Abstract
Description
特許文献2には、式(B)の化合物がカテプシンS阻害作用を示し、種々の代謝性疾患或いはSLE等の免疫疾患の治療に有用であることが記載されている。
非特許文献1には、式(C)の化合物がSLEやループス腎炎の進行を妨げることが記載されている。
特許文献4には、式(E)の化合物がカテプシンS阻害作用を示し、糖尿病等の治療に有用であることが記載されている。
即ち、本発明は、式(I)の化合物又はその塩、並びに、式(I)の化合物又はその塩、及び一以上の賦形剤を含有する医薬組成物に関する。
R1は、低級アルキル又はハロゲノ低級アルキルであり、
R2は、ハロゲン又はハロゲノ低級アルキルであり、
Lは、結合又は-CH2-であり、
Aは、CH又はNであり、
R3は、H又は低級アルキルであり、
R4及びR5は、同一または異なって、H又は低級アルキルであり、

なお、特に記載がない限り、本明細書中のある化学式中の記号が他の化学式においても用いられる場合、同一の記号は同一の意味を示す。
また、本発明は、
(1) カテプシンS阻害剤である、式(I)の化合物又はその塩、
(2) カテプシンS阻害剤として使用するための式(I)の化合物又はその塩、
(3) 式(I)の化合物又はその塩を含有するカテプシンS阻害剤、
(4) SLEやループス腎炎を含む自己免疫疾患、アレルギー、又は、臓器、骨髄若しくは組織の移植片拒絶の予防及び/又は治療用医薬組成物の製造のための式(I)の化合物又はその塩の使用、
(5) SLEやループス腎炎を含む自己免疫疾患、アレルギー、又は、臓器、骨髄若しくは組織の移植片拒絶の予防及び/又は治療のための式(I)の化合物又はその塩の使用、
(6) SLEやループス腎炎を含む自己免疫疾患、アレルギー、又は、臓器、骨髄若しくは組織の移植片拒絶の予防及び/又は治療に用いるための式(I)の化合物又はその塩、並びに、
(7) 式(I)の化合物又はその塩の有効量を対象に投与することからなるSLEやループス腎炎を含む自己免疫疾患、アレルギー、又は、臓器、骨髄若しくは組織の移植片拒絶の予防及び/又は治療方法、に関する。
なお、「対象」とは、その予防又は治療を必要とするヒト又はその他の動物であり、ある態様としては、その予防又は治療を必要とするヒトである。
(1) 式(I)が以下の式(Ia)である化合物又はその塩。

(1)粉末X線回折で2θ(°)2.7、5.3、9.8、10.4、13.5、14.0、15.1、16.6、17.4 、及び24.4付近にピークを有する。
(2)粉末X線回折で特徴的には2θ(°) 5.3、9.8、15.1、16.6、及び24.4付近にピークを有する。
(3)示差走査熱量計分析(DSC分析)で134.3℃付近に吸熱ピークを有する。
ここで、化合物Aとコハク酸を1:2のモル比で含有する結晶には、化合物Aの二コハク酸塩の結晶及び化合物Aの1コハク酸塩と1コハク酸との共結晶も包含される。
また、式(I)の化合物には、不斉中心又は軸不斉を有する場合があり、これに基づくエナンチオマー(光学異性体)が存在しうる。式(I)の化合物又はその塩は、単離された個々の(R)体、(S)体などのエナンチオマー、それらの混合物(ラセミ混合物又はラセミでない混合物を含む)のいずれも包含する。ある態様では、エナンチオマーは、「立体化学的に純粋」である。「立体化学的に純粋」とは、当業者が、実質的に立体化学的に純粋と認識できる程度の純度をいう。別の態様としては、エナンチオマーは、例えば、90% ee(enantiomeric excess)以上、95% ee以上、98% ee以上、99% ee以上の立体化学的純度を持つ化合物である。
同位体でラベルされた本願発明の化合物は、薬物及び/又は基質の組織分布研究等の研究等に使用しうる。例えば、トリチウム(3H)、炭素14(14C)等の放射性同位体は、ラベルの容易さ及び検出の簡便さから、本目的で使用しうる。
より重い同位体への置換、例えば、水素の重水素(2H)への置換は、代謝安定性が向上することにより治療上有利(例えば、in vivoでの半減期の増加、必要用量の減少、薬物相互作用の減少)な場合がある。
陽電子放出同位体(11C, 18F, 15O及び13N等)への置換は、基質受容体占有率を試験するためにポジトロン断層法(PET)試験で使用しうる。
本発明の同位体でラベルされた化合物は、一般的に、当業者に知られた従来の手法により、又は、ラベルされていない試薬の代わりに同位体ラベルされた適切な試薬を用いて、実施例又は製造例と同様の製法等により製造することができる。
式(I)の化合物及びその塩は、その基本構造あるいは置換基の種類に基づく特徴を利用し、種々の公知の合成法を適用して製造することができる。その際、官能基の種類によっては、当該官能基を原料から中間体へ至る段階で適当な保護基(容易に当該官能基に転化可能な基)に置き換えておくことが製造技術上効果的な場合がある。このような保護基としては、例えば、ウッツ(P. G. M. Wuts)及びグリーン(T. W. Greene)著、「Greene's Protective Groups in Organic Synthesis(第4版、2006年)」に記載の保護基等を挙げることができ、これらの反応条件に応じて適宜選択して用いればよい。このような方法では、当該保護基を導入して反応を行ったあと、必要に応じて保護基を除去することにより、所望の化合物を得ることができる。
製薬学的に許容されるプロドラッグとは、加溶媒分解により又は生理学的条件下で、アミノ基、水酸基、カルボキシル基等に変換されうる基を有する化合物である。プロドラッグを形成する基としては、例えば、Prog. Med., 5, 2157-2161 (1985)や、「医薬品の開発」(廣川書店、1990年)第7巻 分子設計163-198に記載の基が挙げられる。
また、式(I)の化合物のプロドラッグは、上記保護基と同様、原料から中間体へ至る段階で特定の基を導入、あるいは得られた式(I)の化合物を用いてさらに反応を行うことで製造できる。反応は通常のエステル化、アミド化、脱水等、当業者に公知の方法を適用することにより行うことができる。
以下、式(I)の化合物の代表的な製造法を説明する。各製法は、当該説明に付した参考文献を参照して行うこともできる。なお、本発明の製造法は以下に示した例には限定されない。

Ac=アセチル、BOC=tert-ブトキシカルボニル、t-Bu=tert-ブチル、Me=メチル、Et=エチル、CF3=トリフルオロメチル、Ms=メタンスルホニル、Ts=p-トルエンスルホニル、Tf=トリフルオロメタンスルホニル、Ph=フェニル。
式(I)の化合物は、化合物(1)と化合物(2)とのカップリング反応により得ることができる。脱離基の例としては、ハロゲン、TfO等が挙げられる。
この反応では、化合物(1)と化合物(2)を、等量或いは一方を過剰量用い、これらの混合物を、反応に不活性な溶媒中、塩基及びパラジウム触媒の存在下、室温から加熱還流下、ある態様としては室温から150℃において、通常0.1時間から5日間撹拌する。
溶媒の例としては、特に限定されないが、トルエン等の芳香族炭化水素類、THF、1,4-ジオキサン等のエーテル類、ジクロロメタン等のハロゲン化炭化水素類、アルコール類、DMF、DMSO、EtOAc、MeCN、H2O、及びこれらの混合溶媒が挙げられる。塩基の例としては、Cs2CO3、K3PO4、K2CO3、Na2CO3、KOH等の無機塩基が好ましい。パラジウム触媒としては、酢酸パラジウムとXPhos、RuPhos等のホスフィン配位子により系中で調整されるパラジウム触媒、テトラキス(トリフェニルホスフィン)パラジウム、ジクロロビス(トリフェニルホスフィン)パラジウム、1,1'-ビス(ジフェニルホスフィノ)フェロセン-パラジウム(II)ジクロリド等が挙げられる。

式(I-1)の化合物は、化合物(3)に脱離基を導入した後、化合物(4)と反応させることにより得ることができる。
この反応では、化合物(3)を、MsClやTsCl等のハロゲン化スルホニル化合物と、反応に不活性な溶媒中、塩基の存在下で反応させて得られる化合物と、化合物(4)を、等量或いは一方を過剰量用い、これらの混合物を、反応に不活性な溶媒中、塩基の存在下、氷冷下から加熱還流下、好ましくは0℃から120℃において、通常0.1時間から5日間撹拌する。
溶媒の例としては、特に限定されないが、トルエン等の芳香族炭化水素類、1,4-ジオキサン等のエーテル類、ジクロロメタン等のハロゲン化炭化水素類、DMF、DMSO、EtOAc、MeCN、及びこれらの混合物が挙げられる。塩基の例としては、TEA、DIPEA若しくはNMM等の有機塩基、又はK2CO3、Na2CO3若しくはKOH等の無機塩基等が挙げられる。
また、化合物(5)を反応性誘導体へ変換した後に化合物(6)と反応させる方法も用いることができる。カルボン酸の反応性誘導体の例としては、オキシ塩化リン、塩化チオニル等のハロゲン化剤と反応して得られる酸ハロゲン化物、クロロギ酸イソブチル等と反応して得られる混合酸無水物、1-ヒドロキシベンゾトリアゾール等と縮合して得られる活性エステル等が挙げられる。これらの反応性誘導体と化合物(6)との反応は、ハロゲン化炭化水素類、芳香族炭化水素類、エーテル類等の反応に不活性な溶媒中、冷却下~加熱下、好ましくは、-20℃~60℃で行うことができる。

Step1-1乃至Step1-3で示される工程は、それぞれ、化合物(17)からWittig試薬(リンイリド)として用いる化合物(15)を得る反応、及び、化合物(15)と化合物(14)とのWittig反応により化合物(13)を得る反応である。Step1-1及びStep1-2では、化合物(17)をN-ブロモスクシンイミドや臭素等のハロゲン化剤と反応させ、ハロゲン化物(16)とした後、トリフェニルホスフィンとの混合物を、反応に不活性な溶媒中、冷却下から加熱還流下、ある態様として0℃から120℃において、通常0.1時間から5日間撹拌する。ハロゲン化反応時にジハロゲン化された場合には、ホスホン酸ジエチルと反応させることで、モノハロゲン化物(16)を得ることができる。Step1-3では、化合物(15)と化合物(14)との混合物を、反応に不活性な溶媒中、塩基の存在下、冷却下から加熱還流下、好ましくは0℃から100℃において、通常0.1時間から10日間撹拌する。溶媒の例としては、芳香族炭化水素類、エーテル類、ジクロロメタン等のハロゲン化炭化水素類、アルコール類、DMF、DMSO、EtOAc、MeCN、及びこれらの混合物が挙げられる。塩基の例としては、ナトリウムメトキシド、カリウムtert-ブトキシド、n-ブチルリチウム、リチウムヘキサメチルジシラジド等の有機塩基、又はK2CO3、Na2CO3、KOH等の無機塩基が挙げられる。

Step1-4で示される工程は、化合物(13)のアルケンのハイドロボレーションに続く酸化反応により化合物(12)を得る反応である。この反応では、化合物(13)と、ボラン-THF錯体、9-ボラビシクロ[3.3.1]ノナン、ジシアミルボラン、デキシルボラン等との混合物を、反応に不活性な溶媒中、好ましくは10℃から80℃で通常0.1時間から3日間撹拌することにより得た反応物を、反応に不活性な溶媒中、塩基の存在下で、冷却下から加熱還流下、好ましくは-20℃から80℃で、通常0.1時間から3日間、等量若しくは過剰量の酸化剤で処理することにより、化合物(12)を得ることができる。

Step1-5で示される工程は、化合物(12)の酸化反応により化合物(11)を得る反応である。この反応では、反応に不活性な溶媒中、冷却下から加熱下、好ましくは、-20℃から80℃で、化合物(12)を、通常0.1時間から3日間、等量若しくは過剰量の酸化剤で処理する。本反応においては、テトラプロピルアンモニウムペルルテナート(TPAP)酸化、スワーン(Swern)酸化等のDMSO酸化、又は、デスマーチン(Dess-Martin)試薬を用いた酸化が好適に用いられる。TPAP酸化においては、酸化触媒であるテトラプロピルアンモニウムペルルテナート、脱水剤であるモレキュラーシーブ4A、及び再酸化剤であるN-メチルモルホリン-N-オキシド(NMMO)の存在下で、化合物(12)を処理する。溶媒の例としては、芳香族炭化水素類、エーテル類、ジクロロメタン等のハロゲン化炭化水素類、DMF、DMSO、EtOAc、MeCN、及びこれらの混合物が挙げられる。他の酸化剤の例としては、過酸化水素、クメンヒドロペルオキシド、過酢酸、過安息香酸、m-クロロ過安息香酸、オキソン、活性二酸化マンガン、クロム酸、過マンガン酸カリウム、過ヨウ素酸ナトリウム等が挙げられる。

Step1-9で示される工程は、化合物(7)と化合物(6)を縮合することにより化合物(1)を得る反応である。第3製法と同様の方法で行うことができる。
Pg3の例としては、2,4,6-トリクロロフェニルが挙げられる。
Step2-1で示される工程は、化合物(1)と化合物(19)との反応により化合物(18)を得る反応である。第1製法と同様の方法で、化合物(2)の代わりに化合物(19)を用いることで行うことができる。
溶媒としては、トルエン又はベンゾトリフルオリドが好ましい。塩基としては、TEA又はトリブチルアミンが好ましい。パラジウム触媒としては、酢酸パラジウムとXantPhos等のホスフィン配位子により系中で調整されるパラジウム触媒が好ましい。

Step2-2で示される工程は、化合物(18)の還元反応により化合物(3)を得る反応である。この反応では、反応に不活性な溶媒中、冷却下から加熱下、好ましくは-20℃から80℃で、化合物(18)を等量若しくは過剰量の還元剤で、通常0.1時間から3時間処理する。溶媒の例としては、芳香族炭化水素類、エーテル類、ハロゲン化炭化水素類、MeOH等のアルコール類、DMF、DMSO、EtOAc、MeCN、及びこれらの混合物が挙げられる。還元剤の例としては、水素化ホウ素ナトリウム、水素化ホウ素リチウム等のヒドリド還元剤が挙げられる。

Step3-2で示される工程は、化合物(20)の脱保護により化合物(5)を得る反応である。原料合成1のStep1-8と同様の方法で行うことができる。
単離、精製は、抽出、分別結晶化、各種分画クロマトグラフィー等、通常の化学操作を適用して行なわれる。
各種の異性体は、適当な原料化合物を選択することにより製造でき、あるいは異性体間の物理化学的性質の差を利用して分離することができる。例えば、光学異性体は、ラセミ体の一般的な光学分割法(例えば、光学活性な塩基又は酸とのジアステレオマー塩に導く分別結晶化や、キラルカラム等を用いたクロマトグラフィー等)により得られ、また、適当な光学活性な原料化合物から製造することもできる。
5μLのヒトカテプシンS酵素(R&D 1183-CY-010)を20 ng/wellとなるように96ウェルプレートに添加した。次に、被験化合物(10 mM DMSO溶液)を、アッセイ用緩衝液(50mM酢酸ナトリウム、250mM塩化ナトリウム及び5mMジチオトレイトール(DTT)、pH=4.5)で最終濃度が0.1 nM~1μMになるように10倍希釈系列にて5段階もしくは0.01 nM~10 nMとなるように3倍希釈系列にて7段階に希釈し、当該ウェルへ10 μL添加し(最終DMSO濃度は0.1%)、続いて25 μLの合成基質VVR-AMC(ペプチド研究所 3211-V)を最終濃度40 μMとなるように添加して酵素反応を開始した。分光蛍光光度計(SPECTRA MAX GEMINI、モレキュラーデバイス)を用いて反応開始直後から5~10分間、37℃にて蛍光強度(励起波長:380 nm、蛍光波長:460 nm)を測定し、直線性の認められる時点(5分)での反応速度を被験化合物の各濃度において求めた。被験化合物を加えない酵素非添加時の反応速度及び被験化合物を加えない酵素添加時の反応速度を、それぞれ100%抑制、0%抑制として各濃度の阻害率を求め、シグモイドEmax非線形回帰法にてIC50値を算出した。結果を表1に示す。表中Exは実施例化合物番号、Dat1はヒトカテプシンS阻害活性のIC50値(nM)を示す。
抗原提示細胞において、カテプシンSの阻害はMHC class II分子の発現を抑制する。その結果、CD4陽性T細胞への抗原提示が抑制されることで外来抗原に対する免疫応答の低下が引き起こされる。B細胞におけるMHC class IIの発現上昇に対し、式(I)の化合物について阻害作用を検討した。
雄性C57BL/6Jマウス(日本チャールスリバー)より採取した脾細胞を、1×105cells/wellとなるように96ウェルプレートに播種し、被験化合物の10 mM DMSO溶液をRPMI1640培地(10%ウシ胎児血清(FCS)、5×10-5 M 2-メルカプトエタノール、50 IU/mLペニシリン、及び、50 μg/mLストレプトマイシンを含む)で最終濃度が0.026 nM~10 μMになるように5倍希釈系列にて9段階希釈、もしくは0.205 pM~10 μMになるように5倍希釈系列にて12段階希釈して添加した(最終DMSO濃度は0.1%)。同時にLPS(Sigma L4005)を最終濃度2 μg/mLとなるように当該ウェルへ添加し、37℃、5%CO2下で48時間培養を行った。培養後、ビオチン標識YAe抗体(EBIOSCIENCE 13-5741-85)で細胞を4℃、20分間染色し、洗浄後さらにFITC蛍光標識抗マウスB220抗体(BD BIOSCIENCES 553088)及びPE蛍光標識ストレプトアビジン(BD BIOSCIENCES 554061)で細胞を4℃、20分間染色し、フローサイトメトリーシステム(Guava EasyCyte Plus System、ミリポア)を用いてB220陽性B細胞におけるEaペプチドを結合したMHC class IIの発現量(YAe-ビオチン/ストレプトアビジン-PEの蛍光強度)を測定した。被験化合物を加えないLPS非刺激時の値及び被験化合物を加えないLPS刺激時の値を、それぞれ100%抑制、0%抑制として各濃度の阻害率を求め、シグモイドEmax非線形回帰法によりIC50値を算出した。結果を表2に示す。表中Exは実施例化合物番号、Dat2はIC50値(nM)を示す。
MHC class II分子の発現抑制作用をex vivoの系で評価した。
雄性C57BL/6Jマウス(日本チャールスリバー)に被験化合物を経口投与し、経口投与後の末梢血中のB細胞におけるMHC class IIの発現上昇に対する抑制作用を検討した。すなわち雄性C57BL/6Jマウスへvehicle[30%のプロピレングリコール溶剤{プロピレングリコール:硬化ヒマシ油(HCO40):Tween80=4:2:1}/HCl(被験化合物に対して2当量)/water]で溶解した被験化合物を10 mL/kgで経口投与(投与用量0.3 mg/kg、1群4例)し、6時間後に末梢血を回収した。当該末梢血90 μLにPBSを10 μLまたはLPS(Sigma L4005)を10 μL(最終濃度100μg/mL)添加し、37℃、5%CO2下で15時間培養を行った。培養後、FITC蛍光標識抗マウスI-A/I-E抗体(BD BIOSCIENCES 553623)及びPE蛍光標識抗マウスB220抗体(BD BIOSCIENCES 553090)で細胞を冷蔵下、30分間染色した後、バッファー(BD BIOSCIENCES Phosflow Lyse/Fix Buffer 558049)を用いて37℃、11~12分間溶血及び固定を行った。洗浄後、フローサイトメトリーシステム(FACSCanto II、BD BIOSCIENCES)を用いてB220陽性B細胞表面上のMHC class IIの発現量をFITCの平均蛍光強度(以下MFIと表記する)を指標に測定した。LPS刺激時のMFIと無刺激時のMFIとの差をΔMFIとし、vehicleを10 mL/kg投与したマウスのΔMFIを1として、被験化合物投与によるΔMFIの抑制率を算出した。
NZB/W F1マウス(日本エスエルシー)はヒトに近い病態を自然発症するSLEのモデルとして用いられる(「JAMA」、1966年、第195巻、p.145;「Advances in Immunology」、1985年、第37巻、p.269-390;「Journal of Biomedicine and Biotechnology」2011年、第2011巻、271694)。
NZB/W F1雌性マウスを、19週齢時点の尿タンパク値(クレアチニン補正値)、血漿中抗double stranded DNA (dsDNA)抗体価(IgG)、末梢血B細胞上MHC class IIの発現量、体重をもとに群分けを行った。この時点で尿タンパク値(クレアチニン補正値)が3以上の個体は除外した。20週齢より被験化合物を1日2回経口投与し、その後経時的に採尿、採血を行って尿タンパク値、抗dsDNA抗体価の経時的推移を評価した。
上記試験において、SLE様病態に伴い産生される血漿中抗dsDNA IgG抗体価を、ELISA法(ALPHA DIAGNOSTIC 5120)により測定した。各個体の28週齢と32週齢における抗体価の幾何平均値において、vehicle投与群10例の幾何平均値が216662 U/mLであったのに対し、実施例2の化合物1 mg/kg b.i.d.投与群10例の幾何平均値は26827 U/mLと有意差(P値=0.0009、Dunnett多重比較)をもって低値であった。また40週齢における尿タンパク値(クレアチニン補正値)を測定した。Vehicle投与群10例の幾何平均値が13.0であったのに対し、実施例2の化合物1mg/kg b.i.d.投与群10例の幾何平均値は0.7と有意差(P値=0.0005、Dunnett多重比較)をもって低値であった。この事から、実施例2の化合物がNZB/W F1雌性マウスにおけるSLE様病態発症を抑制する効果が確認された。
NZB/W F1マウス(日本エスエルシー)にToll様受容体3のリガンドであるpoly(I:C)を投与する事により、SLE様病態に伴うタンパク尿増加を加速化する事ができる。poly(I:C)投与によりタンパク尿を誘導した状態から被験化合物の投与を開始し、SLE様病態の治療作用を評価する。
22週齢のNZB/W F1マウスに、poly(I:C) (InvivoGen tlrl-picw-250)200μgを週3回、4週間、計12回投与する。その後2週の間に尿タンパク値(クレアチニン補正値)が原則2~50となった個体を試験に組み入れ、尿タンパク値をもとに群分けを行う。群分け後より被験化合物を1日2回、5週間経口投与し、経時的に採尿を行って尿タンパク値の経時的推移を評価する。
投与は錠剤、丸剤、カプセル剤、顆粒剤、散剤、液剤等による経口投与、又は、関節内、静脈内、筋肉内等の注射剤、坐剤、点眼剤、眼軟膏、経皮用液剤、軟膏剤、経皮用貼付剤、経粘膜液剤、経粘膜貼付剤、吸入剤等による非経口投与のいずれの形態であってもよい。
経口投与のための液体組成物は、薬剤的に許容される乳濁剤、溶液剤、懸濁剤、シロップ剤又はエリキシル剤等を含み、一般的に用いられる不活性な希釈剤、例えば精製水又はエタノールを含む。当該液体組成物は不活性な希釈剤以外に可溶化剤、湿潤剤、懸濁剤のような補助剤、甘味剤、風味剤、芳香剤、防腐剤を含有していてもよい。
DSC測定は、DSC Q2000(TA Instruments製)を用い、測定温度範囲:室温~300℃、昇温速度:10℃/min、窒素流量:50 mL/minの条件で、アルミニウム製サンプルパンを用い,サンプルパンには蓋をしない状態で測定を行った。
なお、粉末X線回折パターンはデータの性質上、結晶の同一性認定においては、結晶格子間隔や全体的なパターンが重要であり、粉末X線回折における回折角(2θ(°))の誤差範囲は、通常±0.2°であるが、回折角及び回折強度は結晶成長の方向、粒子の大きさ、測定条件によって多少変わりうるものであるから、厳密に解されるべきではない。

構造式中のHClは塩酸塩であることを示し、HClの前の数字はモル比を示す。例えば2HClは二塩酸塩であることを意味する。同様に、SAは、コハク酸塩であることを示し、2SAは二コハク酸塩(化合物とコハク酸を1:2でのモル比含有する共結晶を含む)であることを意味する。
製造例番号及び実施例番号に付記したgmは、幾何異性体の混合物であることを表す。同様に、emは、ピロリジン環4位エピマーの混合物又はピロリジン環3位エピマーの混合物、dmは、ベンジル基α位及びピロリジン環4位の立体配置の異なるジアステレオマーの混合物であることを、それぞれ表す。
アルゴンガス雰囲気下、4-ブロモ-1-メチル-2-(トリフルオロメチル)ベンゼン (100 g)のMeCN (700 mL)溶液に室温にて、N-ブロモスクシンイミド (148.9 g)及び47%臭化水素酸 (5 mL)を加えた。混合物に、2,2'-アゾビス(イソブチロニトリル) (3.43 g)を加え、70℃で10分間撹拌し、100℃で終夜撹拌した。混合物を室温まで冷却した。混合物を氷冷し、飽和チオ硫酸ナトリウム水溶液を加え、10分間撹拌した。混合物に、水及びEtOAcを加えた。有機層を分離し、飽和炭酸水素ナトリウム水溶液及びbrineで洗浄した。有機層を無水硫酸ナトリウムで乾燥し、減圧下濃縮した。残渣にn-ヘキサンを加え、析出した固体を濾別した。濾液を減圧下濃縮した。残渣のTHF (500 mL)溶液に、氷冷下にて、DIPEA (79 mL)及びホスホン酸ジエチル (54 mL)を加えた。混合物を室温で2時間撹拌した。混合物を氷冷し、水を加えた。混合物にEtOAcを加え、有機層を分離し、塩酸 (1M)、水、飽和炭酸水素ナトリウム水溶液及びbrineで洗浄した。有機層をMgSO4で乾燥し、減圧下濃縮した。残渣のトルエン(700 mL)溶液に、トリフェニルホスフィン (115 g)を加え、100℃で終夜撹拌した。析出物を濾取し、トルエンで洗浄し、臭化[4-ブロモ-2-(トリフルオロメチル)ベンジル](トリフェニル)ホスホニウム (198.51 g)を固体として得た。
アルゴンガス雰囲気下、臭化[4-ブロモ-2-(トリフルオロメチル)ベンジル](トリフェニル)ホスホニウム (150 g)のジクロロメタン (600 mL)溶液に、氷冷下にて、カリウムtert-ブトキシド (27.5 g)を加え、室温で6時間撹拌した。混合物を氷冷し、(2S)-4-オキソピロリジン-1,2-ジカルボン酸1-tert-ブチルエステル2-メチルエステル (50 g)のジクロロメタン (150 mL)溶液を加え、室温で4日間撹拌した。混合物に飽和塩化アンモニウム水溶液を加え、15分間撹拌した。有機層を分離し、水層をクロロホルムで抽出した。合わせた有機層をbrineで洗浄した。有機層をMgSO4で乾燥し、減圧下濃縮した。残渣にEtOAc (40 mL)及びn-ヘキサン (200 mL)を加え、室温で1時間撹拌した。析出物を濾別し、濾液を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(溶出溶媒;n-ヘキサン/EtOAc)で精製し、(2S)-4-[4-ブロモ-2-(トリフルオロメチル)ベンジリデン]ピロリジン-1,2-ジカルボン酸1-tert-ブチルエステル2-メチルエステル(幾何異性体の混合物) (51.74 g)を油状物として得た。

窒素ガス雰囲気下、(2S)-4-{[4-ブロモ-2-(トリフルオロメチル)フェニル](ヒドロキシ)メチル}ピロリジン-1,2-ジカルボン酸1-tert-ブチルエステル2-メチルエステル(ベンジル基α位及びピロリジン環4位の立体配置の異なるジアステレオマーの混合物) (20.96 g)のジクロロメタン (105 mL)溶液に、氷冷下にて、4-メチルモルホリンN-オキシド (6.1 g)及びモレキュラーシーブ 4A (8.7 g)を加え、10分間撹拌した。混合物に、氷冷下にて、テトラプロピルアンモニウムペルルテナート (1.5 g)を加え、室温で1時間撹拌した。混合物にEtOAcを加え、減圧下濃縮した。残渣をシリカゲルを用いて、濾過し、EtOAcで洗浄した。濾液に飽和チオ硫酸ナトリウム水溶液を加え、撹拌した。有機層を分離し、brineで洗浄した。有機層をMgSO4で乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(溶出溶媒;n-ヘキサン/EtOAc)で精製し、(2S)-4-[4-ブロモ-2-(トリフルオロメチル)ベンゾイル]ピロリジン-1,2-ジカルボン酸1-tert-ブチルエステル2-メチルエステル(ピロリジン環4位エピマーの混合物) (19.6 g)を油状物として得た。
(2S)-4-[4-ブロモ-2-(トリフルオロメチル)ベンゾイル]ピロリジン-1,2-ジカルボン酸1-tert-ブチルエステル2-メチルエステル(ピロリジン環4位エピマーの混合物) (78.9 g)のMeOH (395 mL)懸濁液に氷冷下にて、塩化水素 (4M 1,4-ジオキサン溶液、395 mL)を加え、室温で1時間撹拌した。混合物を減圧下濃縮した。混合物にトルエンを加え、減圧下濃縮した。窒素ガス雰囲気下、残渣のジクロロメタン (630 mL)溶液に氷冷下にて、1-メチルシクロプロパンカルボン酸 (20 g)、HATU (76 g)及びDIPEA (86 mL)を加え、室温で5時間撹拌した。混合物を氷冷し、飽和炭酸水素ナトリウム水溶液を加えた。混合物にクロロホルムを加え、有機層を分離し、塩酸 (1M)、飽和炭酸水素ナトリウム水溶液及びbrineで洗浄した。有機層を無水硫酸ナトリウムで乾燥し、減圧下濃縮した。残渣を、アミノシリカゲルカラムクロマトグラフィー(溶出溶媒;n-ヘキサン/EtOAc)及びシリカゲルカラムクロマトグラフィー(溶出溶媒;n-ヘキサン/EtOAc)で精製し、4-[4-ブロモ-2-(トリフルオロメチル)ベンゾイル]-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンメチルエステル(ピロリジン環4位エピマーの混合物) (57.65 g)を油状物として得た。
アルゴンガス雰囲気下、テフロン(登録商標)製反応容器を用いて、4-[4-ブロモ-2-(トリフルオロメチル)ベンゾイル]-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンメチルエステル(ピロリジン環4位エピマーの混合物) (64 g)のジクロロメタン (320 mL)溶液に4-tert-ブチル-2,6-ジメチルフェニルサルファートリフルオリド (138.6 g)及びフッ化水素ピリジン (101.5 mL)を加え、室温で15時間撹拌した。反応容器の撹拌を止め、室温で75時間静置した。混合物を氷及び28% アンモニア水溶液 (1400 mL)の混合物に加えた。混合物にクロロホルムを加え、8時間撹拌した。有機層を分離し、水層をクロロホルムで抽出した。合わせた有機層を飽和炭酸水素ナトリウム水溶液、塩酸 (1M)、飽和炭酸水素ナトリウム水溶液及びbrineで洗浄した。有機層をMgSO4で乾燥し、減圧下濃縮した。残渣にn-ヘキサンを加え、室温で1時間撹拌した。不溶物を濾別し、濾液を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(溶出溶媒;n-ヘキサン/EtOAc)で精製した。得られた油状物をn-ヘキサン (253 mL)及びEtOAc (2.5 mL)と混合した。混合物を加熱し、固体が析出した時点で放冷し、室温で15時間撹拌した。析出物を濾取し、n-ヘキサン及びEtOAc(99 : 1)の混合液で洗浄し、(4R)-4-{[4-ブロモ-2-(トリフルオロメチル)フェニル](ジフルオロ)メチル}-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンメチルエステル (26.6 g)を固体として得た。
(4R)-4-{[4-ブロモ-2-(トリフルオロメチル)フェニル](ジフルオロ)メチル}-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンメチルエステル (26.57 g)及びTHF (265 mL)の混合液に、氷冷下にて、水酸化リチウム一水和物水溶液 (1M、82 mL)を加え、室温で2時間撹拌した。混合物を氷冷し、塩酸 (1M、約85 mL)を加えた。混合物にEtOAcを加え、有機層を分離し、brineで洗浄した。有機層を無水硫酸ナトリウムで乾燥し、減圧下濃縮し、(4R)-4-{[4-ブロモ-2-(トリフルオロメチル)フェニル](ジフルオロ)メチル}-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリン (26.03 g)を固体として得た。
(4R)-4-{[4-ブロモ-2-(トリフルオロメチル)フェニル](ジフルオロ)メチル}-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリン (25.80 g)のジクロロメタン (258 mL)溶液に、氷冷下にて、1-アミノシクロプロパンカルボニトリル一塩酸塩 (7.19 g)、HATU (21.9 g)、DIPEA (23.5 mL)を加えた。混合物を室温で15時間撹拌した。混合物に、クロロホルム及び飽和塩化アンモニウム水溶液を加えた。有機層を分離し、飽和炭酸水素ナトリウム水溶液及びbrineで洗浄した。有機層を無水硫酸ナトリウムで乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(溶出溶媒;n-ヘキサン/EtOAc)で精製し、(4R)-4-{[4-ブロモ-2-(トリフルオロメチル)フェニル](ジフルオロ)メチル}-N-(1-シアノシクロプロピル)-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンアミド (28.68 g)を固体として得た。
アルゴンガス雰囲気下、テフロン(登録商標)製反応容器を用いて、4-[4-ブロモ-2-(トリフルオロメチル)ベンゾイル]-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンメチルエステル(ピロリジン環4位エピマーの混合物) (12.9 g)のジクロロメタン (64 mL)溶液に4-tert-ブチル-2,6-ジメチルフェニルサルファートリフルオリド (25 g)およびフッ化水素ピリジン (18 mL)を加え、室温で9時間撹拌した。反応容器の撹拌を止め、室温で14時間静置した。混合物を室温で、11時間撹拌した。反応容器の撹拌を止め、室温で14時間静置した。混合物を氷、28% アンモニア水溶液 (220 mL)及びクロロホルムの混合物に加え、3時間撹拌した。有機層を分離し、水層をクロロホルムで抽出した。合わせた有機層を無水硫酸ナトリウムで乾燥し、減圧下濃縮した。残渣にn-ヘキサンを加え、室温で1時間撹拌した。不溶物を濾別し、濾液を減圧下濃縮した。残渣にトルエンを加え、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(溶出溶媒;n-ヘキサン/EtOAc)で精製し、4-{[4-ブロモ-2-(トリフルオロメチル)フェニル](ジフルオロ)メチル}-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンメチルエステル(ピロリジン環4位エピマーの混合物) (11.67 g)を油状物として得た。
4-{[4-ブロモ-2-(トリフルオロメチル)フェニル](ジフルオロ)メチル}-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンメチルエステル(ピロリジン環4位エピマーの混合物) (3.69 g)をカラムクロマトグラフィー (カラム:CHIRALPAK IA、溶出溶媒:n-ヘキサン/EtOAc)で精製し、(4R)-4-{[4-ブロモ-2-(トリフルオロメチル)フェニル](ジフルオロ)メチル}-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンメチルエステル (1.91 g)を油状物として得た。
アルゴンガス雰囲気下、4-{[4-ブロモ-2-(トリフルオロメチル)フェニル](ジフルオロ)メチル}-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンメチルエステル(ピロリジン環4位エピマーの混合物) (478 mg)、トリフルオロ[(4-メチルピペラジン-1-イル)メチル]ホウ酸(1-)カリウム (435 mg)、酢酸パラジウム(II) (23 mg)、RuPhos (97 mg)及びK3PO4 (839 mg)の1,4-ジオキサン (10 mL)及び水 (0.072 mL)混合物を80℃で終夜撹拌した。混合物に飽和炭酸水素ナトリウム水溶液を加えた。混合物にクロロホルムを加え、有機層を分離した。有機層をbrineで洗浄し、減圧下濃縮した。残渣をアミノシリカゲルカラムクロマトグラフィー(溶出溶媒;クロロホルム/MeOH)で精製し、4-(ジフルオロ{4-[(4-メチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}メチル)-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンメチルエステル(ピロリジン環4位エピマーの混合物) (340 mg)を油状物として得た。
4-ブロモ-1-(ブロモメチル)-2-クロロベンゼン (10 g)のトルエン (70 mL)溶液に、室温にてトリフェニルホスフィン (9.75 g)を加え、80℃で6時間撹拌した。混合物を氷冷し、30分間撹拌した。析出物を濾取し、トルエンで洗浄し、臭化(4-ブロモ-2-クロロベンジル)(トリフェニル)ホスホニウム (18.9 g)を固体として得た。
アルゴンガス雰囲気下、4-{[4-ブロモ-2-(トリフルオロメチル)フェニル](ジフルオロ)メチル}-N-(1-シアノシクロプロピル)-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンアミド(ピロリジン環4位エピマーの混合物) (2.1 g)のトルエン (30 mL)溶液に100℃にて、ギ酸2,4,6-トリクロロフェニルエステル (1 g)、酢酸パラジウム(II) (120 mg)、XantPhos (300 mg)及びDIPEA (1.5 mL)を加え、1時間撹拌した。混合物を氷冷し、クロロホルム及びbrineを加えた。不溶物をセライトで濾別し、濾液の有機層を分離した。有機層を無水硫酸ナトリウムで乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(溶出溶媒;n-ヘキサン/EtOAc)で精製し、4-{[(5S)-5-[(1-シアノシクロプロピル)カルバモイル]-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}ピロリジン-3-イル](ジフルオロ)メチル}-3-(トリフルオロメチル)安息香酸2,4,6-トリクロロフェニルエステル(ピロリジン環3位エピマーの混合物) (2.4 g)を固体として得た。
4-{[(5S)-5-[(1-シアノシクロプロピル)カルバモイル]-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}ピロリジン-3-イル](ジフルオロ)メチル}-3-(トリフルオロメチル)安息香酸2,4,6-トリクロロフェニルエステル(ピロリジン環3位エピマーの混合物) (1 g)及びTHF (20 mL)の混合物に氷冷下にて、水素化ホウ素ナトリウム (103 mg)を加えた。混合物に氷冷下にて、MeOH (2 mL)を加え、室温で30分間撹拌した。混合物に水を加えた。混合物にEtOAcを加え、有機層を分離し、brineで洗浄した。有機層をMgSO4で乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(溶出溶媒;n-ヘキサン/EtOAc)で精製し、N-(1-シアノシクロプロピル)-4-{ジフルオロ[4-(ヒドロキシメチル)-2-(トリフルオロメチル)フェニル]メチル}-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンアミド(ピロリジン環4位エピマーの混合物) (290 mg)を固体として得た。
アルゴンガス雰囲気下、亜鉛 (365 mg)のN,N-ジメチルアセトアミド (2 mL)懸濁液に室温にて1,2-ジブロモエタン (0.032 mL)およびクロロ(トリメチル)シラン (0.047 mL)の混合物を加え、50℃で、15分間撹拌した。混合物に、室温にて、4-ヨードピペリジン-1-カルボン酸tert-ブチルエステル (1.73 g)のN,N-ジメチルアセトアミド (2 mL)溶液を加え、50℃で、15分間撹拌した。上記の混合物を、アルゴンガス雰囲気下、室温にて、4-{[4-ブロモ-2-(トリフルオロメチル)フェニル](ジフルオロ)メチル}-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンメチルエステル(ピロリジン環4位エピマーの混合物) (1 g), 1,1'-ビス(ジフェニルホスフィノ)フェロセン-パラジウム(II)ジクロリド-ジクロロメタン錯体 (151 mg) 及びヨウ化銅(I) (71 mg)のN,N-ジメチルアセトアミド (5 mL)懸濁液に加え、80℃で終夜撹拌した。混合物を室温まで冷却し、EtOAcを加え、不溶物を濾別した。濾液を水及びbrineにて洗浄し、有機層をMgSO4で乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(溶出溶媒;n-ヘキサン/EtOAc)で精製し、4-[4-{ジフルオロ[(5S)-5-(メトキシカルボニル)-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}ピロリジン-3-イル]メチル}-3-(トリフルオロメチル)フェニル]ピペリジン-1-カルボン酸tert-ブチルエステル(ピロリジン環3位エピマーの混合物) (1.15 g)を油状物として得た。
4-[4-{[(5S)-5-[(1-シアノシクロプロピル)カルバモイル]-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}ピロリジン-3-イル](ジフルオロ)メチル}-3-(トリフルオロメチル)フェニル]ピペリジン-1-カルボン酸tert-ブチルエステル(ピロリジン環3位エピマーの混合物) (675 mg)のジクロロメタン (15 mL)及びMeCN (7.5 mL)溶液に、氷冷下にて、トリフルオロ酢酸 (1.5 mL)を加え、室温で3時間撹拌した。混合物に室温にて、トリフルオロ酢酸 (2.25 mL)を加え、4時間撹拌した。混合物を飽和炭酸水素ナトリウム水溶液に加えた。混合物にクロロホルムを加え、有機層を分離し、brineで洗浄した。有機層をMgSO4で乾燥し、減圧下濃縮し、N-(1-シアノシクロプロピル)-4-{ジフルオロ[4-(ピペリジン-4-イル)-2-(トリフルオロメチル)フェニル]メチル}-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンアミド(ピロリジン環4位エピマーの混合物) (509 mg)を固体として得た。
窒素ガス雰囲気下、臭化(4-ブロモ-2-クロロベンジル)(トリフェニル)ホスホニウム (18.9 g)のジクロロメタン(80 mL)溶液に室温にて、炭酸カリウム (5.5 g) を加え、20分間撹拌した。混合物に室温にて、(2S)-4-オキソピロリジン-1,2-ジカルボン酸1-tert-ブチルエステル2-メチルエステル (6.5 g)及び18-クラウン-6 (110 mg)を加え、3日間加熱還流下、撹拌した。混合物を氷冷し、飽和炭酸水素ナトリウム水溶液を加えた。有機層を分離し、brineで洗浄した。有機層を無水硫酸ナトリウムで乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(溶出溶媒;n-ヘキサン/EtOAc)で精製し、(2S)-4-(4-ブロモ-2-クロロベンジリデン)ピロリジン-1,2-ジカルボン酸1-tert-ブチルエステル2-メチルエステル(幾何異性体の混合物) (5.17 g)を油状物として得た。
アルゴンガス雰囲気下、4-(4-ブロモ-2-クロロベンゾイル)-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンメチルエステル(ピロリジン環4位エピマーの混合物) (2.8 g)にビス(2-メトキシエチル)アミノサルファートリフルオリド (15 mL)を加え、90℃で3時間撹拌した。混合物を氷冷し、氷及び飽和炭酸水素ナトリウム水溶液の混合物に加えた。混合物にEtOAcを加え、有機層を分離し、brineで洗浄した。有機層を無水硫酸ナトリウムで乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(溶出溶媒;n-ヘキサン/EtOAc)で精製し、4-[(4-ブロモ-2-クロロフェニル)(ジフルオロ)メチル]-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンメチルエステル(ピロリジン環4位エピマーの混合物) (2.37 g)を油状物として得た。
4-(4-{ジフルオロ[(5S)-5-(メトキシカルボニル)-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}ピロリジン-3-イル]メチル}-3-フルオロフェニル)ピペリジン-1-カルボン酸tert-ブチルエステル(ピロリジン環3位エピマーの混合物) (105 mg)のMeOH (0.7 mL)及びTHF (0.7 mL)溶液に、室温にて、水酸化リチウム一水和物 (20 mg)を加え、室温で1時間撹拌した。混合物を減圧下濃縮し、(2S)-4-[{4-[1-(tert-ブトキシカルボニル)ピペリジン-4-イル]-2-フルオロフェニル}(ジフルオロ)メチル]-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}ピロリジン-2-カルボン酸リチウム(ピロリジン環4位エピマーの混合物) (102 mg)を固体として得た。
アルゴンガス雰囲気下、(4R)-4-{[4-ブロモ-2-(トリフルオロメチル)フェニル](ジフルオロ)メチル}-N-(1-シアノシクロプロピル)-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンアミド (5 g)の1,4-ジオキサン (50 mL)及び水(10 mL)溶液に室温にて、トリフルオロ[(4-メチルピペラジン-1-イル)メチル]ホウ酸(1-)カリウム (4.1 g)、XPhos (892 mg)、炭酸セシウム(12 g)及び酢酸パラジウム(II) (214 mg)を加え、加熱還流下にて、3時間撹拌した。混合物を室温まで冷却し、水を加えた。不溶物をセライトを用いて濾別し、クロロホルムで洗浄した。濾液に水及びクロロホルムを加えた。有機層を分離し、brineで洗浄した。有機層をMgSO4で乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(溶出溶媒;n-ヘキサン/EtOAc、及びクロロホルム/MeOH)で精製し、(4R)-N-(1-シアノシクロプロピル)-4-(ジフルオロ{4-[(4-メチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}メチル)-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンアミド (4.7 g)を固体として得た。
(4R)-N-(1-シアノシクロプロピル)-4-(ジフルオロ{4-[(4-メチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}メチル)-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンアミド (1.43 g)のEtOAc (7 mL)及びエタノール (7 mL)溶液に、塩化水素 (4M 1,4-ジオキサン溶液、1.39 mL)を加え、室温で30分間撹拌した。混合物を減圧下濃縮した。残渣にエタノールを加え、混合物を減圧下濃縮した。残渣にEtOAcを加え、混合物を1時間程度撹拌した。混合物を減圧下濃縮した。残渣にエタノール (10 mL)を加え、95℃で加熱し、溶液とした。混合物を室温で終夜撹拌した。析出物を濾取し、エタノールで洗浄し、(4R)-N-(1-シアノシクロプロピル)-4-(ジフルオロ{4-[(4-メチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}メチル)-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンアミド二塩酸塩 (516 mg)を固体として得た。
アルゴンガス雰囲気下、(4R)-4-{[4-ブロモ-2-(トリフルオロメチル)フェニル](ジフルオロ)メチル}-N-(1-シアノシクロプロピル)-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンアミド (627.4 mg)、トリフルオロ[(4-メチルピペラジン-1-イル)メチル]ホウ酸(1-)カリウム (705 mg)、酢酸パラジウム(II) (25 mg)、RuPhos (105 mg)、K3PO4 (905 mg)、1,4-ジオキサン (13 mL)及び水 (1.3 mL)の混合物を80℃で6時間撹拌した。混合物に飽和炭酸水素ナトリウム水溶液及びクロロホルムを加えた。有機層を分離し、brineで洗浄した。有機層をMgSO4で乾燥し、減圧下濃縮した。残渣をアミノシリカゲルカラムクロマトグラフィー(溶出溶媒;クロロホルム/MeOH)で精製し、(4R)-N-(1-シアノシクロプロピル)-4-(ジフルオロ{4-[(4-メチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}メチル)-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンアミド (491.4 mg)を油状物として得た。

4-(ジフルオロ{4-[(4-メチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}メチル)-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリン(ピロリジン環4位エピマーの混合物) (415 mg)、1-アミノシクロプロパンカルボニトリル一塩酸塩 (117 mg)、HATU (375 mg)、DIPEA (0.338 mL)及びDMF (8 mL)の混合物を、室温で終夜撹拌した。混合物に水を加え、クロロホルムで抽出した。有機層を減圧下濃縮した。残渣をアミノシリカゲルカラムクロマトグラフィー(溶出溶媒;クロロホルム/MeOH)で精製した。得られた油状物を逆相高速液体クロマトグラフィー(溶出溶媒;0.1%ギ酸水溶液/MeCN)で精製した。得られた残渣のジクロロメタン (1 mL)溶液に、塩化水素 (4M 1,4-ジオキサン溶液、0.115 mL)を加え、室温で5分間撹拌した。混合物を減圧下濃縮し、N-(1-シアノシクロプロピル)-4-(ジフルオロ{4-[(4-メチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}メチル)-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンアミド二塩酸塩(ピロリジン環4位エピマーの混合物) (62 mg)を固体として得た。
4-[4-{[(3R,5S)-5-[(1-シアノシクロプロピル)カルバモイル]-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}ピロリジン-3-イル](ジフルオロ)メチル}-3-(トリフルオロメチル)ベンジル]ピペラジン-1-カルボン酸tert-ブチルエステル (35 mg)のジクロロメタン (0.42 mL)及びMeCN (0.11 mL)溶液に氷冷下にて、トリフルオロ酢酸 (0.11 mL)を加え、室温で終夜撹拌した。混合物を減圧下濃縮した。残渣をアミノシリカゲルカラムクロマトグラフィー(溶出溶媒;クロロホルム/MeOH)で精製した。得られた残渣の1,4-ジオキサン (0.35 mL)溶液に塩化水素 (4M 1,4-ジオキサン溶液、0.015 mL)を加え、室温で5分間撹拌した。混合物を減圧下濃縮し、残渣にジイソプロピルエーテルを加えた。析出物を濾取し、(4R)-N-(1-シアノシクロプロピル)-4-{ジフルオロ[4-(ピペラジン-1-イルメチル)-2-(トリフルオロメチル)フェニル]メチル}-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンアミド二塩酸塩 (22 mg)を固体として得た。
N-(1-シアノシクロプロピル)-4-{ジフルオロ[4-(ヒドロキシメチル)-2-(トリフルオロメチル)フェニル]メチル}-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンアミド(ピロリジン環4位エピマーの混合物) (50 mg)のジクロロメタン (2 mL)溶液に塩化メタンスルホニル (0.011 mL)及びTEA (0.039 mL)を加え、室温で30分間撹拌した。混合物を減圧下濃縮した。残渣にDMF (2 mL)、1-エチルピペラジン (0.024 mL)及び炭酸カリウム (86 mg)を加え、室温で20時間撹拌した。混合物に水及びEtOAcを加え、有機層を分離し、水及びbrineで洗浄した。有機層を無水硫酸ナトリウムで乾燥し、減圧下濃縮した。残渣をアミノシリカゲルカラムクロマトグラフィー(溶出溶媒;n-ヘキサン/クロロホルム、及びクロロホルム/MeOH)で精製し、N-(1-シアノシクロプロピル)-4-[{4-[(4-エチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}(ジフルオロ)メチル]-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンアミド(ピロリジン環4位エピマーの混合物) (52 mg)を固体として得た。
(4R)-N-(1-シアノシクロプロピル)-4-(ジフルオロ{4-[(4-メチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}メチル)-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンアミド (565.9 mg)のEtOAc (12 mL)及びエタノール (1.2 mL)溶液に塩化水素 (4M EtOAc溶液、 0.455 mL)を加え、室温で終夜撹拌した。析出物を濾取し、EtOAcで洗浄し、(4R)-N-(1-シアノシクロプロピル)-4-(ジフルオロ{4-[(4-メチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}メチル)-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンアミド二塩酸塩 (508 mg)を固体として得た。
N-(1-シアノシクロプロピル)-4-{ジフルオロ[2-フルオロ-4-(ピペリジン-4-イル)フェニル]メチル}-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンアミド(ピロリジン環4位エピマーの混合物) (32 mg)のジクロロエタン (1 mL)溶液に、アルゴンガス雰囲気下、氷冷下にて、酢酸 (0.010 mL)及びトリアセトキシ水素化ホウ素ナトリウム (50 mg)を加え、数分間撹拌した。混合物に氷冷下にて、アセトアルデヒド (0.030 mL)を加え、30分間撹拌した。混合物に、氷冷下にて、飽和炭酸水素ナトリウム水溶液を加え、有機層を分離し、brineで洗浄した。有機層を無水硫酸ナトリウムで乾燥し、減圧下濃縮した。残渣をアミノシリカゲルカラムクロマトグラフィー(溶出溶媒;n-ヘキサン/EtOAc)で精製し、N-(1-シアノシクロプロピル)-4-{[4-(1-エチルピペリジン-4-イル)-2-フルオロフェニル](ジフルオロ)メチル}-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンアミド(ピロリジン環4位エピマーの混合物) (27 mg)を固体として得た。
N-(1-シアノシクロプロピル)-4-{ジフルオロ[4-(ピペリジン-4-イル)-2-(トリフルオロメチル)フェニル]メチル}-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンアミド(ピロリジン環4位エピマーの混合物) (95 mg)のジクロロエタン (1.9 mL)溶液に、酢酸 (0.019 mL)及びアセトアルデヒド (0.045 mL)を室温で加え、5分間撹拌した。混合物に室温にて、トリアセトキシ水素化ホウ素ナトリウム (136 mg)を加え、2時間撹拌した。混合物に水及びクロロホルムを加え、有機層を分離し、brineで洗浄した。有機層をMgSO4で乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(溶出溶媒;クロロホルム/MeOH)で精製した。得られた油状物のジクロロメタン (2 mL)溶液に塩化水素 (4M 1,4-ジオキサン溶液、 0.025 mL)を加え、30分間撹拌した。混合物を減圧下濃縮し、N-(1-シアノシクロプロピル)-4-{[4-(1-エチルピペリジン-4-イル)-2-(トリフルオロメチル)フェニル](ジフルオロ)メチル}-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンアミド一塩酸塩(ピロリジン環4位エピマーの混合物) (60.6 mg)を固体として得た。
(4R)-N-(1-シアノシクロプロピル)-4-(ジフルオロ{4-[(4-メチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}メチル)-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンアミド (250 mg)の2-プロパノール (2.5 mL)溶液に室温で、コハク酸 (104 mg)を加えた。混合物を60℃で5分間撹拌した後、室温で終夜撹拌した。混合物に2-プロパノール (2.5 mL)を加え、10分間撹拌した。析出物を濾取し、少量の2-プロパノールで洗浄し、(4R)-N-(1-シアノシクロプロピル)-4-(ジフルオロ{4-[(4-メチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}メチル)-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンアミドとコハク酸を1:2で含有する結晶(本明細書中で、「二コハク酸塩」と記載する場合がある) (281 mg)を得た。
Claims (18)
- Lが-CH2-である、請求項1記載の化合物又はその塩。
- R2がハロゲノ低級アルキルであり、AがNであり、R3が低級アルキルであり、R4がHであり、R5がHであり、R6がHであり、R7がHである、請求項2記載の化合物又はその塩。
- 式(I)が式(Ib)である、請求項3記載の化合物又はその塩。
- (4R)-N-(1-シアノシクロプロピル)-4-(ジフルオロ{4-[(4-メチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}メチル)-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンアミド、
(4R)-N-(1-シアノシクロプロピル)-4-(ジフルオロ{4-[(4-メチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}メチル)-1-{[1-(トリフルオロメチル) シクロプロピル] カルボニル}-L-プロリンアミド、
(4R)-N-(1-シアノシクロプロピル)-4-[{4-[(4-エチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}(ジフルオロ)メチル]-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンアミド、
(4R)-N-(1-シアノシクロプロピル)-4-[{4-[(4-エチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}(ジフルオロ)メチル]-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンアミド、及びこれらの塩からなる群から選択される、請求項1に記載の化合物又はその塩。 - (4R)-N-(1-シアノシクロプロピル)-4-(ジフルオロ{4-[(4-メチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}メチル)-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンアミド又はその塩である、請求項7に記載の化合物又はその塩。

- (4R)-N-(1-シアノシクロプロピル)-4-[{4-[(4-エチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}(ジフルオロ)メチル]-1-{[1-(トリフルオロメチル)シクロプロピル]カルボニル}-L-プロリンアミド又はその塩である、請求項7に記載の化合物又はその塩。
- (4R)-N-(1-シアノシクロプロピル)-4-[{4-[(4-エチルピペラジン-1-イル)メチル]-2-(トリフルオロメチル)フェニル}(ジフルオロ)メチル]-1-[(1-メチルシクロプロピル)カルボニル]-L-プロリンアミド又はその塩である、請求項7に記載の化合物又はその塩。
- 請求項1に記載の化合物又はその塩、及び一以上の賦形剤を含有する医薬組成物。
- カテプシンS阻害剤である請求項12に記載の医薬組成物。
- SLEやループス腎炎を含む自己免疫疾患、アレルギー、又は、臓器、骨髄若しくは組織の移植片拒絶の予防もしくは治療用医薬組成物である請求項12に記載の医薬組成物。
- SLEやループス腎炎を含む自己免疫疾患、アレルギー、又は、臓器、骨髄若しくは組織の移植片拒絶の予防もしくは治療用医薬組成物の製造のための請求項1に記載の化合物又はその塩の使用。
- SLEやループス腎炎を含む自己免疫疾患、アレルギー、又は、臓器、骨髄若しくは組織の移植片拒絶の予防もしくは治療のための請求項1に記載の化合物又はその塩の使用。
- SLEやループス腎炎を含む自己免疫疾患、アレルギー、又は、臓器、骨髄若しくは組織の移植片拒絶の予防もしくは治療に用いるための請求項1に記載の化合物又はその塩。
