우리나라는 핵가족화, 개인화, 급속한 경제성장, 빠른 고령화 등 많은 변화를 겪고 있고, 경제협력개발기구(OEDC) 국가에서 자살률이 현재 1위로서 우울증에 대한 사회적 관심 및 보건정책 수립의 중요성이 어느 때보다 강조되는 시점이다.
현재까지는 우울증의 발생기전이 명확히 밝혀져 있지 않으며, 중추신경계 시냅스의 모노아민계 신경전달물질인 세로토닌, 노르에피네프린, 도파민 등의 결핍을 가장 유력한 가설로 여기고 있다. 따라서 대부분의 우울증 치료제는 중추신경계 시냅스의 모노아민계 신경전달 물질의 농도를 높이는 약리작용을 가지고 있다. 현재 상용되는 대표적인 우울증 치료제로는 벤조디아제핀(benxodiazepine), 디아제팜(diazepam), 옥사제팜(oxazepam), 로라제팜(lorazepam), 알프라졸람(alprazolam), 헬라제팜(helazepam), 클로나제팜(clonazepam), 인도메타신(indomethacin) 등이 있는데 이 약품들은 진정 및 수면유도제로도 사용된다.

본 발명의 목적을 달성하기 위하여, 본 발명은 장내 미생물 균주, 상기 균주 유래 소포체, 상기 균주의 단백체 및 대사체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상을 유효성분으로 함유하는 우울증의 예방 또는 치료용 약학 조성물을 제공한다.
본 발명에서, 상기 장내 미생물 균주는 아커만시아 뮤시니필라(Akkermansia muciniphila) 및 아가토바쿨룸 부티리시프로두센스(Agathobaculum butyriciproducens) 중에서 선택된 어느 하나 이상의 균주일 수 있다.
본 발명의 한 구현예로서, 아커만시아 뮤시니필라(Akkermansia muciniphila) 균주는 미국균주은행(ATCC, American Type Culture Collection)으로부터 분양받아서 본 발명에 사용하였으며(미국균주은행 기탁번호 ATCC BAA-835, 상기 균주는 독일생물자원은행 기탁번호 DSMZ 22959와 동일한 균주임), 아가토바쿨룸 부티리시프로두센스 SR79(Agathobaculum butyriciproducens SR79) 균주는 한국생명공학연구원(KCTC)에 2016년 06월 07일자로 기탁한 균주를 사용하였다(기탁번호: KCTC13036BP, 한국등록특허 10-1799830).
상기 약학 조성물은 상기 균주 유래 소포체, 상기 균주의 단백체 및 대사체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액으로부터 유래하는 추출물 또는 분획물, 현탁액 등의 형태를 제한 없이 포함할 수 있다.
본 발명의 용어 "배양액"은 균주를 배양하여 수득된 배양액 자체, 또는 이로부터 균주를 제거하여 수득된 배양 상층액의 농축물 또는 동결건조물을 포함할 수 있다.
본 발명에 있어서, 상기 배양액은 본 발명의 장내 미생물 균주를 배양하여 얻어진 결과물을 의미한다. 넓은 의미로 상기 배양물은 상기 장내 미생물 균주의 전체 배양물, 배양상등액, 파쇄물, 이들의 분획물 등이 될 수 있다. 이때, 상기 배양상등액은 상기 균주의 배양물을 원심분리하여 수득할 수 있고, 상기 파쇄물은 상기 균주를 물리적으로 또는 초음파처리하여 수득할 수 있으며, 상기 분획물은 상기 배양물, 배양상등액, 파쇄물 등을 원심분리, 크로마토그래피 등의 방법에 적용하여 수득할 수 있다.
본 발명의 용어 "배양"이란, 미생물을 적당히 인공적으로 조절한 환경조건에서 생육시키는 일련의 행위를 의미한다.
본 발명에 있어서, 상기 배양은 본 발명에서 제공하는 장내 미생물 균주를 배양하는 방법을 의미하는 것으로 해석될 수 있는데, 상기 배양방법은 당업계에 널리 알려져 있는 방법을 이용하여 수행할 수 있다. 구체적으로 상기 배양은 배치 공정 또는 주입 배치 또는 반복 주입 배치 공정(fed batch or repeated fed batch process)에서 연속식으로 수행할 수 있다.
상기 장내 미생물 균주를 배양하기 위하여는 뮤신, 적당한 탄소원, 질소원, 아미노산, 비타민 등을 함유한 통상의 배지 내에서 혐기성 조건 하에서 온도, pH 등을 조절하면서 적절한 방식으로 특정 균주의 생존요건을 충족시켜야 한다. 사용될 수 있는 탄소원으로는 주로 뮤신이나 N-아세틸글루코사민이며, 글루코즈 및 자일로즈의 혼합당을 탄소원으로 사용할 수 있고, 이외에 갈락토사민, 수크로즈, 락토즈, 프락토즈, 말토즈, 전분, 셀룰로즈와 같은 당 및 탄수화물, 대두유, 해바라기유, 피마자유, 코코넛유 등과 같은 오일 및 지방, 팔미트산, 스테아린산, 리놀레산과 같은 지방산, 글리세롤, 에탄올과 같은 알코올, 아세트산과 같은 유기산이 포함된다. 이들 물질은 개별적으로 또는 혼합물로서 사용될 수 있다. 사용될 수 있는 질소원으로는 암모니아, 황산암모늄, 염화암모늄, 초산암모늄, 인산암모늄, 탄산안모늄, 및 질산암모늄과 같은 무기질소원; 글루탐산, 메티오닌, 글루타민과 같은 아미노산 및 소이톤(soytone), 트립톤, 펩톤, NZ-아민, 육류 추출물, 효모 추출물, 맥아 추출물, 옥수수 침지액, 카세인 가수분해물, 어류 또는 그의 분해생성물, 탈지 대두 케이크 또는 그의 분해생성물 등 유기질소원이 사용될 수 있다. 이들 질소원은 단독 또는 조합되어 사용될 수 있다. 상기 배지에는 인원으로서 인산 제1칼륨, 인산 제2칼륨 및 대응되는 소듐-함유 염이 포함될 수 있다. 사용될 수 있는 인원으로는 인산이수소칼륨 또는 인산수소이칼륨 또는 상응하는 나트륨-함유 염이 포함된다. 또한, 무기화합물로는 염화나트륨, 염화칼슘, 염화철, 황산마그네슘, 황산철, 황산망간 및 탄산칼슘 등이 사용될 수 있다. 마지막으로, 상기 물질에 더하여 아미노산 및 비타민과 같은 필수 성장 물질이 사용될 수 있다. 또한, 지방산 폴리글리콜 에스테르와 같은 소포제를 사용하여 기포 생성을 억제할 수 있다.
본 발명의 용어 "소포체(vesicle)"는 세포 내에 존재하는 비교적 작은 주머니 형태의 구획으로, 화학 물질을 감싸서 이동시키는 데 사용하는 구조물을 의미한다. 상기 소포체는 소낭과 혼용되어 사용될 수 있다.
본 발명의 용어 "단백체"는 특정한 세포, 조직 또는 생물체 등 다양한 생물학적 개체에서 발현하고 있는 모든 단백질의 총집합을 의미한다.
본 발명의 용어 "대사체"는 세포, 조직, 체액과 같은 생물학적 시료 내에 존재하는 대사물질들의 총체를 의미한다. 대사체는 일반적으로 1500달톤(Dalton) 이하의 물질들로 구성되어 있고 생체 내 대사물질(endogenous metabolome; 예를 들면, 아미노산, 핵산, 지방산, 당류, 아민류, 당지질, 짧은 펩티드, 비타민, 호르몬 등)과 생체 외 대사물질(exogenous metabolome; 예를 들면, 약물, 음식, 첨가제, 독성 물질 등)로 분류된다. 이때 DNA, RNA, 단백질이 분해되어 생성되는 표적물질도 대사체로 분류되기도 한다. 대사체는 체내 대사의 영향을 받아 매 초마다 빠르고 역동적으로 변하기 때문에 유전체나 단백체에 비해 약물의 반응이나 음식으로 인한 영양분의 변화와 같은 표현형(phenotype)을 즉각적으로 반영한다.
본 발명의 용어 "추출물"은 본 발명에서 제공하는 장내 미생물 균주를 추출 처리하여 얻어지는 추출액, 상기 추출액의 희석액이나 농축액, 상기 추출액을 건조하여 얻어지는 건조물, 상기 추출액의 조정제물이나 정제물, 또는 이들의 혼합물 등, 추출액 자체 및 추출액을 이용하여 형성 가능한 모든 제형의 추출물을 포함한다. 상기 추출물을 얻는 추출 방법은 특별히 제한되지 않으며, 당해 기술 분야에서 통상적으로 사용하는 방법에 따라 추출할 수 있다. 상기 추출 방법의 비제한적인 예로는, 열수 추출법, 초음파 추출법, 여과법, 환류 추출법 등을 들 수 있으며, 이들은 단독으로 수행되거나 2종 이상의 방법을 병용하여 수행될 수 있다. 상기 추출물을 얻는 데 사용되는 추출 용매의 종류는 특별히 제한되지 않으며, 당해 기술 분야에서 공지된 임의의 용매를 사용할 수 있다. 상기 추출 용매의 비제한적인 예로는 물, 알코올 또는 이들의 혼합 용매 등을 들 수 있으며, 이들은 단독으로 사용되거나 1종 이상 혼합하여 사용될 수 있다.
본 발명의 용어 "분획물"은 여러 다양한 구성 성분들을 포함하는 혼합물로부터 특정 성분 또는 특정 성분 그룹을 분리하기 위하여 분획을 수행하여 얻어진 결과물을 의미한다. 상기 분획물을 얻는 분획 방법은 특별히 제한되지 않으며, 당해 기술 분야에서 통상적으로 사용하는 방법에 따라 수행될 수 있다. 상기 분획물을 얻는 데 사용되는 분획 용매의 종류는 특별히 제한되지 않으며, 당해 기술 분야에서 공지된 임의의 용매를 사용할 수 있다. 상기 분획 용매의 비제한적인 예로는 물, 알코올 등의 극성 용매; 헥산(Hexan), 에틸 아세테이트(Ethyl acetate), 클로로포름(Chloroform), 디클로로메탄(Dichloromethane) 등의 비극성 용매 등을 들 수 있다. 이들은 단독으로 사용되거나 1종 이상 혼합하여 사용될 수 있다.
본 발명의 용어 "현탁액"은 액체 속에 미세한 고체의 입자가 용해되지 않은 채 액체 속에 균일하게 분산되어 있는 혼합물을 의미한다.
본 발명에 있어서, 상기 현탁액은 동결건조된 일정 수 이상의 균주가 물 또는 액체 배양배지(액체 배지, media)에 분산된 것일 수 있다.
본 발명에서 용어 "우울증"은 의욕 저하와 우울감을 주요 증상으로 하여 다양한 인지 및 정신 신체적 증상을 일으켜 일상 기능의 저하를 가져오는 질환을 의미한다.
본 발명에서의 용어 "예방"이란 본 발명에 따른 약학적 조성물의 투여에 의해 우울증의 발병을 억제 또는 지연시키는 모든 행위를 의미하고, "치료"란 상기 약학적 조성물의 투여에 의해 우울증의 의심 및 발병 개체의 증상이 호전되거나 이롭게 변경되는 모든 행위를 의미한다.
본 발명의 상기 약학조성물은 약학적 조성물의 제조에 통상적으로 사용하는 적절한 담체, 부형제 및 희석제를 더 포함할 수 있다.
본 발명에 따른 상기 약학조성물은, 각각 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있다. 본 발명에 따른 상기 약학조성물에 포함될 수 있는 담체, 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유 등을 포함한 다양한 화합물 혹은 혼합물을 들 수 있다. 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다.
경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 상기 균주, 상기 균주 유래 소포체, 상기 균주의 단백체 및 대사체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 또는 상기 배양액의 추출물에 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘카보네이트(calcium carbonate), 수크로스(sucrose) 또는 락토오스(lactose), 젤라틴 등을 섞어 조제된다. 또한 단순한 부형제 이외에 마그네슘 스테아레이트, 탈크 같은 윤활제들도 사용된다. 경구를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다.
비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 비수성용제, 현탁제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로젤라틴 등이 사용될 수 있다.
본 발명의 약학적 조성물은 약제학적으로 유효한 양으로 투여될 수 있는데, 본 발명의 용어 "약제학적으로 유효한 양"이란 의학적 예방 또는 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 예방 또는 치료하기에 충분한 양을 의미하며, 유효 용량 수준은 질환의 중증도, 약물의 활성, 환자의 연령, 체중, 건강, 성별, 환자의 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 배합 또는 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 약학적 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적으로 또는 동시에 투여될 수 있다. 그리고 단일 또는 다중 투여될 수 있다. 상기 요소를 모두 고려하여 부작용없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하다.
본 발명의 약학적 조성물의 투여량은 예를 들어, 본 발명의 약학 조성물을 사람을 포함하는 포유동물에 하루 동안 1.0×109 CFU로 투여하는 것이 좋다. 본 발명의 조성물의 투여 빈도는 특별히 이에 제한되지 않으나, 1일 1회 투여하거나 또는 용량을 분할하여 수회 투여할 수 있다. 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다.
상기 목적을 달성하기 위한 또 다른 실시양태로서, 본 발명은 상기 약학적 조성물을 약제학적으로 유효한 양으로 개체에 투여하는 단계를 포함하는 우울증 예방 또는 치료방법을 제공한다.
상술한 바와 같이, 본 발명에서 제공하는 상기 장내 미생물 아커만시아 뮤시니필라(Akkermansia muciniphila) 균주 또는 아가토바쿨룸 부티리시프로두센스(Agathobaculum butyriciproducens) 균주, 상기 균주 유래 소포체, 상기 균주의 단백체 및 대사체, 상기 균주의 배양물, 상기 배양물의 농축액, 상기 배양물의 건조물 및 상기 배양물의 추출물로 이루어지는 군에서 선택된 1종 이상은 우울증 예방 또는 치료용 약학적 조성물의 유효성분으로서 사용할 수 있으므로, 상기 조성물은 우울증을 예방 또는 치료하는데 사용될 수 있다.
본 발명의 용어 "개체"란 우울증이 발병될 가능성이 있거나 또는 발병된 쥐, 가축, 인간 등을 포함하는 포유동물을 제한 없이 포함한다.
본 발명의 우울증을 치료하는 방법에 있어서, 상기 약학적 조성물의 투여 경로는 목적 조직에 도달할 수 있는 한 어떠한 일반적인 경로를 통하여도 투여될 수 있다. 본 발명의 약학적 조성물은 특별히 이에 제한되지 않으나, 경구 투여, 직장내 투여 등의 경로를 통해 투여할 수 있으며 경우에 따라 목적에 따라 다른 경로로도 투여될 수 있다. 다만, 경구 투여 시에는 위산에 의하여 상기 장내 미생물 아커만시아 뮤시니필라 및 아가토바쿨룸 부티리시프로두센스 균주가 변성될 수 있기 때문에 경구용 조성물은 활성 약제를 코팅하거나 위에서의 분해로부터 보호되도록 제형화 되어야 한다. 또한, 상기 조성물은 활성 물질이 표적 세포로 이동할 수 있는 임의의 장치에 의해 투여될 수 있다.
또한, 본 발명은 장내 미생물 균주, 상기 균주 유래 소포체, 상기 균주의 단백체 및 대사체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상을 유효성분으로 함유하는 우울증의 예방 또는 개선용 건강기능식품 조성물을 제공한다.
상기 균주에 대해서는 전술한 바와 같으며, 우울증 예방 또는 개선을 목적으로 건강기능식품에 첨가할 수 있다.
본 발명의 일 구현 예에 따른 우울증 예방 또는 개선용 건강기능식품 조성물은 상기 유효성분 이외에 추가로, 영양제, 비타민, 전해질, 풍미제, 착색제, 증진제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올 및 탄산음료에 사용되는 탄산화제 중에서 선택된 하나 이상을 더 함유할 수 있다.
상기에서 언급한 본 발명의 상기 균주 또는 이의 배양액 등을 건강기능식품 조성물로 사용할 경우, 상기 균주 또는 이의 배양액을 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용할 수 있으며, 통상적인 방법에 따라 적절하게 사용될 수 있다. 유효 성분의 혼합양은 그 사용 목적 (예방, 건강 또는 치료적 처치)에 따라 적합하게 결정될 수 있다.
본 발명의 식품(또는 건강기능식품)은, 식품 제조 시에 통상적으로 첨가되고 식품학적으로 허용되는 성분을 더 포함할 수 있다. 예컨대, 음료수로 제조되는 경우에는 본 발명의 균주 이외에 구연산, 액상과당, 설탕, 포도당, 초산, 사과산, 과즙 등에서 하나 이상의 성분을 추가로 포함시킬 수 있다.
본 발명에 따른 식품(또는 건강기능식품)의 유효성분으로 포함될 수 있는 양은 우울증 예방 또는 개선용 식품을 원하는 사람의 연령, 성별, 체중, 상태, 질병의 증상에 따라 적절히 선택될 수 있으며, 바람직하게는 성인기준 1일 0.01 g 내지 10.0 g 정도로 포함되는 것이 좋으며, 이러한 함량을 갖는 식품을 섭취함으로써 우울증 예방 또는 개선 효과를 얻을 수 있다.
또한, 본 발명은 아커만시아 뮤시니필라(Akkermansia muciniphila) 및 아가토바쿨룸 부티리시프로두센스(Agathobaculum butyriciproducens) 중 어느 하나 이상의 균주를 배양하는 단계를 포함하는 우울증 예방 또는 치료용 미생물 제제를 제조하는 방법을 제공한다.
본 발명의 균주를 배양하는 방법은 당업계에 통상적으로 이용되는 방법에 따라 배양할 수 있다. 이에 대해서는 전술한 바와 같다.
본 발명에 따른 우울증 예방 또는 치료용 미생물 제제는 용액, 분말, 현탁액, 분산액, 에멀젼, 유성 분산액, 페이스트, 분진, 분사제 또는 과립제로 제조할 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 또 다른 하나의 양태는, 본 발명의 장내 미생물 균주, 상기 균주 유래 소포체, 상기 균주의 단백체 및 대사체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상을 유효성분으로 함유하는 우울증의 예방 또는 개선용 사료첨가제 조성물을 제공한다.
상기 균주에 대해서는 전술한 바와 같으며, 우울증 예방 또는 개선을 목적으로 사료첨가제 조성물로 사용될 수 있다. 본 발명의 사료첨가제는 사료관리법상의 보조사료에 해당한다.
본 발명의 사료첨가제 조성물은 사료에 첨가될 수 있으며, "사료"는 동물이 먹고, 섭취하며, 소화시키기 위한 또는 이에 적당한 임의의 천연 또는 인공 규정식, 한끼식 등 또는 상기 한끼식의 성분을 의미할 수 있다. 상기 사료의 종류는 특별히 제한되지 아니하며, 당해 기술 분야에서 통상적으로 사용되는 사료를 사용할 수 있다. 상기 사료의 비제한적인 예로는, 곡물류, 근과류, 식품 가공 부산물류, 조류, 섬유질류, 제약 부산물류, 유지류, 전분류, 박류 또는 곡물 부산물류 등과 같은 식물성 사료; 단백질류, 무기물류, 유지류, 광물성류, 유지류, 단세포 단백질류, 동물성 플랑크톤류 또는 음식물 등과 같은 동물성 사료를 들 수 있다.
이하, 실시예를 통하여 본 발명의 구성 및 효과를 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것일 뿐, 본 발명의 범위가 이들 실시예에 의해 한정되는 것은 아니다.
실시예 1. 아커만시아 뮤시니필라(
Akkermansia muciniphila

) SR79 균주의 항우울증 효능 확인
실시예 1-1: 우울증 모델 마우스 제조 및 균주 배양액 처리
실험동물은 5주령의 ICR 수컷 마우스를 22-24℃의 온도를 유지하며 12시간씩 낮과 밤이 교대되는 환경에서 사육하였으며, 실험 개시 전 1주일 동안 사육시키며 적응기를 거친 후, 아커만시아 뮤시니필라(Akkermansia muciniphila, AK: 미국균주은행 기탁번호 ATCC BAA-835, 상기 균주는 독일생물자원은행 기탁번호 DSM 22959와 동일한 균주임) 또는 아가토바쿨룸 부티리시프로두센스(Agathobaculum butyriciproducens) SR79(KCTC13036BP) 균주 배양액을 1.0×109 CFU로 1회/1일씩 3일 동안 마우스에 각각 경구 투여하였다. 균주 배양액은 매일 오전 9시에서 9시 반 사이에 투여하였으며, 균주 배양액 이틀째에는 각각의 마우스에 LPS를 복강주입하였으며(1mg/kg, 복강주사), LPS 주입 30분 후 구속(restraint) 스트레스를 2시간 동안 가하여 마우스에 우울증을 유도하였다. 실험은 그룹당 마우스 9마리에서 19마리 사용하였으며, 균주 배양액의 항우울 효과에 대한 비교대조군으로서 항우울제로 사용되고 있는 인도메타신(indomethacin) 및 플루옥세틴(fluoxetine)을 각각 경구 투여하였다(도 1). 균주 배양액의 투여 마지막 날에 행동실험을 실시하였다. 모든 실험값은 평균±표준오차로 표시하였으며, 통계분석은 분산분석(ANOVA)와 스튜던트 티 검정(Student's t-test)으로 처리하였으며, 통계적인 유의적인 p<0.05 범위 내에서 허용하여 처리하였다.
실시예 1-2: 강제수영실험(Forced Swimming Test)
본 발명의 아커만시아 뮤시니필라(Akkermansia muciniphila, AK) 또는 아가토바쿨룸 부티리시프로두센스(Agathobaculum butyriciproducens) SR79(KCTC13036BP) 균주 배양액의 항우울 효과를 확인하기 위해서, 강제수영실험(Forced Swimming Test; FST)을 실시하였다. 구체적으로, 투명 비이커(지름 13cm 및 높이 19 cm)에 14 cm만큼 물을 채우고 마우스를 입수시켜 6분 동안 강제 수영시킨 다음, 4분을 기준으로 마우스가 움직임 없이 가만히 있는 시간(immobility time, sec, 마우스가 어떠한 미동도 없이 수면에 부유되는 상태; 부동시간)을 측정하였다. 통상, 우울증 마우스는 정상 마우스에 비해 부동시간이 긴 경향이 있다.
그 결과, 도 2에 개시한 바와 같이 우울증 모델 마우스(control)는 우울증을 유발하지 않은 마우스(vehicle)에 비해 부동시간이 현저히 증가한 것을 알 수 있었고, 이를 통해 LPS 투여 및 구속 스트레스 조건은 우울증 모델 마우스 제조에 충분하였음을 알 수 있었다. 또한, control군에 비해 본 발명의 AK 또는 SR79 균주 배양액을 투여한 실험군에서 부동시간이 유의미한 수준으로 감소된 것을 관찰할 수 있었는데, 이는 AK 또는 SR79 균주 배양액이 항우울 효과를 나타냄을 의미하였다. 특히, AK 또는 SR79 균주 배양액 처리 실험군은 항우울제로 사용되고 있는 인도메타신 및 플루옥세틴 처리군의 부동시간 감소와 유사하거나 보다 좋은 효과를 나타내어, AK 또는 SR79 균주 또는 이의 배양액이 우울증의 예방, 개선 또는 치료에 사용될 수 있음을 시사하였다.
실시에 1-3: 자발적 운동 활성(locomotor activity)
상기 강제수영실험에 앞서 우울증 모델 마우스와 균주 배양액 또는 약물을 처리한 우울증 모델 마우스를 대상으로 OFT(open-field test) 시험을 실시하여 자발적 운동 활성(locomotor activity)을 측정하였다. 구체적으로, OFT는 네모난 상자(전체 크기: 45 cm × 45 cm × 40 cm; 15 cm × 15 cm로 이루어진 9개의 구획을 바닥에 표시하여 구획을 나눔)의 정 중앙에 실험동물을 한 마리씩 넣어서 10분간 자발적으로 보행시킨 후, 네 개의 발을 이용하여 이동한 칸의 횟수를 측정하여 각 군마다 평균값을 이용하여 활동성을 측정하였다.
그 결과, 도 3에 개시한 바와 같이 우울증 모델 마우스(control)는 우울증을 유발하지 않은 마우스(vehicle)에 비해 운동성이 감소된 것으로 확인되었으나, control군과 균주 배양액 또는 약물을 처리한 우울증 모델 마우스간에는 자발적 운동 활성 차이가 없음을 확인하였다. 이를 통해, 본 발명의 AK 및 SR79 균주 투여에 의한 항우울 효능은 정신의학적 효과(psychiatric effect)이며, 물리적 효과(physical effect)에 의한 것이 아님을 확인할 수 있었다.
실시예 2. 아커만시아 뮤시니필라(
Akkermansia muciniphila
, AK) 및 아가토바쿨룸 부티리시프로두센스(
Agathobaculum butyriciproducens
) SR79 균주의 만성 우울증 예방 및 치료 효능 확인
실시예 2-1: 만성 우울증 모델 마우스 제조 및 균주 처리
실험동물은 5주령의 ICR 수컷 마우스를 22-24℃의 온도를 유지하며 12시간씩 낮과 밤이 교대되는 환경에서 사육하였으며, 실험 개시 전 1주일 동안 사육시키며 적응기를 거친 후 사용하였다.
아커만시아 뮤시니필라(Akkermansia muciniphila, AK: 미국균주은행 기탁번호 ATCC BAA-835, 상기 균주는 독일생물자원은행 기탁번호 DSM 22959와 동일한 균주임) 또는 아가토바쿨룸 부티리시프로두센스(Agathobaculum butyriciproducens) SR79(KCTC13036BP) 균주 배양액을 마우스 당 150 uL 투여량으로 1회/1일씩 21일간 매일 투여하였다. 균주 배양액 투여 후, 마우스에 LPS투여 없이, 3주간 매일 2시간씩 구속(restraint) 스트레스를 가하여 마우스에 만성 우울증을 유도하였다. 스트레스를 가한 후 행동실험을 수행하였다. 한편 균주 배양액의 항우울 효과에 대한 비교대조군으로서, 항우울제로 사용되고 있는 플루옥세틴(fluoxetine)을 10㎎/㎏(in 33% DMSO)용량으로 경구 투여하였다. 실험은 그룹당 마우스 10마리를 사용하였고, 모든 실험값은 평균±표준오차로 표시하였으며, 통계분석은 분산분석(ANOVA)와 스튜던트 티 검정(Student's t-test)으로 처리하였으며, 통계적인 유의적인 p<0.05 범위 내에서 허용하여 처리하였다.
실시예 2-2: 강제수영실험(Forced Swimming Test)
실시에 2-1에서 제조한 마우스 모델의 항우울 효과를 확인하기 위해서, 강제수영실험(Forced Swimming Test; FST)을 실시하였다. 강제수영 실험 방법은 실시예 1-2와 동일하게 수행하였다.
그 결과, 도 5에 개시한 바와 같이 우울증 모델 마우스(control)는 우울증을 유발하지 않은 마우스(vehicle)에 비해 부동시간이 현저히 증가한 것을 알 수 있었고, 이를 통해 LPS 없이도 구속 스트레스 조건만으로 우울증 모델 마우스가 제조되었음을 확인하였다.
또한, control군에 비해 본 발명의 AK 또는 SR79 균주 배양액을 투여한 실험군에서 부동시간이 유의미한 수준으로 감소된 것을 관찰할 수 있었는데, 이는 AK 또는 SR79 균주가 만성 우울증 동물 모델에서도 항우울 효과를 나타냄을 의미한다. 특히, AK 또는 SR79 균주 처리 실험군은 항우울제로 사용되고 있는 플루옥세틴 처리군의 부동시간 감소와 유사한 효과를 나타내었으며, 우울증을 유도하지 않은 Vehicle 군과 유사한 수준으로 감소하여 거의 정상수준으로 회복된 것을 확인하였다.
이를 통해, AK 또는 SR79 균주 또는 이의 배양액이 우울증의 예방, 개선 또는 치료에 사용될 수 있음을 확인하였으며, 만성 우울증의 치료에도 사용될 수 있음을 확인하였다.
실시예 2-3: 자발적 운동 활성(locomotor activity)
실시에 2-1에서 제조한 마우스 모델의 항우울 효과를 확인하기 위해서, 상기 실시예 2-2의 강제수영실험 전에, OFT(open-field test) 시험을 실시하여 자발적 운동 활성(locomotor activity)을 측정하였다. 실시예 1-3과 동일한 방법으로 실험을 수행하였다.
그 결과, 도 6에 개시한 바와 같이 우울증 모델 마우스(control)는 우울증을 유발하지 않은 마우스(vehicle)에 비해 운동성이 감소된 것으로 확인되었으나, control군과 균주 배양액 또는 약물을 처리한 우울증 모델 마우스간에는 자발적 운동 활성 차이가 없음을 확인하였다. 이를 통해, 본 발명의 AK 및 SR79 균주 투여에 의한 만성적 항우울 효능 역시 정신의학적 효과(psychiatric effect)이며, 물리적 효과(physical effect)에 의한 것이 아님을 확인할 수 있었다.
이상의 설명으로부터, 본 발명이 속하는 기술분야의 당업자는 본 발명이 그 기술적 사상이나 필수적 특징을 변경하지 않고서 다른 구체적인 형태로 실시될 수 있다는 것을 이해할 수 있을 것이다. 이와 관련하여, 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적인 것이 아닌 것으로 이해해야만 한다. 본 발명의 범위는 상기 상세한 설명보다는 후술하는 특허 청구범위의 의미 및 범위 그리고 그 등가 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.
Claims (8)
- 장내 미생물 균주, 상기 균주 유래 소포체, 상기 균주의 단백체 및 대사체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상을 유효성분으로 함유하는 우울증의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서, 상기 장내 미생물 균주는 아커만시아 뮤시니필라(Akkermansia muciniphila) 균주 및 아가토바쿨룸 부티리시프로두센스(Agathobaculum butyriciproducens) 균주 중에서 선택되는 1 이상의 균주인 것을 특징으로 하는 우울증의 예방 또는 치료용 약학 조성물.
- 제2항에 있어서, 상기 아커만시아 뮤시니필라(Akkermansia muciniphila) 균주는 기탁번호가 DSM22959이고, 상기 아가토바쿨룸 부티리시프로두센스(Agathobaculum butyriciproducens) 균주는 기탁번호가 KCTC13036BP인 것을 특징으로 하는 우울증의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서, 상기 유효성분 이외에 추가로 담체, 부형제 또는 희석제를 포함하는 것을 특징으로 하는 우울증의 예방 또는 치료용 약학 조성물.
- 장내 미생물 균주, 상기 균주 유래 소포체, 상기 균주의 단백체 및 대사체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상을 유효성분으로 함유하는 우울증의 예방 또는 개선용 건강기능식품 조성물.
- 제5항에 있어서, 상기 유효성분 이외에 추가로, 영양제, 비타민, 전해질, 풍미제, 착색제, 증진제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올 및 탄산음료에 사용되는 탄산화제 중에서 선택된 하나 이상을 더 함유하는 것을 특징으로 하는 우울증의 예방 또는 개선용 건강기능식품 조성물.
- 장내 미생물 균주, 상기 균주 유래 소포체, 상기 균주의 단백체 및 대사체, 상기 균주의 배양액, 상기 배양액의 농축액, 상기 배양액의 건조물 및 상기 배양액의 추출물로 이루어지는 군에서 선택된 1종 이상을 유효성분으로 함유하는 우울증의 예방 또는 개선용 사료첨가제 조성물.
- 제1항 내지 제4항 중 어느 한 항의 조성물을 개체에 투여하는 단계를 포함하는, 우울증의 예방 또는 치료 방법.
