세포 현탁액을 배양하였다. 항-CD3(1μg/ml) 항체를 실온에서 배양 접시에 3시간 동안 플레이팅하고, 세포를 접종(5 x 105 세포/well)하였다. 1시간 후, 항-CD28 항체(1㎍/ml), IL-23(20ng/ml), IL-6(20ng/ml) 및 TGF-β(5ng/ml)를 첨가하였다. 그 후, 3일 동안 배양한 세포에 A900-FAM 펩타이드(1μg/mL)를 1시간 및 24시간 동안 처리 후, 공초점 현미경 및 FACS을 통해 세포 내 발현 및 빈도를 분석한 결과, 도 8에 나타난 바와 같이 1시간 배양 그룹에서 10%, 24시간 배양 그룹에서 15%의 빈도로 펩타이드가 발현하였으며, 시간의 경과에 따라 펩타이드의 발현 빈도가 증가하는 것으로 나타났다.

아토피피부염 마우스의 피부 병변에 펩타이드를 외용제(A900~A916)로 처리한 후, 비장(spleen)에서 Real time PCR을 통해 펩타이드의 효과를 분석한 결과, 도 9에 나타난 바와 같이, A900, A901, A902, A903, A907, A911 및 A914 펩타이드가 아토피 관련 cytokine인 IL-4, IL-31 및 IL-17 발현을 억제하는 것으로 나타났다.
<실험예 7> ex vivo를 통한 FACS 결과 분석
6주령 Balb/c male 마우스에 털 제거 후, 1% DNCB 1회/일 2일간, 0.5% DNCB 1회/일 3일간, 총 5일간 바른 후 아토피 증상을 유도하였다. 아토피 마우스에서 PBMC를 분리하여 24 well plate에 seeding한 후, 펩타이드를 처리하고 24시간 동안 배양한 다음 FACS로 분석하였다. 대조군으로 정상 마우스 및 아토피 유도 마우스에서 분리한 PBMC를 사용하였고, 아토피 마우스에서 분리한 PBMC 배양액에 펩타이드를 처리하였다. 펩타이드 대조군은 듀피센트를 사용하였다(Dupixent 5μg/ml). 펩타이드는 A900, A900P, A900H, A900S, A900M, A900MAL, A900PEG2 및 A900PEG4를 사용하였고, 농도는 3μg/ml로 설정하였다. 항-CD4 항체로 surface을 염색하였고, 사이토카인(cytokine)들은 intracellular 염색하였으며, double stained population 빈도를 측정하였다.
그 결과, 도 10에 나타난 바와 같이, 처리하지 않은 대조군(ADC)에 비하여 A900, A900M, A900MAL, A900PEG4, A900P 및 A900S 펩타이드가 아토피 관련 사이토카인(cytokine)을 분비하는 CD4 T cell의 빈도를 억제하여 아토피피부염 증상에 관련된 면역 조절 효과를 보이는 것으로 나타났다.
<실험예 8> 세포 내 사이토카인(cytokine)의 Real time PCR 결과 분석
도 10과 동일한 마우스에서 분리한 비장세포(splenocytes) 및 림프절세포(lymph Node cell) 각각에 A900 및 A900 펩타이드 terminal conjugated 펩타이드(A900M, A900S, A900P, A900H, A900MAL, A900PEG2 및 A900PEG4 펩타이드)를 처리하고 24시간 배양한 후, real time PCR로 아토피피부염 증상 유발 관련 cytokine 발현 정도를 분석한 결과, 도 11 및 도 12에 나타난 바와 같이, 처리하지 않은 대조군(ADC)에 비하여 A900, A900M, A900P, A900H, A900MAL, A900PEG2 및 A900PEG4 펩타이드가 아토피 관련 cytokine인 IL-4, IL-13 및 IL-21을 억제하는 것으로 나타났다.

실험을 통한 세포 내 사이토카인(

결과 분석
6주령 Balb/c male 마우스에 털 제거 후, 1% DNCB 1회/일 2일간, 0.5% DNCB 1회/일 3일간 총 5일간 바른 후 아토피 증상을 유도하였다(각 그룹당 N=5~8마리). 아토피 증상 마우스에 5일간 마우스 피부에 외용제로 투여 후 말초혈액세포(peripheral blood leukocytes, PBL)를 분리하여 FACS용 항체로 intracellular stain 후 각 사이토카인 발현 세포의 빈도를 분석하고 비교하였다. 대조군은 항염 보습크림 외용제(5회) 및 anti-mouse IL-4 receptor antibody(100ug/mouse) 복강 주사(1회)를 투여하였다. 펩타이드는 A900, A900M, A900MAL 및 A900PEG4를 사용하였고, 농도는 1μg/ml로 설정하였다. 사이토카인(cytokine)들은 intracellular 염색하여 stained cell population 빈도를 측정하였다.
그 결과, 도 13에 나타난 바와 같이, 대조군인 PBS를 투여한 아토피피부염 마우스(ADC)에 비하여 A900, A900M, A900MAL 및 A900PEG4 펩타이드가 아토피 관련 사이토카인(cytokine)을 분비하는 cell의 빈도를 억제하여 아토피피부염 증상에 관련된 면역 조절 효과를 보이는 것으로 나타났다.
도 13과 동일한 마우스에서 분리한 말초혈액세포(peripheral blood leukocytes, PBL)를 분리하여 FACS용 항체로 CD4 surface stain 및 intracellular stain 후 각 사이토카인 발현 CD4 세포의 빈도를 분석하고 비교하였다.
그 결과, 도 14에 나타난 바와 같이, A900MAL 펩타이드가 아토피 관련 사이토카인(cytokine) IL-13을 분비하는 CD4 cell 및 IL-31을 분비하는 CD4 cell의 빈도를 억제하여 아토피피부염 증상에 관련된 면역 조절 효과를 보이는 것으로 나타났다.
