Description
本発明は、タンパク質製剤を収納するための医薬品容器に関する。
タンパク質製剤は、ガン、糖尿病、C型肝炎、慢性腎不全などのような多くの慢性病だけではなく、血友病、ファブリー病、発育不全、多発性硬化症、クローン病などの稀な病気に対しても高い治療効果が期待されている。しかし、タンパク質製剤の有効成分は、タンパク質などに由来するため、物理的・化学的刺激に敏感で不安定である。
タンパク質製剤を収納する容器としては、重くて破損しやすいガラス製の容器に代えて、軽くて破損しにくく、かつ取扱い性に優れるプラスチック製の容器が広く用いられている。
医療用容器を製造するためのプラスチック素材としては、ポリプロピレン、ポリエチレン、環状オレフィン系重合体、ポリ塩化ビニル、ポリエステル、ポリアミド、ポリカーボネート、ポリメタクリレートなどが挙げられる。これらのプラスチックのうち、環状オレフィン系重合体は、医薬品容器用の素材として注目され、その使用が色々試みられている。
特開2014-51502号公報(米国特許第7253142号明細書)には、環状オレフィンとオレフィンの共重合体であるシクロオレフィンコポリマー、シクロオレフィン類開環重合体、またはシクロオレフィン類開環重合体に水素添加したものを用いた医薬品容器が開示されている。
しかしながら、特開2014-51502号公報(米国特許第7253142号明細書)に記載の環状オレフィン系重合体を用いる医薬品容器では、酸素や水蒸気のガス透過性が比較的高く、収納されるタンパク質製剤が液剤の場合には容器の外側から脱酸素剤などによって酸素を吸収するようにしても十分に吸収することができず、収納されるタンパク質製剤が凍結乾燥剤等の固形剤である場合には容器の外部から吸水剤などによって水分を吸収するようにしても十分に吸収することができないため、容器内の製剤が、溶存酸素や残存水分によって不安定化する恐れがある。
したがって、本発明は、上記事情を鑑みてなされたものであり、収納するタンパク質製剤の安定化向上に寄与する医薬品容器を提供することを目的とする。
本発明者らは、上記課題を解決すべく鋭意研究を積み重ねた。その結果、水素化ブロックコポリマーを含む、タンパク質製剤を収納するための医薬品容器によって上記課題を解決することを見出し、本発明の完成に至った。
以下、本発明の一形態に係る実施の形態を説明する。本発明は、以下の実施の形態のみには限定されない。
本明細書において、範囲を示す「X~Y」は「X以上Y以下」を意味する。また、特記しない限り、操作および物性等の測定は室温(20~25℃)/相対湿度40~50%RHの条件で測定する。
<医薬品容器>
本発明の一形態は、水素化ブロックコポリマーを含む、タンパク質製剤を収納するための医薬品容器である。かかる構成により、収納するタンパク質製剤の安定化向上に寄与する医薬品容器を提供することができる。本明細書中、「タンパク質製剤を収納するための医薬品容器」を単に「医薬品容器」とも称する。本形態の医薬品容器は、環状オレフィン系重合体製容器と比べて、酸素や水蒸気のガス透過性が高いため、脱酸素剤や脱水剤または脱酸素機能や脱水機能を有する包装材と組み合わせて用いることで、医薬品容器に収納されるタンパク質製剤中の溶存酸素または残存水分を大幅に除去することができる。したがって、タンパク質製剤をより安定して保管することができる。
本発明の一形態は、水素化ブロックコポリマーを含む、タンパク質製剤を収納するための医薬品容器である。かかる構成により、収納するタンパク質製剤の安定化向上に寄与する医薬品容器を提供することができる。本明細書中、「タンパク質製剤を収納するための医薬品容器」を単に「医薬品容器」とも称する。本形態の医薬品容器は、環状オレフィン系重合体製容器と比べて、酸素や水蒸気のガス透過性が高いため、脱酸素剤や脱水剤または脱酸素機能や脱水機能を有する包装材と組み合わせて用いることで、医薬品容器に収納されるタンパク質製剤中の溶存酸素または残存水分を大幅に除去することができる。したがって、タンパク質製剤をより安定して保管することができる。

水素化ブロックコポリマーの数平均分子量は、本発明の効果をより発揮するとの観点から、好ましくは20,000~300,000である。数平均分子量は、ゲルパーミエーションクロマトグラフィー(GPC)により測定することができる。
水素化ブロックコポリマーのメルトマスフローレイト(MFR)は、本発明の効果をより発揮するとの観点から、好ましくは0.5~300g/10minである。MFRは、JIS K 7210-1:2014、ASTM D1238、あるいはISO 1133-1:2011に準じて測定することができる。
特に、水素化ブロックコポリマーにおいて、その数平均分子量が20,000以上である、および/またはそのMFRが300g/10minであると、水素化ブロックコポリマーが水素化共役ジエンポリマーブロック(後述)を有する場合、側鎖を有さない共役ジエン由来の構成単位の割合が多くなり、側鎖を有する共役ジエン由来の構成単位を多く含むポリマーに比べて、立体的障害がなく、ポリマー主鎖の分子運動性が高くなるため、医薬品容器のガス透過性を高くすることができる。また、側鎖を含まないことにより、光などによる側鎖の開裂がなく耐候性が高くなるため、医薬品容器の透明性も高く維持することができる。
また、水素化ブロックコポリマーにおいて、その数平均分子量が300,000以下である、またはそのMFRが0.5g/10minであると、射出成形などを用いる医薬品容器の成形性の点で好ましい。
水素化ブロックコポリマーを構成するポリマーブロックは、例えば上記密度や数平均分子量を満たすものであれば、特に制限されないが、本発明の効果をより発揮するとの観点から、好ましくは水素化ビニル芳香族ポリマーブロックと水素化共役ジエンポリマーブロックとを有する。水素化ブロックコポリマーが水素化ビニル芳香族ポリマーブロック(ハードセグメント)と水素化共役ジエンポリマーブロック(ソフトセグメント)とを有することにより、特開2014-51502号公報(米国特許第7253142号明細書)に記載の環状オレフィン系重合体を用いる医薬品容器に比べて、低密度化を実現し、酸素や水蒸気のガス透過性がより高い医薬品容器を得ることができる。そのため、このような医薬品容器を脱酸素剤や脱水剤または脱酸素機能や脱水機能を有する包装材と組み合わせて用いることで、医薬品容器に収納されるタンパク質製剤中の溶存酸素または残存水分をより大幅に除去することができる。したがって、タンパク質製剤をより安定して保管することができる。
さらに、医薬品容器が水素化ブロックコポリマーを含むことにより、優れた耐候性および耐滅菌性を発揮することができる。
また、ポリマーが不飽和炭化水素(2重結合または3重結合を含む炭化水素化合物)からなる場合、光による2重結合または3重結合の開裂により変色が発生し、また2重結合または3重結合によりポリマー主鎖の分子運動性の低下し、ガス透過性が低くなる。そのため、本形態の医薬品容器では、ソフトセグメントとハードセグメントとからなるブロックコポリマー(耐衝撃性に優れる)を水添して得られた水素化ブロックコポリマーを用いることで、医薬品容器の透明性およびガス透過性を向上することができる。
以下、好ましい実施形態である水素化ビニル芳香族ポリマーブロックと水素化共役ジエンポリマーブロックとを有する水素化ブロックコポリマーについて、説明する。
(水素化ビニル芳香族ポリマーブロック)
水素化ビニル芳香族ポリマーブロックは、ビニル芳香族化合物由来の構成単位を含む。水素化ビニル芳香族ポリマーブロックは、ビニル芳香族化合物由来の構成単位を50質量%以上、好ましくは80質量%以上、より好ましくは90質量%以上、特に好ましくは100質量%含む。
水素化ビニル芳香族ポリマーブロックは、ビニル芳香族化合物由来の構成単位を含む。水素化ビニル芳香族ポリマーブロックは、ビニル芳香族化合物由来の構成単位を50質量%以上、好ましくは80質量%以上、より好ましくは90質量%以上、特に好ましくは100質量%含む。
ビニル芳香族化合物としては、例えばスチレン、α-メチルスチレン、2-メチルスチレン、3-メチルスチレン、4-メチルスチレン、4-プロピルスチレン、4-シクロヘキシルスチレン、4-ドデシルスチレン、2-エチル-4-ベンジルスチレン、4-(フェニルブチル)スチレン、ビニルトルエン、1-ビニルナフタレン、2-ビニルナフタレンなどが挙げられる。ビニル芳香族化合物は、好ましくはスチレンおよびα-メチルスチレンから選択され、より好ましくはスチレンである。
水素化ビニル芳香族ポリマーブロックは、上記ビニル芳香族化合物の1種のみから構成されていてもよく、2種以上から構成されていてもよい。
水素化ビニル芳香族ポリマーブロックは、ビニル芳香族化合物由来の構成単位以外のその他の構成単位を含むことができる。その他の構成単位としては、例えばイソプレン、ブタジエン、2,3-ジメチル-ブタジエン、1,3-ペンタジエン、1,3-ヘキサジエンなどに由来する構成単位が挙げられる。
(水素化共役ジエンポリマーブロック)
水素化共役ジエンポリマーブロックは、共役ジエン由来の構成単位を含む。水素化共役ジエンポリマーブロックは、共役ジエン由来の構成単位を50質量%以上、好ましくは80質量%以上、より好ましくは90質量%以上、特に好ましくは100質量%含む。

共役ジエンとしては、例えばブタジエン、2-メチル-1,3-ブタジエン(イソプレン)、2,3-ジメチル-ブタジエン、1,3-ペンタジエン、1,3-ヘキサジエンなどが挙げられる。共役ジエンは、好ましくはブタジエンおよびイソプレンから選択され、より好ましくはブタジエンである。
一実施形態では、水素化共役ジエンポリマーブロックは、側鎖を有さない共役ジエン由来の構成単位を含むことが好ましく、側鎖を有さない共役ジエン由来の構成単位からなることがより好ましい。ポリマーの分子設計において、ガス透過性を付与するためには、ポリマー主鎖の分子運動性を高くする必要がある。側鎖を有する共役ジエンを使用すると、側鎖を有さない共役ジエンに比べて、立体的障害がなく、ポリマー主鎖の分子運動性が高くなるため、医薬品容器のガス透過性を高くすることができる。また、側鎖を含まないことにより、光などによる側鎖の開裂がなく耐候性が高くなるため、医薬品容器の透明性も高く維持することができる。側鎖を有さない共役ジエンとしては、例えばブタジエン、1,3-ペンタジエン、1,3-ヘキサジエンなどが挙げられる。側鎖を有さない共役ジエンとしては、好ましくはブタジエンである。
水素化共役ジエンポリマーブロックにおいて、共役ジエンの結合形態、すなわちミクロ構造は特に制限されない。例えば、ブタジエンの場合、1,2-結合および1,4-結合の結合形態をとることができる。また、イソプレンの場合、1,2-結合、3,4-結合および1,4-結合の結合形態をとることができる。これらの結合形態は、1種のみが存在していてもよく、2種以上が存在していてもよい。2種以上の結合形態が存在する場合、結合形態の存在の割合は、特に制限されない。
水素化共役ジエンポリマーブロックは、共役ジエン由来の構成単位以外のその他の構成単位を含むことができる。その他の構成単位としては、スチレン、α-メチルスチレン、2-メチルスチレン、3-メチルスチレン、4-メチルスチレン、4-プロピルスチレン、4-シクロヘキシルスチレン、4-ドデシルスチレン、2-エチル-4-ベンジルスチレン、4-(フェニルブチル)スチレン、ビニルトルエン、1-ビニルナフタレン、2-ビニルナフタレンなどに由来する構成単位が挙げられる。
水素化共役ジエンポリマーブロックの含有量は、水素化ビニル芳香族ポリマーブロックおよび水素化共役ジエンポリマーブロックの合計100質量%に対して、好ましくは10~90質量%である。水素化ブロックコポリマーが上記範囲の水素化共役ジエンポリマーブロックを含むことにより、酸素や水蒸気のガス透過性をより高くすることができる。
(水素添加率)
本実施形態に係る水素化ビニル芳香族ポリマーブロックにおいて、水素化ビニル芳香族ポリマーブロックの芳香環の水素添加率は、好ましくは50モル%以上であり、より好ましくは80モル%以上であり、さらに好ましくは90モル%以上である。また、水素化共役ジエンポリマーブロックの共役ジエン由来の炭素-炭素二重結合の水素添加率は、好ましくは95モル%以上であり、より好ましくは100モル%である。
本実施形態に係る水素化ビニル芳香族ポリマーブロックにおいて、水素化ビニル芳香族ポリマーブロックの芳香環の水素添加率は、好ましくは50モル%以上であり、より好ましくは80モル%以上であり、さらに好ましくは90モル%以上である。また、水素化共役ジエンポリマーブロックの共役ジエン由来の炭素-炭素二重結合の水素添加率は、好ましくは95モル%以上であり、より好ましくは100モル%である。
芳香環の水素添加率は、例えば1H-NMRにより、0.5~2.5ppm付近の脂肪族由来のピークと、6.0~8.0ppm付近の芳香環由来のピークの積分値から算出することができる。また、炭素-炭素二重結合の水素添加率は、水素化共役ジエンポリマーブロック中の共役ジエン由来の炭素-炭素二重結合の含有量を、水素添加の前後において1H-NMRスペクトルによって測定し、その測定値から求めることができる。
(水素化ブロックコポリマーの結合様式)
水素化ブロックコポリマーにおけるポリマーブロックの結合様式は、線状、分岐状および放射状のいずれであってもよく、これらの組み合わせであってもよい。

例えば、水素化ビニル芳香族ポリマーブロックを「X」、水素化共役ジエンポリマーブロックを「Y」で表すと、結合様式としては、ジブロックポリマー(X-Y)、トリブロックコポリマー(X-Y-X)、テトラブロックコポリマー(X-Y-X-Y)、ペンタブロックコポリマー(X-Y-X-Y-XまたはY-X-Y-X-Y)、(X-Y)nZ型コポリマー(Zはカップリング剤残基であり、nは2以上の整数である)などが挙げられる。結合様式は、製造容易性の観点から、好ましくはトリブロックコポリマーまたはテトラブロックコポリマーであり、より好ましくはトリブロックコポリマーである。
(水素化ブロックコポリマーの製造方法)
水素化ブロックコポリマーの製造方法は、特に制限されず、アニオン重合法などの公知の方法を用いることができる。

具体的には、本発明に係る水素化ブロックコポリマーは、アルキルリチウム化合物を開始剤として用いて、ビニル芳香族化合物と、共役ジエンとを逐次重合させる方法;アルキルリチウム化合物を開始剤として用いて、ビニル芳香族化合物と、共役ジエンとを逐次重合させ、次いでカップリング剤を加えてカップリングする方法;ジリチウム化合物を開始剤として用い、共役ジエン、次いでビニル芳香族化合物を逐次重合させる方法などにより重合反応を行った後、水素添加反応を行うことによって製造することができる。
アルキルリチウム化合物としては、例えばメチルリチウム、エチルリチウム、n-ブチルリチウム、sec-ブチルリチウム、tert-ブチルリチウム、ペンチルリチウムなどが挙げられる。
カップリング剤としては、ジビニルベンゼン;エポキシ化1,2-ポリブタジエン、エポキシ化大豆油、1,3-ビス(N,N-グリシジルアミノメチル)シクロヘキサンなどの多価エポキシ化合物;ジメチルジクロロシラン、ジメチルジブロモシラン、トリクロロシラン、メチルトリクロロシラン、テトラクロロシラン、テトラクロロスズなどのハロゲン化合物;安息香酸メチル、安息香酸エチル、安息香酸フェニル、シュウ酸ジエチル、マロン酸ジエチル、アジピン酸ジエチル、アジピン酸ジオクチル、フタル酸ジメチル、フタル酸ジエチル、イソフタル酸ジメチル、テレフタル酸ジメチルなどのエステル化合物;炭酸ジメチル、炭酸ジエチル、炭酸ジフェニル等の炭酸エステル化合物;ジメチルジメトキシシラン、メチルトリメトキシシラン、メチルトリエトキシシラン、テトラメトキシシラン、テトラエトキシシラン、ビス(トリメトキシシリル)ヘキサン、ビス(トリエトキシシリル)エタンなどのアルコキシシラン化合物などが挙げられる。
ジリチウム化合物としては、ナフタレンジリチウム、ジリチオヘキシルベンゼンなどが挙げられる。
重合反応は、溶媒の存在下で行うのが好ましい。溶媒としては、開始剤に対して不活性で、反応に悪影響を及ぼさなければ特に制限はなく、例えばヘキサン、シクロヘキサン、ヘプタン、オクタン、デカンなどの飽和脂肪族炭化水素;トルエン、ベンゼン、キシレンなどの芳香族炭化水素が挙げられる。また、重合反応の温度は、通常、ミクロ構造制御の観点から、好ましくは0~100℃であり、より好ましくは30~90℃であり、さらに好ましくは40~80℃であり、特に好ましくは50~80℃である。重合反応の時間は、ミクロ構造制御の観点から、好ましくは0.5~50時間である。
また、重合反応の際に共触媒としてルイス塩基を用いてもよい。ルイス塩基としては、例えばジメチルエーテル、ジエチルエーテル、テトラヒドロフラン等のエーテル類;エチレングリコールジメチルエーテル、ジエチレングリコールジメチルエーテル等のグリコールエーテル類;トリエチルアミン、N,N,N’,N’-テトラメチルエチレンジアミン、N-メチルモルホリン等のアミン類などが挙げられる。これらのルイス塩基は、1種単独で用いてもよいし、2種以上を併用してもよい。
水素添加反応は、重合反応に引き続き行ってもよく、重合反応後にブロックコポリマーを一旦単離してから行ってもよい。
重合反応後にブロックコポリマーを一旦単離する場合、重合反応後に得られた重合反応液をメタノールなどのブロックコポリマーの貧溶媒に注いでブロックコポリマーを凝固させる、または重合反応液をスチームと共に熱水中に注いで溶媒を共沸によって除去(スチームストリッピング)した後、乾燥させることにより、ブロックコポリマーを単離することができる。
ブロック共重合体の水素添加反応は、例えば水素添加触媒の存在下、反応温度20~200℃および水素圧力0.1~20MPaの条件下で、0.1~100時間反応させることにより実施できる。
水素添加触媒としては、ラネーニッケル;白金(Pt)、パラジウム(Pd)、ルテニウム(Ru)、ロジウム(Rh)、ニッケル(Ni)等の金属をカーボン、アルミナ、珪藻土等の担体に担持させた不均一触媒;遷移金属化合物(オクチル酸ニッケル、ナフテン酸ニッケル、ニッケルアセチルアセトナート、オクチル酸コバルト、ナフテン酸コバルト、コバルトアセチルアセトナート等)とトリエチルアルミニウム、トリイソブチルアルミニウム等の有機アルミニウム化合物または有機リチウム化合物等との組み合わせからなるチーグラー系触媒;チタン、ジルコニウム、ハフニウム等の遷移金属のビス(シクロペンタジエニル)化合物とリチウム、ナトリウム、カリウム、アルミニウム、亜鉛またはマグネシウム等からなる有機金属化合物の組み合わせからなるメタロセン系触媒などが挙げられる。
水素添加反応を重合反応に引き続き行う場合、水素化ブロックコポリマーの単離は、水素添加反応液を、メタノールなどの水素化ブロックコポリマーの貧溶媒に注いで凝固させる、または水素添加反応液をスチームと共に熱水中に注いで溶媒を共沸によって除去(スチームストリッピング)した後、乾燥することにより単離することができる。
本形態に係る水素化ブロックコポリマーは、上記の方法で製造してもよく、市販品を用いてもよい。市販品としては、例えばViviOnTM(USI Corporation製)などが挙げられる。
[タンパク質製剤]
本発明の一実施形態では、上記医薬品容器がタンパク質製剤を収納している。

本明細書において、「タンパク質製剤」は、生物由来のタンパク質やペプチドからなる有効成分を含有する。有効成分としては、抗体、血液凝固線溶系因子、ホルモン、酵素、サイトカイン類、インターフェロン類、血清タンパク質、ワクチン、エリスロポエチン類、融合タンパク質などが挙げられる。本発明に係るタンパク質製剤容器に収納されるタンパク質製剤は、特に制限されず、公知のタンパク質製剤を用いることができる。タンパク質製剤は、液体であっても、固体であってもよい。
抗体としては、例えばアダリムマブ、ムロモナブ-CD3、トラスツズマブ、リツキシマブ、パリビズマブ、インフリキシマブ、バシリキシマブ、トシリズマブ、ゲムツズマブオゾガマイシン、ベバシズマブ、イブリツモマブチウキセタンセツキシマブ、ラニビズマブ、オマリズマブ、エクリズマブ、パニツムマブ、ウステキヌマブ、ゴリムマブ、カナキヌマブ、デノスマブなどが挙げられる。
血液凝固線溶系因子としては、例えばオクトコグアルファ、ルリオクトコグアルファ、ノナコグアルファ、トロンボモデュリンアルファなどが挙げられる。
ホルモンとしては、例えばインスリングラルギン、インスリンヒト、インスリンリスプロ、インスリンアスパルト、インスリンデテミル、インスリングルリジン、ソマトロピン、ペグビソマント、ホリトロピンアルファ、フォリトロピンベータ、リラグルチド、テリパラチドなどが挙げられる。
酵素としては、例えばアルテプラーゼ、イミグルセラーゼ、アガルシダーゼアルファ、ラロニダーゼ、アルグルコシダーゼアルファ、イデュルスルファーゼ、ガルスルファーゼ、ラスブリカーゼなどが挙げられる。
サイトカイン類としては、例えばフィルグラスチム、セルモロイキン、トラフェルミンなどが挙げられる。
インターフェロン類としては、例えばインターフェロンアルファ、インターフェロンベータなどが挙げられる。
血清タンパク質としては、例えば人血清アルブミンが挙げられる。

タンパク質製剤の配合内容およびpHなどの物性は、特に制限されず、使用するタンパク質製剤の種類に応じて、適宜調整することができる。
タンパク質製剤は、上記成分に加えて、必要に応じて安定化剤、緩衝剤、溶解補助剤、等張化剤、pH調整剤、無痛化剤、還元剤、酸化防止剤などの添加剤を含んでもよい。

医薬品容器に収納されるタンパク質製剤の量は、タンパク質製剤の種類、用途、使用形態などに応じて適宜調整することができる。
医薬品容器の形態、サイズなどは、特に制限されず、収納されるタンパク質製剤の種類、量、用途、使用形態などに応じて適宜選択することができる。
医薬品容器の形態としては、例えばバッグ、バイアル、アンプル、シリンジ、カートリッジ、ボトルなどが挙げられ、好ましくはシリンジまたはカートリッジである。
医薬品容器は、本形態に係る水素化ブロックコポリマー単独で形成された構造を有してもよく、また本発明の効果を損なわない限り、本形態に係る水素化ブロックコポリマーで形成された層を有する積層構造、本形態に係る水素化ブロックコポリマーと他の重合体との混合物から形成された構造を有してもよい。
また、医薬品容器の形態に応じて、容器本体以外の部分、例えば栓体、ガスケット、プランジャーなどは、本形態に係る水素化ブロックコポリマー以外の材料で形成されていてもよい。
医薬品容器は、射出成形、押出成形、ブロー成形、回転成形、吹き込み成形、トランスファー成形、プレス成形、溶液キャスト法などの公知の方法を用いて製造することができる。
医薬品容器の滅菌方法は、特に制限されず、放射線滅菌、エチレンオキサイド滅菌、二酸化窒素滅菌、高圧蒸気滅菌などの従来公知の方法を用いることができる。
医薬品容器がタンパク質製剤を収納している場合、医薬品容器へのタンパク質製剤の収納は、常法に従って行うことができる。
<包装体>
本発明の一形態は、上述の医薬品容器を包装材で包装した包装体である。本形態の包装体は、上記医薬品容器と共に、脱酸素剤および脱水剤の少なくとも一方とが包装材で包装されている;または前記包装材が脱酸素機能および脱水機能の少なくとも一方の機能を有する。かかる構成によって、医薬品容器に収納されるタンパク質製剤中の溶存酸素または残存水分を大幅に除去することができる。したがって、タンパク質製剤をより安定して保管することができる。
本発明の一形態は、上述の医薬品容器を包装材で包装した包装体である。本形態の包装体は、上記医薬品容器と共に、脱酸素剤および脱水剤の少なくとも一方とが包装材で包装されている;または前記包装材が脱酸素機能および脱水機能の少なくとも一方の機能を有する。かかる構成によって、医薬品容器に収納されるタンパク質製剤中の溶存酸素または残存水分を大幅に除去することができる。したがって、タンパク質製剤をより安定して保管することができる。
本形態に係る包装材は、本発明の効果をより発揮するとの観点から、バリア性包装材である。バリア性包装材は、酸素バリア性および水分バリア性の少なくとも一方を有する。バリア性包装材は、単層から形成されてもよく、2層以上の積層構造で形成されてもよい。バリア性包装材が積層構造である場合、少なくとも1つの層が酸素バリア性および水分バリア性の少なくとも一方を有する。
一実施形態では、本形態の包装体は、前記医薬品容器の外表面から包装材または包装材を構成するバリア層の間に脱酸素剤および脱水剤の少なくとも一方を備えている。バリア性包装材のバリア性は脱酸素剤を備える場合は、少なくとも酸素バリア性を有し、脱水剤を備える場合は、少なくとも水分バリア性を有する。
一実施形態では、本形態の包装体は、前記包装材が包装する内部に対して脱酸素機能および脱水機能の少なくとも一方の機能を発揮させるようにその一部に脱酸素剤および脱水剤の少なくとも一方を有する。
脱酸素剤としては、特に制限されず、従来公知の脱酸素剤を使用することができる。脱酸素剤としては、例えば水酸化鉄、酸化鉄、炭化鉄などの鉄化合物、グルコースおよびグルコースオキシダーゼの酵素作用を利用する脱酸素剤などが挙げられる。市販品としては、エージレス(登録商標)(三菱ガス化学株式会社製)、モデュラン(日本化薬フードテクノ株式会社製)、セキュール(登録商標)(ニッソーファイン株式会社製)などが挙げられる。
脱水剤としては、特に制限されず、従来公知の脱水剤を使用することができる。脱水剤としては、例えば塩化カルシウム、シリカゲル、カリウムアルミニウムシリケート、カルシウムアルミニウムシリケートなどが挙げられる。
包装材としては、特に制限されず、医薬品容器の包装材として一般に汎用されているフィルム、シートなどを用いることができる。包装材の材質としては、酸素や水蒸気に不安定な医薬品を安定に保管できるようにガスバリア性を有するものが好ましい。ガスバリア性を有する材質としては、例えばポリエチレン、ポリプロピレン、ポリエチレンテレフタレート、ポリビニルアルコール、ポリエチレンナフタレート、エチレン・ビニルアルコール共重合体、ポリ塩化ビニリデン、塩化ビニリデン・塩化ビニル共重合体、塩化ビニリデン・アクリル酸エステル共重合体、ポリアクリロニトリル、ポリアミド、ポリエステルなどが挙げられる。
包装材は、上記材質の少なくとも1つを層成分として有する積層体であってもよい。また、包装材は、アルミ箔層、アルミ蒸着層、酸化アルミ蒸着層、酸化ケイ素蒸着層などの遮光能を有する層を有してもよい。
本形態の包装体は、脱酸素機能および脱水機能の少なくとも一方の機能を有する包装材を使用することができる。このような包装材としては、例えば少なくとも一層が脱酸素剤や脱水剤を含むフィルムを使用することができる。
脱酸素機能を有する包装材の市販品としては、オキシキャッチ(登録商標)(共同印刷株式会社製)、ハイスターO2(スタープラスチック工業株式会社製)、エージレス・オーマック(登録商標)(三菱ガス化学株式会社製)、オキシデック(東洋製罐株式会社製)などが挙げられる。
脱水機能を有する包装材の市販品としては、シリカゲル(豊田化工株式会社製)などが挙げられる。
本形態の包装体の構造、形状などは、特に制限されず、包装する医薬品容器に応じて適宜選択することができる。例えば医薬品容器を脱酸素剤または脱水剤と共にブリスター方式によって包装する場合、医薬品容器を収納する凹部を有する本体部分と前記凹部を覆う被覆部分とを、各部分に求められる機能に応じて選択したフィルムまたはシートから形成し、本体部分と被覆部分とをその周縁などで密封した構造にすることができる。
本発明の効果を、以下の実施例および比較例を用いて説明する。ただし、本発明の技術的範囲が以下の実施例のみに制限されるわけではない。特記しない限り、「%」および「部」は、それぞれ、「質量%」および「質量部」を意味する。
[実施例]
(シリンジの作製)
プレフィルドシリンジ製剤用のルアーロックシリンジ本体(シリンジサイズ:ISO規格11040-6の2.25mLに準拠)を、水素化ブロックコポリマー(「ViviOnTM」、USI Corporation製、密度:0.94g/cm3、数平均分子量:82,000、MFR:7g/10min)を用いて、インサート射出成形によって製造した。
(シリンジの作製)
プレフィルドシリンジ製剤用のルアーロックシリンジ本体(シリンジサイズ:ISO規格11040-6の2.25mLに準拠)を、水素化ブロックコポリマー(「ViviOnTM」、USI Corporation製、密度:0.94g/cm3、数平均分子量:82,000、MFR:7g/10min)を用いて、インサート射出成形によって製造した。
[比較例]
(シリンジの作製)
プレフィルドシリンジ製剤用のルアーロックシリンジ本体(シリンジサイズ:ISO規格11040-6の2.25mLに準拠)を、環状ポリオレフィン重合体(「ZEONEX(登録商標)」、日本ゼオン株式会社製、密度:1.01g/cm3、数平均分子量:45,000、MFR:17g/10min)を用いて、インサート射出成形によって製造した。
(シリンジの作製)
プレフィルドシリンジ製剤用のルアーロックシリンジ本体(シリンジサイズ:ISO規格11040-6の2.25mLに準拠)を、環状ポリオレフィン重合体(「ZEONEX(登録商標)」、日本ゼオン株式会社製、密度:1.01g/cm3、数平均分子量:45,000、MFR:17g/10min)を用いて、インサート射出成形によって製造した。
[水分吸収量および水分蒸散量の測定]
(水分吸収量の測定)
上記で得られた各シリンジに、塩化カルシウムを0.9g加えて、針先をゴム栓で封止した。封止した直後のシリンジの重量(0週間)を測定した後、40℃、75%RHの環境下で保管した。一定期間(1、2、4、8、12、16、20および24週間)保管後のシリンジの重量を測定して、0週間からの重量変化量を水分変化量とした。シリンジの重量は、精密天秤にて測定した。
(水分吸収量の測定)
上記で得られた各シリンジに、塩化カルシウムを0.9g加えて、針先をゴム栓で封止した。封止した直後のシリンジの重量(0週間)を測定した後、40℃、75%RHの環境下で保管した。一定期間(1、2、4、8、12、16、20および24週間)保管後のシリンジの重量を測定して、0週間からの重量変化量を水分変化量とした。シリンジの重量は、精密天秤にて測定した。
(水分蒸散量の測定)
上記で得られた各シリンジに、水を1g加えて、針先をゴム栓で封止した。封止した直後のシリンジの重量(0週間)を測定した後、40℃、20%RHの環境下で保管した。一定期間(1、2、4、8、12、16、20および24週間)保管後のシリンジの重量を測定して、0週間からの重量変化量を水分蒸散量とした。シリンジの重量は、精密天秤にて測定した。
上記で得られた各シリンジに、水を1g加えて、針先をゴム栓で封止した。封止した直後のシリンジの重量(0週間)を測定した後、40℃、20%RHの環境下で保管した。一定期間(1、2、4、8、12、16、20および24週間)保管後のシリンジの重量を測定して、0週間からの重量変化量を水分蒸散量とした。シリンジの重量は、精密天秤にて測定した。
水分吸収量の測定結果および水分蒸散量の測定結果を図1に示す。図1中、「CBC」は、実施例のシリンジでの測定結果を示し、「COP」は、比較例のシリンジでの測定結果を示す。
図1に示すように、実施例のシリンジは、比較例のシリンジと比べて、水分を透過しやすいことが分かる。
[液体製剤中の溶存酸素濃度の測定]
アダリムマブプラセボ溶液(AbbVie社製)を添付文書に従い調製し、タンパク質製剤のモデル液体製剤を調製した。

上記で得られた各シリンジに、モデル液体製剤を1mL加えて、溶存酸素濃度(0週間)を測定した後、針先をゴム栓で封止した。
封止したシリンジと脱酸素剤(「エージレス(登録商標)」、三菱ガス化学株式会社製)とを包装したブリスターパック(トップフィルム(ポリエチレン層およびポリエチレンテレフタレート層からなる2層フィルム)、ボトムフィルム(ポリエチレンフィルム))を作製した。
一定期間(0.5、1、2、4、8および12週間)保管後、シリンジ内のモデル液体製剤の溶存酸素濃度を測定した。
溶存酸素濃度は、OXY-4(PreSens Precision Sensing GmbH製)を用いて測定した。
結果を図2に示す。図2中、「CBC」は、実施例のシリンジでの測定結果を示し、「COP」は、比較例のシリンジでの測定結果を示す。
図2に示すように、実施例のシリンジを用いることで、比較例のシリンジと比べて、液体製剤中の溶存酸素濃度をより低下させることができる、すなわち液体薬剤中の残存酸素をより除去できることが分かる。
[粉末製剤中の水分濃度の測定]
オクトコグアルファプラセボ溶液(バイエル社製)添付文書に従い調製し、オクトコグアルファプラセボ液体製剤を調製した。

上記で得られた各シリンジに、オクトコグアルファ液体製剤を1mL加えて、凍結乾燥することで、オクトコグアルファプラセボからなるタンパク質製剤のモデル粉末製剤を含むシリンジを作製した。シリンジ内のモデル粉末製剤の残存水分濃度(0週間)を測定した後、針先をゴム栓で封止した。
封止したシリンジと脱水剤(包装用乾燥剤シリカゲル)を包装したブリスターパック(トップフィルム(ポリエチレン層、エチレン・ビニルアルコール共重合体層、およびポリエチレンテレフタレート層からなる3層フィルム)、ボトムフィルム(ポリエチレン層、およびエチレン・ビニルアルコール共重合体層からなる2層フィルム))を作製した。
一定期間(0.5、1、2、4、8および12週間)保管後、シリンジ内のオクトコグアルファ粉末製剤の残存水分濃度を測定した。
残存水分濃度は、カールフィッシャー水分計を用いて測定した。
結果を図3に示す。図3中、「CBC」は、実施例のシリンジでの測定結果を示し、「COP」は、比較例のシリンジでの測定結果を示す。
図3に示すように、実施例のシリンジを用いることで、比較例のシリンジと比べて、粉末製剤中の水分濃度を低下させることができる、すなわち液体薬剤中の残存水分をより除去できることが分かる。

Claims (8)
- 水素化ブロックコポリマーを含む、タンパク質製剤を収納するための医薬品容器。
- 前記水素化ブロックコポリマーの密度が0.91~0.97g/cm3である、請求項1に記載の医薬品容器。
- 前記水素化ブロックコポリマーの数平均分子量が20,000~300,000である、請求項1または2に記載の医薬品容器。
- 前記水素化ブロックコポリマーが水素化ビニル芳香族ポリマーブロックと水素化共役ジエンポリマーブロックとを有する、請求項1~3のいずれか1項に記載の医薬品容器。
- 前記水素化ビニル芳香族ポリマーブロックの水素添加率が50モル%以上であり、前記水素化共役ジエンポリマーブロックの水素添加率が95モル%以上である、請求項4に記載の医薬品容器。
- 前記水素化共役ジエンポリマーブロックの含有量が、前記水素化ビニル芳香族ポリマーブロックおよび前記水素化共役ジエンポリマーブロックの合計100質量%に対して、10~90質量%である、請求項4または5に記載の医薬品容器。
- 前記医薬品容器が、タンパク質製剤を収納する、請求項1~6のいずれか1項に記載の医薬品容器。
- 請求項1~7のいずれか1項に記載の医薬品容器を包装材で包装した包装体であって、
前記医薬品容器と共に、脱酸素剤および脱水剤の少なくとも一方とが包装材で包装されている;または
前記包装材が脱酸素機能および脱水機能の少なくとも一方の機能を有する、包装体。
