化合物(6)は、適切な不活性溶媒中で通常用いられるニトロ基の還元条件により化合物(5)を還元して製造することができる。本反応の条件は、例えばパラジウム-炭素等による水素添加条件下での接触還元や、亜鉛、鉄等による金属還元、水素化アルミニウムリチウム等によるヒドリド還元等が挙げられる。溶媒はそれぞれの還元条件で通常用いられる溶媒が挙げられ、接触還元の場合は、メタノール、エタノール、THF、酢酸エチル等、金属還元の場合はTHF、酢酸、メタノール、エタノール等、ヒドリド還元の場合はジエチルエーテル、THF等が挙げられる。反応時間は通常、約10分から24時間であり、反応温度としては0℃から溶媒の沸点までである。

化合物(7)は、化合物(16)を用いて、以下の2つの反応条件により製造することができるが、これらの方法に限定されない。
1)塩基を使用する条件として、化合物(7)を化合物(16)およびRa-X5(Raは、前記と同じであり、X5は、ハロゲン、トリフルオロメタンスルホニルオキシ、メタンスルホニルオキシ等の脱離基である。)を用い工程(2-1)と同様の方法で製造する工程が挙げられる。

化合物(22)は、化合物(24)と、Rb基を持つハライド化合物またはトリフラート化合物(Rb-X(X:ハロゲン原子)またはCF3SO2O-Rb等)を、触媒および塩基の存在下で反応に付すことにより製造することができる。具体的には鈴木カップリング反応を用いる工程である。塩基としては炭酸ナトリウム、炭酸カリウム、炭酸セシウム、リン酸三カリウム等が挙げられる。触媒としては酢酸パラジウム、テトラキス(トリフェニルホスフィン)パラジウム、トリス(ジベンジリデンアセトン)ジパラジウム等が挙げられる。溶媒としては1,4-ジオキサン、トルエン、1,2-ジメトキシエタン等が挙げられる。反応時間は約30分から48時間であり、反応温度は室温から溶媒の沸点までである。
また、光学異性体は前記製造法の適切な工程で、光学活性カラムを用いた方法、分別結晶化法等の公知の分離工程を実施することで分離することができる。また、出発原料として光学活性体を使用することもできる。
本発明の化合物は、光学異性体、立体異性体、ケトエノール体のような互変異性体、および/または幾何異性体を有する場合もあるが、本発明の化合物は、これらを含め全ての可能な異性体およびそれらの混合物を包含する。

電位依存性Naチャネル遺伝子発現細胞を用いたNaイオン電流の測定
Nav1.7由来電流は、ヒトSCN9A安定発現細胞を用いて、マニュアルパッチクランプ法により測定した。
テトラサイクリン誘導型SCN9A安定発現細胞はChanTest社より入手した。細胞は、10%ウシ胎児血清、100units/mL Penicillin-Streptomycin、10μg/mL Blasticidin、400μg/mL Zeocinを含むHam’s F-12培地で継代維持した。スライドグラスに播種した細胞を1μg/mL テトラサイクリン、1μg/mL Sodium butyrate、10%ウシ胎児血清、100units/mL Penicillin-Streptomycinを含むHam’s F-12培地で培養し、翌日マニュアルパッチクランプ法にてNaイオン電流を測定した。
Naイオン電流は、マニュアルパッチクランプ法により、以下の細胞外液、細胞内液を用いて測定した。

6-(4-クロロフェノキシ)-1-[(3-ヒドロキシオキセタン-3-イル)メチル]-1,3-ジヒドロ-2H-ベンズイミダゾール-2-オン

1-[(3-ヒドロキシオキセタン-3-イル)メチル]-3-メチル-5-[4-(トリフルオロメトキシ)フェノキシ]-1,3-ジヒドロ-2H-ベンズイミダゾール-2-オン。 - 請求項1~13のいずれか一項に記載の化合物またはその製薬学的に許容される塩を含有する医薬組成物。
- 請求項1~13のいずれか一項に記載の化合物またはその製薬学的に許容される塩を有効成分として含有する、Nav1.7(SCN9A)が関与する疾患の治療薬。
- 請求項1~13のいずれか一項に記載の化合物またはその製薬学的に許容される塩を有効成分として含有する、神経障害性疼痛、侵害受容性疼痛、炎症性疼痛、小径線維ニューロパチー、肢端紅痛症、発作性激痛症、排尿障害または多発性硬化症の治療薬。
- 請求項1~13のいずれか一項に記載の化合物またはその製薬学的に許容される塩と、抗てんかん薬、抗うつ薬、麻薬性鎮痛薬、抗炎症薬、還元酵素阻害剤およびプロスタグランジン誘導体製剤から選択される少なくとも1種以上の薬剤とを組み合わせてなる医薬。
- 神経障害性疼痛、侵害受容性疼痛、炎症性疼痛、小径線維ニューロパチー、肢端紅痛症、発作性激痛症、排尿障害または多発性硬化症の治療剤を製造するための、請求項1~13のいずれか一項に記載の化合物またはその製薬学的に許容される塩の使用。
- 請求項1~13のいずれか一項に記載の化合物またはその製薬学的に許容される塩の有効量を哺乳動物に投与することを含む、当該哺乳動物における神経障害性疼痛、侵害受容性疼痛、炎症性疼痛、小径線維ニューロパチー、肢端紅痛症、発作性激痛症、排尿障害または多発性硬化症の治療方法。
