Abstract
본 발명은 피부 미백 조성물 및 그 제조방법에 관한 것으로 피부에 자극을 줄 수 있는 화학 첨가물을 최소화하면서 미백 효과를 증대시킬 수 있다. 또한, 멜라닌의 생성 및 멜라닌의 합성에 관여하는 물질을 저해시킴으로써 피부의 흑화를 효과적으로 예방 및 개선시킬 수 있으므로, 다양한 제품에 세포 독성 없이 유용하게 적용할 수 있다.
Description
본 발명은 식물 추출물을 함유하는 피부 미백 조성물에 관한 것이다.
또한, 본 발명은 상기 조성물의 제조방법에 관한 것이다.
색소침착과 같은 피부 흑화는 피부에 존재하는 멜라닌 (melanin)의 합성이 원인이 되는 것으로 알려져 있다. 멜라닌은 멜라닌 형성 세포 내에 위치하는 멜라노좀(melanosome)에 의해 형성되며 형성된 멜라닌은 각질 형성 세포로 이동하여 피부 상피층에 축적되어 색소 침착 현상을 나타낸다. 멜라닌 합성은 아미노산의 하나인 티로신(tyrosin)을 기질로 티로시나아제(tyrosinase)에 의하여 3,4-디히드록시페닐알라닌(3,4-dihydroxyphenylalanine, DOPA)를 거쳐 DOPA 퀴논(quinone)으로 전환되고 효소반응으로 DOPA 크롬(chrome)을 거쳐 멜라닌이 생성되는 과정으로 형성된다 Lee et al., Kor. J. Pharmacogn. 34(1) 33-39, 2003). 이때 생성된 멜라닌은 피부의 색을 검게 하므로 티로시나아제의 활성을 효과적으로 저해하는 실험과 멜라닌 함량을 측정하는 실험이 미백효과를 찾아내려는 연구방법으로 평가 받고 있다.
피부의 심미적 기능을 중요시하는 현대에는 밝고 투명한 피부를 위하여 레이저, 박피 등의 의학 기술을 이용하거나, 미백 성분이 포함된 화장품 등을 이용하고 있다.
따라서, 환한 피부를 원하는 소비자들의 기대에 따라 천연 미백 성분을 포함하는 제품 등의 개발이 이루어지고 있으며, 더욱 효과적인 천연 미백 조성물의 연구가 요구된다.
본 발명의 목적은 피부 미백 조성물을 제공하는 것이다.
또한, 상기 피부 미백 조성물의 제조방법을 제공하는 것이다.
상기 과제를 해결하기 위하여 본 발명은,
배초향, 석류 및 로즈힙을 혼합하여 혼합물을 제조하는 단계;
상기 혼합물을 열수 추출하여 추출물을 제조하는 단계;
상기 추출물에 효소를 첨가하여 반응시키는 효소 처리 단계; 및
상기 효소 처리 단계 후 농축하여 조성물을 수득하는 단계를 포함하는 피부 미백 조성물 제조방법을 제공한다.
일구현예에 따르면, 상기 조성물을 건조시키는 단계를 포함할 수 있다. 또한, 상기 건조시키는 단계는 동결건조 또는 분무건조 단계를 포함할 수 있다.
일구현예에 따르면, 상기 배초향, 석류 및 로즈힙의 중량비는 2:2:1일 수 있다.
일구현예에 따르면, 상기 효소는 글루코아밀라아제(glucoamylase), Peclyve CP 및 β-글루코시다아제(β-glucosidase)로 이루어지는 군으로부터 선택되는 하나 이상을 포함할 수 있다.

본 발명의 피부 미백 조성물 및 그 제조방법에 따르면, 피부에 자극을 줄 수 있는 화학 첨가물을 최소화하면서 미백 효과를 증대시킨 조성물을 제공할 수 있다. 또한, 멜라닌의 생성 및 멜라닌의 합성에 관여하는 물질을 저해함으로써 피부의 흑화를 효과적으로 예방 및 개선시킬 수 있다. 그러므로, 다양한 제품에 세포 독성 없이 유용하게 적용할 수 있다.

배초향(
Agastache
rugosa)은 꿀풀과의 다년생 초본으로, 독특한 향을 가지고 있는 식물이다. 배초향은 유리당 성분으로 프룩토스(fructose), 글루코스(glucose), 수크로스(sucrose), 말토스(maltose) 등을 포함하고, 아미노산으로 글루탐산(glutamic acid), 아스파트산(aspartic acid), 류신(leucine) 등을 포함하고, 무기질로 칼륨, 칼슘, 나트륨, 마그네슘 등을 포함하고, 아스코르브산(ascorbic acid), 폴리페놀류 등을 포함한다. 배초향을 달인 물은 신체의 옴이나 버짐에 외용제로서 사용되기도 한다.
석류(

일구현예에 따르면, 본 발명의 화장품 조성물은 당업계에서 통상적으로 제조되며, 피부학적으로 적용이 가능한 어떠한 제형으로도 제조될 수 있다. 피부학적으로 적용이 가능하다는 것은 적용 대상에게 비교적 비독성이고 무해한 유효작용을 할 수 있는 조성물로서, 피부에 적용할 수 있는 외용제를 포함할 수 있으며, 상기 조성물로부터 기인하는 부작용이 유효 성분의 효능을 저하시키기 않고, 적용 대상에 심각한 자극을 유발하지 않으면서 유효성분들의 활성 및 물성을 손상시키지 않는다는 것을 의미할 수 있다. 본 발명의 피부학적으로 적용이 가능한 화장품 조성물로는 예를 들어, 용액, 현탁액, 유액, 유탁액, 페이스트, 겔, 팩, 크림, 로션, 파우더, 비누, 계면활성제-함유 클렌징, 오일, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션, 스프레이 및 모발 화장품 등으로 제형화될 수 있으나, 이에 한정되는 것은 아니다. 구체적으로, 상기 화장품 조성물은 스킨로션, 스킨소프너, 스킨토너, 아스트린젠트, 로션, 겔, 밀크로션, 모이스쳐 로션, 영양로션, 맛사지크림, 영양크림, 모이스처크림, 핸드크림, 파운데이션, 에센스, 엠플, 영양에센스, 팩, 비누, 헤어샴푸, 풋샴푸, 클렌징폼, 클렌징로션, 클렌징크림, 바디로션, 바디클린저 등의 제형 등으로 제조될 수 있다.
본 발명의 다른 구현예에 따르면, 상기와 같은 조성물을 포함하는 기능성 식품 조성물을 제공할 수 있다. 본 발명에 따른 조성물이 기능성 식품 조성물에 함유되는 경우 그대로 첨가되거나 다른 식품 성분과 함께 함유될 수 있고, 통상적인 방법에 따라 적절하게 포함될 수 있다.
일구현예에 따르면, 식품 또는 기능성 식품 조성물의 형태는 산제, 과립제, 환, 정제, 캡슐제의 형태 등으로 제조될 수 있고, 일반적인 식품 또는 음료의 형태라면 특별히 제한되지 않는다. 식품 조성물의 종류로는 통상적인 의미의 식품을 모두 포함할 수 있으며, 예를 들면, 육류, 소세지, 빵, 쵸콜렛, 캔디류, 스넥류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차, 드링크제, 알콜 음료, 비타민 복합제 등을 포함할 수 있으나, 이에 한정되지는 않는다.
상기 음료수 및 음료는 통상의 음료와 같이 일반적인 향미제 또는 천연 탄수화물 등을 추가 성분으로 포함할 수 있다. 상기 천연 탄수화물은 포도당, 과당과 같은 모노사카라이드, 말토스, 슈크로스와 같은 디사카라이드 및 덱스트린, 사이클로덱스트린과 같은 폴리사카라이드, 자일리톨, 소르비톨, 에리트리톨 등의 당알콜을 포함할 수 있다. 감미제로는 타우마틴, 스테비아 추출물과 같은 천연 감미제, 사카린, 아스파르탐과 같은 합성 감미제 등을 포함할 수 있다.
또한, 본 발명의 식품 조성물은 일반적인 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산음료에 사용되는 탄산화제 등을 1종 이상 포함할 수 있고, 천연 과일쥬스, 과일쥬스 음료 및 야채 음료에 함유되는 과육을 포함할 수 있다.
이하, 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있도록 본 발명의 실시예에 대하여 상세히 설명한다. 그러나 본 발명은 여러 가지 상이한 형태로 구현될 수 있으며 여기에서 설명하는 실시예에 한정되지 않는다.
비교예 1: 배초향, 석류 및 로즈힙 열수 추출물 제조
배초향, 석류, 로즈힙을 고형분 비율 2:2:1로 혼합하여 혼합물을 제조한 다음, 상기 혼합물 중량의 20배수의 물을 넣고 95℃에서 240분간 교반하면서 추출물을 수득하였다. 상기 추출물을 와트만(Whatman) 4번 여과지로 감압 여과하고, 여과된 추출액을 진공 회전 농축기로 농축하여 용매 성분을 제거하여 조성물을 제조하였다.
비교예 2: 비스코자임(Viscozyme L) 처리 조성물
배초향, 석류, 로즈힙을 고형분 비율 2:2:1로 혼합하여 혼합물을 제조한 다음, 상기 혼합물 중량의 20배수의 물을 넣고 95℃에서 240분간 교반하면서 추출물을 수득하였다. 상기 추출물에 비스코자임(Viscozyme L)을 질량 대비, 0.2%(w/w)가 되도록 첨가하였다. 상기 효소를 첨가한 추출물을 40℃가 되도록 유지시키고, 3시간 동안 교반하면서 가수분해 반응을 진행시켰다. 효소 처리된 복합 추출물은 와트만(Whatman) 4번 여과지로 감압 여과시키고, 여과된 추출액을 진공 회전 농축기로 농축하여 용매 성분을 제거하여 조성물을 제조하였다.
실시예 1: 글루코아밀라아제(glucoamylase) 처리 조성물
배초향, 석류, 로즈힙을 고형분 비율 2:2:1로 혼합하여 혼합물을 제조한 다음, 상기 혼합물 중량의 20배수의 물을 넣고 95℃에서 240분간 교반하면서 추출물을 수득하였다. 상기 추출물에 글루코아밀라아제(glucoamylase)를 질량 대비, 0.2%(w/w)가 되도록 첨가하였다. 상기 효소를 첨가한 추출물을 60℃가 되도록 유지시키고, 3시간 동안 교반하면서 가수분해 반응을 진행시켰다. 효소 처리된 복합 추출물은 와트만(Whatman) 4번 여과지로 감압 여과시키고, 여과된 추출액을 진공 회전 농축기로 농축하여 용매 성분을 제거하여 피부 미백 조성물을 제조하였다.
실시예 2: Peclyve CP 처리 조성물
배초향, 석류, 로즈힙을 고형분 비율 2:2:1로 혼합하여 혼합물을 제조한 다음, 상기 혼합물 중량의 20배수의 물을 넣고 95℃에서 240분간 교반하면서 추출물을 수득하였다. 상기 추출물에 Peclyve CP를 질량 대비, 0.2%(w/w)가 되도록 첨가하였다. 상기 효소를 첨가한 추출물을 45℃가 되도록 유지시키고, 3시간 동안 교반하면서 가수분해 반응을 진행시켰다. 효소 처리된 복합 추출물은 와트만(Whatman) 4번 여과지로 감압 여과시키고, 여과된 추출액을 진공 회전 농축기로 농축하여 용매 성분을 제거하여 피부 미백 조성물을 제조하였다.
실시예 3: β글루코시다아제(β-glucosidase) 처리 조성물
배초향, 석류, 로즈힙을 고형분 비율 2:2:1로 혼합하여 혼합물을 제조한 다음, 상기 혼합물 중량의 20배수의 물을 넣고 95℃에서 240분간 교반하면서 추출물을 수득하였다. 상기 추출물에 β글루코시다아제(β-glucosidase)를 질량 대비, 0.2%(w/w)가 되도록 첨가하였다. 상기 효소를 첨가한 추출물을 50℃가 되도록 유지시키고, 3시간 동안 교반하면서 가수분해 반응을 진행시켰다. 효소 처리된 복합 추출물은 와트만(Whatman) 4번 여과지로 감압 여과시키고, 여과된 추출액을 진공 회전 농축기로 농축하여 용매 성분을 제거하여 피부 미백 조성물을 제조하였다.
실험예 1: 멜라닌 농도 측정
멜라닌 농도 측정에 사용할 세포로, 마우스 흑색종 세포주인 B16F10 세포를 페니실린(penicillin) 100 IU/ml 및 스트렙토마이신(streptomycin) 100 μg/ml과 10%의 소 태아 혈청(FBS)을 함유하는 Dulbeco's Modified Eagle Media(DMEM)를 이용하여 100mm 세포 배양 접시(cell culture dish)에 70 내지 80%의 밀도로 배양하였다.
배양시킨 B16F10 세포를 페니실린(penicillin) 100 IU/ml 및 스트렙토마이신(streptomycin) 100 μg/ml과 10%의 소 태아 혈청(FBS)을 함유하는 Dulbeco's Modified Eagle Media(DMEM)를 이용하여 5Х10
4 cell/ml의 농도로 조절한 후, 12 웰 플레이트(well plate)에 접종하고, 5% CO
2 및 37℃에서 전 배양하였다. 세포가 처리된 플레이트에 각각의 실시예 및 비교예에 따른 조성물을 50ug/ml 농도로 처리하였고, 대조군으로 조성물을 무처리한 것을 사용하였다. 각각의 웰에 α-멜라노사이트 자극 호르몬(α-melanocyte-stimulating hormone, α-MSH)을 처리하여 배양시켰다. 72시간 후 배지를 제거하고 멜라닌의 용해를 위하여 1N NaOH를 첨가하였다. 멜라닌의 함량은 Elisa microplate reader를 사용하여 405nm에서 흡광도를 측정함으로써 분석하였고, 그 결과는 도 1에 나타내었다. 도 1에 나타난 바와 같이, 효소를 처리하지 않은 혼합 추출물인 비교예 1의 경우 멜라닌의 함량 감소율이 매우 적게 나타난 반면, 실시예의 조성물의 멜라닌 억제 효과가 우수한 것을 확인할 수 있다. 특히, 실시예 3의 경우 멜라닌 생성 억제 효과가 뛰어난 것을 알 수 있다.
실험예 2: 티로시나아제(tyrosinase) 활성 측정
실험예 1과 같이 배양시킨 B16F10 세포를 사용하였다. 상기의 세포가 처리된 플레이트에 각각의 실시예 및 비교예에 따른 조성물을 50ug/ml 농도로 처리하였고, 대조군으로 조성물을 무처리한 것을 사용하였다. 각각의 웰에 α-멜라노사이트 자극 호르몬(α-melanocyte-stimulating hormone, α-MSH)을 처리하여 배양시켰다. 72시간 후, RIPA 버퍼(buffer)를 이용하여 세포를 파괴시킨 후 14,000rpm에서 20분 동안 원심분리하여 상등액을 얻었다. 상등액에 0.1% L-DOPA(L-3,4-dihydroxyphenylalanine)를 첨가한 후 37℃에서 3시간 동안 배양하였다. 티로시나아제의 활성은 Elisa microplate reader로 475nm에서 흡광도를 측정함으로써 분석하였으며, 그 결과는 도 2에 나타내었다. 도 2에 나타난 바와 같이, 비교예 1 및 2를 사용한 경우 티로시나아제의 활성 감소율이 매우 적게 나타난 반면, 실시예예 따른 조성물을 사용한 경우 티로시나아제의 활성이 크게 감소하는 것을 확인할 수 있다. 특히, 실시예 3을 처리한 경우의 티로시나아제 활성 억제 효과가 뛰어난 것을 알 수 있다.
상기와 같은 결과로부터 확인할 수 있는 바와 같이, 본 발명은 멜라닌의 생성을 감소시킬 수 있다. 또한, 멜라닌 형성에 관여하는 티로시나아제의 활성을 감소시킬 수 있으므로 멜라닌의 생성을 예방하여 피부 미백 효능을 증대시킬 수 있음을 확인하였다.
이상의 설명은 본 발명의 기술 사상을 예시적으로 설명한 것에 불과한 것으로서, 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자라면 본 발명의 본질적인 특성에서 벗어나지 않는 범위에서 다양한 수정 및 변형이 가능하다. 또한, 본 발명에 개시된 실시 예들은 본 발명의 기술 사상을 한정하기 위한 것이 아니라 설명하기 위한 것이고, 이러한 실시 예에 의하여 본 발명의 기술 사상의 범위가 한정되는 것은 아니다. 본 발명의 보호 범위는 아래의 청구범위에 의하여 해석되어야 하며, 그와 동등한 범위 내에 있는 모든 기술 사상은 본 발명의 권리범위에 포함되는 것으로 해석되어야 할 것이다.
Claims (12)
- 배초향, 석류 및 로즈힙을 혼합하여 혼합물을 제조하는 단계;상기 혼합물을 열수 추출하여 추출물을 제조하는 단계;상기 추출물에 효소를 첨가하여 반응시키는 효소 처리 단계; 및상기 효소 처리 단계 후 농축하여 조성물을 수득하는 단계를 포함하는 피부 미백 조성물 제조방법.
- 제1항에 있어서,상기 조성물을 건조시키는 단계를 포함하는 것인, 조성물 제조방법.
- 제2항에 있어서,상기 건조시키는 단계가 동결건조 또는 분무건조 단계를 포함하는 것인, 조성물 제조방법.
- 제1항에 있어서,상기 배초향, 석류 및 로즈힙의 중량비가 2:2:1인 것인, 피부 미백 조성물 제조방법.
- 제1항에 있어서,상기 효소가 글루코아밀라아제(glucoamylase), Peclyve CP 및 β-글루코시다아제(β-glucosidase)로 이루어지는 군으로부터 선택되는 하나 이상을 포함하는 것인, 피부 미백 조성물 제조방법.
- 제1항에 있어서,상기 추출물을 제조하는 단계가 상기 혼합물 중량의 1 내지 100배수의 물을 첨가하는 단계; 및50 내지 120℃에서 100 내지 300분 동안 교반시키는 단계를 포함하는 것인, 피부 미백 조성물 제조방법.
- 제1항에 있어서,상기 효소 처리 단계가 30 내지 70℃에서 2 내지 10시간 동안 교반시키는 단계를 포함하는 것인, 피부 미백 조성물 제조방법.
- 제1항 내지 제7항 중 어느 한 항에 따라 제조되는 피부 미백 조성물.
- 제8항에 있어서,상기 조성물이 멜라닌의 생성을 억제하는 것인, 피부 미백 조성물.
- 제8항에 있어서,상기 조성물이 티로시나아제의 활성을 억제하는 것인, 피부 미백 조성물.
- 제8항에 따른 조성물을 포함하는 식품 조성물.
- 제8항에 따른 조성물을 포함하는 화장품 조성물.
