一种辅助降脂减肥的海藻果蔬配方 技术领域
本发明属于保健技术领域,具体涉及一种辅助降脂减肥的海藻果蔬配方。
背景技术
据《世界粮食安全和营养状况》的数据,肥胖每年导致约400万人死亡,学龄段人群成为肥胖高发群体,主要是水果、蔬菜摄入不足,缺乏体育锻炼。肥胖主要是由于能量摄入过多造成的,能量摄入超过机体处理负荷时,就会导致相应的病理生理改变,如高血糖、高血脂、脂肪肝等。如今市面上有多种减肥药,其减肥效果虽然明显,但存在很多问题如副作用大、依赖性强等。因此,为了避免减肥药给身体带来的副作用,开发出辅助降脂减肥的保健食品具有非常重要的意义。
海藻产量丰富、易获得,是一种具有食药两用价值的海洋生物资源。近年来关于海藻的研究越来越多,研究表明来自海藻的许多天然产物具有抗炎、降脂减肥、降血糖等生物活性。例如褐藻中的羊栖菜,关于其活性成分及相应的生理功能已有较为广泛的研究;其中有文章提到羊栖菜中的岩藻黄质提取物可以通过调节血糖代谢途径,起到降低肥胖小鼠体重的效果,但笔者通过实验验证时发现,羊栖菜仅能使附睾脂肪显著减少,对皮下脂肪没有作用,单一食用效果不佳,所以需要一种配方食品来补足羊栖菜在减脂方面的不足。
发明内容
本发明要解决的技术问题是羊栖菜仅能使附睾脂肪显著减少,对皮下脂肪没有作用,单一食用效果不佳,所以需要一种配方食品来补足羊栖菜在减脂方面的不足。
为解决上述问题,本发明提供一种辅助降脂减肥的海藻果蔬配方,通过在羊栖菜中复配姜黄提取物和石榴皮提取物,充分利用海藻和果蔬中的天然活性物质,实现辅助降脂减肥的效果,并进一步提高自然资源的利用率。同时,海藻果蔬的天然属性会极大降低合成物质带来的毒副作用,其来源的天然产物安全无毒、无害,并且能够达到辅助降脂减肥的目的,能够改善人群的健康状况。
为达到上述目的,本发明具体通过以下技术方案实现:一种辅助降脂减肥的配方,包括以下质量百分比的组分:羊栖菜95-96%;姜黄提取物2-3%;石榴皮提取物1-2%。
羊栖菜(Sargassum fusiforme)是褐藻门马尾藻科马尾藻属植物,在我国沿海地带均有分布,是一种低脂低热、高蛋白、高膳食纤维和含丰富矿物元素的天然食品。研究发 现羊栖菜具有广泛的生物活性,如抗氧化、抗癌和减肥作用等。有研究表明羊栖菜中岩藻黄质提取物具有良好的减肥作用,其作用机理还待研究,但实验结果表明可能是通过调节血糖代谢途径,从而降低血糖含量,实现减肥的作用。
姜黄素(Curcumin)是从姜科植物姜黄中提取的一种常见的食用色素,具有抗氧化、抗炎等功能。研究表明姜黄素还可以调节体内脂质代谢,显著降低血清和肝脏中的胆固醇含量。而脂质代谢时部分产物会通过糖酵解或产生糖而进入血糖代谢,姜黄素对于羊栖菜的协同作用可能是在于通过脂质代谢的调节,将体内的脂肪,特别是皮下脂肪分解,再通过血糖代谢途径进行代谢,从而解决羊栖菜不能作用皮下脂肪的问题。本发明所用姜黄提取物源自陕西藤迈生物科技有限公司,主要成分:姜黄素,含量约为20%。
从石榴皮中提取的多酚类物质安石榴苷被人体吸收后,可以分解为鞣花酸,其抗氧化性优良,且在抗炎抗癌等方面具有重要的作用,同时有研究表明安石榴苷也具有降脂减肥的功效。安石榴苷在配方中的作用,一方面可能是通过改善炎性反应和肝脏氧化应激,加强肝脏对胆固醇的转化,辅助姜黄素的降胆固醇作用,从而影响脂质代谢;另一个方面可能是改善胰岛素抵抗,使细胞葡萄糖摄取能力下降,从而辅助羊栖菜改善血糖代谢,从而实现辅助减脂的最佳效果。本发明所用石榴皮提取物源自陕西藤迈生物科技有限公司,主要成分:安石榴苷,含量约为20%。
进一步的,所述配方包括以下质量百分比的组分:羊栖菜95.88%;姜黄提取物2.68%;石榴皮提取物1.44%。
以体外糖脂代谢关键酶α-葡萄糖苷酶和胰脂肪酶抑制率为评价指标,抑制率越高效果越好。通过单因素试验和D优化响应面分析,得出最优配方。单因素试验结果如图1所示。在单因素试验的基础上,对配方进行响应面分析,试验因素与水平设计见表1,试验分组及结果如表2所示。对于α-葡萄糖苷酶抑制率和胰脂肪酶抑制率两个响应值,模型的P值均小于0.05,判断模型显著;模型失拟项p值均大于0.05,说明模型失拟项不显著。两个响应值的模型相关系数R 2分别为0.99、0.94,校正后R 2分别为0.97、0.82,表示模型相关度良好;模型的变异系数均小于10%,说明模型对响应值的置信度良好,该模型可以较好地反应试验的真实性。对响应面试验的数据进行分析处理,结果如表3和图1所示。由配方对两种酶的抑制效果可以看出,A1水平(羊栖菜水提物)时,配方对两种酶的抑制效果优于A2和A3水平;B3水平(姜黄素)时,配方对两种酶的抑制效果优于B1和B2;C3水平(安石榴苷)时,配方对两种酶的抑制效果优于C1和C2。经Design-Expert 8.0.6软件分析得出最佳配方为:羊栖菜水提物配比为150,姜黄 素配比为6;安石榴苷配比为3;相应的模型预测α-葡萄糖苷酶抑制率为45.98%,胰脂肪酶抑制率为73.88%。选取预测所得的最佳配比进行复配,检测复配配方2种酶活性抑制率,配方α-葡萄糖苷酶抑制率为47.77±2.15%,胰脂肪酶抑制率为74.18±0.46%,实际值与预测值相近,说明该模型能够较好地预测配方最佳配比。
由此确定的最优配比为羊栖菜水提物∶姜黄素∶安石榴苷=150∶6∶3(w/w/w)。此配方中姜黄素纯度为95%,安石榴苷纯度为98%。因为配方成分均为高纯度的功能因子,没有食用的习惯,不是新食品原料,不能直接用于保健食品。据此制定保健食品级配方,其中羊栖菜水提物对应羊栖菜(羊栖菜水提物含量约为15%),姜黄素对应姜黄提取物(姜黄素含量约为20%),安石榴苷对应石榴皮提取物(安石榴苷含量约为20%)。即羊栖菜∶姜黄提取物∶石榴皮提取物=1000∶28∶15(w/w/w)。因此最佳配方组成为:羊栖菜95.88%;姜黄提取物2.68%;石榴皮提取物1.44%。

进一步的,所述配方所用的姜黄提取物的姜黄素含量为20%,石榴皮提取物的安石榴苷含量为20%。
进一步的,一种辅助降脂减肥配方的应用,按照配方质量百分比称取各组分,按照5%的比例添加入饮食中。根据美国FDA标准规定饲料中受试物的添加量不得超过5%,以不影响正常饮食结构为目的。因此按照5%的比例添加入饮食中,不影响正常饮食结构的情况下又可发挥辅助降血脂、缓解肥胖的作用。
本发明的有益效果在于:
本发明以羊栖菜为主要原料,利用海藻果蔬优良自然资源研发了一种海藻果蔬复配配方,对高脂饮食诱导的肥胖小鼠体重增长有显著的抑制作用,能够减少小鼠体内脂质 积累,降低血脂水平,从而达到辅助降血脂、缓解肥胖的目的;并且实现了羊栖菜的高值化利用。

实施例1:
本实施例的配方为羊栖菜95%;姜黄提取物3%;石榴皮提取物2%。按照配方质量百分比称取各组分,混合后以5%添加量加入饮食中。

实施例3:

对比实施例:采用实施例5的配方做以下实验。
(1)动物实验:高脂饮食诱导的肥胖小鼠模型建立与分组。C57BL/6小鼠适应性喂养1周,按体重随机分为6组,每组10只,即正常组(Control),肥胖模型组(Model),羊栖 菜组(S),羊栖菜与石榴皮提取物复配组(SP),羊栖菜与姜黄提取物复配组(ST),三组分配方组(STP)。正常组小鼠给予10%低脂对照饲料(D14060603),其余5组给予45%高脂饲料(D14060606),小鼠自由摄食9周。室内温度21℃-23℃,相对湿度40%-55%,空气流通,光照时间12h。9周后,按照表4所示给予各组小鼠饲料,持续喂养4周。饲养期间每周称1次体重,每2天记录1次摄食量。实验结束后,称体重,处死,解剖取相应内脏称重并记录,并保存于-80℃,备用。

实验结果:
(1)统计各实验组小鼠饲养期间体重变化情况。结果如图2所示,高脂饮食导致小鼠体重显著增加,Lee指数显著高于正常组,表明长期的高脂饮食导致小鼠肥胖。而摄入配方后(STP组),小鼠体重增长缓慢,体重增长和Lee指数均显著低于模型组。将配方组(STP)与S、SP和ST组分别进行比较后,发现羊栖菜与姜黄提取物和/或石榴皮提取物复配后均可延缓小鼠体重增长。但三组分复配的效果最佳,与模型组相比差异达到显著性。既优于羊栖菜单一成分,也优于两种成分(羊栖菜与姜黄提取物或石榴皮提取物复配),对肥胖小鼠具有一定的延缓体重增长的作用。
(2)称量小鼠各脂肪组织的重量并计算体脂率。结果如图3所示,高脂饮食导致小鼠脂肪组织:附睾脂肪、皮下脂肪、肾周脂肪积累量显著增加,体脂率显著高于正常组。本发明配方则显著减少了附睾脂肪和皮下脂肪的积累量,显著降低了体脂率。将配方组(STP)与S、SP和ST组分别进行比较后,发现羊栖菜与姜黄提取物和/或石榴皮 提取物复配后均减少了附睾脂肪和皮下脂肪的积累量,降低了体脂率,且达到显著性。相比之下,羊栖菜组仅附睾脂肪显著减少,三组分复配的效果要明显优于羊栖菜单一成分。同时也优于两种成分(羊栖菜与姜黄提取物或石榴皮提取物复配),说明配方具有辅助肥胖小鼠减肥的作用。
(3)测定各实验组小鼠的血脂水平,结果如图4所示。羊栖菜与姜黄提取物和/或石榴皮提取物复配后相应地降低了小鼠血清总胆固醇(TC)水平,虽然未达到显著水平,但依照羊栖菜单一成分、两种成分复配、三组分配方的次序具有逐渐降低的趋势。本发明配方能够显著降低肥胖小鼠血液中甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)的含量,能够将LDL-C∶HDL-C比例恢复到正常水平,且效果优于羊栖菜单一成分和两种成分复配(羊栖菜与姜黄提取物或石榴皮提取物复配)。说明本发明配方能够辅助降低肥胖小鼠的血脂水平。
以上所述,仅是本发明的较佳实施例,并非是对本发明作其它形式的限制,任何熟悉本专业的技术人员可以利用上述技术内容加以变更或改型为等同变化的等效实施例。但是凡是未脱离本发明技术方案内容,依据本发明的技术实质对以上实施例所作的任何简单修改、等同变化与改型,仍属于本发明技术方案的保护范围。

Claims (4)

  1. 一种辅助降脂减肥的配方,其特征在于包括以下质量百分比的组分:羊栖菜95-96%;姜黄提取物2-3%;石榴皮提取物1-2%。
  2. 如权利要求1所述的辅助降脂减肥的配方,其特征在于包括以下质量百分比的组分:羊栖菜95.88%;姜黄提取物2.68%;石榴皮提取物1.44%。
  3. 如权利要求1或2所述的辅助降脂减肥的配方,其特征在于:所述配方所用的姜黄提取物的姜黄素含量为20%,石榴皮提取物的安石榴苷含量为20%。
  4. 一种权利要求1中的辅助降脂减肥的配方的应用,其特征在于:按照配方质量百分比称取各组分,按照5%的比例添加入饮食中。
