本工程では、上記工程Aの反応液に、化合物(IV)を添加し、化合物(II)と化合物(IV)とを縮合して化合物(VI)を得る。上記工程Aの反応液へは、上記工程Bの反応液や、精製または粗精製した化合物(IV)を添加すればよいが、精製または粗生成した化合物(IV)を上記工程Aの反応液へ添加することが好ましい。即ち、少なくとも上記工程Aと本工程Cは同一系内(ワンポット)で行う。

(1)Boc-Gly-Pro-Ser(Bzl)-Ser(Bzl)-Gly-OHの製造
Boc-Gly-Pro-Ser(Bzl)-Ser(Bzl)-Gly-O(4-NBn)(0.82g,1.0mmol)に、Na2S2O4(0.86g,5.0mmol)、DMA(5mL)および水(0.5mL)を添加し、90℃で2時間撹拌した。反応溶液に酢酸エチル(8mL)と水(8mL)を添加して分液と抽出を行い、フラグメントF2としてBoc-Gly-Pro-Ser(Bzl)-Ser(Bzl)-Gly-OHを得た。
Boc-Ala-Pro-Pro-Pro-Ser(Bzl)-NH2(0.69g,1.1mmol)にジクロロメタン(5mL)とメタンスルホン酸(0.58g,6.0mmol)を添加し、室温で1.5時間撹拌した。反応溶液を氷冷し、トリエチルアミン(0.62g,6.1mmol)を添加し室温で10分撹拌した後、該反応液を上記実施例1(1)で得られたBoc-Gly-Pro-Ser(Bzl)-Ser(Bzl)-Gly-OHのジクロロメタン溶液(6mL)に添加した。続けてHOBt・H2O(0.23g,1.5mmol)を添加し、最後にEDC塩酸塩(0.29g,1.5mmol)を添加し、室温で16時間撹拌した。次いで反応を停止させ、抽出によりフラグメントF9としてBoc-Gly-Pro-Ser(Bzl)-Ser(Bzl)-Gly-Ala-Pro-Pro-Pro-Ser(Bzl)-NH2を得た(0.93mmol,純度95.0%)。
(1)Boc-Phe-Ile-Glu(OBzl)-Trp(CHO)-Leu-Lys(Cl-Z)-OHの製造
Boc-Phe-Ile-Glu(OBzl)-Trp(CHO)-Leu-Lys(Cl-Z)-O(4-NBn)(1.0g,0.94mmol)に、DMA(6g)、Na2S2O4(0.82g,4.7mmol)および水(0.6g)を添加し、90℃で2時間撹拌した。その後、20℃まで冷却し、5%硫酸水素カリウム水溶液(40g)を添加した。析出固体を濾取し、減圧乾燥することで、フラグメントF4として0.87gのBoc-Phe-Ile-Glu(OBzl)-Trp(CHO)-Leu-Lys(Cl-Z)-OHを得た。
Boc-Asn-Gly-O(4-NBn)(3.5g,8.3mmol)にジクロロメタン(160mL)とメタンスルホン酸(5.4g,56mmol)を添加し、3時間撹拌した。反応溶液を氷冷し、トリエチルアミン(5.8g,57mmol)を添加して5分間撹拌することにより、N末端を脱保護した。
H-Asn-Gly-O(4-NBn)溶液に、上記実施例2(1)で得られたBoc-Phe-Ile-Glu(OBzl)-Trp(CHO)-Leu-Lys(Cl-Z)-OH(8.5g,7.0mmol)とHOOBt(2.0g,13mmol)を添加し、最後にEDC塩酸塩(2.0g,10mmol)の水溶液(5.8mL)を3時間かけて添加した。20℃で4時間撹拌後、水(160g)を添加して析出物を濾取することで、フラグメントF10としてBoc-Phe-Ile-Glu(OBzl)-Trp(CHO)-Leu-Lys(Cl-Z)-Asn-Gly-O(4-NBn)を得た(9.7g,6.3mmol)。
(1)Boc-His(BOM)-Gly-Glu(OBzl)-Gly-OHの製造
Boc-His(BOM)-Gly-Glu(OBzl)-Gly-O(4-NBn)(5.0g,5.9mmol)に、DMA(15g)、Na2S2O4(5.2g,30mmol)および水(1.1g)を添加し、100℃で3時間撹拌した。10℃まで冷却後、水(30g)とジクロロメタン(30g)を添加して分液と抽出を行い、フラグメントF8としてBoc-His(BOM)-Gly-Glu(OBzl)-Gly-OHを得た。
Boc-Thr(Bzl)-Phe-Thr(Bzl)-Ser(Bzl)-Asp(OBzl)-Leu-O(4-NBn)(6.3g,4.9mmol)にジクロロメタン(11g)とメタンスルホン酸(2.4g,25mmol)を添加し、2時間撹拌した。反応溶液を氷冷し、トリエチルアミン(2.6g,25mmol)を添加して15分間撹拌した後、上記実施例3(1)で得られたBoc-His(BOM)-Gly-Glu(OBzl)-Gly-OH溶液(42g,5.9mmol)、HOBt・H2O(1.1g,7.4mmol)、EDC塩酸塩(1.4g,7.4mmol)をこの順で添加した。0℃で18時間撹拌後、5%炭酸ナトリウム水溶液(60g)を添加した。析出した固体を濾取することで、フラグメントF11としてBoc-His(BOM)-Gly-Glu(OBzl)-Gly-Thr(Bzl)-Phe-Thr(Bzl)-Ser(Bzl)-Asp(OBzl)-Leu-O(4-NBn)を取得した(8.6g,4.6mmol,純度93.5%)。
(1)Boc-Glu(OBzl)-Ala-Val-Arg(Z)2-Leu-OHの製造
Boc-Glu(OBzl)-Ala-Val-Arg(Z)2-Leu-O(4-NBn)(8.5g,7.2mmol)に、DMA(60mL)、Na2S2O4(6.2g,36mmol)および水(6mL)を添加し、90℃で4時間撹拌した。その後、水(60mL)を添加して室温まで冷却し、酢酸エチル(60mL)を添加して、室温で2時間撹拌した。析出した固体を濾取し、水と酢酸エチルで洗浄することにより、フラグメントF5としてBoc-Glu(OBzl)-Ala-Val-Arg(Z)2-Leu-OHを取得した(7.5g,7.2mmol,純度:96.5%)。
上記実施例2で得られたBoc-Phe-Ile-Glu(OBzl)-Trp(CHO)-Leu-Lys(Cl-Z)-Asn-Gly-O(4-NBn)(9.5g,6.2mmol)にジクロロメタン(105mL)とメタンスルホン酸(3.0g,31mmol)を添加し、1時間撹拌した。反応溶液を氷冷し、トリエチルアミン(2.8g,28mmol)を添加して5分間撹拌した。更にDMA(146g)を添加して20℃まで昇温した。
上記反応液に、上記実施例4(1)で得られたBoc-Glu(OBzl)-Ala-Val-Arg(Z)2-Leu-OH(6.5g,6.2mmol)およびHOOBt(1.5g,9.3mmol)を添加し、最後にEDC塩酸塩(1.8g,9.3mmol)のDMA溶液(311mL)を5時間かけて添加した。20℃で13時間撹拌後、30℃で17時間撹拌した。更に、HOOBt(0.25g,1.6mmol)、EDC塩酸塩(0.30g,1.6mmol)、Boc-Glu(OBzl)-Ala-Val-Arg(Z)2-Leu-OH(0.97g,0.94mmol)、およびトリエチルアミン(0.22g,2.2mmol)を添加し、15時間撹拌した。その後、水(570mL)を添加し、一部を40℃で減圧濃縮した後に、水(285mL)を再び添加した。0℃に冷却し、析出物を濾取することで、フラグメントF12としてBoc-Glu(OBzl)-Ala-Val-Arg(Z)2-Leu-Phe-Ile-Glu(OBzl)-Trp(CHO)-Leu-Lys(Cl-Z)-Asn-Gly-O(4-NBn)を得た(15.7g)。
(1)Boc-Glu(OBzl)-Ala-Val-Arg(Z)2-Leu-Phe-Ile-Glu(OBzl)-Trp(CHO)-Leu-Lys(Cl-Z)-Asn-Gly-OHの製造
上記実施例4で得られたBoc-Glu(OBzl)-Ala-Val-Arg(Z)2-Leu-Phe-Ile-Glu(OBzl)-Trp(CHO)-Leu-Lys(Cl-Z)-Asn-Gly-O(4-NBn)(15g,6.1mmol)にDMA(90g)を添加し、55℃まで加温した。続けて水(9.0g)とNa2S2O4(5.3g,30mmol)を添加し、90℃になるまで加熱した。90℃で2時間撹拌後、水(90g)を添加して析出物を濾取することにより、フラグメントF12としてBoc-Glu(OBzl)-Ala-Val-Arg(Z)2-Leu-Phe-Ile-Glu(OBzl)-Trp(CHO)-Leu-Lys(Cl-Z)-Asn-Gly-OHを得た(13.4g,5.8mmol)。
上記実施例1で得られたBoc-Gly-Pro-Ser(Bzl)-Ser(Bzl)-Gly-Ala-Pro-Pro-Pro-Ser(Bzl)-NH2(7.1g,5.8mmol)のジクロロメタン溶液(96g)にメタンスルホン酸(3.7g,39mmol)を添加し、3時間撹拌した。反応溶液を氷冷し、トリエチルアミン(4.2g,41mmol)を添加し、5分間撹拌した。更にDMA(106g)を添加した後、ジクロロメタンを減圧留去した。
次いで、上記実施例5(1)で得られたBoc-Glu(OBzl)-Ala-Val-Arg(Z)2-Leu-Phe-Ile-Glu(OBzl)-Trp(CHO)-Leu-Lys(Cl-Z)-Asn-Gly-OH(11.2g,4.8mmol)、DMA(106g)およびHOOBt(1.6g,9.7mmol)を添加し、最後にEDC塩酸塩(1.9g,9.7mmol)を添加し、2時間撹拌した。更にEDC塩酸塩(0.46g,2.4mmol)とトリエチルアミン(0.24g,2.4mmol)を1時間おきに3度添加し、1時間撹拌した後に、トリエチルアミン(0.49g,4.8mmol)を添加し、13時間撹拌した。水(106g)を添加して析出物を濾取することで、フラグメントF13としてBoc-Glu(OBzl)-Ala-Val-Arg(Z)2-Leu-Phe-Ile-Glu(OBzl)-Trp(CHO)-Leu-Lys(Cl-Z)-Asn-Gly-Gly-Pro-Ser(Bzl)-Ser(Bzl)-Gly-Ala-Pro-Pro-Pro-Ser(Bzl)-NH2を得た(13.3g,3.9mmol)。
(1)Boc-Ser(Bzl)-Lys(Cl-Z)-Gln-Met(O)-Glu(OBzl)-Glu(OBzl)-OHの製造
Boc-Ser(Bzl)-Lys(Cl-Z)-Gln-Met(O)-Glu(OBzl)-Glu(OBzl)-O(4-NBn)(1.0g、0.69mmol)に、DMA(6.0g)、水(0.6g)およびNa2S2O4(1.2g,6.9mmol)を添加し、90℃で4時間撹拌した。続けて水(60g)を添加して析出した固体を濾取することで、フラグメントF6としてBoc-Ser(Bzl)-Lys(Cl-Z)-Gln-Met(O)-Glu(OBzl)-Glu(OBzl)-OHを得た(0.89g,0.68mmol,純度94.6%)。
上記実施例5で得られたBoc-Glu(OBzl)-Ala-Val-Arg(Z)2-Leu-Phe-Ile-Glu(OBzl)-Trp(CHO)-Leu-Lys(Cl-Z)-Asn-Gly-Gly-Pro-Ser(Bzl)-Ser(Bzl)-Gly-Ala-Pro-Pro-Pro-Ser(Bzl)-NH2(13g,3.8mmol)にジクロロメタン(104g)を添加した。続けて、メタンスルホン酸(7.3g,76mmol)を添加し、2時間撹拌した。反応溶液を氷冷し、トリエチルアミン(8.3g,82mmol)を添加して5分間撹拌した。DMA(169g)を添加し、上記実施例6(1)で得られたBoc-Ser(Bzl)-Lys(Cl-Z)-Gln-Met(O)-Glu(OBzl)-Glu(OBzl)-OH(3.9g,3.0mmol)およびHOOBt(1.2g,7.6mmol)を添加した。続けて30分ごとにEDC塩酸塩(0.36g,1.9mmol)を4回添加し、1時間撹拌した後、Boc-Ser(Bzl)-Lys(Cl-Z)-Gln-Met(O)-Glu(OBzl)-Glu(OBzl)-OH(2.0g,1.5mmol)を添加し、2時間撹拌した。再びトリエチルアミン(0.19g,1.9mmol)およびEDC塩酸塩(0.36g,1.9mmol)を添加し、13時間撹拌した。水(273g)を添加して減圧濃縮した後、再び水(104g)を添加して析出した固体を濾取することで、フラグメントF14としてBoc-Ser(Bzl)-Lys(Cl-Z)-Gln-Met(O)-Glu(OBzl)-Glu(OBzl)-Glu(OBzl)-Ala-Val-Arg(Z)2-Leu-Phe-Ile-Glu(OBzl)-Trp(CHO)-Leu-Lys(Cl-Z)-Asn-Gly-Gly-Pro-Ser(Bzl)-Ser(Bzl)-Gly-Ala-Pro-Pro-Pro-Ser(Bzl)-NH2を得た(16.1g)。
(1)Boc-His(BOM)-Gly-Glu(OBzl)-Gly-Thr(Bzl)-Phe-Thr(Bzl)-Ser(Bzl)-Asp(OBzl)-Leu-OHの製造
上記実施例3で得られたBoc-His(BOM)-Gly-Glu(OBzl)-Gly-Thr(Bzl)-Phe-Thr(Bzl)-Ser(Bzl)-Asp(OBzl)-Leu-O(4-NBn)(11mmol)にDMSO(81g)を添加し、45℃に昇温した。Na2S2O4(5.6g,32mmol)を添加し、65℃で7時間撹拌後、再びNa2S2O4(1.9g,11mmol)を添加し、3時間撹拌した。水(0.58g)およびNa2S2O4(2.8g,16mmol)を添加し、1時間撹拌後、70℃で3時間撹拌した。20℃まで冷却後、水(302g)を添加して析出物を濾取することで、フラグメントF11としてBoc-His(BOM)-Gly-Glu(OBzl)-Gly-Thr(Bzl)-Phe-Thr(Bzl)-Ser(Bzl)-Asp(OBzl)-Leu-OHを得た(20g)。
上記実施例6で得られたBoc-Ser(Bzl)-Lys(Cl-Z)-Gln-Met(O)-Glu(OBzl)-Glu(OBzl)-Glu(OBzl)-Ala-Val-Arg(Z)2-Leu-Phe-Ile-Glu(OBzl)-Trp(CHO)-Leu-Lys(Cl-Z)-Asn-Gly-Gly-Pro-Ser(Bzl)-Ser(Bzl)-Gly-Ala-Pro-Pro-Pro-Ser(Bzl)-NH2(15g,3.2mmol)にジクロロメタン(177g)を添加した。続けて、メタンスルホン酸(9.3g,96mmol)を添加し、2時間撹拌した。反応溶液を氷冷し、トリエチルアミン(10g,103mmol)を添加し、5分間撹拌した。DMA(177g)を添加し、更に上記実施例7(1)で得られたBoc-His(BOM)-Gly-Glu(OBzl)-Gly-Thr(Bzl)-Phe-Thr(Bzl)-Ser(Bzl)-Asp(OBzl)-Leu-OH(6.1g,3.5mmol)およびHOOBt(1.1g,6.4mmol)を添加した。続けてEDC塩酸塩(1.2g,6.4mmol)を添加し、4時間撹拌した。更にBoc-His(BOM)-Gly-Glu(OBzl)-Gly-Thr(Bzl)-Phe-Thr(Bzl)-Ser(Bzl)-Asp(OBzl)-Leu-OH(0.56g,0.32mmol)を添加し、3時間撹拌し、続けて反応溶液を減圧濃縮した。残渣に水(354g)を添加し、10℃に冷却した後、析出した固体を濾取することにより、フラグメントF15としてBoc-His(BOM)-Gly-Glu(OBzl)-Gly-Thr(Bzl)-Phe-Thr(Bzl)-Ser(Bzl)-Asp(OBzl)-Leu-Ser(Bzl)-Lys(Cl-Z)-Gln-Met(O)-Glu(OBzl)-Glu(OBzl)-Glu(OBzl)-Ala-Val-Arg(Z)2-Leu-Phe-Ile-Glu(OBzl)-Trp(CHO)-Leu-Lys(Cl-Z)-Asn-Gly-Gly-Pro-Ser(Bzl)-Ser(Bzl)-Gly-Ala-Pro-Pro-Pro-Ser(Bzl)-NH2を得た(19.3g)。
上記実施例7で得られたBoc-His(BOM)-Gly-Glu(OBzl)-Gly-Thr(Bzl)-Phe-Thr(Bzl)-Ser(Bzl)-Asp(OBzl)-Leu-Ser(Bzl)-Lys(Cl-Z)-Gln-Met(O)-Glu(OBzl)-Glu(OBzl)-Glu(OBzl)-Ala-Val-Arg(Z)2-Leu-Phe-Ile-Glu(OBzl)-Trp(CHO)-Leu-Lys(Cl-Z)-Asn-Gly-Gly-Pro-Ser(Bzl)-Ser(Bzl)-Gly-Ala-Pro-Pro-Pro-Ser(Bzl)-NH2(1.0g,0.2mmol)にジクロロメタン(15g)を添加した。続けて、メタンスルホン酸(1.2g,13mmol)を添加し、1時間撹拌した。反応溶液を氷冷後、DMF(10g)を加えて減圧濃縮した。次に水素雰囲気下、Pd/Cを用いて水素添加反応を実施後、ピペリジンを加えてエキセナチドを取得した。

スルホン酸類を用いて下記式(I)で表される化合物のN末端を選択的に脱保護して下記式(II)で表される化合物を得た後、塩基性化合物を添加して反応液を中和する工程A、

F15: His-Gly-Glu-Gly-Thr-Phe-Thr-Ser-Asp-Leu-Ser-Lys-Gln-Met-Glu-Glu-Glu-Ala-Val-Arg-Leu-Phe-Ile-Glu-Trp-Leu-Lys-Asn-Gly-Gly-Pro-Ser-Ser-Gly-Ala-Pro-Pro-Pro-Ser
および、上記ペプチド断片F15のN末端および反応性側鎖官能基を脱保護する工程を含む請求項12に記載の方法。
