一般式(I)で表されるヒドラジノプリン化合物(I)において、R1、R2及びR3が全て水素原子であるヒドラジノプリン化合物3a,b(表1、化合物番号I-1~I-22)の製造法は特に限定されないが、次に示す反応式に従って合成することができる(Scheme 1)。
(Scheme 1中、R4は水素原子又はメチル基を示し、R5はアルキル基又はアリール基を示す。)
本明細書中において、Acはアセチル基を示し、AcOHは酢酸を示し、及びTFAはトリフルオロ酢酸を示す。
この工程は公知であり、化合物2a,bは既知文献(T.Nagamatsu,et al,J.Chem.Soc.Perkin Trans.1,3117(1999)、該文献の全記載はここに開示として援用される)及び(T.Nagamatsu,et al,PCT Int.Appl.(1996),WO9626208、該文献の全記載はここに開示として援用される)に従いそれぞれ調製することができる。
公知合成法(T.Nagamatsu,et al,J.Chem.Soc.Perkin Trans.1,3117(1999))に従い、表1に記した種々のプリンアルデヒドヒドラゾン類化合物3a,b(表1、化合物番号I-1~I-22)を調製することができる。なお、化合物番号I-1~I-7及びI-14~I-18は公知の化合物であり、化合物番号I-8~I-13及びI-19~I-22の化合物は新規の化合物である。
本明細書中において、Etはエチル基を示し、EtOHはエタノールを示し、及びDMFはN,N’-ジメチルホルムアミドを示す。
この工程は、常法に従い化合物4より化合物5を調製することができる。
エタノール中、化合物4(1g)に対して包水ヒドラジン(4mL)を加え、30分間加熱還流する。反応後、析出した結晶を濾取し、これを水から再結晶すると無色の針状結晶の化合物5を得ることができる。
第2工程で述べた同様の合成法により、表1に記した種々の新規のプリンアルデヒドヒドラゾン類化合物6(表1、化合物番号I-23~I-28)を調製することができる。
化合物5(2.6mmol)に対してR5-CHO(式中、R5はアルキル基又はアリール基を示す。)で表されるアルデヒド(3.1mmol)を用い、N,N’-ジメチルホルムアミド(DMF)(40mL)中、10~30°Cで0.5~2時間反応させて化合物6(表1、化合物番号I-23~I-28)を得ることができる(Scheme 2)。
(Scheme 3中、R2はアルキル又はアリール基を示し、R5はアリール基を示す。)
この工程は、常法に従い化合物7a-cより化合物8a-cを調製することができる。
ピリジン又はβ-ピコリン(200mL)に化合物7a-c(15mmol)と五硫化二りん(45mmol)を加え、8時間加熱還流する。反応後、減圧下溶媒を留去し、熱水で処理し、析出した結晶を濾取する。これをDMFと水の混合溶媒中活性炭処理及び再結晶して黄色の粉末結晶の化合物8a-cを得ることができる。
この工程は、常法に従い化合物8a-cより化合物9a-cを調製することができる。
エタノール中(3~4mL)、化合物8a-c(1g)に対して包水ヒドラジン(3~4mL)を加え、1時間加熱還流する。反応後、析出した結晶を濾取し、これを水から再結晶すると無色の針状結晶の化合物9a-cを得ることができる。
この工程の化合物は第2工程で述べた合成法により、表1に記した種々のプリンアルデヒドヒドラゾン類化合物10a-c(表1、化合物番号I-29~I-43)を調製することができる。
TFA(8mL)に化合物9a-c(1.8~2.8mmol)と各種アルデヒド(2.2~3.3mmol)を加え、それぞれ室温で0.5~1時間撹拌する。反応後、減圧下溶媒を留去し、ジエチルエーテルで処理し、析出した結晶を濾取する。これを1%aq.KHCO3で洗浄し、得られた固体をDMFと水の混合溶媒から再結晶して化合物10a-c(表1、化合物番号I-29~I-43)を得ることができる(Scheme 3)。
(ii-1)R6、R7及びR8が全て水素原子であり、R9が水素原子、アルキル基又はアリール基である。

この工程は、ヒドラジノ化合物2aにオルトエステル類を加え加熱して化合物12(表2、化合物番号II-1~II-3)を調製することができる。
酢酸(8mL)に6-ヒドラジノ-7H-プリン-2(3H)-オン化合物2a(1.2mmol)と各種オルトエステル(4.8mmol)を加え、それぞれ80℃で10~20分間加熱撹拌する。反応後、室温まで冷却して析出した結晶を濾取し、これをDMFと水の混合溶媒から再結晶して化合物12(表2、化合物番号II-1~II-3)を得ることができる。
この工程は、適切な酸化剤を用いて化合物3aを酸化し、化合物3aが閉環した化合物12(表2、化合物番号II-3~II-10)を調製することができる。
(Route i):TFA(6mL)に6-n-オクチリデンヒドラジノ-7H-プリン-2(3H)-オン又は6-アリールメチリデンヒドラジノ-7H-プリン-2(3H)-オン類化合物3a(1mmol)と70%硝酸(1.5mmol)を加え、それぞれ室温で10分間撹拌する。反応後、減圧下溶媒を留去し、ジエチルエーテルで処理し、析出した結晶を濾取する。これを1%aq.KHCO3で洗浄し、得られた固体をDMFと水の混合溶媒から再結晶して化合物12(表2、化合物番号II-3~II-10)を得ることができる。
(Route ii):DMF(20mL)に6-アリールメチリデンヒドラジノ-7H-プリン-2(3H)-オン類化合物3a(1mmol)とクロラニル(1.5mmol)を加え、それぞれ10分間加熱還流する。反応後、減圧下溶媒を留去し、エタノールとジエチルエーテルで処理し、析出した結晶を濾取する。これをDMFと水の混合溶媒中活性炭処理及び再結晶して化合物12(表2、化合物番号II-3~II-10)を得ることができる。
(Scheme 5中、R10とR12は両方ともメチル基を示し、R11は水素原子を示し、R13は水素原子、メチル基又はアリール基を示す。)
本明細書中において、DMFDMAはN,N’-ジメチルホルムアミドジメチルアセタールを示し、DNPAは、O-(2,4-ジニトロフェニル)ヒドロキシルアミンを示し、MeONaMeOHはナトリウムメトキシドメタノール溶液を示す。
この工程は、ヒドラジノプリン化合物5にオルトエステル類を加え加熱して化合物13(表3、化合物番号III-1~III-3)を調製することができる。
DMF(20mL)に6-ヒドラジノ-3,7-ジメチル-7H-プリン-2(3H)-オン化合物5(1.03mmol)と各種オルトエステル(2.06mmol)を加え、それぞれ1時間加熱還流する。反応後、減圧下溶媒を留去し、酢酸エチルで処理し、析出した結晶を濾取する。これをDMFと水の混合溶媒から再結晶して化合物13(表3、化合物番号III-1~III-3)を得ることができる。
この工程は、化合物6(表1、化合物番号I-23~I-28)を適切な酸化剤を用いて酸化し、閉環することで化合物13(表3、化合物番号III-2~III-7)を調製することができる。
DMF(30mL)に6-アルキリデンヒドラジノ-3,7-ジメチル-7H-プリン-2(3H)-オン又は6-アリールメチリデンヒドラジノ-3,7-ジメチル-7H-プリン-2(3H)-オン類化合物6(1mmol)と70%硝酸(1.5mmol)を加え、それぞれ100℃で0.5~1時間撹拌する。反応後、減圧下溶媒を留去し、水で処理し、析出した結晶を濾取する。これをDMFと水の混合溶媒から再結晶すると化合物13(表3、化合物番号III-2~III-7)を得ることができる。
この工程は、化合物13(表3、化合物番号III-1~III-7)をアルカリ加水分解することによってその開環化合物である化合物14a-gを調製できる。
0.1Nナトリウムメトキシドメタノール溶液(10~15mL)に6,9-ジメチル-9H-1,2,4-トリアゾロ[3,4-i]プリン-5(6H)-オン類化合物13(表3、化合物番号III-1~III-7)(1mmol)を加え、それぞれ室温で0.5~1時間撹拌する。反応後、浮遊物を濾去して減圧下溶媒を留去し、水を加えて10%aq.HClで中和すると結晶が析出する。これを濾取し、適切な有機溶媒から再結晶して化合物14a-gを得ることができる。
この工程は、上述の化合物14a-gをジフェニルエーテル中で加熱閉環することで化合物12(表2、化合物番号II-11~II-17)を調製できる。
ジフェニルエーテル(8mL)に化合物14a-g(1mmol)を加え、それぞれ200℃で2時間加熱撹拌する。反応後、室温まで冷却して析出した結晶を濾取し、ジエチルエーテルで洗浄し、これをDMFから再結晶すると化合物12(表2、化合物番号II-11~II-17)を得ることができる。
この工程は、前述の第9工程で得た化合物12(II)の構造確定のための工程であり、化合物12(表2、化合物番号II-1~II-3)をDMFDMAによるメチル化で得た化合物12(表2、化合物番号II-11~II-13)は、第13工程から得られた化合物12(表2、化合物番号II-11~II-13)と同定できる。
DMF(10mL)に対応する1H-[1,2,4]トリアゾロ[5,1-i]プリン-5(4H)-オン類化合物12(表2、化合物番号II-1~II-3)(1mmol)とN,N’-ジメチルホルムアミドジメチルアセタール(DMFDMA)(10mmol)を加え、それぞれ2~3時間加熱還流する。反応後、減圧下溶媒を留去し、酢酸エチルで処理し、析出した結晶を濾取する。これをDMFから再結晶して化合物12(表2、化合物番号II-11~II-13)を得ることができる。
この工程は、さらに化合物12の構造確定のための工程で、別途合成法での出発物質である化合物16の合成である。
DMF(100mL)に既知の化合物である6-アミノ-2,3-ジヒドロ-3,7-ジメチル-2-オキソ-7H-プリン化合物15(1.12mmol)とO-(2,4-ジニトロフェニル)ヒドロキシルアミン(DNPA)(1.68mmol)を加え、80℃で1時間加熱撹拌する。反応後、減圧下溶媒を留去し、残渣をシリカゲルカラムクロマトグラフィー(Kieselgel 70-230 mesh)(酢酸エチル:エタノール=4:1)によって分離精製し、得られた固体をエタノールから再結晶して無色の粉末結晶の1-アミノ-1,2,3,6-テトラヒドロ-6-イミノ-3,7-ジメチル-2-オキソ-7H-プリン化合物16を得ることができる。
この工程は、第15工程で得た化合物16にオルトギ酸エステル類を加え加熱して化合物12(表2、化合物番号II-11~II-13)を調製することができる。
ジフェニルエーテル(8mL)に1-アミノ-1,2,3,6-テトラヒドロ-6-イミノ-3,7-ジメチル-2-オキソ-7H-プリン化合物16(1.03mmol)と各種オルトエステル(3.09mmol)を加え、それぞれ200℃で20分間加熱撹拌する。反応後、室温まで冷却して析出した結晶を濾取し、ジエチルエーテルで洗浄し、これをDMFから再結晶して化合物12(表2、化合物番号II-11~II-13)を得ることができる。
(Scheme 6中、R9は水素原子、メチル基又はアリール基を示す。)
本明細書中において、DEADはアゾジカルボン酸ジエチルを示す。
さらに、第7工程で得たプリンアルデヒドヒドラゾン誘導体化合物10a-cに酸化剤のジエチルアゾジカルボン酸エステルを加え加熱すると、閉環転位して化合物17a-c(表2、化合物番号II-20~II-24、化合物番号II-27~II-31、及び化合物番号II-34~II-38)を得ることができる(第18工程)。以下、各工程毎に説明する。
この工程は、ヒドラジノプリン化合物9a-cにオルトエステル類を加え加熱して化合物17a-c(表2、化合物番号II-18~II-20,II-25~II-27及びII-32~II-34)を調製することができる。
即ち、酢酸(5~8mL)にヒドラジノプリン化合物9a-c(0.7~1.1mmol)と各種オルトエステル(2.2~3.3mmol)を加え、それぞれ80℃で10分間加熱撹拌する。反応後、室温まで冷却して析出した結晶を濾取する。これをDMFと水の混合溶媒から再結晶して化合物17a-cを得ることができる。
この工程は、プリンアルデヒドヒドラゾン化合物10a-cに適切な酸化剤を用いて酸化し、その閉環化合物である化合物17a-c(表2、化合物番号II-20~II-24,化合物番号II-27~II-31及び化合物番号II-34~II-38)を調製することができる。
(Scheme 7中、R9は水素原子、メチル基又はフェニル基を示す。)
本明細書中において、HAOSはヒドロキシルアミン-O-スルホン酸を示す。
この工程は、28%アンモニア水(50mL)にチオキソプリン化合物8b,c(8mmol)を加え、封管中160℃で48時間加熱する。反応後、氷中冷却し、析出した結晶を濾取し、水及びエタノールで洗浄して無色の粉末結晶の化合物18b,cを得ることができる。
この工程は、アミノ化合物18b,c(2mmol)を2N NaOH(15mL)に溶解させ、水3mLに溶解させたヒドロキシルアミン-O-スルホン酸(HAOS)(7.6~8.6mmol)を0~10℃で加え、反応後析出した結晶を濾取し、これを水に溶解させ10%aq.HClで中和すると結晶が析出する。これをエタノールから再結晶して無色の粉末結晶の1-アミノ-6-イミノ化合物である化合物19b,cを得ることができる。
この工程は、トリアゾロ[5,1-i]プリン類の構造確実な別途合成法として下記の3ルートよりトリアゾロ[5,1-i]プリン類化合物17b,c(表2、化合物番号II-25~II-27及び化合物番号II-32~II-34)を合成できる。
(Route ii):無水酢酸(4mL)に8-置換1-アミノ-1,2,3,6-テトラヒドロ-6-イミノ-2-オキソ-7H-プリン化合物19b,c(0.8mmol)を加え、3時間加熱還流する。反応後、室温まで冷却して析出した結晶を濾取する。これをDMFとエタノールの混合溶媒から再結晶して無色の粉末結晶の化合物17b,c(表2、化合物番号II-26及びII-33)を得ることができる。
(Route iii):DMF(15mL)に8-置換1-アミノ-1,2,3,6-テトラヒドロ-6-イミノ-2-オキソ-7H-プリン化合物19b,c(0.8mmol)とベンズアルデヒド(1.2mmol)を加え、30分間加熱還流した後、DEAD(1.25mmol)を加え、1時間加熱還流する。反応後、減圧下溶媒を濃縮し、水で処理し、析出した結晶を濾取する。これをDMFとエタノールの混合溶媒から再結晶して無色の粉末結晶の化合物17b,c(表2、化合物番号II-27及びII-34)を得ることができる。
前述のScheme 1に記載の反応式に従い、化合物3a(表1、化合物番号I-1~I-13、以下、記載の化合物番号は前記表1~3を参照する)及び化合物3b(化合物番号I-14~I-22)で表されるヒドラジノプリン化合物を合成した(Scheme 1中、R5は表1参照)。
6-n-オクチリデンヒドラジノ-7H-プリン-2(3H)-オン化合物3a(化合物番号I-8:R5=n-C7H15)の合成
TFA(10mL)に6-ヒドラジノ-7H-プリン-2(3H)-オン化合物2a(0.50g,3.0mmol)とオクタナール(0.42g,3.3mmol)を加え、室温で30分間撹拌した。反応後、減圧下溶媒を留去し、水で処理し、析出した結晶を濾取した。これを1%aq.KHCO3で洗浄し、得られた固体をエタノールから再結晶して無色の粉末結晶の化合物3a(化合物番号I-8)を得た(表4及び5)。
6-アリールメチリデンヒドラジノ-7H-プリン-2(3H)-オン類化合物3a(化合物番号I-9~I-13:R5=アリール基)の一般合成
TFA(10mL)に6-ヒドラジノ-7H-プリン-2(3H)-オン化合物2a(0.50g,3.0mmol)と各種アルデヒド(3.6mmol)を加え、それぞれ室温で30分間撹拌した。反応後、減圧下溶媒を留去し、ジエチルエーテルで処理し、析出した結晶を濾取した。これを1%aq.KHCO3で洗浄し、得られた固体をDMFと水の混合溶媒から再結晶して化合物3a(化合物番号I-9~I-13)を得た(表4及び5)。
6-(1-メチル-2-n-オクチリデンヒドラジノ)-7H-プリン-2(3H)-オン化合物3b(化合物番号I-19:R5=n-C7H15)の合成
TFA(10mL)に6-(1-メチルヒドラジノ)-7H-プリン-2(3H)-オン化合物2b(0.50g,2.78mmol)とオクタナール(0.39g,3.06mmol)を加え、室温で30分間撹拌した。反応後、減圧下溶媒を留去し、水で処理し、析出した結晶を濾取した。これを1%aq.KHCO3で洗浄し、得られた固体をエタノールから再結晶して無色の粉末結晶の化合物3b(化合物番号I-19)を得た(表4及び5)。
6-(2-アリールメチリデン-1-メチルヒドラジノ)-7H-プリン-2(3H)-オン類化合物3b(化合物番号I-20~I-22:R5=アリール基)の一般合成
TFA(10mL)に6-(1-メチルヒドラジノ)-7H-プリン-2(3H)-オン化合物2b(0.50g,2.78mmol)各種アルデヒド(3.34mmol)を加え、それぞれ室温で30分間撹拌した。反応後、減圧下溶媒を留去し、ジエチルエーテルで処理し、析出した結晶を濾取した。これを1%aq.KHCO3で洗浄し、得られた固体をDMFと水の混合溶媒から再結晶して化合物3b(化合物番号I-20~I-22)を得た(表4及び5)。
6-ヒドラジノ-3,7-ジメチル-7H-プリン-2(3H)-オン化合物5の合成
エタノール(4mL)に1,2,3,6-テトラヒドロ-3,7-ジメチル-2-オキソ-6-チオキソ-7H-プリン化合物4(1g,5.1mmol)とヒドラジン水和物(4mL,117mmol)を加え、30分間加熱還流した。反応後、析出した結晶を濾取し、これを水から再結晶して無色の針状結晶の化合物5 0.73g(収率74%)を得た。
6-アルキリデンヒドラジノ-3,7-ジメチル-7H-プリン-2(3H)-オン又は6-アリールメチリデンヒドラジノ-3,7-ジメチル-7H-プリン-2(3H)-オン類化合物6(化合物番号I-23~I-28)の一般合成
DMF(40mL)に6-ヒドラジノ-3,7-ジメチル-7H-プリン-2(3H)-オン化合物5(0.50g,2.57mmol)と各種アルデヒド(3.08mmol)を加え、それぞれ室温で0.5~2時間撹拌した。反応後、減圧下溶媒を留去し、酢酸エチルで処理し、析出した結晶を濾取した。これをエタノール又はDMFと水の混合溶媒から再結晶して化合物6(化合物番号I-23~I-28)を得た(表6及び7)。
8-(4-クロロフェニル)-1,2,3,6-テトラヒドロ-2-オキソ-6-チオキソ-7H-プリン化合物8c(R2=4-Cl-C6H4)の合成
ピリジン(200mL)に8-(4-クロロフェニル)-1,2,3,6-テトラヒドロ-2,6-ジオキソ-7H-プリン化合物7c(4g,15.2mmol)と五硫化リン(10g,45.6mmol)を加え、8時間加熱還流した。反応後、減圧下溶媒を留去し、熱水で処理し、析出した結晶を濾取した。これをDMFと水の混合溶媒中活性炭処理及び再結晶して黄色の粉末結晶の化合物8c(3.5g,82%,mp>330℃)を得た。
6-ヒドラジノ-8-メチル-7H-プリン-2(3H)-オン化合物9a(R2=メチル基)の合成
エタノール(3mL)に1,2,3,6-テトラヒドロ-8-メチル-2-オキソ-6-チオキソ-7H-プリン化合物8a(1g,5.49mmol)とヒドラジン水和物(3mL,85.6mmol)を加え、1時間加熱還流した。反応後、析出した結晶を濾取し、水及びエタノールで洗浄して無色の粉末結晶の化合物9a(0.66g,67%,mp285℃(分解))を得た。汎用溶媒に対する溶解性が悪いため、再結晶は不可能であり、純度の指標となる元素分析は不可能であったが、単体化合物であることをTLC及び1H-NMRスペクトルより確認した。
6-ヒドラジノ-8-フェニル-7H-プリン-2(3H)-オン化合物9b(R2=フェニル基)の合成
エタノール(4mL)に1,2,3,6-テトラヒドロ-2-オキソ-8-フェニル-6-チオキソ-7H-プリン化合物8b(1g,4.09mmol)とヒドラジン水和物(4mL,114mmol)を加え、1時間加熱還流した。反応後、析出した結晶を濾取し、水及びエタノールで洗浄して無色の粉末結晶の化合物9b(0.65g,66%,mp280℃(分解))を得た。汎用溶媒に対する溶解性が悪いため、再結晶は不可能であり、純度の指標となる元素分析は不可能であったが、単体化合物であることをTLC及び1H-NMRスペクトルより確認した。
8-(4-クロロフェニル)-6-ヒドラジノ-7H-プリン-2(3H)-オン化合物9c(R2=4-Cl-C6H4)の合成

6-アリールメチリデンヒドラジノ-8-メチル-7H-プリン-2(3H)-オン類化合物10a(R2=Me)(化合物番号I-29~I-33)の一般合成
TFA(8mL)に6-ヒドラジノ-8-メチル-7H-プリン-2(3H)-オン化合物9a(0.50g,2.77mmol)と各種アルデヒド(3.32mmol)を加え、それぞれ室温で30分間撹拌した。反応後、減圧下溶媒を留去し、ジエチルエーテルで処理し、析出した結晶を濾取した。これを1%aq.KHCO3で洗浄し、得られた固体をDMFと水の混合溶媒から再結晶して化合物10a(化合物番号I-29~I-33)を得た(表8及び9)。
6-アリールメチリデンヒドラジノ-8-フェニル-7H-プリン-2(3H)-オン類化合物10b(R2=フェニル基)(化合物番号I-34~I-38)の一般合成
TFA(8mL)に6-ヒドラジノ-8-フェニル-7H-プリン-2(3H)-オン化合物9b(0.50g,2.06mmol)と各種アルデヒド(2.47mmol)を加え、それぞれ室温で0.5~1時間撹拌した。反応後、減圧下溶媒を留去し、ジエチルエーテルで処理し、析出した結晶を濾取した。これを1%aq.KHCO3で洗浄し、得られた固体をDMFと水の混合溶媒から再結晶して化合物10b(化合物番号I-34~I-38)を得た(表10及び11)。
6-アリールメチリデンヒドラジノ-8-(4-クロロフェニル)-7H-プリン-2(3H)-オン類化合物10c(R2=4-Cl-C6H4)(化合物番号I-39~I-43)の一般合成
TFA(8mL)に8-(4-クロロフェニル)-6-ヒドラジノ-7H-プリン-2(3H)-オン化合物10c(0.50g,1.81mmol)と各種アルデヒド(2.17mmol)を加え、それぞれ室温で0.5~1時間撹拌した。反応後、減圧下溶媒を留去し、ジエチルエーテルで処理し、析出した結晶を濾取した。これを1%aq.KHCO3で洗浄し、得られた固体をDMFとエタノールの混合溶媒から再結晶して化合物10c(化合物番号I-39~I-43)を得た(表12及び13)。
前述のScheme 4に記載の反応式に従い、化合物12(化合物番号II-1~II-10)で表されるトリアゾロ[5,1-i]プリン化合物を合成した。(Scheme 4中、R9は表2参照)
1H-[1,2,4]トリアゾロ[5,1-i]プリン-5(4H)-オン類化合物12(化合物番号II-1~II-3)の一般合成
酢酸(8mL)に6-ヒドラジノ-7H-プリン-2(3H)-オン化合物2a(0.20g,1.2mmol)と各種オルトエステル(4.8mmol)を加え、それぞれ80℃で10~20分間加熱撹拌した。反応後、室温まで冷却して析出した結晶を濾取した。これをDMFと水の混合溶媒から再結晶して化合物12(化合物番号II-1~II-3)を得た(表14及び15)。
8-置換1H-[1,2,4]トリアゾロ[5,1-i]プリン-5(4H)-オン類化合物12(化合物番号II-3~II-10)の一般合成
(Route i):TFA(6mL)に対応する6-n-オクチリデンヒドラジノ-7H-プリン-2(3H)-オン又は6-アリールメチリデンヒドラジノ-7H-プリン-2(3H)-オン類化合物3a(1mmol)と70%硝酸(0.10mL,1.5mmol)を加え、それぞれ室温で10分間撹拌した。反応後、減圧下溶媒を留去し、ジエチルエーテルで処理し、析出した結晶を濾取した。これを1%aq.KHCO3で洗浄し、得られた固体をDMFと水の混合溶媒から再結晶して化合物12(化合物番号II-3~II-10)を得た(表14及び15)。
(Route ii):DMF(20mL)に対応する6-アリールメチリデンヒドラジノ-7H-プリン-2(3H)-オン類化合物3a(1mmol)とクロラニル(0.37g,1.5mmol)を加え、それぞれ10分間加熱還流した。反応後、減圧下溶媒を留去し、エタノールとジエチルエーテルで処理し、析出した結晶を濾取した。これをDMFと水の混合溶媒中活性炭処理及び再結晶して化合物12(化合物番号II-3~II-10)を得た(表14及び15)。
6,9-ジメチル-9H-1,2,4-トリアゾロ[3,4-i]プリン-5(6H)-オン類13(化合物番号III-1~III-3)の一般合成
DMF(20mL)に6-ヒドラジノ-3,7-ジメチル-7H-プリン-2(3H)-オン化合物5(0.20g,1.03mmol)と各種オルトエステル(2.06mmol)を加え、それぞれ1時間加熱還流した。反応後、減圧下溶媒を留去し、酢酸エチルで処理し、析出した結晶を濾取した。これをDMFと水の混合溶媒から再結晶して化合物13(化合物番号III-1~III-3)を得た(表16及び17)。
3-置換6,9-ジメチル-9H-1,2,4-トリアゾロ[3,4-i]プリン-5(6H)-オン類化合物13(化合物番号III-2~III-7)の一般合成
DMF(30mL)に6-アルキリデンヒドラジノ-3,7-ジメチル-7H-プリン-2(3H)-オン又は6-アリールメチリデンヒドラジノ-3,7-ジメチル-7H-プリン-2(3H)-オン類化合物6(1mmol)と70%硝酸(0.10mL,1.5mmol)を加え、それぞれ100℃で0.5~1時間撹拌した。反応後、減圧下溶媒を留去し、水で処理し、析出した結晶を濾取した。これをDMFと水の混合溶媒から再結晶して化合物13(化合物番号III-2~III-7)を得た(表16及び17)。
メチル(1-メチル-5-(4H-1,2,4-トリアゾール-3-イル)-1H-イミダゾール-4-イル)カルバミン酸メチル化合物14aの合成
0.1Nナトリウムメトキシドメタノール溶液(10mL)に6,9-ジメチル-9H-1,2,4-トリアゾロ[3,4-i]プリン-5(6H)-オン化合物13(化合物番号III-1)(0.20g,0.98mmol)を加え、室温で30分間撹拌した。反応後、浮遊物を濾去して減圧下溶媒を留去し、水を加えて10%aq.HClで中和した後、酢酸エチルで抽出し(20mL×4)、有機層をMg2SO4で乾燥させた。これを減圧下留去し、得られた結晶を酢酸エチルから再結晶して化合物14aを得た(表18及び19)。
メチル(1-メチル-5-(5-メチル-4H-1,2,4-トリアゾール-3-イル)-1H-イミダゾール-4-イル)カルバミン酸メチル化合物14bの合成
0.1Nナトリウムメトキシドメタノール溶液(10mL)に3,6,9-トリメチル-9H-1,2,4-トリアゾロ[3,4-i]プリン-5(6H)-オン類化合物13(化合物番号III-2)(0.20g,0.92mmol)を加え、室温で30分間撹拌した。反応後、浮遊物を濾去して減圧下溶媒を留去し、水を加えて10%aq.HClで中和した後、酢酸エチルで抽出し(20mL×4)、有機層をMg2SO4で乾燥させた。これを減圧下留去し、得られた結晶を酢酸エチルから再結晶して化合物14bを得た(表18及び19)。
メチル(1-メチル-5-(5-アリール-4H-1,2,4-トリアゾール-3-イル)-1H-イミダゾール-4-イル)カルバミン酸メチル類化合物14c-gの一般合成
0.1Nナトリウムメトキシドメタノール溶液(15mL)に3-アリール-6,9-ジメチル-9H-1,2,4-トリアゾロ[3,4-i]プリン-5(6H)-オン類化合物13(化合物番号III-3~III-7)(1mmol)を加え、それぞれ室温で0.5~1時間撹拌した。反応後、浮遊物を濾去して減圧下溶媒を留去し、水を加えて10%aq.HClで中和すると結晶が析出した。これを濾取し、エタノール又はDMFとエタノールの混合溶媒から再結晶して化合物14c-gを得た(表18及び19)。
1,4-ジメチル-1H-[1,2,4]トリアゾロ[5,1-i]プリン-5(4H)-オン類化合物12(化合物番号II-11~II-17)の一般合成
ジフェニルエーテル(8mL)に化合物14a-g(1mmol)を加え、それぞれ200℃で2時間加熱撹拌した。反応後、室温まで冷却して析出した結晶を濾取し、ジエチルエーテルで洗浄し、これをDMFから再結晶して化合物12(化合物番号II-11~II-17)を得た(表20及び21)。
1,4-ジメチル-1H-[1,2,4]トリアゾロ[5,1-i]プリン-5(4H)-オン類化合物12(化合物番号II-11~II-13)の一般合成
DMF(10mL)に対応する1H-[1,2,4]トリアゾロ[5,1-i]プリン-5(4H)-オン類化合物12(化合物番号II-1~II-3)(1mmol)とN,N-ジメチルホルムアミドジメチルアセタール(1.2g,10mmol)を加え、それぞれ2~3時間加熱還流した。反応後、減圧下溶媒を留去し、酢酸エチルで処理し、析出した結晶を濾取した。これをDMFから再結晶して化合物12(化合物番号II-11~II-13)を得た(表20及び21)。
1-アミノ-1,2,3,6-テトラヒドロ-6-イミノ-3,7-ジメチル-2-オキソ-7H-プリン化合物16の合成
DMF(100mL)に既知(Z.Kazimierczuk,et al,Acta Biochim.Pol.,21,455(1974))の6-アミノ-2,3-ジヒドロ-3,7-ジメチル-2-オキソ-7H-プリン化合物15(0.20g,1.12mmol)とO-(2,4-ジニトロフェニル)ヒドロキシルアミン(DNPA)(0.33g,1.68mmol)を加え、80℃で1時間加熱撹拌した。反応後、減圧下溶媒を留去し、残渣をシリカゲルカラムクロマトグラフィー(Kieselgel 70-230 mesh)(酢酸エチル:エタノール=4:1)によって分離精製し、得られた固体をエタノールから再結晶して無色の粉末結晶の化合物16(0.12g,55%,mp226℃(分解))を得た。
1,4-ジメチル-1H-[1,2,4]トリアゾロ[5,1-i]プリン-5(4H)-オン類化合物12(化合物番号II-11~II-13)の一般合成
ジフェニルエーテル(8mL)に1-アミノ-1,2,3,6-テトラヒドロ-6-イミノ-3,7-ジメチル-2-オキソ-7H-プリン化合物16(0.20g,1.03mmol)と各種オルトエステル(3.09mmol)を加え、それぞれ200℃で20分間加熱撹拌した。反応後、室温まで冷却して析出した結晶を濾取し、ジエチルエーテルで洗浄し、これをDMFから再結晶して化合物12(化合物番号II-11~II-13)を得た(表20及び21)。
2-メチル-1H-[1,2,4]トリアゾロ[5,1-i]プリン-5(4H)-オン類化合物17a(化合物番号II-18~II-20)の一般合成
酢酸(5mL)に6-ヒドラジノ-8-メチル-7H-プリン-2(3H)-オン化合物9a(0.20g,1.11mmol)と各種オルトエステル(3.33mmol)を加え、それぞれ80℃で10分間加熱撹拌した。反応後、室温まで冷却して析出した結晶を濾取した。これをDMFと水の混合溶媒から再結晶して化合物17a(化合物番号II-18~II-20)を得た(表22及び23)。
8-アリール-2-メチル-1H-[1,2,4]トリアゾロ[5,1-i]プリン-5(4H)-オン類化合物17a(化合物番号II-20~II-24)の一般合成
A:DMF(15mL)に6-アリールメチリデンヒドラジノ-8-メチル-7H-プリン-2(3H)-オン類化合物10a(化合物番号I-29~I-32)(1mmol)とDEAD(0.26g,1.5mmol)を加え、それぞれ1時間加熱還流した。反応後、減圧下溶媒を濃縮し、氷中冷却し、析出した結晶を濾取した。これをDMFと水の混合溶媒から再結晶して化合物17a(化合物番号II-20~II-23)を得た(表22及び23)。
B:DMF(20mL)に8-メチル-6-(4-ニトロベンジリデンヒドラジノ)-7H-プリン-2(3H)-オン化合物10a(化合物番号I-33)(0.30g,0.96mmol)とDEAD(0.17g,0.96mmol)を加え、3時間加熱還流した。但し、DEADは1時間おきに0.17gを2回に分けて追加した。反応後、減圧下溶媒を濃縮し、氷中冷却し、析出した結晶を濾取した。これをDMFと水の混合溶媒から再結晶して無色の粉末結晶の化合物17a(化合物番号II-24)を得た(表22及び23)。
2-フェニル-1H-[1,2,4]トリアゾロ[5,1-i]プリン-5(4H)-オン類化合物17b(化合物番号II-25~II-27)の一般合成
酢酸(8mL)に6-ヒドラジノ-8-フェニル-7H-プリン-2(3H)-オン化合物9b(0.20g,0.83mmol)と各種オルトエステル(2.49mmol)を加え、それぞれ80℃で10分間加熱撹拌した。反応後、室温まで冷却して析出した結晶を濾取した。これをDMFとエタノールの混合溶媒から再結晶して化合物17b(化合物番号II-25~II-27)を得た(表24及び25)。
8-アリール-2-フェニル-1H-[1,2,4]トリアゾロ[5,1-i]プリン-5(4H)-オン類化合物17b(化合物番号II-27~II-31)の一般合成
A:DMF(20mL)に対応する6-アリールメチリデンヒドラジノ-8-フェニル-7H-プリン-2(3H)-オン類化合物10b(化合物番号I-34~I-37)(1mmol)とDEAD(0.26g,1.5mmol)を加え、それぞれ1~1.5時間加熱還流した。反応後、減圧下溶媒を濃縮し、水で処理し、析出した結晶を濾取した。これをDMFとエタノールの混合溶媒から再結晶して化合物17b(化合物番号II-27~II-30)を得た(表24及び25)。
B:DMF(20mL)に6-(4-ニトロベンジリデンヒドラジノ)-8-フェニル-7H-プリン-2(3H)-オン化合物10b(化合物番号I-38)(0.30g,0.80mmol)とDEAD(0.14g,0.80mmol)を加え、2時間加熱還流した。但し、DEADは1時間後に0.14gを追加した。反応後、減圧下溶媒を濃縮し、水で処理し、析出した結晶を濾取した。これをDMFとエタノールの混合溶媒から再結晶して無色の粉末結晶の化合物17b(化合物番号II-31)を得た(表24及び25)。
2-(クロロフェニル)-1H-[1,2,4]トリアゾロ[5,1-i]プリン-5(4H)-オン類化合物17c(化合物番号II-32~II-34)の一般合成
酢酸(8mL)に8-(4-クロロフェニル)-6-ヒドラジノ-7H-プリン-2(3H)-オン化合物9c(0.20g,0.72mmol)と各種オルトエステル(2.16mmol)を加え、それぞれ80℃で10分間加熱撹拌した。反応後、室温まで冷却して析出した結晶を濾取した。これをDMFとエタノールの混合溶媒から再結晶して化合物17c(化合物番号II-32~II-34)を得た(表26及び27)。
8-アリール-2-(クロロフェニル)-1H-[1,2,4]トリアゾロ[5,1-i]プリン-5(4H)-オン類化合物17c(化合物番号II-34~II-38)の一般合成
A:DMF(20mL)に対応する6-アリールメチリデンヒドラジノ-8-(4-クロロフェニル)-7H-プリン-2(3H)-オン類化合物10c(化合物番号I-39~I-42)(1mmol)とDEAD(0.26g,1.5mmol)を加え、それぞれ1~1.5時間加熱還流した。反応後、減圧下溶媒を濃縮し、水で処理し、析出した結晶を濾取した。これをDMFとエタノールの混合溶媒から再結晶して化合物17c(化合物番号II-34~II-37)を得た(表26及び27)。
B:DMF(20mL)に8-(4-クロロフェニル)-6-(4-ニトロベンジリデンヒドラジノ)-7H-プリン-2(3H)-オン化合物10c(化合物番号I-43)(0.30g,0.73mmol)とDEAD(0.13g,0.73mmol)を加え、2時間加熱還流した。但し、DEADは1時間後に0.13gを追加した。反応後、減圧下溶媒を濃縮し、水で処理し、析出した結晶を濾取した。これをDMFとエタノールの混合溶媒から再結晶して無色の粉末結晶の化合物17c(化合物番号II-38)を得た(表26及び27)。
6-アミノ-2,3-ジヒドロ-2-オキソ-8-フェニル-7H-プリン化合物18bの合成
28%アンモニア水(50mL)に1,2,3,6-テトラヒドロ-2-オキソ-8-フェニル-6-チオキソ-7H-プリン化合物8b(2g,8.19mmol)を加え、封管中160℃で48時間加熱した。反応後、氷中冷却し、析出した結晶を濾取し、水及びエタノールで洗浄して無色の粉末結晶の化合物18b(1.4g,75%,mp>330℃)を得た。汎用溶媒に対する溶解性が悪いため、再結晶は不可能であり、純度の指標となる元素分析は不可能であったが、単体化合物であることをTLC及び1H-NMRスペクトルより確認した。
6-アミノ-8-(4-クロロフェニル)-2,3-ジヒドロ-2-オキソ-7H-プリン化合物18cの合成
28%アンモニア水(50mL)に8-(4-クロロフェニル)-1,2,3,6-テトラヒドロ-2-オキソ-6-チオキソ-7H-プリン化合物8c(2g,7.18mmol)を加え、封管中160℃で48時間加熱した。反応後、氷中冷却し、析出した結晶を濾取し、水及びエタノールで洗浄して無色の粉末結晶の化合物18c(1.44g,77%,mp>330℃)を得た。汎用溶媒に対する溶解性が悪いため、再結晶は不可能であり、純度の指標となる元素分析は不可能であったが、単体化合物であることをTLC及び1H-NMRスペクトルより確認した。
1-アミノ-1,2,3,6-テトラヒドロ-6-イミノ-2-オキソ-8-フェニル-7H-プリン化合物19bの合成
6-アミノ-2,3-ジヒドロ-2-オキソ-8-フェニル-7H-プリン化合物18b(0.50g,2.20mmol)を2N NaOH(15mL)に溶解させ、水3mLに溶解させたヒドロキシルアミン-O-スルホン酸(1g,8.80mmol)を0~10℃で滴下し、30分間撹拌した。反応後、析出した結晶を濾取し、これを水に溶解させ10%aq.HClで中和すると結晶が析出した。これを濾取し、エタノールから再結晶して無色の粉末結晶の化合物19b(0.34g,64%,mp270℃(分解))を得た。
1-アミノ-8-(4-クロロフェニル)-1,2,3,6-テトラヒドロ-6-イミノ-2-オキソ-7H-プリン化合物19cの合成
6-アミノ-8-(4-クロロフェニル)-2,3-ジヒドロ-2-オキソ-7H-プリン化合物18c(0.50g,1.91mmol)を2N NaOH(15mL)に溶解させ、水3mLに溶解させたヒドロキシルアミン-O-スルホン酸(0.86g,7.64mmol)を0~10℃で滴下し、30分間撹拌した。反応後、析出した結晶を濾取し、これを水に溶解させ10%aq.HClで中和すると結晶が析出した。これを濾取し、エタノールから再結晶して無色の粉末結晶の化合物19c(0.33g,62%,mp292℃(分解))を得た。
2-フェニル-1H-[1,2,4]トリアゾロ[5,1-i]プリン-5(4H)-オン類化合物17b(化合物番号II-25~II-27)の一般合成
(Route i):TFA(4mL)に1-アミノ-1,2,3,6-テトラヒドロ-6-イミノ-2-オキソ-8-フェニル-7H-プリン化合物19b(0.20g,0.83mmol)とオルトギ酸トリエチル(0.62g,4.15mmol)を加え、室温で3時間撹拌した。反応後、減圧下溶媒を留去し、ジエチルエーテルで処理し、析出した結晶を濾取した。これを0.5%aq.KHCO3で洗浄し、得られた固体をDMFとエタノールの混合溶媒から再結晶して無色の粉末結晶の化合物17b(化合物番号II-25)を得た(表24及び25)。
(Route ii):無水酢酸(4mL)に1-アミノ-1,2,3,6-テトラヒドロ-6-イミノ-2-オキソ-8-フェニル-7H-プリン化合物19b(0.20g,0.83mmol)を加え、3時間加熱還流した。反応後、室温まで冷却して析出した結晶を濾取した。これをDMFとエタノールの混合溶媒から再結晶して無色の粉末結晶の化合物17b(化合物番号II-26)を得た(表24及び25)。
(Route iii):DMF(15mL)に1-アミノ-1,2,3,6-テトラヒドロ-6-イミノ-2-オキソ-8-フェニル-7H-プリン化合物19b(0.20g,0.83mmol)とベンズアルデヒド(0.13g,1.25mmol)を加え、30分間加熱還流した後、DEAD(0.22g,1.25mmol)を加え、1時間加熱還流した。反応後、減圧下溶媒を濃縮し、水で処理し、析出した結晶を濾取した。これをDMFとエタノールの混合溶媒から再結晶して無色の粉末結晶の化合物17b(化合物番号II-27)を得た(表24及び25)。
2-(クロロフェニル)-1H-[1,2,4]トリアゾロ[5,1-i]プリン-5(4H)-オン類化合物17c(化合物番号II-32~II-34)の一般合成
(Route i):TFA(4mL)に1-アミノ-8-(4-クロロフェニル)-1,2,3,6-テトラヒドロ-6-イミノ-2-オキソ-7H-プリン化合物19c(0.20g,0.72mmol)とオルトギ酸トリエチル(0.53g,3.6mmol)を加え、室温で3時間撹拌した。反応後、減圧下溶媒を留去し、ジエチルエーテルで処理し、析出した結晶を濾取した。これを0.5%aq.KHCO3で洗浄し、得られた固体をDMFとエタノールの混合溶媒から再結晶して無色の粉末結晶の化合物17c(化合物番号II-32)を得た(表26及び27)。
(Route ii):無水酢酸(4mL)に1-アミノ-8-(4-クロロフェニル)-1,2,3,6-テトラヒドロ-6-イミノ-2-オキソ-7H-プリン化合物19c(0.20g,0.72mmol)を加え、3時間加熱還流した。反応後、室温まで冷却して析出した結晶を濾取した。これをDMFとエタノールの混合溶媒から再結晶して無色の粉末結晶の化合物17c(化合物番号II-33)を得た(表26及び27)。
(Route iii):DMF(15mL)に1-アミノ-8-(4-クロロフェニル)-1,2,3,6-テトラヒドロ-6-イミノ-2-オキソ-7H-プリン化合物19c(0.20g,0.72mmol)とベンズアルデヒド(0.11g,1.08mmol)を加え、30分間加熱還流した後、DEAD(0.19g,1.08mmol)を加え、1時間加熱還流した。反応後、減圧下溶媒を濃縮し、水で処理し、析出した結晶を濾取した。これをDMFとエタノールの混合溶媒から再結晶して無色の粉末結晶の化合物17c(化合物番号II-34)を得た(表26及び27)。
例1~2で合成した化合物の物理データ及び1H-NMRデータを以下表4~27に示す。
試験例1:キサンチンオキシダーゼ阻害作用
例1~2で合成した化合物についてキサンチンオキシダーゼ阻害作用を測定した。
50mMリン酸緩衝液(pH7.4)を用いて、キサンチン(100μM)、ウシミルク由来キサンチンオキシダーゼ(XOD)(10mU/mL)及び検体化合物を混合し、室温にて15分間インキュベートした後、キサンチンから尿酸への変化を292nmにおける吸光度(O.D.)変化として測定し、検体化合物の尿酸生成阻害作用を検討した。
尿酸生成阻害率を次式により求め、各検体化合物の用量反応曲線を作成し、50%阻害濃度(IC50,μM)を算出した。なお、対照化合物としてはアロプリノールを用いた。
尿酸生成阻害率(%)=100-[(D-DB)/T]×100
T : (キサンチン+ XOD)溶液のO.D.
D : (検体化合物+ キサンチン+ XOD)溶液のO.D.
DB: (検体化合物+ XOD)溶液のO.D.
例1~2で合成した化合物について、キサンチン酸化還元酵素へのドッキングスコア(結合自由エネルギー、kcal/mol)を計算した。
J Comput Aided Mol Des(2010)24:57-75に記載の計算方法に従って、例1~2で合成した化合物のキサンチン酸化還元酵素(XOR)(PDBコード 1N5X及び1V97)へのドッキングスコアを計算した。対照化合物として、アロプリノール、オキシプリノール、尿酸及びフェブキソスタットも同様に計算を行った。
本発明の化合物のキサンチンオキシダーゼ阻害用組成物、並びに高尿酸血症、痛風及び高尿酸血症に起因する疾患の予防又は治療のための製剤の調製例を以下に示す。
本発明化合物 30.0 mg
微粉末セルロース 25.0 mg
乳糖 39.5 mg
スターチ 40.0 mg
タルク 5.0 mg
ステアリン酸マグネシウム 0.5 mg
上記組成から常法によって錠剤を調整する。
本発明化合物 30.0 mg
乳糖 40.0 mg
スターチ 15.0 mg
タルク 5.0 mg
上記組成から常法によってカプセル剤を調整する。
本発明化合物 30.0 mg
グルコース 100.0 mg
上記組成を注射用精製水に溶解して注射剤を調整する。
Claims (16)
- R1、R2、R3及びR4が全て水素原子であり、R5がアルキル基又はアリール基である、請求項1に記載の化合物。
- R1、R2及びR3が全て水素原子であり、R4がメチル基であり、R5がアルキル基又はアリール基である、請求項1に記載の化合物。
- R1及びR3が両方ともメチル基であり、R2及びR4が両方とも水素原子であり、R5がアルキル基又はアリール基である、請求項1に記載の化合物。
- R1、R3及びR4が全て水素原子であり、R2及びR5が、それぞれ独立して、アルキル基又はアリール基である、請求項1に記載の化合物。
- R6、R7及びR8が全て水素原子であり、R9が水素原子、アルキル基又はアリール基である、請求項6に記載の化合物。
- R6及びR8が両方ともアルキル基であり、R7が水素原子であり、R9が水素原子、アルキル基又はアリール基である、請求項6に記載の化合物。
- R6及びR8が両方とも水素原子であり、R7がアルキル基又はアリール基であり、R9が水素原子、アルキル基又はアリール基である、請求項6に記載の化合物。
- R10及びR12が両方ともメチル基であり、R11が水素原子であり、R13が水素原子、メチル基又はアリール基である、請求項10に記載の化合物。
- 請求項1~5に記載のヒドラジノプリン化合物及び請求項6~11に記載のトリアゾロプリン化合物からなる群から選ばれる少なくとも1種の化合物を有効成分として含有する、キサンチンオキシダーゼ阻害用組成物。
- 請求項1~5に記載のヒドラジノプリン化合物及び請求項6~11に記載のトリアゾロプリン化合物からなる群から選ばれる少なくとも1種の化合物を有効成分として含有する、医薬用組成物。
- 前記医薬用組成物は、高尿酸血症、痛風、腎不全や慢性腎臓病の腎障害、高血圧、心血管疾患、脂質異常及びメタボリックシンドロームからなる群から選ばれる少なくとも1種の疾患を予防又は治療するための医薬用組成物である、請求項13に記載の医薬用組成物。
- 医薬用組成物を製造するための、請求項1~5に記載のヒドラジノプリン化合物及び請求項6~11に記載のトリアゾロプリン化合物からなる群から選ばれる少なくとも1種の化合物の使用。
- 被験体に、請求項1~5に記載のヒドラジノプリン化合物及び請求項6~11に記載のトリアゾロプリン化合物からなる群から選ばれる少なくとも1種の化合物の有効量を投与することを含む、高尿酸血症、痛風、腎不全や慢性腎臓病の腎障害、高血圧、心血管疾患、脂質異常及びメタボリックシンドロームの予防方法又は治療方法。
