Abstract
Description
[2] 水溶性高分子が、その10重量%以下の水溶液の25℃における粘度が1000mPa・s以上の高分子である、上記[1]に記載の放出制御製剤;
[3] 水溶性高分子がヒプロメロース、ヒドロキシプロピルセルロースおよびポリビニルアルコールからなる群から選択される少なくとも一つである、上記[1]または[2]に記載の放出制御製剤;
[4] 製剤中に含まれる水溶性高分子の量が、製剤の総重量に対して5重量%~70重量%の範囲内である、上記[1]~[3]のいずれかに記載の放出制御製剤;
[5] 機能性デンプンが(a)または(b)の場合である、上記[1]~[4]のいずれかに記載の放出制御製剤:
(a) 7重量%水懸濁液の25℃における粘度が100mPa・s~1500mPa・sの範囲内であるデンプン類、または
(b) デンプン類を圧縮形成した場合に、第十七改正日本薬局方に基づく補助盤を使用する崩壊試験において1時間後でも崩壊しないものを形成しうるデンプン類;

[10] 液量が900mLのpH6.8の試験液、パドル回転数50rpmの溶出試験条件における、製剤中の化合物Aの40~60%が溶出する時間をt、その時の溶出率をRとした場合において、下記要件(x)~要件(z)のすべてを満たすことを特徴とする、上記[1]~[9]のいずれかに記載の放出制御製剤:
要件(x) 液量が900mLのpH6.8の試験液、パドル回転数200rpmの溶出試験条件において、時間tにおける溶出率(R(x))がR±15%の範囲内であること、
要件(y) 液量が900mLのpH5.0の試験液、パドル回転数50rpmの溶出試験条件において、時間tにおける溶出率(R(y))がR±15%の範囲内であること、および
要件(z) 液量が500mLのpH1.2の試験液、パドル回転数200rpmの溶出試験条件において、時間tで製剤が崩壊しないこと;

[13] 糖尿病性神経障害、糖尿病性壊疽、末梢循環障害、慢性動脈閉塞症、間欠性跛行、強皮症、血栓症、肺高血圧症、心筋梗塞、狭心症、糸球体腎炎、糖尿病性腎症、慢性腎不全、気管支喘息、間質性肺炎、肺線維症、慢性閉塞性肺疾患、尿細管間質性腎炎、炎症性腸疾患、または脊柱管狭窄症に伴う症状の治療のための、上記[1]~[12]のいずれかに記載の放出制御製剤;
[14] 液量が900mLのpH6.8の試験液、パドル回転数50rpmの溶出試験条件における、製剤中の2-{4-[N-(5,6-ジフェニルピラジン-2-イル)-N-イソプロピルアミノ]ブチルオキシ}-N-(メチルスルホニル)アセトアミドの40~60%が溶出する時間をt、その時の溶出率をRとした場合において、下記要件(x)~(z)のすべてを満たすことを特徴とする、2-{4-[N-(5,6-ジフェニルピラジン-2-イル)-N-イソプロピルアミノ]ブチルオキシ}-N-(メチルスルホニル)アセトアミドを有効成分として含有する、放出制御製剤:
要件(x) 液量が900mLのpH6.8の試験液、パドル回転数200rpmの溶出試験条件において、時間tにおける溶出率(R(x))がR±15%の範囲内であること、
要件(y) 液量が900mLのpH5.0の試験液、パドル回転数50rpmの溶出試験条件において、時間tにおける溶出率(R(y))がR±15%の範囲内であること、および
要件(z) 液量が500mLのpH1.2の試験液、パドル回転数200rpmの溶出試験条件において、時間tで製剤が崩壊しないこと。
[15] 医薬の製造における、2-{4-[N-(5,6-ジフェニルピラジン-2-イル)-N-イソプロピルアミノ]ブチルオキシ}-N-(メチルスルホニル)アセトアミド、水溶性高分子、機能性デンプンおよびアルカリ物質を含有し、そして放出制御組成物のpHが10以上である、放出制御組成物の使用;
[16] 2-{4-[N-(5,6-ジフェニルピラジン-2-イル)-N-イソプロピルアミノ]ブチルオキシ}-N-(メチルスルホニル)アセトアミド、水溶性高分子、機能性デンプンおよびアルカリ物質を含有し、組成物のpHが10以上であることを特徴とする放出制御組成物の治療有効量を、それを必要とする患者に投与することを含む、疾患の治療または予防方法。
本発明放出制御製剤の有効成分である化合物Aは、例えば、特許文献7、特許文献18に記載された公知化合物であり、当業者はこれら特許文献に記載の方法に従って製造することにより容易に入手することができる。
本発明放出制御製剤において、化合物Aは、化合物Aの光学異性体、化合物Aの薬学的に許容される塩、化合物Aの非晶形(アモルファス:WO2017029594、WO2017042731、WO2018015975等を参照)、化合物Aの結晶形、またはそれらの混合物であってもよく、特に化合物Aの結晶形が好ましい。
本発明放出制御製剤において、本発明の化合物Aの結晶形は、後述の3つの形態の化合物AのI型結晶、II型結晶、III型結晶の他にも、例えば、化合物AのIV型~IX型結晶が挙げられる(WO2017040872、WO2018022704、WO2018015974等を参照)。
本発明の化合物Aの結晶形としては、中でも、下記の化合物AのI型結晶、II型結晶、III型結晶が好ましく、化合物AのI型結晶が更に好ましい。
(1) 粉末X線回折図がCu Kα放射線(λ=1.54Å)を用いて得られるものであって、化合物Aの粉末X線回折スペクトルにおいて、次の回折角2θ:9.4度、9.8度、17.2度および19.4度で回折ピークを示す、化合物AのI型結晶。
(2) 粉末X線回折図がCu Kα放射線(λ=1.54Å)を用いて得られるものであって、化合物Aの粉末X線回折スペクトルにおいて、次の回折角2θ:9.0度、12.9度、20.7度および22.6度で回折ピークを示す、化合物AのII型結晶。
(3) 粉末X線回折図がCu Kα放射線(λ=1.54Å)を用いて得られるものであって、化合物Aの粉末X線回折スペクトルにおいて、次の回折角2θ:9.3度、9.7度、16.8度、20.6度および23.5度で回折ピークを示す、化合物AのIII型結晶。
本発明放出制御製剤は、有効成分として、化合物B、化合物Bの光学異性体、化合物Bの薬学的に許容される塩、化合物Bの非晶形(アモルファス)、化合物Bの結晶形、またはそれらの混合物を含有することができる。
本明細書中における「水溶性高分子」とは、水に溶解し高粘性となる高分子を意味する。
本発明放出制御製剤で用いられる水溶性高分子としては、例えば、水に水溶性高分子を溶解させ、10重量%以下の水溶液にしたときの粘度が、25℃で1000mPa・s以上である高分子を挙げることができる。特に、水溶性高分子の水溶液の濃度が、0.01~10重量%、好ましくは0.1~10重量%、更に好ましくは1.0~10重量%であり、水溶性高分子の水溶液の粘度が、25℃で1000mPa・s以上である高分子を挙げることができる。
また、本発明放出制御製剤で用いられる水溶性高分子としては、例えば、水に水溶性高分子を溶解させ、10重量%以下の水溶液にしたときの粘度が、25℃で1000mPa・s~2000mPa・sである高分子を挙げることができる。
本発明放出制御製剤で用いられる水溶性高分子は、10重量%以下の水溶性高分子の水溶液の粘度が、25℃で2000mPa・sを越える場合、水溶液の濃度を再調整して、水溶液の粘度が25℃で2000mPa・s以下になる高分子を含む。
本発明において、水溶性高分子の水溶液の粘度は、水溶性高分子を水に溶解させ、25℃で約24時間静置したものを試料とし、回転式粘度計(RHEOMETER R/S plus、Brookfield製)を用いて、測定温度25℃、回転数10rpmで行い、測定開始から300秒後に計測する。
本発明放出制御製剤で用いられるPVAとしては、例えば、ゴーセノール(登録商標) EG-48P(日本合成化学工業(株)製)およびパーテック SRP80(メルク社製)を挙げることができ、これらはいずれも市販品として入手可能である。
本明細書中における「機能性デンプン」の一例としては、例えば、水と接触することで粘性を増すデンプン類を挙げることができ、特にアルファー化デンプンが挙げられる。具体的な例としては、7重量%水懸濁液の25℃における粘度が100mPa・s~1500mPa・sの範囲内、好ましくは300mPa・s~1000mPa・sの範囲内であるデンプン類を挙げることができる。特に、7重量%水懸濁液の25℃における粘度が100mPa・s~1500mPa・sの範囲内、好ましくは300mPa・s~1000mPa・sの範囲内であるアルファー化デンプンが好ましい。
7重量%水懸濁液の粘度は、デンプン類を水によく撹拌して分散させた後、25℃で約24時間静置したものを試料とし、回転式粘度計(RHEOMETER R/S plus、Brookfield製)を用いて測定することができる。測定は、測定温度25℃、回転数10rpmで行い、測定開始から300秒後の粘度を計測することとする。
ここで、「圧縮成形」とは、例えば、オートグラフ(AG-50kNXD、島津製作所製)を用い、デンプン類190mgまたはデンプン類と糖類との混合物190mgを1000kgfで圧縮し直径8mmφの円柱状に成形することを意味する。
「糖類」としては、例えば、D-マンニトールまたは乳糖水和物を挙げることができる。
「保水量」は、乾燥した粉末のデンプン類[W0(g)]を純水に分散し、24時間振盪したものを遠心分離(3000G、10分)した後、直ちに分離した上層を切り捨て、下層に残る水を保持したデンプン類(デンプン類+デンプン類が保持する純水の量)[W(g)]の重量を計測し、下式により求めることができる。
保水量(%)=100×[W-W0]/W0
なお、遠心分離後に純水と粉末成分が分離されない場合は溶解とする。
本発明放出制御製剤で用いられる「アルカリ物質」としては、例えば、アルカリ物質を水に溶解させ、0.1重量%水溶液にしたときのpHが10以上となる物質を挙げることができる。中でも、0.1重量%水溶液にしたときのpHが10~13の範囲内となる物質が好ましく、10~12.5の範囲内となる物質がより好ましい。
本発明放出制御製剤で用いられる「アルカリ物質」としては、例えば、乾燥炭酸ナトリウム、炭酸カリウム、水酸化カルシウム、水酸化マグネシウム、酸化マグネシウムおよびメグルミンを挙げることができる。中でも、乾燥炭酸ナトリウムが好ましい。
本発明放出制御製剤で用いられる「アルカリ物質」は、例えば、本発明放出制御製剤のpHを10以上とすることができる。中でも、当該製剤のpHを10~13の範囲内とするものが好ましく、10~12の範囲内とするものがより好ましい。
ここで、「製剤のpH」とは、本発明放出制御製剤を粉末とし、それを3mLのメタノールと7mLの純水の混合液に分散させ、遠心後、上澄みのpHを測定した時の値を意味する。pH測定には、例えば、pH METER HM-30R(東亜ディーケーケー(株)製)を使用することができる。なお、製剤の総重量が100mg以下の場合は粉末100mgを量り取り測定する。
本発明放出制御製剤は、上記成分の他に、本発明の効果を阻害しない限り、薬学的に許容される添加剤を配合することができる。例えば、賦形剤、結合剤、崩壊剤、流動化剤、滑沢剤、可塑剤、着色剤、矯味剤または香料といった添加剤を必要に応じて、適当量配合することができる。これらの添加剤は、単独で用いても、2種以上組み合わせてもよい。
本発明放出制御製剤の添加剤の含有量は特に限定されないが、例えば、放出制御製剤総重量の15重量%~75重量%の範囲内が適当である。
本発明放出制御製剤で用いられるD-マンニトールとしては、例えば、マンニットC(三菱商事フードテック(株)製)、マンニットP(三菱商事フードテック(株)製)、マンニットS(三菱商事フードテック(株)製)、ペアリトール25C(ROQUETTE社製)、PEARLITOL(登録商標)50C(ROQUETTE社製)、PEARLITOL(登録商標)160C(ROQUETTE社製)、ノンパレル(登録商標)108(100)(フロイント産業株式会社)、ノンパレル(登録商標)108(200)(フロイント産業(株)製)を挙げることができ、市販品として入手可能である。中でもマンニットP、マンニットS、PEARLITOL(登録商標)50C、PEARLITOL(登録商標)160Cが好ましい。
本発明放出制御製剤において結合剤に用いられるHPMCとしては、例えば、TC-5R(信越化学工業(株)製)を挙げることができ、市販品として入手可能である。
本発明放出制御製剤において結合剤に用いられるPVAとしては、例えば、ゴーセノール(登録商標) EG-05P(日本合成化学工業(株)製)を挙げることができ、市販品として入手可能である。
本発明放出制御製剤の平面形状は特に限定されず、例えば、円形の他、楕円形、ドーナツ形などでもよい。
本発明放出制御製剤の錠厚は特に制限されないが、例えば、1mm~10mmの範囲内のものが適当であり、2mm~9mmの範囲内のものが好ましい。
本発明放出制御製剤の大きさは特に制限されないが、例えば、短径(円形の錠剤の場合は直径)1mm~20mmの範囲内のものが適当であり、例えば、2mm~14mmの範囲内のものが好ましい。
本発明放出制御製剤の重量は特に限定されないが、5mg~500mgの範囲内のものが適当であり、10mg~300mgの範囲内のものが好ましい。
本発明の一態様としては、液量が900mLのpH6.8の試験液、パドル回転数50rpmの溶出試験条件における、製剤中の化合物Aの約50%(40~60%)が溶出する時間をt、その時の溶出率をRとした場合において、例えば、下記要件(x)~要件(z)の条件を少なくとも要件(x)および要件(y)を満たし、好ましくは要件(x)~要件(z)を満たす放出制御製剤である。
要件(x) 液量が900mLのpH6.8の試験液、パドル回転数200rpmの溶出試験条件において、時間tにおける溶出率(R(x))がR±15%の範囲内であること、
要件(y) 液量が900mLのpH5.0の試験液、パドル回転数50rpmの溶出試験条件において、時間tにおける溶出率(R(y))がR±15%の範囲内であること、および
要件(z) 液量が500mLのpH1.2の試験液、パドル回転数200rpmの溶出試験条件において、時間tで製剤の崩壊が観察されないこと。
上記要件(x)~要件(z)の条件において、時間tは、例えば2時間~24時間の範囲内が挙げられ、好ましくは2時間~12時間である。
「pH6.8の試験液」は第17改正日本薬局方に記載の溶出試験第2液であり、「pH5.0の試験液」は薄めたMcIlvain緩衝液(0.05mol/Lのリン酸水素二ナトリウム水溶液と0.025mol/Lのクエン酸水溶液を用いて、pHを5.0に調製する。)であり(「医療用医薬品の品質に係る再評価の予試験について(医薬審第599号)」を参照)、「pH1.2の試験液」は第17改正日本薬局方に記載の溶出試験第1液である。
また、溶出試験(要件(Z))における「製剤の崩壊」とは、崩壊の様子を観察して、製剤がその形状を維持しない(溶解、消失、分離または明らかに形状を認めない軟体の物質となる)とき、崩壊したものとすることをいう。
本発明放出制御製剤は、上記した各種添加剤を用いて、製剤技術分野において慣用の方法によって製造することができる。
本発明放出制御製剤は、例えば、有効成分である化合物Aと各種添加剤を混合し直接圧縮成形するか、または、有効成分である化合物Aと各種添加剤とを顆粒とし、そのまま、もしくは他の添加剤と混合したものを圧縮成形物とすることで製造することができる。
前記圧縮成形の方法としては、特に制限はなく、公知の装置を適宜選択することができ、例えば、圧縮試験装置、油圧プレス、打錠機などの装置を用いる方法などを挙げることができる。圧縮試験装置としては、例えば、万能試験機(オートグラフ、株式会社島津製作所製)を挙げることができる。打錠機としては、例えば、ロータリー式打錠機(クーリーンプレスコレクト12、菊水製作所製)を挙げることができる。
圧縮成形時の圧力としては、所望の装置および圧縮成形物に適した条件を適宜選択すればよい。
また、上記の圧縮成形物をカプセルに充填することによって、カプセル剤を製造することができる。カプセルとしては公知のものを用いることができ、例えば、ゼラチンカプセルやHPMCカプセルを挙げることができる。カプセルの大きさは本発明放出制御製剤を充填できる大きさであれば特に制限されないが、00号から5号のカプセルを市販品として入手することができる。
本発明放出制御製剤は、必要に応じて、製剤技術分野において慣用の方法によってコーティングを行ってもよい。また、識別性のためのマーク、文字さらには分割用の割線を付してあってもよい。
ここで、コーティング基剤としては、例えば、糖衣基剤および水溶性フィルムコーティング基剤を挙げることができる。
糖衣基剤としては、ショ糖が用いられ、さらに、例えば、タルク、沈降炭酸カルシウム、ゼラチン、アラビアゴム、プルランおよびカルナバロウからなる群から選択される1種または2種以上を併用してもよい。
水溶性フィルムコーティング基剤としては、例えば、(i)ヒドロキシプロピルセルロース、ヒドロキシプロピルメチルセルロース、ヒドロキシエチルセルロース、メチルヒドロキシエチルセルロースなどのセルロース系高分子、(ii)ポリビニルアルコール、ポリビニルアセタールジエチルアミノアセテート、アミノアルキルメタアクリレートコポリマーE〔オイドラギット(登録商標)E〕、ポリビニルピロリドンなどの合成高分子および(iii)プルランなどの多糖類を挙げることができる。
上記したコーティング基剤は、その2種以上を適宜の割合で混合して用いてもよい。また、コーティングの際に、コーティング添加剤を用いてもよい。
該コーティング添加剤としては、例えば、(i)酸化チタン、タルク、三二酸化鉄などの遮光剤および/または着色剤および(ii)ポリエチレングリコール、クエン酸トリエチル、ヒマシ油、ポリソルベート類などの可塑剤を挙げることができる。
化合物Aは、優れたPGI2受容体作動作用を有し、血小板凝集抑制作用、血管拡張作用、気管支筋拡張作用、脂質沈着抑制作用、白血球活性化抑制作用等、種々の薬効を示す(例えば、特許文献7-12参照)。
また、本発明放出制御製剤は、遺伝子治療または自己骨髄細胞移植などの血管新生療法の促進剤、末梢血管再建術または血管新生療法における血管形成促進剤としても有用である。
本発明放出制御製剤は、特に、糖尿病性神経障害、糖尿病性壊疽、末梢循環障害、慢性動脈閉塞症、間欠性跛行、強皮症、血栓症、肺高血圧症、心筋梗塞、狭心症、糸球体腎炎、糖尿病性腎症、慢性腎不全、気管支喘息、間質性肺炎、肺線維症、慢性閉塞性肺疾患、尿細管間質性腎炎、炎症性腸疾患、または脊柱管狭窄症に伴う症状の予防剤または治療剤として有用である。
本発明放出制御製剤は、上記成分の他に、本発明の効果を阻害しない限り、医薬有効成分を配合することができ、例えば、上記の疾患の治療薬もしくは予防薬または治験薬を配合することができる。
本発明の放出制御剤は、本発明の効果を阻害しない限り、投与量は、例えば、症状、投与対象の年齢、性別等を考慮して個々の場合に応じて適宜決定されるが、通常経口投与の場合、例えば、成人1日あたり化合物Aとして0.05mg~5.0mgであり、これを1回であるいは2~4回に分けて投与することができ、好ましくは1日1回の投与である。
本発明放出制御製剤は、本発明の効果を阻害しない限り、その他上記の疾患の治療薬もしくは予防薬または治験薬と併用することができる。
本発明の一態様として、上記疾患の治療または予防における本放出制御製剤の使用、上記疾患の治療または予防のための医薬の製造における本放出制御製剤の使用、本放出制御製剤の有効量をそれを必要とする患者に投与することを含む上記疾患の治療または予防方法などが挙げられる。本放出制御製剤の使用について、上記の本放出制御製剤の記載を適用することができる。
本発明放出制御製剤は、放出制御組成物または放出制御処方の語と交換可能であり、本発明放出制御製剤に係る記載は、放出制御組成物または放出制御処方にも適用される。
実施例および比較例で使用した化合物Aは、前記化合物AのI型結晶である。
水溶性高分子の濃度と粘度(mPa・s)を、下記の表1に示す。
なお、水溶性高分子水溶液の粘度(mPa・s)は、以下の方法にしたがって測定した。
<測定方法>
水溶性高分子を水に溶解させ、25℃で約24時間静置したものを試料とし、回転式粘度計(RHEOMETER R/S plus、Brookfield製)を用いて測定した。測定は、測定温度25℃、回転数10rpmで行い、測定開始から300秒後の粘度を測定した。
水溶性高分子水溶液の粘度は、用いる水溶性高分子により大きく異なる。水溶性高分子の濃度が、水溶性高分子水溶液の総重量の10重量%以下の水溶液で、水溶性高分子水溶液の粘度が1000mPa・s以上となる場合には、溶液調製の操作性を考慮し、1000~2000mPa・sとなるように濃度を調節した。
デンプン類の7重量%懸濁液粘度(mPa・s)および保水量(%)を、下記の表2に示す。
なお、7重量%懸濁液粘度(mPa・s)および保水量(%)は、以下の方法にしたがって測定した。
<デンプン類の7重量%懸濁液粘度(mPa・s)の測定方法>
デンプン類を水に分散させ、7重量%水懸濁液を調製し、当該懸濁液をよく撹拌し、25℃で約24時間静置したものを試料とした。測定前に試料を緩やかに撹拌し、回転式粘度計(RHEOMETER R/S plus、Brookfield製)を用いて粘度を測定した。
なお、測定は、測定温度25℃、回転数10rpmで行い、測定開始から300秒後の値をデンプン類の粘度とした。
<デンプン類の保水量(%)の測定方法>
約80℃で5時間乾燥した粉末のデンプン類[W0(g)](約1g)を純水に分散し、24時間振盪したものを遠心分離(3000G、10分)した後、すぐに分離した上層を切り捨て、下層に残る水を保持したデンプン類(デンプン類+デンプン類が保持する純水の量)[W(g)]の重量を計測し、下式により求めた。
保水量(%)=100×[W-W0]/W0
なお、十の位を切り捨て、百単位で表示し、遠心分離後に純水と粉末成分が分離されない場合は溶解とした。
なお、圧縮成形したデンプン類および圧縮成形したデンプン類と糖類との混合物の1時間後の崩壊性は、以下の方法に従って測定した。
<測定方法>
190mgのデンプン類およびデンプン類と糖類とを重量比1対1で混合した物190mgを1000kgfで圧縮し、直径8mmφ(レンズ面、12R)の円柱状の圧縮成形物を得た。これら圧縮成形物を第十七改正日本薬局方 崩壊試験法(試験液:水)に従い、補助盤を使用して崩壊試験を実施し、試験開始1時間の崩壊性を確認した。
なお、糖類にはD-マンニトール(マンニットP(三菱商事フードテック(株)製))又は、乳糖水和物(Pharmatose(登録商標) 200M(DFE Pharma))を使用した。
アルカリ物質のpHを、下記表4に示す。
なお、アルカリ物質のpHは、以下の方法にしたがって測定した。
<測定方法>
アルカリ物質を0.1重量%となるように水に溶解(または分散)させた溶液(または分散液)のpHを、pH METER HM-30R(東亜ディーケーケー(株)製)を用いて測定した値をアルカリ物質のpHとした。
表5に記載の錠剤を製造し、それら錠剤を評価した。
次の表5に1錠あたりの錠剤の処方を示す。それぞれ処方に従い添加剤を秤量し、乳鉢を用いて混合し、混合末をオートグラフ(AG-50kNXD、島津製作所製)を用いて1000kgfで圧縮し錠径8mmφの錠剤を得た。
表5に記載の錠剤について、第17改正日本薬局方 溶出試験 パドル法に準じて、評価を行った。なお、試験には、シンカーを使用した。
各錠剤について、液量が900mLのpH6.8の試験液、パドル回転数50rpmの溶出試験条件における、錠剤中の化合物Aの約50%(40~60%)が溶出する時間(試験時間t)、その時の溶出率をRと定義し、以下の評価を行った。
要件(x)について
液量が900mLのpH6.8の試験液、パドル回転数200rpmの溶出試験条件において、試験時間tにおける溶出率R(x)がR±15%の範囲内である場合には「適」と判定し、R±15%の範囲外である場合には「不適」と判定した。
要件(y)について
液量が900mLのpH5.0の試験液、パドル回転数50rpmの溶出試験条件において、試験時間tにおける溶出率R(y)がR±15%の範囲内である場合には「適」と判定し、R±15%の範囲外である場合には「不適」と判定した。
要件(z)について
液量が500mLのpH1.2の試験液、パドル回転数200rpmの溶出試験条件において、試験時間tで錠剤が崩壊しない場合には「適」と判定し、錠剤が崩壊する場合には「不適」と判定した。
要件(x)~要件(z)のすべてにおいて「適」と判定された場合には、総合評価を「適」とし、それ以外の場合には「不適」とした。
比較例1-1、比較例1-2、比較例1-3および比較例1-4の評価結果を表6に示す。
表6に記載の通り、徐放性基剤として汎用される、HPMC、HPC、アルファー化デンプン、ポビドン/酢酸ビニル樹脂をそれぞれ単体で使用した錠剤は、要件(x)~要件(z)のすべてを満たすことはできなかった。
表7に記載の錠剤を製造し、それら錠剤を評価した。
表7に1錠あたりの錠剤の処方を示す。それぞれ処方に従い、試験例1に記載の方法と同様の方法を用いて、錠径8mmφの錠剤を得た。
表7に記載の錠剤について、試験例1と同様の方法で評価を行い、またそれら製剤のpHを以下に記載の方法にしたがって測定した。
<製剤のpHの測定方法>
1製剤を粉末とし、それを3mLのメタノールと7mLの純水の混合液に分散させ、遠心後、上澄みのpHを測定した。pH測定には、pH METER HM-30R(東亜ディーケーケー(株)製)を使用した。
比較例2-1、比較例2-2、実施例2-1および実施例2-2の評価結果を表8に示す。また、比較例2-1の溶出プロファイルを図4に、比較例2-2の溶出プロファイルを図5に、実施例2-1の溶出プロファイルを図6に、実施例2-2の溶出プロファイルを図7に示す。
表8に記載の通り、水溶性高分子(HPMCまたはPVA)および機能性デンプンを含み、アルカリ物質を含まない錠剤は、要件(x)~要件(z)のすべてを満たすことはできなかった。
一方、水溶性高分子(HPMCまたはPVA)、機能性デンプンおよびアルカリ物質を含む錠剤は、上記要件(x)~要件(z)のすべてを満たすことができた。
表9に記載の錠剤を製造し、それら錠剤を評価した。
表9に1錠あたりの錠剤の処方を示す。それぞれ処方に従い、試験例1に記載の方法と同様の方法を用いて、錠径8mmφの錠剤を得た。
表9に記載の錠剤について、試験例2と同様の方法で評価を行った。
比較例3-1および実施例3-1の評価結果を表10に示す。
表10に記載の通り、水溶性高分子、機能性デンプおよびアルカリ物質を含む錠剤のうち、機能性デンプンの添加量が5重量%である比較例3-1は、要件(x)~要件(z)のすべてを満たすことはできなかったが、機能性デンプンの添加量が15重量%である実施例3-1は、要件(x)~(z)のすべてを満たすことができた。
表11に記載の錠剤を製造し、それら錠剤を評価した。
表11に1錠あたりの錠剤の処方を示す。それぞれ処方に従い、試験例1に記載の方法と同様の方法を用いて、錠径8mmφの錠剤を得た。
表11に記載の錠剤について、試験例1と同様の方法で評価を行った。
比較例4-1、比較例4-2、比較例4-3および比較例4-4の評価結果を表12に示す。
表12に記載の通り、機能性デンプおよびアルカリ物質を含むが、水溶性高分子を含まない比較例4-1、水溶性高分子およびアルカリ物質を含むが、機能性デンプンを含まない比較例4-2、比較例4-3および比較例4-4は、要件(x)~要件(z)のすべてを満たすことができなかった。
表13に記載の錠剤を製造し、それら錠剤を評価した。
表13に1錠あたりの錠剤の処方を示す。それぞれ処方に従い、試験例1に記載の方法と同様の方法を用いて、錠径8mmφの錠剤を得た。
表13に記載の錠剤について、試験例2と同様の方法で評価を行った。
比較例5-1、比較例5-2および実施例5-1の評価結果を表14に示す。
表14に記載の通り、水溶性高分子、機能性デンプンに該当しないアルファー化デンプンおよびアルカリ物質を含む比較例5-1は、2時間後の溶出率が70%以上となり、徐放性を示すことができなかった。
また、機能性デンプおよびアルカリ物質を含み、水溶性高分子を含まない比較例5-2は要件(x)~要件(z)のすべてを満たすことができなかったが、それに水溶性高分子を加えた実施例5-1は要件(x)~要件(z)のすべてを満たすことができた。
表15に記載の錠剤を製造し、それら錠剤を評価した。
表15に1錠あたりの錠剤の処方を示す。それぞれ処方に従い、試験例1に記載の方法と同様の方法を用いて、錠径8mmφの錠剤を得た。
表15に記載の錠剤について、試験例2と同様の方法で評価を行った。
比較例6-1、比較例6-2、実施例6-1および実施例6-2の評価結果を表16に示す。
表16に記載のとおり、水溶性高分子および機能性デンプンを含み、アルカリ物質を含まない比較例6-1は、要件(x)~要件(z)のすべてを満たすことはできなかったが、それにアルカリ物質を含む実施例6-1は要件(x)~要件(z)のすべてを満たすことができた。
また、水溶性高分子、機能性デンプン以外の徐放性基剤(ポビドン/酢酸ビニル樹脂)およびアルカリ物質を含む比較例6-2は、要件(x)~要件(z)のすべてを満たすことはできなかったが、水溶性高分子、機能性デンプンおよびアルカリ物質を含む実施例6-2は、要件(x)~(z)のすべてを満たすことができた。
表17および表18に記載の錠剤を製造し、それら錠剤を評価した。
表17および表18に1錠あたりの錠剤の処方を示す。それぞれ処方に従い、試験例1に記載の方法と同様の方法を用いて、錠径8mmφの錠剤を得た。
表17および表18に記載の錠剤について、試験例2と同様の方法で評価を行った。
比較例7-1および比較例7-2の評価結果を表19に、また実施例7-1、実施例7-2および実施例7-3の評価結果を表20に示す。
表19及び表20に記載の通り、機能性デンプンおよびアルカリ物質を含み、水溶性高分子を含まない比較例7-1は要件(y)および要件(z)を満たすことができなかったが、水溶性高分子、機能性デンプンおよびアルカリ物質を含む実施例7-1(水溶性高分子の添加量:5重量%)および実施例7-2(水溶性高分子の添加量:10重量%)は要件(x)~要件(z)のすべてを満たすことができた。
また、水溶性高分子と機能性デンプンを含み、アルカリ物質を含まない比較例7-2は要件(y)を満たすことができなかったが、水溶性高分子、機能性デンプンおよびアルカリ物質を含む実施例7-3は要件(x)~要件(z)のすべてを満たすことができた。
表21および表22に記載の錠剤を製造し、それら錠剤を評価した。
表21および表22に1錠あたりの錠剤の処方を示す。それぞれ処方に従い、試験例1に記載の方法と同様の方法を用いて、錠径8mmφの錠剤を得た。
表21および表22に記載の錠剤について、試験例2と同様の方法で評価を行った。
比較例8-1、比較例8-2、実施例8-1および実施例8-2の評価結果を表23に、また実施例8-3、実施例8-4、実施例8-5および実施例8-6の評価結果を表24に示す。
表23および表24に記載の通り、アルカリ物質としてリン酸水素二ナトリウムまたは炭酸カルシウムを含む錠剤のpHは10.0より小さく、要件(y)を満たすことができなかった(表23の比較例8-1および8-2の評価結果を参照)が、アルカリ物質として水酸化カルシウム、水酸化マグネシウムまたは乾燥炭酸ナトリウムを含む錠剤のpHは10.0以上となり、要件(x)~要件(z)のすべてを満たすことができた(表23の実施例8-1および実施例8-2の評価結果、および表24の実施例8-3~実施例8-6の評価結果を参照)。
表25に記載の錠剤を製造し、それら錠剤を評価した。
実施例9-1
表25に1錠あたりの錠剤の処方を示す。それぞれ処方に従い化合物A、乳糖水和物
、結晶セルロース、HPMC、アルファー化デンプン、乾燥炭酸ナトリウムおよびステアリン酸マグネシウムを、乳鉢を用いて混合し、混合末をオートグラフ(AG-50kNXD、島津製作所製)を用いて1000kgfで圧縮し錠径8mmφの錠剤を得た。得られた錠剤を、錠剤コーティング機(DRC-200、パウレック(株)製)に入れ、コーティング液(乳糖水和物・HPMC・酸化チタン・マクロゴール混合物を水に分散し、固形分濃度20重量%としたもの)を噴霧してフィルムコート錠を得た。なお、錠剤コーティング機における仕込み量が適切な量となるように、適宜、模擬錠を使用した。
表26に1錠あたりの錠剤の処方を示す。それぞれ処方に従い化合物A、結晶セルロース、HPMC(メトローズ(登録商標)90SH-100000SR)、アルファー化デンプン、乾燥炭酸ナトリウムおよびステアリン酸マグネシウムを、乳鉢を用いて混合し、混合末をオートグラフ(AG-50kNXD、島津製作所製)を用いて800kgfで圧縮し錠径6.5mmφの錠剤を得た。得られた錠剤を、錠剤コーティング機(DRC-200、パウレック(株)製)に入れ、コーティング液(HPMC(TC-5R(登録商標))、プロピレングリコール、酸化チタンを水に分散し、固形分濃度10重量%としたもの)を噴霧してフィルムコート錠を得た。なお、錠剤コーティング機における仕込み量が適切な量となるように、適宜、模擬錠を使用した。
表25および表26に記載の錠剤について、試験例2と同様の方法で評価を行った。
実施例9-1および実施例9-2の評価結果を表27に示す。
水溶性高分子、機能性デンプンおよびアルカリ物質を含むフィルムコーティング錠は、要件(x)~要件(z)のすべてを満たすことができた。
表28に記載の錠剤を製造し、それら錠剤を評価した。
表28に1錠あたりの錠剤の処方を示す。それぞれ処方に従い添加剤を秤量し、乳鉢を用いて混合し、混合末をオートグラフ(AG-50kNXD、島津製作所製)で圧縮した。実施例10-1、実施例10-2および実施例10-4は1000kgfで圧縮し錠径8mmφの錠剤を得た。実施例10-3は1200kgfで圧縮し錠径9.5mmφの錠剤を得た。
表28に記載の錠剤について、試験例2と同様の方法で評価を行った。
実施例10-1、実施例10-2、実施例10-3および実施例10-4の評価結果を表29に示す。
水溶性高分子、機能性デンプンおよびアルカリ物質を含むすべての実施例において、要件(x)~要件(z)のすべてを満たすことができた。
したがって、本発明放出制御製剤は、有効成分の放出を長時間一定速度に保つことが必要である徐放性製剤として有用である。
Claims (16)
- 2-{4-[N-(5,6-ジフェニルピラジン-2-イル)-N-イソプロピルアミノ]ブチルオキシ}-N-(メチルスルホニル)アセトアミド、水溶性高分子、機能性デンプンおよびアルカリ物質を含有し、製剤のpHが10以上であることを特徴とする放出制御製剤。
- 水溶性高分子が、その10重量%以下の水溶液の25℃における粘度が1000mPa・s以上である、請求項1に記載の放出制御製剤。
- 水溶性高分子がヒプロメロース、ヒドロキシプロピルセルロースおよびポリビニルアルコールからなる群から選択される少なくとも一つである、請求項1または2に記載の放出制御製剤。
- 製剤中に含まれる水溶性高分子の量が、製剤の総重量に対して5重量%~70重量%の範囲内である、請求項1~3のいずれか一項に記載の放出制御製剤。
- 機能性デンプンが(a)または(b)の場合である、請求項1~4のいずれか一項に記載の放出制御製剤:
(a)7重量%水懸濁液の25℃における粘度が100mPa・s~1500mPa・sの範囲内であるデンプン類、または
(b)デンプン類を圧縮形成した場合に、第十七改正日本薬局方に基づく補助盤を使用する崩壊試験において1時間後でも崩壊しないものを形成しうるデンプン類。 - 製剤中に含まれる機能性デンプンの量が、製剤の総重量に対して15重量%~70重量%の範囲内である、請求項1~5のいずれか一項に記載の放出制御製剤。
- 製剤中に含まれる水溶性高分子と機能性デンプンとの合計量が、製剤の総重量に対して25重量%~85重量%の範囲内である、請求項1~6のいずれか一項に記載の放出制御製剤。
- アルカリ物質が、その0.1重量%水溶液のpHが10以上である、請求項1~7のいずれか一項に記載の放出制御製剤。
- 製剤中に含まれるアルカリ物質の量が、製剤の総重量に対して1重量%~15重量%の範囲内である、請求項1~8のいずれか一項に記載の放出制御製剤。
- 液量が900mLのpH6.8の試験液、パドル回転数50rpmの溶出試験条件における、製剤中の2-{4-[N-(5,6-ジフェニルピラジン-2-イル)-N-イソプロピルアミノ]ブチルオキシ}-N-(メチルスルホニル)アセトアミドの40~60%が溶出する時間をt、その時の溶出率をRとした場合において、下記要件(x)~(z)のすべてを満たすことを特徴とする、請求項1~9のいずれか一項に記載の放出制御製剤:
要件(x) 液量が900mLのpH6.8の試験液、パドル回転数200rpmの溶出試験条件において、時間tにおける溶出率(R(x))がR±15%の範囲内であること、
要件(y) 液量が900mLのpH5.0の試験液、パドル回転数50rpmの溶出試験条件において、時間tにおける溶出率(R(y))がR±15%の範囲内であること、および
要件(z) 液量が500mLのpH1.2の試験液、パドル回転数200rpmの溶出試験条件において、時間tで製剤が崩壊しないこと。 - 徐放性製剤である、請求項1~10のいずれか一項に記載の放出制御製剤。
- 錠剤またはカプセル剤である、請求項1~11のいずれか一項に記載の放出制御製剤。
- 糖尿病性神経障害、糖尿病性壊疽、末梢循環障害、慢性動脈閉塞症、間欠性跛行、強皮症、血栓症、肺高血圧症、心筋梗塞、狭心症、糸球体腎炎、糖尿病性腎症、慢性腎不全、気管支喘息、間質性肺炎、肺線維症、慢性閉塞性肺疾患、尿細管間質性腎炎、炎症性腸疾患、または脊柱管狭窄症に伴う症状の治療のための請求項1~12のいずれか一項に記載の放出制御製剤。
- 液量が900mLのpH6.8の試験液、パドル回転数50rpmの溶出試験条件における、製剤中の2-{4-[N-(5,6-ジフェニルピラジン-2-イル)-N-イソプロピルアミノ]ブチルオキシ}-N-(メチルスルホニル)アセトアミドの40~60%が溶出する時間をt、その時の溶出率をRとした場合において、下記要件(x)~(z)のすべてを満たすことを特徴とする、2-{4-[N-(5,6-ジフェニルピラジン-2-イル)-N-イソプロピルアミノ]ブチルオキシ}-N-(メチルスルホニル)アセトアミドを有効成分として含有する、放出制御製剤:
要件(x) 液量が900mLのpH6.8の試験液、パドル回転数200rpmの溶出試験条件において、時間tにおける溶出率(R(x))がR±15%の範囲内であること、
要件(y) 液量が900mLのpH5.0の試験液、パドル回転数50rpmの溶出試験条件において、時間tにおける溶出率(R(y))がR±15%の範囲内であること、および
要件(z) 液量が500mLのpH1.2の試験液、パドル回転数200rpmの溶出試験条件において、時間tで製剤が崩壊しないこと。 - 医薬の製造における、2-{4-[N-(5,6-ジフェニルピラジン-2-イル)-N-イソプロピルアミノ]ブチルオキシ}-N-(メチルスルホニル)アセトアミド、水溶性高分子、機能性デンプンおよびアルカリ物質を含有し、そして放出制御組成物のpHが10以上である、放出制御組成物の使用。
- 2-{4-[N-(5,6-ジフェニルピラジン-2-イル)-N-イソプロピルアミノ]ブチルオキシ}-N-(メチルスルホニル)アセトアミド、水溶性高分子、機能性デンプンおよびアルカリ物質を含有し、組成物のpHが10以上であることを特徴とする放出制御組成物の治療有効量を、それを必要とする患者に投与することを含む、疾患の治療または予防方法。
