Abstract
본 발명은 특정 펩타이드를 유효성분으로 포함하는 피지 과다분비의 억제 또는 개선용 화장료 조성물 및 피지 과다분비의 예방, 개선 또는 치료용 약학 조성물을 제공한다. 상기 펩타이드는 p53과 MDM2의 상호작용을 현저하게 감소시키고. 그 결과 p53과 RNA 폴리머라아제 II(RNA Polymerase II, POLII)의 상호작용을 현저하게 증가시킴으로써, 피지세포의 사멸(apoptosis)을 유도하는 사이토카인의 발현을 유도할 수 있다.
Description
본 발명은 p53을 활성화하는 펩타이드를 포함하는 조성물에 관한 것이다. 더욱 상세하게는, 본 발명은 p53과 MDM2의 상호작용을 현저하게 감소시키고. 그 결과 p53과 POLII의 상호작용을 증가시키는 활성을 갖는 펩타이드를 유효성분으로 포함하는 피지 과다분비의 억제 또는 개선용 화장료 조성물; 및 피지 과다분비의 예방, 개선 또는 치료용 약학 조성물에 관한 것이다.
피지선(sebaceous gland)의 피지세포(sebocytes)에서 분비되는 피지는 피부 외부의 유해 물질의 침투를 막아주고 유수분을 조절하는 순기능을 한다. 그러나 사춘기 호르몬의 변화, 스트레스 등에 의해 피지(sebum)가 과잉으로 분비될 경우, 역기능을 초래하여 다양한 피부장애를 야기하게 된다. 피지 과다분비(seborrhea)는 피지의 과도한 분비를 특징으로 하는 피지선의 질환 또는 피부 트러블(skin troubles)로서, 피부 상에 번들거림(oily coating), 피딱지(crusts 혹은 scales) 등을 야기한다.
피지 과다분비의 억제 또는 개선은 주로 개인적인 비누 세안, 샴푸 사용 등의 위생적인(hygienic) 방법에 의해 이루어지고 있는 실정이다. 약물 치료법으로서 셀레늄 설파이드, 아졸계 화합물, 소듐 설파세트아마이드, 터비나핀 등의 항진균제, 스테로이드 등의 항염증제가 사용되고 있으나, 이는 증상의 경감을 위한 것으로 피지 과다분비를 근본적으로 억제, 개선, 또는 치료할 수는 없다. 따라서, 피지 과다분비를 근본적으로 억제, 개선, 또는 치료할 수 있는 방법을 개발하는 것이 당업계에 요구된다.
CD99는 당질화된 세포 밖 부위, 세포막 횡단부위와 짧은 세포 내 부위로 구성된 제1형 막횡단 단백질(Banting, G. S., Pym, B., Darling, S. M.,and Goodfellow P. N. (1989). The MIC2 gene product: epitope mapping and structural prediction analysis define an integral membrane protein. Mol. Immunol. 26: 181)로서, Ewing sarcoma (EWS) 세포에서 발현되는 CD99 분자를 항 CD99 단일클론항체로 결합시키면 E3 ubiquitin ligase인 MDM2가 소실되며, 그 결과 MDM2와 p53의 결합이 억제된다고 보고된 바 있다(Guerzoni C. et. al. (2015). CD99 triggering in Ewing sarcoma delivers a lethal signal through p53 pathway reactivation and cooperates with doxorubicin. Clin Cancer Res 21:146-156.). MDM2으로부터 풀려난 p53은 세포핵에서 RNA 폴리머라아제 II(RNA Polymerase II, POLII)을 포함한 RNA 폴리머라아제 복합체를 활성화하여 피지세포의 사멸(apoptosis)을 유도하는 TRAIL(tumour necrosis factor-related apoptosis inducing ligand)을 비롯한 하위 표적유전자의 발현을 증가시킨다(Melnik B.C. (2017). p53: key conductor of all anti-acne therapies. J Transl Med 15:195-206.). 또한, p53은 피지세포의 증식과 분화를 유도하는 IGF-1 신호전달과 c-Myc의 기능을 억제한다.
따라서, MDM2와 p53의 결합을 억제하고 p53과 POLII의 상호작용을 증가시킬 경우, 피지세포의 증식과 분화를 억제하고 사멸(apoptosis)을 유도함으로써 피지 과다분비를 근본적으로 억제, 개선, 또는 치료할 수 있을 것으로 기대된다.
본 발명자들은 특정 펩타이드가 PKA(protein tyrosine kinase)와 SHP2(Src homology region 2 domain-containing phosphatase-2)의 상호작용을 현저하게 증가시킴으로써, β1 인테그린 신호전달을 매개로 한 암세포 전이를 억제할 수 있으며, 또한 염증 질환을 예방 또는 치료에 유용하게 적용될 수 있다는 것을 밝혀낸 바 있다(대한민국 특허공개 제10-2018-0099092호). 놀랍게도, 본 발명자들은 상기 특정 펩타이드가 p53과 MDM2의 상호작용을 현저하게 감소시키고. 그 결과 p53과 POLII의 상호작용을 현저하게 증가시킴으로써, 피지 과다분비에 대한 억제, 개선, 또는 치료활성을 갖는다는 것을 발견하였다.
따라서, 본 발명은 특정 펩타이드를 유효성분으로 포함하는 피지 과다분비의 억제 또는 개선용 화장료 조성물을 제공하는 것을 목적으로 한다.
또한, 본 발명은 특정 펩타이드를 유효성분으로 포함하는 피지 과다분비의 예방, 개선 또는 치료용 약학 조성물을 제공하는 것을 목적으로 한다.

본 발명의 화장료 조성물 및 약학 조성물에 있어서, 바람직하게는 상기 펩타이드는 서열번호 2, 3, 4, 6, 7, 9 또는 18의 아미노산 서열로 구성된 펩타이드일 수 있다.
화학식 1의 펩타이드, 즉 화학식 1로 표시되는 3개의 아미노산으로 구성된 펩타이드가 p53과 MDM2의 상호작용을 현저하게 감소시키고. 그 결과 p53과 POLII의 상호작용을 현저하게 증가시킨다는 것이 본 발명에 의해 밝혀졌다. 따라서, 상기 펩타이드는 피지세포의 사멸(apoptosis)을 유도하는 사이토카인, 예를 들어 TRAIL(tumor-necrosis factor-related apoptosis-inducing ligand)의 발현을 유도함으로써, 피지 과다분비의 억제 또는 개선용 화장료 조성물 및 피지 과다분비의 예방, 개선 또는 치료용 약학 조성물에 유용하게 적용될 수 있다.

본 명세서에서, "피지 과다분비(seborrhea)"라 함은 피지의 과도한 분비를 특징으로 하는 피지선의 질환 또는 피부 트러블(skin troubles)을 말하며, 과잉분비된 피지에 의해 야기되는 여드름(acne derived from oversecretion of sebum)을 포함한다.

본 발명의 화장료 조성물 및 약학 조성물에 있어서, 서열번호 2, 3, 4, 6, 7, 9 또는 18의 아미노산 서열로 구성된 펩타이드는 p53과 MDM2의 상호작용을 현저하게 감소시키고. 그 결과 p53와 POLII의 상호작용을 증가시키는데 있어서 특히 우수한 활성을 갖는다는 것이 밝혀졌다. 따라서, 일 구현예에서, 상기 펩타이드는 서열번호 2, 3, 4, 6, 7, 9 또는 18의 아미노산 서열로 구성된 펩타이드일 수 있다.
본 발명은 따른 화장료 조성물은 예를 들어 기능성 화장료 조성물의 형태로 제조될 수 있다. 상기 화장료 조성물은 통상의 화장료 제조방법에 따라, 다양한 형태로 제조될 수 있다. 예를 들어, 상기 화장료 조성물은 상기 펩타이드를 함유하는 향장 제품, 화장수, 크림, 로오숀 등의 형태로 제조될 수 있으며, 이는 통상의 클렌징액, 수렴액 및 보습액으로 희석하여 사용될 수 있다. 또한, 상기 화장료 조성물은 화장료 조성물 분야에서 통상적으로 사용되는 안정화제, 용해화제, 비타민, 안료, 및 향료와 같은 통상적인 보조제를 포함할 수 있다. 상기 화장료 조성물에 있어서, 상기 조성물의 함량은 피지 과다분비의 억제 또는 개선 효과를 달성하기에 유효한 양, 예를 들면 조성물 총 중량에 대하여 약 0.00001 ∼ 1 중량%의 함량으로 함유될 수 있고, 바람직하게는 약 0.001 ∼ 0.1 중량%의 함량으로 함유될 수 있다.
본 발명의 약학 조성물은 약학적으로 허용가능한 담체를 포함할 수 있으며, 예를 들어, 락토즈, 옥수수전분 등의 부형제, 마그네슘 스테아레이트 등의 윤활제, 공지되어 사용가능한 유화제, 현탁제, 완충제, 등장화제 등을 포함할 수 있다. 본 발명의 약학 조성물은 경구투여 제제 형태(oral dosage form) 또는 비경구투여 제제 형태(parenteral dosage form) 형태로 제제화될 수 있으며, 바람직하게는 예를 들어, 경피 투여, 피하 투여, 근육내 투여 등의 비경구 투여형태로 제제화될 수 있다. 비경구 투여형태의 경우, 필요에 따라 완충제, 등장화제 등의 첨가제가 사용될 수 있으며, 인산완충식염수(phosphate buffered saline, PBS) 등의 담체를 포함하는 수용액제의 형태일 수 있다. 본 발명의 약학 조성물은 피지 과다분비를 예방, 개선 또는 치료하기에 적합한 양으로 환자에게 투여될 수 있으며, 그 투여용량은 통상 각 환자의 연령, 체중 및 환자의 증상에 따라 일반적으로 변화시킬 수 있는 용량, 예를 들어 1일 약 0.01 내지 10 mg/kg의 용량으로 투여될 수 있다.
이하, 본 발명을 실시예 및 시험예를 통하여 더욱 상세히 설명한다. 그러나, 이들 실시예 및 시험예는 본 발명을 예시하기 위한 것으로, 본 발명이 이들 실시예 및 시험예에 한정되는 것은 아니다.
실시예 1. 펩타이드의 합성
서열번호 1 내지 20의 펩타이드(하기 표 1 참조)는 자동화합성기(PeptrEx-R48, 펩트론사, 대전, 대한민국)를 이용하여 FMOC 고체-상 방법(FMOC solid-phase method)으로 합성하였다. 합성된 펩타이드는 C18 분석용 RP 컬럼(Shiseido capcell pak)을 사용한 역상 고속액체크로마토그래피(reverse-phase HPLC) (Prominence LC-20AB, Shimadzu사, 일본)로 정제 및 분석하였으며, 질량분석기(HP 1100 Series LC/MSD, Hewlett-Packard사, Roseville, 미국)를 이용하여 동정하였다.

실시예 2. 펩타이드를 포함하는 조성물의 제조
서열번호 1 내지 20의 펩타이드를 PBS에 각각 용해시켜, 1 M의 농도가 되도록 제조하였다. 얻어진 단백질 용액을 하기 시험예에서 사용하였다.
시험예 1. p53과 MDM2의 물리적 결합에 미치는 펩타이드의 활성 평가
본 발명의 펩타이드를 세포에 처리한 후, in situ PLA (proximity ligation assay) 방법을 통하여 p53과 MDM2의 물리적 결합에 대한 영향을 평가하였다. 24-웰 마이크로플레이트의 각 웰에 4.5 X 104 개의 HaCaT 세포(사람각질세포, CLS)를 10% 우태아혈청을 함유하는 DMEM 배지에 분주하고, 37℃, 5% CO2 인큐베이터에서 24시간 배양한 후, 배지 중 펩타이드의 농도가 1, 5, 10, 20 μM이 되도록 실시예 2의 펩타이드 용액을 가한 다음, 30분 동안 동일한 조건에서 인큐베이션하였다. 대조군은 아무것도 처리하지 않았다. 각 웰의 세포를 PBS로 세정하고 2% 포름알데히드로 15분 동안 처리하여 고정시킨 후, 0.1% TritonX-100을 5분간 처리하여 세포 안으로의 항체 투과성을 높였다. anti-p53 다클론 항체(abcam, UK) 및 anti-MDM2 다클론 항체(Santa cruz, CA, USA)를 첨가하고, In situ PLA 키트(Sigma-Aldrich)를 사용하여 제조사의 지침에 따라 PLA 프로브를 가한 다음, 하이브리디제이션(hybridization), 라이게이션(ligation), 증폭(amplification) 및 마운팅 단계를 수행하였다. 각각의 세포에서 검출되는 발광 신호(PLA signals)를 공초점 레이져 현미경(Olympus fluoview FW1000; Olympus, Tokyo, Japan)으로 측정하여 p53과 MDM2의 물리적 상호작용을 정량하였다. 그 결과는 도 1과 같다.
도 1의 결과로부터, 본 발명의 펩타이드 처리군은 미처리 대조군에 비하여 p53와 MDM의 상호작용을 유의성 있게 감소시켰음을 확인할 수 있다.
시험예 2. p53 인산화와 MDM2 소실에 미치는 펩타이드의 활성 평가
웨스턴 블럿(western blot)기법을 사용하여 p53의 인산화와 MDM2의 발현을 확인하였다. HaCaT 세포를 웰당 5 x 106이 되도록 준비한 다음 배지 중 펩타이드의 농도가 20μM이 되도록 실시예 2의 펩타이드 용액을 가한 다음 동일조건에서 5분, 15분, 30분, 60분 동안 인큐베이션하였다. 세포들을 PBS로 3회 세척한 후, 0.1 μM PMSP(phenylmethylsulfonyl fluoride), 1 ㎍/㎖ 펩스타틴 A(pepstatin A), 10 ㎍/㎖ 류펩틴(leupeptin), 1 ㎍/㎖ 아프로티닌(aprotinin)과 1 mM Na3VO4가 포함된 1% NP40 용해 완충액(lysis buffer)(1% Nonidet P40, 0.1M NaCl, 0.05M tris (pH 8.0), 5 mM EDTA)으로 용해하였다. 세포 용해물을 브래드포드 분석(Bradford assay) 방법을 이용해 정량한 후 전기영동을 위한 샘플을 준비하였다. 면역침전물을 10% 폴리아크릴아미드 겔 상에서 전기영동을 실시하였으며, 전개된 단백질을 니트로셀룰로오스 막으로 이송한 후, 블록킹 용액(blocking solution) (0.05% Tween 20과 3% 소혈청알부민이 함유된 Tris-완충 식염수(Tris-buffered saline, TBS)으로 상온에서 1시간 가량 처리하였다. 이 후 항-p53 다클론항체(Santa cruz, CA, USA)와 항-MDM2 다클론항체(Santa cruz, CA, USA)가 각각 첨가된 TBS (0.5% 소혈청알부민이 함유) 완충액에서 2시간 반응시킨 후, 0.05% Tween 20이 포함된 TBS 완충액으로 세척하였다. horseradish peroxidase conjugated anti-rabbit IgG (Santa cruz, CA, USA)로 상온에서 1시간 처리한 후, 0.05% Tween 20을 함유하는 TBS로 5회 세척하고 antibody detection kit (Ab frontier, Korea)를 사용하여 현상하였다. 동일량의 세포용해물을 전기영동한 것을 확인하기 위해 항-beta actin 단일클론항체(Santa cruz, CA, USA)를 사용하여 액틴을 인식하였다. 그 결과는 도 2와 같다.
도 2의 결과로부터 알 수 있는 바와 같이, 펩타이드 처리 시간이 15분이 되었을 때 p53의 인산화가 가장 증가하였고, 60분이 되었을 때 MDM2의 발현은 소실되었다.
시험예 3. p53과 RNA Polymerase II(POLII)의 물리적 결합에 미치는 펩타이드의 활성 평가
본 발명의 펩타이드를 세포에 처리한 후, in situ PLA (proximity ligation assay) 방법을 통하여 p53과 RNA Polymerase II(POLII)의 물리적 결합에 대한 영향을 평가하였다. 24-웰 마이크로플레이트의 각 웰에 4.5 X 104 개의 HaCaT 세포(사람각질세포, CLS)를 10% 우태아혈청을 함유하는 DMEM 배지에 분주하고, 37℃, 5% CO2 인큐베이터에서 24시간 배양한 후, 배지 중 펩타이드의 농도가 20 μM이 되도록 실시예 2의 펩타이드 용액을 가한 다음, 1시간 동안 동일한 조건에서 인큐베이션하였다. 대조군은 아무것도 처리하지 않았다. 각 웰의 세포를 PBS로 세정하고 2% 포름알데히드로 15분 동안 처리하여 고정시킨 후, 0.1% TritonX-100을 5분간 처리하여 세포 안으로의 항체 투과성을 높였다. anti-p53 다클론 항체(abcam, UK) 및 anti-POLII 다클론 항체(Santa cruz, CA, USA)를 첨가하고, In situ PLA 키트(Sigma-Aldrich)를 사용하여 제조사의 지침에 따라 PLA 프로브를 가한 다음, 하이브리디제이션(hybridization), 라이게이션(ligation), 증폭(amplification) 및 마운팅 단계를 수행하였다. 각각의 세포에서 검출되는 발광 신호(PLA signals)를 공초점 레이져 현미경(Olympus fluoview FW1000; Olympus, Tokyo, Japan)으로 측정하여 p53과 POLII의 물리적 상호작용을 정량하였다. 그 결과는 도 3과 같다.
도 3의 결과로부터, 본 발명의 펩타이드 처리군은 미처리 대조군에 비하여 p53와 POLII의 상호작용을 유의성 있게 증가시켰음을 확인할 수 있다. 특히, Pep 2, 3, 4, 6, 7, 9 및 18(즉, 서열번호 2, 3, 4, 6, 7, 9 및 18의 펩타이드)는 대조군에 비하여 p53와 POLII의 상호작용을 2.5배 이상 현저하게 증가시켰음을 확인할 수 있다.
Claims (4)
- 하기 화학식 1로 표시되는 3개의 아미노산으로 구성된 펩타이드를 유효성분으로 포함하는 피지 과다분비의 억제 또는 개선용 화장료 조성물:<화학식 1>Leu-X-Asp식 중,X는 글루타민산(Glu), 세린(Ser), 글라이신(Gly), 알라닌(Ala), 글루타민(Gln), 아르기닌(Arg), 라이신(Lys), 류신(Leu), 타이로신(Tyr), 아스파르트산(Asp), 페닐알라닌(Phe), 아스파라긴(Asn), 시스테인(Cys), 히스티딘(His), 아이소류신(Ile), 메싸이오닌(Met), 프롤린(Pro), 트레오닌(Thr), 트립토판(Trp), 또는 발린(Val)이고,-는 펩타이드 결합이다.
- 제1항에 있어서, 상기 펩타이드가 서열번호 2, 3, 4, 6, 7, 9 또는 18의 아미노산 서열로 구성된 펩타이드인 것을 특징으로 하는 화장료 조성물.
- 하기 화학식 1로 표시되는 3개의 아미노산으로 구성된 펩타이드를 유효성분으로 포함하는 피지 과다분비의 예방, 개선 또는 치료용 약학 조성물:<화학식 1>Leu-X-Asp식 중,X는 글루타민산(Glu), 세린(Ser), 글라이신(Gly), 알라닌(Ala), 글루타민(Gln), 아르기닌(Arg), 라이신(Lys), 류신(Leu), 타이로신(Tyr), 아스파르트산(Asp), 페닐알라닌(Phe), 아스파라긴(Asn), 시스테인(Cys), 히스티딘(His), 아이소류신(Ile), 메싸이오닌(Met), 프롤린(Pro), 트레오닌(Thr), 트립토판(Trp), 또는 발린(Val)이고,-는 펩타이드 결합이다.
