Abstract
본 발명은 β1 인테그린의 활성화를 억제시키는 신규의 펩타이드 단편 및 이를 유효성분으로 포함하는 약학 조성물을 제공한다. 본 발명의 펩타이드는 PKA와 SHP2의 상호작용을 현저하게 증가시킴으로써, 암세포의 전이 억제 및 염증 질환의 예방 또는 치료에 유용하게 적용될 수 있다.
Description
본 발명은 β1 인테그린 신호전달 억제 활성을 갖는 펩타이드 및 이를 유효성분으로 포함하는 암세포의 전이 억제용 약학 조성물 또는 염증 질환의 예방 또는 치료용 약학 조성물에 관한 것이다.
인테그린(integrins)은 암 성장의 다양한 단계에 관여하는 주요 세포 부착 분자(cell adhesion molecules) 중 하나이다(Hakanpaa L et al., Nat Commun 6:5962; Hongu T et al., Nat Commun 6:7925; Vitorino P et al., Nature 519:425-30; Yamamoto H et al., Nat Commun 6:6429; Vellon L et al, Oncogene 24:3759-73). β1 인테그린은 종양세포에서 가장 일반적으로 발현되는 인테그린이며, 암 전이에 있어서 중요한 역할을 한다(Paschos KA et al, Cell Signal 21:665-74; Desgrosellier JS et al, Nat Rev Cancer 10:9-22). 예를 들어, 전이성 암(metastatic tumors)은 비-전이성 암에 비하여 현저하게 높은 β1 인테그린 발현을 나타낸다(Wang D et al, Clin Cancer Res 18:4589-99). 또한, 인테그린은 염증 부위에 대한 백혈구의 혈관외 유출(extravasation)의 조절에 있어서 핵심적인 역할을 한다(Campbell DJ et al, J Exp Med . 2002;195:135-41; Do JS et al, Immunol Cell Biol. 2014;92(1):90-8; Doll K et al, Veterinary journal. 2009;181(2):90-6). 염증 매개인자(inflammatory mediators) 또는 신호전달에 의해 자극된 인테그린 활성화는 염증 세포로의 백혈구 이동(leukocyte migration)을 개시하는데 있어서 핵심적인 단계이다. 따라서, β1 인테그린 등의 인테그린의 활성을 억제하는 것은 암 전이 및 염증 질환에 대한 치료제 개발의 중요한 표적이 될 수 있다.
Suh JS. 등은 CD99 분자가 활성화되면 β1 인테그린의 기능을 변화시켜, 암세포가 세포외 기질(extracellular matrix, ECM)에 부착하는 것을 억제함으로써, 암전이 과정에 관여할 수 있음을 개시한 바 있다(Suh JS., 2001. Control of invasiveness of human breast carcinoma cell line MCF-7 by CD99 molecule. Kangwon National University). 또한, Lee KJ 등은 CD99가 SHP2(Src homology region 2 domain-containing phosphatase-2)에 의해 매개되는 FAK 탈인산화(SHP2-mediated FAK dephosphorylation)를 통한 β1 인테그린 신호전달을 억제함을 보고한 바 있다(Lee KJ et al, Exp Mol Med 798 36:534-44).
국제특허공개 제WO2007/037601호는 CD99로부터 유래된 특정 서열을 갖는 펩타이드 단편이 CD99를 효과적으로 활성화시킴으로써, 백혈구의 혈관외유출 또는 암세포의 성장 및/또는 전이를 억제할 수 있다는 것을 개시한 바 있다. 또한, 국제특허공개 제WO2011/049289호는 CD99, CD99L2, 및 PBDX의 세포밖부위 중 고도보존부위(highly conserved resion, HCR) I - III에서 유래한 펩타이드(예를 들어, LSD, LGD, LAD, LED 등) 또는 그의 융합단백질 및 암세포의 성장 및 전이 억제용 약학 조성물 및 염증 질환의 예방 또는 치료용 약학 조성물을 개시한 바 있다.
본 발명자들은 SHP2-매개되는 FAK 탈인산화를 통한 CD99에 의한 β1 인테그린 신호전달 억제에 대한 분자생물학적 메커니즘을 규명하기 위하여 다양한 연구를 수행하였다. 본 발명자들은 활성화된 CD99가 PKA(protein tyrosine kinase)를 활성화시킴으로써, SHP2 불러모음(SHP2 recruitment) 및 SHP2 활성화를 야기한다는 것을 발견하였다. 또한, 본 발명자들은 CD99를 활성화시킬 수 있는 다양한 펩타이드 단편을 제작하여 PKA와 SHP2의 상호작용에 미치는 영향을 평가하였으며, 그 결과 특정 아미노산 서열을 갖는 펩타이드가 PKA와 SHP2의 상호작용을 현저하게 증가시킨다는 것을 발견하였다.
따라서, 본 발명은 상기 PKA와 SHP2의 상호작용을 증가시키는 펩타이드를 제공하는 것을 목적으로 한다.
또한, 본 발명은 상기 펩타이드를 유효성분으로 포함하는 암세포 전이 억제용 약학 조성물을 제공하는 것을 목적으로 한다.
또한, 본 발명은 상기 펩타이드를 유효성분으로 포함하는 염증 질환의 예방 또는 치료용 약학 조성물을 제공하는 것을 목적으로 한다.

활성화된 CD99가 PKA(protein tyrosine kinase)를 활성화시킴으로써, SHP2를 불러모으고 또한 SHP2를 활성화시켜 β1 인테그린 신호전달을 억제한다는 것이 본 발명에 의해 밝혀졌다. 또한, 본 발명의 펩타이드가 PKA와 SHP2의 상호작용을 현저하게 증가시킴으로써, β1 인테그린 신호전달을 매개로 한 암세포 전이를 억제할 수 있으며, 또한 염증 질환을 예방 또는 치료에 유용하게 적용될 수 있다는 것이 밝혀졌다. 따라서, 본 발명의 펩타이드는 암세포 전이 억제 및 염증 질환의 예방 또는 치료에 유용하게 적용될 수 있다.
도 1은 인간 유방암 세포 MCF-7를 CD99 효능제인 Pep4와 SHP-2, MEK 또는 아데닐 사이클레이즈(Adenyl Cyclase) 특이적 저해제인 NSC87877, PD98059와 KH7을 처리하였을 때, CD99-ezrin, CD99-PKA, 및 PKA-SHP2의 상호작용에 미치는 영향을 평가하여 얻어진 결과를 나타낸다.

본 발명의 약학 조성물은 약학적으로 허용가능한 담체를 포함할 수 있으며, 예를 들어, 락토즈, 옥수수전분 등의 부형제, 마그네슘 스테아레이트 등의 윤활제, 공지되어 사용가능한 유화제, 현탁제, 완충제, 등장화제 등을 포함할 수 있다. 본 발명의 약학 조성물은 경구투여 제제 형태(oral dosage form) 또는 비경구투여 제제 형태(parenteral dosage form) 형태로 제제화될 수 있으며, 바람직하게는 비경구 투여형태로 제제화될 수 있다. 예를 들어, 근육내, 복강내, 피하 및 정맥내 투여 형태의 경우, 통상 활성 성분의 멸균 용액을 제조하고, 용액의 pH를 적합하게 조절할 수 있는 완충제를 포함할 수 있으며, 정맥내 투여의 경우 제제에 등장성이 부여되도록 등장화제를 포함할 수 있다. 또한, 본 발명의 약학 조성물은 pH가 7.4인 염수와 같은 약학적으로 허용되는 담체를 포함하는 수용액제의 형태가 될 수 있으며, 용액제의 형태로 국소적으로 환자의 근육내 혈류에 도입할 수 있다.
본 발명의 약학 조성물은 암세포의 전이를 효과적으로 억제함으로써, 유방암, 위암, 대장암, 결장암, 직장암, 췌장암을 비롯한 다양한 고형암 또는 골수성 백혈병 등의 암환자에게 투여될 수 있으며, 그 투여용량은 통상 각 환자의 연령, 체중 및 환자의 증상에 따라 일반적으로 변화시킬 수 있는 용량, 예를 들어 1일 약 0.01 내지 10 mg/kg의 용량으로 투여될 수 있다. 또한, 본 발명의 약학 조성물은 다양한 염증 질환을 앓고 있는 환자에게 투여될 수 있으며, 상기 투여용량은 통상 각 환자의 연령, 체중 및 환자의 증상에 따라 일반적으로 변화시킬 수 있는 용량, 예를 들어 1일 약 0.01 내지 10 mg/kg의 용량으로 투여될 수 있다.
이하, 본 발명을 실시예 및 시험예를 통하여 더욱 상세히 설명한다. 그러나, 이들 실시예 및 시험예는 본 발명을 예시하기 위한 것으로, 본 발명이 이들 실시예 및 시험예에 한정되는 것은 아니다.
실시예
1.
펩타이드의
합성
서열번호 1 내지 20의 펩타이드(하기 표 1 참조)는 자동화합성기(PeptrEx-R48, 펩트론사, 대전, 대한민국)를 이용하여 FMOC 고체-상 방법(FMOC solid-phase method)으로 합성하였다. 합성된 펩타이드는 C18 분석용 RP 컬럼(Shiseido capcell pak)을 사용한 역상 고속액체크로마토그래피(reverse-phase HPLC) (Prominence LC-20AB, Shimadzu사, 일본)로 정제 및 분석하였으며, 질량분석기(HP 1100 Series LC/MSD, Hewlett-Packard사, Roseville, 미국)를 이용하여 동정하였다.

실시예 2. 펩타이드를 포함하는 조성물의 제조
서열번호 1 내지 20의 펩타이드를 PBS에 각각 용해시켜, 1 M의 농도가 되도록 제조하였다. 얻어진 단백질 용액을 하기 시험예에서 사용하였다.

1. CD99의 활성화에 의한
SHP2의
불러모음 및 활성화 메커니즘
내피세포 CD99(endothelial CD99)는 에즈린(ezrin)과 수용성 아데닐릴 사이클레이즈(soluble adenylyl cyclase, sAC)와 신호 복합체를 형성하여 PKA 활성화를 촉진한다는 것이 보고된 바 있다(Watson RL et al, J Exp Med 212:1021-41). CD99의 활성화가 sAC/PKA 신호 전달을 활성화시키는지 그리고 PKA가 SHP2에 어떻게 작용하는지를 평가하였다.

물리적 상호작용 평가
CD99 효능제(Pep4)를 처리하여 CD99를 활성화시켰을 때 CD99-ezrin, CD99-PKA 및 SHP2-PKA의 물리적 상호작용이 어떻게 달라지는지 관찰하였다. 인간 유방암 세포 MCF-7(ATCC, HTB-22)를 특이적 저해제(NSC87877; SHP2 저해제, PD98059; MEK저해제, KH7; sAC저해제) 또는 CD99 효능제(Pep4)로 처리한 후 in situ PLA (proximity ligation assay)를 이용하여 CD99-ezrin , CD99-PKA 및 SHP2-PKA의 물리적 결합을 관찰하였다. 24-웰 마이크로플레이트의 각 웰에 4.5 X 104 개의 MCF-7 세포(ATCC, HTB-22)를 DMEM 혈청배지에 분주하고 37℃, 5% CO2 인큐베이터에서 24시간 배양한 후, 무혈청배지 중 CD99 효능제의 농도가 20 μM이 되도록 실시예 2의 펩타이드 용액과 특이적 저해제을 가한 다음, 30분간 동일한 조건에서 인큐베이션하였다. 대조군은 아무 처리를 하지 않았다. 각 웰의 세포를 PBS로 세정하고 2% 포름알데히드로 15분 동안 처리하여 고정시킨 후, 0.1% TritonX-100을 5분간 처리하여 세포 안으로의 항체 투과성을 높였다. 항-CD99 단일클론항체(Santa Cruz, CA, USA)와 항-Ezrin 다클론항체(Santa Cruz, CA, USA) 및 항-CD99 단일클론항체(Santa Cruz, CA, USA) 와 anti-PKA α cat 다클론항체(Santa Cruz, CA, USA) 그리고 anti-SHP2 다클론항체(Santa Cruz, CA, USA)와 anti-PKA α cat 다클론항체(Santa Cruz, CA, USA)를 각각 첨가하고, In situ PLA 키트(Sigma-Aldrich)를 사용하여 제조사의 지침에 따라 PLA 프로브를 가한 다음, 하이브리디제이션(hybridization), 라이게이션(ligation), 증폭(amplification) 및 마운팅 단계를 수행하였다. 각각의 세포에서 검출되는 발광 신호(PLA signals)를 공초점 레이져 현미경(Olympus fluoview FW1000; Olympus, Tokyo, Japan)으로 측정하여 CD99-ezrin 및 CD99-PKA 그리고 SHP2-PKA항체의 물리적인 결합을 정량하였다. 그 결과는 도 1과 같다.
도 1에서 알 수 있는 바와 같이, CD99 효능제를 사용한 CD99의 활성화는 CD99-ezrin 및 CD99-PKA 상호작용을 유의성 있게 증가시켰다. 놀랍게도, CD99 효능제 처리는 PKA 및 SHP2 사이의 강한 상호작용을 유도하였다. CD99와 ezrin 및 PKA의 상호작용은 SHP2의 저해제인 NSC87877의 처리에 의해 영향을 받지 않았다. 또한, PD98059에 의한 MEK의 표적화된 저해(targeted inhibition)도 CD99-ezrin 혹은 SHP2-PKA 상호작용에 어떠한 영향도 미치지 않았다. 대조적으로, sAC 저해제인 KH7의 처리는 PKA와 CD99 및 SHP2의 상호작용을 특이적으로 억제하였다.
또한, siRNA를 이용하여 Ezrin 또는 PKA의 발현을 억제시킨 후 CD99와 PKA의 상호작용을 in situ PLA를 통하여 관찰하였다. MCF-7(ATCC, HTB-22)를 DMEM 혈청배지에서 50∼60% 콘플루언시까지 배양한 후, Ezrin 또는 PKA siRNA(Santa Cruz, CA, USA)(무처리, 1 nM 처리, 또는 10 nM 처리) 및 리포펙타민 RNAiMAX (Lipofectamine RNAiMAX)(Invitrogen)를 사용하여 제조사 지침에 따라 일시적으로 형질감염(transfection)시켜 각각의 단백질 발현을 억제시킨 다음, CD99 효능제(Pep4)를 20 μM의 농도로 30분 동안 처리하였다. 도 1의 결과로부터 알 수 있는 바와 같이, Ezrin 또는 PKA siRNA를 일시적으로 형질감염시킨 시험군에서 CD99 및 PKA의 CD99 효능제-유도된 상호작용을 완전히 차단하였다.
(2) 면역침전(
Immunoprecipitation
)을 사용한
SHP2와
PKA의
상호결합 분석
면역침전(Immunoprecipitation)기법을 사용하여 SHP2와 PKA의 상호결합을 확인하였다. MCF-7 세포를 웰당 5 x 106이 되도록 준비한 다음, 또는 PKA siRNA(Santa cruz, CA, USA) 및 리포펙타민 RNAiMAX (Lipofectamine RNAiMAX)(Invitrogen)를 사용하여 제조사 지침에 따라 일시적으로 형질감염(transfection)시켜 세포를 준비한 다음, 20 μM 농도의 CD99 효능제(Pep4)와 특이적 저해제(NSC87877; SHP2 저해제, KH7; sAC저해제)를 세포에 처리한 후 30분 동안 인큐베이션하였다. 세포들을 PBS로 3회 세척한 후, 0.1 μM PMSP(phenylmethylsulfonyl fluoride), 1 ㎍/㎖ 펩스타틴 A(pepstatin A), 10 ㎍/㎖ 류펩틴(leupeptin), 1 ㎍/㎖ 아프로티닌(aprotinin)과 1 mM Na3VO4가 포함된 1% NP40 용해 완충액(lysis buffer) (1% Nonidet P40, 0.1M NaCl, 0.05M tris (pH 8.0), 5 mM EDTA)으로 용해하였다. 세포용해물에 anti-SHP2 다클론항체(Santa cruz, CA, USA) 1㎍을 첨가하여 4℃ 회전교반기에 16시간 이상 인큐베이션하여 세포 용해물 내에 존재하는 SHP2단백질과 항체를 결합시킨 후, Protein A/G bead(Santa cruz, CA, USA)를 40 μl첨가하고 4℃ 회전교반기에 3시간 인큐베이션하여 항체와 결합시켰다. TBS 완충액으로 면역침전물(SHP2결합단백질-항체-Protein A/G bead)외의 다른 용해물을 세척한 후 원심분리 하여 면역침전물을 추출한 후 전기영동을 위한 샘플을 준비하였다. 면역침전물을 10% 폴리아크릴아미드 겔 상에서 전기영동을 실시하였으며, 전개된 단백질을 니트로셀룰로오스 막으로 이송한 후, 블록킹 용액(blocking solution) (0.05% Tween 20과 3% 소혈청알부민이 함유된 Tris-완충 식염수(Tris-buffered saline, TBS)으로 상온에서 1시간 가량 처리하였다. 이 후 항-PKA α cat 다클론항체(Santa cruz, CA, USA)또는 항-SHP2 다클론항체(Santa cruz, CA, USA)가 첨가된 TBS 완충액에서 2시간 반응시킨 후, 0.05% Tween 20이 포함된 TBS 완충액으로 세척하였다. horseradish peroxidase conjugated anti-rabbit IgG (Santa cruz, CA, USA)로 상온에서 1시간 처리한 후, 0.05% Tween 20을 함유하는 TBS로 5회 세척하고 antibody detection kit (Ab frontier, Korea)를 사용하여 현상하였다. 동일량의 세포용해물을 대상으로 실험한 것을 확인하기 위하여 면역침전을 하기 전 세포용해물을 전기영동하여 항-SHP2 다클론항체(Santa cruz, CA, USA)와 항-PKA α cat 다클론항체(Santa cruz, CA, USA) 및 항-beta actin 단일클론항체(Sigma-Aldrich Ltd.; cat No. A54441)를 사용하여 액틴을 인식하였다. 또한 SHP2의 인산화 정도를 확인하기 위해 항-SHP2-p 다클론항체(Santa cruz, CA, USA)를 사용하여 인식하였다. 그 결과는 도 2와 같다.
도 2의 결과로부터 알 수 있는 바와 같이, sAC 또는 SHP2의 저해제는 CD99 효능제에 의해 자극된 PKA에 대한 SHP2의 결합을 저해하였다. 이들 2개의 분자들 사이의 물리적인 결합은 PKA의 넉다운(knockdown)에 의해 유의성 있게 감소하였다. 또한, SHP2의 CD99 효능제-유도된 인산화는 sAC의 저해 또는 PKA의 넉다운에 의해 현저하게 억제되었다.
상기 도 1 및 도 2의 결과는 CD99 효능제에 의해 활성화된 CD99가 ezrin-sAC-PKA 신호 전달 복합체의 형성을 자극함으로써, SHP2의 불러모음(recruitment) 및 활성화(activation)로 이어진다는 것을 나타낸다.

펩타이드의
활성 평가
본 발명의 펩타이드를 세포에 처리한 후, in situ PLA (proximity ligation assay)방법을 통하여 PKA와 SHP2의 물리적 결합에 대한 영향을 평가하였다. 시험예 1과 동일한 방법으로 세포를 전처리한 후, 항-PKA α cat 다클론항체(Santa cruz, CA, USA) 및 항-SHP2 다클론항체(Santa cruz, CA, USA)를 첨가하고, In situ PLA 키트(Sigma-Aldrich)를 사용하여 제조사의 지침에 따라 PLA 프로브를 가한 다음, 하이브리디제이션(hybridization), 라이게이션(ligation), 증폭(amplification) 및 마운팅 단계를 수행하였다. 각각의 세포에서 검출되는 발광 신호(PLA signals)를 공초점 레이져 현미경(Olympus fluoview FW1000; Olympus, Tokyo, Japan)으로 측정하여 PKA 및 SHP2 항체의 물리적인 상호작용을 정량하였다. 그 결과는 도 3a 내지 도 3d와 같다. 도 3a 내지 도 3d의 결과로부터, 본 발명의 펩타이드 처리군은 미처리 대조군에 비하여 PKA와 SHP2의 상호작용을 현저하게 증가시켰음을 확인할 수 있다.
