Abstract
본 발명은 자폐 범주성 장애의 예방 또는 치료 효과를 가지는 아가토바쿨룸 속 균주 및 이의 용도에 관한 것으로, 본 발명의 장내 미생물 아가토바쿨룸 속 균주는 VPA(valproic acid) 유도성 자폐증 모델 마우스 및 BTBR 자폐증 모델 마우스에 경구투여한 결과, 비처리 대조구에 비해 마우스의 반복적 행동, 과활동성, 사회성 결핍 및 인지-기억 기능 장애 행동을 포함한 자폐 범주성 장애를 현저히 개선시키는 효과를 나타내므로, 자폐 범주성 장애의 예방이나 치료를 위한 식품, 의약품 또는 사료로 유용하게 사용될 수 있어, 관련 산업에 매우 유용하다.
Description
본 발명은 아가토바쿨룸 속 균주를 유효성분으로 함유하는 자폐 범주성 장애의 예방, 개선 또는 치료용 조성물에 관한 것이다.
자폐증은 사회적 상호작용의 장애, 상동적 행동 및 관심 범위의 제한, 언어적, 비언어적 의사소통의 장애를 주 증상으로 하는 뇌 발달 장애이다. 자폐증은 보통 3세 이하에 시작되고 진단될 수 있으며 발달의 광범위한 범주에 영향을 주는 질환이므로 대체로 일생 동안 기능의 이상이 계속되어 환자 본인과 가족에게 심각한 고통을 초래하는 질환으로 의학적 및 사회적 손실비용이 매우 큰 질환이다. 최근에는 자폐의 정도와 예후가 매우 다양함을 강조하여 과거 사용되던 자폐증의 진단명을 자폐 범주성 장애(Autism Spectrum Disorder, ASD)로 수정한 용어를 사용한다. 자폐 범주성 장애(ASD)에는 자폐증을 비롯하여 아스퍼거 증후군, Rett's Disorder, 아동기 붕괴성 장애, 달리 분류되지 않는 전반적 발달장애(PDD NOS) 등을 포함하는 전반적 발달장애(pervasive developmental disorder, PDD)이 포함된다.
자폐증은 최근 약 20여년 간에 걸쳐 그 유병률이 급격히 증가하고 있는 질환으로 2012년 미국 질병 관리국 CDC 보고에 의하면 자폐증의 유병률은 100명당 1명을 이미 넘어선 것으로 확인되었으며, 특히 한국의 아동들을 대상으로 한 역학 연구에서는 유병율이 매우 높아, 7~12세 아동의 2.64%가 자폐증 또는 기타 자폐 범주성 장애를 갖고 있는 것으로 보고되었다 (Kim et al., Am J Psychiatry 2011 168:904-912).
최근 연구 결과들은 장내 미생물이 사람의 장 건강 및 장 질환은 물론이고 뇌기능 조절 및 뇌질환의 발병 등에 적극적으로 영향을 주는 것이 밝혀지고 있다. 그러나 장내 미생물인 신규한 아가토바쿨룸 부트리시프로두센스(Agathobaculum butyriciproducens) SR79 균주에 의한 자폐 범주성 장애의 예방 또는 치료효과에 대한 영향은 알려진 바 없다.
한편, 한국공개특허 제2004-0074053호에서는 '새로운 젖산균 균주, 이를 함유하는 식용 조성물, 약물 및 수의학적 제품'에 대해 개시하고 있고, 한국공개특허 제2017-0086492호에서는 '신경발달 장애에서의 행동을 개선시키기 위한 박테리아를 포함하는 조성물 및 방법'에 대해 개시하고 있으나, 본 발명에서와 같이, '자폐 범주성 장애의 예방 또는 치료 효과를 가지는 아가토바쿨룸 속 균주 및 이의 용도'에 대해서는 밝혀진 바가 전혀 없다.
본 발명은 상기와 같은 요구에 의해 도출된 것으로서, 본 발명에서 분리한 아가토바쿨룸(Agathobaculum sp.) 속 균주를 VPA(valproic acid) 유도성 자폐증 모델 마우스에 경구투여한 결과, 비처리 대조구에 비해 마우스의 반복적 행동, 과활동성, 사회성 결핍 및 인지-기억 기능 장애 행동을 포함한 자폐 범주성 장애를 현저히 개선시키는 효과를 확인함으로써, 본 발명을 완성하였다.
상기 과제를 해결하기 위해, 본 발명은 아가토바쿨룸 속(Agathobaculum sp.) 균주, 상기 균주 유래 소포체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상을 유효성분으로 함유하는 자폐 범주성 장애의 예방 또는 치료용 약학 조성물을 제공한다.
또한, 본 발명은 아가토바쿨룸 속(Agathobaculum sp.) 균주, 상기 균주 유래 소포체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상을 유효성분으로 함유하는 자폐 범주성 장애의 예방 또는 개선용 건강기능식품 조성물을 제공한다.
또한, 본 발명은 아가토바쿨룸 속(Agathobaculum sp.) 균주, 상기 균주 유래 소포체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상을 유효성분으로 함유하는 자폐 범주성 장애의 예방 또는 개선용 사료첨가제 조성물을 제공한다.
또한, 본 발명은 아가토바쿨룸 속(Agathobaculum sp.) 균주, 상기 균주 유래 소포체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상을 유효성분으로 함유하는 자폐 범주성 장애의 예방 또는 치료용 동물의약품 조성물을 제공한다.
본 발명의 장내 미생물 아가토바쿨룸 속 균주는 VPA(valproic acid) 유도성 자폐증 모델 마우스 및 BTBR 자폐증 모델 마우스에 경구투여한 결과, 비처리 대조구에 비해 마우스의 반복적 행동, 과활동성, 사회성 결핍 및 인지-기억 기능 장애 행동을 포함한 자폐 범주성 장애를 현저히 개선시키는 효과를 나타내므로, 자폐 범주성 장애의 예방이나 치료를 위한 식품, 의약품 또는 사료로 유용하게 사용될 수 있어, 관련 산업에 매우 유용하다.
도 1은 본 발명에서 분리한 아가토바쿨룸 속 SR79 균주 처리에 따른 모델 마우스의 반복적인 행동(A) 및 과활동성(B) 대한 행동을 분석한 결과이다. VPA는 발프로산(valproic acid) 처리를 의미하고, Vehicle는 비처리 대조구이고, SR은 아가토바쿨룸 속 SR79 균주 처리를 의미한다.
도 2는 본 발명에서 분리한 아가토바쿨룸 속 SR79 균주 처리에 따른 모델 마우스의 사회성 행동의 개선 효과를 측정하기 위해 다른 마우스와의 상호작용 행동(sniffing behavior)을 관찰하여 시간을 측정한 결과이다.
도 3은 본 발명에서 분리한 아가토바쿨룸 속 SR79 균주 처리에 따른 모델 마우스의 사회성 행동의 개선 효과를 측정하기 위해 BTBR 마우스를 이용하여 다른 마우스와의 상호작용 행동(sniffing behavior)을 관찰하여 시간을 측정한 결과이다.

본 발명의 목적을 달성하기 위하여, 본 발명은 아가토바쿨룸 속(Agathobaculum sp.) 균주, 상기 균주 유래 소포체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상을 유효성분으로 함유하는 자폐 범주성 장애의 예방 또는 치료용 약학 조성물을 제공한다.
본 발명의 일 구현 예에 따른 균주에서, 상기 아가토바쿨룸 속 균주는 아가토바쿨룸 부트리시프로두센스(Agathobaculum butyriciproducens)일 수 있고, 바람직하게는 상기 아가토바쿨룸 부트리시프로두센스는 기탁번호가 KCTC 13036BP인 아가토바쿨룸 부트리시프로두센스 SR79 균주일 수 있으나, 이에 제한되지 않는다.
본 발명에 따른 상기 아가토바쿨룸 부트리시프로두센스(Agathobaculum butyriciproducens) SR79 균주는 본 발명자가 건강한 한국인 장내에서 직접 분리한 절대혐기성 미생물로서 기존에 보고된 표준균주와는 다른 새로운 속(Genus) 및 종(Species)으로 분류되는 신규 미생물임을 분자생물학적, 생리/생화학적 미생물 동정방법으로 확인하였고, 이의 미생물명을 아가토바쿨룸 부트리시프로두센스(Agathobaculum butyriciproducens) SR79로 제안하였는데, 본 발명에서 분리한 SR79 균주에 대해서는 본 발명자가 제안한 새로운 학명인 아가토바쿨룸 부트리시프로두센스(Agathobaculum butyriciproducens)에 대한 속 및 종에 대한 표준균주로 국제저널 (International Journal of Systematic and Evolutionary Microbiology)에서 수락하였다.
또한, 상기 아가토바쿨룸 부트리시프로두센스(Agathobaculum butyriciproducens) SR79 균주를 한국생명공학연구원에 기탁번호가 KCTC 13036BP로 2016년 6월 7일자로 기탁하였다.
신규 분리 미생물 균주인 아가토바쿨룸 부트리시프로두센스(Agathobaculum butyriciproducens) SR79 균주는 자폐 범주성 장애 관련 질환 등의 예방 또는 치료의 용도에 대해 개시된 바는 없으며, 본 발명자들에 의해 최초로 자폐 범주성 장애 관련 질환의 예방 또는 치료의 용도가 있음이 규명되었다.
본 발명에서는 자폐 범주성 장애와 주의력결핍 과잉행동장애 행동 표현형을 보이는 발프로산 유도 마우스 모델에서 SR79 균주의 경구 투여가 마우스의 반복적 행동(도 1A)와 과활동성(도 1B), 사회성 결핍(도 2 및 도 3), 인지-기억 기능 장애(도 4 및 도 5) 행동을 현저히 개선시키는 효능이 있음을 확인하였다. 이러한 결과들을 바탕으로 SR79 균주가 자폐 범주성 장애와 주의력결핍 과잉행동장애 등 신경발달 질환의 예방 및 치료 효과를 가지는 것을 확인하였다.
본 발명의 "자폐 범주성 장애"는 자폐증을 비롯하여 아스퍼거 증후군, Rett's Disorder, 아동기 붕괴성 장애, 달리 분류되지 않는 전반적 발달장애(PDD NOS) 등을 포함하는 전반적 발달장애(pervasive developmental disorder, PDD)를 포함하는 것으로, 일반적으로 자폐 범주성 장애의 증상으로서 보고된 증상이라면 어느 것이든 포함할 수 있으며, 바람직하게는 과잉행동 증상, 사회성 결여 증상, 인지-기억 기능 장애 행동 및 간질성 경련 증상으로 이루어진 군으로부터 선택되는 어느 하나 이상의 증상을 포함할 수 있으나, 이에 제한되지 않는다.
본 발명에서의 용어 "예방"이란 본 발명에 따른 약학적 조성물의 투여에 의해 자폐 범주성 장애의 발병을 억제 또는 지연시키는 모든 행위를 의미하고, "치료"란 상기 약학적 조성물의 투여에 의해 자폐 범주성 장애의 의심 및 발병 개체의 증상이 호전되거나 이롭게 변경되는 모든 행위를 의미한다.
본 발명의 상기 약학조성물은 약학적 조성물의 제조에 통상적으로 사용하는 적절한 담체, 부형제 및 희석제를 더 포함할 수 있다.
본 발명에 따른 상기 약학조성물은, 각각 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있다. 본 발명에 따른 상기 약학조성물에 포함될 수 있는 담체, 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유 등을 포함한 다양한 화합물 혹은 혼합물을 들 수 있다. 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다. 경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 상기 균주 또는 상기 균주 유래 소포체에 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘카보네이트(calcium carbonate), 수크로스(sucrose) 또는 락토오스(lactose), 젤라틴 등을 섞어 조제된다. 또한 단순한 부형제 이외에 마그네슘 스테아레이트, 탈크 같은 윤활제들도 사용된다. 경구를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 비수성용제, 현탁제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로젤라틴 등이 사용될 수 있다.
본 발명의 약학적 조성물은 약제학적으로 유효한 양으로 투여될 수 있는데, 본 발명의 용어 "약제학적으로 유효한 양"이란 의학적 예방 또는 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 예방 또는 치료하기에 충분한 양을 의미하며, 유효 용량 수준은 질환의 중증도, 약물의 활성, 환자의 연령, 체중, 건강, 성별, 환자의 약물에 대한 민감도, 사용된 본 발명 조성물의 투여 시간, 투여 경로 및 배출 비율, 치료기간, 사용된 본 발명의 조성물과 배합 또는 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 약학적 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적으로 또는 동시에 투여될 수 있다. 그리고 단일 또는 다중 투여될 수 있다. 상기 요소를 모두 고려하여 부작용없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하다.
본 발명의 약학적 조성물의 투여량은 예를 들어, 본 발명의 약학 조성물을 사람을 포함하는 포유동물에 하루 동안 1.0Х109 CFU로 투여하는 것이 좋으나, 이에 제한되지 않는다. 본 발명의 조성물의 투여 빈도는 특별히 이에 제한되지 않으나, 1일 1회 투여하거나 또는 용량을 분할하여 수회 투여할 수 있다. 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다.
상기 목적을 달성하기 위한 또 다른 실시양태로서, 본 발명은 상기 약학적 조성물을 약제학적으로 유효한 양으로 자폐 범주성 장애가 발병될 가능성이 있거나 또는 발병된 개체에 투여하는 단계를 포함하는 자폐 범주성 장애의 예방 또는 치료방법을 제공한다.
상술한 바와 같이, 본 발명에서 제공하는 상기 장내 미생물 SR79 균주, 상기 균주 유래 소포체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상은 자폐 범주성 장애의 예방 또는 치료용 약학적 조성물의 유효성분으로서 사용할 수 있으므로, 상기 조성물은 자폐 범주성 장애를 예방 또는 치료하는데 사용될 수 있다.
본 발명의 용어 "개체"란 자폐 범주성 장애가 발병될 가능성이 있거나 또는 발병된 쥐, 가축, 인간 등을 포함하는 포유동물을 제한 없이 포함한다.
본 발명의 자폐 범주성 장애를 치료하는 방법에 있어서, 상기 약학적 조성물의 투여 경로는 목적 조직에 도달할 수 있는 한 어떠한 일반적인 경로를 통하여도 투여될 수 있다. 본 발명의 약학적 조성물은 특별히 이에 제한되지 않으나, 경구 투여, 직장내 투여 등의 경로를 통해 투여할 수 있으며 경우에 따라 목적에 따라 다른 경로로도 투여될 수 있다. 다만, 경구 투여 시에는 위산에 의하여 상기 장내 미생물 SR79 균주가 변성될 수 있기 때문에 경구용 조성물은 활성 약제를 코팅하거나 위에서의 분해로부터 보호되도록 제형화 되어야 한다. 또한, 상기 조성물은 활성 물질이 표적 세포로 이동할 수 있는 임의의 장치에 의해 투여될 수 있다.
또한, 본 발명은 상기 균주, 상기 균주 유래 소포체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상을 유효성분으로 함유하는 자폐 범주성 장애의 예방 또는 개선용 건강기능식품 조성물을 제공한다.
본 발명의 일 구현 예에 따른 건강기능식품 조성물에서, 상기 아가토바쿨룸 속 균주는 아가토바쿨룸 부트리시프로두센스(Agathobaculum butyriciproducens)일 수 있고, 바람직하게는 상기 아가토바쿨룸 부트리시프로두센스는 기탁번호가 KCTC 13036BP인 아가토바쿨룸 부트리시프로두센스 SR79 균주일 수 있으나, 이에 제한되지 않는다.
본 발명의 일 구현 예에 따른 건강기능식품 조성물에서, 상기 "자폐 범주성 장애"는 자폐증을 비롯하여 아스퍼거 증후군, Rett's Disorder, 아동기 붕괴성 장애, 달리 분류되지 않는 전반적 발달장애(PDD NOS) 등을 포함하는 전반적 발달장애(pervasive developmental disorder, PDD)를 포함하는 것으로, 일반적으로 자폐 범주성 장애의 증상으로서 보고된 증상이라면 어느 것이든 포함할 수 있으며, 바람직하게는 과잉행동 증상, 사회성 결여 증상, 인지-기억 기능 장애 행동 및 간질성 경련 증상으로 이루어진 군으로부터 선택되는 어느 하나 이상의 증상을 포함할 수 있으나, 이에 제한되지 않는다.
상기 균주에 대해서는 전술한 바와 같으며, 자폐 범주성 장애의 예방 또는 개선을 목적으로 건강기능식품에 첨가할 수 있다.
상기에서 언급한 본 발명의 상기 균주 또는 이의 배양액 등을 건강기능식품 조성물로 사용할 경우, 상기 균주 또는 이의 배양액을 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용할 수 있으며, 통상적인 방법에 따라 적절하게 사용될 수 있다. 유효 성분의 혼합양은 그 사용 목적 (예방, 건강 또는 치료적 처치)에 따라 적합하게 결정될 수 있다.
본 발명의 식품(또는 건강기능식품)은, 식품 제조 시에 통상적으로 첨가되고 식품학적으로 허용되는 성분을 더 포함할 수 있다. 바람직하게는 상기 유효성분 이외에 추가로, 영양제, 비타민, 전해질, 풍미제, 착색제, 증진제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올 및 탄산음료에 사용되는 탄산화제 중에서 선택된 하나 이상을 더 함유할 수 있으나, 이에 제한되지 않는다. 예컨대, 음료수로 제조되는 경우에는 본 발명의 균주 이외에 구연산, 액상과당, 설탕, 포도당, 초산, 사과산, 과즙 등에서 하나 이상의 성분을 추가로 포함시킬 수 있다.
본 발명에 따른 식품(또는 건강기능식품)의 유효성분으로 포함될 수 있는 양은 자폐 범주성 장애의 예방 또는 개선용 식품을 원하는 사람의 연령, 성별, 체중, 상태, 질병의 증상에 따라 적절히 선택될 수 있으며, 바람직하게는 성인기준 1일 0.01 g 내지 10.0 g 정도로 포함되는 것이 좋으며, 이러한 함량을 갖는 식품을 섭취함으로써 자폐 범주성 장애의 예방 또는 개선 효과를 얻을 수 있다.
또한, 본 발명은 상기 균주를 배양하는 단계를 포함하는 자폐 범주성 장애의 예방 또는 치료용 미생물 제제를 제조하는 방법을 제공한다.
본 발명의 균주를 배양하는 방법은 당업계에 통상적으로 이용되는 방법에 따라 배양할 수 있다.
본 발명의 자폐 범주성 장애의 예방 또는 치료용 미생물 제제는 유효성분으로서 아가토바쿨룸 속(Agathobaculum sp.) 균주, 바람직하게는 아가토바쿨룸 부트리시프로두센스(Agathobaculum butyriciproducens) 균주, 가장 바람직하게는 기탁번호가 KCTC 13036BP인 아가토바쿨룸 부트리시프로두센스 SR79 균주를 이용하여 제조될 수 있다. 본 발명에 따른 자폐 범주성 장애의 예방 또는 치료용 미생물 제제는 용액, 분말, 현탁액, 분산액, 에멀젼, 유성 분산액, 페이스트, 분진, 분사제 또는 과립제로 제조할 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 또 다른 하나의 양태는, 상기 균주, 상기 균주 유래 소포체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상을 포함하는 자폐 범주성 장애의 예방 또는 개선용 사료첨가제 조성물을 제공한다.
상기 균주에 대해서는 전술한 바와 같으며, 자폐 범주성 장애의 예방 또는 개선을 목적으로 사료첨가제 조성물로 첨가할 수 있다. 본 발명의 사료첨가제는 사료관리법상의 보조사료에 해당한다.
본 발명에서 용어 "사료"는 동물이 먹고, 섭취하며, 소화시키기 위한 또는 이에 적당한 임의의 천연 또는 인공 규정식, 한끼식 등 또는 상기 한끼식의 성분을 의미할 수 있다. 상기 사료의 종류는 특별히 제한되지 아니하며, 당해 기술 분야에서 통상적으로 사용되는 사료를 사용할 수 있다. 상기 사료의 비제한적인 예로는, 곡물류, 근과류, 식품 가공 부산물류, 조류, 섬유질류, 제약 부산물류, 유지류, 전분류, 박류 또는 곡물 부산물류 등과 같은 식물성 사료; 단백질류, 무기물류, 유지류, 광물성류, 유지류, 단세포 단백질류, 동물성 플랑크톤류 또는 음식물 등과 같은 동물성 사료를 들 수 있다. 이들은 단독으로 사용되거나 2종 이상을 혼합하여 사용될 수 있다.
또한, 본 발명은 아가토바쿨룸 속(Agathobaculum sp.) 균주, 상기 균주 유래 소포체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상을 유효성분으로 함유하는 자폐 범주성 장애의 예방 또는 치료용 동물의약품 조성물을 제공한다.
상기 조성물은 바람직하게는 자폐 범주성 장애의 예방 또는 치료 효과를 가짐으로써 달성되는 것일 수 있으며, 이에 대해서는 상기에서 설명한 바와 같다.
본 발명의 조성물을 동물의약품 조성물로 사용할 경우, 상기 조성물을 그대로 사용하거나 다른 의약품 또는 의약외품 성분과 함께 사용할 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있으며 이에 제한되는 것은 아니다. 유효성분의 혼합양은 사용 목적(예방, 건강, 개선 또는 치료적 처치)에 따라 적합하게 결정될 수 있다.
이하, 실시예를 통하여 본 발명의 구성 및 효과를 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것일 뿐, 본 발명의 범위가 이들 실시예에 의해 한정되는 것은 아니다.
동물모델과 SR79 균주 투여
실험동물로서, C57BL6/J 마우스를 사용하였으며 임신마우스에 VPA를 투여하여 태어난 자폐증 모델 수컷 마우스에 대해 생후 3주부터 장 유래 미생물 SR79 균주를 1.0 × 109 CFU 농도로 매일 경구로 투여하고 미생물 투여 4주 이후부터 미생물 투여에 대한 영향을 마우스의 신경발달 질환에 관련된 행동 실험을 수행하였다. 이때, 대조군(vehicle)에는 25% 글리세롤/PBS를 SR79와 동일 부피로 경구 투여하였다.
또한, 자폐 범주성 장애 동물모델로는 세계적으로 널리 사용되고 있는 inbred BTBR T+tf/J (BTBR) 마우스 모델 수컷을 사용하여 생후 3주부터 장 유래 미생물 SR79 균주를 1.0 × 109 CFU 농도로 매일 경구로 투여하고 미생물 투여 4주 이후부터 미생물 투여에 대한 영향을 마우스의 신경발달 질환에 관련된 행동 실험을 수행하였다. 이때, 대조군 (vehicle)에는 25% 글리세롤/PBS를 SR79와 동일 부피로 경구투여 하였다. 이때, 대조군 마우스는 동일 주령의 C57BL6/J 수컷 마우스를 사용하였다.
실시예 1. 마우스의 반복적행동과 과활동성 분석
도 1은 마우스의 반복적인 행동(A) 및 과활동성(B) 대한 마우스의 행동을 분석한 결과이다. VPA 투여 마우스는 grooming 행동을 반복적으로 하는 횟수가 통계적으로 유의하게 증가되어 있으나 VPA 투여 그룹에 SR79 균주를 먹인 그룹은 반복적인 행동이 현저히 감소되는 것을 확인하였다(A, ##p <0.01). 또한, 마우스의 rearing 행동 분석을 통해 마우스의 과활동성 행동을 분석한 결과에서도 VPA 투여 마우스에서 나타나는 과활동성 행동이 SR79 투여에 의해 의미있게 감소하는 것을 확인하였다(B, ##p <0.01).
실시예 2. 마우스의 사회성행동 분석
도 2는 마우스의 사회성 행동을 측정하기 위해 다른 마우스와의 상호작용 행동(sniffing behavior)을 관찰하여 시간을 측정한 것으로 VPA 투여 마우스의 감소된 사회성 행동이 SR79 투여에 의해 유의미하게 증가하는 것을 확인하였다(##p <0.01).
또한, 도 3은 마우스의 사회성 행동을 측정하기 위해 다른 마우스와의 상호작용 행동(sniffing behavior)을 관찰하여 시간을 BTBR 마우스에서 측정한 것으로 대조군인 C57BL6/J 마우스에 비해 감소된 상호작용 행동(sniffing behavior)이 SR79 투여에 의해 유의미하게 증가하는 것을 확인하였다(#p <0.05).
실시예 3. 마우스의 인지기억력 테스트
도 4는 마우스의 인지기능 개선 효과를 테스트한 것으로 이전에 본 물체와 함께 새로운 물체를 제공하였을 때 새로운 물체에 대한 인지기억력을 테스트하는 novel object recognition test를 사용하였다. 이러한 새로운 물체 인지 학습 기억력이 VPA 투여 마우스에서 현저히 감소되어 있는 것을 확인할 수 있었으며, 이러한 인지학습 기억력은 SR79 균주 투여에 의해 의미있게 개선되는 것을 확인하였다(A, 새로운 물체 탐색 시간; B, 새로운 물체 탐색 횟수, ##p <0.01). 또한 마우스의 작업행동에 대한 Y-maze alteration test에서 새로운 공간으로 들어가는 alteration 퍼센트를 측정한 결과(도 4C), VPA 투여에 의한 작업기억력 감소가 SR79 균주 투여에 의해 의미있게 증가하는 것을 관찰하였다(#p <0.05). 또한, 도 5는 BTBR 마우스의 인지기능 개선 효과를 새로운 물체 탐색 시간을 측정하여 테스트한 것으로, SR79 균주 투여에 의해 의미있게 개선되는 것을 확인하였다.
Claims (9)
- 아가토바쿨룸 속(Agathobaculum sp.) 균주, 상기 균주 유래 소포체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상을 유효성분으로 함유하는 자폐 범주성 장애의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서, 상기 아가토바쿨룸 속 균주는 아가토바쿨룸 부트리시프로두센스(Agathobaculum butyriciproducens)인 것을 특징으로 하는 자폐 범주성 장애의 예방 또는 치료용 약학 조성물.
- 제2항에 있어서, 상기 아가토바쿨룸 부트리시프로두센스는 기탁번호가 KCTC 13036BP인 아가토바쿨룸 부트리시프로두센스 SR79 균주인 것을 특징으로 하는 자폐 범주성 장애의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서, 상기 자폐 범주성 장애는 반복적 행동, 과활동성, 사회성 결핍 또는 인지-기억 기능 장애 행동인 것을 특징으로 하는 자폐 범주성 장애의 예방 또는 치료용 약학 조성물.
- 아가토바쿨룸 속(Agathobaculum sp.) 균주, 상기 균주 유래 소포체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상을 유효성분으로 함유하는 자폐 범주성 장애의 예방 또는 개선용 건강기능식품 조성물.
- 제5항에 있어서, 상기 아가토바쿨룸 속 균주는 아가토바쿨룸 부트리시프로두센스(Agathobaculum butyriciproducens)인 것을 특징으로 하는 자폐 범주성 장애의 예방 또는 개선용 건강기능식품 조성물.
- 제6항에 있어서, 상기 아가토바쿨룸 부트리시프로두센스는 기탁번호가 KCTC 13036BP인 아가토바쿨룸 부트리시프로두센스 SR79 균주인 것을 특징으로 하는 자폐 범주성 장애의 예방 또는 개선용 건강기능식품 조성물.
- 아가토바쿨룸 속(Agathobaculum sp.) 균주, 상기 균주 유래 소포체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상을 유효성분으로 함유하는 자폐 범주성 장애의 예방 또는 개선용 사료첨가제 조성물.
