Abstract
本発明によれば、善玉菌となり得るビフィドバクテリウム属細菌の増殖促進が可能な組成物が提供される。この組成物は、ラクト-N-ネオテトラオース、ラクト-N-テトラオース、2'-フコシルラクトース、3'-シアリルラクトース及び6'-シアリルラクトースからなるヒトミルクオリゴ糖の混合物と、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスとを含む。

本発明は、組成物に関する。

近年、動物に良い影響を与える細菌(以下、「善玉菌」ともいう。)を積極的に摂取して腸内環境を整えることで、病気の発生抑制又は健康増進等を目的とするプロバイオティクス及び善玉菌の増殖を助ける成分等のプレバイオティクスの研究が盛んに行われるようになってきている。プロバイオティクスは、腸内で有益な働きをする細菌をいうのに対して、プレバイオティクスは、これらのプロバイオティクスの選択的な栄養源となり、それらの増殖を促進する物質をいう。プレバイオティクスにより、乳酸菌・ビフィドバクテリウム属細菌増殖促進作用、整腸作用及び炎症性腸疾患への予防・改善作用等のヒトの健康に有益な効果があることが知られている。
ヒトミルクオリゴ糖(以下、「HMOs」ともいう。)は、母乳中に10~20g/L含まれ、130種類以上のオリゴ糖の混合物と言われている。それらの構造は、13種類のコア構造にフコース及びシアル酸が付加した構造をとる。ヒトミルクにおいては、フコシル化された中性糖の割合が高く、2’-フコシルラクトース、ラクト-N-フコペンタオースI、ラクト-N-ジフコヘキサオースI及びラクト-N-テトラオースが代表的である。これら4種類のオリゴ糖だけで全ヒトミルクオリゴ糖の1/3~1/4を占めている。ラクト-N-ビオースを含むオリゴ糖をタイプI型といい、N-アセチルラクトサミン(LacNAc)を含むオリゴ糖をタイプII型という。ヒトミルク中では、タイプI型のラクト-N-テトラオース及びラクト-N-フコペンタオースIが、タイプII型のラクト-N-ネオテトラオース及びラクト-N-フコペンタオースIIIよりも、含有量が高い。タイプI型及びタイプII型の共存、並びにタイプI型の優先性は、他種ミルクオリゴ糖とは異なるヒトミルクオリゴ糖の際だった特徴である(非特許文献1及び2)。
母乳に含まれるHMOsである2’-FL及びLNTは、ビフィズス菌増殖活性を有する。2’-FL及びLNTは、ビフィズス菌に選択的に利用されることによりビフィズス菌の増殖因子として作用することが知られており、ビフィズス菌優勢な腸内フローラの形成に重要である。また、HMOsは合成が可能で、工業的に利用できる状況にある(特許文献1)。
ビフィズス菌は一部のHMOsをビフィズス菌に特異的なGNB/LNB経路を利用して代謝している。この経路はヒト腸管でよく見られる種の多くが持っているが、ヒト以外の動物及び昆虫の腸管でよく見られるビフィズス菌のほとんどは持っていない。HMOsを単独の炭素源として利用し、生育可能な菌種としては、乳児によく見られるビフィズス菌であるビフィドバクテリウム・ロンガム・サブスピーシーズ・ロンガム(Bifidobacterium longum subspecies longum)、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス(Bifidobacterium longum subspecies infantis)、ビフィドバクテリウム・ブレーベ(Bifidobacterium breve)及びビフィドバクテリウム・ビフィダム(Bifidobacterium bifidum)の4菌種が代表例として挙げられる。これらのなかでも、特にビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス及びビフィドバクテリウム・ビフィダムの2菌種は様々なHMOsを利用することが報告されている(非特許文献3)。

善玉菌を積極的に摂取して腸内環境を整えるには、その善玉菌の増殖が促進されるような環境において、善玉菌を摂取することが重要である。
本発明では、善玉菌となり得るビフィドバクテリウム属細菌の増殖促進が可能な組成物を提供することを課題とする。
本発明者らは、上記課題を解決すべく、各種HMOsを含む環境下において、ビフィドバクテリウム属細菌の増殖を検討したところ、HMOsの種類及び組み合わせに応じて、ビフィドバクテリウム属細菌の増殖の程度に違いがあることを見出した。そして、さらに、ビフィドバクテリウム属細菌の増殖に適したHMOsの種類及び組み合わせについて鋭意研究を行った結果、ビフィドバクテリウム属細菌の増殖に適したHMOsの混合物の組成を構築することに成功し、本発明を完成させるに至った。
即ち、本発明では、まず、ラクト-N-ネオテトラオースと、
ラクト-N-テトラオースと、
2’-フコシルラクトースと、
3’-シアリルラクトースと、
6’-シアリルラクトースと、
からなるヒトミルクオリゴ糖の混合物と、
ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスと、
を含む組成物を提供する。
本発明に係る組成物に用いる上記ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスとしては、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス NITE BP-02623を用いることができる。
本発明に係る組成物に用いる上記ヒトミルクオリゴ糖の混合物中には、
上記ラクト-N-ネオテトラオースを、2~10質量%、
上記ラクト-N-テトラオースを、10~20質量%、
上記2’-フコシルラクトースを、55~75質量%、
上記3’-シアリルラクトースを、1~10質量%、
上記6’-シアリルラクトースを、5~15質量%、含ませることができる。
本発明に係る組成物に用いる上記ヒトミルクオリゴ糖には、さらに、ジフコシルラクトース又は3’-フコシルラクトースを含ませることができる。
この場合、本発明に係る組成物に用いる上記ヒトミルクオリゴ糖の混合物中には、
上記ジフコシルラクトース又は上記3’-フコシルラクトースを、1~10質量%、含ませることができる。
本発明に係る組成物には、さらに、ラクトースを含ませることができる。

本発明に係る組成物を用いれば、善玉菌となり得るビフィドバクテリウム属細菌の増殖促進が可能である。なお、ここに記載された効果は、必ずしも限定されるものではなく、本発明中に記載されたいずれかの効果であってもよい。
以下、本発明を実施するための実施形態について説明する。なお、以下に説明する実施形態は、本開示の代表的な実施形態の一例を示したものであり、これにより本発明の範囲が狭く解釈されることはない。なお、本明細書において、数値範囲を「下限~上限」で表現するものに関しては、上限は「以下」であっても「未満」であってもよく、下限は「以上」であっても「超」であってもよい。また、本明細書において百分率は特に断りのない限り質量による表示である。
1.組成物
本発明に係る組成物は、ヒトミルクオリゴ糖の混合物と、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスと、を少なくとも含む。また、本発明に係る組成物は、ラクト-N-テトラオースと、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスと、を少なくとも含む。本発明に係る組成物は、さらに、ラクトース等を含むこともできる。
本発明に係る組成物は、ヒトミルクオリゴ糖の混合物と、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスと、を少なくとも含む。また、本発明に係る組成物は、ラクト-N-テトラオースと、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスと、を少なくとも含む。本発明に係る組成物は、さらに、ラクトース等を含むこともできる。
(1)ヒトミルクオリゴ糖の混合物
本発明で用いるヒトミルクオリゴ糖の混合物には、以下のHMOsを含む。
a.ラクト-N-ネオテトラオース(以下、「LNnT」ともいう。)
b.ラクト-N-テトラオース(以下、「LNT」ともいう。)
c.2’-フコシルラクトース(以下、「2’-FL」ともいう。)
d.3’-シアリルラクトース(以下、「3’-SL」ともいう。)
e.6’-シアリルラクトース(以下、「6’-SL」ともいう。)
本発明で用いるヒトミルクオリゴ糖の混合物には、以下のHMOsを含む。
a.ラクト-N-ネオテトラオース(以下、「LNnT」ともいう。)
b.ラクト-N-テトラオース(以下、「LNT」ともいう。)
c.2’-フコシルラクトース(以下、「2’-FL」ともいう。)

また、本発明で用いるヒトミルクオリゴ糖の混合物には、以下のHMOsを含んでもよい。
f.ジフコシルラクトース(以下、「DFL」ともいう。)又は3’-フコシルラクトース(以下、「3’-FL」ともいう。)

HMOsの混合物中の各HMOsの含有量は、目的に応じて適宜設定することができるが、それぞれ下記の範囲に設定することが好ましい。
a.LNnT:2~10質量%
b.LNT:10~20質量%
c.2’-FL:55~75質量%
d.3’-SL:1~10質量%
e.6’-SL:5~15質量%
f.DFL又は3’-FL:1~10質量%

本発明で用いるHMOsの混合物には、これらの各種HMOsの他に、一般的に知られている各種HMOsを1種又は2種以上含ませることもできる。上記各種HMOsとしては、例えば、3-ジフコシルラクトース、3-フコシル-3’-シアリルラクトース、ラクト-N-フコペンタオースI、ラクト-N-フコペンタオースII、ラクト-N-フコペンタオースIII、ラクト-N-フコペンタオースV、ラクト-N-ジフコシルヘキサオースI、ラクト-N-ジフコシルヘキサオースII、ラクト-N-シアリルペンタオース、LSTa、LSTb及びLSTcが挙げられる。
本発明で用いるHMOsの混合物に含ませる各種HMOsは、市販品を使用してもよいし、乳から調製してもよいし、公知の有機合成又は酵素処理等の方法によって得てもよい。また、各種HMOsを含む乳(例えば、母乳、粉ミルク、牛乳又は乳製品)を使用してもよい。また、各種HMOsを乳(例えば、牛乳、粉ミルク又は育乳用ミルク)に配合した組成物(すなわち、HMOsを含む乳)を使用してもよい。
なお、本発明で用いるLNTは、以上説明したHMOsの混合物として用いなくてもよく、本発明に係る組成物は、LNTと、後述するビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌と、からなる組成物であってもよい。
(2)ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌
本発明で用いることができるビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス(なお、「ビフィドバクテリウム・インファンティス」は、「ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス」に再分類されており、同義のものである)に属する細菌としては、本発明の効果を損なわない限り、公知又は未知のビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌を1種又は2種以上、自由に選択して用いることができる。具体的に挙げるとすれば、例えば、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス(NITE BP-02623)、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス(ATCC15697T)等を挙げることができる。
本発明で用いることができるビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス(なお、「ビフィドバクテリウム・インファンティス」は、「ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス」に再分類されており、同義のものである)に属する細菌としては、本発明の効果を損なわない限り、公知又は未知のビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌を1種又は2種以上、自由に選択して用いることができる。具体的に挙げるとすれば、例えば、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス(NITE BP-02623)、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス(ATCC15697T)等を挙げることができる。
ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス(NITE BP-02623)は、2018年1月26日に独立行政法人製品評価技術基盤機構 特許微生物寄託センター(〒292-0818 千葉県木更津市かずさ鎌足2-5-8 122号室)にブダペスト条約に基づく国際寄託がなされている。
ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス(ATCC15697T)は、ATCCアメリカ国立菌培養収集所(アメリカ合衆国ヴァージニア州20110-2209マナサス10801ユニバーシティブルバード)から入手可能である。
なお、上記例示した菌株名で特定される菌株には、当該菌株名で所定の機関に寄託及び登録がなされている株そのもの(以下、説明の便宜上、「寄託株」ともいう。)に限られず、それと実質的に同等な株(以下、「派生株」又は「誘導株」ともいう。)も包含される。すなわち、例えば、「ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス(NITE BP-02623)」には、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス(NITE BP-02623)の寄託番号で上記寄託機関に寄託されている株そのものに限られず、それと実質的に同等な株も包含される。
上記寄託株と実質的に同等の株は、例えば、当該寄託株を親株とする派生株であってよい。派生株としては、寄託株から育種された株又は寄託株から自然に生じた株が挙げられる。

派生株は下記のような株が挙げられる。

(2)当該寄託菌株由来の遺伝子のみ保有し、外来由来の遺伝子を持たず、DNAの同一性が95%以上である菌株
(3)当該菌株から育種された株若しくは、遺伝子工学的改変、突然変異又は自然突然変異を含む同一の形質を有する株

これらのビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌は、前述したHMOsの混合物の存在下において、増殖が促進される。即ち、前述したHMOsの混合物を用いることで、腸内において、これらのビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌を容易に増殖することができる。
ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌は、様々な生理機能が報告されており、その機能は腸内での増殖及び産生する物質(例えば、酢酸)によるものであると報告されている。従って、本発明のビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌も、安全性が高く、かつ幅広い年齢層において、一般的に云われているビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌による効能を期待することができる。このため、本発明の組成物は、幅広い用途(飲食品用、機能性食品用、医薬品用、飼料用等)に用いることが可能である。また、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌は、プロバイオティクス効果を期待することができるので、本発明の組成物は、健康増進、食生活改善、腸内環境改善、腸内感染予防・治療等の目的に使用することも可能である。
本発明に用いるビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌は、例えば、同菌株を培養することにより増殖させることができる。
培養する方法は、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌が増殖できる限り特に限定されず、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌の培養に通常用いられる方法を必要により適宜修正して用いることができる。例えば、培養温度は30~50℃でよく、35~45℃であることが好ましい。また培養は嫌気条件下で行うことが好ましく、例えば、炭酸ガス等の嫌気ガスを通気しながら培養することができる。また、液体静置培養等の微好気条件下で培養してもよい。

本発明に用いるビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌を増殖させるために培養する培地としては、特に限定されず、本発明に用いるビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌の培養に通常用いられる培地を必要により適宜修正して用いることができる。すなわち、各種HMOs以外の炭素源としては、例えば、ガラクトース、グルコース、フルクトース、マンノース、セロビオース、マルトース、ラクトース、スクロース、トレハロース、デンプン加水分解物又は廃糖蜜等の糖類を資化性に応じて使用できる。窒素源としては、例えば、アンモニア、硫酸アンモニウム、塩化アンモニウム若しくは硝酸アンモニウム等のアンモニウム塩類又は硝酸塩類を使用できる。また、無機塩類としては、例えば、塩化ナトリウム、塩化カリウム、硫酸カリウム、硫酸マグネシウム、塩化カルシウム、硝酸カルシウム、塩化マンガン又は硫酸第一鉄を用いることができる。また、ペプトン、大豆粉、脱脂大豆粕、肉エキス又は酵母エキス等の有機成分を用いてもよい。また、調製済みの培地としては、例えば、MRS培地を用いることができる。
(3)その他
本発明に係る組成物には、本発明の効果を損なわない限り、任意の成分を含ませることも可能である。例えば、ヤシ油、大豆油、モノグリセリド及びジグリセリド等の脂肪分;グルコース、ラクトース及びデンプン加水分解物等の炭水化物;大豆タンパク質及び乳タンパク質等のタンパク質;ビタミン及びミネラル(カルシウム、リン、カリウム、ナトリウム、塩化物、マグネシウム、マンガン、鉄、銅、亜鉛、セレン、ヨウ素、ビタミンA、E、D、C、B、及びこれらの複合体)を適宜用いることができる。
本発明に係る組成物には、本発明の効果を損なわない限り、任意の成分を含ませることも可能である。例えば、ヤシ油、大豆油、モノグリセリド及びジグリセリド等の脂肪分;グルコース、ラクトース及びデンプン加水分解物等の炭水化物;大豆タンパク質及び乳タンパク質等のタンパク質;ビタミン及びミネラル(カルシウム、リン、カリウム、ナトリウム、塩化物、マグネシウム、マンガン、鉄、銅、亜鉛、セレン、ヨウ素、ビタミンA、E、D、C、B、及びこれらの複合体)を適宜用いることができる。
2.本発明に係る組成物の用途
本発明の組成物をヒトに投与することにより、善玉菌に作用するプロバイオティクスの増殖促進効果が期待できる。即ちプロバイオティクスが腸内で増殖することにより、腸内細菌叢が改善される。腸内細菌叢が改善されることにより、腸内細菌叢に起因する疾患が改善又は予防され、治療が可能になる。
腸内細菌叢に起因する疾患としては、炎症性大腸炎、潰瘍性大腸炎、過敏性大腸炎、大腸がん、糖尿病、及び動脈硬化等が挙げられる。
さらに、本発明の組成物を投与することにより、免疫機能が増進される。免疫機能が増進することにより、花粉症、喘息、アトピー性皮膚炎等のアレルギーの予防又は改善、治療が可能になる。
本発明に係る組成物は、飲食品、調製乳、医薬品、医薬部外品又は飼料等に好適に用いることができる。
本発明の組成物をヒトに投与することにより、善玉菌に作用するプロバイオティクスの増殖促進効果が期待できる。即ちプロバイオティクスが腸内で増殖することにより、腸内細菌叢が改善される。腸内細菌叢が改善されることにより、腸内細菌叢に起因する疾患が改善又は予防され、治療が可能になる。
腸内細菌叢に起因する疾患としては、炎症性大腸炎、潰瘍性大腸炎、過敏性大腸炎、大腸がん、糖尿病、及び動脈硬化等が挙げられる。
さらに、本発明の組成物を投与することにより、免疫機能が増進される。免疫機能が増進することにより、花粉症、喘息、アトピー性皮膚炎等のアレルギーの予防又は改善、治療が可能になる。
本発明に係る組成物は、飲食品、調製乳、医薬品、医薬部外品又は飼料等に好適に用いることができる。
なお、本実施形態の用途は、治療目的使用であっても、非治療目的使用であってもよい。
「非治療目的」とは、医療行為、すなわち、治療による人体への処置行為を含まない概念である。例えば、健康増進及び美容行為が挙げられる。
「改善」とは、疾患、症状又は状態の好転;疾患、症状又は状態の悪化防止、遅延;疾患又は症状の進行の逆転、防止又は遅延をいう。
「予防」とは、適用対象における疾患若しくは症状の発症の防止若しくは遅延、又は適用対象の疾患若しくは症状の危険性の低下をいう。
「非治療目的」とは、医療行為、すなわち、治療による人体への処置行為を含まない概念である。例えば、健康増進及び美容行為が挙げられる。
「改善」とは、疾患、症状又は状態の好転;疾患、症状又は状態の悪化防止、遅延;疾患又は症状の進行の逆転、防止又は遅延をいう。
「予防」とは、適用対象における疾患若しくは症状の発症の防止若しくは遅延、又は適用対象の疾患若しくは症状の危険性の低下をいう。
本発明に係る組成物において、上記HMOsの混合物又はLNTの含有量は、本発明の効果を損なわない限り、自由に設定することができる。本発明では、特に、組成物中の上記HMOsの混合物又はLNTの含有量を、最終組成物に対し、0.05%~20.0%に設定することができる。
また、本発明において、上記HMOsの混合物又はLNTの1日の投与量についても、本発明の効果を損なわない限り、自由に設定することができる。本発明では、特に、一般的な母乳中に含まれるHMOs濃度と同等に設定することが好ましい。具体的には、例えば、200mg~20g/1日、好ましくは300mg~15g/1日、より好ましくは400mg~10g/1日、さらに好ましくは500mg~10g/1日、最も好ましくは1g~10g/1日に設定することができる。
本発明に係る組成物において、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌は、最終組成物に対し、約1×103~約1×1012CFU/g又はmLで投与されることが好ましいが、これよりも高い数値でも許容される。組成物が成人のヒト(すなわち、16歳を超えるヒト)に投与される場合、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌は、好ましくは少なくとも1×106CFU/g又はmL以上、より好ましくは少なくとも1×108CFU/g又はmL以上で投与されることが好ましい。組成物が小児(すなわち、16歳未満のヒト)に投与される場合、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌は、好ましくは約1×106~約1×1011CFU/g又はmL、より好ましくは約1×106~約1×109CFU/g又はmLの範囲で投与されることが好ましい。なお、本発明において、CFUとは、Colony forming unitの意味であり、コロニー形成単位を表す。上記細菌が死菌の場合、CFUは、個細胞(cells)と置き換えることができる。
また、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌の使用量は、安全性が高いので特に制限されないが、例えば、1×106~1×1012CFU/kg体重/日が好ましく、1×107~1×1011CFU/kg体重/日がより好ましく、1×108~1×1010CFU/kg体重/日がさらに好ましい。又は、1個体(Body weight)あたりの使用量(投与量)としては、107~1014CFU/日が好ましく、108~1013CFU/日がより好ましく、109~1012CFU/日がさらに好ましい。
本発明の投与対象は、乳幼児が好ましい。
乳幼児とは、乳児及び幼児を含み、さらに詳細には、乳児、幼児及び新生児を含み、さらに詳細には、乳児、幼児、新生児、未熟児、早産児及び低出生体重児を含む。本発明において、乳幼児の種には、特に断りがない限り、ヒトが含まれる。乳児とは、乳児期にある子供を指し、乳児期とは母乳等の乳を主な栄養源としている時期を意味し、ヒトの場合、通常では1歳未満が乳児期にあたる。幼児とは、一般には就学前までの時期にある子供を指す。新生児とは、新生児期にある子供を指し、新生児期とは出生後の間もない時期を意味し、ヒトの場合、通常では出生後から4週間以内が新生児期にあたる。
乳幼児とは、乳児及び幼児を含み、さらに詳細には、乳児、幼児及び新生児を含み、さらに詳細には、乳児、幼児、新生児、未熟児、早産児及び低出生体重児を含む。本発明において、乳幼児の種には、特に断りがない限り、ヒトが含まれる。乳児とは、乳児期にある子供を指し、乳児期とは母乳等の乳を主な栄養源としている時期を意味し、ヒトの場合、通常では1歳未満が乳児期にあたる。幼児とは、一般には就学前までの時期にある子供を指す。新生児とは、新生児期にある子供を指し、新生児期とは出生後の間もない時期を意味し、ヒトの場合、通常では出生後から4週間以内が新生児期にあたる。
さらに本発明においては、前述した各種HMOsと、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌に加えて、プレバイオティクスも併せて用いてもよい。プレバイオティクスとは、大腸内の特定の細菌の増殖及び活性を選択的に変化させることより、宿主に有利な影響を与え、宿主の健康を改善する難消化性食品成分のことである。
プレバイオティクスは、本細菌又はその培養物とともに摂取させたときに本発明の効果をより発揮し得るものであれば特に制限されないが、例えば、ラクチュロース、ラフィノース、ガラクトオリゴ糖、フラクトオリゴ糖、大豆オリゴ糖、乳果オリゴ糖、キシロオリゴ糖、イソマルオリゴ糖、コーヒー豆マンノオリゴ糖、グルコン酸、ポリデキストロース及びイヌリンが挙げられ、ラクチュロース、ラフィノース及びガラクトオリゴ糖が好ましく、ラクチュロース、ラフィノース及びガラクトオリゴ糖がより好ましい。
プレバイオティクスは、本細菌又はその培養物とともに摂取させたときに本発明の効果をより発揮し得るものであれば特に制限されないが、例えば、ラクチュロース、ラフィノース、ガラクトオリゴ糖、フラクトオリゴ糖、大豆オリゴ糖、乳果オリゴ糖、キシロオリゴ糖、イソマルオリゴ糖、コーヒー豆マンノオリゴ糖、グルコン酸、ポリデキストロース及びイヌリンが挙げられ、ラクチュロース、ラフィノース及びガラクトオリゴ糖が好ましく、ラクチュロース、ラフィノース及びガラクトオリゴ糖がより好ましい。
<栄養組成物>
本発明に係る組成物は、栄養組成物として用いることができる。本発明において、「栄養組成物」は、経口又は静脈摂取され、接種者に栄養を与える飲食品を指す。本発明で用いることができる栄養組成物の種類は特に限定されないが、好ましくは調製乳又は流動食等であり、より好ましくは調製乳である。摂取対象は、乳児、幼児、小児及び成人を問わないが、好ましくは乳児及び幼児である。
調製乳には、調製粉乳及び調製液状乳が含まれる。
調製粉乳は、乳及び乳製品の成分規格等に関する省令(乳等省令)において、「生乳、牛乳、特別牛乳、又はこれらを原料として製造した食品を加工し、又は主要原料とし、乳幼児に必要な栄養分を加え粉末状にしたもの」として定義される。
調製液状乳は、上記省令において、「生乳、牛乳、特別牛乳、又はこれらを原料として製造した食品を加工し、又は主要原料とし、乳幼児に必要な栄養分を加え液状にしたもの」として定義される。
また、調製乳は、各種の蛋白質、油脂、炭水化物、ミネラル類又はビタミン類等の栄養成分が配合されたものであって、粉末状又は液状に加工されたものも含まれる。
また、調製乳にはさらに、健康増進法で規定される特別用途食品における「乳児用調製粉乳」、「乳児用調製液状乳」及び「妊産婦、授乳婦用粉乳」が含まれ、0~12か月の乳児を対象とする乳幼児用調製粉乳、6~9か月以降の乳児及び年少幼児(3歳まで)を対象とするフォローアップミルク、出生時の体重が2500g未満の新生児(低出生体重児)を対象とする低出生体重児用調製粉乳、牛乳アレルギー又は乳糖不耐症等の病的状態を有する乳幼児の治療に用いられるアレルギー用調製乳、乳糖不耐症用調製乳、先天代謝異常用調製乳等の各種治療用ミルク、幼児向け調製粉乳、学童用調製乳、成人用調製乳、高齢者用調製乳、ベビーフード、幼児用フード、母乳又は調製乳用強化剤、成人用栄養粉末、及び高齢者用栄養粉末等の態様も含まれる。さらに、上記組成物を保健機能食品又は病者用食品に適用することができる。保健機能食品制度は、内外の動向、及び従来からの特定保健用食品制度との整合性を踏まえて、通常の食品のみならず錠剤、又はカプセル等の形状をした食品を対象として設けられたもので、特定保健用食品(個別許可型)と栄養機能食品(規格基準型)の2種類の類型からなる。
なお、本発明において、各種栄養組成物、各種調製乳、各種食品等の定義は、各国、各国際機関、各連合機関、各連合機構、及びこれらに属する各種機関、各種機構、各種団体、各種会等が定義する内容を包含する。
本発明に係る組成物は、栄養組成物として用いることができる。本発明において、「栄養組成物」は、経口又は静脈摂取され、接種者に栄養を与える飲食品を指す。本発明で用いることができる栄養組成物の種類は特に限定されないが、好ましくは調製乳又は流動食等であり、より好ましくは調製乳である。摂取対象は、乳児、幼児、小児及び成人を問わないが、好ましくは乳児及び幼児である。
調製乳には、調製粉乳及び調製液状乳が含まれる。
調製粉乳は、乳及び乳製品の成分規格等に関する省令(乳等省令)において、「生乳、牛乳、特別牛乳、又はこれらを原料として製造した食品を加工し、又は主要原料とし、乳幼児に必要な栄養分を加え粉末状にしたもの」として定義される。
調製液状乳は、上記省令において、「生乳、牛乳、特別牛乳、又はこれらを原料として製造した食品を加工し、又は主要原料とし、乳幼児に必要な栄養分を加え液状にしたもの」として定義される。
また、調製乳は、各種の蛋白質、油脂、炭水化物、ミネラル類又はビタミン類等の栄養成分が配合されたものであって、粉末状又は液状に加工されたものも含まれる。
また、調製乳にはさらに、健康増進法で規定される特別用途食品における「乳児用調製粉乳」、「乳児用調製液状乳」及び「妊産婦、授乳婦用粉乳」が含まれ、0~12か月の乳児を対象とする乳幼児用調製粉乳、6~9か月以降の乳児及び年少幼児(3歳まで)を対象とするフォローアップミルク、出生時の体重が2500g未満の新生児(低出生体重児)を対象とする低出生体重児用調製粉乳、牛乳アレルギー又は乳糖不耐症等の病的状態を有する乳幼児の治療に用いられるアレルギー用調製乳、乳糖不耐症用調製乳、先天代謝異常用調製乳等の各種治療用ミルク、幼児向け調製粉乳、学童用調製乳、成人用調製乳、高齢者用調製乳、ベビーフード、幼児用フード、母乳又は調製乳用強化剤、成人用栄養粉末、及び高齢者用栄養粉末等の態様も含まれる。さらに、上記組成物を保健機能食品又は病者用食品に適用することができる。保健機能食品制度は、内外の動向、及び従来からの特定保健用食品制度との整合性を踏まえて、通常の食品のみならず錠剤、又はカプセル等の形状をした食品を対象として設けられたもので、特定保健用食品(個別許可型)と栄養機能食品(規格基準型)の2種類の類型からなる。
なお、本発明において、各種栄養組成物、各種調製乳、各種食品等の定義は、各国、各国際機関、各連合機関、各連合機構、及びこれらに属する各種機関、各種機構、各種団体、各種会等が定義する内容を包含する。
<飲食品>
本発明に係る組成物は、公知の飲食品に添加して調製することもできる。飲食品の原料中に組成物を混合して新たな飲食品を製造することもできる。

本発明に係る組成物を含む飲食品は、液状、ペースト状、固体又は粉末等の形態を問わず、錠菓及び流動食等のほか、例えば、小麦粉製品、即席食品、農産加工品、水産加工品、畜産加工品、乳・乳製品、油脂類、基礎調味料、複合調味料・食品類、冷凍食品、菓子類、飲料及びこれら以外の市販品が挙げられる。
上記小麦粉製品としては、例えば、パン、マカロニ、スパゲッティ、めん類、ケーキミックス、から揚げ粉及びパン粉が挙げられる。
上記即席食品類としては、例えば、即席めん、カップめん、レトルト・調理食品、調理缶詰め、電子レンジ食品、即席スープ・シチュー、即席みそ汁・吸い物、スープ缶詰め、フリーズ・ドライ食品及びその他の即席食品が挙げられる。
上記農産加工品としては、例えば、農産缶詰め、果実缶詰め、ジャム・マーマレード類、漬物、煮豆類、農産乾物類及びシリアル(穀物加工品)が挙げられる。
上記水産加工品としては、例えば、水産缶詰め、魚肉ハム・ソーセージ、水産練り製品、水産珍味類及びつくだ煮類が挙げられる。
上記畜産加工品としては、例えば、畜産缶詰め・ペースト類及び畜肉ハム・ソーセージが挙げられる。
上記乳・乳製品としては、例えば、発酵乳、加工乳、乳飲料、ヨーグルト類、乳酸菌飲料類、チーズ、アイスクリーム類、調製乳類、クリーム及びその他の乳製品が挙げられる。
上記油脂類としては、例えば、バター、マーガリン類及び植物油が挙げられる。
上記基礎調味料としては、例えば、しょうゆ、みそ、ソース類、トマト加工調味料、みりん類及び食酢類が挙げられる。
上記複合調味料・食品類として、調理ミックス、カレーの素類、たれ類、ドレッシング類、めんつゆ類、スパイス類及びその他の複合調味料が挙げられる。
上記冷凍食品としては、例えば、素材冷凍食品、半調理冷凍食品及び調理済冷凍食品が挙げられる。
上記菓子類としては、例えば、キャラメル、キャンディー、チューインガム、チョコレート、クッキー、ビスケット、ケーキ、パイ、スナック、クラッカー、和菓子、米菓子、豆菓子、デザート菓子及びその他の菓子が挙げられる。
上記飲料としては、例えば、炭酸飲料、天然果汁、果汁飲料、果汁入り清涼飲料、果肉飲料、果粒入り果実飲料、野菜系飲料、豆乳、豆乳飲料、コーヒー飲料、お茶飲料、粉末飲料、濃縮飲料、スポーツ飲料、栄養飲料、アルコール飲料及びその他の嗜好飲料が挙げられる。
上記以外の市販食品としては、例えば、ベビーフード、ふりかけ及びお茶漬けのりが挙げられる。
上記即席食品類としては、例えば、即席めん、カップめん、レトルト・調理食品、調理缶詰め、電子レンジ食品、即席スープ・シチュー、即席みそ汁・吸い物、スープ缶詰め、フリーズ・ドライ食品及びその他の即席食品が挙げられる。

〔機能性表示飲食品〕
また、本発明で定義される飲食品等は、特定の用途(特に保健の用途)又は機能が表示された飲食品として提供・販売されることも可能である。
「表示」行為には、需要者に対して上記用途を知らしめるための全ての行為が含まれ、上記用途を想起・類推させうるような表現であれば、表示の目的、表示の内容、表示する対象物・媒体等の如何に拘わらず、全て本発明の「表示」行為に該当する。
また、本発明で定義される飲食品等は、特定の用途(特に保健の用途)又は機能が表示された飲食品として提供・販売されることも可能である。
「表示」行為には、需要者に対して上記用途を知らしめるための全ての行為が含まれ、上記用途を想起・類推させうるような表現であれば、表示の目的、表示の内容、表示する対象物・媒体等の如何に拘わらず、全て本発明の「表示」行為に該当する。
また、「表示」は、需要者が上記用途を直接的に認識できるような表現により行われることが好ましい。具体的には、飲食品に係る商品又は商品の包装に上記用途を記載したものを譲渡し、引き渡し、譲渡若しくは引き渡しのために展示し、輸入する行為、商品に関する広告、価格表若しくは取引書類に上記用途を記載して展示し、若しくは頒布し、又はこれらを内容とする情報に上記用途を記載して電磁気的(インターネット等)方法により提供する行為等が挙げられる。
一方、表示内容としては、行政等によって認可された表示(例えば、行政が定める各種制度に基づいて認可を受け、そのような認可に基づいた態様で行う表示)であることが好ましい。また、そのような表示内容を、包装、容器、カタログ、パンフレット、若しくはPOP等の販売現場における宣伝材、又はその他の書類等へ付することが好ましい。
また、「表示」には、健康食品、機能性食品、病者用食品、経腸栄養食品、特別用途食品、保健機能食品、特定保健用食品、機能性表示食品、栄養機能食品、又は医薬用部外品等としての表示も挙げられる。この中でも特に、消費者庁によって認可される表示、例えば、特定保健用食品制度、機能性表示食品制度、又はこれらに類似する制度にて認可される表示が挙げられる。より具体的には、特定保健用食品としての表示、条件付き特定保健用食品としての表示、機能性表示食品としての表示、身体の構造及び機能に影響を与える旨の表示、又は疾病リスク低減表示等を挙げることができる。この中でも典型的な例としては、健康増進法施行規則(平成15年4月30日日本国厚生労働省令第86号)に定められた特定保健用食品としての表示(特に保健の用途の表示)、又は食品表示法(平成25年法律第70号)に定められた機能性表示食品としての表示及びこれらに類する表示である。
本発明に係る組成物を投与することにより、投与対象の腸内中にビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌が増殖する。このプロバイオティクスが腸内で増殖することにより、免疫力が向上し、さらに腸内フローラが改善する。特に乳幼児においては、本発明に係る組成物を投与することにより、腸内のビフィズス菌が有効に増加し、腸内環境が整う。さらにビフィズス菌とともに悪玉菌又は日和見菌を取り入れることにより免疫力が獲得でき、出来上がった腸内細菌フローラ等の腸内環境はそのまま大人になっても引き継がれる。
調製乳の製造方法は、本発明の効果を損なわない限り、公知の製造方法を1種又は2種以上自由に組み合わせて用いることができる。具体的には、例えば、原料乳に本発明に係る組成物及び各種添加物が混合され、均質化、殺菌、濃縮、乾燥、造粒、及び篩別等の工程を経て製造することができる。
本発明の調製乳は、具体的には、例えば、以下の方法により製造できる。
すなわち、本発明は、上記HMOsの混合物又はLNTと、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌を含む菌体粉末と、を混合し、調製乳を得る、調製乳の製造方法を提供する。
具体的には、例えば、下記の工程A及び工程Bを含む、調製乳の製造方法を提供する:
工程A:任意の成分を含む培地でビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌を培養し、培養物を得る工程;
工程B:上記培養物と、上記HMOsの混合物又はLNTと、を混合し、調製乳を得る工程。
具体的には、例えば、下記の工程A及び工程Bを含む、調製乳の製造方法を提供する:
工程A:任意の成分を含む培地でビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌を培養し、培養物を得る工程;
工程B:上記培養物と、上記HMOsの混合物又はLNTと、を混合し、調製乳を得る工程。
また、本発明の食品組成物は、具体的には、例えば、乳幼児用サプリメントであってよい。乳幼児用サプリメントは、例えば、以下の方法により製造できる。
すなわち、本発明は、例えば、下記工程C~工程Eを含む、サプリメントの製造方法を提供する:
工程C:任意の成分を含む培地でビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌を培養し、培養物を得る工程;
工程D:上記培養物と、上記HMOsの混合物又はLNTと、賦形剤と、を混合し、混合物を得る工程;
工程E:上記混合物を打錠等により製剤化する工程。
工程C:任意の成分を含む培地でビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌を培養し、培養物を得る工程;
工程D:上記培養物と、上記HMOsの混合物又はLNTと、賦形剤と、を混合し、混合物を得る工程;
工程E:上記混合物を打錠等により製剤化する工程。
なお、上記工程A及び工程Cに用いる培地には、上記HMOsの混合物又はLNTを含ませることも可能である。
また、上記いずれの製造方法においても、適宜、上記工程で言及した成分以外の成分を併用してよい。
本発明に係る組成物の添加方法は、本発明の効果を損なわない限り、特に限定されないが、例えば、本発明に係る組成物を、上記のように混合時に液状で添加する他に、粉末化後に添加して粉体混合し、製造することができる。
本発明に係る調製乳中の上記HMOsの混合物又はLNTの含有量は、本発明の効果を損なわない限り、自由に設定することができる。本発明では、特に、調製乳中の上記HMOsの混合物又はLNTの含有量を、調製乳の最終組成物に対し、0.05%~20.0%に設定することができる。
本発明に係る調製乳中のビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌の含有量は、本発明の効果を損なわない限り、自由に設定することができる。本発明では、特に、調製乳中のビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌の含有量を、調製乳の最終組成物に対し、1×103~1×1012CFU/mL又はgに設定することができる。
<医薬品、医薬部外品>
本発明に係る組成物を、公知の医薬品又は医薬部外品(以下、「医薬品等」ともいう。)に添加してもよい。医薬品等の原料中に、本発明に係る組成物を混合して新たな医薬品等を製造することができる。

医薬品等に本発明に係る組成物を含む場合、該医薬品等は、経口投与又は非経口投与等の投与方法に応じて、適宜所望の剤形に製剤化することができる。その剤形は特に限定されないが、経口投与の場合、例えば、散剤、顆粒剤、錠剤、トローチ剤又はカプセル剤等の固形製剤;溶液剤、シロップ剤、懸濁剤又は乳剤等の液剤等に製剤化することができる。非経口投与の場合、例えば、座剤、噴霧剤、吸入剤、軟膏剤、貼付剤、又は注射剤に製剤化することができる。本発明では、経口投与の剤形に製剤化することが好ましい。
なお、製剤化は剤形に応じて、適宜、公知の方法により実施できる。

製剤化に際しては、適宜製剤担体を配合する等して製剤化してもよい。また、本発明のペプチドのほか、通常製剤化に用いられている賦形剤、pH調整剤、着色剤又は矯味剤等の成分を用いることができる。さらに、公知の又は将来的に見出される疾患又は症状の予防、改善及び/又は治療の効果を有する成分を、適宜目的に応じて併用することも可能である。
上記製剤担体としては、剤形に応じて、各種有機又は無機の担体を用いることができる。固形製剤の場合の担体としては、例えば、賦形剤、結合剤、崩壊剤、滑沢剤、安定剤、及び矯味矯臭剤が挙げられる。
上記賦形剤としては、例えば、乳糖、白糖、ブドウ糖、マンニット及びソルビット等の糖誘導体;トウモロコシデンプン、馬鈴薯デンプン、α-デンプン、デキストリン及びカルボキシメチルデンプン等のデンプン誘導体;結晶セルロース、ヒドロキシプロピルセルロース、ヒドロキシプロピルメチルセルロース、カルボキシメチルセルロース及びカルボキシメチルセルロースカルシウム等のセルロース誘導体;アラビアゴム;デキストラン;プルラン;軽質無水珪酸、合成珪酸アルミニウム及びメタ珪酸アルミン酸マグネシウム等の珪酸塩誘導体;リン酸カルシウム等のリン酸塩誘導体;炭酸カルシウム等の炭酸塩誘導体;並びに硫酸カルシウム等の硫酸塩誘導体が挙げられる。
上記結合剤としては、例えば、上記賦形剤の他、ゼラチン、ポリビニルピロリドン及びマクロゴールが挙げられる。
上記崩壊剤としては、例えば、上記賦形剤の他、クロスカルメロースナトリウム、カルボキシメチルスターチナトリウム、及び架橋ポリビニルピロリドン等の化学修飾されたデンプン又はセルロース誘導体が挙げられる。
上記滑沢剤としては、例えば、タルク;ステアリン酸;ステアリン酸カルシウム及びステアリン酸マグネシウム等のステアリン酸金属塩;コロイドシリカ;ピーガム及びゲイロウ等のワックス類;硼酸;グリコール;フマル酸及びアジピン酸等のカルボン酸類;安息香酸ナトリウム等のカルボン酸ナトリウム塩;硫酸ナトリウム等の硫酸塩類;ロイシン;ラウリル硫酸ナトリウム及びラウリル硫酸マグネシウム等のラウリル硫酸塩;無水珪酸及び珪酸水和物等の珪酸類;並びにデンプン誘導体が挙げられる。
上記安定剤としては、例えば、メチルパラベン及びプロピルパラベン等のパラオキシ安息香酸エステル類;クロロブタノール、ベンジルアルコール及びフェニルエチルアルコール等のアルコール類;塩化ベンザルコニウム;無水酢酸;並びにソルビン酸が挙げられる。
上記矯味矯臭剤としては、例えば、甘味料、酸味料及び香料が挙げられる。
なお、経口投与用の液剤の場合に使用する担体としては、例えば、水等の溶剤、及び矯味矯臭剤が挙げられる。

<飼料>
本発明に係る組成物を、公知の飼料に添加してもよい。飼料の原料中に組成物を混合して飼料を製造することができる。

飼料に本発明に係る組成物を含む場合、飼料の原料としては、例えば、トウモロコシ、小麦、大麦及びライ麦等の穀類;ふすま、麦糠、米糠及び脱脂米糠等の糠類;コーングルテンミール及びコーンジャムミール等の製造粕類;脱脂粉乳、ホエイ、魚粉及び骨粉等の動物性飼料類;ビール酵母等の酵母類;リン酸カルシウム及び炭酸カルシウム等の鉱物質飼料;油脂類;アミノ酸類;並びに糖類が挙げられる。また、上記飼料の形態としては、例えば、ペットフード等の愛玩動物用飼料、家畜飼料及び養魚飼料が挙げられる。
以下、実施例に基づいて本発明をさらに詳細に説明する。なお、以下に説明する実施例は、本発明の代表的な実施例の一例を示したものであり、これにより本発明の範囲が狭く解釈されることはない。
ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスに属する細菌の一例として、森永乳業株式会社所有のビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス NITE BP-02623について、糖源を表1に記載のヒトミルクオリゴ糖(Jennewein Biotechnologie GmbH社製)に変更したMRS(de Man-Rogosa-Sharpe)液体培地(組成:表2)をそれぞれ作成し、16時間脱気した培地200μLに菌液を1v/v%ずつ接種し、嫌気条件下で37℃にて培養した。培養16時間後の濁度(OD660)を測定し、実施例1及び比較例1の糖比率におけるビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスの増殖を比較した。
結果を、図1に示す。図1のグラフに示す通り、比較例1の糖比率の場合と比較して、実施例の糖比率の場合の方が、高い増殖活性を示した。この結果から、LNnT、LNT、2’-FL、3’-SL、及び6’-SLからなるHMOsの混合物を用いることで、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスの増殖が促進されることが証明された。
[製造例]
下記に組成物、医薬品組成物、発酵乳及び調製乳の製造例を示す。

<製造例1>
ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスを、MRS培地3mLに添加し、37℃で24時間嫌気培養後、培養液を濃縮し、凍結乾燥を行い、凍結乾燥粉末を得る。得られた凍結乾燥粉末と、LNnT、LNT、2’-FL、3’-SL、及び6’-SLを均一に混合して組成物を得る。
ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスを、MRS培地3mLに添加し、37℃で24時間嫌気培養後、培養液を濃縮し、凍結乾燥を行い、凍結乾燥粉末を得る。得られた凍結乾燥粉末と、LNnT、LNT、2’-FL、3’-SL、及び6’-SLを均一に混合して組成物を得る。
<製造例2>
ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスを、MRS培地3mLに添加し、37℃で24時間嫌気培養後、培養液を濃縮し、凍結乾燥を行い、凍結乾燥粉末を得る。次に、得られた凍結乾燥粉末と、LNnT、LNT、2’-FL、3’-SL、6’-SL、及び結晶セルロースを撹拌造粒機に投入し混合する。その後、精製水を加え造粒、造粒物を乾燥し、造粒物(医薬品組成物)を得る。
ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスを、MRS培地3mLに添加し、37℃で24時間嫌気培養後、培養液を濃縮し、凍結乾燥を行い、凍結乾燥粉末を得る。次に、得られた凍結乾燥粉末と、LNnT、LNT、2’-FL、3’-SL、6’-SL、及び結晶セルロースを撹拌造粒機に投入し混合する。その後、精製水を加え造粒、造粒物を乾燥し、造粒物(医薬品組成物)を得る。
<製造例3>
ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス、及び本発明で用いる各種HMOsを添加した発酵乳の製造法を下記に示す。
まず、乳原料、及び必要に応じた水、その他の成分等を混合し、好ましくは均質化処理を行い、加熱殺菌処理する。均質化処理及び加熱殺菌処理は常法により行うことができる。加熱殺菌された殺菌調乳液に乳酸菌スターターを添加(接種)し、所定の発酵温度に保持して発酵させ、発酵物を得る。発酵によりカードが形成される。
ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス、及び本発明で用いる各種HMOsを添加した発酵乳の製造法を下記に示す。
まず、乳原料、及び必要に応じた水、その他の成分等を混合し、好ましくは均質化処理を行い、加熱殺菌処理する。均質化処理及び加熱殺菌処理は常法により行うことができる。加熱殺菌された殺菌調乳液に乳酸菌スターターを添加(接種)し、所定の発酵温度に保持して発酵させ、発酵物を得る。発酵によりカードが形成される。
乳酸菌スターターとしては、例えば、ラクトバチルス・ブルガリクス、ラクトコッカス・ラクチス、ストレプトコッカス・サーモフィラス等のヨーグルト製造に通常用いられている乳酸菌を用いることができる。pHが目標の値に達したら、形成されたカードを撹拌により破砕し、10℃以下に冷却して発酵物を得る。10℃以下に冷却することにより、乳酸菌の活性を低下させて酸の生成を抑制することができる。
次いで、発酵工程で得られた発酵物を加熱処理して加熱処理後の発酵物を得る。発酵物を適度に加熱することにより、加熱処理後の発酵物中の乳酸菌による酸の生成を抑えることができる。これによって、その後の製造工程中及び/又は濃縮発酵乳の保存中のpHの低下を抑えることができ、その結果、ビフィズス菌の生残性を向上させることができる。
上記加熱処理工程で得られた加熱後発酵物に、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス、LNnT、LNT、2’-FL、3’-SL、及び6’-SLを添加する。ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスの添加量は、加熱処理後の発酵物に対して1×107~1×1011CFU/mLが好ましく、1×108~1×1010CFU/mLがより好ましい。ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスが死菌の場合、CFU/mLは、個細胞/mLと置き換えることができる。
加熱処理後の発酵物に、ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス、及び各種HMOsを添加した後、濃縮を行う。濃縮工程は公知の濃縮方法を適宜用いて行うことができる。遠心分離法では、被濃縮物(ビフィズス菌及びプレバイオティクスが添加された加熱後発酵物)中のホエーが除去されて、固形分濃度が高められたビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス、及び本発明で用いる各種HMOsが添加された濃縮発酵乳が得られる。得られた発酵乳を摂取することにより、腸内フローラの改善が可能になる。
<製造例4>
ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス、及び本発明で用いる各種HMOsを添加した調製乳の製造法を下記に示す。
脱塩牛乳乳清蛋白質粉末(ミライ社製)10kg、牛乳カゼイン粉末(フォンテラ社製)6kg、乳糖(ミライ社製)48kg、ミネラル混合物(富田製薬社製)920g、ビタミン混合物(田辺製薬社製)32g、ラクチュロース(森永乳業社製)500g、ラフィノース(日本甜菜製糖社製)500g、ガラクトオリゴ糖液糖(ヤクルト薬品工業社製)900gを温水300kgに溶解し、さらに90℃で10分間加熱溶解し、調製脂肪(太陽油脂社製)28kgを添加して均質化する。その後、殺菌、濃縮の工程を行って噴霧乾燥し、調製乳約95kgを調製する。これに、澱粉に倍散したビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスの菌体粉末100g、LNnT、LNT、2’-FL、3’-SL、及び6’-SLを加えて、調製乳約95kgを調製する。得られた調製乳を水に溶解して、標準調乳濃度である総固形分濃度14%(w/V)の調乳液を得る。得られた調乳液中のビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスの菌数として、2.7×109CFU/100mlを得ることができる。上述のようにして得られた調製乳を投与することにより、プロバイオティクスが腸内で増殖促進可能になる。
ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス、及び本発明で用いる各種HMOsを添加した調製乳の製造法を下記に示す。

上述の製造例1~4で製造された組成物、医薬品組成物、発酵乳又は調製乳をヒトに投与することにより、腸内でビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスが増殖し、腸内フローラの改善につながる。

- ラクト-N-ネオテトラオースと、
ラクト-N-テトラオースと、
2’-フコシルラクトースと、
3’-シアリルラクトースと、
6’-シアリルラクトースと、
からなるヒトミルクオリゴ糖の混合物と、
ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスと、
を含む組成物。 - 前記ビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティスがビフィドバクテリウム・ロンガム・サブスピーシーズ・インファンティス NITE BP-02623である、請求項1に記載の組成物。
- 前記ヒトミルクオリゴ糖の混合物中に、
前記ラクト-N-ネオテトラオースが、2~10質量%、
前記ラクト-N-テトラオースが、10~20質量%、
前記2’-フコシルラクトースが、55~75質量%、
