Abstract
본 발명은 MLS(Mitochondria Localization Sequence) 펩타이드와 STAT3(Signal Transducer and Activator of Transcription 3)이 융합된 재조합 펩타이드; 상기 재조합 펩타이드를 암호화하는 폴리뉴클레오타이드를 포함하는 재조합 벡터; 및 상기 재조합 펩타이드 또는 상기 재조합 벡터를 유효성분으로 포함하는 자가면역질환 또는 염증성질환의 예방 또는 치료용 조성물으로서, 상기 재조합 펩타이드 또는 상기 재조합 벡터는 STAT3가 미토콘드리아 내에서 과발현되어 미토콘드리아 기능 상향을 통한 IL-17를 포함한 염증성 사이토카인의 발현을 억제함으로써 자가면역질환 또는 염증성질환의 예방 또는 치료에 유용하게 사용할 수 있다.
Description
본 발명은 MLS(Mitochondria Localization Sequence) 펩타이드와 STAT3(Signal Transducer and Activator of Transcription 3)이 융합된 재조합 펩타이드; 상기 재조합 펩타이드를 암호화하는 폴리뉴클레오타이드를 포함하는 재조합 벡터; 및 상기 재조합 펩타이드 또는 상기의 재조합 벡터를 포함하는 자가면역질환 또는 염증성질환의 예방 또는 치료용 조성물에 관한 것이다.
전 세계적으로 면역과민반응으로 인한 질환이 증가하고 있지만, 이러한 질환들의 발생에 대한 근본적인 원인 규명이 충분히 이루어지지 않은 상태이다. 현재 과도한 면역반응에 의한 질환의 치료방법으로는 면역억제제를 단독 또는 병용 투여함으로써 상기 질환에 의해 야기되는 각종 증상을 완화 내지 감소시키는 것이다.
면역억제제란 항원의 작용에 대하여 숙주가 항체를 만드는 능력(체액성 면역반응) 또는 세포성 면역반응을 일으키는 능력을 저하시키거나 차단하기 위해 사용되는 다양한 물질들을 말한다. 이러한 면역억제제는 장기이식분야 뿐만 아니라 루푸스, 류마티스 관절염 등과 같은 자가면역질환과, 아토피, 알러지 등의 피부과민 반응에도 유용하게 사용될 수 있다. 우수한 면역억제제는 면역반응의 불균형을 조절할 수 있어야 하고, 인체에 대한 안전성이 확보되어야 하며, 장기간 치료시에 질환의 재발 발생 빈도가 낮아야 한다.
현재 사용되고 있는 면역억제제로는 사이클로스포린 A와 FK506 등이 있는데, 이들은 복잡한 화학구조를 가진 천연물 유래의 화합물로서 원료 수급의 측면에서 고비용이 드는 문제점이 있어 비경제적이고, 장기투여로 인해 각종 부작용이 야기될 수 있다는 위험성을 내포하고 있다. 따라서 낮은 독성 및 면역관용 유도와 함께 경제적인 생산이 가능한 새로운 면역억제제의 개발이 절실히 요구되고 있는 실정이다.
한편, 각종 병원체에 대한 생체 방어 시스템으로 면역계에서 중심적 역할을 담당하는 세포군의 하나로 T 세포가 있다. T 세포는 인체의 흉선에서 생성되며 일련의 분화 과정을 거치면서 고유의 특성을 지닌 T 세포로 분화하게 되는데, 분화를 완료한 T 세포는 그 기능에 따라 크게 1형 보조 세포(Th1)와 2형 보조 세포(Th2)로 구분된다. 이 중에서 Th1 세포의 주된 기능은 세포 매개성 면역에 관여하고, Th2 세포는 체액성 면역에 관여하며, 면역계에서 이러한 두 세포 집단은 서로 과 활성화되지 않도록 서로 견제를 통해 면역계의 균형을 유지하고 있다.
따라서 면역질환의 대부분은 이러한 두 가지 면역 세포간의 불균형에 기인하는 것으로 볼 수 있는데, 예를 들어 Th1 세포의 활성이 비정상적으로 증가하는 경우 자가면역질환이 발생할 수 있고, Th2 세포의 활성이 비정상적으로 증가하는 경우 과민반응에 의한 면역질환이 발생하는 것으로 알려져 있다.
한편, Th1 세포의 분화에 대한 최근 연구 결과에 따르면, Th1 세포의 활성을 조절할 수 있는 새로운 그룹인 면역조절 T 세포(Treg)의 존재가 알려지면서 이를 이용한 면역질환의 치료에 대한 연구가 대두되고 있는데, Treg 세포는 비정상적으로 활성화된 면역세포의 기능을 억제하여 염증 반응을 제어하는 특성이 있어, Treg 세포의 활성을 증가시키는 작용을 통해 면역질환을 치료하고자 하는 연구들이 많이 진행되고 있다.
또한, Treg 세포 이 외에 T 세포의 분화 과정에서 만들어지는 또 다른 그룹으로 Th17 세포가 있는데, Th17 세포는 미분화 T세포의 분화 과정에서 Treg 세포의 분화와 유사한 과정을 거치며 형성되는 것으로 알려져 있다. 즉, Treg 세포와 Th17 세포의 분화는 공통적으로 TGF-β의 존재 하에서 이루어지지만, Treg 세포의 경우 IL-6을 필요로 하지 않는 반면, Th17 세포의 경우에는 TGF-β와 함께 IL-6가 존재하는 상황에서 분화를 한다. 또한, 분화된 Th17 세포는 IL-17을 분비하는 것을 특징으로 한다.
Th17 세포는 Treg 세포와는 달리 면역질환에서 보이는 염증반응의 최전방에서 관여하여 염증 반응의 신호를 최대화시켜 질병의 진행을 가속화시키는 것이 밝혀지고 있다. 그러므로 자가면역질환 중 Treg 세포에 의해 제어되지 않는 자가면역질환의 경우, Th17 세포 활성의 억제를 표적으로 한 자가면역질환의 치료제 개발이 크게 부각되고 있다.
현재 사용되고 있는 면역질환치료제로는 T 세포에서의 신호변환 경로를 차단하는 면역 억제제가 가장 많이 사용되고 있는데, 이러한 면역억제제들은 독성, 감염, 임파종, 당뇨병, 진전(tremor), 두통, 설사, 고혈압, 오심, 신기능 장애 등의 부작용이 발생하는 문제점이 있다.
또한, T 세포의 활성화를 억제하는 방법을 통해 면역질환을 치료하는 방법 이외에도 면역 세포로부터 분비되는 사이토카인의 양을 조절하는 치료법 및 면역 세포로부터 분비되는 사이토카인을 표적으로 하는 항체를 이용한 치료법이 개발 중에 있다. 그러나 이러한 방법은 실제적으로 임상실험을 거쳐 환자들에게 적용하기까지 많은 시간이 소요되어야 하고, 항체를 이용하는 방법은 항체 제작 과정에서 너무 많은 비용이 든다는 문제점이 있다.
본 발명의 목적은 MLS(Mitochondria Localization Sequence) 펩타이드와 STAT3(Signal Transducer and Activator of Transcription 3)이 융합된 재조합 펩타이드를 제공하는 것이다.
본 발명의 또 다른 목적은 상기 재조합 펩타이드를 암호화하는 폴리뉴클레오타이드를 포함하는 재조합 벡터를 제공하는 것이다.
본 발명의 또 다른 목적은 상기 재조합 펩타이드 또는 상기 재조합 벡터를 유효성분으로 포함하는 자가면역질환 또는 염증성질환의 예방 또는 치료용 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 재조합 펩타이드 또는 상기 재조합 벡터의 유효한 양을 개체에 투여하는 단계를 포함하는 자가면역질환 또는 염증성질환의 예방 또는 치료 방법을 제공하는 것이다.
상기 목적을 달성하기 위하여, 본 발명은 MLS(Mitochondria Localization Sequence) 펩타이드와 STAT3(Signal Transducer and Activator of Transcription 3)이 융합된 재조합 펩타이드를 제공한다.
본 발명의 일 실시예에 있어서, 상기 재조합 펩타이드는 서열번호 1의 아미노산 서열로 이루어진 것일 수 있다.
또한, 본 발명은 상기 재조합 펩타이드를 암호화하는 폴리뉴클레오타이드를 포함하는 재조합 벡터를 제공한다.
본 발명의 일 실시예에 있어서, 상기 폴리뉴클레오타이드는 서열번호 2의 염기서열로 이루어진 것일 수 있다.
또한, 본 발명은 상기 재조합 펩타이드 또는 상기 재조합 벡터를 유효성분으로 포함하는 자가면역질환 또는 염증성질환의 예방 또는 치료용 조성물을 제공한다.

본 발명의 일 실시예에 있어서, 상기 재조합 펩타이드 또는 상기 재조합 벡터는 염증성 사이토카인인 IL-17의 발현을 억제하는 것일 수 있다.
본 발명의 일 실시예에 있어서, 상기 조성물은 비경구로 투여되는 것일 수 있다.

본 발명에 따른 조성물은 STAT3가 미토콘드리아 내에서 과발현되도록 하여 IL-17를 포함한 염증성 사이토카인의 발현을 억제함으로써 자가면역질환 또는 염증성질환의 예방 또는 치료에 유용하게 사용할 수 있다.

본 발명은 MLS(Mitochondria Localization Sequence) 펩타이드와 STAT3(Signal Transducer and Activator of Transcription 3)이 융합된 재조합 펩타이드; 상기 재조합 펩타이드를 암호화하는 폴리뉴클레오타이드를 포함하는 재조합 벡터; 및 상기 재조합 펩타이드 또는 상기의 재조합 벡터를 포함하는 자가면역질환 또는 염증성질환의 예방 또는 치료용 조성물을 제공한다.
본 발명의 재조합 펩타이드는 서열번호 1의 아미노산 서열로 이루어진 것일 수 있고, 서열번호 1의 아미노산 서열로 이루어진 폴리펩타이드에 대해 기능적 동등물을 포함한다.
상기 "기능적 동등물"은 아미노산의 부가, 치환 또는 결실의 결과, 상기 상기 서열번호 1의 아미노산 서열과 적어도 70% 이상, 바람직하게는 80% 이상, 더욱 바람직하게는 90% 이상, 더욱 바람직하게는 95% 이상의 서열 상동성을 갖는 것으로서, 서열번호 1의 아미노산 서열로 이루어진 단백질과 실질적으로 동질의 생리활성을 나타내는 단백질을 말한다. 상기 "실질적으로 동질의 활성"이란 상기 MLS-STAT3의 활성을 의미한다. 상기 기능적 동등물에는, 예를 들어, 본 발명에 따른 아미노산 서열의 아미노산 중 일부가 치환되거나, 결실 또는 부가된 아미노산 서열 변형체가 포함될 수 있다. 아미노산의 치환은 바람직하게는 보존적 치환일 수 있으며, 천연에 존재하는 아미노산의 보존적 치환의 예는 다음과 같다; 지방족 아미노산(Gly, Ala, Pro), 소수성 아미노산(Ile, Leu, Val), 방향족 아미노산(Phe, Tyr, Trp), 산성 아미노산(Asp, Glu), 염기성 아미노산(His, Lys, Arg, Gln, Asn) 및 황함유 아미노산(Cys, Met). 아미노산의 결실은 바람직하게는 본 발명의 재조합 펩타이드 활성에 직접 관여하지 않는 부분에 위치할 수 있다.
본 발명의 재조합 펩타이드를 암호화하는 폴리뉴클레오타이드는 플라스미드 또는 바이러스 벡터와 같은 발현 벡터 내로 공지의 방법에 의해 도입시킨 후 상기 발현 벡터를 당업계에 공지된 다양한 방법으로 형질도입(transduction) 또는 형질주입(transfection)에 의해 발현형으로 표적 세포 내에 도입시킬 수 있다.
플라스미드 발현 벡터는 사람에게 사용할 수 있는 FDA의 승인된 유전자 전달방법으로 사람 세포에 직접적으로 플라스미드 DNA를 전달하는 방법으로서(Nabel, E G et al, Science, 249:1285-1288, 1990), 플라스미드 DNA는 바이러스 벡터와는 달리 균질하게 정제될 수 있는 장점이 있다. 본 발명에서 사용할 수 있는 플라스미드 발현 벡터로는 당업계에 공지된 포유동물 발현 플라스미드를 사용할 수 있다.
본 발명에 따른 핵산을 포함하는 플라스미드 발현 벡터(plasmid expression vector)는 당업계에 공지된 방법, 예를 들어, 일시적 형질주입(transient transfection), 미세주사(microinjection), 형질도입(transduction), 세포융합, 칼슘 포스페이트 침전법, 리포좀 매개된 형질주입(liposome-mediated transfection), DEAE 덱스트란-매개된 형질주입(DEAE Dextran-mediated transfection), 폴리브렌-매개된 형질주입(polybrene-mediated trans fection), 전기침공법(electropora tion), 유전자 건(gene gun) 및 세포 내로 DNA를 유입시키기 위한 다른 공지의 방법에 의해 목적세포 내로 도입할 수 있고, 이에 한정되는 것은 아니다(Wu et al, J Bio Chem, 267:963-967, 1992; Wu et al, Bio Chem, 263:14621-14624, 1988).
또한, 상기 벡터는 공지의 방법에 의해 세포, 조직 또는 체내로 투여될 수 있는데, 예를 들면, 국소적으로, 비경구, 비강, 정맥, 근육 내, 피하 내 또는 다른 적절한 수단에 의해 투여될 수 있다. 특히, 상기 벡터는 표적 조직 또는 표적 세포를 치료하기 위한 유효량으로 직접 주사할 수 있다.
본 발명에 따른 조성물은 자가면역질환 또는 염증성질환을 예방 및 치료할 수 있는 약학 조성물로 사용될 수 있으며, 상기 약학 조성물은 약학적으로 허용되는 담체를 추가로 포함할 수 있다. 상기에서 "약학적으로 허용되는"이란 생리학적으로 허용되고 인간에게 투여될 때, 통상적으로 위장 장애, 현기증 등과 같은 알레르기 반응 또는 이와 유사한 반응을 일으키지 않는 조성물을 말한다. 약학적으로 허용되는 담체로는, 예를 들면, 락토스, 전분, 셀룰로스 유도체, 마그네슘 스테아레이트, 스테아르산 등과 같은 경구투여용 담체 및 물, 적합한 오일, 식염수, 수성 글루코스 및 글리콜 등과 같은 비경구 투여용 담체 등이 있으며, 안정화제 및 보존제를 추가로 포함할 수 있다. 적합한 안정화제로는 아황산수소나트륨, 아황산나트륨 또는 아스코르브산과 같은 항산화제가 있다. 적합한 보존제로는 벤즈알코늄 클로라이드, 메틸- 또는 프로필-파라벤 및 클로로부탄올이 있다. 그 밖의 약학적으로 허용되는 담체로는 다음의 문헌에 기재되어 있는 것을 참고로 할 수 있다(Remington's Pharmaceutical Sciences, 19th ed, Mack Publishing Company, Easton, PA, 1995).
본 발명에 따른 약학 조성물은 상술한 바와 같은 약학적으로 허용되는 담체와 함께 당업계에 공지된 방법에 따라 적합한 형태로 제형화될 수 있다. 즉, 본 발명의 약학 조성물은 공지의 방법에 따라 다양한 비경구 또는 경구 투여용 형태로 제조될 수 있으며, 비경구 투여용 제형의 대표적인 것으로는 주사용 제형으로 등장성 수용액 또는 현탁액이 바람직하다. 주사용 제형은 적합한 분산제 또는 습윤제 및 현탁화제를 사용하여 당업계에 공지된 기술에 따라 제조할 수 있다. 예를 들면, 각 성분을 식염수 또는 완충액에 용해시켜 주사용으로 제형화될 수 있다. 또한, 경구 투여용 제형으로는, 분말, 과립, 정제, 환약 및 캡슐 등이 있고, 이에 한정되는 것은 아니다.
또한, 본 발명은 상기 재조합 펩타이드 또는 상기 재조합 벡터의 유효한 양을 개체에 투여하는 단계를 포함하는 자가면역질환 또는 염증성질환의 예방 또는 치료 방법을 제공한다.
본 발명의 치료 방법은 상기 재조합 펩타이드 또는 상기 재조합 벡터를 치료적 유효량으로 개체에 투여하는 것을 포함한다. 특정 개체에 대한 구체적인 치료적 유효량은 달성하고자 하는 반응의 종류와 정도, 경우에 따라 다른 제제가 사용되는지의 여부를 비롯한 구체적 조성물, 개체의 연령, 체중, 일반건강 상태, 성별 및 식이, 투여 시간, 투여 경로 및 조성물의 분비율, 치료기간, 구체적 조성물과 함께 사용되거나 동시 사용되는 약물을 비롯한 다양한 인자와 의약 분야에 잘 알려진 유사 인자에 따라 다르게 적용하는 것이 바람직하다. 일일 투여량은 본 발명의 약학조성물의 양을 기준으로 0.0001 내지 100 ㎎/㎏이고, 바람직하게는 0.01 내지 100 ㎎/㎏이며, 하루 1 ~ 6 회 투여될 수 있다. 다만, 각 유효성분의 복용량 또는 투여량이 각 유효성분의 함량을 지나치게 높게 포함하여 부작용을 초래하지 않을 정도이어야 함은 당업자에게 자명하다. 따라서 본 발명의 목적에 적합한 조성물의 유효량은 전술한 사항을 고려하여 결정하는 것이 바람직하다.
상기 개체는 임의의 포유동물에 적용가능하며, 상기 포유동물은 인간 및 영장류뿐만 아니라, 소, 돼지, 양, 말, 개 및 고양이 등의 가축을 포함한다.
본 발명의 재조합 펩타이드 또는 재조합 벡터는, 쥐, 생쥐, 가축, 인간 등의 포유동물에 다양한 경로로 투여될 수 있다. 투여의 모든 방식은 예상될 수 있는데, 예를 들면, 경구, 직장 또는 정맥, 근육, 피하, 자궁내 경막 또는 뇌혈관내(intracerebroventricular) 주사에 의해 투여될 수 있다.
이하, 본 발명을 실시예를 통하여 더욱 상세히 설명하기로 한다. 이들 실시예는 본 발명을 보다 구체적으로 설명하기 위한 것으로서, 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예 1. MLS-STAT3 (Mitochondria Localization Sequence-Signal Transducer and Activator of Transcription 3)
과발현의 재조합 벡터 제작 및 과발현 확인
본 발명자들은 STAT3가 미토콘드리아 내로 이동하여 발현되는 벡터(이하, "MLS-STAT3"라 함)를 제조하기 위하여, MLS(Mitochondria Localization Sequence) 서열이 존재하는 GFP 벡터인 pEGFP-N3 벡터에서 BamHI-NotI의 제한효소 부위를 자른 후 상기 부위에 mouse STAT3의 DNA 서열을 삽입하였다. MLS-mouse STAT3 과발현의 재조합 벡터의 개열 지도는 도 1에 나타내었고, MLS-mouse STAT3 과발현의 재조합 벡터의 DNA 서열정보 및 아미노산 서열정보는 도 2에 나타내었다.
이후, 본 발명자들은 제작된 MLS-STAT3가 미토콘드리아 내에서 과발현되는지 확인하기 위하여, 제작된 재조합 벡터를 HEK293 세포에 도입하고, 도입된 세포의 미토콘드리아를 분리한 후, 웨스턴블럿팅을 통해 STAT3의 발현 여부를 확인하는 실험을 수행하였다. 그 결과, MLS-STAT3 재조합 벡터가 삽입된 HEK293 세포의 미토콘드리아 내에서는 STAT3가 과발현됨을 확인하였고, 이는 대조군(Mock) 또는 MLS 서열이 없는 STAT3 재조합 벡터(WT-STAT3)가 삽입된 경우보다 현저하게 발현량이 증가됨을 확인하였다 (도 3).
또한, 본 발명자들은 MLS-STAT3가 HEK293 세포의 미토콘드리아 내에서 과발현되는지 확인하기 위하여 공초점(confocal) 분석을 수행하였다. 그 결과, MLS-STAT3 재조합 벡터가 HEK293 세포에 삽입된 경우, STAT3는 HEK293 세포의 미토콘드리아 내에서 과발현됨을 확인하였다 (도 4).
실시예 2. MLS-STAT3 과발현의 재조합 벡터에 의한 류마티스 관절염의 치료 효과
본 발명자들은 MLS-STAT3 과발현의 재조합 벡터에 의한 류마티스 관절염의 치료 효과를 확인하기 위하여, 류마티스 관절염이 유발된 동물모델에 상기 벡터를 주입한 후 치료 효과를 분석하는 실험을 수행하였다. 간단하게, DBA1/J 마우스에 Type II Collagen을 주입하여 CIA(collagen-induced arthritis)가 유도된 마우스 동물모델을 준비하였고, 관절염을 유발시킨 후 1주일 후부터 마우스에 Mock 또는 MLS-STAT3 재조합 벡터를 전기천공법(electroporation)으로 11주 동안 매주 주입(100μg)하였다. 이후, 관절염 지수(arthritis score) 및 관절염 발병 정도(incidence)를 분석하였다. 관절염 평가 기준은 다음과 같다.
-평가 기준-
0점: 부종이나 종창이 없다.
1점: 발 또는 발목관절에 국한된 경한 부종과 발적
2점: 발목관절에서 족근골(metatarsal)에 걸친 경한 부종과 발적
3점: 발목관절에서 족근골에 걸친 중등도의 부종과 발적
4점: 발목에서 다리 전체에 걸쳐 부종과 발적이 있는 경우
5점 이상: 부종과 발적의 증상이 심각하게 계속 진행되는 경우
그 결과, 관절염이 유도된 마우스에 MLS-STAT3 재조합 벡터를 주입한 경우 대조군(Mock)에 비해 현저하게 관절염 지수가 낮아짐을 확인하였고, 관절염 발병 정도 역시 대조군에 비해 현저하게 낮아짐을 확인하였다 (도 5A).
따라서, MLS-STAT3 재조합 벡터에 의해 류마티스 관절염을 치료할 수 있음을 확인하였다.
실시예 3. MLS-STAT3 과발현의 재조합 벡터에 의한 항체 조절 효과
본 발명자들은 류마티스 관절염이 유도된 동물모델에서 MLS-STAT3 과발현의 재조합 벡터에 의하여 항체 조절 효과가 있는지 확인하는 실험을 수행하였다. 간단하게, 상기 실시예 2에서 사용한 관절염이 유도된 마우스에서 혈액을 수득하고 다시 혈청을 분리한 후, IgG, IgG1 및 IgG2a의 발현량을 ELISA를 통해 분석하였다.
그 결과, MLS-STAT3 재조합 벡터가 주입된 마우스에서는 대조군(Mock)에 비해 IgG의 발현이 감소되었고, Th2 반응과 관련있는 IgG1의 발현은 약간 증가하였으며, Th1 반응과 관련있는 IgG2a의 발현은 감소하였다 (도 5B).
따라서, MLS-STAT3 재조합 벡터가 주입된 경우, Th2와 같은 항염증성 항체는 증가되고, Th1과 같은 염증성 항체의 생성은 억제됨으로써, 류마티스 관절염을 치료할 수 있음을 확인하였다.
실시예 4. MLS-STAT3 과발현의 재조합 벡터에 의한 류마티스 관절염의 연골 손상 억제 효과
본 발명자들은 류마티스 관절염이 유도된 동물모델에서 MLS-STAT3 과발현의 재조합 벡터에 의하여 연골 손상을 억제하는 효과가 있는지 확인하는 실험을 수행하였다. 간단하게, 관절염이 유도된 마우스에서 수득된 관절 조직을 10% 포르말린으로 고정시키고 뼈에서 석회질을 제거한 후 파라핀에 포매한 후 조직을 7 μm 두께의 절편을 만들어서 슬라이드에 부착하였다. 이후, H&E 염색 및 Safranin O 염색을 진행하고, 광학현미경을 통해 골 손상 정도(bone damage) 및 연골 손상 정도(cartillage damage)를 분석하였다.
그 결과, 관절염이 유도된 마우스에 MLS-STAT3 재조합 벡터를 주입한 경우 대조군(Mock)에 비해 골 손상 정도가 약 75% 정도로 낮아졌고, 연골 손상 정도는 약 40% 정도로 현저히 낮아졌음을 확인하였다 (도 6).
따라서, MLS-STAT3 재조합 벡터가 주입된 경우 연골 손상을 억제함으로써 류마티스 관절염을 치료할 수 있음을 확인하였다.
실시예 5. MLS-STAT3 과발현의 재조합 벡터에 의한 관절 내 염증성 사이토카인의 발현 억제 효과
본 발명자들은 류마티스 관절염이 유도된 동물모델에서 MLS-STAT3 과발현의 재조합 벡터에 의하여 관절 내의 염증성 사이토카인의 변화가 있는지 확인하는 실험을 수행하였다. 간단하게, 관절염이 유도된 마우스에서 수득된 관절 조직으로부터 면역조직화학염색(Immunohistochemical Staining)을 수행하여 염증성 사이토카인인 IL-17, TNF-α, IL-6 및 IL-1β의 발현량을 분석하였다.
그 결과, 관절염이 유도된 마우스에 MLS-STAT3 재조합 벡터를 주입한 경우 대조군(Mock)에 비해 IL-17, TNF-α, IL-6 및 IL-1β의 발현량이 유의적으로 감소됨을 확인하였다 (도 7).
따라서, MLS-STAT3 재조합 벡터가 주입된 경우 염증성 사이토카인의 발현량을 감소시킴으로써 류마티스 관절염을 치료할 수 있음을 확인하였다.
실시예 6. 염증성 사이토카인인 IL-17에 의한 미토콘드리아 유전자의 발현 감소 효과
본 발명자들은 염증성 사이토카인인 IL-17에 미토콘드리아의 기능 및 활성에 관여되는 효과를 확인하는 실험을 수행하였다. 우선, FLS(fibroblast-like synoviocyte) 세포에 IL-17을 처리한 후, 미토콘드리아 유전자인 CYCS(Cytochrome C, Somatic), COX5B(cytochrome c oxidase subunit 5B) 및 ATP5O(ATP synthase subunit O)의 발현량을 qPCR를 통해 분석하였다.
그 결과, IL-17을 처리한 경우에는 대조군(Nil)에 비해 미토콘드리아 유전자의 발현량이 감소되었고, H2O2에 의한 ROS 유도와 함께 IL-17을 처리한 경우에는 미토콘드리아 유전자의 발현량이 더욱 감소됨을 확인하였다 (도 8A).
또한, 본 발명자들은 IL-17에 의한 RA(Rheumatoid Arthritis) 유래 FLS 세포 내에서 네크롭토시스(Necroptosis)가 유도되는지 분석하기 위하여, OA(osteoarthritis) 유래 FLS 세포(대조군) 또는 RA 유래 FLS 세포에 IL-17을 처리하고 PI/Annexin V 염색하여 유세포 분석을 수행하였다.
그 결과, IL-17을 처리한 경우, RA 유래 FLS 세포에서는 대조군인 OA 유래 FLS 세포에 비해 초기 사멸(early apoptotic)되는 염증성 세포가 현저히 증가되었고, H2O2 산화 스트레스 하에서 IL-17이 처리된 경우, RA 유래 FLS 세포에서 초기 사멸되는 세포의 수가 특이적으로 증가됨을 확인하였다 (도 8B). 초기 사멸된 세포는 네크롭토시스가 유도된 세포로서, IL-17은 RA 유래 FLS 세포에서 염증성 세포사멸인 네크롭토시스를 유도하는 것으로 확인하였다. 또한, 공초점(confocal) 분석에서는 IL-17을 처리한 경우 항-세포사멸인자인 Bcl2를 활성시키는 것으로 확인되었다 (도 8C).
따라서, 염증성 사이토카인인 IL-17은 미토콘드리아 유전자의 발현을 억제하고, 염증성 세포의 사멸은 증가시키나, 정상 세포의 사멸은 억제시키는 것으로 확인되었다.
실시예7. MLS-STAT3과발현 재조합 벡터에 의한 Th17세포 활성 제어 및 Treg 세포활성 확인
Th17 세포는 염증성 질환의 질병을 가속화 시키는 것으로 알려져 있어, 본 발명의 MLS-STAT3 과발현 벡터가 Th17의 활성을 억제시키는지 확인하였다. 또한 Th17과 유사한 환경에서 분화하는 Treg 세포는, Th1 세포를 조절하여, 염증을 억제하는 것으로 알려져 있어, Treg세포의 활성이 MLS-STAT3 과발현 벡터에 의해 증가하는지도 확인하였다. Th17, Treg 활성 조절과 미토콘드리아 내 STAT3 이동을 확인하기 위하여, 관절염이 유도된 마우스에서, 관절염 유도 후 11주째에 마우스 비장 세포를 수득하여, FITC-conjugated anti-CD4, PE-conjugated antiforkhead box P3(Foxp3), APC-conjugated anti-CD25, FITC-conjugated anti-CD4, PE-conjugated anti-IL-17 및 FITC-conjugated anti-CD4, PE-conjugated anti-pSTAT3(S727), APC-conjugated anti-COX4(eBiosciences, San Diego, CA, USA)로 염색하였다. 염색된 섹션을 Zeiss microscope(LSM 510 Meta; Carl Zeiss, Oberkochen, Germany)를 이용하여 시각화 시켰다.
그 결과, 병인 Th17(CD4+IL-17+) 발현이 대조군 대비 유의적으로 감소한 반면, 면역억제반응을 유도하는 Treg(CD4+CD25+Foxp3+) 세포의 발현이 증가함으로써 관절염 치료 효과를 보일 수 있는 것으로 나타났다(도 9). 또한 미토콘드리아 내에 pSTAT3 727의 이동이 증가됨을 확인하였다. 미토콘드리아 내로의 pSTAT 727의 이동에 의해서 향상된 미토콘드리아 기능에 의해서 류마티스관절염이 제어 됨을 확인 하였다.
실시예8. MLS-STAT3과발현 재조합 벡터에 의한 활막조직 세포 사멸 제어 확인
실시예8-1. MLS-STAT3과발현 재조합 벡터에 의한 염증성 세포사멸 제어 확인
활막조직의 염증성 세포 사멸을 본 발명의 MLS-STAT3 과발현 벡터가 제어하는지 확인하고자 마우스에서 수득된 활막 조직에, RIP1, RIP3 및 pMKL을 처리하여 염증성 세포사멸을 유도하였다. 염증성 세포사멸이 유도된 활막조직에, MLS-STAT3 과발현 벡터를 처리하여, 10% 포르말린으로 고정시키고, 파라핀에 포매한 후 조직을 7 μm 두께의 절편으로 만들어서 슬라이드에 부착하였다. 이후 H&E염색을 진행하고, 광학현미경을 통해, 활막조직의 염증성 세포 사멸을 분석하였다.
그 결과, 도 10에 나타낸 것과 같이 MLS-STAT3 과발현 벡터가 처리된 활막조직에서 RIP1, RIP3 및 pMKL에 의해 유도된 염증성 세포사멸을 조절하는 것을 확인하였다.
실시예8-2. MLS-STAT3과발현 재조합 벡터에 의한 정상 세포사멸 증가 확인
정상 세포사멸은, 관절염이 치료되는 과정중 하나로서, 본 발명의 MLS-STAT3 과발현 벡터가 정상 세포사멸을 증가시키는지 확인하고자, 관절염 유도 마우스의 관절 시노비움 조직내에서 세포사멸인자 BAX의 발현을 조직 분석 하였다. MLS-STAT3 과발현 벡터 주입된 류마티스관절염 동물 모델의 관절 시노비움 조직 내에서 BAX 세포사멸인자가 증가됨을 확인하였다.
그 결과, 대조군에서 보다 MLS-STAT3 과발현 벡터를 처리한 마우스 관절 시노비움 조직에서 정상 세포사멸이 유의하게 증가하는 것을 확인하였다(도 11).
실시예 9. MLS-mouse STAT3 과발현 벡터 제작
본 발명자들은 MLS-STAT3 과발현 마우스를 제작하기 위하여, MLS 서열이 존재하는 pCMV/myc/mito 벡터에서 XhoI-NotI의 제한효소 부위를 자른 후, 상기 부위에 mouse STAT3의 DNA 서열을 삽입하여 MLS-mouse STAT3 과발현의 재조합 벡터를 제작하였다. MLS-mouse STAT3 과발현의 재조합 벡터의 개열지도는 도 12에 나타내었다.
실시예 10. MLS-mSTAT3 과발현 Th17 억제 및 Treg 유도 조절과 병인-조절 마크로파지 조절 확인
상기 실시예 9에서 제작된 MLS-mSTAT3 과발현 트렌스 제닉 마우스의 비장세포에서 병인-조절 세포의 활성 조절 효력을 분석하였다. 정상 마우스와 MLS-mSTAT3 과발현 트렌스 제닉 마우스의 비장세포를 수득하여 PerCP-conjugated anti-CD4, PE-conjugated antiforkhead box P3(Foxp3), APC-conjugated anti-CD25, PerCP-conjugated anti-CD4, FITC-conjugated anti-IL-17 및 FITC-conjugated anti-CD11c, PE-conjugated anti-CD206를 염색하여 유세포 분석 하였다.
그 결과, MLS-mSTAT3 과발현된 상태에서 Th17의 활성이 억제된 것을 확인하였으며, Treg의 활성(Foxp3)은 증가한 것을 확인하였다(도 13). 또한 병인 마크로파지 1 (M1)의 활성은 저해되고 항염증성 마크로파지 2 (M2)의 발현이 증가된것을 확인 하였다.
실시예 11. STAT3 과발현 유도에 따른 류마티스관절염 모델에서의 효과 확인
실시예 11-1. STAT3로 유도된 류마티스관절염에 대한 MLS-mSTAT3 과발현 벡터의 치료효과 확인
STAT3의 과발현으로 유도된 류마티스관절염에서도 본 발명의 MLS-mSTAT3 과발현 벡터의 치료효과가 있는지 확인하였다. DBA1/J 마우스에 STAT3를 과발현하는 벡터를 주입하여, 류마티스관절염이 유도된 동물모델을 준비하였고, 관절염을 유발시킨 후 1주일 후부터 마우스에 STAT3 OVN 단독 또는 MLS-mSTAT3와 재조합된 벡터를 전기천공법으로 11주 동안 매주 주입(100 μg)하였다. 이후, 관절염 지수(arthritis score) 및 관절염 발명 정도(incidence)를 분석하였다. 관절염 평가 기준은 다음과 같다.
-평가 기준-
0점: 부종이나 종창이 없다.
1점: 발 또는 발목관절에 국한된 경한 부종과 발적
2점: 발목관절에서 족근골(metatarsal)에 걸친 경한 부종과 발적
3점: 발목관절에서 족근골에 걸친 중등도의 부종과 발적
4점: 발목에서 다리 전체에 걸쳐 부종과 발적이 있는 경우
5정 이상: 부종과 발적의 증상이 심각하게 계속 진행되는 경우
그 결과, STAT3에 의해 유도된 류마티스관절염 마우스에 MLS-STAT3 재조합 벡터를 주입한 경우, 대조군인 STAT3 OVN 단독 처리군에서보다, 낮은 관절염 지수를 확인하였으며, 관절염 발병 정도 역시 대조군에 비해 낮아짐을 확인함으로서, 본 발명의 MLS-mSTAT3 과발현 벡터는 STAT3 과발현으로 유도된 관절염에서도 효과가 있는 것을 확인하였다(도 14).
실시예11-2. STAT3로 유도된 류마티스관절염에 대한 MLS-mSTAT3 과발현 벡터의 치료효에서 Th17 및 Treg 조절 효과 확인
STAT3의 과발현으로 유도된 류마티스관절염에서도 본 발명의 MLS-mSTAT3 과발현 벡터의 치료효과에서 병인-조절 세포의 활성을 조절 할 수 있는지 확인하고자 하였다. Th17 및 Treg 조절 효과를 확인하기 위하여, 관절염이 유도된 후 35일째에 마우스 비장세포를 수득하여, FITC-conjugated anti-CD4, PE-conjugated antiforkhead box P3(Foxp3), APC-conjugated anti-CD25, FITC-conjugated anti-CD4, PE-conjugated anti-IL-17 및 FITC-conjugated anti-CD4, PE-conjugated anti-pSTAT3(S727), APC-conjugated anti-COX4(eBiosciences, San Diego, CA, USA)로 염색하였다. 염색된 섹션을 Zeiss microscope(LSM 510 Meta; Carl Zeiss, Oberkochen, Germany)를 이용하여 시각화 시켰다.
그 결과, STAT3 과활성 대비 MLS-mSTAT3 투여된 마우스의 비장 세포내에 병인 Th17(CD4+IL-17+) 세포 억제 및 면역억제반응을 유도하는 Treg(CD4+CD25+Foxp3+) 세포의 발현이 증가함을 확인 하였다. 또한 핵 내의 p-STAT3 727 발현이(CD4+p-STAT3 727+) 대조군 대비 감소한 반면, 미토콘드리아 내에 존재하는 p-STAT3 727(CD4+COX4+p-STAT3 727+)은 증가한 것을 확인 하였다. 따라서 미토콘드리아 내로의 STAT3 727의 이동에 의해서 류마티스관절염 질환 제어 및 병인-조절 면역 세포의 조절 효과가 있음을 확인하였다 (도 15).
실시예 12. MLS-mSTAT3 과발현 트랜스 제닉 마우스에서 염증성 장질환 치료 확인
본 발명자들은 본 발명의 MLS-mSTAT3 과발현 트랜스제닉 마우스에서 염증성 장질환에 대한 효과를 확인하기 위하여, 2.5% 덱스트란 설페이트 소듐(dextran sulfate sodium, DSS)물을 1주일간 음용하여, 염증성 장 질환이 유도된 마우스 모델에, 대조군으로 C57BL/6 처리한 마우스 및 본 발명의 MLS-mSTAT3 트랜스 제닉군으로 나누어 확인하여였다. 매일 마우스 개체별 몸무게를 측정하여, 염증성 장질환에 의한 체중 변화를 확인하였다.
그 결과, 대조군의 무게는 투여 5일 이후 급격히 감소하는 것으로 나타났고, MLS-mSTAT3 트랜스 제닉군에서는, 무게의 변화가 없었으며, 체중이 유지되는 것을 확인할 수 있었다(도 16).
실시예 13. 염증성 장질환 유발된 MLS-mSTAT3 트랜스제닉 마우스 내염증형 마크로파지 M1 억제 및 항염증형 마크로파지 M2 조절 확인
염증성 장질환에서 MLS-mSTAT3에 의한 마크로파지에 대한 영향을 확인하기 위하여, 림프구 내의 염증형 마크로파지 M1 및 항염증형 마크로파지 M2를 확인하였다. 상기 실험예 12에서 장질환이 유도된 마우스 실험군을 대상으로, 비장세포를 분리한 후, 비장 세포에서 FITC-conjugated anti-CD11c, PE-conjugated anti-CD206를 염색하여 유세포 분석 하였다.
그 결과, 염증성 장질환 유발된 본 발명의 MLS-mSTAT3 과발현 트랜스제닉 마우스에서, 염증형 마크로파지 M1은 대조군 보다 감소된 것을 확인하였으며, 항염증형 마크로파지 M2는 대조군과 유사한 수준으로 유지된 것을 확인하였다(도 17).
실시예 14. 뇌척수염 질환(EAE)에서의 MLS-mSTAT3 유전자 주입을 통한 EAE자가 항체 억제 확인
MLS-mSTAT3 과발현 트랜스제닉 마우스에 MOG 특이적인 뇌척수염 질환을 유발하였으며, 4주 후에 마우스의 혈청을 채취하여 마우스 항 MOG specific IgG 농도를 ELISA 방법으로 측정하였다. 대조군으로는 C57BL/6 정상 마우스 및 EAE 유발된 C57BL/6을 사용하였다. 항 MOG specific IgG 항체 측정방법은 다음과 같다. MOG를 4μg/ml 농도로 0.05M sodium carbonate coating buffer (pH 9.6)에 희석하여 96 웰(well) 마이크로티터 플레이트(microtiter plate)에 도포한 다음 4℃에서 18시간 방치하였다. 도포된 용액을 제거한 후 비특이적인 결합을 억제하기 위하여 1% 소 혈청 알부민(BSA: Amre-sco, solon, Ohio)이 포함된 TBS(pH 8.0)를 200μl씩 넣고 1시간 실온에서 반응시켰다. 항 MOG specific IgG 측정을 위해서 검체를 1:1,000으로 희석하였으며, 이때 희석 용액은 1% BAS, 0.05% 트윈 20(Amreco)이 포함된 TBS(pH 8.0) 용액을 사용하였다. 다음으로, 희석해 둔 혈청 샘플을 웰(well)당 50μl씩 넣고 실온에서 1시간 동안 반응시켰다. 반응이 끝나면 0.05% 트윈 20(Amresco)이 포함된 TBS 용액(pH 8.0)으로 5회 수세한 후, detection IgG-HRP conjugate(anti-mouse IgG HRP)를 1:75,000으로 희석하여 각 웰당 50μl씩 넣어 실온에서 1시간 동안 반응시켰다. 반응 후, washing buffer로 3회 수세한 후, TMB+H2O2 시스템(KPL, Gaithersbufg, MD)으로 발색하였고, 1N H2SO4를 같은 양으로 넣어 반응을 중지시켰다. 이는 ELISA reader를 이용하여 450nm에서 흡광도로 읽었으며, 항체 측정 결과는 흡광도 자체로 표시하였다.
그 결과, 본 발명의 뇌척수염 발병 MLS-mSTAT3 과발현 트랜스제닉 마우스는 대조군에 비하여 MOG 특이적 항체 MOG specific IgG 항체가 유의적으로 감소된 것을 optical density(O.D) 값을 통해 확인할 수 있었다. 이를 통해 본 발명의 MLS-STAT3가 자가항체 활성 반응을 효과적으로 억제시킬 수 있음을 알 수 있었다(도 18).
실시예 15. MLS-human STAT3 과발현의 재조합 벡터 제작 및 과발현 확인
본 발명자들은 MLS-STAT3 과발현 벡터의 인간 세포 내에서의 검증을 위하여, MLS 서열이 존재하는 pCMV/myc/mito 벡터에서 XhoI-NotI의 제한효소 부위를 자른 후, 상기 부위에 human STAT3의 DNA 서열을 삽입하여 MLS-human STAT3 과발현 재조합 벡터를 제작하였다. MLS-human STAT3 과발현 벡터의 개열지도는 도 19A에 나타내었고, MLS-human STAT3 과발현 벡터의 구체적인 DNA 서열정보 및 아미노산 서열정보는 도 20에 나타내었다.
이후, 본 발명자들은 제작된 MLS-human STAT3 과발현 벡터가 미토콘드리아 내에서 과발현되는지 확인하기 위하여, 제작된 재조합 벡터를 인간유래 NIH3T3 세포에 도입하고, 도입된 세포의 미토콘드리아를 분리한 후, 웨스턴블럿팅을 통해 STAT3의 발현 여부를 확인하는 실험을 수행하였다. 그 결과, MLS-human STAT3 과발현 벡터가 삽입된 NIH3T3 세포의 미토콘드리아 내에서는 대조군(Mock)에 비해 human STAT3가 현저하게 과발현됨을 확인하였다 (도 19B).
또한, 본 발명자들은 MLS-human STAT3 과발현 재조합 벡터를 주입한 후, 염증성 세포사멸인자인 pMLKL(phospho MLKL, mixed lineage kinase domain like pseudokinase) 및 caspase의 발현량을 확인하는 실험을 수행하였다. 그 결과, MLS-human STAT3 과발현 재조합 벡터는 염증성 사멸인자인 pMLKL의 발현을 현저하게 감소시키는 효과가 있음을 확인하였다 (도 19C).
따라서, MLS-human STAT3 과발현 벡터가 주입된 경우 염증성 사멸인자의 발현량을 감소시킴으로써 류마티스 관절염을 치료할 수 있음을 확인하였다.
실시예 16. MLS-human STAT3 과발현 벡터 주입 T세포의 미토콘드리아 기능회복 및 Th17 세포 활성 제어
MLS-human STAT3 과발현 벡터가 T세포에서 미토콘드리아의 기능회복 및 Th17 세포의 활성을 제어하는지 확인하기 위하여, 정상인의 혈액으로부터 PBMC 를 분리하였으며, 분리된 PBMC 에 anti-CD3 와 anti-CD28을 자극하여 T 세포를 활성 시켰으며, T 세포 활성 후 MOCK 및 MLS-human STAT3 과발현 벡터를 각각 과발현 시켰다. 벡터 발현 후 미토콘드리아 포텐셜 분석을 위해서 JC1 염색 하였으며, 유세포 분서하였다. 또한 병인 Th17 세포 및 Th1 세포를 분석하기 위해 각 벡터 발현된 PBMC 에 PerCP-conjugated anti-CD4, PE-conjugated anti-IL-17, FITC-conjugated anti-IFNγ를 염색하였으며, 유세포 분석하였다.
그 결과, MLS-human STAT3 과발현 벡터가 투여된 정상인의 PBMC에서 미토콘드리아 포텐셜이 증가 한것을 확인 하였으며, 병인 Th17 세포는 감소함을 확인 하였다(도 21).
실시예 17. MLS-human STAT3 과발현 벡터의 류마티스관절염 질환 제어
류마티스 관절염이 유발된 마우스 모델을 제조하였는데, DBA1/J 마우스에 제2형 콜라겐을 피하 주사하여(intradermal)관절염을 유도하였으며, 관절염 유도 후 매주 1회씩 총 8회 MLS-human STAT3 과발현 벡터 및 대조군 벡터를 각각 하이드로다이나믹 주입(hydrodynamic injection) 방법을 통해 50ul/mice의 양으로 마우스에 주사하였다. 하이드로다이나믹 주입법은 물리적인 힘을 사용하여 유전자를 세포막을 통과하여 세포질 내로 도입시키는 방법으로 생리 식염수에 희석된 본 발명의 재조합 벡터를 꼬리 정맥으로 주사하는 방법이다. 질환의 유발촉진(Booster)을위해 제2형 콜라겐을 보조제(IFA)와 1:1의 부피비로 섞어 꼬리에 주사하였으며 2일 뒤 오른쪽 허벅지에 MLS-human STAT3를 주입하였고 1주일 뒤 왼쪽 허벅지에 다시 MLS-human STAT3를 주입하였다. 이때 상기 대조군으로는 Human STAT3 유전자를 포함하지 않은 mock 벡터를 주입한 군을 사용하였다. 이후 관절염 증상을 측정하였다.
그 결과, 도 22에 나타낸 바와 같이 MLS-human STAT3 과발현 벡터가 주입된 관절염 마우스 모델의 경우, 관절염 지수가 대조군 보다 유의적으로 낮아지는 것을 확인하였다.
따라서, 본 발명의 MLS-STAT3과발현 벡터는 Th17의 활성 억제 및 Treg를 유도하며, STAT3에 의해 유도된 류마티스 관절염에서도 개선 효과를 가지며, 염증성 장질환에서의 염증형 마크로파지 M1 억제 및 항염증형 마크로파지 M2를 조절하고, 자가면역성 뇌척수 질환에서 자가 항체에 대한 억제능을 가지는 것을 확인하였기에, 면역질환의 예방 또는 치료용 조성물로 이용할 수 있다.

- MLS(Mitochondria Localization Sequence) 펩타이드와 STAT3(Signal Transducer and Activator of Transcription 3)이 융합된 재조합 펩타이드.
- 제 1 항에 있어서,상기 재조합 펩타이드는 서열번호 1의 아미노산 서열로 이루어진 것을 특징으로 하는 재조합 펩타이드.
- 제 1 항의 재조합 펩타이드를 암호화하는 폴리뉴클레오타이드를 포함하는 재조합 벡터.
- 제 3 항에 있어서,상기 폴리뉴클레오타이드는 서열번호 2의 염기서열로 이루어진 것을 특징으로 하는 재조합 벡터.
