Abstract
SAMの評価系の評価能力の向上を追求して自然発生がんの評価系に用いることができるかの確認と、この評価系を用いて、特許第6013670号公報でも用いられた植物発酵物に自然発がん予防効果があるかどうかを確認する。 次の(a)ないし(g) (a)大麦、黒大豆、赤米、黒米、小豆、はと麦、ヒエ、アワおよびキビよりなる群から選ばれる1種又は2種以上の豆・穀物類の麹菌発酵物 (b)ミカン、ブドウ、リンゴ、ヤマブドウ、モモ、カキ、パパイヤ、ナシ、スイカ、ウメ、イチジク、カリン、カボチャ、キンカン、ユズ、ビワ、アンズ、ナツメ、クリ、マタタビおよびスモモよりなる群から選ばれる1種又は2種以上の果実類の酵母および/又は乳酸菌発酵物 (c)紫イモ、菊芋、ニンジン、タマネギ、サツマイモ、里芋、自然薯、大根、赤カブ、ゴボウ、レンコン、ヤーコン、ユリ根、クワイ、しょうが、にんにくおよびウコンよりなる群から選ばれる1種又は2種以上の根菜類・イモ類の酵母および/又は乳酸菌発酵物 (d)キャベツ、紫蘇、桑葉、どくだみ、ヨモギ、クマザサおよびタンポポよりなる群から選ばれる1種又は2種以上の花・葉菜類の酵母および/又は乳酸菌発酵物 (e)コンブ、ワカメおよびモズクよりなる群から選ばれる1種又は2種以上の海藻類の酵母および/又は乳酸菌発酵物 (f)黒ゴマ、クルミおよびギンナンよりなる群から選ばれる1種又は2種以上の種子類の酵母および/又は乳酸菌発酵物 (g)マイタケおよびシイタケよりなる群から選ばれる1種又は2種のキノコ類の酵母および/又は乳酸菌発酵物 の混合物である植物発酵物を有効成分として含有することを特徴とする自然発がん予防剤ならびにSAMのP系マウスを、6週齢からSPF条件を維持しながら、60週齢以上まで飼育して得られることを特徴とする自然発がんマウスおよびこのマウスを利用した被験物質の自然発がんに対する効果の評価方法。
Description
本発明は、植物発酵物を利用した自然発がん予防剤に関する。
ヒトのがんの発生は高齢化や老化に伴って急激に増加する自然発生がんが大部分である。老化に伴って急激に増加する自然発生がんの原因として慢性炎症が認識されるようになった(非特許文献1~4)。
がんの究極の抑制は、発生を抑制することであるが、老化に伴った自然発生がんの発生抑制の評価に長期間を要する事が開発を妨げている。発生までに時間が掛かる自然発生がんの発生抑制試験をヒトで実施する事は時間的および倫理的にもほぼ不可能である。このため、寿命の短いげっ歯類を用いることは可能であるが、老化に伴う自然発生がんを評価する試験系は限定され、しかも2年に近い時間を要する。
このため、様々な動物モデル試験系が考案されて効果は上がってはいるものの、ヒトのがんの大部分を占める老化を伴うがん自然発生がんの発生抑制効果を評価するには不十分である。したがって、発がん予防の評価には老化を伴った評価系を用いた自然発がんの評価系が必須である(非特許文献5)。
ところで、我々はすでに老化促進マウス(Senescence Accelerated Mouse:SAM)を用いて植物発酵物が有意に寿命延長と老化の抑制をすることを証明し、老化抑制剤として特許を取得している(特許文献1)。この評価系で用いたSAMは、SAM学会(http://www.samrc.jp/)指導の下に販売供給され、その性質はSAMの発見者、開発者である竹田俊男により報告されている(非特許文献6)。この報告によると、SAMの寿命は9.7ヶ月(38.8週齢)であり、マウスで一般的に発がんが観察される60週齢に達していないため、自然発ガンの評価系に用いることはできなかった。

本発明者らは、このSAMの評価系の評価能力の向上を追求して自然発生がんの評価系に用いることができるかの確認と、この評価系を用いて、特許文献1でも用いられた植物発酵物に自然発がん予防効果があるかどうかを確認することを課題とした。
本発明者らは、上記課題を解決するため鋭意研究した。この研究過程において、複数のマウスの鼠径部や下腹部に腫瘤状の突起が出現し、高濃度の植物発酵物摂取群では出現が抑制される傾向が見られることが分かった。全てのマウスを解剖して腫瘤が観察された全部位を解剖観察したが、嚢胞状の脂肪塊が観察されたのみで、腫瘍は発見されないことが分かった。対照として飼育された正常型のSAM系マウスの4匹中1匹のみは長期(157週齢)まで飼育できることが分かった。剖検するとこのマウスのみ腸間膜リンパ節に白色の固形物が観察されることが分かった。その形状は本明細書に示したリンパ腫に酷似していたことが分かった。そして、SAM系マウスは飼育環境を調整することにより長期飼育が可能であることが分かった。それによりSAM系マウスを用いた系により自然発がんの評価系が構築可能であることを見出した。
また、本発明者らは、上記で見出したSAMの自然発生がんの評価系を用いて特許文献1でも用いられた植物発酵物に自然発生がんの予防効果があることを見出した。
すなわち、本発明は、次の(a)ないし(g)

本発明は、SAMのP系マウスを、6~14週齢からSPF条件を維持しながら、60週齢以上まで飼育して得られることを特徴とする自然発がんマウスである。

本発明は、被験物質を、上記自然発がんマウスに投与することを特徴とする被験物質の自然発がんに対する効果の評価方法である。
本発明は、SAMのP系マウスを、6~14週齢からSPF条件を維持しながら、60週齢以上まで飼育する最中に、被験物質を投与することを特徴とする被験物質の自然発がんに対する効果の評価方法である。
本発明により、特定の植物発酵物に自然発生がんの発生抑制効果があることが明瞭に示された。植物発酵物では初めての証明である。
本発明の自然発がん予防剤は、これまでも食経験の豊かな特定の植物発酵物を有効成分としているため飲食品、医薬品等としても好適である。
本発明の自然発がんマウスは、70週齢以上の長期飼育を可能であるため、ヒトでの大部分を占める老化に伴う自然発生がんに対する効果の評価が可能である。
また、このマウスは試験開始時期を14週齢まで遅らせても自然発がんが多発する60週齢までの試験期間を46週間にまで短縮可能である。そのため、遺伝子改変されたrasH2マウスの試験期間である36週齢(試験期間は30週)に迫る試験期間を達成できるため有用である。
本発明の自然発がん予防剤は、下記の植物発酵物を有効成分として含有するものである。この植物発酵物は、以下の(a)~(g)の植物発酵物の混合物である。
(a)大麦、黒大豆、赤米、黒米、小豆、はと麦、ヒエ、アワおよびキビよりなる群から選ばれる1種又は2種以上の豆・穀類に麹菌を作用させることにより得られる発酵物。
(b)ミカン、ブドウ、リンゴ、ヤマブドウ、モモ、カキ、パパイヤ、ナシ、スイカ、ウメ、イチジク、カリン、カボチャ、キンカン、ユズ、ビワ、アンズ、ナツメ、クリ、マタタビおよびスモモよりなる群から選ばれる1種又は2種以上の果実類に酵母および/又は乳酸菌を作用させることにより得られる発酵物。
(c)紫イモ、菊芋、ニンジン、タマネギ、サツマイモ、里芋、自然薯、大根、赤カブ、ゴボウ、レンコン、ヤーコン、ユリ根、クワイ、しょうが、にんにくおよびウコンよりなる群から選ばれる1種又は2種以上の根菜類・イモ類に酵母および/又は乳酸菌を作用させることにより得られる発酵物。
(d)キャベツ、紫蘇、桑葉、どくだみ、ヨモギ、クマザサおよびタンポポよりなる群から選ばれる1種又は2種以上の花・葉菜類に酵母および/又は乳酸菌を作用させることにより得られる発酵物。
(e)コンブ、ワカメおよびモズクよりなる群から選ばれる1種又は2種以上の海藻類に酵母および/又は乳酸菌を作用させることにより得られる発酵物。
(f)黒ゴマ、クルミおよびギンナンよりなる群から選ばれる1種又は2種以上の種子類に酵母および/又は乳酸菌を発酵させることにより得られる発酵物。
(g)マイタケおよびシイタケよりなる群から選ばれる1種又は2種のキノコ類に酵母および/又は乳酸菌を作用させることにより得られる発酵物。
本発明の自然発がん予防剤は、上記(a)~(g)の発酵物を混合した植物発酵物をそのまま有効成分としてもよいが、さらに多段階発酵させることにより、呈味性や製剤性を向上させることができる。
酵母としては、サッカロマイセス属、ジゴサッカロマイセス属等に属する酵母が挙げられ、中でもサッカロマイセス・セレビシエ(S. cervisiae)、ジゴサッカロマイセス・ロキシー(Z. rouxii)、サッカロマイセス・エキシグス(S.exiguus)等が好ましく用いられる。乳酸菌としては、例えば、ぺディオコッカス(Pediococcus)属、ロイコノストック(Leuconostoc)属、ラクトバチルス(Lactobacillus)属、ラクトコッカス(Lactococcus)属等に属する乳酸菌が挙げられ、中でも、ぺディオコッカス・アシディラクティ(P.acidilacti)、ラクトバチルス・ブレビス(L. brevis)、ロイコノストック・メセンテロイデス(L. mesenteroides)、ラクトバチルス・プランタラム(L. plantarum)、ラクトコッカス・ラクチス(L. lactis)、ラクトバチルス・サケイ(L. sakei)、ペディオコッカス・ペントサセウス(P. pentosaceus)、ラクトバチルス・カゼイ(L. casei)、ラクトバチルス・ロイテリ(L.reuteri)およびラクトバチルス・クルバタス(L. curvatus)等が好ましく用いられ、これらの1種または2種以上を使用することができる。麹菌としては、アスペルギルス・オリゼー(Aspergillus oryzae)、アスペルギルス・ニガー(Aspergillus niger)、アスペルギルス・カワチ(Aspergillus kawachii)等が例示でき、これらの1種又は2種以上を使用できる。これらは市販されているものも使用することができる。
発酵に供する豆・穀類、果実等の植物は、そのまま使用してもよく、あるいは必要に応じて粉砕、乾燥等の前処理を行ってもよい。また必要に応じて水を添加して希釈してもよい。
上記(a)~(g)の発酵物は、原料となる植物に、乳酸菌、酵母または麹菌を接種し、培養させることによって得られる。培養は、常法によって行えばよく、例えば、1種または2種以上の植物の混合物に対し、乳酸菌、酵母または麹菌を0.001~1質量%程度添加し、20~50℃で70~140時間程度発酵処理させればよい。
このようにして得られた(a)~(g)の発酵物を混合してそのまま有効成分とすることもできるが、必要に応じさらに20~40℃で200~300時間程度培養することが好ましい。さらにこの混合物を後発酵(熟成)させることが好ましい。後発酵は、必要に応じて、酢酸菌を作用させてもよく、例えば、上記(a)~(g)に含まれる植物のうち1種または2種以上に上記酵母を作用させたものに、アセトバクター・アセチ(A.aceti)等の酢酸菌を作用させた酢酸発酵物を添加すればよい。後発酵は、25~35℃で70時間~約1年程度行えばよい。後発酵による熟成過程を経ることより、呈味性や製剤性を改善することができ、また抗酸化活性の増大を図ることもできる。
上記植物発酵物を製造するための好適な方法の例として、多段階複合発酵方式が挙げられる。この方法は、果実類、野菜類、海藻類を主原料とする複合乳酸菌発酵物と、野菜類、根菜類、種子類、キノコ類、果実類を主要原料とする酵母発酵物を混合し、これに穀類、豆類を主原料とする麹菌発酵物を加え、さらに酢酸発酵物を加え混合した後、ろ過濃縮し、さらに約1年程度の後発酵を行うものである。このような多段階複合発酵によって、それぞれの発酵生産物がさらに別種の菌類によって資化・変換されることにより、風味が向上するとともに、抗酸化活性などの効果が高められると考えられる。
本発明に用いる有効成分の植物発酵物は、下記(1)から(4)の性質を有する。
(1)呈味性
果物、野菜等原料由来の甘味や有機酸に加え麹由来の甘味を有する。原料由来のポリフェノール類を含むものの苦味は少ない。
(2)水に対する溶解性
水に対して容易に溶解する。
(3)安定性
熱、酸に対して安定であり、ペーストは1年間室温保存しても腐敗や呈味性に変化はない。
(4)安全性
原料に用いた野菜、果物、ハーブ等は日常食しているものであり、また発酵に用いた酵母、乳酸菌、麹菌はいずれも食品の醸造や漬物等に由来する菌種を用いているため食経験が豊富である。
(1)呈味性

本発明の自然発がん予防剤は、かくして得られた植物発酵物に、公知の製剤学的製法に準じて、薬理学的に許容され得る担体、賦形剤、水分活性調整剤などを加え、製剤化することにより得られる。植物発酵物は、必要に応じ濃縮して濃度を調整したり、噴霧または凍結乾燥等により粉末化してもよい。製剤化において用いられる担体や賦形剤としては、たとえば乳糖、ブドウ糖、蔗糖、デンプン、糖類混合物などが用いられる。最終的形態としては、溶液、ペースト、ソフトカプセル、チュアブル、カプセルが用いられる。用量・用法としては、例えば、有効成分の植物発酵物として大人1日当たり、0.1g以上、好ましくは0.1~12g、より好ましくは0.6~10g(固形分換算)程度経口摂取すればよい。
また、本発明の自然発がん予防剤は医薬品、医薬部外品等の他、公知の食品素材とともに配合することにより飲食品の形態とすることもできる。上記植物発酵物はそのまま摂取することも可能であるが、日持ち性向上のために殺菌後にろ過、濃縮してもよく、あるいは必要に応じて賦形剤を添加して噴霧または凍結乾燥した粉末状としてもよい。さらに、流通過程におけるシェルフライフ向上のためには、濃縮して水分活性を低下させたものが望ましい。かかる飲食品の形態としては、ペースト、ソフトカプセル、錠剤、ドリンク剤等が例示できる。大人1日当たり植物発酵物として0.1g以上、好ましくは0.1~12g、より好ましくは0.6~10g(固形分換算)程度経口摂取することにより、優れた自然発がん予防効果等を得ることができる。このような上記植物発酵物を製剤化、食品の形態とした市販品として、天生酵素金印、天生酵素カプセル(日本自然発酵社製)等が挙げられる。
また、本発明の自然発がん予防剤を含有する飲食品は、上記のものの他に、この有効成分である植物発酵物を、調味料等の一種として用いて、みそ等の調味料、食パン等のパン類、せんべい、クッキー、チョコレート、飴、饅頭、ケーキ等の菓子類、ヨーグルト、チーズ等乳製品、たくわん等の漬物類、ラーメン、そば、うどん等の麺類、コーンポタージュ、わかめスープ等のスープ類、清涼飲料、健康飲料、炭酸飲料、果汁飲料等の飲食品に含有させたものであってもよい。
更に、本発明の自然発がん予防剤には、更にがんの治療薬および/またはがんの代替治療薬を含有させて、効果を増強させてもよい。がんの治療薬としては、特に限定されないが、例えば、代謝拮抗剤、アルキル化剤、抗がん性抗生物質、微小管阻害薬等の細胞障害性抗がん剤、分子標的治療薬、白金製剤、トポイソメラーゼ阻害剤等が挙げられる。がんの代替治療薬としては、特に限定されないが、例えば、十全大補湯、大柴胡湯、補中益気湯等の漢方製剤、フコイダン、メシマコブ、冬虫夏草、田七人参、鹿角霊芝、百花蛇舌草、発酵アガリクス等が挙げられる。これらがんの治療薬および/またはがんの代替治療薬を本発明の自然発がん予防剤に含有させる場合、含有量は目的とする効果にあわせて適宜設定すればよい。
この植物発酵物を有効成分とする自然発がん予防剤は、自然発がんの発生抑制作用だけでなく、前がん病変の発生抑制作用を有する。
本発明の自然発がんマウスは、SAMのP系マウスを、6~14週齢、好ましくは8~12週齢、より好ましくは10週齢からSPF条件を維持しながら、60週齢以上、好ましくは70週齢まで飼育することにより得られる。SAMのP系マウスとしては、例えば、SAMP1/SkuSlc(三協ラボサービス株式会社から購入可能)等が挙げられる。SPF条件を維持しながらとは、SPFを維持するため、環境浄化方法をさらに強化することである。通常、SPF条件と言いながらも、厳密にその条件が維持されていることはほとんどない。
ここでSPF条件とは、動物実験室の運用では常識的な条件である。例えば、飼育者はUV照射装置のある前室で殺菌した実権衣への着替えや室内履きの履き替え、手術用手袋の着用、帽子の着用をする。飼育設備は、例えば、動物飼育室と廊下とのバリヤー兼着替え用の前室(UVライトで殺菌する)を備え、更に、差圧計、出口フィルター、HEPAフィルター等のフィルター付きの空調設備等を備えた飼育設備で行う。前室と動物飼育室の内圧に差をつけて動物室が汚染されないような空調システムとする。前室と動物飼育室は、消毒用アルコールによる殺菌、粘着マットによるほこりの吸着の他に、床敷や餌は使用するまで前室で保管しておく。
環境浄化方法としては、例えば、空調の出口フィルターの交換頻度を増やす、プラズマクラスター等の殺菌機能付きの空気清浄機の導入、ヒビテン等の殺菌液による床の定期的な清拭等を行うこと等が挙げられる。特に空調の出口フィルターの交換は、前室にある差圧計が1Pa増加あるいは床敷の粉末によるフィルターの変色を目安にした。
上記のようにして飼育されたSAMのP系マウスは、60週齢を超えるとがんを自然発生(56週齢ではがんは確認されない)する。がんが発生したかどうかは、従来公知の方法により確認することができる。
本発明の自然発がんマウスを利用すれば、被験物質の自然発がんに対する効果の評価を行うことができる。なお、被験物質は特に限定されない。
具体的に、自然発がんマウスを利用した、被験物質の自然発がんに対する効果の評価方法としては、被験物質を、自然発がんマウスに投与する方法や、SAMのP系マウスを、上記条件で飼育する最中に、被験物質を投与する方法等が挙げられる。
以下、本発明を実施例を挙げて詳細に説明するが、本発明はこれら実施例に何ら限定されるものではない。
製 造 例 1
植物発酵物の製造:
以下の植物を原料として使用した。
(a)豆・穀類(大麦、黒大豆、赤米、黒米、小豆、はと麦、ヒエ、アワ、キビ)

上記(c)、(d)および(g)の原料(730g)に対し、乳酸菌類(P. acidilacti, L. brevis, L. mesenteroides, L. plantarum, L. lactis, L. sakei, L. casei)が約1.0×105cfu/gの菌濃度となるように調製した培養液を0.2質量%接種し、30℃で50時間培養を行った。また上記(b)、(e)および(f)の原料(900kg)に対し、酵母類(S. cervisiae 5種、Z. rouxii 2種)が約1.0×105cfu/gの菌濃度となるように調製した培養液を0.4質量%接種し30℃で50時間培養を行った。(a)豆類および穀類系原料(1000kg)には、麹菌類(黄麹菌、黒麹菌、白麹菌)を0.1質量%接種して35℃で70時間培養を行った。次いでそれぞれの培養物を混合後、30℃で200時間培養を行った。発酵を終了したモロミについて固液分離操作を行い、得られた濾液を濃縮してペースト状にしたものを容器に分注し、更に1年間後発酵(熟成)させ植物発酵物を得た。

発がん抑制試験:

SAMP1/SkuSlc(♂)マウス(以下、これを「SAMP1」という)を三協ラボサービス株式会社から購入して使用した。この試験では12週齢のマウスを用いた。飼育は、SPF仕様のマウス飼育施設(23±2℃、湿度50±10%、20:00~8:00の照明の明暗サイクル)を用い、マウスは各サンプル1群8~9匹を用い日本クレア製のポリカーボネートケージを用い単独飼育した。餌は三協ラボサービスから購入した500N(γ線滅菌済)を自由摂取させた。床敷きは日本クレア製の高温殺菌済みを用いた。ケージ交換は週1回行った。飼育は73週齢相当まで行った。
発がん抑制試験:
(1)マウス

(3)がんの評価

異常な腫瘤の有無な外観と触診、腹部の膨満などの腹水貯留傾向の外観、触診を行って記録した。腹部膨満は、1匹に観察された。サンプル群の1例の大腿部に約10mm角の固い腫瘤が71週目に発生し(老化度=5.5)、急激に膨大したが72週目には増大は緩やかになった。行動にやや支障はあるものの老化評価点数も高くなく、良好なQOLであった(老化度=6.0、皮膚外観と背骨の曲がりが主)。最終的の老化度評価は総合点数6.0であり、外観の老化(毛艶がやや低下する程度および腫瘤のために歩行不十分な程度)を示す程度であった。なお、老化度は症状別に点数化された学会での基準に基づいて評価した(“The Grading Score System: A Method for evaluating the degree of senescence in SAM strains of mice” M. Hosokawa et al, “The Senescence-accelerated Mouse (SAM) achievements and future directions” Elsevier ed. by Toshio Takeda, (2013)p.561-567)。

2)体腔内臓器
73週目に生存していた全てのマウスにソムノペンチル70μl/100gを腹腔投与してから、開腹し、体腔内臓器を観察した。頚部まで切開し、体腔内他の臓器を観察したところ、対照群(イオン交換水)の腸間膜部分に小指指頭程度の白色腫瘤が1個/匹見出された(図1)。この図において、毛細血管の浸襲を伴うものも見られた。これらの病理組織検査を株式会社バイオ病理研究所へ委託検査に供したところ、B細胞リンパ腫と判定された。マウスではリンパ腫とはヒトの悪性リンパ腫に相当する。

他の臓器には腫瘍は観察されず、腸間膜リンパ節が原発と推定された。1例は直径5mm程度のブドウ房状に増殖したものも見られ、これもB細胞リンパ腫であった。他に、解剖前日に死亡した個体では胸腺が解剖直前に急な頸部肥大が見られた例があり、重量は0.6354gであった。病理組織検査では異常なTリンパ球の増殖体であったが、死後サンプルのため悪性か否かの判定は出来なかった。死因は胸腺肥大による心臓と肺臓の極度の萎縮が見られ、圧迫死と判定された。2%群の1例で観察された右大腿部の固形腫瘤は切開したところ、強固な繊維化した間質に繭状に包嚢され、この包皮を切開して摘出したところ骨状の組織が見られた。病理組織検査で骨肉腫と判定された。がん組織においてがん微小環境において線溶系が亢進してがんの転移や播種の促進になるという報告も見られるが、一方、生体防御としてがん組織を強固に取り囲み、増殖を抑制することも知られている(「原発性後腹膜ならびに腸間膜腫瘍の臨床病理学的検討」松下昌裕ら、日臨外会誌47巻(8)59-66(1986))。このマウス個体の場合は他の部位に腫瘍組織の存在が認められず、また老化評価点も高くなく、QOLの低下も見られず、生体防御の反応例と思われた。表1に腫瘍発生個体数を体腔内と体腔外に分けて示した。表2では、腫瘍平均重量の解析を示した。表3には各腫瘍の特性の一覧を示した。表4には、異常増殖体の数を示した。
表1において、0%群の老化点数は2%群に比べて有意に高かった(t-検定のP=0.0099)。特に2%群は毛艶が優れていた。
表2に示したように、73週齢において生存していたマウスの腫瘍あるいは異常増殖物の発生頻度は、0%群の水対照群で高く、6匹中4匹であり、発生率は約66%であった。なお、外観から腹水貯留が著しかった個体については、剖検では強度の肝臓肥大が見られ、腹水貯留は無かった。肝臓の色調にも異常はなく、割面内部に腫瘍は観察されなかった。水投与群には、脾臓の肥大が見られる個体が多発したが、腫瘍の存在は見出されなかった。この打切り時点での平均生存率は0%群の385.85日に比べ2%群は456.25日と長かったが、カプランマイヤー解析では有意ではなかった。
サンプル濃度2%群では腫瘍発生は6匹中1匹であり、0%投与のマウスで頻発した体腔内リンパ腫は0%であった。このように経口投与された植物発酵物は自然腫瘍の発生を顕著に抑制した。少なくとも植物発酵物で自然発生がんの抑制を示したはじめての例である。サンプル群で1例発生した骨肉腫についても生体防御を示す間質由来と思われる強固な膜で周囲全体が覆われ、防御機能が働いていると思われる。このマウスは、見かけの老化度評価点数が低く(6.0)、腫瘍と共存しているように思われた。
表3からも明らかなように、2%植物発酵物投与群の腫瘍重量は0%群に比べ少なかった。2%植物発酵物は自然発がんの増殖抑制効果が認められた。
表4に示したように、0%植物発酵物群の異常増殖物は10個であるのに対し、2%植物発酵物群では1個であった。これにより2%植物発酵物の自然発生がんの発生抑制効果は明らかであった。
以上のことから、2%植物発酵物の自然発生がんの発生抑制効果があることが分かった。そのため、2%植物発酵物は自然発生がんの予防剤となることも分かった。また、SAMP1はがんを自然発生したため、自然発生がんのモデル動物となりうることも分かった。
試 験 例 2
自然発生がんの発生時期の短縮:
試験例1でSAMP1はがんを自然発生したものの、飼育期間が長かった。そのため、飼育条件などを調整して自然発生がんの発生時期の短縮を狙った。

基本的な飼育条件は試験例1と同様であるが、植物発酵物の飲水中濃度については、0%、0.4%、2%とした。なお、0.4%はヒト換算だと約2g/日である。この試験では0%群を9匹、0.4%を9匹、2%群を9匹用いた。また、対照として若齢マウス6匹を用いた(9週齢入荷、10週齢解剖)。マウスの飼育は、動物飼育室と廊下とのバリヤー兼着替え用の前室(UVライトで殺菌する)を備えた実験設備で行った。前室と動物飼育室の内圧に差をつけて動物室が汚染されないような空調システムとした。飼育条件は、SPF条件を維持するため、環境浄化方法をさらに強化し、空調の出口フィルターの交換頻度の増加(前室にある差圧計が1Pa増加あるいは床敷の粉末によるフィルターの変色で交換)、プラズマクラスター付きの空気清浄機の導入に加えて、ヒビテン500倍希釈液による床の清拭を追加した。飼育は72週齢相当まで行った。
表5に自然発生がん発生率の高まる60週齢以上生存数を各サンプル群ごとに示した。なお、早期死亡例の多くは、SAMP1(♂)個別飼育時に頻発する自虐行為による消耗死と思われた。
12週齢開始の場合は、担癌体%は明らかに2%の方が小さく、腫瘍総重量も低い。14週齢開始群では、担癌体%も濃度依存的であるが、腫瘍総重量では明らかな濃度依存性をしめした。発生した腫瘍は病理組織判定をバイオ病理研究所に委託した結果Bリンパ腫が殆どであり、1例のみ肝細胞腺腫と判定された。即ち、植物発酵物の老化を伴う自然発生がんの発生抑制効果は明らかである。
SAMP1(♂)の72週齢までの自然発生がんの性質と腫瘤の組織検査結果をみると、試験例1の結果と同様な傾向であり、腸間膜に発生するリンパ腫が主であるが、発生したがんには腸間膜部発生のリンパ腫だけでなく、肝がんも見られ、発がん発生抑制効果はリンパ腫に限定されない事を示唆している。
以上の結果より、SAMP1をSPF条件下を厳しく維持しながら飼育することにより自然発生がんの発生時期を短縮できることが分かった。
試 験 例 3
大腸の異常腺窩(ACF)の抑制試験:
異常腺窩巣(ACF)は結腸および直腸の内壁表面にみられることのある異常な管状の腺の集合体である。ACFは、大腸ポリープよりも早い時期に形成され、結腸内で最も早期に発見される前がん病変(がん化の可能性のある変化)のひとつである。SAMP1の長期飼育により自然発生するリンパ腫等の腫瘍を本植物発酵物が発生抑制を示したことから、大腸における前がん病変であるACFを本植物発酵物が発症予防を示すかを調べた。

SAMP1(♂)を1群9匹個別飼育をし、植物発酵物は0%と2%水溶液を試験例1と同様に調製し、これを自由摂取させた。開始時は19週齢を用い、これらを自然発がん発生が観察される73週齢まで長期飼育した。対照として、若齢マウスは9週齢のものを6匹購入し、10週齢を用いた。これらSAMP1(♂)を麻酔後、大腸を摘出し、10%ホルマリン液に1晩浸漬後、シャーレの中で3回イオン交換水で洗浄し、0.02%メチレンブルー染色液(Scy Tek社)に浸漬し、実体顕微鏡下で濃染されたACFをカウントした。麻酔は公知の三種混合液を用いた。悪性度の高い4細胞以上/ACFと4細胞以下/ACFを分けて計数した(染色方法は、「熟成ニンニク抽出液による大腸腫瘍の抑制効果に関する研究」直原 寛、広島大学博士論文(2015)を参考に、判定方法は、「大腸癌:診断と治療の進歩、I.疫学と病態;4.大腸癌の前癌状態」高山哲治、勝木伸一、新津洋司郎、日本内科学会誌、96巻24-29(平成19年2月10日)を参考にした)。これらの結果を表6に示した。
高週齢(73週齢)飼育で0%の場合は、悪性度の高い4細胞以上/Foci数は4.3個であったが、2%植物発酵物を摂取した高週齢マウスは、1.0個と1/4以下低下し、その値は自然発生がんが見られない10週齢の若齢マウスと同程度の値であった。本植物発酵物はリンパ腫などの自然発生がんの発生抑制だけでなく、前がん病変であるACFの段階でも発生抑制効果を示した。

製造例1で製造したペースト状の植物発酵物とみそを混合し、調味料で味を調整してみそ様食品を製造した。

ソフトカプセル:
製造例1で製造したペースト状の植物発酵物と、植物油、レシチンを混合したものを常法でソフトカプセルに充填してソフトカプセルを製造した。

製造例1で製造したペースト状の植物発酵物と、植物油、レシチンを混合したものを常法でソフトカプセルに充填してソフトカプセルを製造した。
実 施 例 4
チュアブル錠:
製造例1で製造したペースト状の植物発酵物と、結晶セルロース、白糖を混合したものを常法で打錠してチュアブル錠を製造した。

本発明によれば、自然発がんを予防することができる。また、本発明の自然発がんモデル動物は、被験物質の自然発がんに対する効果を評価することができる。
Claims (12)
- 次の(a)ないし(g)
(a)大麦、黒大豆、赤米、黒米、小豆、はと麦、ヒエ、アワおよびキビよりなる群から選ばれる1種又は2種以上の豆・穀物類の麹菌発酵物
(b)ミカン、ブドウ、リンゴ、ヤマブドウ、モモ、カキ、パパイヤ、ナシ、スイカ、ウメ、イチジク、カリン、カボチャ、キンカン、ユズ、ビワ、アンズ、ナツメ、クリ、マタタビおよびスモモよりなる群から選ばれる1種又は2種以上の果実類の酵母および/又は乳酸菌発酵物
(c)紫イモ、菊芋、ニンジン、タマネギ、サツマイモ、里芋、自然薯、大根、赤カブ、ゴボウ、レンコン、ヤーコン、ユリ根、クワイ、しょうが、にんにくおよびウコンよりなる群から選ばれる1種又は2種以上の根菜類・イモ類の酵母および/又は乳酸菌発酵物
(d)キャベツ、紫蘇、桑葉、どくだみ、ヨモギ、クマザサおよびタンポポよりなる群から選ばれる1種又は2種以上の花・葉菜類の酵母および/又は乳酸菌発酵物
(e)コンブ、ワカメおよびモズクよりなる群から選ばれる1種又は2種以上の海藻類の酵母および/又は乳酸菌発酵物
(f)黒ゴマ、クルミおよびギンナンよりなる群から選ばれる1種又は2種以上の種子類の酵母および/又は乳酸菌発酵物
(g)マイタケおよびシイタケよりなる群から選ばれる1種又は2種のキノコ類の酵母および/又は乳酸菌発酵物
の混合物である植物発酵物を有効成分として含有することを特徴とする自然発がん予防剤。 - 植物発酵物が、植物発酵物100g当たり以下の組成のアミノ酸を含有するものである請求項1に記載の自然発がん予防剤。
アルギニン 0.2~0.6g
リジン 0.1~0.7g
ヒスチジン 0.1~0.4g
フェニルアラニン 0.2~0.8g
チロシン 0.1~0.6g
ロイシン 0.3~1.2g
イソロイシン 0.2~0.8g
メチオニン 0.05~0.30g
バリン 0.2~0.9g
アラニン 0.2~0.9g
グリシン 0.2~0.7g
プロリン 0.4~1.2g
グルタミン酸 1.2~3.0g
セリン 0.2~0.8g
スレオニン 0.2~0.7g
アスパラギン酸 0.4~1.5g
トリプトファン 0.03~0.15g
シスチン 0.05~0.40g - 植物発酵物が、植物発酵物100g当たり以下の組成の有機酸を含有するものである請求項1又は2に記載の自然発がん予防剤。
クエン酸 0.5~1.2g
リンゴ酸 0.05~0.5g
コハク酸 0.04~0.3g
乳酸 0.5~6.0g
ギ酸 0.01~0.1g
ピルビン酸 0.005~0.05g
遊離γ-アミノ酪酸 0.01~0.05g - 乳酸菌が、ぺディオコッカス・アシディラクティ(P.acidilacti)、ラクトバチルス・ブレビス(L. brevis)、ロイコノストック・メセンテロイデス(L. mesenteroides)、ラクトバチルス・プランタラム(L. plantarum)、ラクトコッカス・ラクチス(L. lactis)、ラクトバチルス・サケイ(L. sakei)、ペディオコッカス・ペントサセウス(P. pentosaceus)、ラクトバチルス・カゼイ(L. casei)およびラクトバチルス・クルバタス(L. curvatus)よりなる群から選ばれた1種または2種以上である請求項1~3のいずれかの項記載の自然発がん予防剤。
- 酵母が、サッカロマイセス・セレビシエ(S. cervisiae)および/又はジゴサッカロマイセス・ロキシー(Z. rouxii)である請求項1~4のいずれかの項記載の自然発がん予防剤。
- 更に、がんの治療薬および/またはがんの代替治療薬を含有する請求項1~5のいずれかの項記載の自然発がん予防剤。
- 請求項1~6のいずれかの項記載の自然発がん予防剤を含有する飲食品。
- 医薬品である請求項1~6のいずれかの項記載の自然発がん予防剤。
- SAMのP系マウスを、6~14週齢からSPF条件を維持しながら、60週齢以上まで飼育して得られることを特徴とする自然発がんマウス。
- SAMのP系マウスを、6~14週齢からSPF条件を維持しながら、60週齢以上まで飼育することを特徴とする自然発がんマウスの製造方法。
- 被験物質を、請求項9に記載の自然発がんマウスに投与することを特徴とする被験物質の自然発がんに対する効果の評価方法。
- SAMのP系マウスを、6~14週齢からSPF条件を維持しながら、60週齢以上まで飼育する最中に、被験物質を投与することを特徴とする被験物質の自然発がんに対する効果の評価方法。
