겨우살이는 스스로 광합성을 하며 부족한 영양분을 숙주식물로부터 얻는 반기생식물로 다양한 숙주나무에 기생하고 있다. 겨우살이 속의 식물들은 온 세계 전역에 분포하여 자라는 다년생의 반기생 식물로서 전 세계적으로 30속 1500종의 식물이 있는 것으로 알려져 있으며, 로란타세(Loranthaccae), 비스카세(Viscaccae) 및 에레몰레피다세(Eremolepidaccae)의 3개의 과로 나누어진다. 로란타세과에 속하며 특히 한국 등 아시아 지역에서 자라는 겨우살이를 비스컴 알붐 콜로라툼(Viscum album coloratum)이라 분류하며 우리나라 한방에서는 곡기생으로 알려져 있다. 본 발명에는 한국산 겨우살이가 사용되었다. 한국산 겨우살이는 유럽산 겨우살이와 효능과 성분에서 차이를 가지고 있으며 현재까지 항암 효능(대한민국 등록특허 10-0644381, 10-0659771), 항당뇨 효능(대한민국 등록특허 10-0547187), 항비만 효능(대한민국 등록특허 10-1325515), 운동수행능력 증가효능(대한민국 등록특허 10-1191081) 등이 밝혀져 있다.
겨우살이의 주요한 성분물질로는 겨우살이 렉틴 2형(mistletoe lectin type 2), 호모플라보야도리린 B(homoflavoyadorinin B), 클로로겐산(chlorogenic acid), 베툴린산(betulinic acid), 올레아놀산(oleanolic acid) 등이 있다. 이 중 베툴린산과 올레아놀산은 오환성 트리테르페노이드 (pentacyclic triterpenoid)의 일종으로 최근 근위축 억제 효과를 가지고 있는 것으로 밝혀진 우르솔 산(ursolic acid)과 구조적으로 유사하다. 우르솔 산(ursolic acid) 역시 오환성 트리테르페노이드 (pentacyclic triterpenoid)의 구조를 가지고 있으며 일찍이 사과 등의 껍질에 존재하는 것으로 알려져 있다. 최근 미국 아이오와 대학 크리스토퍼 애덤스 (Christopher Adams) 박사 연구팀은 우르솔 산이 근육 형성에 중요한 역할을 하는 두 가지 호르몬인 인슐린과 인슐린 유사성장인자-1(IGF-1)을 활성화시킴을 발표하였다. 연구팀은 질병 또는 노화에 의해 근육이 쇠약해지는 근위축 현상을 유발하는 유전자변화를 억제하는 물질을 찾아본 결과 우르솔 산의 근 위축 억제 효과가 있음을 확인하였으며 동물 모델을 통해 이를 증명하였다.
근 위축은 다양한 원인에 의해서 발생하는 근육의 손실 현상을 일컫는 용어로 근육을 사용할 수 없거나 사용하지 않는 현상에 의해 ubiquitin-proteasome pathway에 의해 단백질 분해가 가속화되는 결과를 말한다. 일반적으로 노화에 따라 근 위축은 진행이 되며 장기 입원 환자 등의 경우와 같이 근육을 사용하지 못할 때 근육의 양, 크기, 근력, 기능의 전반적인 감소를 가져온다. 노화에 의한 자연스런 근 감소 외에 각종 질병, 대사성 질환, 흡연, 지나친 음주 등에 의해 근 감소가 가속될 수 있다. 노화에 따라 근 위축이 오면 무능력해지고 다른 사람의 수발이 필요하게 될 수도 있다.
근위축증 가운데 많이 알려진 형태로는 근위축성 측삭경화증과 척수성 진행성 근위축증이 있다. 두 가지 형태 모두 척수에 있는 운동신경섬유 및 세포의 진행성 병성에 의한 것으로 알려져 있지만 정확한 원인은 아직 불분명하다. 근위축성 측삭경화증은 19세기 말 유럽에서 처음 보고되었으며 아직까지 그 원인이 밝혀지지 않은 불치병이다. 스티븐 호킹 박사가 이 병을 앓고 있으며 미국의 야구선수 L. 게릭이 이 질병으로 사망하여 ‘루게릭병’이라고 부른다. 이 병은 척수신경이나 간뇌의 운동세포가 서서히 파괴됨으로 인하여 근육이 위축되어 힘을 쓰지 못하게 된다. 척수성 진행성 근위축병은 척수 전각세포의 변성이 천천히 진행된다.
근육 파괴 단백질이라고 불리는 atrogin-1과 MuRF-1은 그 발현에 의해서 근육을 이루는 단백질을 분해함으로써 근육의 양이 줄어들고 위축되게 하는 역할을 한다. 이와 반대로 PGC-1은 골격근의 손상과 이상을 예방하는 단백질로서 PGC-1의 발현이 증가하면 근 위축을 억제할 수 있다.
본 발명과 관련된 선행기술로는 대한민국 등록특허 10-0923217호에 겨우살이 추출물 또는 이로부터 분리된 활성성분을 함유하는 신경병증성 통증의 예방 및 치료용 조성물에 관한 것으로 현재까지 근 위축 및 근 비대 활성을 갖는 한국산 겨우살이 추출물에 대하여는 공지된 바 없다.
따라서, 본 발명의 목적은 근 위축을 유도하는 atrogin-1과 MuRF-1을 감소시켜 근 위축을 억제하는 뛰어난 효과가 있는 한국산 겨우살이 잎, 줄기, 눈 및 열매로부터 물 추출하여 수득한 추출물을 유효성분으로 함유하는 근 위축억제 및 근 비대 활성용 조성물을 제공하는 데 있다.
본 발명의 상기 목적은 12월에서 2월 사이에 채취한 1 ~ 2년생 겨우살이의 잎, 줄기 및 열매를 증류수에서 세척하는 단계와; 상기 단계에서 얻은 겨우살이를 정제수를 가한 후 100℃에서 추출하는 단계와; 상기 단계에서 얻은 추출물을 다시 정제수를 가하여 2차 추출하는 단계와; 상기 단계에서 얻은 추출물을 50~60℃에서 진공 농축하는 단계와; 상기 단계에서 얻은 농축물을 여과 과정을 통해 이물질을 제거한 후 분무건조하여 본 발명 겨우살이 추출물을 수득하는 단계를 통해 이루어지고 상기에서 얻은 추출물을 공시재료로 근 위축과 관련된 유전자외 발현억제 실험 결과를 평가함으로써 달성하였다.
본 발명은 겨우살이 수추출물을 제공하는 효과가 있고 이를 통하여 근 위축과 관련된 유전자들의 발현을 억제시킴으로써 근 위축을 억제하는 뛰어난 효과가 있다.

본 발명은 근 위축 억제 및 근 비대 활성을 갖는 한국산 겨우살이 추출물을 제공한다.
본 발명에 따르면 한국산 겨우살이 추출물(KMH) 실험 결과를 도면에 일일이 제시하지는 않았으나, 기존의 근위축 억제 효과가 있는 것으로 알려진 우르솔 산(UA)과 비교 할 수 없을 정도로 탁월하였다.
본 발명에서 사용한 겨우살이는 강원도 일대에서 참나무를 숙주로 하여 자생하는 한국산 겨우살이를 12월에서 2월 사이에 채취한 것이 본 발명에 따른 효과에 가장 바람직하였다.
또, 본 발명에 따르면 상기 겨우살이는 잎, 줄기 및 열매를 취하여 증류수로 세척 후 응달에서 건조 한 후 -20℃에서 냉동 보관하면서 사용하는것이 바람직하였다.
또, 본 발명에 따라 상기 건조된 겨우살이 100kg을 정제수 7배를 가한 후 100℃에서 추출한 후 2차로 4배의 정제수를 가해 추출한 후, 바람직하게는 50~60℃에서 농축하고 더욱 바람직하게는 55℃에서 농축하여 30kg을 수득하였다.
본 발명에 따라 본 발명자들은 한국산 겨우살이 추출물을 사용한 동물모델을 통하여 신경절단에 의한 근 위축의 억제와 장기투여에 의한 근 비대효과를 최초로 확인하였다.
본 발명에 따르면 상기 겨우살이 추출물은 일반적으로 천연 식물에서 추출물을 수득할 때 사용되는 추출법이라면 어떠한 방법을 사용하여도 무방하다. 그러나, 유기용매 추출의 경우 그 후속절차들에 의하여 본원 발명의 목적을 달성하는 데 수반되는 경제적 비용과 기술적인 문제적 및 효과가 좋지 않아 실험 결과는 여기에 제시하는 것을 생략하였고, 열수 추출법, 냉수 추출법 또는 이들의 혼용법 중 어느 하나의 수추출법을 사용하는 것이 바람직하였다.
본 발명에 따른 상기 추출물은 음료, 차 및 선식 중 어느 하나의 제형인 것이 특징인 식품조성물을 제공한다.
또, 본 발명에 따른 상기 추출물은 정제, 환제, 산제, 과립제, 캅셀제, 현탁제, 유제, 시럽제, 비수성용제, 현탁제 및 동결건조제제로 이루어진 군으로부터 선택되는 어느 하나의 제형을 가지는 것이 특징인 약학적 조성물을 제공한다.
본 발명의 상기 약학적 조성물은 약제학적 조성물 형태로 제조할 수 있으며, 하나 또는 그 이상의 무독성 및 약제학적으로 허용 가능한 담체, 부형제, 보조제 또는 희석제 또는 다른 활성성분을 추가로 더 포함할 수 있다.
본 발명은 하기의 구체적인 실시예, 실험예 및 제조예에 의해 보다 구체적으로 설명되나, 하기 실시예, 실험예 및 제조예는 본 발명을 예시하기 위한 것으로 본 발명의 권리범위가 이에 한정하는 것으로 의도되지는 않는다.
본 발명의 제조예1 내지 제조예3에는 가장 바람직하게는 겨우살이의 수추출물 함유량의 수치를 1중량%로 한정하였으나 0.5~2.5중량%까지 첨가하여도 그 효능에는 무방하였다.
실시예 1. 한국산 겨우살이(Korean Viscum album Coloratum) 전처리
강원도 일대에서 참나무를 숙주로 하여 자생하는 한국산 겨우살이를 12월에서 2월 사이에 1, 2년생을 잎 및 줄기를 채취하여 증류수에서 가장 바람직하게는 3회까지 세척 후 응달에서 건조하고 진공포장하여 추출시까지 -20℃에서 냉동보관하였다.
실시예 2. 한국산 겨우살이 물 추출(Water extract)
상기 실시예1의 한국산 겨우살이 건조물 100kg을 추출탱크에 넣고 7배의 정제수를 가한 후 100℃에서 추출한 후 다시 4배의 정제수를 가하여 2차 추출하고 여과 과정을 통해 이물질을 제거한 후 50~60℃에서 진공 농축하여 120L의 농축물을 수득하였다. 다시 여과 과정을 통해 이물질을 제거한 후 분무건조하여 수득한 분말 30kg을 얻고 밀봉포장하여 냉장 보관하고 공시재료로 사용하였다.
실험예1. 본 발명 한국산 겨우살이 수추출물의 마우스 근육 세포에서 근 위축 억제 효과
실험예1-1 본 발명 한국산 겨우살이 추출물의 Akt 신호 전달 경로 활성화
본 발명 한국산 겨우살이 추출물의 근 위축 억제 효과를 세포 단계에서 확인하기 위해 상기 실시예2에서 수득한 수추출물을 마우스 근육 세포인 C2C12 세포에 100ug/mL의 농도로 처리하였다.
실험 결과, 도1에서 알 수 있듯이 한국산 겨우살이의 수추출물을 처리한 그룹에서 비처리구에 비해 Akt의 인산화가 증가하는 것을 확인하였고 이를 통해 겨우살이 수추출물이 Akt 신호 전달 경로를 활성화하여 근 질량과 관련의 조절하는 효과를 확인하였다.
실험예1-2 본 발명 한국산 겨우살이 추출물의 근 위축 억제 효과
Erk는 MAPK 단백질의 아과(subfamily)로 Akt/mTOR 신호전달 경로를 통한 단백질 합성을 촉진하여 근육의 비대와 관련이 있고, FoxO는 단백질 분해를 활성화하는 근 위축의 주요한 물질이다. FoxO가 E3 ubiquitin ligase인 Murf1 유전자와 Atrogin-1을 촉진시켜 근 위축을 유도한다. 본 발명 한국산 겨우살이 수추출물의 근 위축 신호전달과정을 확인하기 위해 상기 실시예2에서 수득한 한국산 겨우살이 수추출물과 양성대조군인 ursolic acid를 마우스 근육 세포인 C2C12 세포에 100ug/mL의 농도로 처리하여 단백질의 변화를 확인하였다.
실험 결과, 도2 내지 3에서 알 수 있듯이 본 발명 한국산 겨우살이 수추출물은 대조군보다 FoxO 및 Erk의 인산화를 더 촉진함을 확인하였다.
실험예1-3 본 발명 한국산 겨우살이 수추출물의 근 위축 억제 효과
Atrogene이라 불리는 Atrogin-1과 Murf1 유전자는 근 위축을 유도하며 PGC-1α, GLUT4, SREBP-1c 등에 의해 조절되는 것으로 알려져 있다. 본 발명 한국산 겨우살이 수추출물이 근육 세포에서 Atrogin-1과 Murf1 유전자의 발현에 관여하는지 확인하기 위해 상기 실시예2에서 수득한 한국산 겨우살이 수추출물(KMH)을 마우스 근육 세포인 C2C12 세포에 100ug/mL의 농도로 처리한 후 16시간 배양하고 Real-time PCR로 mRNA의 변화를 확인하였다.
실험 결과, 도4 내지 5에서 알 수 있듯이 본 발명 한국산 겨우살이 수추출물은 Atrogin-1 및 Murf1의 발현을 감소시킴을 확인하였다.
실험예2. 본 발명 한국산 겨우살이 수추출물의 신경절단 마우스 모델을 이용한 근 위축 억제 효과
본 발명 한국산 겨우살이 수추출물의 근 위축 억제 효과를 확인하기 위하여 상기 실시예2에서 수득한 추출물을 신경절단 마우스 모델을 이용하여 실험하였다.
이를 위해 6주령 ICR 마우스의 오른쪽 뒷다리의 궁둥신경(sciatic nerve)를 절단한 후 본 발명 시료를 200mg/kg과 500mg/kg의 농도로 각각 2주간 하루 두 번씩 경구로 섭취시켰고, 2주 후 희생(sacrifice)하여 대퇴사두근(quadriceps)을 적출하여 무게를 측정하고 실험시까지 -80℃에 보관하였다.
실험 결과, 도6에서 알 수 있듯이 각 그룹간 대퇴사두근 무게는 본 발명 겨우살이 추출물 투여에 따른 근육 무게가 신경절단을 하지 않은 대조군과 유사할 정도로 회복되었음을 보아 근육이 위축되는 것을 억제함을 확인하였다.
실험예3. 본 발명 한국산 겨우살이 수추출물의 근 위축 억제 유전자 발현량 확인
상기 실시예2에서 수득한 한국산 겨우살이 수추출물의 근 위축 억제 유전자 발현량 확인을 위하여 신경절단 마우스 모델에서 적출한 대퇴사두근에서 RNA를 추출하여 근 위축 억제 유전자로 알려진 atrogin-1과 MuRF-1의 발현을 확인하였다.
실험 결과, 도7에 알 수 있듯이 저용량의 본 발명 시료 투여시 atrogin-1의 발현량이 PBS를 투여한 대조군에 비해 현저히 감소함을 확인할 수 있었다. 이러한 결과를 통해 근 위축을 유도하는 atrogin-1의 발현이 본 발명 한국산 겨우살이 수추출물(KMH) 투여에 억제됨을 확인하였다.
실험예4. 마우스 모델을 이용한 본 발명 한국산 겨우살이 수추출물의 근 비대 효과
본 발명 한국산 겨우살이 수추출물의 근 비대 효과를 확인하기 위하여 상기 실시예2에서 수득한 본 발명 시료를 마우스 모델에 투여하여 실험하였다.
이를 위해 6주령 ICR 마우스에 본 발명 시료 0.3% 및 1.5%가 함유된 사료를 4주간 자유급여 하였다. 4주 후, 전신 악력 테스트(whole body grip strength test)를 통해 악력을 확인한 후 체중을 확인하고 희생(sacrifice) 하여 대퇴사두근(quadriceps)을 적출하여 무게를 확인한 후 실험시까지 -80℃에 보관하였다.
실험 결과, 도8에서 알 수 있듯이 본 발명 시료의 고용량 섭취군에서 악력이 증가함을 확인하였다.
또, 도9에서 알 수 있듯이 본 발명 한국산 겨우살이 수추출물 섭취군에서 대조군에 비해 근육의 무게가 증가함을 확인하였으며 이는 한국산 겨우살이 수추출물이 근 위축뿐만 아니라 근 비대에도 영향을 나타내었음이 확인되었다.
실험예5. 근육 관련 유전자 발현량 확인 실험
상기 실시예2에서 수득한 추출물의 근육 관련 유전자 발현량 확인을 위하여 Sirt1의 기질 중 하나인 PGC-1α는 미토콘드리아가 형성되는데 주 조절자 역할을 하는 근육 관련 유전자인 PGC-1α 및 Sirt1을 대퇴사두근에서 추출한 RNA를 토대로 확인하였다. Sirt1의 기질 중 하나인 PGC-1α는 미토콘드리아가 형성되는데 주 조절자 역할을 한다.
실험 결과, 도10 내지 11에서 알 수 있듯이 상호연결된 근육 관련 유전자인 PGC-1α와 Sirt1 모두 본 발명 시료를 급여한 그룹에서 증가함을 확인하였으며 본 발명 한국산 겨우살이 수추출물은 미토콘드리아를 형성 촉진하는데 영향을 주는 것이 확인되었다.
실험예6. PI3K/Akt 경로 확인 실험
상기 실시예2에서 수득한 한국산 겨우살이 수추출물(KMH)이 근 비대에 미치는 영향을 확인하기 위해 PI3K/Akt 경로의 활성화를 확인하였다. PI3K/Akt 경로는 근 비대와 관련되어 잘 알려진 신호전달 경로로서 Akt 가 활성화되면 그 하위 신호인 mTOR가 활성화되면 이에 따라 S6K가 활성화된다. 활성화된 S6K는 단백질 합성을 자극하여 근 비대를 유도한다. Erk는 PI3K/Akt 경로의 또 다른 사이드 체인으로서 이 또한 근 비대 및 근 위축과 관련되어 있다.
실험 결과, 도12 내지 15에서 알 수 있듯이 PI3K/Akt 경로를 따라 Akt, mTOR, S6K가 모두 활성화되고 Erk 또한 활성화되어 본 발명 시료의 수추출물 투여가 근 비대와 관련된 신호 전달 경로를 따라 활성화하는 것을 확인하였다.
이하에서는 본 발명 한국산 겨우살이 수추출물 시료를 함유한 음료, 차 및 선식의 제형의 배합비를 나타낸 식품 제조예를 보인 것이다. 제조 실시예에 따르면 적어도 0.5중량% 수준 이상이면 본 발명 실험예 1~6을 충족시킬 수 있었다.

- 한국산 겨우살이(Viscum album var. coloratum)의 잎, 줄기 및 열매 중 하나 이상의 재료를 증류수에서 세척하는 단계와; 상기 단계에서 얻은 겨우살이 시료에 7배의 정제수를 가한 후 수추출하는 단계와; 상기 단계에서 얻은 수추출물을 진공 농축하는 단계와; 상기 단계에서 얻은 농축물을 여과 과정을 통해 이물질을 제거한 다음 분무건조하는 단계로 이루어진 한국산 겨우살이 추출물의 제조방법
- 제 1항의 방법에 있어서, 상기 수추출 단계 후에 다시 4배의 정재수를 가하여 2차 추출하는 단계를 더 포함하는 것이 특징인 방법.
