(1)前記生物試料を、第一のプローブと第二のプローブと混合する手順1、
ここで、前記第一のプローブは、インフルエンザウイルス由来のノイラミニダーゼ及びバクテリア由来のノイラミニダーゼの双方により分解されて光学的に検出可能な信号を生成するものであり、
前記第二のプローブは、バクテリア由来のノイラミニダーゼにより分解されて光学的に検出可能な信号を生成しかつインフルエンザ由来のノイラミニダーゼにより分解されないものであり、
前記第一のプローブから生成する信号と前記第二のプローブから生成する信号は光学的に区別して検出可能である、と、
(2)前記第一のプローブ及び前記第二のプローブから生成する信号を検出する手順2と、を含み、
(A)前記第二のプローブから生成する信号の強度に対する前記第一のプローブから生成する信号の強度の比が所定値以上である場合に、前記生物試料中のインフルエンザウイルスあるいはインフルエンザウイルス由来ノイラミニダーゼの存在を検出し、
前記比が前記所定値未満である場合に、前記生物試料中のインフルエンザウイルスあるいはインフルエンザウイルス由来ノイラミニダーゼの不存在を検出する、又は、
(B)前記第一のプローブから生成する信号が検出され、前記第二のプローブから生成する信号が検出されない場合に、前記生物試料中のインフルエンザウイルスあるいはインフルエンザウイルス由来ノイラミニダーゼの存在を検出し、

本発明に係るインフルエンザウイルス検出方法は、インフルエンザウイルス由来のノイラミニダーゼによるプローブの加水分解の有無に基づいてインフルエンザウイルスを検出するものである。したがって、本発明において、「インフルエンザウイルスの検出」と「インフルエンザウイルス由来ノイラミニダーゼ」の検出とは同義に用いられる。
手順1:生物試料を、第一のプローブと第二のプローブと混合する手順。
手順2:第一のプローブ及び第二のプローブから生成する信号を検出する手順。
[生物試料]
本発明において、生物試料は、インフルエンザウイルスが含まれ得る、生体由来の材料であれば特に限定されない。
生物試料としては、例えば、鼻腔吸引液、鼻腔ぬぐい液、咽頭ぬぐい液、気管ぬぐい液、唾液、喀痰、血液(全血、血清及び血漿を含む)、尿、細胞や組織、臓器の抽出液等が挙げられる。
本発明に係るインフルエンザ感染の診断方法においては、生物試料は、インフルエンザウイルスに感染した対象又は感染した疑いがある対象から分離される。
対象は、ヒトに限定されず、サル及びブタを含む哺乳類や、アヒル及びニワトリを含む鳥類であってよい。
第一のプローブ(以下「プローブ1」と称する)は、インフルエンザウイルス由来のノイラミニダーゼ及びバクテリア由来のノイラミニダーゼの双方により加水分解されて光学的に検出可能な信号を生成する。
鼻腔、咽喉頭、口腔及び泌尿器等には、Actinomyces, Bacteroides, Clostridium, Corynebacterium, Enterobacteriaceae, Eubacterium, Fusobacterium, Haemophilus, Lactobacillus, Micrococcus, Mycobacterium, Mycoplasma, Neisseria, Peptostreptococcus, Porphyromonas, Prevotella, Propinibacterium, Salmonella, Staphylococcus, Streptococcus, Spirohaetaceae及びVeillonella等の各属に属する多種のバクテリアが常在しており、これらのバクテリアはノイラミニダーゼを有する場合がある。
ここで、光学特性への変化とは、吸光度、旋光度及び屈折率の変化や、蛍光の有無あるいは強度の変化などを意味する。
発色団は、蛍光物質又は化学発光物質であってよいが、蛍光物質とされること好ましい。蛍光物質には、例えば4-メチルウンベリフェロン(4-Methylumbelliferone)、フルオレセイン(Fluorescein)、レゾルフィン(Resorufin)及びローダミン(Rhodamine)などの公知の物質を用いることができる。
このようなプローブ1としては、例えば、2´-(4-メチルウンベリフェリル)-α-D-N-アセチルノイラミン酸(4MU-NANA)が汎用されている。4MU-NANAは、インフルエンザウイルス及びバクテリアに由来するノイラミニダーゼと接触して反応すると、蛍光物質である4-メチルウンベリフェロンを遊離させる。
第二のプローブ(以下「プローブ2」と称する)は、インフルエンザ由来のノイラミニダーゼにより分解されず、バクテリア由来のノイラミニダーゼによってのみ分解されて光学的に検出可能な信号を生成する。
具体的には、例えばプローブ1が生成する信号が、吸光度、旋光度及び屈折率の変化である場合、プローブ2が生成する信号は、蛍光の有無あるいは強度の変化であってよい。あるいは、この場合、プローブ2が生成する信号は、プローブ1が生成する信号とは異なる波長域での吸光度旋光度及び屈折率の変化であってよい。
また、例えばプローブ1が生成する信号が、蛍光の有無あるいは強度の変化である場合、プローブ2が生成する信号は、吸光度、旋光度及び屈折率の変化であってよい。あるいは、この場合、プローブ2が生成する信号は、プローブ1が生成する信号とは異なる波長域での蛍光の有無あるいは強度の変化であってよい。
本発明の1つの好ましい側面においては、mが0であり、R1存在せずに無置換のベンゼン環である。
R2及びR3がアルキル基を示す場合には、該アルキル基にはハロゲン原子、カルボキシ基、スルホニル基、水酸基、アミノ基、アルコキシ基などが1個又は2個以上存在していてもよく、例えばR2又はR3が示すアルキル基はハロゲン化アルキル基、ヒドロキシアルキル基、カルボキシアルキル基などであってもよい。R2及びR3がそれぞれ独立に水素原子又はハロゲン原子であることが好ましく、R2及びR3がともにフッ素原子であるか、塩素原子である場合がより好ましい。
本発明の1つの好ましい側面においては、R6は、-CH2-CF3である。
R7又はR8がアリール基を示す場合には、アリール基は単環の芳香族基又は縮合芳香族基のいずれであってもよく、アリール環は1個又は2個以上の環構成ヘテロ原子(例えば窒素原子、酸素原子、又は硫黄原子など)を含んでいてもよい。アリール基としてはフェニル基が好ましい。アリール環上には1個又は2個以上の置換基が存在していてもよい。置換基としては、例えばハロゲン原子、カルボキシ基、スルホニル基、水酸基、アミノ基、アルコキシ基などが1個又は2個以上存在していてもよい。
一価の置換基としては、ハロゲン、置換されていてもよいアルキル基等が挙げられる。
本発明の1つの好ましい側面においては、tが0であり、R10は存在せずに無置換のベンゼン環である。
本発明の1つの好ましい側面においては、Xは酸素原子である。
酸付加塩としては、例えば、塩酸塩、硫酸塩、硝酸塩などの鉱酸塩、又はメタンスルホン酸塩、p-トルエンスルホン酸塩、シュウ酸塩、クエン酸塩、酒石酸塩などの有機酸塩などを挙げることができる。
塩基付加塩としては、ナトリウム塩、カリウム塩、カルシウム塩、マグネシウム塩などの金属塩、アンモニウム塩、又はトリエチルアミン塩などの有機アミン塩などを挙げることができる。
式(1)で表される化合物は、グリシンなどのアミノ酸との塩を形成する場合もある。
化合物(1)は、水和物又は溶媒和物として存在する場合もある。
したがって、化合物(1)は、インフルエンザウイルスあるいはインフルエンザ由来ノイラミニダーゼを、バクテリアあるいはバクテリア由来ノイラミニダーゼと識別検出するための蛍光プローブとして利用できる。
本発明は、化合物(1)を含む、インフルエンザウイルスあるいはインフルエンザ由来ノイラミニダーゼをバクテリアあるいはバクテリア由来ノイラミニダーゼと識別検出するための試薬をも提供するものである。この試薬は、乾燥された化合物(1)あるいは適当な緩衝液に溶解された化合物(1)であってよい。なお、緩衝液には後述のものを適宜用いることができる。

親水性溶媒には、酵素の反応溶液として従来用いられているものであれば特に限定されず、例えば、水、あるいは、水に酵素反応の最適化のために必要な濃度で緩衝物質を添加したバッファー溶液が用いられる。
プローブ1及びプローブ2から生成する信号は、イメージセンサ、分光光度計、旋光計、分光蛍光光度計及び蛍光顕微鏡等の従来公知の機器を用いて検出し、数値化することができる。生物試料、プローブ1及びプローブ2の混合液(以下、「サンプル液」とも称する)を機器にロードし、機器の操作手順書に従って測定を行えばよい。
一方、プローブ2は、バクテリア由来ノイラミニダーゼによってのみ加水分解されて光学的に検出可能な信号を生成する。プローブ2からの信号強度をS2とする。
一方、サンプル液中に存在するノイラミニダーゼがインフルエンザウイルス由来である場合(対象が感染の場合)、プローブ1からは信号が生成するが、プローブ2から生成する信号は理論的には0であり、プローブ2の自然分解(ノイラミニダーゼの酵素活性によらない分解)により生成する信号を考慮しても十分に小さい。
したがって、S2に対するS1の比(S1/S2)は、サンプル液中にバクテリア由来ノイラミニダーゼが存在する場合(対象が非感染の場合)に比べて、サンプル液中にインフルエンザウイルス由来ノイラミニダーゼが存在する場合(対象が感染の場合)では、より大きくなる。
すなわち、プローブ1から生成する信号の強度S1に対するプローブ2から生成する信号の強度S2の比(S1/S2)が所定値(閾値)以上であれば、サンプル液中にインフルエンザウイルスが存在する(対象がインフルエンザウイルスに感染している)と検出できる。
一方、比(S1/S2)が閾値未満であれば、サンプル液中にインフルエンザウイルスが存在しない(対象がインフルエンザウイルスに感染していない)と検出できる。
これにより、生物試料中に存在するノイラミニダーゼがインフルエンザウイルス由来のものであるかバクテリア由来のものであるかを明確に区別して、バクテリア由来ノイラミニダーゼに起因する信号を擬陽性として検出することなく、インフルエンザウイルスを高確度に検出することができる。
すなわち、プローブ1から生成する信号が検出され、プローブ2から生成する信号が検出されない場合に、サンプル液中のインフルエンザウイルスの存在(対象のインフルエンザ感染)を検出し、プローブ1から生成する信号及びプローブ2から生成する信号が検出された場合には、サンプル液中のインフルエンザウイルスの不存在(対象のインフルエンザウイルス非感染)を検出することができる。
手順2にデジタル法を適用した実施形態を説明する。
デジタル法では、多数の極小空間(例えば微小液滴)中に検出対象物を最大で1分子ずつ存在するように導入する。そして、各極小空間内の検出対象物を検出し、その存在を0又は1で数値化する。これによって、デジタル法では、検出対象物を高感度かつ定量的に検出することが可能である。
手順2A1:インフルエンザウイルス及びバクテリアを収容可能な複数の収容部が、疎水性の上面を有する隔壁によって互いに隔てられて形成されている下層部と、当該下層部における当該収容部が形成されている面に対向している上層部との間の空間に、サンプル液を導入する手順。
手順2A2:前記空間に疎水性溶媒を導入して、前記収容部内に、疎水性溶媒で被覆されかつインフルエンザウイルス及び/又はバクテリアを包含する、サンプル液の液滴を形成する手順と、
手順2A3:前記液滴中でプローブ1及びプローブ2から生成する信号を検出する手順。
図2Aを参照して導入手順2A1を説明する。
アレイ101は、下層部111と上層部121が空間131を隔てて対向して配置されており、空間131を流体流路とするフローセル構造を有している。
下層部111には、インフルエンザウイルス及び/又はバクテリアを収容可能な収容部114が複数形成されている。収容部114は、疎水性の上面を有する隔壁113によって互いに隔てられている。
また、上層部121は、空間131を隔てて、下層部111における収容部114が形成されている面に対向している。
サンプル液4には、界面活性剤を添加してもよい。界面活性剤を添加することで、サンプル液4が、空間131内及び収容部114内へ導入され易くなる傾向がある。

図2Bを参照して封入手順2A2を説明する。本手順では、下層部111と上層部121との間の空間131に疎水性溶媒5を導入する。
疎水性溶媒5として、例えば飽和炭化水素、不飽和炭化水素、芳香族炭化水素、シリコーンオイル、パーフルオロカーボン、ハロゲン系溶媒、及び疎水性イオン液体からなる群より選択される少なくとも1つ又はこれを含む混合物等を好適に用いることができる。飽和炭化水素としては、例えばアルカン、シクロアルカンなどが挙げられる。アルカンとしては、例えばデカン、ヘキサデカン等が挙げられる。不飽和炭化水素としては、例えばスクアレン等が挙げられる。芳香族炭化水素としては、例えばベンゼン、トルエン等が挙げられる。パーフルオロカーボンとしては、例えばフロリナート(登録商標)FC40(SIGMA社製)等が挙げられる。ハロゲン系溶媒としては、例えばクロロホルム、塩化メチレン、クロロベンゼン等が挙げられる。疎水性イオン液体とは少なくとも水中では解離しないイオン液体をさし、例えば1―Butyl―3―methylimidazolium Hexafluorophosphate等が挙げられる。イオン液体とは、室温において液体で存在する塩をさす。
サンプル液4の液滴の極小容積中で、インフルエンザウイルス又はバクテリアのノイラミニダーゼとプローブ1及びプローブ2との反応が進行する。
導入手順2A1と封入手順2A2との間に、空間131内の脱気を行ってもよい。脱気方法としては、例えば、アレイ101を減圧環境下において放置する方法、あるいはアレイ101を冷却する方法等を好適に用いることができる。具体的には、約0.1気圧の減圧デシケータ内にアレイ101を約30秒放置する方法などである。
本手順では、サンプル液4の液滴中で生成したプローブ1及びプローブ2からの信号が検出される。
各液滴においてプローブ1から生成する信号が検出され、プローブ2から生成する信号が検出されない場合(図2C左図参照)に、当該液滴中にインフルエンザウイルスを検出できる。一方、プローブ1から生成する信号及びプローブ2から生成する信号が検出された場合(図2C右図参照)には、当該液滴中にバクテリアの存在(インフルエンザウイルスの不存在)を検出できる。
プローブ1に4MU-NANAを、プローブ2に化合物(1)としてHMRef-S-Neu5Acを用いたデジタル法において、インフルエンザウイルスが封入された液滴から生じた4-メチルウンベリフェロン(4MU)の蛍光をCCDカメラにより検出したイメージの一例を図2D左図に示す。検出領域に信号陽性の液滴が2つ確認できる。インフルエンザウイルスが存在する液滴から4MUが発する青色蛍光(波長443 nm)が検出されている。液滴にバクリテリアが存在する場合には、4MUが発する青色蛍光(波長443 nm)とHMRefが発する緑色蛍光(波長:518nm)が検出される。図2D右図は、信号陽性の液滴が存在しない領域を撮像したイメージの一例である。
なお、インフルエンザウイルスが検出された収容部114が1つでも存在すれば、被検試料についてインフルエンザ陽性と判定してよい。このような判定方法は、検体中のインフルエンザウイルスの数が極めて少ない感染初期の場合にインフルエンザの罹患の有無を判定するため有用である。
手順2にデジタル法を適用した他の実施形態を説明する。
本実施形態に係る手順2は、以下の手順2B1~2B3を含む。
手順2B1:インフルエンザウイルス及びバクテリアを収容可能な複数の収容部が、疎水性の上面を有する隔壁によって互いに隔てられて形成されている基板上に、サンプル液を導入する手順。
手順2B2:サンプル液よりも比重が大きい疎水性溶媒をサンプル液層の上に積層するようにして導入し、サンプル液層と疎水性溶媒層との層置換を生じさせることにより、前記収容部内に、疎水性溶媒で被覆されかつインフルエンザウイルス及び/又はバクテリアを包含する、サンプル液の液滴を形成する手順。
手順2B3:前記液滴中でプローブ1及びプローブ2から生成する信号を検出する検出手順。
図3-1~図3-3を参照して導入手順2B1を説明する。図は、アレイ201の側方断面図である。
アレイ201は、下層部211を含んでなる。アレイ201は、下層部211と側壁221,221によって下方及び両側方を区画されておりかつ上方が開放されている領域21を有する。すなわち、下層部211と側壁221,221は、上方に開口を有する開口ウェルを形成している。当該開口ウェル内の空間を、図中、領域21として示す。
下層部211には、インフルエンザウイルス及び/又はバクテリアを収容可能な収容部214が複数形成されている。収容部214は、疎水性の上面を有する隔壁213によって互いに隔てられている。下層部211は、それに対向する面を有さない。
図3-1~図3-3B-Hを参照して封入手順2B2を説明する。本手順では、疎水性溶媒5をサンプル液4の層の上に積層するようにして導入する。
疎水性溶媒5の導入方法としては、特に限定されないが、例えば、開口ウェルの開口から領域21内に導入することができる。この際、疎水性溶媒5は、図3-1Bに示すように、サンプル液4の層上に疎水性溶媒5の層が積層されるようにして導入することが好ましい。
疎水性溶媒5は、サンプル液4と同様に、層置換を促進するために界面活性剤を含んでいてもよい。
サンプル液4の液滴の極小容積中で、インフルエンザウイルス又はバクテリアのノイラミニダーゼとプローブ1及びプローブ2との反応が進行する。
本手順では、サンプル液4の液滴中で生成したプローブ1及びプローブ2からの信号が検出される。
各液滴においてプローブ1から生成する信号が検出され、プローブ2から生成する信号が検出されない場合に、当該液滴中にインフルエンザウイルスを検出できる。一方、プローブ1から生成する信号及びプローブ2から生成する信号が検出された場合には、当該液滴中にバクテリアの存在(インフルエンザウイルスの不存在)を検出できる。
本発明に係るインフルエンザウイルスの検出キットは、インフルエンザウイルスに感染した対象又は感染した疑いがある対象から分離された生物試料中のインフルエンザウイルスを検出するためのキットである。本発明に係るインフルエンザウイルスの検出キットは、対象におけるインフルエンザ感染の有無の診断に用いられる。本発明に係るインフルエンザウイルスの検出キットは、上述のプローブ1及びプローブ2を含んでなるものであり、これらに加えて任意に上述した疎水性溶媒、及び、アレイ101又はアレイ201を含んでいてよい。これらのキット構成要素(コンポーネント)は、ひとまとまりで販売されるものであってよく、それぞれ単独で販売されるものであってもよい。
アレイ101及びアレイ201の作製のためのフォトリソグラフィー、エッチング、基板積層などの技術は、汎用のマイクロチップやアレイを作成するための技術を適用できる。
上層部121の材質は、例えばガラス、シリコン、高分子樹脂等とできる。
上層部121の厚みは、特に限定されない。
上層部121の下面(空間131に臨む面)は、疎水性であることが好ましい。「疎水性」とは、ここでは「親油性」と同じ意味で用いられ、疎水性溶媒との親和性が親水性溶媒との親和性よりも高いことをいう。
板状部材112の材質は、例えばガラス、シリコン、高分子樹脂等とできる。
板状部材112の厚みは、特に限定されない。
上記アモルファスフッ素樹脂としては、例えば、CYTOP(登録商標)、TEFLON(登録商標)AF2400、およびTEFLON(登録商標)AF1600から選択した少なくとも1つを好適に用いることができる。中でも、微細加工が容易であるという理由で、CYTOP(登録商標)が最も好ましい。
収容部114の大きさは、インフルエンザウイルス及び/又はバクテリアを収容可能であれば特に限定されないが、水平方向の幅及び垂直方向の高さ(隔壁113の高さ)は、それぞれ、例えば100nm-100μm、200nm-90μm、300nm-80μm、400nm-70μm、500nm-60μm、600nm-50μm、700nm-40μm、800nm-30μm、900nm-20μm、1000nm-10μm、2μm-9μm、3μm-8μmの範囲である。
収容部114は、例えば100万個以上が高密度に配置されていてよい。このような超高密度アレイであっても、先に説明した封入手順によれば、疎水性溶媒5で被覆されかつプローブ1、プローブ2、及びインフルエンザウイルス又はバクテリア3を包含する、サンプル液4の液滴を各収容部114に効率よく形成できる。
収容部114の底面が親水性であり、かつ隔壁113の上面及び上層部121の下面が疎水性であることにより、導入手順2A1において、サンプル液4を円滑に収容部114内に導入することができる。また、収容部114の底面が親水性であり、かつ隔壁113の上面が疎水性であることにより、封入手順2A2において疎水性溶媒5が収容部114内に入り込むことを防止できる。
なお、隔壁113は、その上面が疎水性であればよく、その側面(収容部114に臨む面)は、疎水性であっても親水性であってもよい。
アレイ201の下層部211は、アレイ101の下層部111と同様の構成であってよく、下層部211を構成する板状部材212及び隔壁213も、アレイ101の板状部材112及び隔壁113と同様の構成であってよい。
また、収容部214も、アレイ101の収容部114と同様の構成であってよい。
側壁221の高さは、任意であるが、サンプル液4の層と疎水性溶媒5の層とが積層された状態での合計の厚みを上回る必要がある。
以下の反応スキームの手順に従って化合物3を合成した。次に、化合物3を用いてHMRef-S-Neu5Acを合成した。
合成に使用した全ての化学物質は、東京化成工業(株)、和光純薬工業(株)、シグマアルドリッチ(株)から購入し、追加的な精製を行うことなく使用した。
NMRスペクトルは、重水素化溶媒中を用い、Bruker NMR AVANCE III 400分光計[1H(400MHz)、13C(101MHz)]で得た。
高分解能ESI質量スペクトルは、microOTOF II(Bruker)で得た。
HPLC精製は、Inertstil-ODS-3カラム(Φ10×250mm(セミ分取)およびΦ20×250mm(分取))を備えたJASCO PU-2087 Plusポンプ(GL Science Co.、Ltd.)およびUVIDEC-100-V検出器(JASCO)で行った。
HPLCに使用した溶媒は、和光(株)より入手した。シリカゲルカラムクロマトグラフィーは、シリカゲル60N(球状、中性、63~210μm;関東化学株式会社製)を用いて行った。
TLCは、シリカゲルプレートF254(0.25mm(分析);Merck、AKG)で行った。
UV-visスペクトルは、Shimadzu UV-2450分光光度計で得た。
4-ヒドロキシベンズアルデヒド(S、479mg、3.9mmol)およびジイソプロピルエチルアミン(5mL)の溶液に、N-アセチル-2-クロロ-2-デオイズノイラミン酸メチルエステル4,7,8,9-テトラアセテート3(0.5mL、3.1mmol)のMeCN溶液に加え、反応混合物を室温で3時間撹拌した。この後、すべての溶媒を除去し、トルエン(×3)で希釈した。得られた残渣をシリカゲルクロマトグラフィー(EtOAc:DCM、1:1~EtOAc)を用いて精製して、化合物1を白色泡状物として得た(145mg、62%)。
化合物1(247mg、0.42mmol)を乾燥THF(5mL)に溶解し、0℃でLiAlH(OtBu)3(0.83mLの1.0M THF溶液)を加えた。0℃で1時間撹拌した後、飽和NH4Cl(aq)(5mL)およびEtOAc(10mL)を混合物に加え、室温でさらに1時間撹拌した。これにロッシェル塩(水性)(10mL)の飽和溶液を加え、粗生成物をCHCl3(×3)で抽出したのち、有機層を減圧残去した。次いで残留物をシリカゲルクロマトグラフィー(MeOH:DCM 1:99から5:95)を用いて精製して、化合物2を白色泡状物として得た(156mg、63%)。
化合物2(150mg、0.25mmol)をDCM(3mL)に溶解し、三臭化リン(12μL、0.13mmol)を0℃で溶液に添加した。0℃で2時間撹拌後、溶液を飽和NaHCO3水溶液(×3)および飽和食塩水(×1)で洗浄した。有機層を硫酸ナトリウムで乾燥させ、減圧残去して化合物3を白色泡状物(136mg、82%)として得た。
以下の反応スキームの手順に従って、HMRef-S-Neu5Acを合成した。
有機層を硫酸ナトリウム上で乾燥させ、減圧留去した。残渣をMeOH(6mL)に溶解し、1M NaOH(水溶液)を加えた(3mL)。混合物を室温で5時間撹拌し、その後、有機溶媒を蒸発させた。残留水溶液を2MのHClで中和し、次いで凍結乾燥させた。次いで、残留物をMeOH:DCM、1:4溶液に溶解し、濾過して不溶性塩を除去した。この溶液を減圧留去させ、残渣をHPLC(A液:100mM TEAA緩衝液、B液:CH3CN 99%、H2O 1%。A/B=90/10で5分間、その後15分で10/90に勾配をかけ、次いで10/90で15分間)で精製し、凍結乾燥後に橙色の粉末(2ステップで36%)を得た。
直線勾配(0min、20%CH3CN/0.1%TFAaq. To 15min,100%CH3CN 0.1%TFA aq;flow rate=1.0mL/min)を用いてHPLC分析を行った。520nmでの蛍光を調べた。
合成例1で得られたHMRef-S-Neu5Acについて、アルスロバクター・ウレアファシエンス(Arthrobacter ureafaciens)由来のノイラミニダーゼとの酵素反応に伴う蛍光強度の経時変化を調べた。
測定溶液の蛍光スペクトルを、日立F-7000を用いて1秒ごとに測定した(励起波長490nm、発光波長520nm)。測定開始後60秒後にアルスロバクター・ウレアファシエンス由来のノイラミニダーゼ0.01Uを、単独であるいは酵素阻害剤との組み合わせで添加した。測定時の温度は37℃、酵素阻害剤(DANA)の濃度は100μMとした。
ウェルシュ菌(Clostridium perfringens)由来のノイラミニダーゼを用いた場合にも同様の結果を得た。ウェルシュ菌は、ヒトに常在する菌であり、口腔を含む上部気道粘膜にも常在している。
HMRef-S-Neu5Acについて、試験例1と同様にしてインフルエンザイウルス由来のノイラミニダーゼとの酵素反応に伴う蛍光強度の経時変化を調べた。
測定溶液にインフルエンザウイルス(A/PR8 医科研II株)を5×107PFU/mLを添加し、蛍光スペクトルの測定を開始した(励起波長360nm、発光波長448nm)。測定時の温度は25℃とした。
Claims (9)
- 生物試料中のインフルエンザウイルスを検出する方法であって、
(1)前記生物試料を、第一のプローブと第二のプローブと混合する手順1、
ここで、前記第一のプローブは、インフルエンザウイルス由来のノイラミニダーゼ及びバクテリア由来のノイラミニダーゼの双方により分解されて光学的に検出可能な信号を生成するものであり、
前記第二のプローブは、バクテリア由来のノイラミニダーゼにより分解されて光学的に検出可能な信号を生成しかつインフルエンザ由来のノイラミニダーゼにより分解されないものであり、
前記第一のプローブから生成する信号と前記第二のプローブから生成する信号は光学的に区別して検出可能である、と、
(2)前記第一のプローブ及び前記第二のプローブから生成する信号を検出する手順2と、を含み、
前記第二のプローブから生成する信号の強度に対する前記第一のプローブから生成する信号の強度の比が所定値以上である場合に、前記生物試料中のインフルエンザウイルスの存在を検出し、

(1)インフルエンザウイルスに感染した対象又は感染した疑いがある対象から分離された生物試料を、第一のプローブと第二のプローブと混合する手順1、
ここで、前記第一のプローブは、インフルエンザウイルス由来のノイラミニダーゼ及びバクテリア由来のノイラミニダーゼの双方により分解されて光学的に検出可能な信号を生成するものであり、
前記第二のプローブは、バクテリア由来のノイラミニダーゼにより分解されて光学的に検出可能な信号を生成しかつインフルエンザ由来のノイラミニダーゼにより分解されないものであり、
前記第一のプローブから生成する信号と前記第二のプローブから生成する信号は光学的に区別して検出可能である、と、
(2)前記第一のプローブ及び前記第二のプローブから生成する信号を検出する手順2と、を含み、
前記第二のプローブから生成する信号の強度に対する前記第一のプローブから生成する信号の強度の比が所定値以上である場合に、前記対象のインフルエンザウイルスの感染を決定し、
前記比が前記所定値未満である場合に、前記対象のインフルエンザウイルスの非感染を決定する、診断方法。
